AB  - Soft glucocorticoids are compounds that are biotransformed to inactive and non-toxic metabolites and have fewer side effects than traditional glucocorticoids. A new class of 17-carboxamide steroids has been recently introduced by our group. In this study, local anti-inflammatory activity of these derivatives was evaluated by use of the croton oil-induced ear edema test. Glucocorticoids with the highest maximal edema inhibition (MEI) were pointed out, and the systemic side effects of those with the lowest EC50 values were significantly lower in comparison to dexamethasone. A 3D-QSAR model was created and employed for the design of 27 compounds. By use of the sequential combination of ligand-based and structure-based virtual screening, three compounds were selected from the ChEMBL library and used as a starting point for the design of 15 derivatives. Molecular docking analysis of the designed derivatives with the highest predicted MEI and relative glucocorticoid receptor binding affinity (20, 22, 24-1, 25-1, 27, VS7, VS13, and VS14) confirmed the presence of interactions with the glucocorticoid receptor that are important for the activity.

Synthesis, crystal structure and local anti-inflammatory activity of the L-phenylalanine methyl ester derivative of dexamethasone-derived cortienic acid

AB  - The L-phenylalanine methyl ester derivative of dexamethasone-derived cortienic acid (DF) was synthesized and its crystal structure characterized by the X-ray diffraction method. The crystal system is orthorhombic with space group P2(1)2(1)2(1) and cell constants a = 8.2969(3) angstrom, b = 18.9358(8) angstrom, c = 20.0904(6) angstrom, V = 3156.4(2) angstrom(3) and Z = 4. Ring A of the steroid nucleus and phenyl ring in the 17 beta-side chain are almost planar. Rings B and C have a slightly distorted chair conformation, whereas ring D has an envelope conformation. The packing of DF is characterized by a network of intermolecular hydrogen bonds involving the O4 atom from one side of the steroid nucleus and O1 and F1 atoms from the other side as hydrogen bond acceptors. Apart from the intermolecular hydrogen bonds in the crystal packing, there are also numerous intramolecular hydrogen bonds of the N-H center dot center dot center dot O, C-H center dot center dot center dot O and C-H center dot center dot center dot F type. The local anti-inflammatory activity of DF was evaluated using the croton oil-induced ear oedema test. This derivative achieved maximal inhibition of ear oedema at significantly lower concentration in comparison with dexamethasone.

AB  - Molecular docking studies were performed on 18 17-carboxamide steroids in order to select compounds with potential local anti-inflammatory activity. These derivatives are amides of cortienic acids (obtained from hydrocortisone, prednisolone, and methylprednisolone) with methyl or ethyl esters of six amino acids. Interactions with the glucocorticoid receptor (GR), binding energies and ligand efficiency values of these compounds were compared with dexamethasone and cortienic acid obtained from prednisolone (inactive metabolite). On the basis of molecular docking studies, seven compounds were selected and their binding affinities for the GR were predicted by use of the exponential model created in this study. Subsequently, selected compounds were synthesized in good yields by use of modified N,N-dicyclohexylcarbodiimide (DCC)/1-hydroxybenzotriazole (HOBt) coupling procedure. Finally, the local anti-inflammatory activity of the synthesized compounds was examined by use of the croton oil-induced ear edema test. In vivo evaluation of systemic side effects as well as in silico prediction of metabolism were performed on the derivative with the best local anti-inflammatory activity. The combination of molecular docking studies and the exponential model for the GR binding affinity prediction could be used as an in silico tool for the rational design of novel 17-carboxamide steroids with potentially better biological profile than dexamethasone.

The impact of prolonged cadmium exposure and co-exposure with polychlorinated biphenyls on thyroid function in rats

AB  - Endocrine-disrupting chemicals currently represent one of the major concerns and this study was aimed to investigate the effects of different doses of cadmium, widespread toxic metal, on the levels of thyroid hormones and to calculate Benchmark doses for these effects. Furthermore, the effects of co-exposure to cadmium and polychlorinated biphenyls on thyroid function were investigated. Six orally-treated groups of rats were receiving 0.3, 0.6, 1.25, 2.5, 5 and 10 mg Cd/kg b.w./day, five groups were orally treated with 0.5, 1, 2, 4 and 8 mg PCBs/kgb.w./day, while nine groups of rats were orally-treated with different dose combinations of Cd and PCBs (0.6, 1.25 and 2.5 mg Cd/kgb.w. and 2,4 and 8 mg PCBs/kg b.w./day), during 28 days. Thyroid hormones were adversely affected by cadmium, with most prominent effect observed on triiodothyroxine levels indicating Cd interference with thyroid function at extrathyroidal level. Calculated Benchmark doses for Cd effects on thyroid hormones indicate triiodothyroxine as the most sensitive one that can be used as a basis for risk assessment. This study also implicates possible synergistic effects of Cd and PCBs on thyroid function as a consequence of their interference at different levels of thyroid homeostasis.

abstract = "Endocrine-disrupting chemicals currently represent one of the major concerns and this study was aimed to investigate the effects of different doses of cadmium, widespread toxic metal, on the levels of thyroid hormones and to calculate Benchmark doses for these effects. Furthermore, the effects of co-exposure to cadmium and polychlorinated biphenyls on thyroid function were investigated. Six orally-treated groups of rats were receiving 0.3, 0.6, 1.25, 2.5, 5 and 10 mg Cd/kg b.w./day, five groups were orally treated with 0.5, 1, 2, 4 and 8 mg PCBs/kgb.w./day, while nine groups of rats were orally-treated with different dose combinations of Cd and PCBs (0.6, 1.25 and 2.5 mg Cd/kgb.w. and 2,4 and 8 mg PCBs/kg b.w./day), during 28 days. Thyroid hormones were adversely affected by cadmium, with most prominent effect observed on triiodothyroxine levels indicating Cd interference with thyroid function at extrathyroidal level. Calculated Benchmark doses for Cd effects on thyroid hormones indicate triiodothyroxine as the most sensitive one that can be used as a basis for risk assessment. This study also implicates possible synergistic effects of Cd and PCBs on thyroid function as a consequence of their interference at different levels of thyroid homeostasis.",

AB  - In relation to commonly used no-observed-adverse-effect-level (NOAEL) point of departure, because of its statistical power and reliability, Benchmark dose (BMD) approach has been proposed as an alternative in dose-response assessment. The aim of this study was to derive BMD (10%) by means of PROAST software to quantify influence of cadmium (Cd) and/or decabrominated diphenyl ether (BDE209) on thyroid hormones homestasis. Study was conducted on male Wistar rats treated orally by gavage for 28 days by either single substances or their combination. Three groups of animals were dosed with Cd at levels of 2.5, 7.5 and 15 mg/kg b.w./day, three groups of animals were dosed with BDE209 at levels of 1000, 2000 or 4000 mg/kg b.w./day, while 9 groups received different dose mixtures of previously given dose levels of Cd and BDE209 (design 3 x 3). Results of the study have indicated that Cd+BDE209 mixtures are likely more potent to disrupt thyroid function than would be expected from the chemicals individually. Derived BMD - lower confidence limits (BMDL), if ratio BMD/BMDL is  lt  10, were 9.4 mg Cd/kg b.w./day and 2155 mg BDE209/kg b.w./day for the effect on T3; and 6.22 mg Cd/kg b.w./day in the mixture with BDE209 2000 mg/kg b.w./day for the effect on FT3.
AB  - Primena statistički dobijene Benchmark doze (BMD) u proceni rizika predstavlja alternativu najčešće korišćenoj 'dozi bez štetnog efekta' (NOAEL) zbog veće pouzdanosti u analizi odnosa doze i toksičnog efekta. Cilj ovog rada bio je izračunavanje BMD (10%) primenom PROAST softvera radi kvantitativne procene uticaja Cd i/ili BDE209 na homeostazu hormona štitaste žlezde. Ispitivanje je sprovedeno na mužjacima Wistar pacova koji su putem oralne sonde, tokom 28 dana, primali pojedinačne supstance ili njihove kombinacije. Kadmijum je primenjivan u dozama od 2,5, 7,5 i 15 mg Cd/kg t.m./dan, a BDE209 u dozama od 1000, 2000 i 4000 mg BDE209/kg t.m./dan, dok je ostalih devet grupa životinja primalo kombinacije hemikalija (dizajn 3 x 3). AB  - Polychlorinated biphenyls (PCBs) and brominated flame retardants, polybrominated diphenylethers (PBDEs), are widespread environmental contaminants as a result of anthropogenic activities and due to their persistency and resistance to degradation. Both groups of chemicals are placed on the list of persistent organic pollutants (POPs), covered by the Stockholm convention which is aimed to limit or ban the production, use, emission, import and export of POPs in order to protect human health and the environment. Taking into account the structural similarity between PCBs and PBDEs, and the known effects of PCBs, these two groups of chemicals could have similar mechanisms of action. Also, because of real simultaneous exposure to these compounds, an examination of possible interactions is of great importance. Interactions of the compounds from these groups are likely at the system level of hormones, particularly thyroid and reproductive ones. As there are mechanisms of adverse effects of both groups of chemicals on the nervous system, including functional, neurological and behavioral changes, there are influences on neurotransmiters, changes in signal transduction and apoptosis. There are interactions affecting the occurrence of metabolic disorders as well. Data on the toxicity of mixtures of PCBs and PBDEs, as well as interactions of these chemicals would contribute to the processes of risk evaluation and risk characterisation.
AB  - Polihlorovani bifenili (PCBs) i bromovani usporivači gorenja, polibromovani difenil etri (PBDEs) su široko rasprostranjeni u životnoj sredini kao posledica antropogenih aktivnosti i zahvaljujući njihovoj postojanosti i otpornosti na degradaciju. Obe grupe jedinjenja se nalaze na listi perzistentnih organskih zagađivača (POPs) usvojenoj Stokholmskom konvencijom sa ciljem da zabrani ili ograniči proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs u svrhu zaštite zdravlja ljudi i životne sredine. Uzimajući u obzir strukturnu sličnost između PBDEs i PCBs, kao i činjenicu da je istovremena ekspozicija ovim jedinjenima realna, ispitivanja njihovih interakcija su od značaja za predviđanje rezultujućeg efekta. Saznanja u oblasti toksikologije smeša, takođe doprinose procesu evaluacije i karakterizacije rizika. Interakcije jedinjenja iz ovih grupa su verovatne na nivou endokrinog sistema, naročito tiroidnog i reproduktivnog. AB  - The influence of methanol extracts of four Stachys species from Balkan on carbon tetrachloride induced hepatotoxicity in rats was investigated. The extracts were obtained from aerial parts of S. beckeana Dorfler & Hayek, S. anisochila Vis. et Pancic, S. plumosa Griseb. and S. alpina L. subsp. dinarica Murb. The liver damage was induced by s.c. injections of carbon tetrachloride (2.5 ml/kg b.w.) and the extracts were then consecutively applied for five days. Their effects were evaluated through alteration of biochemical parameters (liver enzymes in the serum), as well as through histopathological changes in the liver (liver damage score, LDS). Treatment of CCl(4)-intoxicated rats with methanol extracts of investigated Stachys taxa (200 and 100 mg/kg b.w. p.o.) significantly reduced increased level of marker enzymes aspartate transaminase (AST), alanine transaminase (ALT) and alkaline phosphatase (ALP) in the serum (with the exception of AST and ALT after 100 mg/kg of S. anisochila extract was administered). Vast degenerative and vascular changes in CCl(4)-treated rats were also notably reduced after the treatment with investigated extracts, corroborating the biochemical observations and confirming their hepatoprotective effects. The best dose-dependant activity was achieved by the methanol extract of S. alpina subsp. dinarica.

AB  - Polybrominated diphenyl ethers (PBDEs) represent group of persistant organic chemicals (POPs), usually used as flame retardant additives in a variety of matherials and commercial products. Since 2009, PBDEs have been placed on the list of POPs chemicals, covered by Stockholm convention which is aimed to limit or ban production, use, emission, import and export of persistent organic pollutants, due to human health and environmental protection. Predominant routes of human exposure by PBDEs are ingestion by food, inhalation or dermal. Based on experimental studies, targets for PBDEs toxicity are nervous system, liver, thyroid gland and development. According to International Agency for Research on Cancer (IARC) PBDEs are not classified as a carcinogens, whereas according to Environmental Protection Agency (EPA) only deca BDE is classified as possible human carcinogen. So far, PBDEs have not been incoporated in national legislative framework. There are no data on environmental contamination, occupational exposure and exposure of general population. Therefore, placing the data on PBDEs in the focus of publics could contribute to the process of risk characterisation, and give directions for risk management in the Republic of Serbia.
AB  - Polibromovani difeniletri (PBDEs) su hemikalije koje se dodaju različitim proizvodima kojima smo okruženi u svakodnevnom životu da bi usporili i/ili sprečili izbijanja požara. Polibromovani difeniletri su 2009. godine pridruženi listi perzistentnih organskih polutanata (POPs) usvojenoj Stokholmskom konvencijom čiji je cilj da ograniči ili zabrani proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs radi zaštite zdravlja ljudi i životne sredine. Do ekspozicije ljudi PBDEs može doći ingestijom, inhalacijom, dermalno. Eksperimentalne studije ukazuju da PBDEs najznačajnije toksične efekte ispoljavaju na jetru, štitnu žlezdu, nervni sistem, rast i razvoj. Međunarodna agencija za istraživanje karcinoma (IARC) ne klasifikuje PBDEs kao karcinogene za ljude, a Američka agencija za zaštitu životne sredine (EPA) klasifikuje samo dekaBDE kao moguć karcinogen za ljude. U našoj zemlji PBDEs nisu inkorporirani u nacionalnu regulativu, nema podataka o nivou zagađenja životne sredine, o sadržaju ovih jedinjenja u radnom okruženju i ekpoziciji opšte populacije, stoga bi saznanja u ovoj oblasti doprinela procesu evaluacije i karakterizacije rizika, ali i dala smernice za upravljanje rizikom PBDEs hemikalijama u Republici Srbiji.

Međunarodna agencija za istraživanje karcinoma (IARC) ne klasifikuje PBDEs kao karcinogene za ljude, a Američka agencija za zaštitu životne sredine (EPA) klasifikuje samo dekaBDE kao moguć karcinogen za ljude. U našoj zemlji PBDEs nisu inkorporirani u nacionalnu regulativu, nema podataka o nivou zagađenja životne sredine, o sadržaju ovih jedinjenja u radnom okruženju i ekpoziciji opšte populacije, stoga bi saznanja u ovoj oblasti doprinela procesu evaluacije i karakterizacije rizika, ali i dala smernice za upravljanje rizikom PBDEs hemikalijama u Republici Srbiji.",

AB  - In this study it was described synthesis of new C-21 esters of fluocinolone acetonide with alpha-alkoxyalcanoic and alpha-aryloxyalkanoic acids. Esterification with these branched oxyacids has to improve benefit-risk ratio. All synthesized compound were shown some topical anti-inflammatory activity.
