AB  - Three seaweeds (Halimeda tuna, Codium bursa and Cystoseira barbata) and one seagrass (Cymodocea nodosa) were collected from the Coast of Montenegro, Gulf of Boka Kotorska and their chemical analysis was performed. In seagrass C. nodosa, three phenolic compounds were identified (diosmetin 7-sulfate, caftaric and coutaric acid). The content of β-glucan, fatty acids, sterols and micro- and macro-elements were investigated among all samples. The highest content of β-glucan was detected in C. nodosa seagrass (13.04±0.42 g/100 g). The highest polyunsaturated fatty acids (PUFAs) level was reported in C. barbata, the brown alga (7.157 mg/g), which also had the significant sterol content (fucosterol, 21.76±0.1 μg/g). Green algae, C. bursa and H. tuna, showed the highest level of sterols (β-sitosterol, 95.21±0.16 μg/g and 73.90±0.08 μg/g, respectively). H. tuna had the highest content of calcium (Ca) in amount of 55125 μg/g. In C. bursa, C. barbata and C. nodosa, the Na/K ratio was low (0.43, 0.46 and 0.69, respectively).

AB  - Pronalazak se odnosi na nov postupak ekstrakcije elaginske kiseline iz herbe biljne vrste gospin plašt (Alchemilla vulgaris L.). Postupak ekstrakcije pokazuje neočekivanu selektivnost za elaginsku kiselinu, dok je sadržaj drugih jedinjenja ispod granica detekcije. Ispitivanja ekstrakta A. vulgaris pokazuju zadovoljavajuću antioksidativnu aktivnost, a određen je i sadržaj ukupnih flavonoida. Dobijeni ekstrakt herbe A. vulgaris može biti primenjen kao prirodni aditiv u prehrambenoj, kozmetičkoj i farmaceutskoj industriji, jer ima jak antioksidativni i zaštitni efekat.

AB  - The essential oil from birch (Betula pendula Roth.) buds was obtained by Clevenger-type hydrodistillation. Its qualitative and quantitative composition was determined by gas chromatography in combination with mass spectrometry (GC/MS) and flame ionization detection (GC/FID), respectively. Twenty-seven compounds, mainly sesquiterpene hydrocarbons (78.7%) and oxygenated sesquiterpenes (14.8%), were identified comprising 93.5% of total identified components in the essential oil. The most abundant compounds were germacrene D (21.7%) and -cadinene (17.0%). Antimicrobial activity of isolated essential oil against Gram-positive and Gram-negative bacteria as well as fungus Candida albicans was determined using the disc-diffusion method. The isolated essential oil has shown antimicrobial activity against the following microorganisms: Proteus vulgaris, Bacillus luteus and Klebsiella pneumoniae. Antioxidative activity of the essential oil was determined using the DPPH assay, after adding DPPH radical and after 20min, 30min and 60min incubation with radical. Essential oil showed antioxidant activity.

Growth rings width of bald cypress stems from two alluvial sites in Serbia

AB  - The paper shows the differences in growth rings width between bald cypress stems grown on two alluvial sites in Serbia - Veliko ratno ostrvo and Backa Palanka. Performing this anatomical research included falling of overall six trunks (3 from one and 3 from another location). After that, appropriate wood cross-sections were made and they served for making permanent anatomical preparations. Obtained results proved a big dependence between radial increment from one side and available water content together with nutrients content on the other side. It caused a very significant correlation between growth rings width and qualitative site features. Bearing on mind this fact, it was established that Veliko ratno ostrvo is more suitable for bald cypress growing. As for behavior of radial increment with ages, there are some differences between these locations, first of all because stems from Veliko ratno ostrvo still did not reach its culmination.

AB  - Pronalazak se odnosi na novu primenu hidrogelova poli(N-izopropilakrilamid-ko-2-hidroksipropilmetakrilata), p(NIPAM-HPMet)-a i poli(N-izopropilakrilamida), p(NIPAM)-a, kao nosača za modifikovano oslobađanje elaginske kiseline i ekstrakta Alchemilla vulgaris L. sa visokim sadržajem elaginske kiseline. Analiza FTIR spektara homopolimera p(NIPAM)-a i kopolimera p(NIPAM-HPMet)-a sa uklopljenim standardom elaginske kiseline i ekstraktom Alchemilla vulgaris L., ukazuju na formiranje intermolekulskih vodoničnih veza između lanaca homopolimera, tj. kopolimera, s jedne strane i elaginske kiseline, s druge strane. Hidrogelovi p(NIPAM) i p(NIPAM-HPMet) su se pokazali kao pogodni nosači elaginske kiseline, jer omogućavaju njeno modifikovano oslobađanje.

AB  - Stimulus-sensitive hydrogels are used as carriers for modified release of pharmaceuticals. The synthesis of thermosensitive hydrogels poly(N-isopropylacrylamide), p(NIPAM), and poly(N-isopropylacrylamide-co-2-hydroxypropyl methacrylate), p(NIPAM-HPMet), is performed. The synthesized hydrogels are characterized using FTIR and scanning electron microscope methods and swelling properties, and applied for modified release of ellagic acid (EA). This work presents the selective extraction of EA, as a natural antioxidant, from the aerial parts of Alchemilla vulgaris L. EA and A. vulgaris extract are incorporated into p(NIPAM) and p(NIPAM-HPMet) hydrogels and characterized by FTIR method. The EA content in the extract by the UHPLC-DAD-HESI-MS/MS method is determined (0.64mgcm(-3)). The total flavonoids content in the A. vulgaris extract was determined by the spectrophotometric method. Antioxidant activity of the A. vulgaris extract and EA is examined using the 2,2-diphenyl-1-picrylhydrazyl assay. The p(NIPAM-HPMet) shows a better incorporation and release at 37 degrees C of EA standard and A. vulgaris extract (98.87 and 96.45% respectively), compared with p(NIPAM). [GRAPHICS]

Elemental composition and nutritional value of three edible mushrooms from Serbia

AB  - Nutrition value, as well as macro- and microconstituents of three edible mushrooms (Pleurotus ostreatus, Lentinula edodes, Macrolepiota procera) collected in Serbia were tested. It has been determined that all three mushroom samples were low in energy, fat and carbohydrates, but rich in β-glucans content. The most abundant elements in these three mushrooms were essential marcroelements Ca, K and P, but also there is a significant amount of Li, Se and Zn. On the other hand, toxic elements, such as Pb, As and Cd were also detected in level higher than allowed.
AB  - Nutritivna vrednost, kao i sadržaj makro- i mikroelemenata testirana je u uzorcima tri jestive pečurke, sakupljene u Srbiji (Pleurotus ostreatus, Lentinula edodes, Macrolepiota procera). Pokazano je da su sve tri gljive bile siromašne energijom, mastima i ugljenimhidratima, ali bogate β-glukanima. Najzastupljeniji elementi u sva tri uzorka bili su esencijalni makroelementi Ca, K i P, ali je takođe je detektovana značajna količina Li, Se i Zn. Sa druge strane, koncentracija toksičnih metala, Pb, As i Cd, bila je iznad dozvoljenih granica.

AB  - Antibacterial and cytotoxic activities of cyclohexane, dichloromethane, methanol, and aqueous extracts of Cantharellus cibarius were tested. Broth microdilution assay was performed against 10 bacterial strains (Staphylococcus aureus, S. epidermidis, Micrococcus luteus, Bacillus subtilis, Enterococcus feacalis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella abony), with emphasis on Helicobacter pylori. Methanol extract was the most active against H. pylori strains with minimal inhibitory concentration values between 4 and 32 mu g/mL. All extracts were active against antibiotic resistant H. pylori. Methanol and aqueous extracts had no cytotoxicity against tested cell lines, whereas cyclohexane and dichloromethane extracts were active against HeLa and N87 cells, but also against healthy MRC-5 cells (IC50 39.26 +/- 1.24-134.79 +/- 0.01 mu g/mL). The tested aqueous extracts have shown 68% of angiotensin-converting enzyme inhibitory activity in doses of 1.25 mg/mL. Chemical analysis has shown the presence of linoleic, cis-vaccenic, and oleic acids, sterols, beta-glucans, and polyphenolic compounds.

AB  - The aim of this paper was to estimate the biological activity of the plum seed extract and to define the chemical composition by using the ESI-MS method. During the investigation of the antioxidant activity, the extract showed a better ability to inhibit DPPH radicals compared with amygdalin standard. The results of the antimicrobial study indicate that the extract has a greater effect on Gram-negative bacteria compared with amygdalin. Gram-positive bacteria and fungi remained resistant in both cases. Unlike amygdalin, the extract showed a greater antimicrobial activity against the same microbiological strains. Amygdalin was isolated from the extract before the investigation of antiproliferative activity against MDA-MB-361, MDA-MB-453, HeLa and LS174 cell lines using MTT assay. Unlike the MDA-MB-453 cell line, the activity of standard and isolated amygdalin was not observed at the concentrations lower than 400 μg cm-3 on MDA-MB-361 cell line. The isolated amygdalin has not shown the activity in other cell lines. The antiproliferative activity of amygdalin standard was also noticed on HeLa and LS-174 cell lines at the concentrations of 333.27 and 333.17 μg cm-3, respectively.
AB  - Cilj ovog rada bila je procena biološke aktivnosti ekstrakta semena šljive i definisanje hemijskog sastava primenom ESI-MS metode. Tokom ispitivanja antioksidativne aktivnosti, ekstrakt je pokazao bolju sposobnost inhibiranja DPPH radikala u poređenju sa standardom amigdalina. Rezultati antimikrobne studije pokazali su da ekstrakt ima veći uticaj na rast Gram-negativnih bakterija u odnosnu na amigdalin. AB  - The process of amygdalin extraction from plum seeds was optimized using central composite design (CCD) and multilayer perceptron (MLP). The effect of time, ethanol concentration, solid-to-liquid ratio, and temperature on the amygdalin content in the extracts was estimated using both mathematical models. The MLP 4-3-1 with exponential function in hidden layer and linear function in output layer was used for describing the extraction process. MLP model was more superior compared with CCD model due to better prediction ability. According to MLP model, the suggested optimal conditions are: time of 120 min, 100% (v/v) ethanol, solid-to liquid ratio of 1:25 (m/v) and temperature of 34.4 degrees C. The predicted value of amygdalin content in the dried extract (25.42 g per 100 g) at these conditions was experimentally confirmed (25.30 g per 100 g of dried extract). Amygdalin (>90%) was isolated from the complex extraction mixture and structurally characterized by FT-IR, UV, and MS methods.

AB  - Molecular inclusion complexes of usnic acid (UA) with beta-cyclodextrin (beta-CD) and 2-hydroxypropyl beta-cyclodextrin (HP beta-CD) were prepared by the co-precipitation method in the solid state in the molar ratio of 1:1. Structural complexes characterization was based on different methods, FTIR, H-1 NMR, XRD and DSC. Parallel to the complex by the above methods, corresponding physical mixtures of UA with cyclodextrins and complexing agents (beta-CD, HP beta-CD and UA) were analyzed. The results of DSC analysis showed that, at around 200 A degrees C, the endothermal peak in the complexes with cyclodextrins originating from the UA melting has disappeared. Complex diffractogram patterns do not contain peaks characteristic for the pure UA. They are more appropriate to cyclodextrin diffractogram. This fact points to the molecular encapsulation of UA in the cyclodextrin cavity. Chemical shifts in H-1 NMR spectra after the inclusion of UA into the cyclodextrin cavity, especially H-3 protons (0.0012 and 0.0102 ppm in the beta-CD and HP beta-CD, respectively) and H-5 and H-6 (0.0134 ppm) and hydrogen from CH3 (0.0073 ppm) HP beta-CD also points to the formation of molecular inclusion complexes. The improved solubility of UA in water was achieved by molecular incapsulation. In the complex with beta-CD the solubility is 0.3 mg/cm(3), with HP beta-CD 4.2 mg/cm(3) while the uncomplexed UA solubility is 0.06 mg/cm(3). The microbial activity of UA and both complexes was tested against eight bacteria and two fungi and during the test no reduced activity of UA in the complexes was observed.

AB  - Background: Venous ulcers (ulcus cruris venosum) are a common chronic disease that requires continuing treatment and significantly influences a patient's way of life. The therapeutic effects of the ointment Herbadermal on epithelialization and microbial flora of venous ulcers in 25 patients (10 men and 15 women) were tested over a 7-week period. The major components of the ointment are extracts of garlic, St. John's wort, and calendula. Material and Methods: The patients were over 18 years of age, with ulceration of the lower leg no longer than 2 months or recurrent ulceration during the last 6 months. The involved patients did not use any other phytomedicines or supportive therapies. Parameters were evaluated before the treatment and every 2 weeks during the period of 7 weeks. The total treatment response was evaluated on the basis of epithelialization, granulation, fibrin deposits, exudation, and edema. Results: The percentage of epithelialization was 99.1% after 7 weeks, without significant effects on the microbial flora. Conclusion: This combination of extracts can be recommended as topical treatment for wound healing because of its epithelizing, anti-erythematous, and anti-edematous properties.

AB  - According to the tested antimicrobial activity of constituents and their chemical characteristics, a new formulation of oral antiseptic was made based on garlic bulb powder and its active principles with strong antimicrobial activity against a wide range of bacteria and fungi. The antimicrobial activity of garlic bulb powder, allicin and the lozenge with 15% of garlic powder was tested using broth microdilution method. The tested garlic powder, as well the lozenge, have shown very high antimicrobial activity with MIC 1.25-5.00 mg/ml, and 1.87-7.50 mg/ml, respectively. The major compound, allicin, was highly active at a very low concentration. The minimal inhibitory concentration of allicin was from 6.25-12.50 μg/ml for antibacterial activity and 0.4 μg/ml for antifungal activity. Those concentrations are comparable with concentrations of commercially available antibiotics and fungicides. The formulation of an antiseptic with herbal constituents, high antibacterial and antifungal activity and pleasant taste could be an alternative to classical pharmaceutical oral antiseptics.
AB  - Na osnovu ispitane antimikrobne aktivnosti komponenata i njihovih hemijskih karakteristika, napravljena je nova formulacija oralnog antiseptika koji kao aktivnu komponentu ima prašak belog luka čiji sastojci pokazuju veoma izraženu aktivnost protiv širokog spektra bakterija i gljive Candida albicans. Antimikrobna aktivnost praška belog luka, alicina i lozengi sa 15% praška belog luka je ispitivana bujon mikrodilucionom metodom. Testiran prašak belog luka, kao i lozenge imaju veoma jaku antimikrobnu aktivnost sa minimalnim inhibitornim koncentracijama (MIC) 1,25-5,00 mg/ml (prašak belog luka) i 1,87-7,50 mg/ml (lozenge). Glavna aktivna komponeta belog luka, alicin, pokazuje antimikrobnu aktivnost u veoma niskim koncentracijama. Minimalne inhibitorne koncentracije alicina se kreću od 6,25-12,5 μg/ml protiv bakterija i 0,4 μg/ml protiv gljivice Candida albicans. Ove koncentracije se mogu uporediti sa koncentracijama komercijalnih antibiotika i fungicida. Naši rezultati su u skladu sa prethodnim ispitivanjima alicina i različitih ekstrakta belog luka, i potvrđuju najvažniju ulogu alicina u antimikrobnoj aktivnosti. Veoma je važna visoka aktivnost ovako formulisanog oralnog preparata protiv gljivice C. albicans koja je čest uzročnik infekcija usne duplje. Formulisan proizvod sa antiseptičkim osobinama karakteriše stanje čvrstog rastvora gde su aktivne supstance molekularno-disperzno ugrađene u obliku dobro rastvornog polimernog matriksa (sorbitola) velike specifične površine. Dobra rastvorljivost polimernog matriksa i velika specifična površina omogućuju zadovoljavajuću brzinu otpuštanja aktivnih principa iz belog luka (alicin). Pored visoke antimikrobne aktivnosti, lozenge poseduju prijatan ukus i osvežavaju dah, pa se mogu koristiti kao prirodni antiseptik u prevenciji i terapiji infekcija sluzokože usta i ždrela. Formulacija antiseptika sa biljnim sastojcima i visokom antibakterijskom i antifungalnom aktivnošću može biti alternativa farmaceutskim oralnim antisepticima.

AB  - Paclitaxel is isolated from the Pacific yew. It can be obtained from the European yew, but only after chemical modification of the isolated compound by a semi-synthesis procedure. The procedure for total synthesis of paclitaxel is very complicated, involving multiple steps, and the yields of paclitaxel are meagre. This substance is also a metabolite of certain kinds of fungus. The microbiological pathway for producing paclitaxel compared with isolation from plant material involves shorter procuction times but a small yield. Cyclodextrins are usually used for improving the solubility of paclitaxel in aqueous media, with polymeric and other substances added. Paclitaxel has anticancer activity and use for preparing the formulations intravenously administrated to patients with tumors. The paclitaxel concentration in these formulations is determined using validated HPLC methods.
