HPTLC-based metabolomics approach for the detection of chokeberry (Aronia melanocarpa (Michx.) Elliott) adulteration

AB  - Introduction: Chokeberry is a plant belonging to the family Rosaceae which has a long history of edible and medicinal use. The aims of this study are: i) to investigate of HPTLC profile of the berry samples using HPTLC technique based metabolomics approach, ii) to determine the botanical origin of investigated chokeberry adulterations iii) to identify the most important botanical markers responsible for classification. Methods: A simple, rapid, and high-throughput planar chromatography (HPTLC) method was applied in combination with multivariate techniques for differentiating chokeberry samples and identification of the main metabolites of chokeberry and four common adulterants. Images of HPTLC chromatograms obtained using two different detections i.e. visible light and derivatization with natural products reagent were used as a dataset for multivariate analysis. Results: Principal component analysis and Orthogonal Partial Least Squares Discriminant Analysis confirmed the discrimination of five botanically different samples and recognized their main markers such as responsible for differences between chokeberry and four common adulterants. Conclusion: The obtained results showed that the HPTLC-based metabolomics approach can be a very reliable technique for the detection of chokeberry adulteration.

Comparative Analysis of Phenolic Compounds in Seven Hypericum Species and Their Antioxidant Properties

AB  - A comparative analysis of the phenolic compounds in the 70% EtOH extracts of Hypericum acutum, H. androsaemum, H. barbatum, H hirsutum, H maculatum, and H richeri has been carried out using high-performance liquid chromatography coupled with photodiode array UV detection and high resolution TOF mass spectrometry. Quercetin, astilbin, I3,II8-biapigenin, orientin, 2"-O-acetylorientin, three phenolcarboxylic acids, and eight flavonols 3-O-glycosides were identified in the extracts on the basis of their on-line UV spectra, accurate mass spectral data, and in comparison of retention times with those from the standards. Fingerprint analysis of the extracts revealed significant differences in the qualitative and quantitative chemical composition of the studied species. Antioxidant assays with various reaction mechanisms were used including ferric reducing antioxidant power (FRAP) assay, DPPH, ABTS, superoxide anion radical scavenging capacity and inhibition of liposome peroxidation induced by Fe2+. The most potent were extracts of H acutum and H maculatum indicating this Hypericum species interesting for further research aimed as a potentially new source of biologically active compounds.

AB  - The chemical composition and free radical scavenging properties of the MeOH extract of Hypericum richeri and its subsequent CHCl3, n-BuOH, EtOAc and aqueous extracts were investigated. Four quercetin-3-O-glycosides, three myricetin-3-O-glycosides, 3-O- and 5-O-caffeoylquinic acid, quercetin, I3,II8-biapigenin, pseudohypericin and hypericin were identified. The constituents of the extracts were quantified using HPLC-DAD (high-performance liquid chromatography-diode array detector). Isolation of the compounds from n-BuOH and EtOAc extracts was conducted by column chromatography. Antioxidant capacity of the extracts was evaluated by superoxide radical and ABTS root+ scavenging assays and inhibition of lipid peroxidation. The EtOAc extract of H. richeri was the most potent antioxidant in all experiments. Free radical scavenging compounds were identified by HPLC-DPPH root (high-performance liquid chromatography-2,2-diphenyl-1-picrylhydrazyl) post-column derivatisation method, where all phenolic compounds except I3,II8-biapigenin exhibited radical scavenging properties. This is the first report of the presence of myricetin-3-O-rutinoside, myricetin-3-O-galactoside, myricetin-3-O-glucoside, 5-O-caffeoylquinic acid and I3,II8-biapigenin in H. richeri.

Anti-inflammatory and Gastroprotective Properties of Hypericum richeri Oil Extracts

AB  - Oil extracts of flowering tops of Hypericum richeri Vill. prepared in three different ways were evaluated for chemical composition, and anti-inflammatory and gastroprotective activities. An HPLC method was developed for determination of two dominant flavonoids, quercetin and I3,II8-biapigenin. The carrageenan-induced rat paw edema test was used for screening the anti-inflammatory activity, while indomethacin-induced rat gastric mucosa damage test was used for evaluation of gastroprotective activity. The oil extract prepared by maceration with 96% ethanol, followed by extraction with sunflower oil by heating on a water bath, exhibited the highest anti-inflammatory (38.4 %) and gastroprotective activities (gastric damage score of 0.9). The same oil extract had the highest content of quercetin (49 mu g/mL) and I3,II8-biapigenin (60 mu g/mL). These results approve the usage of oil extracts of H. richeri as an anti-inflammatory and gastroprotective agent.

Pyrrolizidine alkaloids from seven wild-growing Senecio species in Serbia and Montenegro

AB  - The genus Senecio (family Asteraceae) is one of the largest in the world. It comprises about 1100 species which are the rich source of pyrrolizidine alkaloids. Plants containing pyrrolizidine alkaloids are among the most important sources of human and animal exposure to plant toxins and carcinogens. The pyrrolizidine alkaloids of seven Senecio species (S. erucifolius, S. othonnae, S. wagneri, S. subalpinus, S. carpathicus, S. paludosus and S. rupestris) were studied. Fourteen alkaloids were isolated and their structures determined from spectroscopic data (1H- and 13C-NMR, IR and MS). Five of them were identified in S. erucifolius, four in S. othonnae, two in S. wagneri, four in S. subalpinus, two in S. carpathicus, three in S. paludosus and three in S. rupestris. Seven pyrrolizidine alkaloids were found for the first time in particular species. The results have chemotaxonomic importance. The cytotoxic activity and antimicrobial activity of some alkaloids were also studied.
AB  - Jedan od najbrojnijih rodova na svetu je rod Senecio (familija Asteraceae). Obuhvata oko 1100 vrsta koje predstavljaju bogat izvor pirolizidinskih alkaloida. Najznačajnija izloženost ljudi i životinja biljnim toksinima i karcinogenima potiče od biljaka koje sadrže pirolizidinske alkaloide. U ovom radu izolovani su pirolizidinski alkaloidi iz sedam vrsta roda Senecio (S. erucifolius, S. othonnae, S. wagneri, S. subalpinus, S. carpathicus, S. paludosus i S. rupestris), a njihove strukture određene na bazi spektroskopskih podataka (1H- i 13C-NMR, IR i MS). Izolovano je ukupno 14 alkaloida, pet iz S. erucifolius, četiri iz S. othonnae, dva iz S. wagneri, četiri iz S. subalpinus, dva iz S. carpathicus, tri iz S. paludosus i tri iz S. rupestris. Evaluation of Hypericum perforatum Oil Extracts for an Antiinflammatory and Gastroprotective Activity in Rats

AB  - Oil extracts of Hypericum perforatum L. (Oleum Hyperici) were prepared in three different ways according to the prescriptions from traditional medicine. Variability of constituents and biological activity were evaluated in the obtained oil extracts. The carrageenan-induced rat paw edema test has been used for screening the antiinflammatory activity, while the indomethacin-induced rat gastric mucosa damage test was used for evaluation of gastroprotective activity. All examined oil extracts possessed antiinflammatory and gastroprotective activity. Among them, the oil extract prepared by maceration with 96% ethanol, followed by extraction with sunflower oil by heating on a water bath (extract 2), in a dose of 1.25 mL/kg p.o., exhibited the highest antiinflammatory effect (95.24 +/- 11.66%) and gastroprotective activity (gastric damage score of 0.21 +/- 0.12). The same oil extract had the highest content of quercetin and I3,II8-biapigenin (1.29 +/- 9 mu g/mL and 52 +/- 4 mu g/mL, respectively). Quercetin and I3,II8-biapigenin exhibited antiinflammatory activity similar to those of indomethacin as well as significant gastroprotective activity. The results provide evidence for the usage of Oleum Hyperici as an antiinflammatory and gastroprotective agent, Which has been based previously only on ethnopharmacological claims. Copyright

AB  - In this paper the first complexes of M2+ ions (M2+ = Zn2+, Cd2+ and Co2+) with N-benzyloxycarbonyl-S-phenylalaninato ligand (1-3) are described. The new complexes were characterized by means of elemental analysis, IR and UV-vis spectroscopy, molar conductivity measurements and H-1, C-13 and N-15 NMR spectroscopy (1D and 2D). The Co(II) complex adopts an octahedral geometry, and the Zn(II) and Cd(II) complexes adopt a tetrahedral one. For the first time, the antimicrobial activity of N-benzyloxycarbonyl-S-phenylalaninato ligand (N-Boc-S-phe) and the complexes 1-3 was investigated against Gram-positive: Micrococcus luteus, Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis and Bacillus subtilis and Gram-negative bacteria: Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and two strains of the yeast Candida albicans. It was shown that the complexes were effective against most strains. The best activity was detected against the yeast C. albicans.
