AB  - Metals exert detrimental effects on various systems within the body, including the nervous system. Nevertheless, the dose-response relationship concerning the administration of low doses of metal mixtures remains inadequately explored. The assessment of neurotoxic effects of lead, cadmium, mercury, and arsenic mixture (MIX) administered at low dose ranges, was conducted using an in vivo approach. A subacute study was conducted on a rat model consisting of a control and five treatment groups subjected to oral exposure with gradually increasing doses (from MIX 1 to MIX 5). The results indicated that behavioural patterns in an already developed nervous system displayed a reduced susceptibility to the metal mixture exposure with tendency of higher doses to alter short term memory. However, the vulnerability of the mature brain to even minimal amounts of the investigated metal mixture was evident, particularly in the context of oxidative stress. Moreover, the study highlights superoxide dismutase's sensitivity as an early-stage neurotoxicity marker, as indicated by dose-dependent induction of oxidative stress in the brain revealed through Benchmark analysis. The narrowest Benchmark Dose Interval (BMDI) for superoxide dismutase (SOD) activity (1e-06 - 3.18e-05 mg As/kg b.w./day) indicates that arsenic may dictate the alterations in SOD activity when co-exposed with the other examined metals. The predicted Benchmark doses for oxidative stress parameters were very low, supporting “no-threshold” concept. Histopathological alterations were most severe in the groups treated with higher doses of metal mixture. Similarly, the brain acetylcholinesterase (AChE) activity demonstrated a dose-dependent decrease significant in higher doses, while BMDI suggested Cd as the main contributor in the examined metal mixture. These findings imply varying susceptibility of neurotoxic endpoints to different doses of environmentally relevant metal mixtures, advocating for risk assessment and regulatory measures to address metal pollution and enhance remediation strategies.

AB  - This study aimed to assess the redox status and trace metal levels in 49 shoe industry workers (11 men and 38 women) occupationally exposed to a mixture of volatile organic compounds (VOCs), which includes aliphatic hydrocarbons, aromatic hydrocarbons, ketones, esters, ethers, and carboxylic acids. All measured VOCs were below the permitted occupational exposure limits. The control group included 50 unexposed participants (25 men and 25 women). The following plasma parameters were analysed: superoxide anion (O2•-), advanced oxidation protein products (AOPP), total oxidative status (TOS), prooxidant-antioxidant balance (PAB), oxidative stress index (OSI), superoxide dismutase (SOD) and paraoxonase-1 (PON1) enzyme activity, total SH group content (SHG), and total antioxidant status (TAS). Trace metal levels (copper, zinc, iron, magnesium, and manganese) were analysed in whole blood. All oxidative stress and antioxidative defence parameters were higher in the exposed workers than controls, except for PON1 activity. Higher Fe, Mg, and Zn, and lower Cu were observed in the exposed vs control men, while the exposed women had higher Fe and lower Mg, Zn, and Cu than their controls. Our findings confirm that combined exposure to a mixture of VOCs, even at permitted levels, may result in additive or synergistic adverse health effects and related disorders. This raises concern about current risk assessments, which mainly rely on the effects of individual chemicals, and calls for risk assessment approaches that can explain combined exposure to multiple chemicals.
AB  - Cilj istraživanja bio je procijeniti redoks status i koncentracije metala u tragovima kod 49 radnika (11 muškaraca i 38 žena) zaposlenih u
industriji obuće, izloženih mješavini hlapljivih organskih spojeva (engl. volatile organic compounds, krat. VOC) koju su činili alifatski i aromatični
ugljikovodici, ketoni, esteri, eteri i karboksilne kiseline. Sve izmjerene razine VOC bile su ispod dozvoljenih granica profesionalne izloženosti.
U kontrolnoj je skupini bilo 50 neizloženih ispitanika (25 muškaraca i 25 žena). U plazmi obućarskih radnika analizirani su sljedeći parametri:
superoksid anion radikal (O2
•-
), uznapredovali produkti oksidacije proteina (AOPP), totalni oksidacijski status (TOS), prooksidacijsko-
antioksidacijski balans (PAB), indeks oksidacijskoga stresa (OSI), aktivnost superoksid-dismutaze (SOD), aktivnost enzima paraoksonaze-1
(PON1), ukupni sadržaj sulfhidrilnih grupa (SHG) i totalni antioksidacijski status (TAS). Koncentracije metala u tragovima (bakar, cink,
željezo, magnezij i mangan) analizirane su u punoj krvi. U usporedbi s kontrolnom skupinom, svi parametri oksidacijskoga stresa i
antioksidacijske obrane u izloženih radnika bili su povišeni, osim aktivnosti PON1. Povećana koncentracija Fe, Mg i Zn te smanjena
koncentracija Cu uočene su u izloženih muškaraca u usporedbi s kontrolnima, a u žena je primijećena povišena koncentracija Fe, a niža
koncentracija Mg, Zn i Cu u odnosu na kontrolnu skupinu. Naši nalazi potvrđuju da kombinirano izlaganje mješavini VOC, čak i pri
dozvoljenim koncentracijama, može dovesti do aditivnih ili sinergističkih štetnih učinaka na zdravlje i popratnih poremećaja. Trenutna
procjena rizika, koja se uglavnom oslanja na učinke pojedinačnih kemikalija, izaziva zabrinutost i upućuje na potrebu za novim pristupima
koji će objasniti kombiniranu izloženost većem broju kemikalija.

Antioxidative Effects of Black Currant and Cornelian Cherry Juices in Different Tissues of an Experimental Model of Metabolic Syndrome in Rats

AB  - A Western-style diet, rich in fat and simple sugars, is the main risk factor for a significant number of chronic diseases and disorders, as well as for a progression of metabolic syndrome (MetS). One of the key mechanisms involved in MetS development is increased oxidative stress caused by the accumulation of body fat. Some dietary polyphenols have shown a protective role in preventing oxidative-stress-induced damage. We investigated the difference in the oxidative response of plasma, liver, and visceral adipose tissue in rats fed with a high-fat high-fructose (HFF) diet for ten weeks, and the effectiveness of polyphenol-rich juices (black currant (BC) and cornelian cherry (CC)) in HFF-diet-induced oxidative stress prevention. The most prominent impact of the HFF diet on redox parameters was recorded in the liver, whereas adipose tissue showed the most potent protection mechanisms against oxidative stress. Consumption of both juices decreased advanced oxidation protein product (AOPP) level in plasma, increased paraoxonase1 (PON1) activity in the liver, and significantly decreased total oxidative status (TOS) in adipose tissue. BC exerted stronger antioxidative potential than CC and decreased the superoxide anion radical (O2•−) level in the liver. It also reduced TOS, total antioxidative status (TAS), and malondialdehyde (MDA) concentration in adipose tissue. The multiple linear regression analysis has shown that the best predictors of MetS development, estimated through the increase in visceral adiposity, were superoxide dismutase (SOD), AOPP, TOS, and TAS. The consumption of polyphenol-rich juices may provide a convenient approach for the systemic reduction of oxidative stress parameters.

We investigated the difference in the oxidative response of plasma, liver, and visceral adipose tissue in rats fed with a high-fat high-fructose (HFF) diet for ten weeks, and the effectiveness of polyphenol-rich juices (black currant (BC) and cornelian cherry (CC)) in HFF-diet-induced oxidative stress prevention. The most prominent impact of the HFF diet on redox parameters was recorded in the liver, whereas adipose tissue showed the most potent protection mechanisms against oxidative stress. Consumption of both juices decreased advanced oxidation protein product (AOPP) level in plasma, increased paraoxonase1 (PON1) activity in the liver, and significantly decreased total oxidative status (TOS) in adipose tissue. BC exerted stronger antioxidative potential than CC and decreased the superoxide anion radical (O2•−) level in the liver. It also reduced TOS, total antioxidative status (TAS), and malondialdehyde (MDA) concentration in adipose tissue. The multiple linear regression analysis has shown that the best predictors of MetS development, estimated through the increase in visceral adiposity, were superoxide dismutase (SOD), AOPP, TOS, and TAS. The consumption of polyphenol-rich juices may provide a convenient approach for the systemic reduction of oxidative stress parameters.",

AB  - Background: The aim of the study was to explore the mutual relationship between oxidative stress, inflammation and
metabolic biomarkers in subjects with prediabetes (PRE),
newly diagnosed type 2 diabetes patients (NT2D) and overt
type 2 diabetes (T2D) using principal component analysis
(PCA) as a thorough statistical approach.
Methods: Glycated hemoglobin, lipid parameters, inflammation (IL-6, CRP and fibrinogen) and oxidative stress
markers  pro-oxidants (AOPP, PAB, TOS) and antioxidants
(PON1, tSHG, TAS)  were measured. PCA was applied to
explore the factors that the most strongly influenced glucoregulation.
Results: A total of 278 subjects were (i.e., 37 PRE, 42
NT2D and 99 T2D) were compared with 100 healthy subjects as a control group (CG). PCA emphasized 4 different
factors explaining 49% of the variance of the tested parameters: oxidative stress-dyslipidemia related factor (with positive loading of TG and tSHG, and with negative loading of
HDL-c and TAS), dyslipidaemia related factor (i.e., total
cholesterol and LDL-c, both with positive loading),
Anthropometric related factor (i.e., waist and hip circumference, both with positive loading) and oxidative stress-
Inflammation related factor (i.e., PAB, fibrinogen, and CRP, all with positive loading). Out of these 4 factors, only oxidative stress – dyslipidaemia related factor showed a significant predictive capability towards poor glucoregulation. An
increase in this factor by one unit showed a 1.6 times higher probability for poor glucoregulation.
Conclusions: Redox imbalance (determined with lower TAS
and higher tSHG), in addition to higher TG and lower HDL-
c was associated with poor glucoregulation.
AB  - Uvod: Cilj istraživanja je bio da se ispita povezanost oksidativnog stresa, inflamacije i metaboličkih biomarkera kod pacijenata sa predijabetesom (PRE), de novo dijabetesom (NT2D) i ranije dijagnostikovanim dijabetesom (T2D) pomoću glavne komponentne analize (PCA). Metode: Glikozilirani hemoglobin, lipidni status, markeri inflamacije (IL-6, CRP i fibrinogen), parametri oksidativnog stresa pro-oksidanti (AOPP, PAB, TOS) i antioksidansi (PON1, tSHG, TAS) su mereni. PCA je primenjena da bi se ispitali faktori koji najviše utiču na glikoregulaciju. Rezultati: U istraživanje je uključeno 278 ispitanika: 37 PRE, 42 NT2D i 99 T2D, kao i 100 zdravih osoba koji su činili kontrolnu grupu (CG). AB  - Introduction: Phenols are a large family of natural and synthetic compounds with known antioxidant activity. The aim of this study was to perform in vitro screening of natural and natural-like phenol monomers and their C2-symmetric dimers (hydroxylated biphenyls) in order to identify those representatives whose pharmacophores have the strongest antioxidant and the lowest prooxidant activity. Material and methods: Antioxidative properties of 36 compounds (monomers and their C2-symmetric dimers) were evaluated in vitro. Different (red/ ox) assays were used to measure their total oxidative potential (TOP), their total antioxidative capacity (TAC), the pro-oxidative-antioxidant balance (PAB) and total SH-group content (SHG) in a biologically relevant environment. The Pro-oxidative Score, Antioxidative Score and the Oxy Score were also calculated. Trolox, a water soluble analogue of α-tocopherol, was used as a positive control. Results: In an assay consisting of pooled human serum, 6 of the 36 compounds showed significant antioxidant activity (compounds 6, 7, 12, 13, 26, and 27), whereas 4 showed extremely weak antioxidant activity (compounds 2, 29, 30, and 31). Within the 36 compounds comprising zingerone, dehydrozingerone, aurone, chalcone, and magnolol derivatives, in both monomeric and dimeric forms, the 2 compounds that indicated the highest antioxidant activity were dehydrozingerone derivatives (compounds 6 and 12). Trolox’s activity was found between the strong and weak antioxidant compounds analysed in our study. Conclusions: In this study selected dehydrozingerones were identified as good candidates for in-depth testing of their biological behaviour and for possible precursors for synthesis of novel polyphenolic molecules with potential therapeutic applications.

AB  - Introduction: Markers of iron homeostasis are related to insulin resistance (IR) in adults. However, studies in children and adolescents are scarce and show contradictory results. The aim was to evaluate the potential relationship between iron status markers and IR. Additionally, no previous study has explored the mutual effect of biomarkers of iron homeostasis and inflammation (i.e. high sensitivity C-reactive protein (hsCRP)), and adipokines (i.e. retinol-binding protein 4 (RBP4)) on IR in the cohort of adolescent girls. Material and methods: A total of 60 girls age between 16 and 19 years were included in the study. Serum levels of ferritin, transferrin, soluble transferrin receptor (sTfR), hsCRP, and RBP4 were measured by immunonephelometry. Homeostasis model assessment of insulin resistance (HOMA-IR) and iron homeostasis indexes were calculated. Univariate and multivariate binary logistic regression analysis were used to investigate the possible independent associations of the examined biomarkers. Principal component analysis was used to examine their mutual effect on HOMA-IR in the studied girls. Results: Ferritin, sTfR, hsCRP and RBP4 were significant predictors for higher HOMA-IR in univariate analysis (p = 0.020, p = 0.009, p = 0.007, p = 0.003, respectively). Multivariate regression analysis after adjustment for waist circumference (WC) showed that serum sTfR levels remained positively associated with higher HOMA-IR (p = 0.044). Factorial analysis revealed that the obesity-inflammation related factor (i.e., WC and hsCRP) and adipokine- acute phase protein related factor (i.e., RBP4 and ferritin) showed significant differences between HOMA-IR < 2.5 and HOMA-IR ≥ 2.5. Conclusions: Serum sTfR levels are independently associated with HOMA-IR, whereas higher serum ferritin levels together with higher RBP4 are related to higher HOMA-IR in adolescent girls.

AB  - Introduction: Telomeres are protective chromosomal ends. Short telomeres are a proven biomarker of biological aging. We aimed to find an association of telomere length and telomerase activity in circulating leukocytes and thromboaspirates of patients with acute myocardial infarction. Furthermore, association of the telomere-telomerase system with oxidative stress markers (as common risk factors for coronary artery disease (CAD)) was tested. Material and methods: Patients were selected from the patients admitted to the intensive care unit with acute myocardial infarction with ST-segment elevation (STEMI), with the following inclusion criteria – STEMI patients between 18 and 80 years old of both genders and candidates for primary percutaneous coronary intervention, with infarction pain present for a maximum of 12 h. In all the patients leukocyte telomere length, telomerase activity and scores related to oxidative-stress status (Protective, Damage and OXY) were evaluated. Results: Patients were divided into different groups: with stable angina pectoris (AP) (n = 22), acute myocardial infarction with: STEMI (n = 93), non-obstructive coronary arteries (MINOCA) (n = 7), blood vessel rupture (n = 6) at three time points, and compared to the group of 84 healthy subjects. Telomerase activity was significantly higher in all CAD sub-groups compared to the control group (AP = 0.373 (0.355–0.386), STEMI = 0.375 (0.349–0.395), MINOCA = 0.391 (0.366–0.401), blood vessel rupture = 0.360 (0.352–0.385) vs. CG = 0.069 (0.061–0.081), p < 0.001), while telomeres were significantly shorter in STEMI, MINOCA and blood vessel rupture groups compared to the control group (STEMI = 1.179 (0.931–1.376), MINOCA = 1.026 (0.951–1.070), blood vessel rupture = 1.089 (0.842–1.173) vs. CG = 1.329 (1.096–1.624), p = 0.030]. Values of OXY score were significantly higher in STEMI and MINOCA patients compared to the control group and AP patients (5.83 (4.55–7.54) and 10.28 (9.19–10.72) vs. 4.94 (3.29–6.18) and 4.18 (2.58–4.86), p < 0.001). Longer telomeres and higher telomerase activity were found in thromboaspirates, compared to the peripheral blood leukocytes in the same patients (1.25 (1.01–1.84) vs. 1.18 (0.909–1.516), p = 0.036; and 0.366 (0.367–0.379) vs. 0.366 (0.367–0.379), p < 0.001, respectively). In addition, telomere length and telomerase activity had good diagnostic ability to separate STEMI patients from healthy persons. Conclusions: Leukocyte telomere length and telomerase activity can differentiate CAD patients from healthy persons, and relate CAD to oxidative stress.

AB  - The clinical efficacy of chemotherapy, as a recognized therapeutic approach for malignant 
diseases,   usually   has   certain   limitations   due   to   its   cardiotoxicity   (CT)   and   consequent   
cardiomyopathy,  or  even  heart  failure.  CT
  is  defined  as  any  cardiac  injury  connected  with  
oncology treatment, whether it is chemo
-, radio
-, targeted or immunotherapy, or cancer by itself, 
and it represents a great challenge for clinicians in everyday practice. A wide spectrum of factors 
related t
o chemotherapy (type of drug, dose during each cycle, cumulative dose, schedule, method 
of  application,  combination  with  other  cardiotoxic  drugs  or  association  with  radiotherapy)  and  
patient  characteristics  (age,  presence  of  cardiovascular  risk  factors,  pr
evious  cardiovascular  
disease)  are  the  determining  factors  that  influence  the  frequency  of  CT.  Imaging  methods  for  
morphological  and  functional  monitoring  of  the  heart  muscle  are  used  for  monitoring  CT.  The  
quest  for  diagnostic  tools  for  early  CT  detection 
is  of  great  significance.  In  line  with  this,  the  
measurement  of  some  cardiac  biomarkers  has  found  its  place  in  clinical  settings  as  an  early  
determinant of myocardial injury. Therefore, in this review article, special attention will be paid 
to  certain  wel
l-established,  as  well  as  certain  novel  cardiac  biomarkers,  and  their  role  in  
recognizing asymptomatic CT, in order to gain deeper insight into their diagnostic utility.
AB  - Klinička  efikasnost  hemioterapije,  kao  priznatog  terapeutskog  pristupa  malignim 
bolestima, obično ima određena ograničenja zbog svoje kardiotoksičnosti (KT), te posledične 
kardiomiopatije, pa čak i srčane insuficijencije. KT se definiše kao svaka povreda miokarda 
povezana  s  lečenjem  raka,  uzimajući  u  obzir  hemioterapiju,  radioterapiju,  biološku  ili 
imunoterapiju,  kao  i  sam  karcinom,  te  predstavlja veliki  izazov  za  kliničare 
u  svakodnevnoj  
praksi. Širok spektar faktora povezanih s hemioterapijom (vrsta leka, doza tokom svakog ciklusa, 
kumulativna doza, raspored, način primene, kombinacija s drugim kardiotoksičnim lekovima ili 
povezanost s radioterapijom) i karakteristike bolesnika (dob, prisutnost faktora kardiovaskularnog 
rizika, prethodne kardiovaskularne bolesti), odlučujući su faktori koji utiču na učestalost KT. Za 
praćenje KT koriste se radiološke metode za morfološko i funkcionalno praćenje srčanog mišića. 
Potraga za dij
agnostičkim alatima za rano otkrivanje KT od velikog je značaja. U skladu s tim, 
merenje nekih srčanih biomarkera kao rane determinante povrede miokarda našlo je svoje mesto 
u kliničkim okruženjima. Stoga 
će u ovom preglednom članku posebna pažnja biti po
sve
ć
ena 
nekim  dobro  utvrđenim,  kao  i  nekim  novim  srčanim  biomarkerima,  i  njihovoj  ulozi  u 
prepoznavanju asimptomatske KT, kako bi se stekao dublji uvid u njihovu dijagnostičku korisnost 
i neželjene efekte lekova koji se primenjuju u terapiji karcinoma.

AB  - Purpose: The aim of this study was to analyze data on gabapentinoid-related attendances to the National Poison Control Center of Serbia (NPCC), particularly abuse cases; to estimate its changes and to compare it with trends in national consumption rates of these drugs. We also aimed to analyze the main characteristics of the study population and to investigate the major clinical effects in poisoned patients. Patients and Methods: This is a retrospective study of patients admitted to the NPCC for acute poisoning involving gabapentinoids from 1 May 2012 to 1 October 2022. Results: There were 357 (95.5%) pregabalin-related and 17 (4.5%) gabapentin-related poisoning cases in 302 patients. Abuse of pregabalin was detected in 27.8% (84/302), while gabapentin abuse occurred in 0.7% (2/302) of all patients. A steady increase in rates of pregabalin poisoning and abuse cases strongly correlated with the increase in overall consumption of this drug, while there were no significant changes in rates of gabapentin consumption, poisoning and abuse rate during the study period. Most patients who abused pregabalin pregabalin were males (84.5%) and the median age was 26 years (range: 15–45 years). Almost 60% of patients who abused pregabalin (48/84) belonged to the migrant population. Co-ingestions occurred in 89.4% of pregabalin-related cases (319/357), resulting in more severe poisoning. The most often co-ingested drugs were benzodiazepines and among them clonazepam was detected in the largest number of cases. Conclusion: The poisoning and abuse cases involving pregabalin are on the rise in Serbia, which coincided with an increase in its overall consumption during the study period. Isolated pregabalin ingestions resulted in mild poisoning, although severe symptoms such as coma and bradycardia were recorded. When prescribing pregabalin to patients at risk of abuse caution is needed. Strengthening the measures for dispensing of pregabalin may reduce the risks associated with its abuse.

AB  - Background A three-year COVID-19 pandemic revealed a spectrum of disease severity and clinical manifestations. The most intriguing part of this phenomenon lays in interindividual variability in COVID-19 course among patients, which is attributable to patient's age, comorbidities and general health status. Focus of this follow-up study was to assess leukocyte telomere length change in mild-to-moderate COVID-19 patients and concomitant influence of inflammation, oxidative stress (OS), pulmonary involvement and implemented therapy on the course of the disease.

Material and methods Routine biochemical/haematological parameters, markers of OS (prooxidants and antioxidants), vitamin D, IgM and IgG antibodies level and relative length of leukocyte telomeres (rLTL) were measured at three time-points (at diagnosis, after 14 and 21 days from the disease onset) in blood samples of 31 consecutive COVID-19 patients, with a mild (n=16) and moderate (n=15) form of the disease, treated on an outpatient basis.

Results Although the patients had reduced rLTL at baseline (median: 0.592; 25th – 75th percentiles: 0.518-0.724), it significantly increased during the follow-up (median: 0.773; 25th – 75th percentiles: 0.615-0.923; P<0.01). The rate of telomere attrition was associated with the extent of OS and pulmonary involvement. During follow-up, the burden of OS was reduced, while antioxidative defence mechanisms were recovered. The use of antibiotics and N-acetylcysteine-propolis supplementation was associated with telomere lengthening.

Conclusion Results of this study revealed significant interaction between OS, inflammation and leukocyte telomere length attrition in COVID-19. Our data suggest that rRTL can be a biomarker that enables more precise therapy decision and accurate patient status estimation.

AB  - Newer research points to alterations in the plasma redox status and the HDL subclass distributions in cancer. We aimed to assess the redox status and the HDL subclass distributions, lipids, and inflammatory markers in lymphoma patients in order to determine whether they were correlated with changes in FDG-PET/CT scans. At the beginning of this study, redox status, HDL subclasses, lipids, and inflammation biomarkers were determined in 58 patients with lymphoma (Hodgkin lymphoma, n=11 and non-Hodgkin lymphoma, n=47), and these same measurements were reassessed during their ensuing treatment (in 25 patients). Initially, the total oxidation status (TOS), the prooxidant–antioxidant balance (PAB), the OS index (OSI), the total protein sulfhydryl groups (SH-groups), and the advanced oxidation protein products (AOPP) were significantly higher in lymphoma patients as compared to healthy subjects, but the total antioxidant status (TAS) was significantly reduced. AB  - The Schlafen 11 (SLFN11) protein has recently emerged as pivotal in DNA damage conditions, with predictive potential for tumor response to cytotoxic chemotherapies. Recent discoveries also showed that the programmed death ligand 1 (PD-L1) protein can be found on malignant cells, providing an immune evasion mechanism exploited by different tumors. Additionally, excessive generation of free radicals, redox imbalance, and consequential DNA damage can affect intestinal cell homeostasis and lead to neoplastic transformation. Therefore, our study aimed to investigate the significance of SLFN11 and PD-L1 proteins and redox status parameters as prognostic biomarkers in CRC patients. This study included a total of 155 CRC patients. SLFN11 and PD-L1 serum levels were measured with ELISA and evaluated based on redox status parameters, sociodemographic and clinical characteristics, and survival. The following redox status parameters were investigated: spectrophotometrically measured superoxide dismutase (SOD), sulfhydryl (SH) groups, advanced oxidation protein products (AOPP), malondialdehyde (MDA), pro-oxidant–antioxidant balance (PAB), and superoxide anion (O2•–). The prooxidative score, antioxidative score, and OXY-SCORE were also calculated. The results showed significantly shorter survival in patients with higher OXY-SCOREs and higher levels of serum SLFN11, while only histopathology-analysis-related factors showed significant prognostic value. OXY-SCORE and SLFN11 levels may harbor prognostic potential in CRC patients.

AB  - In the present study, we examined redox status parameters in arterial and venous blood samples, its potential to predict the prognosis of acute myocardial infarction (AMI) patients assessed through its impact on the comprehensive grading SYNTAX score, and its clinical accuracy. Potential connections between common blood biomarkers, biomarkers of redox status, leukocyte telomere length, and telomerase enzyme activity in the acute myocardial infarction burden were assessed using principal component analysis (PCA). This study included 92 patients with acute myocardial infarction. Significantly higher levels of advanced oxidation protein products (AOPP), superoxide anion (O2•−), ischemia-modified albumin (IMA), and significantly lower levels of total oxidant status (TOS) and total protein sulfhydryl (SH-) groups were found in arterial blood than in the peripheral venous blood samples, while biomarkers of the telomere–telomerase system did not show statistical significance in the two compared sample types (p = 0.834 and p = 0.419). To better understand the effect of the examined biomarkers in the AMI patients on SYNTAX score, those biomarkers were grouped using PCA, which merged them into the four the most contributing factors. The “cholesterol–protein factor” and “oxidative–telomere factor” were independent predictors of higher SYNTAX score (OR = 0.338, p = 0.008 and OR = 0.427, p = 0.035, respectively), while the ability to discriminate STEMI from non-STEMI patients had only the “oxidative–telomere factor” (AUC = 0.860, p = 0.008). The results show that traditional cardiovascular risk factors, i.e., high total cholesterol together with high total serum proteins and haemoglobin, are associated with severe disease progression in much the same way as a combination of redox biomarkers (pro-oxidant-antioxidant balance, total antioxidant status, IMA) and telomere length.

AB  - The aim of this multicentric study was to assess the impacts of oxidative stress, inflam-
mation, and the presence of small, dense, low-density lipoproteins (sdLDL) on the antioxidative
function of high-density lipoprotein (HDL) subclasses and the distribution of paraoxonase-1 (PON1)
activity within HDL in patients with ST-segment elevation acute myocardial infarction (STEMI). In
69 STEMI patients and 67 healthy control subjects, the lipoproteins’ subclasses were separated using
polyacrylamide gradient (3–31%) gel electrophoresis. The relative proportion of sdLDL and each
HDL subclass was evaluated by measuring the areas under the peaks of densitometric scans. The
distribution of the relative proportion of PON1 activity within the HDL subclasses (pPON1 within
HDL) was estimated using the zymogram method. The STEMI patients had significantly lower
proportions of HDL2a and HDL3a subclasses (p = 0.001 and p < 0.001, respectively) and lower pPON1
within HDL3b (p = 0.006), as well as higher proportions of HDL3b and HDL3c subclasses (p = 0.013
and p < 0.001, respectively) and higher pPON1 within HDL2 than the controls. Independent positive
associations between sdLDL and pPON1 within HDL3a and between malondialdehyde (MDA) and
pPON1 within HDL2b were shown in the STEMI group. The increased oxidative stress and increased
proportion of sdLDL in STEMI are closely related to the compromised antioxidative function of small
HDL3 particles and the altered pPON1 within HDL.

Redox homeostasis, oxidative stress and antioxidant system in health and disease: the possibility of modulation by antioxidants

AB  - Redox imbalance occurs when the factors of oxidative stress, known as prooxidants, outweigh
the mechanisms of antioxidant protection. In a healthy state, homeostatic mechanisms ensure the
balanced production of free radicals and a complete series of antioxidants responsible for their safe
removal. The generation of free radicals is a part of physiological processes in a healthy organism,
some of which act as specific signaling molecules, and their presence and activity are necessary in
these processes. In various diseases such as cancer, cardiovascular disease, diabetes, autoimmune
diseases, rheumatic diseases, systemic lupus, and skin diseases, the generation of free radicals
overwhelms the protective mechanisms, leading to the development of "oxidative stress" that damages
cells and tissues. To prevent the harmful effects of free radicals within cells, there exists a system of
enzymatic antioxidant protection composed of superoxide dismutase (SOD), glutathione peroxidase
(GSHPx), glutathione reductase (GR), glutaredoxin, reduced/oxidized glutathione (GSH/GSSG), and
thioredoxin (TRX). The examples of non-enzymatic antioxidants are: antioxidant vitamins such as A,
C and E, dihydrolypoic acid, metallothioneins, ceruloplasmin, coenzyme Q 10, urea, creatinine, etc.
Redox balance is influenced by the circadian rhythm and external factors that constitute the
"exposome", including dietary habits and lifestyle. Antioxidant supplementation has become
increasingly popular for maintaining optimal body function. However, it is important to note that some
antioxidants can exhibit prooxidant activity, emphasizing the need for controlled use. The relationship
between the redox status of the body and the action of antioxidants enables the development of
multidisciplinary research that connects biochemistry, molecular biology, nutritional science, natural
product chemistry, and clinical practice.
AB  - Redoks disbalans se javlja kada činioci oksidativnog stresa – prooksidansi – nadvladaju
mehanizme antioksidativne zaštite. U stanju zdravlja, homeostatski mehanizmi obezbeđuju
uravnoteženo stvaranje slobodnih radikala i čitave serije antioksidanasa koji su zaduženi za njihovo
bezbedno uklanjanje. Stvaranje slobodnih radikala je deo fizioloških procesa u zdravom organizmu;
neki od njih su specifični signalni molekuli i u tim procesima su njihovo prisustvo i aktivnost
neophodni. U različitim bolestima kao što su kancer, kardiovaskularne bolesti, dijabetes,
autoimunske bolesti, reumatske bolesti, sistemski lupus, kožne bolesti, stvaranje slobodnih radikala
nadvladava mehanizme zaštite, pa se razvija „oksidativni stres“ koji oštećuje ćelije i tkiva. Da bi se
sprečilo štetno delovanje slobodnih radikala, u ćeliji postoji sistem enzimske antioksidativne zaštite,
koga čine: superoksid-dismutaza (SOD), glutation-peroksidaza (GSHPx), glutation-reduktaza
(GR), glutaredoksin, redukovani/oksidovani glutation (GSH/GSSG) i tioredoksin (TRX). Primeri
neenzimskih antioksidanasa su: antioksidativni vitamini kao što su A, C i E, dihidrolipoinska
kiselina, metalotioneini, ceruloplazmin, koenzim Q10, urea, kreatinin, itd. Redoks ravnoteža je pod
uticajem cirkadijalnog ritma i spoljašnjih faktora koji čine „ekspozom“ i uključuju način ishrane i
životne navike. Suplementacija antioksidansima je postala sve popularnija praksa za održavanje
optimalne funkcije organizma. Neki od antioksidanasa ispoljavaju prooksidantnu aktivnost i zato je
važno da njihova primena bude kontrolisana. Veza između redoks statusa organizma i delovanja
antioksidanasa omogućava razvoj multidisciplinarnih istraživanja u kojima se povezuju biohemija,
molekularna biologija, nauka o ishrani, hemija prirodnih proizvoda i sama klinička praksa.

thioredoxin (TRX). The examples of non-enzymatic antioxidants are: antioxidant vitamins such as A,
C and E, dihydrolypoic acid, metallothioneins, ceruloplasmin, coenzyme Q 10, urea, creatinine, etc.
Redox balance is influenced by the circadian rhythm and external factors that constitute the
"exposome", including dietary habits and lifestyle. Antioxidant supplementation has become
increasingly popular for maintaining optimal body function. However, it is important to note that some
antioxidants can exhibit prooxidant activity, emphasizing the need for controlled use. The relationship
between the redox status of the body and the action of antioxidants enables the development of
multidisciplinary research that connects biochemistry, molecular biology, nutritional science, natural
product chemistry, and clinical practice., Redoks disbalans se javlja kada činioci oksidativnog stresa – prooksidansi – nadvladaju
mehanizme antioksidativne zaštite. U stanju zdravlja, homeostatski mehanizmi obezbeđuju
uravnoteženo stvaranje slobodnih radikala i čitave serije antioksidanasa koji su zaduženi za njihovo
bezbedno uklanjanje. Stvaranje slobodnih radikala je deo fizioloških procesa u zdravom organizmu;
neki od njih su specifični signalni molekuli i u tim procesima su njihovo prisustvo i aktivnost
neophodni. U različitim bolestima kao što su kancer, kardiovaskularne bolesti, dijabetes,
autoimunske bolesti, reumatske bolesti, sistemski lupus, kožne bolesti, stvaranje slobodnih radikala
nadvladava mehanizme zaštite, pa se razvija „oksidativni stres“ koji oštećuje ćelije i tkiva. Da bi se
sprečilo štetno delovanje slobodnih radikala, u ćeliji postoji sistem enzimske antioksidativne zaštite,
koga čine: superoksid-dismutaza (SOD), glutation-peroksidaza (GSHPx), glutation-reduktaza
(GR), glutaredoksin, redukovani/oksidovani glutation (GSH/GSSG) i tioredoksin (TRX). Primeri
neenzimskih antioksidanasa su: antioksidativni vitamini kao što su A, C i E, dihidrolipoinska
kiselina, metalotioneini, ceruloplazmin, koenzim Q10, urea, kreatinin, itd. Redoks ravnoteža je pod
uticajem cirkadijalnog ritma i spoljašnjih faktora koji čine „ekspozom“ i uključuju način ishrane i
životne navike. Suplementacija antioksidansima je postala sve popularnija praksa za održavanje
optimalne funkcije organizma. Neki od antioksidanasa ispoljavaju prooksidantnu aktivnost i zato je
važno da njihova primena bude kontrolisana. Veza između redoks statusa organizma i delovanja
antioksidanasa omogućava razvoj multidisciplinarnih istraživanja u kojima se povezuju biohemija,
molekularna biologija, nauka o ishrani, hemija prirodnih proizvoda i sama klinička praksa.",

AB  - Purpose: End-stage renal disease patients on chronic hemodialysis (HD) have a shortened life expectancy compared to the general population. The aim of this study was to evaluate a possible link between three new and emerging factors in renal pathophysiology: Klotho protein, telomere length in peripheral blood mononuclear cells (TL) and redox status parameters before HD (bHD) and after HD (aHD), and to test mortality prediction capability of these emerging parameters in a population of HD patients. Methods: The study included 130 adult patients with average age 66 (54–72), on HD (3 times per week; 4–5 h per session). Klotho level, TL, routine laboratory parameters, dialysis adequacy and redox status parameters: advanced oxidation protein products (AOPP), prooxidant–antioxidant balance (PAB), superoxide anion (O2.−), malondialdehyde (MDA), ischemia-modified albumin (IMA), total sulfhydryl group content (SHG), and superoxide dismutase (SOD) were determined. Results: Klotho concentration was significantly higher aHD; 68.2 (22.6–152.9) vs. bHD 64.2 (25.5–119.8) (p = 0.027). The observed increase in TL was not statistically significant. AOPP, PAB, SHG, and SOD activity were significantly increased aHD (p > 0.001). The patients with the highest mortality risk score (MRS) had significantly higher PAB bHD (p = 0.002). Significantly lower O2.− (p < 0.001), SHG content (p = 0.072), and IMA (p = 0.002) aHD were found in patients with the lowest MRS values. Principal component analysis revealed redox balance-Klotho factor as a significant predictor of high mortality risk (p = 0.014). Conclusion: Decreased Klotho and TL attrition as well as redox status disturbance could be connected with higher mortality rate in HD patients.

AB  - Purpose: The aim of this study was to analyze data on gabapentinoid-related attendances to the National Poison Control Center of Serbia (NPCC), particularly abuse cases; to estimate its changes and to compare it with trends in national consumption rates of these drugs. We also aimed to analyze the main characteristics of the study population and to investigate the major clinical effects in poisoned patients. Patients and Methods: This is a retrospective study of patients admitted to the NPCC for acute poisoning involving gabapentinoids from 1 May 2012 to 1 October 2022. Results: There were 357 (95.5%) pregabalin-related and 17 (4.5%) gabapentin-related poisoning cases in 302 patients. Abuse of pregabalin was detected in 27.8% (84/302), while gabapentin abuse occurred in 0.7% (2/302) of all patients. A steady increase in rates of pregabalin poisoning and abuse cases strongly correlated with the increase in overall consumption of this drug, while there were no significant changes in rates of gabapentin consumption, poisoning and abuse rate during the study period. Most patients who abused pregabalin pregabalin were males (84.5%) and the median age was 26 years (range: 15–45 years). Almost 60% of patients who abused pregabalin (48/84) belonged to the migrant population. Co-ingestions occurred in 89.4% of pregabalin-related cases (319/357), resulting in more severe poisoning. The most often co-ingested drugs were benzodiazepines and among them clonazepam was detected in the largest number of cases. Conclusion: The poisoning and abuse cases involving pregabalin are on the rise in Serbia, which coincided with an increase in its overall consumption during the study period. Isolated pregabalin ingestions resulted in mild poisoning, although severe symptoms such as coma and bradycardia were recorded. When prescribing pregabalin to patients at risk of abuse caution is needed. Strengthening the measures for dispensing of pregabalin may reduce the risks associated with its abuse.

Electrochemical Characterisation and Confirmation of Antioxidative Properties of Ivermectin in Biological Medium

AB  - Ivermectin (IVM) is a drug from the group of anthelmintics used in veterinary and human medicine. Recently, interest in IVM has increased as it has been used for the treatment of some malignant diseases, as well as viral infections caused by the Zika virus, HIV-1 and SARS-CoV-2. The electrochemical behaviour of IVM was investigated using cyclic (CV), differential pulse (DPV) and square wave voltammetry (SWV) at glassy carbon electrode (GCE). IVM showed independent oxidation and reduction processes. The effect of pH and scan rate indicated the irreversibility of all processes and confirmed the diffusion character of oxidation and reduction as an adsorption-controlled process. Mechanisms for IVM oxidation at the tetrahydrofuran ring and reduction of the 1,4-diene structure in the IVM molecule are proposed. The redox behaviour of IVM in a biological matrix (human serum pool) showed a pronounced antioxidant potential similar to that of Trolox during short incubation, whereas a prolonged stay among biomolecules and in the presence of an exogenous pro-oxidant (tert-butyl hydroperoxide, TBH) resulted in a loss of its antioxidant effect. The antioxidant potential of IVM was confirmed by voltametric methodology which is proposed for the first time.

The antidepressant drugs vortioxetine and duloxetine differentially and sex-dependently affect animal well-being, cognitive performance, cardiac redox status and histology in a model of osteoarthritis

AB  - Osteoarthritis represents a leading cause of disability with limited treatment options. Furthermore, it is frequently accompanied by cardiovascular and cognitive disorders, which can be exacerbated by osteoarthritis or drugs used for its treatment. Here, we examined the behavioral and cardiac effects of the novel antidepressant vortioxetine in an osteoarthritis model, and compared them to duloxetine (an established osteoarthritis treatment). Osteoarthritis was induced in male and female rats with an intraarticular sodium-monoiodoacetate injection. Antidepressants were orally administered for 28 days following induction. During this period the acetone, burrowing and novel-object-recognition tests (NORT) were used to assess the effects of antidepressants on pain hypersensitivity (cold allodynia), animal well-being and cognitive performance, respectively. Following behavioral experiments, heart muscles were collected for assessment of redox status/histology. Antidepressant treatment dose-dependently reduced cold allodynia in rats with osteoarthritis. Duloxetine (but not vortioxetine) depressed burrowing behavior in osteoarthritic rats in a dose-related manner. Osteoarthritis induction reduced cognitive performance in NORT, which was dose-dependently alleviated by vortioxetine (duloxetine improved performance only in female rats). Furthermore, duloxetine (but not vortioxetine) increased oxidative stress parameters in the heart muscles of female (but not male) rats and induced histological changes in cardiomyocytes indicative of oxidative damage. Vortioxetine displayed comparable efficacy to duloxetine in reducing pain hypersensitivity. Furthermore, vortioxetine (unlike duloxetine) dose-dependently improved cognitive performance and had no adverse effect on burrowing behavior (animal surrogate of well-being) and cardiac redox status/histology. Our results indicate that vortioxetine could be a potential osteoarthritis treatment (with better characteristics compared to duloxetine).

During this period the acetone, burrowing and novel-object-recognition tests (NORT) were used to assess the effects of antidepressants on pain hypersensitivity (cold allodynia), animal well-being and cognitive performance, respectively. Following behavioral experiments, heart muscles were collected for assessment of redox status/histology. Antidepressant treatment dose-dependently reduced cold allodynia in rats with osteoarthritis. Duloxetine (but not vortioxetine) depressed burrowing behavior in osteoarthritic rats in a dose-related manner. Osteoarthritis induction reduced cognitive performance in NORT, which was dose-dependently alleviated by vortioxetine (duloxetine improved performance only in female rats). Furthermore, duloxetine (but not vortioxetine) increased oxidative stress parameters in the heart muscles of female (but not male) rats and induced histological changes in cardiomyocytes indicative of oxidative damage. Vortioxetine displayed comparable efficacy to duloxetine in reducing pain hypersensitivity. Furthermore, vortioxetine (unlike duloxetine) dose-dependently improved cognitive performance and had no adverse effect on burrowing behavior (animal surrogate of well-being) and cardiac redox status/histology. Our results indicate that vortioxetine could be a potential osteoarthritis treatment (with better characteristics compared to duloxetine).",

AB  - A comparative in vitro study of the antioxidant potential of natural phenols (zingerone, curcumin, raspberry ketone, magnolol) and their synthesized derivatives was performed. The antioxidant efficiency was evaluated in blood serum obtained from healthy individuals, by means of spectrophotometry, before and after the addition of pro-oxidant tert-butyl hydroperoxide (TBH). Moreover, the antioxidant effect of an equimolar mixture of curcumin and zingerone was investigated. Interpretation of our results reveals that in the blood serum of healthy individuals curcumin (C1), raspberry ketone (RK1), magnolol (M1) and synthesized derivative of zingerone (Z2) demonstrate remarkable antioxidant effects (p < 0.05). However, in the state of TBH-induced excessive oxidative stress natural magnolol and synthesized derivatives C1, Z1 and RK1 show powerful antioxidant activity and thus can be further investigated to obtain information about their metabolic transformations and their potential influence at the cellular level. Results obtained from measurements in an equimolar mixture of zingerone and curcumin indicate synergism (p < 0.05) between the two compounds. This combination is especially successful due to the fast and efficient neutralization of added pro-oxidant TBH. The commercial availability of turmeric and ginger and their frequent combined use in diet suggest ideas for further broader utilization of the beneficial synergistic effect of their phenolic components.

AB  - Lung cancer (LC) is the second most common malignancy and leading cause of cancer death. The potential "culprit" for local and systemic telomere shortening in LC patients is oxidative stress. We investigated the correlation between the peripheral blood leukocyte (PBL) telomere length (TL) and the presence/severity of LC and oxidative stress, and its usefulness as LC diagnostic marker. PBL TL was measured in 89 LC patients and 83 healthy subjects using the modified Cawthon RTq-PCR method. The relative PBL TL, found to be a potential diagnostic marker for LC with very good accuracy (P < 0.001), was significantly shorter in patients compared to the control group (CG) (P < 0.001). Significantly shorter telomeres were found in patients with LC TNM stage IV than in patients with stages I-III (P = 0.014), in patients without therapy compared to those on therapy (P = 0.008), and in patients with partial response and stable/progressive disease compared to those with complete response (P = 0.039). The total oxidant status (TOS), advanced oxidation protein products (AOPP), prooxidant-antioxidant balance (PAB) and C-reactive protein (CRP) were significantly higher in patients compared to CG (P < 0.001) and correlated negatively with TL in both patients and CG (P < 0.001). PCA showed a relation between PAB and TL, and between the EGFR status and TL. Oxidative stress and PBL telomere shortening are probably associated with LC development and progression.

AB  - The interplay between oxidative stress and inflammation is implicated in many chronic diseases including Alzheimer`s disease, cardiovascular diseases, diabetes and cancer. Thirteen β-hydroxy-β-arylalkanoic acids were previously synthesized and evaluated for their anti-inflammatory activity. The aim of this study was to asses ex vivo antioxidant activity of synthesized acids, as well as ibuprofen and to identify the compounds with the most promising results for further investigation on their capacity to counteract in vivo oxidative stress triggered by inflammation. The antioxidant potential of tested compounds was evaluated by determining the concentrations of total antioxidative status, total oxidative status, prooxidant antioxidant balance and the total sulfhydryl groups. Z score statistics were used to calculate the summary scores for antioxidative activity, prooxidative activity and oxy score. The tested compounds and ibuprofen demonstrated mild prooxidative activity ex vivo. Seven acids with substituents on one benzene ring exhibited better results than ibuprofen and were selected for in vivo testing. In vivo results demonstrated better antioxidant protection compared to ex vivo results. Compound g which contains nitro group on the benzene ring demonstrated the lowest oxy score, and four compounds exhibited better results than ibuprofen.
