AB  - Toxicological research is mostly limited to considering the effects of a single substance, even though the real exposure of people is reflected in their daily exposure to many different chemical substances in low-doses. This in silico toxicogenomic study aims to provide evidence for the selected environmental (organo)metals (lead, cadmium, methyl mercury) + polychlorinated biphenyls mixture involvement in the possible alteration of thyroid, and male reproductive system function, and furthermore to predict the possible toxic mechanisms of the environmental cocktail. The Comparative Toxicogenomic Database, GeneMANIA online software, and ToppGene Suite portal were used as the main tools for toxicogenomic data mining and gene ontology analysis. The results show that 35 annotated common genes between selected chemicals and endocrine system diseases can interact on the co-expression level. Our study highlighted the disruption of the cytokines, the cell's response to oxidative stress, and the influence of the transcription factors as the potential core of toxicological mechanisms of the discussed mixture's effects. The connected toxicological effects of the tested mixture were abnormal sperm cells, a disrupted level of testosterone, and thyroid hormones. The core mechanisms of these effects were inflammation, oxidative stress, disruption of androgen receptor signaling, and the alteration of the FOXO3-Keap-1/NRF2-HMOX1-NQO1 pathway signaling most likely controlled by the co-expression of overlapped genes among used chemicals. This in silico research can be used as a potential core for the determination of biomarkers that can be monitored in future further in vitro and in vivo experiments.

AB  - The current study aimed to assess the connection between the mixture of lead (Pb), cadmium (Cd), arsenic (As), methylmercury (MeHg) and decabrominated diphenyl ether (decaBDE) and thyroid function, by using in silico toxicogenomic data-mining approach. To obtain the linkage between investigated toxic mixture and thyroid diseases (TDs), the Comparative Toxicogenomics Database (CTD) was used, while gene ontology (GO) enrichment analysis was performed by ToppGeneSuite portal. The analysis has shown 10 genes connected to all chemicals present in the mixture and TDs (CAT, GSR, IFNG, IL1B, IL4, IL6, MAPK1, SOD2, TGFB1, TNF), most of which were in co-expression (45.68%), or belonged to the same pathway (30.47%). Top 5 biological processes and molecular functions affected by the investigated mixture emphasized the role of two common mechanisms – oxidative stress and inflammation. Cytokines and inflammatory response was listed as the main molecular pathway that may be triggered by simultaneous exposure to toxic metal(oid)s and decaBDE and connected to TDs. The direct relations between Pb/decaBDE and redox status impairment in thyroid tissue was confirmed by our chemical-phenotype interaction analysis, while the strongest linkage between Pb, As and decaBDE and thyroid disorders was found. The obtained results provide better understanding of molecular mechanisms involved in the thyrotoxicity of the investigated mixture, and can be used to direct further research.

AB  - Benchmark dose (BMD) approach extends the use of data and allows a quantification
of the uncertainties and confirmation of the effect. The objective of this study was to
investigate whether a mixture of lead (Pb) and polychlorinated-biphenyls (PCBs)
has any effect on sperm cells, and whether these effects are dose dependent. The
experiment was conducted in accordance with the approval of Ethical Committee No. 323-
07-11822/2018-05. Male Wistar rats were exposed to the mixture of Pb and PCBs by oral
gavage for 28 days. Doses od Pb in mixture were 0.1 mg/kg/day, 0.5 mg/kg/day and 1
mg/kg/day, while doses of PCBs in mixture were 0.25 mg/kg/day, 0.5 mg/kg/day, and 1
mg/kg/day respectively. Rats were anesthetized by the combination of ketamine/xylazine.
Through the scrotum section, the right testis and epididymis were collected, while sperm
cells were taken from the cauda of epididymis by diffusion in saline solution at 37°C. Sperm
motililty was determined by the light microscope. The aliquot of the sperm cells was fixated
with the combination of formalin in the phosphate buffer and used for morphology and
concentration sperm analysis. The benchmark dose lower confidence limit (BMDL40) of
0.00013 mg/kg/day was derived for the sperm motility effect of Pb and PCBs as co-variant.
From our results, we can conclude that the mortality was the most sensitive parameter. This
value is also one of the lowest BMDL for Pb, which can lead to conclusion that PCBs increase
the potency of Pb toxicity.
AB  - Pristup računanja Benchmark doze (BMD) proširuje upotrebu podataka, omogućava
kvantifikaciju nesigurnosti i potvrđuje efekat. Cilj ove studije bio je da se ispita da li smeša
olova (Pb) i polihlorovanih-bifenila (PCBs) ispoljava efekat na ćelije sperme, kao i da se ispita
da li su efekti dozno zavisni. Eksperiment je sproveden u skladu sa odobrenjem Etičkog
komiteta br. 323-07-11822 / 2018-05. Mužjaci Wistar pacova su bili izloženi smeši Pb i PCBs

AB  - The aim of this study was to evaluate the awareness of livestock breeders on the used
veterinary drugs in order to assess potential environmental contamination in the region of
Sjenica. Veterinary drugs can enter the environment mostly due to widespread usage, but
also as metabolites eliminated from the organisms. In order to evaluate the awareness of
livestock breeders, a survey questionnaire was used. Questions were related to the most
frequently used veterinary drugs without the supervision of a veterinary medical doctor. The
obtained data show that the awareness of domestic animal breeders on the use of veterinary
drugs is very low in the region of Sjenica. The most commonly used veterinary medicines
were antibiotics, streptomycin, and benzylpenicillin-procaine, antihelmintic, albendazole.
Unexpectedly, the animal breeders identified formaldehyde as veterinary medicine and used
it as a disinfectant in poultry raising. The results also show that breeders frequently treat
animals with drugs on their own, without control by veterinary medical doctors. Because of
the wide use of veterinary medicines in the region of Sjenica, it monitoring of the
environmental concentration of streptomycin, benzylpenicillin-procaine, and albendazole, as
well as metabolites in soil and water should be recommended. Moreover, human exposure to
these veterinary medicines could occur because of the abuse of veterinary medicines, no
consultations with veterinary medical doctors, and non-proper handling of waste from
animals.
AB  - Veterinarski lekovi dospevaju u životnu sredinu najčešće usled neadekvatnog
odlaganja otpada, ali i kao metaboliti iz organizma životinja. Cilj studije je bio procena nivoa
svesti uzgajivača stoke o upotrebi veterinarskih lekova u regionu Sjenice sa ciljem procene
potencijalne kontaminacije životne sredine. Za procenu nivoa svesti uzgajivača stoke
korišćen je anketni upitnik. Pitanja iz upitnika su se odnosila na najčešće korišćene
veterinarske lekove i korišćenje bez nadzora veterinara. Dobijeni podaci pokazuju da je nivo
svestu uzgajivača stoke u regionu Sjenice nizak jer koriste veterinarske lekove bez nadzora
veterinara. Najčešće koriste antibiotike, streptomicin i benzoil-penicilin, antihelmintik –
abendazol i, iznenađujuće, koriste formaldehid za dezinfekciju prostorija za smeštaj
životinja, pri čemu ga smatraju veterinarskim lekom. Zbog rasprostranjene upoutrebe
veterinarskih lekova u regionu Sjenice od strane uzgajivača stoke, a bez kontrole veterinara
preporučuje se monitoring streptomicina, benzoilpenicilina i abendazola kao i njihovih
metabolita u životnoj sredini, zemljištu i vodi. Potrebno je preduzeti dodatne mere opreza da
zbog niskog nivoa svesti ne bi došlo do izloženosti ljudi ovim veterinarskim lekovima i
njihovim metabolitima naročito zbog neadekvatnog tretiranja stajnjaka i drugog otpada.

AB  - While experimental animal investigation has historically been the most conventional
approach conducted to assess drug safety and is currently considered the main method for
determining drug toxicity, these studies are constricted by cost, time, and ethical approvals. Over
the last 20 years, there have been significant advances in computational sciences and computer
data processing, while knowledge of alternative techniques and their application has developed
into a valuable skill in toxicology. Thus, the application of in silico methods in drug safety
assessment is constantly increasing. They are very complex and are grounded on accumulated
knowledge from toxicology, bioinformatics, biochemistry, statistics, mathematics, as well as
molecular biology. This review will summarize current state-of-the-art scientific data on the use
of in silico methods in toxicity testing, taking into account their shortcomings, and highlighting
the strategies that should deliver consistent results, while covering the applications of in silico
methods in preclinical trials and drug impurities toxicity testing.
AB  - Ispitivanja na eksperimentalnim životinjama ne samo da su u prošlosti bila smatrana najkonvencionalnijim pristupom za procenu bezbednosti lekova, već su i trenutno osnovna metoda za utvrđivanje njihove toksičnosti. Međutim, ova ispitivanja su skupa, vremenski zahtevna i za njihovo sprovođenje neophodne su etičke dozvole. Tokom poslednjih 20 godina došlo je do napretka u računarskoj nauci i kompjuterskoj obradi podataka, dok se znanje o alternativnim tehnikama i njihovoj primeni razvilo u dragocenu veštinu u toksikologiji. Stoga, primena in silico metoda u proceni bezbednosti lekova neprestano raste. Ove metode su veoma složene i zasnivaju se na saznanjima iz toksikologije, bioinformatike, biohemije, statistike, matematike i molekularne biologije. Ovaj pregledni rad će rezimirati trenutna naučna saznanja koja se tiču upotrebe in silico metoda u ispitivanju toksičnosti lekova, uzimajući u obzir njihova ograničenja i ističući strategije pomoću kojih se mogu dobiti konzistentni rezultati, sa posebnim osvrtom na primenu in silico metoda u pretkliničkim ispitivanjima i ispitivanjima toksičnosti nečistoća u lekovima.
