Role of magnesium in depression?

AB  - Magnesium has a fundamental role in human body and it is necessary for many processes such as: transmembrane ion flux, neuronal activity, neurotransmitter release, regulation of adenylate cyclase and hormone receptor binding. In the central nervous system magnesium blocks the activity of NMDA receptors and magnesium-deficiency leads to NMDA receptors overactivity, possibly causing neuronal injury and neurological dysfunction, which consequently may manifest as major depression. Numerous pre-clinical and clinical studies confirmed that magnesium influences several systems associated with development of depression. On the other hand, standard antidepressants are ineffective in 40% of cases, and have some unpleasant side-effects. Even though the first positive effect of magnesium in patients with agitated depression was reported in 1921, unfortunately, only small number of studies that favor magnesium's effectiveness in treating depression was published in past decades, including one randomized clinical trial in which magnesium was as effective as imipramine in depressed elderly patients that suffered from diabetes. Even though much more clinical research is needed to extend this important line of research and to confirm its efficacy, due to its safety magnesium preparations could be considered as a potential addition for treatment resistant depression cases.
AB  - Magnezijum ima značajnu fiziološku ulogu u humanom organizmu i neophodan je za mnoge procese, uključujući: transmembransko kretanje jona, aktivnost neurona, oslobađanje neurotransmitera, regulaciju adenilat ciklaze i vezivanje pojedinih hormona za receptore. U centralnom nervnom sistemu magnezijum blokira aktivnost NMDA receptora, a deficit magnezijuma dovodi do prekomerne aktivacije NMDA receptora, što za posledicu može imati oštećenja neurona i neurološke disfunkcije i u krajnjoj liniji, može se manifestovati razvojem velike depresije. Brojna pretklinička i klinička ispitivanja potvrđuju da magnezijum utiče na nekoliko sistema uključenih u razvoj depresije. Sa druge strane, oko 40% pacijenata pokazuje rezistenciju na tretman standardnim antidepresivima, kojima se pripisuju i brojni neprijatni neželjeni efekti. Iako je prvi pozitivan učinak magnezijuma kod pacijenata koji pate od agitirane depresije prikazan još 1921, nažalost, samo mali broj studija koje govore u prilog efikasnosti magnezijuma u tretmanu depresije je publikovan tokom proteklih decenija, uključujući jedno randomizovano kliničko ispitivanje koje je pokazalo podjednaku efikasnost magnezijuma i imipramina kod starijih depresivnih pacijenata koji boluju od dijabetesa. Iako je neophodno sprovesti veći broj kliničkih ispitivanja i proširiti ovo polje istraživanja u cilju potvrđivanja njegove efikasnosti, usled svoje bezbednosti, magnezijum bi se potencijalno mogao uvesti kao dodatni tretman u lečenju depresije rezistentne na terapiju.

AB  - In this paper we described the homeostasis of elementary iron: its absorption, utilization, storage and elimination. Special attention was paid to new insights into the regulatory mechanisams involved in its homestasis, especially the role of recently discovered hormone hepcidin, which is synthesized in the liver. Since it is reckoned that hepcidin has a central role in the systemic regulation of iron metabolism, the influences of various factors and conditions on the hepatic synthesis of hepcidin were delineated. Furthermore, we briefly described the most common disorders of iron homeostasis and their possible involvement in the pathogenesis of some diseases.
AB  - U radu je opisan promet gvožđa u organizmu: unošenje, iskorišćavanje, deponovanje i eliminacija. Posebna pažnja posvećena je novim saznanjima o regulaciji prometa tog elementa neophodnog za život. U tom smislu je istaknut, relativno skoro otkriven, hormon hepcidin koji sintetišu hepatociti. Smatra se da hepcidin zauzima centralno mesto u sistemskoj regulaciji metabolizma gvožđa i prikazan je uticaj različitih faktora i stanja na sintezu hepcidina u jetri. Ukratko su opisani i najčešći poremećaji homeostaze gvožđa i njegova eventualna uloga u patogenezi nekih oboljenja.

AB  - DNA damage is a powerful trigger of apoptosis in neurons and is present in patients with Alzheimer's disease (AB). Also, DNA damage is registered in cells that are not neurons, such as peripheral blood leukocytes and fibroblasts. In this paper we examined and compared the impact of AB and chronological aging on DNA damage in peripheral blood leukocytes. We used electrophoresis of individual cells on gel (comet test) as a modern and reliable method for registration of nuclear DNA damage whose fragments form a comet tail, which is not present in undamaged cells. We analyzed leukocytes of these groups: AB patients with sporadic form of the disease, healthy elderly of an appropriate age and healthy younger adults as controls. The results show that the frequency of AB patients with leukocyte DNA damage significantly is higher than in similar control age-matched controls. Also, a significant increase in the frequency of DNA damage has been observed during chronological aging. Based on the obtained data we can conclude that genetic instability, otherwise present in the elderly is more expressed in patients with sporadic AB, which indicates that it is not just an epiphenomenon of aging, but is characteristic of the disease.
AB  - Oštećenja DNK su snažan okidač apoptoze neurona i prisutna su kod obolelih od Alchajmerove bolesti (AB). Ona se registruju i u ćelijama koje nisu neuroni, kao što su leukociti periferne krvi i fibroblasti. U ovom radu smo ispitivali i poredili uticaj AB i hronološkog starenja na oštećenje DNK leukocita periferne krvi. Koristili smo elektroforezu pojedinačnih ćelija na gelu (Komet test), kao savremenu i pouzdanu metodu za registrovanje oštećenja jedarne DNK čiji fragmenti formiraju rep komete, koji nije prisutan u neoštećenim ćelijama. Analizarani su leukociti: AB pacijenata sa sporadičnim oblikom bolesti, zdravih starih osoba odgovarajuće starosti i zdravih odraslih mlađih kontrolnih osoba. Dobijeni rezultati pokazuju da je kod AB ispitanika frekvencija leukocita sa oštećenjima DNK statistički značajno veća nego kod kontrola slične starosti; značajan porast učestalosti oštećenja DNK zapaža se i tokom hronološkog starenja. Na osnovu dobijenih podataka možemo da zaključimo da je genetička nestabilnost, inače prisutna i kod starih osoba, jače izražena kod pacijenata sa sporadičnom AB, što ukazuje na to da ona nije samo epifenomen starenja, već je karakteristika same bolesti.

AB  - Aging is inevitable, and as the percentage of elderly in the population increases it becomes more important to understand physiological mechanisms associated with the normal aging process so the quality of life can be sustained. Maintaining physiological functions or 'health' in aging population will help to reduce the burden on the medical systems. In this work, we briefly consider, in the light of up-to-date scientific data and hypotheses, some key questions on the biology and physiology of aging: what is it and why does it happen. Prospects for delaying and impeding the aging process are disputed as well.
AB  - Starenje je neizbežno i kako zastupljenost starih osoba u populaciji raste, razumevanje fizioloških mehanizama uključenih u proces normalnog starenja ima sve veći značaj u cilju održavanja kvaliteta života u starosti. Takođe, očuvanje fizioloških funkcija ili 'zdravlja' starih ljudi smanjiće pritisak na zdravstvene sisteme i troškove. U ovom radu, u svetlu najnovijih naučnih podataka i hipoteza, sažeto se razmatraju neka ključna pitanja u vezi biologije i fiziologije starenja: šta je starenje i zašto se ono događa, kao i mogućnosti odlaganja i usporavanja starenja.

Differential effects of chronic propranolol treatment on the phenotypic profile of thymocytes from immature and adult rats

AB  - To elucidate a putative role of beta-adrenoceptors in the modulation of intrathymic T-cell maturation, the expression of major differentiational antigene (CD4/CD8 and TCR alpha beta) on the thymocytes from both immature (aged 21 day at the beginning of the treatment) and adult (a,oed 75 days at the beginning of treatment) male rats subjected to a 15-day-long propranolol treatment (0.40 mg/100 g/day, s.c.) was analyzed by two- and one-color flow cytometry, respectively. Rats of matched age injected with saline served as controls. The propranolol treatment in immature but not adult rats caused a significant reduction in both the relative thymus weight and total thymocyte yield. In addition, a significant increase in the percentage of CD4(+)8(+) double-positive cells, with a proportional decrease in the relative proportion of CD4(+)8(-) single positive cells, was found in immature rats. In contrast, a slight but significant decrease in the percentage of CD4(+)8(+) cells with a parallel increase in the relative proportion of CD4+8- cells was found in adult rats. In both groups of rats, the percentage of TCR alpha beta(total) thymocytes was increased: in immature rats this was due to an increase in the percentage of TCR alpha beta(low) thymocytes, while in the adult rats it reflected a rise in the relative proportion of TCR alpha beta(high) cells. In conclusion, the study revealed that propranolol treatment in both immature and adult rats alters the relative proportion of CD4(+)8(+) and CD4(+)8(-) thymocytes, but in opposite fashion, and the data suggest that this treatment affects distinct fractions within the population of CD4(+)8(+) thymocytes with respect to expression of TCR alpha beta. The results also indicate that, regardless of rat sexual maturity, the development of thymocytes towards CD4(-)8(+) T-cells is relatively insensitive to long-lasting P-adrenoceptor blockade.
