Vortioxetine as an analgesic in preclinical inflammatory pain models: Mechanism of action

AB  - Vortioxetine is a novel atypical antidepressant with multimodal activity that has recently demonstrated efficacy against neuropathic pain. There is no published data about its analgesic properties in models characterized by peripheral inflammation and consequent pain pathway sensitization, nor data on its mechanism of antinociceptive action. This study aimed to investigate vortioxetine's antinociceptive/antihyperalgesic effects in trigeminal, visceral, and somatic inflammatory pain models, and provide evidence on its mechanism of action in the modulation of trigeminal nociception. Vortioxetine's effects on the nociceptive behavior in orofacial formalin test (OFT) and acetic acid-writhing test in mice and on mechanical hyperalgesia in carrageenan-induced paw inflammation in rats were examined following peroral single administration. The involvement of serotonergic/adrenergic/cholinergic/cannabinoid/adenosine receptors was evaluated in OFT by intraperitoneally treating mice with an appropriate antagonist immediately after vortioxetine application. We used antagonists of 5-HT1B/1D serotonergic (GR 127935), α1-adrenergic (prazosin), α2-adrenergic (yohimbine), β1-adrenergic (metoprolol), muscarinic (atropine), α7 nicotinic (methyllycaconitine), CB1/CB2 cannabinoid (AM251 and AM630), and adenosine A1 (DPCPX) receptors. Vortioxetine dose-dependently reduced pain behavior in OFT and acetic acid writhing test, as well as inflammatory hyperalgesia in paw pressure test. All examined antagonists except prazosin dose-dependently inhibited vortioxetine's antinociceptive effects. In conclusion, vortioxetine exerted analgesic efficacy in trigeminal, visceral, and somatic inflammatory pain. The effect is at least in part mediated by 5-HT1B/1D serotonergic, α2/β1-adrenergic, muscarinic and nicotinic cholinergic, CB1/CB2 cannabinoid, and adenosine A1 receptors. These findings contribute to better understanding of the analgesic effect of vortioxetine and suggest its potential usefulness for inflammatory pain treatment.

AB  - Chronic pain and depression commonly occur together so dual-acting agents might be particularly useful. The population of patients with chemotherapy-induced neuropathy is increasing in parallel with the increase of population of cancer survivors and there is a compelling need for satisfactory treatment of symptoms of neuropathy and concomitant depression. We examined the effects of vortioxetine, a novel antidepressant with unique mechanism of action, on pain hypersensitivity and depression-like behavior in oxaliplatin-induced neuropathy model in mice (OIPN). Vortioxetine (1–10 mg/kg, p.o.) significantly and dose-dependently reduced mechanical allodynia in von Frey test and cold allodynia in acetone test in OIPN mice, in both repeated prophylactic and acute therapeutic treatment regimens. It also reduced depression-like behavior in the forced swimming test in OIPN mice, in both treatment paradigms. Its antiallodynic and antidepressive-like effects were comparable to those exerted by duloxetine (1–15 mg/kg, p.o.). The antiallodynic and antidepressive-like effects of repeatedly administered vortioxetine might be related to the increased content of 5-hydroxytryptamine (5- HT) and noradrenaline (NA), detected in the brainstem of treated OIPN mice. These results indicate that vortioxetine could be potentially useful in prevention and treatment of chemotherapy-induced neuropathy, for the relief of pain and concomitant depressive symptoms. It should be further tested to this regard in clinical settings.

Investigation of matrix effect in spectrochemical determination of trace elements in biscuits

AB  - In this paper the content of trace elements (Cu, Mn, Fe and Zn) were determined in biscuits with addition of different kinds of dry fruits. For all determinations the pretreatment procedures were optimized. The elements were determined by flame atomic absorption spectrophotometry and atomic emission spectrometry with inductively coupled plasma and argon stabilized U-shaped d.c. arc as excitation sources. It is shown that matrix effect originating from alkaline and alkaline earth elements is different in different excitation sources and depends on analyte type. Therefore matrix matching is indispensable.
AB  - U ovom radu određen je sadržaj tragova elemenata (Cu, Mn, Fe i Zn) u keksima sa dodatkom različitih vrsta sušenog voća. Za sva određivanja optimizovane su procedure pripreme. Elementi su određivani plamenom atomskom apsorpcionom spektrometrijom i atomskom emisionom spektrometrijom, sa induktivno spregnutom plazmom i argonom stabilizovanim U-lukom jednosmerne struje, kao ekscitacionim izvorima. Pokazalo se da je matriks efekat, koji potiče od alkalnih i zemnoalkalnih elemenata, različit sa različitim ekscitacionim izvorima i da zavisi od tipa analita. Stoga je neophodno "poklapanje" matriksa pri tačnom određivanju tragova elemenata.

AB  - The plasma of a vertical argon stabilized DC arc at atmospheric pressure is applied as a spectrochemical source. The lateral distributions of relative spectral line intensities of some trace elements (Zn, Pt, Cd, Mg, Ca and Al) introduced into the plasma in the form of aqueous and ethanol–aqueous solutions were experimentally determined. These distributions were correlated with the calculated equilibrium plasma composition of the arc plasma. On the basis of the obtained results, an explanation of the influence of ethanol addition on the radiation densities from an arc plasma is given.
AB  - Plazma argonom stabilizovanog vertikalnog jednosmernog luka na atmosferskom pritisku korišćena je kao spektrohemijski izvor. Eksperimentalno su određene lateralne raspodele intenziteta emisije nekoliko elemenata prisutnih u tragu (Zn, Pt,Cd, Mg,Ca i Al) u vodenim i etanol–vodenim rastvorima. Ove raspodele su dovedene u vezu sa izračunatim ravnotežnim sastavom lučne plazme i objavšnjen je uticaj etanola na gustinu zračenja iz lučne plazme.

AB  - Some structural properties of a magnesium salt of 12-tungstophosphoric acid, MgHPW12O40.nH(2)O (MgHVvTA-n), are reported. The influence of the Mg2+ ion on the structure, dynamics of the protonic species, and dielectric and conductive characteristics of the salt have been studied. The interaction of protonic species with Keggin anion was discussed. Preliminary results of the crystal structure indicate that the structure is composed of Keggin anions and Mg(OH2)(6) aqua-complexes. Some comparisons are made with other alkaline-earth and alkaline salts of WPA. The results of thermal analysis (TGA and DTA), X-ray diffraction (XRD), scanning electron microscopy (SEM), infrared spectroscopy (IR), conductivity and permittivity measurements of MgHWPA-n are reported and discussed.

AB  - The determination of lead in wines of different origin was performed by means of atomic emission spectroscopy with argon stabilized DC. U-shaped arc and electrothermal atomic absorption spectrometry. The comparison of the results obtained by the direct and standard addition method has indicated the presence of a depressive effect of the complex organic matrix. The effect is avoided successfully by mineralization, as well as by dissolution of the samples. Thus, a relative simple but precise and sensitive method involving the application of a stabilized arc and photoelectric detection with time integration of the emission signals is recommended for the determination of low concentrations of lead in wines. The complex organic matrix was investigated by recording the IR spectra of different wine fractions.
AB  - Određivanje olova u vinima različitog porekla vršeno je primenom atomske emisione spektroskopije sa argonom stabilisanim lukom U-oblika i atomske apsorpcione spektrofotometrije sa grafitnom kivetom. Poređenje rezultata dobijenih direktnom i metodom standardnog dodatka pokazalo je da postoji znatan depresivni efekat složene organske osnove. Efekat je uspešno izbegnut mineralizacijom ili razblaženjem uzorka. Kombinacijom razblaženja uzorka i primenom stabilizovanog luka i fotoelektrične detekcije sa vremenskom integraciijom emisionih signala razvijena je jednostavna i precizna metoda za određeivanje niskih koncentracija olova u vinima. Kompleksan organski matriks ispitivan je snimanjem IC spektara različitih frakcija vina.

AB  - In this paper the latest results of our continuing investigation of heteropoly acids and their salts are reported. Specially attention was paid to the influence of cations on the dynamic equilibrium of protonic species, as well as on the structure of the host lattice itself, i.e., the Keggin anions. The investigations were done by IR and Raman spectroscopy within the range of 1200-40 cm-1.
AB  - U ovom radu saopšteni su poslednji rezultati naših sistematskih ispitivanja heteropolikiselina i njihovih soli. Posebna pažnja posvećena je uticaju katjona na dinamičku ravnotežu protonskih vrsta, kao i na strukturu kristalne rešetke, odnosno Keginovih anjona. Ispitivanja su vršena metodama infracrvene i ramanske spektroskopije u oblasti 1200-40 cm-1. Dobijeni rezultati su važni za objašnjenje mehanizma provodljivosti, kao i za rešavanje problema praktične primene ovih jedinjenja kao čvrstih elektrolita. AB  - In this paper local processes and structural phase transformations of the copper salt of 12-tungstophosphoric acid are investigated. The structural phase transformations were followed through bands, characteristic for the host lattice, in the IR and Raman spectra. The results of these investigations, as well as those of XRPD analysis and impedance measurements as a function of temperature show that some local processes provoke the change in the secondary structure of the Keggin anions.

Strukturne fazne transformacije praćene su preko traka karakterističnih za kristalnu rešetku u infracrvenim i ramanskim spektrima. Rezultati ovih ispitivanja, kao i oni dobiveni rendgeno-strukturnom analizom i iz impedansnih merenja, odnosno provodljivost u funkciji temperature, pokazuju da je fazna transformacija vezana za promene lokalne strukture, tj. sekundarne strukture Keginovih anjona. AB  - This contribution presents the results of the investigation of Mg2+ and Ca2+ 12-tungstophosphoric acid (WPA) salts as superionic proton conductors. We have identified protonic species as charge carriers in several hydration degrees of these salts by IR spectroscopy at low, room and high temperatures. In the IR spectra we have found the characteristic frequencies for H3O+, OH-, H2O, H5O2+. Also, we have investigated the dynamic equilibrium between protonic entities in the function of temperature, as well as the nature and the strength of hydrogen bonds in those compounds. During the process of calcination, these salts undergo a few phase transformations. The temperatures of interest for the further investigations were taken from data of thermal analysis.

AB  - Heteropoly acids arc well known as supcrionic protonic conductors at room temperature. The conductivity depends upon the temperature and relative humidity of the surrounding. Recent results show that the conductivity is very much affected by monovalent exchange counter-ions. In this work the effect of monovalent ions (Li + , Na + , K + and Cs + ) on the nature and structure of hydrogen bonds, the presence of different protonic species as charge carriers, and the dynamic equilibrium of proton species as a function of temperature and ion characteristics were investigated.

abstract = "Heteropoly acids arc well known as supcrionic protonic conductors at room temperature. The conductivity depends upon the temperature and relative humidity of the surrounding. Recent results show that the conductivity is very much affected by monovalent exchange counter-ions. In this work the effect of monovalent ions (Li + , Na + , K + and Cs + ) on the nature and structure of hydrogen bonds, the presence of different protonic species as charge carriers, and the dynamic equilibrium of proton species as a function of temperature and ion characteristics were investigated.",
