Arbutin content and antioxidant activity of some Ericaceae species

AB  - Quantitative analyses and investigation of antioxidant activity of herb and dry ethanolic extracts of five species from Ericaceae family (Arbutus unedo L., Bruckentalia spiculifolia Rchb., Calluna vulgaris Salisb., Erica arborea L. and Erica carnea L.) were performed. Total polyphenols, tannins and flavonoids were determined spectrophotometrically and arbutin content was measured both spectrophotometrically and by HPLC coupled with DAD detection Antioxidative properites of the ethanolic extracts were tested by means of FRAP (total antioxidant capacity), lipid peroxidation and DPPH free radical scavenging activity. A significant amount of arbutin was detected only in Arbutus unedo. All samples investigated showed excellent antioxidant activity. The best inhibition of lipid peroxidation has been shown by Bruckentalia spiculifolia herb extract (62.5 mu g/ml; more than 95%), which contained the highest amount of flavonoids (11.79%). The highest scavenging activity was obtained with leave extract of Arbutus unedd (IC(50) = 7.14 mu g/ml). The leaves of A. unedd contained a small amount of flavonoids but high content of non-tannins polyphenols.

Evaluation of the stimulatory effect of Epimedium alpinum L. methanolic extract on the immune response in vivo

AB  - The effect of the methanolic extract of the underground parts of Epimedium alpinum L. (MEEA) on the immune response to Keyhole Limpet Hemocyanine (KLH) or alloantigens in vivo was studied in AO rats. Immunization of experimental animals with KLH or allogeneic lymphocytes together with MEEA was followed by an increase in cellularity of draining lymph nodes (LN) and enhanced proliferation of LN lymphocytes after their restimulation with specific antigens in vitro, compared to control rats immunized without MEEA. These effects correlated with an increase in relative values of B, MHC class II+, CD25(+) and CD71(+) cells, whereas percentages of T cells and both subsets of T cells (CD4(+) and CD8(+)) were not significantly altered. As a consequence of higher LN cellularity, total numbers of all cell subsets in the MEEA-treated group of rats were significantly increased, compared to the corresponding control. The addition of MEEA together with KLH in vitro to LN lymphocytes of rats immunized with KLH or KLH and MEEA in vivo was manifested by significant increase (0.1 mu g/ml of MEEA) and decrease (50 mu g/ml and 100 mu g/ml of MEEA) of cell proliferation, respectively. However, when LN lymphocytes from rats, immunized in vivo with KLH and MEEA, were stimulated in vitro with MEEA together with an anti-alpha beta T cell receptor monoclonal antibody (R73), their proliferation was siginificantly inhibited. Taken together, obtained results suggest that MEEA possesses immunostimulatory activity in vivo, but some components from the extract exert immunosuppressive effects in vitro on previously in vivo activated T cells.

The HPLC determination of the content of magnoflorine in epimedium alpinum L

AB  - The aporphine alkaloid magnoflorine, known as a constituent of Asian Epimedium species, was isolated for the first time from the wild growing European species, Epimedium alpinum L. Identification was done by comparison with literature data. The HPLC method was applied for the determination of the content of the alkaloid in methanolic extracts of plant material. The underground part of E. alpinum contains 1-2 % of magnoflorine, while its content in methanolic extracts was 9.2-11.8 %. The aerial part of the plant contains less alkaloid (0.06-0.12 %), while the magnoflorine content in methanolic extracts of the aerial part was 0.4-0.8 %. Probably, the high content of magnoflorine in the methanolic extracts of underground parts of E. alpinum influences its dose-dependent cytotoxic activity (300 μg/ml; 39 % survival of cells in an experimental K562 cell culture).
AB  - Aporfinski alkaloid magnoflorin, poznat kao sastojak drugih vrsta roda Epimedium, po prvi put je izolovan iz evropske vrste Epimedium alpinum L. i identifikovan poređenjem sa literaturnim podacima. Ovo jedinjenje je korišćeno kao standard u HPLC analizi metanolnih ekstrakata E. alpinum. Sadržaj magnoflorina je znatno viši u ekstraktima podzemnih organa (9,2-11,8%, što odgovara sadržaju od 1-2 % magnoflorina u biljnom materijalu), nego u ekstraktima nadzemnih organa prevolca (0,4-0,8%,što odgovara sadržaju od 0,06-0,12%magnoflorina u biljnom materijalu). Sasvim je verovatno da je potvrđena citotoksična aktivnost metanolnog ekstrakta podzemnih organa (300 μg/ml; 39 % preživelih ćelija K562 kulture maligno-transformisanih ćelija), kao i imunomodulatorna aktivnost, povezana sa visokom koncentracijom ovog alkaloida u ispitivanom ekstraktu.

Immunomodulatory effects of the methanolic extract of Epimedium alpinum in vitro

AB  - The effect of the methanolic extract of root and rhizome of Epimedium alpinum (MEEA) on phenotype and functions of rat lymphocytes in vitro was studied. It has been found that MEEA at lower concentrations (0.1 mu g/ml and 1 mu g/ml) significantly enhanced proliferation of splenocytes and thymocytes triggered by concanavalin A (Con A), whereas higher concentrations of the extract (50 mu g/ ml-500 mu g/ml) were inhibitory. The stimulatory effect of MEEA on Con A-induced proliferation of splenocytes correlated with the up-regulation of interleukin-2 receptor alpha (IL-2R alpha) expression. In addition, increased production of IL-2 was observed when a blocking IL-2R alpha monoclonal antibody (mAb) was added to cell cultures. MEEA-suppressed proliferation of splenocytes was due to the inhibition of IL-2 production, the down-regulation of IL-2Ra expression and the induction of apoptosis. Cellular proliferation in the presence of inhibitory concentrations of MEEA higher than 50 mu g/ml could not be restored by the addition of exogenous IL-2.
