AB  - Background: The pre-analytical (PA) phase is the most vulnerable phase of laboratory testing procedure, with critical procedures-collection, handling, sample transport, and time and temperature of sample storage. The aim of this study was to examine if different anticoagulants, storage conditions, and freeze-thaw cycles (FTCs) influence the concentrations of basic biochemical parameters. In parallel, the presence and the effect of sample microbiological contamination during routine laboratory work were examined.

Methods: Two plasma pools (EDTA, and sodium-fluoride/potassium oxalate plasma (NaF)) were stored at +4C˚/-20˚C. Total cholesterol (TC), glucose, triglycerides (TG), urea, total protein (TP), and albumin concentrations were measured using Ilab 300+. Sample microbiological contamination was determined by 16S rRNA sequence analysis. The experiment encompassed a 5 day-period: Day 1–fresh sample, Day 2–1st FTC, Day 3–2nd FTC, Day 4–3rd FTC, Day 5–4th FTC. The appearance of bacteria in two consecutive samples was the experiment's endpoint.

Results: During 4 FTCs there were no changes in plasma urea concentrations. Glucose was stable in EDTA+4˚C and NaF- 20˚C until the 3rd FTC (P=0.008, P=0.042, respectively). Changes in protein concentrations followed the zig-zag pattern. TG concentrations changed significantly in the EDTA-20˚C sample after 1st and 4th FTCs (P=0.022, P=0.010, respectively). In NaF samples no contamination was observed during 4 FTCs.

 

Conclusions: Urea and glucose concentrations were robust. Changes in lipid and protein concentrations after FTCs follow complex patterns. Bacterial growth was not observed in NaF plasma samples. This can promote NaF use in analytical procedures in which microbiological contamination affects the quality of analysis.

prikupljanje, rukovanje, transport i skladi{tenje uzoraka, i
predstavlja najzna~ajniji izvor laboratorijskih gre{aka. Cilj
ovog istra`ivanja je bio da se ispita stabilnost osnovnih
biohemijskih parametara u zavisnosti od uslova skladi{tenja
uzoraka i broja ciklusa zamrzavanja-odmrzavanja (FTC).
Pored toga, ispitivano je prisustvo bakterijske kontaminacije
uzoraka tokom rutinskog laboratorijskog rada.
Metode: Dva »pool«-a plazme (etilendiaminotetrasir}etna
kiselina (EDTA) i natrijum-fluorid/kalijum oksalat (NaF)) su
skladi{tena na +4 ˚C/-20 ˚C. Koncentracije ukupnog
holesterola (TC), glukoze, triglicerida (TG), uree i albumina
su odre|ene kori{}enjem BioSystems reagenasa (holesterol

nosno bromkrezol zeleno metodama, sukcesivno) na Ilab
300+ analizatoru. Bakterijska kontaminacija uzoraka je
potvr|ena 16S rRNA sekvencioniranjem. Eksperiment je
sproveden tokom 5 uzastopnih dana: 1. dan – sve` uzorak,
2. dan – 1. FTC, 3. dan –2. FTC, 4. dan – 3. FTC, 5. dan
– 4. FTC. Zavr{nu ta~ku eksperimenta predstavljala je
pojava bakterija u dva uzastopna uzorka.
Rezultati: Tokom 4 FTC koncentracije uree u plazmi se nisu
zna~ajno razlikovale. Koncentracija glukoze je bila stabilna
u EDTA +4 ˚C i NaF -20 ˚C do 3.FTC (P=0,008,
P=0,042, redom). Koncentracije TG su se zna~ajno pro-
menile u uzorku EDTA -20 ˚C nakon 1. i 4. FTC-a (P=0,022, P=0,010, redom). U uzorcima NaF plazme
nije do{lo do bakterijske kontaminacije tokom 4. FTC.
Zaklju~ak: Koncentracije uree i glukoze su bile stabilne
tokom trajanja eksperimenta. Promene u koncentracijama
lipida nakon FTC prate slo`ene obrasce. Rast bakterija nije
prime}en u uzorcima NaF plazme, te upotreba ovog anti-
koagulansa mo`e biti opravdana u analiti~kim proce -
durama podlo`nim uticaju mikrobiolo{ke kontaminacije.

Cholesterol Metabolic Profiling of HDL in Women with Late-Onset Preeclampsia

AB  - A specific feature of dyslipidemia in pregnancy is increased high-density lipoprotein (HDL) cholesterol concentration, which is probably associated with maternal endothelium protection. However, preeclampsia is most often associated with low HDL cholesterol, and the mechanisms behind this change are scarcely explored. We aimed to investigate changes in HDL metabolism in risky pregnancies and those complicated by late-onset preeclampsia. We analyze cholesterol synthesis (cholesterol precursors: desmosterol, 7-dehydrocholesterol, and lathosterol) and absorption markers (phytosterols: campesterol and β-sitosterol) within HDL particles (NCSHDL), the activities of principal modulators of HDL cholesterol’s content, and major HDL functional proteins levels in mid and late pregnancy. On the basis of the pregnancy outcome, participants were classified into the risk group (RG) (70 women) and the preeclampsia group (PG) (20 women). HDL cholesterol was lower in PG in the second trimester compared to RG (p < 0.05) and followed by lower levels of cholesterol absorption markers (p < 0.001 for campesterolHDL and p < 0.05 for β-sitosterolHDL). Lowering of HDL cholesterol between trimesters in RG (p < 0.05) was accompanied by a decrease in HDL phytosterol content (p < 0.001), apolipoprotein A-I (apoA-I) concentration (p < 0.05), and paraoxonase 1 (PON1) (p < 0.001), lecithin–cholesterol acyltransferase (LCAT) (p < 0.05), and cholesterol ester transfer protein (CETP) activities (p < 0.05). These longitudinal changes were absent in PG. Development of late-onset preeclampsia is preceded by the appearance of lower HDL cholesterol and NCSHDL in the second trimester. We propose that reduced capacity for intestinal HDL synthesis, decreased LCAT activity, and impaired capacity for HDL-mediated cholesterol efflux could be the contributing mechanisms resulting in lower HDL cholesterol.

AB  - Introduction: Telomeres are protective chromosomal ends. Short telomeres are a proven biomarker of biological aging. We aimed to find an association of telomere length and telomerase activity in circulating leukocytes and thromboaspirates of patients with acute myocardial infarction. Furthermore, association of the telomere-telomerase system with oxidative stress markers (as common risk factors for coronary artery disease (CAD)) was tested. Material and methods: Patients were selected from the patients admitted to the intensive care unit with acute myocardial infarction with ST-segment elevation (STEMI), with the following inclusion criteria – STEMI patients between 18 and 80 years old of both genders and candidates for primary percutaneous coronary intervention, with infarction pain present for a maximum of 12 h. In all the patients leukocyte telomere length, telomerase activity and scores related to oxidative-stress status (Protective, Damage and OXY) were evaluated. Results: Patients were divided into different groups: with stable angina pectoris (AP) (n = 22), acute myocardial infarction with: STEMI (n = 93), non-obstructive coronary arteries (MINOCA) (n = 7), blood vessel rupture (n = 6) at three time points, and compared to the group of 84 healthy subjects. Telomerase activity was significantly higher in all CAD sub-groups compared to the control group (AP = 0.373 (0.355–0.386), STEMI = 0.375 (0.349–0.395), MINOCA = 0.391 (0.366–0.401), blood vessel rupture = 0.360 (0.352–0.385) vs. CG = 0.069 (0.061–0.081), p < 0.001), while telomeres were significantly shorter in STEMI, MINOCA and blood vessel rupture groups compared to the control group (STEMI = 1.179 (0.931–1.376), MINOCA = 1.026 (0.951–1.070), blood vessel rupture = 1.089 (0.842–1.173) vs. CG = 1.329 (1.096–1.624), p = 0.030]. Values of OXY score were significantly higher in STEMI and MINOCA patients compared to the control group and AP patients (5.83 (4.55–7.54) and 10.28 (9.19–10.72) vs. 4.94 (3.29–6.18) and 4.18 (2.58–4.86), p < 0.001). Longer telomeres and higher telomerase activity were found in thromboaspirates, compared to the peripheral blood leukocytes in the same patients (1.25 (1.01–1.84) vs. 1.18 (0.909–1.516), p = 0.036; and 0.366 (0.367–0.379) vs. 0.366 (0.367–0.379), p < 0.001, respectively). In addition, telomere length and telomerase activity had good diagnostic ability to separate STEMI patients from healthy persons. Conclusions: Leukocyte telomere length and telomerase activity can differentiate CAD patients from healthy persons, and relate CAD to oxidative stress.

abstract = "Introduction: Telomeres are protective chromosomal ends. Short telomeres are a proven biomarker of biological aging. We aimed to find an association of telomere length and telomerase activity in circulating leukocytes and thromboaspirates of patients with acute myocardial infarction. Furthermore, association of the telomere-telomerase system with oxidative stress markers (as common risk factors for coronary artery disease (CAD)) was tested. Material and methods: Patients were selected from the patients admitted to the intensive care unit with acute myocardial infarction with ST-segment elevation (STEMI), with the following inclusion criteria – STEMI patients between 18 and 80 years old of both genders and candidates for primary percutaneous coronary intervention, with infarction pain present for a maximum of 12 h. In all the patients leukocyte telomere length, telomerase activity and scores related to oxidative-stress status (Protective, Damage and OXY) were evaluated. Results: Patients were divided into different groups: with stable angina pectoris (AP) (n = 22), acute myocardial infarction with: STEMI (n = 93), non-obstructive coronary arteries (MINOCA) (n = 7), blood vessel rupture (n = 6) at three time points, and compared to the group of 84 healthy subjects. Telomerase activity was significantly higher in all CAD sub-groups compared to the control group (AP = 0.373 (0.355–0.386), STEMI = 0.375 (0.349–0.395), MINOCA = 0.391 (0.366–0.401), blood vessel rupture = 0.360 (0.352–0.385) vs. CG = 0.069 (0.061–0.081), p < 0.001), while telomeres were significantly shorter in STEMI, MINOCA and blood vessel rupture groups compared to the control group (STEMI = 1.179 (0.931–1.376), MINOCA = 1.026 (0.951–1.070), blood vessel rupture = 1.089 (0.842–1.173) vs. CG = 1.329 (1.096–1.624), p = 0.030]. Values of OXY score were significantly higher in STEMI and MINOCA patients compared to the control group and AP patients (5.83 (4.55–7.54) and 10.28 (9.19–10.72) vs. 4.94 (3.29–6.18) and 4.18 (2.58–4.86), p < 0.001). Upregulation of peripheral blood mononuclear cells resistin gene expression in severe obstructive sleep apnea and obstructive sleep apnea with coexisting type 2 diabetes mellitus

AB  - Purpose: Obstructive sleep apnea (OSA) is characterised by increased systemic inflammation, and is often accompanied with type 2 diabetes mellitus (T2DM) and cardiovascular disease. The aim of this investigation was to evaluate gene expression of resistin, its receptor CAP1 and CD36 as the indicators of the inflammatory changes in PBMCs in relation to the severity of OSA, and the presence of type 2 diabetes mellitus (T2DM) in OSA. Methods: Severity of OSA was defined by the apnea/hypopnea index (AHI): AHI < 30: mild to moderate OSA (MM-OSA), AHI ≥ 30: severe OSA (S-OSA). Presence of T2DM was captured: OSA with T2DM (OSA + T2DM), OSA without T2DM (OSA-T2DM). PBMC resistin, CAP1, and CD36 mRNA were determined by real-time PCR. Results: Resistin mRNA was significantly upregulated in S-OSA (N = 54) compared to the MM-OSA (N = 52, P = 0.043); CAP1 and CD36 mRNA levels did not differ between the groups (P = 0.302; P = 0.166, respectively). Resistin mRNA was significantly upregulated in OSA + T2DM (N = 29) compared to the OSA-T2DM (N = 77, P = 0.029); CAP1 and CD36 mRNA levels did not differ between the groups (P = 0.662; P = 0.108, respectively). AHI and T2DM were independent predictors of resistin mRNA above the 75th percentile (OR = 3.717 [1.152–11.991]; OR = 3.261 [1.000–10.630], P = 0.042 respectively). Conclusion: Resistin gene upregulation in S-OSA indicates its possible contribution to increased inflammation in S-OSA and makes it a possible marker of the disease severity. Resistin gene upregulation in OSA + T2DM suggests that a joint effect of these two comorbidities may have a major contribution to increased inflammation and complications that arise from this state.

AB  - Preventivne mere, rano otkrivanje i potvrda novih slučajeva infekcije, predstavljaju osnov u
sprečavanju širenja i suzbijanju ininfektivne bolesti COVID-19. Primena visoko osetljive, spe-
cifične real-time RT-PCR metode predstavlja zlatni standard u detekciji SARS-CoV-2. Brza i
pouzdana dijanoza su neophodni za efikasno praćenje bolesti, ali veliki broj lažno negativnih
slučajeva omogućio je nekontrolisanu transmisiju infekcije.
Nove metode za detekciju SARS-CoV-2 iz nazofaringealnog brisa zasnovane na principu
tečne hromatografije sa masenom spektrometrijom (HPLC-MS/MS) isključivo se koriste u
istraživačke svrhe. Hromatografski testovi omogućavaju istovremenu detekciju više različitih,
specifičnih peptidnih markera za identifikaciju SARS-CoV-2. Na ovaj način, moguće mutacije
u genskoj sekvenci virusa, lako mogu biti prevaziđene. Upotreba gasne hromatografije sa
spektrometrijom pokretljivosti jona (GC-IMS) za detekciju odabranih molekula u izdahnutom
vazduhu pacijenata sa COVID-19 može omogućiti neinvazivnu, brzu i tačnu, „point of care”
potvrdu dijagnoze bolesti.
Uprkos superiornim analitičkim performansama hromatografskih tehnika, njihova primena
u rutinskoj laboratorijskoj praksi je retka. Pored opreme, njihova primena zahteva obučeno
osoblje i „in house” procedure validacije i verifikacije metoda. Protokoli validacije hromato-
grafskih metoda se oslanjaju na preporuke date u naučnim publikacijama i različitim smer-
nicama, te su istraživački orijentisani. Najčešće korišćene su EMA, FDA i CLSI smernice za
postupke i procedure validacije metoda. Međutim, ove smernice dozvoljavaju različita tuma-
čenja i ostavljaju analitičaru da odluči koji od parametara validacije su neophodni. Različiti
preanalitički i analitički aspekti hromatografskih metoda diktiraju složenost kriterijuma vali-
dacije. Zbog toga je neophodno izdvojiti najvažnije postupke validacije hromatografskih teh-
nika (određivanje linearnosti, LOQ i LOD vrednosti, tačnosti i preciznosti metode) i primeniti
dostupnu opremu i naučna saznanja.

AB  - In order to understand the metabolic and immune potential of adiponectin in
colorectal cancer (CRC), attention is drawn to its receptors: adiponectin receptor 1
(ADIPOR1) and adiponectin receptor 2 (ADIPOR2). Gene set enrichment analysis (GSEA) of
datasets based on malignant tissue samples and peripheral blood monocytes (PBMs) was
used to explore mRNA fingerprints of different signaling pathways best associated with
ADIPOR1/ADIPOR2 gene expression levels. Transcriptomic datasets GSE44076 (1) and
GSE47756 (2) were downloaded from the NCBI Gene Expression Omnibus database. In the
GSE44076 dataset, three groups were formed: 98 colon tumor tissue and matched tumor-
adjacent mucosa samples and tissue samples from 50 healthy volunteers. The other set
(GSE47756) contained information on PBMs' gene signature in CRC, by forming two groups:
38 samples from healthy subjects and 55 CRC patients. GSEA analysis of the GSE44076
dataset implied that ADIPOR1 mRNA levels were in negative association with MTORC1 and
TNF-α NF-κB signaling pathways in tumor tissue. At the same time, ADIPOR2 was positively
associated with metabolic gene sets such as cholesterol homeostasis, glycolysis and PPAR
signaling. Quite opposite to the GSE44076 dataset, GSEA analysisis of GSE47756 revealed
that ADIPOR1 was a metabolically active receptor in PBMs of CRC patients. Surprisingly, the
TNF-α NF-κB signaling pathway gene sets were positively associated with ADIPOR1 mRNA
levels in monocytes of the cancer group. Different types of metabolic and immune regulation
achieved through ADIPOR1/ADIPOR2 in tumor and PBMs suggest possible novel therapeutic
targets in CRC.
AB  - Da bi se razumeo metabolički i imuni potencijal adiponektina u kolorektalnom
karcinomu (CRC), pažnja se mora obratiti i na njegove receptore: adiponektinski receptor 1
(ADIPOR1) i adiponektinski receptor 2 (ADIPOR2). Gene set enrichment analiza (GSEA) dva
skupa podataka, zasnovanih na uzorcima malignog tkiva i na monocitima periferne krvi
(PBM) je korišćena da bi se obezbedili genetski otisci različitih signalnih puteva koji su bili
najbolje povezani sa ekspresijom gena ADIPOR1/ADIPOR2. Skupovi transkriptomskih
podataka GSE44076 (1) i GSE47756 (2) su preuzeti iz NCBI Gene Expression Omnibus baze
podataka. U skupu podataka GSE44076 formirane su tri grupe: 98 uparenih uzoraka
tumorskog tkiva debelog creva i susedne mukoze i uzorci tkiva 50 zdravih dobrovoljaca.
Drugi set (GSE47756) je sadržao informacije o otisku gena PBM u CRC sa formiranim dvema
grupama: 38 uzoraka zdravih subjekata i 55 pacijenata sa CRC. GSEA analiza skupa podataka
GSE44076 je implicira da su nivoi iRNK ADIPOR1 negativno korelirali sa MTORC1 i TNF-α

(GSE47756) contained information on PBMs' gene signature in CRC, by forming two groups:
38 samples from healthy subjects and 55 CRC patients. GSEA analysis of the GSE44076
dataset implied that ADIPOR1 mRNA levels were in negative association with MTORC1 and
TNF-α NF-κB signaling pathways in tumor tissue. At the same time, ADIPOR2 was positively
associated with metabolic gene sets such as cholesterol homeostasis, glycolysis and PPAR
signaling. Quite opposite to the GSE44076 dataset, GSEA analysisis of GSE47756 revealed
that ADIPOR1 was a metabolically active receptor in PBMs of CRC patients. Surprisingly, the
TNF-α NF-κB signaling pathway gene sets were positively associated with ADIPOR1 mRNA
levels in monocytes of the cancer group. Different types of metabolic and immune regulation
achieved through ADIPOR1/ADIPOR2 in tumor and PBMs suggest possible novel therapeutic
targets in CRC., Da bi se razumeo metabolički i imuni potencijal adiponektina u kolorektalnom
karcinomu (CRC), pažnja se mora obratiti i na njegove receptore: adiponektinski receptor 1
(ADIPOR1) i adiponektinski receptor 2 (ADIPOR2). Gene set enrichment analiza (GSEA) dva
skupa podataka, zasnovanih na uzorcima malignog tkiva i na monocitima periferne krvi
(PBM) je korišćena da bi se obezbedili genetski otisci različitih signalnih puteva koji su bili
najbolje povezani sa ekspresijom gena ADIPOR1/ADIPOR2. Skupovi transkriptomskih
podataka GSE44076 (1) i GSE47756 (2) su preuzeti iz NCBI Gene Expression Omnibus baze
podataka. U skupu podataka GSE44076 formirane su tri grupe: 98 uparenih uzoraka

AB  - Adiponectin (ADIPOQ) as both a regulator of metabolic homeostasis and a protein involved in immune response might be of particular interest to contemporary laboratory medicine, especially in terms of minimally invasive diagnostics. The diverse roles of ADIPOQ with regard to the immune and metabolic aspects of colorectal carcinogenesis have been proposed. However, the expression of its receptors ADIPOR1 and ADIPOR2 is scarcely explored in peripheral blood mononuclear cells (PBMCs). Moreover, ADIPORs’ relationships with the immune response mediator TNF-α have not been previously investigated in the PBMCs of CRC patients. This study used both in silico and observational case–control analyses with the aim of exploring the association of ADIPOR gene expression and ADIPOQ single nucleotide polymorphisms (SNPs) with the inflammatory marker TNF-α and lipid status parameters in patients with CRC. Publicly available transcriptomic datasets (GSE47756, GSE44076) obtained from analyses of monocytes and CRC tissue samples were employed for the in silico evaluation of ADIPORs’ specific genetic traits. GSE47756 and GSE44076 datasets were processed with GSEA software to provide a genetic fingertip of different signaling pathways associated with ADIPORs’ mRNA levels. The case–control aspect of the study included the PBMC samples of 73 patients diagnosed with CRC and 80 healthy volunteers. The PCR method was carried out for the PBMC gene expression analysis (ADIPOR1, ADIPOR2, TNF-α mRNA levels) and for the subjects’ genotyping (ADIPOQ rs266729, ADIPOR1 rs7539542). GSEA showed significant associations of ADIPOR mRNA expression with gene sets related to metabolic and immune homeostasis in both datasets. The case–control study revealed the association of ADIPOR1 rs7539542 with reduced lipid status parameters in CRC. In addition, PBMC ADIPOR1 mRNA levels decreased in CRC (p < 0.001), whereas ADIPOR2 mRNA did not differ between the groups (p = 0.442). A reduction in PBMC TNF-α mRNA levels was noted in CRC (p < 0.05). Our results indicate that ADIPOR1 and ADIPOR2 play a significant role in the alteration of both metabolic and immune homeostasis during the progression of CRC. For the first time, ADIPOR1 is shown to be a specific receptor for mediating ADIPOQ’s effects in the PBMCs of CRC patients.

Does Pentraxin-3 contribute to the reduction of low-density lipoprotein levels by statin therapy?

AB  - Statins have been shown to decrease inflammatory markers, especially high sensitivity C reactive protein (hsCRP), in a dose-dependent manner. Pentraxin-3 (PTX3) is another important inflammatory biomarker from the pentraxin family that provides useful prognostic information and facilitates diagnostics of cardiovascular diseases. This study investigated the effect of statin therapy on PTX3 and hsCRP concentrations and whether statins acted synergistically with PTX3 and hsCRP concentrations in lowering LDL-C. The study group consisted of 90 patients undergoing coronary angiography. The results showed that statins reduced PTX3 concentrations (p=0.031). PTX3 and hsCRP levels were significantly different between subclinical and severe stenosis groups (p=0.011 and p=0.009, respectively). Statin therapy was significantly associated with lower PTX3 and LDL-C levels in multiple logistic analyses. The probability that statin therapy would achieve target LDL-C values was highest in patients with low PTX3 values (OR=3.683, p=0.040), while multiplicative interaction was 23.3. The effect of statins on PTX3 reduction was higher than on hsCRP. It can be suggested that statin therapy was more successful in patients with low PTX3 values.
AB  - Pokazano je da statini snižavaju koncentracije inflamatornih markera, posebno visoko osetljivog C reaktivnog proteina (hsCRP), pri čemu je sniženje zavisno od doze leka. Pentraksin- 3 (PTX3) je još jedan važan inflamatorni biomarker iz porodice pentraksina koji ima prognostičke karakteristike i olakšava dijagnozu kardiovaskularnih bolesti. U ovoj studiji je ispitivan efekat terapije statinima na koncentracije PTX3 i hsCRP, kao i sinergistički efekat terapije, koncentracija PTX3 i hsCRP u snižavanju LDL-C. U studiju je uključeno 90 pacijenata kojima je koronarnom angiografijom procenjeno suženje koronarnih krvnih sudova. Rezultati su pokazali da statini smanjuju koncentraciju PTX3 (p=0,031). Koncentracije PTX3 i hsCRP značajno se razlikuju između grupa sa subkliničkim (p=0,011) i teškim oblikom stenoze (p=0,009). Primenom multiple logističke regresione analize uočena je veza između terapije statinima i niskih koncentracija PTX3 i LDL-C. abstract = "Statins have been shown to decrease inflammatory markers, especially high sensitivity C reactive protein (hsCRP), in a dose-dependent manner. Pentraxin-3 (PTX3) is another important inflammatory biomarker from the pentraxin family that provides useful prognostic information and facilitates diagnostics of cardiovascular diseases. This study investigated the effect of statin therapy on PTX3 and hsCRP concentrations and whether statins acted synergistically with PTX3 and hsCRP concentrations in lowering LDL-C. The study group consisted of 90 patients undergoing coronary angiography. The results showed that statins reduced PTX3 concentrations (p=0.031). PTX3 and hsCRP levels were significantly different between subclinical and severe stenosis groups (p=0.011 and p=0.009, respectively). Statin therapy was significantly associated with lower PTX3 and LDL-C levels in multiple logistic analyses. The probability that statin therapy would achieve target LDL-C values was highest in patients with low PTX3 values (OR=3.683, p=0.040), while multiplicative interaction was 23.3. The effect of statins on PTX3 reduction was higher than on hsCRP. It can be suggested that statin therapy was more successful in patients with low PTX3 values., Pokazano je da statini snižavaju koncentracije inflamatornih markera, posebno visoko osetljivog C reaktivnog proteina (hsCRP), pri čemu je sniženje zavisno od doze leka. Pentraksin- 3 (PTX3) je još jedan važan inflamatorni biomarker iz porodice pentraksina koji ima prognostičke karakteristike i olakšava dijagnozu kardiovaskularnih bolesti. U ovoj studiji je ispitivan efekat terapije statinima na koncentracije PTX3 i hsCRP, kao i sinergistički efekat terapije, koncentracija PTX3 i hsCRP u snižavanju LDL-C. U studiju je uključeno 90 pacijenata kojima je koronarnom angiografijom procenjeno suženje koronarnih krvnih sudova. Rezultati su pokazali da statini smanjuju koncentraciju PTX3 (p=0,031). Koncentracije PTX3 i hsCRP značajno se razlikuju između grupa sa subkliničkim (p=0,011) i teškim oblikom stenoze (p=0,009). Primenom multiple logističke regresione analize uočena je veza između terapije statinima i niskih koncentracija PTX3 i LDL-C. AB  - Objectives: Obstructive sleep apnea (OSA) is a common condition closely related to obesity, insulin resistance, dyslipidemia, and cardiovascular disease. The aim of this study was to explore the possible relationship between OSA and proprotein convertase subtilisin/kexin type 9 (PCSK9). Methods: Full-night polysomnography was performed on 150 participants who were divided into three groups: controls, OSA patients on statin therapy, and OSA patients not on statin therapy. Biochemical markers, plasma low-density lipoprotein (LDL) and high-density lipoprotein (HDL) subclasses, and PCSK9 were determined. Results: PCSK9 was highest in OSA patients on statins compared to the control group and to OSA patients not on statins (p = 0.036 and p = 0.039, respectively), after adjustment for body mass index (BMI). LDL diameter was greater in OSA patients not on statins compared to OSA patients on statins (p = 0.032). PCSK9 was highest in the group of patients with all three risk factors (diagnosed OSA, statins, BMI ≥25 kg/m2) compared to groups with no, one, and two risk factors (p = 0.031, p = 0.001, and p = 0.029, respectively). Presence of OSA, statin therapy, and BMI ≥25 kg/m2 when combined were independently associated with higher levels of PCSK9 when adjusted for antihypertensive therapy, small dense LDL, and HDL 3c subclass (odds ratio = 2.849; interquartile range [1.026–7.912], p = 0.044). Conclusion: Statin therapy was closely related to PCSK9. OSA along with obesity and statin use induces elevation of PCSK9.

AB  - Background and aims: Obstructive sleep apnea (OSA) is closely linked to obesity and related adverse metabolic changes, including dyslipidemia. However, it is not clear whether OSA is an independent contributing factor to dyslipidemia, or the observed association is a reflection of a concomitant presence of obesity. Additionally, dyslipidemia is usually evaluated through measurement of parameters of routine lipid status, while more precise evaluation of lipid homeostasis is rarely performed in OSA. In this study, we analyzed markers of cholesterol synthesis and absorption in patients with OSA with respect to the presence of obesity and the disease severity. Methods and results: This study enrolled 116 OSA patients. Concentrations of non-cholesterol sterols (NCS), measured by LC-MS/MS, were used as markers of cholesterol synthesis and absorption. Apnea-hypopnea index (AHI) and oxygen saturation (SaO2) were utilized as markers of OSA severity. Serum lipid status parameters were determined by routine enzymatic methods. Markers of cholesterol synthesis were increased (P = 0.005), whilst markers of cholesterol absorption decreased (P = 0.001) in obese OSA patients. Cholesterol synthesis/absorption ratio was elevated in obese subjects (P < 0.001). Concentration of cholesterol synthesis marker lathosterol was significantly higher in subjects with severe OSA (P = 0.014) and we observed a trend of decreased cholesterol absorption in these patients. AHI was revealed as an independent determinant of lathosterol concentration (P = 0.022). Conclusions: Our results suggest that the presence of obesity and severe forms of OSA is characterized by elevated endogenous cholesterol synthesis. AHI was singled out as an independent determinant of the serum level of cholesterol synthesis marker lathosterol.

AB  - Metabolic disorders in pregnancy, particularly gestational diabetes mellitus (GDM), are associated with an increased risk for adverse pregnancy outcome and long-term cardiometabolic health of mother and child. This study analyzed changes of serum cholesterol synthesis and absorption markers during the course of high-risk pregnancies, with respect to the development of GDM. Possible associations of maternal lipid biomarkers with neonatal characteristics were also investigated. The study included 63 women with high risk for development of pregnancy complications. Size and proportions of small low-density (LDL) and high-density lipoprotein (HDL) particles were assessed across trimesters (T1–T3), as well as concentrations of cholesterol synthesis (lathosterol, desmosterol) and absorption markers (campesterol, β-sitosterol). During the study, 15 women developed GDM, while 48 had no complications (non-GDM). As compared to the non-GDM group, women with GDM had significantly higher triglycerides in each trimester, while having a lower HDL-C level in T3. In addition, they had significantly lower levels of β-sitosterol in T3 (p < 0.05). Cholesterol synthesis markers increased across trimesters in both groups. A decrease in serum β-sitosterol levels during the course of pregnancies affected by GDM was observed. The prevalence of small-sized HDL decreased in non-GDM, while in the GDM group remained unchanged across trimesters. Newborn’s size in the non-GDM group was significantly higher (p < 0.01) and inversely associated with proportions of both small, dense LDL and HDL particles (p < 0.05) in maternal plasma in T1. In conclusion, high-risk pregnancies affected by GDM are characterized by altered cholesterol absorption and HDL maturation. Advanced lipid testing may indicate disturbed lipid homeostasis in GDM.

We hypothesized that lipoprotein particle distributions moved toward less atherogenic profile and that cholesterol homeostasis was achieved. Materials and methods  Thirty-three children, 2 to 9 years old with NS were recruited. Blood sampling took place both in the acute phase and during remission. Serum low-density lipoprotein particles (LDL) and high-density lipoprotein particles (HDL) were separated using non-denaturing polyacrylamide gradient gel (3-31%) electrophoresis. Serum non-cholesterols sterols (NCSs), desmosterol, lathosterol, 7-dehydrocholesterol (7-DHC), campesterol and β-sitosterol were measured by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Results  All patients had desirable serum HDL cholesterol concentrations during remission. The dominant lipoprotein diameters and LDL subclass distribution did not change significantly during follow-up. In contrast, HDL lipoprotein particle distribution shifted towards larger particles. The absolute concentration of desmosterol was significantly lower during remission (P = 0.023). β-sitosterol concentration markedly increased during remission (P = 0.005). Desmosterol/β-sitosterol (P < 0.001) and 7-DHC/β-sitosterol (P = 0.005) ratios significantly declined during disease remission. Conclusions  Favourable changes in the serum lipid profiles, HDL particle subclass distribution and cholesterol metabolism in paediatric patients with NS during remission took place. For the first time, we found that cholesterol homeostasis changed in favour of increased cholesterol absorption during disease remission. Nevertheless, complete cholesterol homeostasis was not achieved during disease remission.

AB  - Colorectal cancer (CRC) is a highly prevalent malignancy. Previous studies suggested that cholesterol might play a signficant role in malignant transformation and proliferation. Non-cholesterol sterols (NCS), which are transported by serum lipoproteins alongside cholesterol, are regarded as cholesterol synthesis and absorption markers. Quantification of NCS in serum and HDL fraction (NCSHDL), could provide a better insight into the cholesterol metabolism. The aim of this study was to examine the status of cholesterol synthesis and cholesterol absorption markers in serum and HDL fraction and explore their interrelation in CRC patients. Current study was designed as observational, case-control study. The study included 73 CRC patients and 95 healthy subjects. NCS and NCSHDL concentrations were determined by HPLC-MS/MS. Based on NCS and NCSHDL concentrations, different cholesterol homeostasis indices were calculated. Patients had significantly lower NCS (P<0.001) and NCSHDL concentrations (P<0.001 for desmosterolHDL; P<0.05 for lathosterolHDL, P=0.001 for campesterolHDL, P<0.001 for β-sitosterolHDL). NCSHDL/NCS (P<0.005 for desmosterolHDL/desmosterol; P<0.05 for lathosterolHDL/lathosterol; P<0.001 for both β-sitosterolHDL/β-sitosterol and campesterolHDL/campesterol) and synthesis to absorption ratio (CSI/CAI) (P<0.005) were increased in CRC patients. Additionally, low serum concentrations of desmosterol (P<0.001; OR=0.329; 95%CI (0.199–0.542)) and campesterol (P<0.001; OR=0.540; 95%CI (0.424–0.687)) were independent predictors of CRC presence. Our data suggest that cholesterol homeostasis in CRC is shifted towards increased synthesis. Relative abundance of NCS in HDL particles is increased, suggesting the possible overproduction of cholesterol precursors in peripheral tissues.

Considering that paraoxonase 1 (PON1) is an anti-oxidative enzyme located on high-density lipoprotein (HDL) particles, the aim of this study was to investigate the connection between oxidative stress (OS) and PON1 activity with lipoprotein subclasses in PCOS depending on obesity. Methods: In 115 PCOS patients lipoprotein subclasses distributions were determined by gradient gel electrophoresis. OS status was assessed by total oxidative status (TOS), advanced oxidation protein products (AOPP), malondialdehyde (MDA), prooxidant-Antioxidant balance (PAB), total antioxidant status (TAS) and superoxide dismutase (SOD) and PON1 activity. Results: Overweight/obese PCOS patients (n=55) had increased OS compared to normal weight patients (n=60). In addition, overweight/obese group had lower HDL size and higher proportion of HDL 3a subclasses (P<0.05). PAB was in negative correlation with HDL 2a (P<0.001), whereas MDA and SOD correlated positively with HDL 3 subclasses (P<0.05). Serum PON1 activity was positively associated with proportions of PON1 activity on HDL 2b (P<0.05) and 2a (P<0.01), but negatively with the proportion on HDL 3 particles (P<0.01). LDL B phenotype patients had increased TAS, SOD and PON1 activity on HDL 2b, but decreased PON1 activity on HDL 3 subclasses. Conclusion: OS is associated with altered lipoprotein subclasses distribution in PCOS patients. Obesity in PCOS affects the profile of HDL subclasses, reflected through the reduced proportion of PON1 activity on HDL 3 subclasses in the presence of sdLDL particles.

AB  - Insulin resistance, hyperlipidemia, oxidative stress and low-grade inflammation are
mechanisms underlying endothelial dysfunction and atherosclerosis development in non-
pregnant state, but in pregnancy they are adaptive mechanisms responsible for fetal
development without health risks for mothers. A special feature of lipids profile in pregnancy
is increase in high density lipoprotein (HDL) cholesterol concentration. Protective role of
HDL particles resides on its complex composition that goes beyond its cholesterol
concentration and exhibits vasodilatory, antioxidant, anti-inflammatory and
antithrombogenic effects. Even the importance of HDL functionality in pathogenesis of
cardiovascular disease has been investigated and confirmed its role in persevering metabolic
health in pregnancy are almost unexplored. Project "HDL research to improve pregnancy

AB  - Insulinska rezistencija, hiperlipidemija, oksidativni stress i stanje inflamacije niskog
stepena, koji se u negravidnom stanju povezuju sa razvojem endotelne disfunkcije i
aterosklerozom, u trudnoći predstavljaju adaptivne mehanizme koji omogućavaju pravilan
rast i razvoj fetusa, a ne ugrožavaju zdravlje trudnice. Specifičnost dislipidemije koja se
razvija u trudnoći, a koja nije karakteristična za dislipidemiju u negravidnim stanjima, jeste
porast koncentracije holesterola u lipoproteinskim česticama visoke gustine (high density
lipoprotein, HDL). Međutim, HDL je mnogo više od sadržaja holesterola u njemu. Ovaj
lipoprotein ima izuzetno kompleksnu i heterogenu strukturu i ispoljava vazodilatatorne,
antioksidativne, antiinflamatorne i antitrombogene efekte. Iako je gubitak funkcionalnosti
ovog lipoproteina u patogenezi kardiovaskularnih bolesti detaljno ispitan i potvrđen,
njegova uloga u očuvanju metaboličkog zdravlja u trudnoći još uvek nije rasvetljena. Projekat
"Ispitivanje HDL-metaboloma u cilju unapređenja ishoda trudnoće" bavi se istraživanjem
povezanosti obrazaca promena strukture i funkcionalnosti HDL (HDL-om) sa različitim

visokorizičnim trudnoćama, kao i trudnoćama bez komplikacija. Izdvajaju se komponente

trudnoće sa budućim kardiometaboličkim zdravljem majke i deteta. Ovako opsežno
ispitivanje trebalo bi da pruži jasniji prikaz uloge HDL u očuvanju metaboličkog zdravlja
tokom trudnoće i, kao takvo, predstavljaće polaznu tačku za dalja istraživanja u ovoj oblasti.

AB  - Background: Type 1 diabetes mellitus (T1DM) is one of the
most common endocrine diseases in children. T-cell autore-
activity toward b-cells is controlled by significant changes in
metabolism of T cells. Mammalian target of rapamycin
(mTOR) is an important intracellular regulator of metabolism
and cell growth. MAPK/MAK/MRK overlapping kinase 1
(MOK1) is one of the less known regulators of mTOR. We
sought to investigate if MOK1 and mTOR mRNA levels in
peripheral blood mononuclear cells (PBMCs) of T1DM pedi-
atric patients are different compared to healthy subjects.
Methods: This study included 172 adolescents with T1DM
and 36 healthy adolescent volunteers designated for control
group (CG). MOK1 and mTOR mRNA levels were deter-
mined in PBMCs by qPCR.
Results: T1DM patients have significant downregulation of
MOK1 mRNA levels in PBMCs compared CG (P=0.018),
while there was no significant difference in mTOR mRNA
levels (P=0.891). Furthermore, in T1DM patients, MOK1
significantly correlated with age, triglycerides and mTOR,
while mTOR correlated significantly with BMI and systolic
blood pressure. Overweight T1DM subjects had significantly
lower MOK1 (P=0.034) and mTOR (P=0.017) mRNA
levels, together with significantly higher levels of systolic
blood pressure (P<0.001), total cholesterol (P=0.001),
LDL-cholesterol (P=0.001) and CRP (P<0.001). Multi-
variate analysis showed that MOK1 was independently
negatively associated with T1DM when adjusted for sex, age, HDL-C and CRP (OR=0.417 (95%CI: 0.175–0.997),
p=0.049).
Conclusions: Our study demonstrated for the first time that
T1DM is associated with MOK1 downregulation. In addition,
downregulation of both mTOR and MOK1 gene expressions
was associated with cardiovascular risk factors in overweight
T1DM patients.

Cilj delovanja rapamicina kod sisara je važan unutarćelijski regulator metabolizma i rasta ćelija. Biomarkers of vitamin D status in healthy adults: associations with serum lipid parameters – a pilot study

AB  - Vitamin D deficiency is among important healthcare challenges today. Traditionally, vitamin D status is assessed through determination of 25-hydroxy metabolite (25(OH)D), but novel data point to 24,25(OH) 2 D and 25(OH)D/24,25(OH) 2 D ratio (VDMR) as promising biomarkers. It is widely accepted that the biological role of vitamin D exceeds its well-known contribution to bone turnover. However, its effects on overall energy metabolism and lipid status alterations are not completely understood. In this study, we analyzed the relationship of vitamin D status assessed as concentrations of 25(OH)D3 and 24,25(OH) 2 D3 determined by liquid chromatography-tandem mass spectrometry, as well as VDMR with advanced lipid status parameters. Vitamin D status biomarkers, routine parameters of lipid status and size and distribution of lipoprotein subclasses were determined in 89 healthy adults (35 with adequate vitamin D status and 54 with vitamin D deficiency). Our results indicated a preponderance of pro- atherogenic small, dense LDL particles (sdLDL) in vitamin D deficient subjects. Both 25(OH)D and 24,25(OH) 2 D were associated with a relative proportion of sdLDL (B: -0.410; SE: 0.154; P=0.010; and B: -2.041; SE: 0.969; P=0.039, respectively). Positive correlation was found for VDMR and relative proportion of HDL 3a particles (ρ=0.251; P=0.024). VDMR value was decreased in subjects with vitamin D deficiency (P=0.001), thus implying its usefulness as a biomarker. A thorough investigation of novel vitamin D biomarkers and advanced lipid status parameters can be useful in the estimation of individual risk for the development of cardiometabolic alterations.
AB  - Deficijencija vitamina D je jedan od značajnih izazova za zdravstvene sisteme današnjice. Tradicionalno, status vitamina D se procenjuje na osnovu određivanja koncentracije njegovog 25- hidroksi metabolita (25(OH)D), ali novi podaci ukazuju na potencijalni značaj 24,25(OH) 2D i odnosa 25(OH)D/24,25(OH) 2 D (VDMR). Danas je široko prihvaćeno da biološka uloga vitamina D uveliko prevazilazi njegove efekte na koštani promet. Međutim, još uvek nije u potpunosti jasan uticaj vitamina D na energetski metabolizam i izmene lipidnog statusa. U ovom istraživanju ispitivali smo povezanost između statusa vitamina D, procenjenog na osnovu nivoa 25(OH)D3 i 24,25(OH) 2 D3 koji su određeni metodom tečne hromatografije – tandem masene spektrometrije, te VDMR sa standardnim i novim parametrima lipidnog statusa. Ispitivani analiti određeni su kod 89 zdravih odraslih osoba. Naši rezultati ukazuju na povećan udeo malih gustih LDL čestica (sdLDL) kod osoba sa deficijencijom vitamina D. Koncentracije 25(OH)D i 24,25(OH) 2 D su bile nezavisno povezane sa relativnim udelom sdLDL (B: -0,410; SE: 0,154; P=0,010; i B: -2,041; SE: 0,969; P=0,039, redom). Traditionally, vitamin D status is assessed through determination of 25-hydroxy metabolite (25(OH)D), but novel data point to 24,25(OH) 2 D and 25(OH)D/24,25(OH) 2 D ratio (VDMR) as promising biomarkers. It is widely accepted that the biological role of vitamin D exceeds its well-known contribution to bone turnover. However, its effects on overall energy metabolism and lipid status alterations are not completely understood. In this study, we analyzed the relationship of vitamin D status assessed as concentrations of 25(OH)D3 and 24,25(OH) 2 D3 determined by liquid chromatography-tandem mass spectrometry, as well as VDMR with advanced lipid status parameters. Vitamin D status biomarkers, routine parameters of lipid status and size and distribution of lipoprotein subclasses were determined in 89 healthy adults (35 with adequate vitamin D status and 54 with vitamin D deficiency). Our results indicated a preponderance of pro- atherogenic small, dense LDL particles (sdLDL) in vitamin D deficient subjects. Both 25(OH)D and 24,25(OH) 2 D were associated with a relative proportion of sdLDL (B: -0.410; SE: 0.154; P=0.010; and B: -2.041; SE: 0.969; P=0.039, respectively). Positive correlation was found for VDMR and relative proportion of HDL 3a particles (ρ=0.251; P=0.024). VDMR value was decreased in subjects with vitamin D deficiency (P=0.001), thus implying its usefulness as a biomarker. A thorough investigation of novel vitamin D biomarkers and advanced lipid status parameters can be useful in the estimation of individual risk for the development of cardiometabolic alterations., Deficijencija vitamina D je jedan od značajnih izazova za zdravstvene sisteme današnjice. Tradicionalno, status vitamina D se procenjuje na osnovu određivanja koncentracije njegovog 25- hidroksi metabolita (25(OH)D), ali novi podaci ukazuju na potencijalni značaj 24,25(OH) 2D i odnosa 25(OH)D/24,25(OH) 2 D (VDMR). Danas je široko prihvaćeno da biološka uloga vitamina D uveliko prevazilazi njegove efekte na koštani promet. Taking  into  consideration  thatobesity  is  an  independent  risk  factor  for  many  cardio  -metabolic  diseases,  we  aimed  to  explore  the  relationshipbetween endocan levels and novel anthropometric indices i.e.,  body  adiposity  index  (BAI),  cardiometabolic  index(CMI), a body shape index, body roundness index, conicityindex,  lipid  accumulation  product  index  and  visceraladiposity  index and  traditional  ones   i.e.,  waist  circum  -ference,  hip  circumference,  body  mass  index,  waist-to-height ratio and waist-to-hip ratio in adult population.Methods:A  total  of  177  participants  were  included.Anthropometric indices and biochemical parametres weremeasured. Results:Univariate  regression  analysis  demonstrated  posi-tive correlations of endocan and almost all anthropometricdata. To explore independent associations of endocan andanthropometric parameters, the Model which fulfilled crite-ria  for  ordinal  regression  testing  was  created.  Adjusted odds  for  BAI  given  in  the  Model  (OR=1.120,  95%  CI1.036–1.212, P=0.004), demonstrated that a rise in BAIby 1 unit increased the probability of higher endocan con-centration  by  12%.  As  well,  a  rise  in  CMI  for  1  unit,increased the probability for higher endocan levels for 2.6times  (OR=2.599,  95%  CI  1.006–6.712,  P=0.049).  Atotal  of  20.1%  of  variation  in  endocan  levels  could  beexplained by this Model.Conclusions:Non-traditional obesity indices, BAI and CMIindependently correlated with higher serum endocan levelsin adult population.
AB  - Uvod: Povezanost endokana sa netradicionalnim antropometrijskim parametrima, kao različitim faktorima rizika za kardiovaskularne bolesti, nije ispitivana u prethodnim studijama. Endokan je novi biomarker inflamacije, čije su veće vrednosti zabeležene kod kardiometaboličkih poremećaja. Znajući da je gojaznost nezavisan faktor rizika za mnoga kardiometabolička oboljenja, cilj je bio da se ispita povezanost endokana i novih antropometrijskih pokazatelja tj. indeks telesne gojaznosti (BAI), kardiometabolički indeks (CMI), indeks oblika tela, indeks zaokruženosti tela, indeks koniciteta, indeks produkata lipidne akumulacije i indeks visceralne gojaznosti i onih tradicionalnih tj. obim struka, obim kukova, indeks telesne mase, odnos obim struka/telesna visina, odnos obim struka/obim kukova u populaciji odraslih. Metode: Ukupno 177 ispitanika je učestvovalo u istraživanju. Mereni su antropometrijski i biohemijski parametri. Rezultati: Univarijantna regresiona analiza je pokazala pozitivnu korelaciju endokana i skoro svih ispitivanih antropometrijskih parametara. U cilju daljeg ispitivanja postojanja nezavisnih korelacija endokana i antropometrijskih parametara, kreiran je Model koji je zadovoljio kriterijume za ordinalnu regresiju. Prilagođeni Odds-ovi za BAI u Modelu (OR=1,120, 95% CI 1,036-1,212, P=0,004), su pokazali da je porast BAI za 1 jedinicu povećao verovatnoću za porast koncentracije endokana za 12%. Takođe, porast CMI za 1 jedinicu, povećao je verovatnoću za veće vrednosti endokana za 2,6 puta (OR=2,599, 95% CI 1,006-6,712, P=0,049). Ukupno 20,1% varijabiliteta u vrednostima koncentracije endokana može biti objašnjeno ovim Modelom. Zaključak: Netradicionalni pokazatelji gojaznosti, BAI i CMI su nezavisno povezani sa većim vrednostima endokana u populaciji odraslih.

AB  - Introduction The link between preeclampsia and dyslipidemia has been established. Even though lipid profile parameters have been intensively investigated in the pathology of preeclampsia, their accurate molecular mechanisms of action have not been fully decoded. Objectives We aimed to identify the specifics of cholesterol metabolism in women affected by late‑onset preeclampsia and single out potential biomarkers associated with late‑onset syndrome. Patient s and methods A total of 90 pregnant women with a priori risk for preeclampsia were monitored at 4 time points during gestation and, based on the outcome of pregnancy, they were classified into the high‑risk group (70 women) and the preeclampsia group (20 women). Cholesterol metabolic profiling was done using liquid chromatography‑tandem mass spectrometry. Result s The only significant change in the preeclampsia group was an increase in the lathosterol level (P = 0.001). The first‑trimester lathosterol level was higher in the preeclampsia group compared with the high‑risk group (P = 0.02). Further, in the preeclampsia group, positive correlations were found between desmosterol and β‑sitosterol (ρ = 0.474; P = 0.03) in the third trimester, desmosterol and campesterol changes between the second and the first (ρ = 0.546; P = 0.02), and the third and first trimesters (ρ = 0.754; P <0.001), as well as between the desmosterol and β‑sitosterol differences between the third and first trimesters (ρ = 0.568; P = 0.01). No similar correlations were found in the high‑risk group. Conclusions Late‑onset preeclampsia could be associated with an altered lipid profile. By studying the quantitative metabolic signatures of cholesterol, we might assume that both cholesterol synthesis and absorption are increased, that is, there is an imbalance in the cholesterol homeostasis regulation in women affected by the disease.

Recent technological advancements empowered transcriptome exploration and led to the discovery of numerous non-coding RNA species of which microRNAs (miRNAs) and long non-coding RNAs (lncRNAs) are the most investigated. They are implicated in the regulation of numerous cellular functions, and as such are being extensively explored as potential biomarkers for various diseases. Altered expression of numerous lncRNAs and miRNAs in placenta has been related to pathophysiological processes that occur in preeclampsia. In the following text we offer summary of the latest knowledge of the molecular mechanism by which lnRNAs and miRNAs (focusing on the chromosome 19 miRNA cluster (C19MC)) contribute to pathophysiology of PE development and their potential utility as biomarkers of PE, with special focus on sample selection and techniques for the quantification of lncRNAs and miRNAs in maternal circulation.

AB  - Objective: Polycystic ovary syndrome (PCOS) is associated with altered lipid profile and increased small, dense LDL particles (sdLDL). Considering that paraoxonase 1 (PON1) is an anti-oxidative enzyme located on high-density lipoprotein (HDL) particles, the aim of this study was to investigate the connection between oxidative stress (OS) and PON1 activity with lipoprotein subclasses in PCOS depending on obesity. Methods: In 115 PCOS patients lipoprotein subclasses distributions were determined by gradient gel electrophoresis. OS status was assessed by total oxidative status (TOS), advanced oxidation protein products (AOPP), malondialdehyde (MDA), prooxidant-Antioxidant balance (PAB), total antioxidant status (TAS) and superoxide dismutase (SOD) and PON1 activity. Results: Overweight/obese PCOS patients (n=55) had increased OS compared to normal weight patients (n=60). In addition, overweight/obese group had lower HDL size and higher proportion of HDL 3a subclasses (P<0.05). PAB was in negative correlation with HDL 2a (P<0.001), whereas MDA and SOD correlated positively with HDL 3 subclasses (P<0.05). Serum PON1 activity was positively associated with proportions of PON1 activity on HDL 2b (P<0.05) and 2a (P<0.01), but negatively with the proportion on HDL 3 particles (P<0.01). LDL B phenotype patients had increased TAS, SOD and PON1 activity on HDL 2b, but decreased PON1 activity on HDL 3 subclasses. Conclusion: OS is associated with altered lipoprotein subclasses distribution in PCOS patients. Obesity in PCOS affects the profile of HDL subclasses, reflected through the reduced proportion of PON1 activity on HDL 3 subclasses in the presence of sdLDL particles.

AB  - Colorectal cancer (CRC) is a highly prevalent malignancy with multifactorial etiology,which includes metabolic alterations as contributors to disease development. Studies have shownthat lipid status disorders are involved in colorectal carcinogenesis. In line with this, previous studieshave also suggested that the serum high-density lipoprotein cholesterol (HDL-C) level decreases inpatients with CRC, but more recently, the focus of investigations has shifted toward the explorationof qualitative properties of HDL in this malignancy. Herein, a comprehensive overview of availableevidences regarding the putative role of HDL in CRC will be presented. We will analyze existingfindings regarding alterations of HDL-C levels but also HDL particle structure and distribution inCRC. In addition, changes in HDL functionality in this malignancy will be discussed.  Moreover,we will focus on the genetic regulation of HDL metabolism, as well as the involvement of HDL indisturbances of cholesterol trafficking in CRC. Finally, possible therapeutic implications related toHDL will be presented. Given the available evidence, future studies are needed to resolve all raisedissues concerning the suggested protective role of HDL in CRC, its presumed function as a biomarker,and eventual therapeutic approaches based on HDL.
