AB  - The present study investigated the effects of PCBs on the rat kidneys with attention given to the determination critical effect dose (CED) using the Benchmark dose (BMD) approach. Male albino Wistar rats (7 animals per group) were given by oral gavage Aroclor 1254 dissolved in corn oil at doses of 0.0, 0.5, 1, 2, 4, 8, or 16 mg/kg b.w./day for 28 days. The PCB nephrotoxicity was manifested by a dose-dependent changes in serum urea levels. The study has also revealed PCB-induced oxidative stress induction in kidneys. The observed nephrotoxic effects can be partly explained by oxidative damage of lipids and proteins in the kidneys due to observed reduced CuZnSOD activity and disturbances in antioxidant protection. Аll the renal oxidative stress parameters showed dependence on PCB oral doses as well as internal, measure kidney PCB levels. Calculated BMDL values were lower than estimated no observed adverse effect levels (NOAEL) based on the study, suggesting the importance of BMD approach use in future risk assessment.

Influence of the mixture of Cd and BDE-209 on Zn and Cu homeostasis in rats

AB  - Humans are exposed to the chemicals from very different sources. Therefore, the
exposure assessment should be focused on mixtures rather than single chemicals. In this
study, bioelements (Zn and Cu) homeostasis effects of mixture of Cd and BDE-209, widely
spread persistent environmental pollutants, has been evaluated. In order to evaluate the
potential of mixture on Zn and Cu homeosthasis in rats kidneys, Wistar rats were treated by
oral gavage during 28 days with three doses of Cd 2.5, 7.5, and 15 mg/bw/day, three
combinations of Cd with 1000 mg BDE-209/kg/day, and water as a control group. Kidney
tissue samples were collected, mineralised, and Cu and Zn levels were measured by flame
atomic absorbtion spectrophotometer. Possible interactions were evaluated comparing the
dose-response curves slopes and lower confidence limit of Benchmark dose of 10%
(BMDL10) were calculated by PROAST software (RIVM, Netherlands). Cd alone can disturb
Zn in kidneys and calculated BMDL10 was 1.713 mg Cd/kg/day, while in combination with
BDE-209 it was 1.719 mg/kg/day. Cd disturbs Cu in kidney in significantly higher dose of
5.679 mg Cd/kg/day. However, in combination with the BDE-209, derived BMDL10 was 10
times lower, 0.517 mg Cd/kg/day. Dose-response curve slope for the effect on Cu was
significantly lower in the case of mixture effect than in the case of Cd effect. Since Cu plays a
major role in the blood cells creation and health of other systems, our results indicate more
potent activity of the mixture than single Cd.
AB  - Ljudi mogu biti izloženi hemikalijama iz različitih izvora, te je potrebno procenjivati
efekte na bazi izloženosti smešama hemikalija, pre nego pojedinačnim hemikalijama. U
studiji je analiziran uticaj smeše Cd i BDE-209, široko rasprostranjenih zagađujućih
perzistentnih supstanci, na homeostazu bioelemenata, cinka (Zn) i bakra (Cu). U cilju
evaluacije potencijala smeše da naruši homeostazu Zn i Cu u bubrezima pacova, Wistar
pacovi su oralnom gavažom bili tretirani tokom 28 dana dozama Cd od 2,5, 7,5 i 15
mg/kg/dan, kombinacijama ove tri doze sa 1000 mg BDE-209/kg/dan i vodom kao
kontrolom. Tkivo bubrega je izdvojeno, zatim mineralizovano, a sadržaj Zn i Cu je meren
atomskom apsorpcionom spektrofotometrijom. Moguće interakcije su procenjivane
poređenjem nagiba krivih doza-odgovor, a izračunate su i donje granice pouzdanosti
Benchmark doze od 10% (BMDL10) korišćenjem PROAST softvera (RIVM, Holandija). Cd
primenjen sam utiče na homeostazu Zn u bubregu i izračunata BMDL10 iznosi 1,713
mg/kg/dan, dok u kombinacji sa BDE-209 iznosi 1,719 mg/kg/dan. Cd ometa homeostazu Cu
u bubregu pacova i BMDL10 iznosi 5,679 mg/kg/dan, dok je u kombinaciji sa BDE-209 BMDL
10 puta niža i iznosi 0,517 mg/kg/dan. Nagib krive doza odgovor za smešu Cd i BDE-209 bio
je daleko niži od nagiba krive doza-odgovor za Cd. S obzirom na to da Cu igra značajnu ulogu
u nastanku krvnih ćelija, ali i u očuvanju durgih sistema organa, dobijeni rezultati iziskuju
pažnju zbog daleko potentnijeg uticaja smeše na nivo Cu u odnosu na uticaj samog Cd.

Human health risk assessment of lead, cadmium, and mercury co-exposure from agricultural soils in the Tuzla Canton (Bosnia and Herzegovina)

AB  - The aim of this study was to assess the risk of human exposure to lead (Pb), cadmium (Cd), and mercury (Hg) through agricultural soil by considering both uncertainty and variability in key exposure parameters. For this reason we collected soil samples from 29 locations in the Tuzla Canton (Bosnia and Herzegovina) and measured their metal levels with inductively coupled plasma atomic emission or absorption spectrometry (ICP-AES and ICP-AAS, respectively). The levels of Pb ranged from 13.33 to 1692.33 mg/kg, of Cd from 0.05 to 3.67 mg/kg, and of Hg from 0.02 to 2.73 mg/kg. To estimate cancer and non-cancer risks we used deterministic and semi-probabilistic methods. Lead was found to involve higher health risk than the other two heavy metals. Its hazard index (HI) decreased between population groups (children>women>men) and exposure routes (ingestion>skin contact>inhalation). Our Monte Carlo simulations indicated that Pb HIs for both adult populations had a 0.6 % probability to exceed the threshold value of 1, while in children this probability was 14.2 %. Cd and Hg showed no probability to exceed the threshold in any scenario. Our simulation results raise concern about possible adverse health effects of heavy metals from soil, especially in children. It is very important to continue monitoring environmental pollution and assess human health risk, not only with respect to soil, but also with other important environmental compartments, such as air and water.
AB  - Cilj ovog istraživanja bio je procijeniti izloženost ljudi olovu (Pb), kadmiju (Cd) i živi (Hg) iz poljoprivrednog tla,
uzimajući u obzir pridružene nesigurnosti i varijabilnosti u ključnim parametrima izloženosti. Primjenom metoda induktivno
spregnute plazme i atomske emisijske ili apsorpcijske spektrometrije (ICP-AES i AAS) određena je koncentracija metala
u uzorcima poljoprivrednoga tla prikupljenog s 29 lokacija u Tuzlanskom kantonu (Bosna i Hercegovina). Koncentracije
Pb kretale su se u rasponu od 13,33 do 1692,33 mg/kg, Cd od 0,05 do 3,67 mg/kg i Hg od 0,02 do 2,73 mg/kg. Za procjenu
kancerogenog i nekancerogenog rizika koristili smo se determinističkim i semiprobabilističkim pristupom u procjeni
rizika. Utvrđeno je da Pb doprinosi povećanom zdravstvenom riziku više nego druga dva teška metala. Indeks opasnosti
(eng. hazard index ‒ HI) smanjivao se među populacijskim skupinama (djeca > žene > muškarci) i putevima izloženosti
(ingestija > dermalni kontakt > inhalacija). Naše Monte Carlo simulacije pokazale su da HI za Pb uključujuči obje
populacije odraslih imaju 0,6 % vjerojatnosti da će prijeći vrijednost praga od 1, dok je u djece ta vjerojatnost bila 14,2 %.
Vjerojatnost da će premašiti prag u bilo kom scenariju nisu pokazali Cd i Hg. Rezultati naših simulacija izazivaju zabrinutost
zbog mogućih štetnih učinaka teških metala iz tla, posebice u djece. Vrlo je važno nastaviti pratiti onečišćenje okoliša i
procijeniti rizik za zdravlje ljudi, ne samo putem tla, već i putem drugih značajnih dijelova okoliša, poput zraka i vode.

The aim of our study was to investigate the associations between periodontal diseases, secretor status, and interleukin-1-β (IL1-ß) and prostaglandine E2 (PGE2) levels in women delivered preterm. Methods. The study included 56 preterm delivery women and 56 women delivered at term as a control group, aged between 17 and 41 years. Periodontal examination, blood and saliva sampling were performed within 48 hours following delivery. Secretor phenotype was determined by hemagglutination inhibition method. The concentrations of IL1-ß and PGE2 were measured by high sensitivity Enzyme-linked Immunosorbent Assay (ELISA). Results. In the pre-term birth group there were 66.1% of women with periodontitis, while in the control one there were 12.5% (p < 0.01). Concentrations of IL1-ß and PGE2 in plasma were significantly higher in the non-secretor group of women who gave birth pre-term and had periodontitis comparing to other groups. There was a significant correlation between salivary and plasma levels of PGE2 and IL1-ß in the preterm birth group (R = 0.416, p = 0.017 and R = -0.592, p < 0.001, respectively). There were no such correlations in women who delivered at term. Conclusion. Our results support the hypothesis that non-secretor phenotype and periodontitis are at least in part responsible for pathogenesis of preterm birth. This probability of negative impact of non-secretor status cannot be ignored. These findings support the need for additional research into the biology of human parturition. © 2020 Inst. Podaci iz literature govore u prilog tome da periodontalni procesi mogu uticati na fetoplacentalnu jedinicu i indukovati preterminski porođaj. Sekretorni status može uticati na stepen periodontalne bolesti. Proinflamatorni citokini imaju uticaj na periodontitis kao i na porođaj. Kombinovani uticaj ovih faktora rizika za prevremeni porođaj nije dovoljno istražen. Cilj ove studije je bio da istraži povezanost između periodontalne bolesti, sekretornog statusa, nivoa interleukina 1-b (IL1-b) i prostaglandina E2 (PGE2) kod žena koje su imale prevremeni porođaj. Naši rezultati podržavaju hipotezu da su sekretorni status i periodontitis, bar delimično, odgovorni za patogenezu preterminskog porođaja. Verovatnoća negativnog uticaja nesekretornog statusa se ne sme ignorisati. Ovi zaključci ukazuju na potrebu za dodatnim istraživanjima porođaja.

AB  - Soil contamination by heavy metals is a serious global environmental problem, especially for developing countries. A large number of industrial plants, which continually pollute the environment, characterize Tuzla Canton, Bosnia and Herzegovina. The aim of this study was to assess the level of soil pollution by heavy metals and to estimate cytotoxicity and genotoxicity of soil leachates from this area. Lead (Pb), cadmium (Cd) and mercury (Hg) were analyzed by ICP-AES and AAS. Soil contamination was assessed using contamination factor, degree of contamination, geoaccumulation index and pollution load index. To determine the connection of variables and understanding their origin in soils, principal component analysis (PCA) and cluster analysis (CA) were used. The results indicate that Cd and Hg originated from natural and anthropogenic activities, while Pb is of anthropogenic origin. For toxicity evaluation, CaCo-2 and HaCaT cells were used. PrestoBlue assay was used for cytotoxicity testing, and cH2A. X for genotoxicity evaluation. Concerning cytotoxicity, Cd and Hg had a positive correlation with cytotoxicity in HaCaT cells, but only Hg induced cytotoxicity in CaCo-2 cells. We also demonstrate that soil leachates contaminated by heavy metals can induce genotoxicity in both used cell lines. According to these results, combining bioassays with standard physicochemical analysis can be useful for evaluating environmental and health risks more accurately. These results are important for developing proper management strategies to decrease pollution. This is one of the first studies from this area and an important indication of soil quality in Southeast Europe.

AB  - It is known that kidneys are target tissue for Cd toxicity but there is not enough literature data on kidney effects of BDE-209. Moreover, Cd as toxic metal and BDE-209 as organic halogenated pollutant can share similar mechanism of action like, oxidative stress which is already assumed. However, how will Cd act as prooxidant in presence of BDE-209 is still not investigated, therefore, the objective of this study was to assess this BDE-209 influence. Results of this study indicated slight decrease in MDA in rat's kidney homogenates after subacute exposure to Cd and/or BDE-209, while SOD activity was increased and content of -SH groups decreased. Based on dose-response assessment, BDE-209 did not influence Cd toxicity mediated by chosen oxidative stress parameters, namely the derived CEDL5 values were the same for single Cd and for Cd given in combination with the BDE-209. In conclusion, dynamic of oxidative process has influence on the parameters levels. Co-exposure with polybrominated organic pollutant did not influence Cd effects on oxidative stress parameters. Having in mind that this is pilot study, performed with only one dose of BDE-209, further investigations are necessary, including changes in dose ranges, duration of exposure etc. Moreover, results contributes to the issue of mixture toxicology even BDE-209 did not influenced oxidative stress as mechanism of Cd toxicity.
AB  - Poznato je da su bubrezi ciljni organ toksičnosti kadmijuma, ali je mnogo manje literaturnih podataka o toksičnim efektima BDE-209 na bubrege. Kadmijum, kao toksičan metal i BDE-209, kao organska halogenovana zagađujuća supstanca mogu imati sličan mehanizam dejstva posredovan oksidativnim stresom. Međutim, kako će Cd da deluje kao prooksidant u prisustvu BDE-209 još uvek nije proučavano, te je cilj ovog rada bio da se proceni uticaj BDE-209 na oksidativni stres posredovan kadmijumom. Rezultati ovog istraživanja ukazuju na blagi pad koncentracije MDA u homogenatima bubrega pacova nakon subakutne ekspozicije Cd i/ili BDE-209, dok se aktivnost SOD povećava i sadržaj - SH grupa smanjuje. Na osnovu procene odnosa doza-odgovor, BDE-209 nije imao uticaja na toksičnost Cd izazvanu posredstvom oksidativnog stresa (posmatrajući odabrane parametre). Potvrda je dobijena i iz izračunatih donjih granica pouzdanosti doze od 5% - CEDL5 jer su vrednosti su bile iste za sam Cd i Cd primenjen u kombinaciji sa BDE-209. Može se zaključiti da dinamika oksidativnih procesa ima uticaj na vrednosti parametara oksidativnog stresa, da stoga istovremena ekspozicija Cd i polibromovanom organskom polutantu nije promenila uticaj samog Cd na parametre oksidativnog stresa. Imajući u vidu da je ovo pilot studija, izvršena sa samo jednom dozom BDE-209, potrebna su dalja istraživanja, uključujući i promene opsega doza, trajanje ekspozicije itd. Pored toga, rezultati ove studije doprinose tumačenju toksikologija smeša, iako BDE-209 nije imao uticaja na oksidativni stres kao mehanizam Cd toksičnosti za date uslove ekspozicije.

AB  - Aim: The aim of this study was to examine the role of IL-33/ST2 pathway in a pathogenesis of acute inflammation and its effects on tissue damage, antioxidative capacity, magnesium concentration and cytokine profile in acutely inflamed tissue. Material and methods: Male mice were randomly divided in four groups: wild-type control group (WT-C), ST2 knockout control group (KO-C), wild-type inflammatory group (WT-I), and ST2 knockout inflammatory group (KO-I). Acute inflammation was induced in WT-I and KO-I by intramuscular injection of turpentine oil, while mice in WT-C and KO-C were treated with saline. After 12 h, animals were euthanized, and blood was collected for determination of creatine kinase (CK) and aspartate transaminase (AST) activity. The treated tissue was used for histopathological analysis, determination of volume density of inflammatory infiltrate (Vdii) and necrotic fiber (Vdnf), gene expression of interleukin (IL)-33, ST2, tumor necrosis factor alpha (TNF-alpha), IL-6, IL-12p35, and transforming growth factor beta (TGF-beta), concentration of magnesium (Mg), copper (Cu), selenium (Se), manganese (Mn) and reduced glutathione (GSH), and superoxide dismutase (SOD) and glutathione peroxidase (GPx) activity. Results: Presence of inflammatory infiltration and necrosis in the treated tissue was histopathologically confirmed in WT-I and KO-I. Vdii was significantly higher in WT-I when compared to KO-I, whereas Vdnf did not significantly differ between WT-I and KO-I. CM and AST significantly increased in both inflammatory groups when compared to corresponding control groups. However, the values of CK and AST were significantly higher in WT-I than in KO-I. Mg in the treated tissue was significantly lower in WT-I in comparison to WT-C and KO-I, while there was no significant difference between KO-C and KO-I. There was no significant difference in Cu, Se, and Mn in the treated tissue between WT-C, KO-C, WT-I and KO-I. Gene expression of IL -33 in the treated tissue increased in both inflammatory groups when compared to the corresponding control groups, but it was significantly higher in KO-I than in WT-I. Gene expression of ST2 in the treated tissue was significantly higher in WT-I than in WT-C. Gene expression of TNF-alpha, IL-6, and IL-12p35 in the treated tissue was significantly higher in WT-I and KO-I than in the corresponding control groups, and IL-6 was significantly higher in KO-C than in WT-C. TGF-beta gene expression in the treated tissue was significantly higher in KO-I when compared to WT-I, while there was no difference between WT-C and KO-C. SOD activity decreased at the site of acute inflammation in both inflammatory groups, while the GPx activity increased. GSH in the treated tissue was significantly higher in KO-I than in KO-C or WT-I. Conclusion: The results of our study have indicated, to our knowledge for the first time, that IL-33/ST2 pathway plays a role in enhancing inflammation and tissue damage at the site of acute inflammation by affecting the concentration of magnesium and GSH, important for antioxidative capacity, as well as gene expression of anti-inflammatory cytokine TGF-beta.

Based on numerous studies in animals, the most prominent toxic effects of decabrominated di-phenyl ether (BDE-209) are observed in the liver, thyroid hormone homeostasis, reproductive and nervous systems. BDE-209 exhibits its toxic effects partly through the aryl hydrocarbon (Ah) receptor and consequent induction of hepatic microsomal enzymes. The aim of this study was to assess the hepatotoxic effect vs target tissue dose of BDE-209 in the subacutely orally exposed Wistar rats. Methods. Effects were examined on male Wistar rats, weighing 200-240 g, exposed to doses of 1,000, 2,000 or 4,000 mg BDE-209/kg body weight (bw)/day by gavage during 28 days. Animals were treated according to the decision of the Ethics Committee of the Military Medical Academy, No 9667-1/2011. Evaluation of the hepatotoxic effect was based on: relative liver weight water and food intake, biochemical parameters of liver function [aspartate amino transferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), gama glutamyl transferase (γ-GT)], and oxidative stress parameters in liver homogenates [malondialdehiyde (MDA), superoxide dismutase (SOD), -SH] and morphological and pathohistological changes in the liver. For the assessment of internal dose - response relationship, lower confidence limit of Benchmark dose (BMDL) of 5% or 10% i.e. BMDL5 or BMDL10, were calculated using PROAST software. Results. After the application of 1,000, 2,000 or 4,000 mg BDE-209/kg bw/day, the concentrations of BDE-209 measured in liver were 0.269, 0.569 and 0.859 mg/kg of liver wet weight, (ww) respectively. Internal doses correlated with external (r = 0.972; p  lt  0.05) according to equation: internal dose (mg BDE-209/kg of liver ww) = 0.0002 ' external dose (mg/kg bw/day) + 0.0622. Hepato-toxicity was demonstrated based on significant increase in AST and γ-GT activities and the degree of histopathological changes. The lowest BMDL5 of 0.07228 mg BDE-209/kg of liver ww, correlating to external dose of 39 mg/kg/day, indicated the increase of AST activity as the most sensitive biomarker of BDE-209 hepatotoxicity in subacutely ex-posed rats. Conclusion. The results of the present work add up to the issue of BDE-209 toxicity profile with a focus on relationship between internal dose and hepatotoxicity. Critical internal dose for the effect on AST of 0.07 mg/kg of liver ww, corresponding to external dose of 39 mg/kg/day, is the lowest dose ever observed among the studies on BDE-209 hepatotoxicity. For the persistent sub-stances with low absorption rate such as BDE-209, critical effect based on internal dose in majority of cases is considered as more precisely defined than the effect established based on external dose, particularly.

Prema podacima iz brojnih studija na životinjama, dekabromovani difeniletar (BDE-209) najznačajnije toksične efekte ispoljava na jetri, homeostazi hormona štitaste žlezde, reproduktivnom i nervnom sistemu. BDE-209 ispoljava toksične efekte delom preko receptora za aromatične ugljovodonike (Ah) i posledične indukcije mikrozomalnih enzima jetre. Cilj rada bio je procena hepatotoksičnog efekta u odnosu na dozu BDE-209 u ciljnom tkivu kod subakutno oralno eksponovanih Wistar pacova. Metode. Efekti su ispitivani na mužjacima Wistar pacova, mase 200-240 g, koji su putem oralne sonde primali doze od 1 000, 2 000 ili 4 000 mg BDE-209/kg telesne mase (tm) dan, tokom 28 dana. Životinje su tretirane u skladu sa odlukom Etičkog komiteta Vojnomedicinske akademije u Beogradu br. 9667-1/2011. Procena hepatotoksičnih efekata bazirana je na merenju relativne mase jetre, unosa vode i hrane, biohemijskih parametara funkcije jetre [aspartat aminotransferaza (AST), alanin amino-transferaza (ALT), alkalna fosfataza (ALP), gama glutamil transferaza (γ-GT)], parametara oksidativnog stresa u homogenatima jetre [malondialdehid (MDA), superoksid dizmutaza (SOD), -SH)] i morfoloških i histoloških promena na jetri. Za procenu odnosa interna doza - odgovor izračunavana je donja granica pouzdanosti granične Benchmark doze (BMDL) od 5% (BMDL5) ili 10% (BMDL10) primenom PROAST softvera. Rezultati. Koncentracije BDE-209 iznosile su 0,269, 0,569 i 0,859 mg/kg jetre nakon aplikacije 1 000, 2 000, odnosno 4 000 mg BDE-209/kg tm/dan. Interna doza u našoj studiji korelisala je sa eksternom dozom prema jednačini: interna doza (mg BDE-209/kg jetre) = 0,0002 ' eksterna doza (mg/kg tm/dan) + 0,0622 (r = 0,972; p  lt  0,05). Hepatotoksičnost je potvrđena na osnovu nalaza o povećanju aktivnosti enzima AST i γ-GT, kao i stepena patohistološkog oštećenja jetre. Najniža BMDL5 u eksperimentu od 0,07228 mg BDE-209/kg jetre, koja koreliše sa eksternom dozom od 39 mg/kg tm/dan izračunata je za aktivnost AST i ukazuje na to da je aktivnost AST ujedno i najosetljiviji biomarker hepatotoksičnosti BDE-209 kod subakutno eksponovanih pacova. Zaključak. Rezultati prezentovane studije daju doprinos pitanju toksikološkog profila BDE-209 sa fokusom na odnos između interne doze i hepatotoksičnih efekata. Kritična interna doza za efekat na AST od 0,07 mg/kg jetre, koja koreliše sa eksternom dozom od 39 mg/kg tm/dan, jeste ujedno i najniža kritična doza do sada definisana za hepatotoksične efekte BDE-209. Kritičan efekat koji se bazira na dozi u ciljnom tkivu u većini slučajeva može se smatrati preciznije definisanim od kritičnog efekta definisanog na bazi oralno primenjene doze, naročito za nedegradabilne supstance sa niskim stepenom apsorpcije, kao što je BDE-209.

AB  - Mackerel is rich in omega-3 fatty acids, but this fish may be contaminated with environmental pollutants. The aim of this study was to evaluate concentrations of non-dioxin like polychlorinated biphenyls (ndl-PCBs) in 160 samples of mackerel available on the Serbian market. Average and maximum ndl-PCB concentrations were 7.9 ng/g and 74 ng/g, respectively. A higher average level of ndl-PCBs was found in mackerel from Spain, but this was still three times lower than the permitted level. We conclude that levels of ndl-PCBs in mackerel available on the Serbian market are acceptable.

AB  - Hepatotoxicity is one of the well-documented adverse health effects of polychlorinated biphenyls (PCBs)persistent organic pollutants widely present in the environment. Although previous studies suggest possible role of oxidative stress, the precise mechanisms of PCB-induced ROS production in liver still remain to be fully assessed. The aim of this study was to evaluate the effects of different doses of PCBs on the parameters of oxidative stress and to investigate whether these effects are dose dependent. Furthermore, a comparison between calculated benchmark doses (BMD) and estimated NOAEL values for investigated parameters, was made. Six groups of male albino Wistar rats (7 animals per group) were receiving Aroclor 1254 dissolved in corn oil in the doses of 0.5, 1, 2, 4, 8, 16 mg PCBs/kg b.w./day by oral gavage during 28 days while control animals were receiving corn oil only. The following parameters of oxidative stress were analyzed in liver homogenates: superoxide dismutase activity, glutathione, malondialdehyde (MDA) and total protein thiol levels. Hepatic enzymes AST, ALT, ALP and protein albumin were also determined in serum as clinical parameters of liver function. Collected data on the investigated parameters were analyzed by the BMD method. The results of this study demonstrate that subacute exposure to PCBs causes induction of oxidative stress in liver with dose-dependent changes of the investigated parameters, although more pronounced adverse effects were observed on enzymatic than on non-enzymatic components of antioxidant protection. The obtained values for BMD and NOAEL support the use of BMD concept in the prediction of health risks associated with PCBs exposure. Furthermore, our results implicate possible use of MDA in PCBs risk assessment, since MDA was the most sensitive investigated parameter with calculated low critical effect dose of 0.07 mg/kg b.w.

AB  - This study deals with the examination of total aescin content in flowers and seeds of Aesculus hippocastanum L. and Aesculus x carnea Hayne (Hippocastanaceae), collected in urban areas, as well as the content of Pb, Cd and Hg as an indicator of potential aerial pollution. Aescin identification was performed by TLC. Total aescin content was determined by UV-VIS spectrophotometry and HPTLC densitometry. The contents of Pb, Cd and Hg was determined by AAS. The concentration of aescin varied in a wide range between 0.17% (A. hippocastanum hulls) and 2.95% (A. hippocastanum cotyledons), as determined by spectrophotometric assay. Slightly lower results were recorded in HPTLC densitometry assay, ranging from 0,10% (the hulls of both plant species) to 2.39% (A. x carnea cotyledons). The extractable matter yield was between 5% and 26%, with a high share of total aescin (5% - 13%, depending on the source). The levels of Pb, Cd and Hg both in the plant material and the extracts remained low, indicating that the health safety of the plant specimens was not compromised. Our results suggest that the seeds and flowers of Chestnut trees in urban areas could be considered as a raw material for chemical and pharmaceutical industry in Serbia.
AB  - Osnovni cilj ovog rada je bio da se utvrdi ukupni sadržaj escina u cvetu i plodu Aesculus hippocastanum L. i Aesculus x carnea Hayne (Hippocastanaceae), sakupljenim u gradskom području, kao i sadržaj Pb, Cd i Hg kao indikatora potencijalnog gradskog aerozagađenja. Identifikacija escina u ispitivanom materijalu je izvršena primenom TLC. Ukupni sadržaj escina je određen primenom UV-VIS spektrofotometrije i HPTLC denzitometrije, kako u biljnom materijalu, tako i u ekstraktima dobijenim ekstrakcijom pomoću 60% etanola. Sadržaj Pb, Cd i Hg je određen primenom AAS. Rezultati spektrofotometrijskog određivanja pokazuju da se koncentracija escina nalazi u širokom rasponu između 0,17% (semenjača ploda A. hippocastanum) i 2,95% (kotiledoni semena A. hippocastanum). Nešto niži rezultati su dobijeni primenom HPTLC denzitometrije, između 0,10% (semenjača ploda obe biljne vrste) i 2,39% (kotiledoni semena A. x carnea). Biljni materijal sadrži između 5% i 26% ekstraktibilnih materija, sa visokim udelom ukupnog escina (5% - 13%, u zavisnosti od izvora). Koncentracija Pb, Cd i Hg, kako u biljnom materijalu, tako i u ekstraktima, bila je niska, što implicira da zdravstvena ispravnost uzoraka biljnog materijala nije ugrožena činjenicom da je biljni materijal sakupljen u gradskoj sredini. Naši rezultati ukazuju na to da seme i cvet obe vrste kestena, koji se gaje u urbanim sredinama radi svojih dekorativnih svojstava, ali proizvode značajnu količinu komunalnog otpada, mogu da se koriste kao sirovina za proizvodnju vrednih proizvoda za upotrebu u hemijskoj i farmaceutskoj industriji.

Determination of hidden gluten in food supplements and foods for particular nutritional uses from Serbian market

AB  - Coeliac disease is characterized by permanent intestinal intolerance to wheat gliadin and related prolamins from barley, rye and oats. The gluten free diet is a life long treatment for coeliac patients. Industrial starch or other ingredients used in manufacturing of dietary products can contain prolamins as impurities. The aim of this work was investigation the presence of gluten in dietary products from the national market: 30 dietary foods for infants and young children and 35 food supplements were analyzed using the Ridascreen Gliadin sandwich R5 enzyme-linked immunosorbent assay with cocktail extraction. The most of the analyzed samples (88%) contained less than limit of quantification of 5 mg/kg for gluten. Gluten content of 5-20 mg/kg was determined in 3 samples. Five of 65 samples (8%) contained mean gluten levels ≥ 20 mg/kg and would not be considered "gluten-free" according to the national Regulation on health safety of dietary products. The systematic control of the presence of gluten in gluten-free dietary products is very important and should be obligatory in order to protect the health of coeliac patients.

abstract = "Coeliac disease is characterized by permanent intestinal intolerance to wheat gliadin and related prolamins from barley, rye and oats. The gluten free diet is a life long treatment for coeliac patients. Industrial starch or other ingredients used in manufacturing of dietary products can contain prolamins as impurities. The aim of this work was investigation the presence of gluten in dietary products from the national market: 30 dietary foods for infants and young children and 35 food supplements were analyzed using the Ridascreen Gliadin sandwich R5 enzyme-linked immunosorbent assay with cocktail extraction. The most of the analyzed samples (88%) contained less than limit of quantification of 5 mg/kg for gluten. Gluten content of 5-20 mg/kg was determined in 3 samples. Five of 65 samples (8%) contained mean gluten levels ≥ 20 mg/kg and would not be considered "gluten-free" according to the national Regulation on health safety of dietary products. The systematic control of the presence of gluten in gluten-free dietary products is very important and should be obligatory in order to protect the health of coeliac patients.",

Assessment of mercury intake associated with fish consumption in Serbia

AB  - Nutritional benefits of fish can be attributed primarily to the content of high-quality proteins, vitamins, elements and omega-3 polyunsaturated fatty acids. On the other hand, fish and fishery products are susceptible to contamination by chemicals that have been recognized as ubiquitous environmental pollutants such as toxic elements and polychlorinated organic compounds. Fish consumption could be therefore considered as one of the major sources of human exposure to all above-mentioned environmental contaminants. This paper is focused on mercury (Hg) that enters the environment by both, natural means (such as volcanic activity, erosions and weathering, factors which contribute to the presence of Hg in water, soil and the atmosphere) and human activities - mining, fossil fuels combustion, industrial emissions, direct application of fertilizers and fungicides as well as disposal of solid waste. Total concentrations of Hg were measured in fish muscle and canned fish products available on Serbian market. Total of 651 samples were analyzed: 350 samples of marine fish (hake, mackerel, sprat, scorpanea, gilthead, salmon), 34 samples of freshwater fish (trout and carp) and 267 samples of canned fish products (tuna and sardines). Data were collected during 2011. For the purpose of intake assessment, we used the data obtained from the GEMS/Food Consumption Cluster Diets database. According to this source, estimated average weekly consumption of marine fish is 106.4 g/week, while freshwater fish and canned fish contribute to the consumption with 29.4 g and 18.2 g/week respectively. Mercury concentrations in marine fish were in the range of 0.005-0.208 μgg-1 (mean 0.040 μgg-1); in freshwater fish 0.005- 0.099 μgg-1 (mean 0.020 μgg-1) and in canned products they were in the range of 0.005-0.642 μgg-1 (mean 0.064 μgg-1). All analyzed samples contained mercury below the maximum level laid down by the European Union and Serbian regulation. The estimated weekly intake for total mercury, based on mean mercury value in fish and average body mass of 70 kg, was 0.095 μg/kg b.w./week. Based on FAO/WHO recommended safe limit and on obtained results, we can conclude that the intake of mercury in the case of consuming fish and canned fish products is lower than the safe limit.
AB  - Nutritivna korist od konzumacije ribe ogleda se, pre svega u sadržaju visoko vrednih proteina, vitamina, makro i mikroelemenata i omega-3 polinezasićenih masnih kiselina. Sa druge strane, riba i proizvodi od ribe su u značajnoj meri podložni hemijskoj kontaminaciji ubikvitarnim zagađivačima kao što su teški metal i polihlorovana organska jedinjenja. Zbog svega ovoga, konzumacija ribe se smatra jednim od najznačajnijih izvora izloženosti navedenim kontaminentima. U radu je ispitivana kontaminacija živom koja se u spoljašnjoj sredini može naći kao posledica prirodnih procesa (vulkanska aktivnost, erozija tla i klimatska dešavanja koja doprinose prisustvu žive u vodi, zemljištu i atmosferi), kao i aktivnosti čoveka - eksploatacija ruda, sagorevanje fosilnih goriva, emisija industrijskih gasova, direktna primena veštačkih đubriva i fungicida, kao i neadekvatno odlaganje čvrstog otpada. Sadržaj ukupne žive je određivan u svežoj i konzervisanoj ribi sa srpskog tržišta. Ispitan je 651 uzorak: 350 uzoraka morske ribe (oslić, skuša, sardela, škarpina, orada i losos), 34 uzorka slatkovodne ribe (šaran i pastrmka) i 267 uzoraka konzervisane ribe (tuna i sardela). Svi uzorci su analizirani tokom 2011. godine. Za procenu unosa korišćeni su podaci iz 'GEMS/Food Consumption Cluster Diets database'. Prema ovom izvoru, procenjena prosečna nedeljna konzumacija morske ribe iznosi 106,4 g, slatkovodne 29,4 g dok je ova vrednost za konzervisane proizvode od ribe 18,2 g. Sardžaj žive u morskoj ribi se kretao u opsegu od 0,005 do 0,208 μgg-1 (srednja vrednost 0,040 μgg-1); u slatkovodnoj ribi 0,005-0,099 μgg-1 (srednja vrednost 0,020 μgg-1), dok se u konzervisanim ribljim proizvodima sadržaj žive kretao od 0,005 do 0,642 μgg-1 (srednja vrednost 0,064 μgg-1). Nivo žive u svim ispitanim uzorcima je bio ispod maksimalno dozvoljenih vrednosti propisanih relevantnom legislativom EU kao i domaćim propisima. Procenjeni nedeljni unos žive baziran na srednjoj vrednosti žive u konzumiranoj ribi i prosečnoj telesnoj težini od 70 kg, bio je 0,095 μg/kg telesne mase nedeljno. Na osnovu preporuka FAO/WHO i dobijenih rezultata, može se zaključiti da je unos žive pri konzumaciji morske i slatkovodne ribe i konzervisanih ribljih proizvoda, niži od preporučenih graničnih vrednosti.

Toxicity mechanisms and interactions of polychlorinated biphenyls and polybrominated diphenylethers

AB  - Polychlorinated biphenyls (PCBs) and brominated flame retardants, polybrominated diphenylethers (PBDEs), are widespread environmental contaminants as a result of anthropogenic activities and due to their persistency and resistance to degradation. Both groups of chemicals are placed on the list of persistent organic pollutants (POPs), covered by the Stockholm convention which is aimed to limit or ban the production, use, emission, import and export of POPs in order to protect human health and the environment. Taking into account the structural similarity between PCBs and PBDEs, and the known effects of PCBs, these two groups of chemicals could have similar mechanisms of action. Also, because of real simultaneous exposure to these compounds, an examination of possible interactions is of great importance. Interactions of the compounds from these groups are likely at the system level of hormones, particularly thyroid and reproductive ones. As there are mechanisms of adverse effects of both groups of chemicals on the nervous system, including functional, neurological and behavioral changes, there are influences on neurotransmiters, changes in signal transduction and apoptosis. There are interactions affecting the occurrence of metabolic disorders as well. Data on the toxicity of mixtures of PCBs and PBDEs, as well as interactions of these chemicals would contribute to the processes of risk evaluation and risk characterisation.
AB  - Polihlorovani bifenili (PCBs) i bromovani usporivači gorenja, polibromovani difenil etri (PBDEs) su široko rasprostranjeni u životnoj sredini kao posledica antropogenih aktivnosti i zahvaljujući njihovoj postojanosti i otpornosti na degradaciju. Obe grupe jedinjenja se nalaze na listi perzistentnih organskih zagađivača (POPs) usvojenoj Stokholmskom konvencijom sa ciljem da zabrani ili ograniči proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs u svrhu zaštite zdravlja ljudi i životne sredine. Uzimajući u obzir strukturnu sličnost između PBDEs i PCBs, kao i činjenicu da je istovremena ekspozicija ovim jedinjenima realna, ispitivanja njihovih interakcija su od značaja za predviđanje rezultujućeg efekta. Saznanja u oblasti toksikologije smeša, takođe doprinose procesu evaluacije i karakterizacije rizika. Interakcije jedinjenja iz ovih grupa su verovatne na nivou endokrinog sistema, naročito tiroidnog i reproduktivnog. S obzirom na mehanizme toksičnosti obe grupe supstanci za nervni sistem, uključujući funkcionalne promene, neurološke poremećaje i promene u ponašanju, moguće je očekivati njihove interakcije i na sistemima neurotransmitera, promene u prenosu signala i izazivanje apoptoze, kao i na pojavu metaboličkih poremećaja.

Interactions of the compounds from these groups are likely at the system level of hormones, particularly thyroid and reproductive ones. As there are mechanisms of adverse effects of both groups of chemicals on the nervous system, including functional, neurological and behavioral changes, there are influences on neurotransmiters, changes in signal transduction and apoptosis. There are interactions affecting the occurrence of metabolic disorders as well. Data on the toxicity of mixtures of PCBs and PBDEs, as well as interactions of these chemicals would contribute to the processes of risk evaluation and risk characterisation., Polihlorovani bifenili (PCBs) i bromovani usporivači gorenja, polibromovani difenil etri (PBDEs) su široko rasprostranjeni u životnoj sredini kao posledica antropogenih aktivnosti i zahvaljujući njihovoj postojanosti i otpornosti na degradaciju. Obe grupe jedinjenja se nalaze na listi perzistentnih organskih zagađivača (POPs) usvojenoj Stokholmskom konvencijom sa ciljem da zabrani ili ograniči proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs u svrhu zaštite zdravlja ljudi i životne sredine. Uzimajući u obzir strukturnu sličnost između PBDEs i PCBs, kao i činjenicu da je istovremena ekspozicija ovim jedinjenima realna, ispitivanja njihovih interakcija su od značaja za predviđanje rezultujućeg efekta. Saznanja u oblasti toksikologije smeša, takođe doprinose procesu evaluacije i karakterizacije rizika. Interakcije jedinjenja iz ovih grupa su verovatne na nivou endokrinog sistema, naročito tiroidnog i reproduktivnog. S obzirom na mehanizme toksičnosti obe grupe supstanci za nervni sistem, uključujući funkcionalne promene, neurološke poremećaje i promene u ponašanju, moguće je očekivati njihove interakcije i na sistemima neurotransmitera, promene u prenosu signala i izazivanje apoptoze, kao i na pojavu metaboličkih poremećaja.",

Use of PROAST software to assess the influence of decabrominated diphenyl ether and/or cadmium on thyroid hormones homeostasis in rats

AB  - In relation to commonly used no-observed-adverse-effect-level (NOAEL) point of departure, because of its statistical power and reliability, Benchmark dose (BMD) approach has been proposed as an alternative in dose-response assessment. The aim of this study was to derive BMD (10%) by means of PROAST software to quantify influence of cadmium (Cd) and/or decabrominated diphenyl ether (BDE209) on thyroid hormones homestasis. Study was conducted on male Wistar rats treated orally by gavage for 28 days by either single substances or their combination. Three groups of animals were dosed with Cd at levels of 2.5, 7.5 and 15 mg/kg b.w./day, three groups of animals were dosed with BDE209 at levels of 1000, 2000 or 4000 mg/kg b.w./day, while 9 groups received different dose mixtures of previously given dose levels of Cd and BDE209 (design 3 x 3). Results of the study have indicated that Cd+BDE209 mixtures are likely more potent to disrupt thyroid function than would be expected from the chemicals individually. Derived BMD - lower confidence limits (BMDL), if ratio BMD/BMDL is  lt  10, were 9.4 mg Cd/kg b.w./day and 2155 mg BDE209/kg b.w./day for the effect on T3; and 6.22 mg Cd/kg b.w./day in the mixture with BDE209 2000 mg/kg b.w./day for the effect on FT3.
AB  - Primena statistički dobijene Benchmark doze (BMD) u proceni rizika predstavlja alternativu najčešće korišćenoj 'dozi bez štetnog efekta' (NOAEL) zbog veće pouzdanosti u analizi odnosa doze i toksičnog efekta. Cilj ovog rada bio je izračunavanje BMD (10%) primenom PROAST softvera radi kvantitativne procene uticaja Cd i/ili BDE209 na homeostazu hormona štitaste žlezde. Ispitivanje je sprovedeno na mužjacima Wistar pacova koji su putem oralne sonde, tokom 28 dana, primali pojedinačne supstance ili njihove kombinacije. Kadmijum je primenjivan u dozama od 2,5, 7,5 i 15 mg Cd/kg t.m./dan, a BDE209 u dozama od 1000, 2000 i 4000 mg BDE209/kg t.m./dan, dok je ostalih devet grupa životinja primalo kombinacije hemikalija (dizajn 3 x 3). Rezultati studije ukazuju da smeša Cd i BDE209 verovatno izaziva intenzivniji poremećaj funkcije štitaste žlezde nego svaka od supstanci pojedinačno.
