Extraction Kinetic of Some Phenolic Compounds During Cabernet Sauvignon Alcoholic Fermentation and Antioxidant Properties of Derived Wines

AB  - In this study, we estimated the usage of Cabernet Sauvignon in microvinifications, obtaining wines with increased phenolic compound content. Kinetic extraction of phenolic compounds during alcoholic fermentation was affected by maceration time (3, 5, 7, 14 and 21 days) and the addition and kinetics of enzyme preparations (EP). The highest extraction rates were observed for catechin (EXV EP – EXV en-zyme preparation and CP EP – Color plus enzyme preparation) and p-hydroxybenzoic acid (Car EP - Ca-ractere enzyme preparation). According to extraction time of the analyzed phenolic compounds, maximal values (ellagic acid, ferulic acid, chlorogenic acid, caffeic acid, naringenin, p-hydroxybenzoic acid, p-coumaric acid, protocatechuic acid, trans-resveratrol, syringic acid, vanillin, and vanillic acid) were ob-tained on day 15 of maceration with addition of CP EP, with exceptions of gallic acid, catechin, and my-ricetin. Prolonged maceration times, up to 21 days, showed the most potent DPPH free radical scavenging activity with Car EP and the highest Ferric Reducing Ability of Plasma (FRAP) values with CP EP.

Streptomyces sp JS520 produces exceptionally high quantities of undecylprodigiosin with antibacterial, antioxidative, and UV-protective properties

AB  - A Gram-positive, red-pigment-producing bacterial strain, designated JS520 was isolated from the pristine sediment from the cave on mountain Miroc in Serbia. Strain was confirmed to belong to Streptomyces genus based on phenotypic and genetic analysis. Streptomyces sp. JS520 has the ability to produce exceptionally high amounts of deep red pigment into both solid and liquid media. Liquid chromatography and mass spectroscopy of the purified pigments revealed the major component to be undecylprodigiosin (93 %) with minor component being oxidatively cyclized derivative. The pigment production was affected by medium composition, temperature, pH, and the aeration rate. By medium optimization, yields of undecylprodigiosin of 138 mg l(-1) were achieved, what is the highest level of undecylprodigiosin production reported for the members of Gram-positive Streptomyces genus. Purified pigment had antimicrobial properties against bacterial Bacillus and Micrococcus species (50 mu g ml(-1)) and against Candida albicans species (100-200 mu g ml(-1) range). The ability to affect auto-oxidation of the linoleic acid was demonstrated for the purified undecylprodigiosin, suggesting antioxidative properties of this pigment. Multiple ecophysiological roles of the pigment were revealed by comparing cultures grown under pigment-producing and pigment-nonproducing conditions. Cells grown under undecylprodigiosin-producing conditions could tolerate presence of hydrogen peroxide exhibiting three times smaller zones of inhibition at 100 mM H2O2. Undecylprodigiosin-producing cells were also less susceptible to tetracycline, kanamycin, chloramphenicol, and 8-hydroxyquinoline. While the growth of the cells not producing pigment was completely inhibited by 15 min of exposure to ultraviolet light (254 nm), cells producing undecylprodigiosin and cells supplied with purified pigment in vitro showed survival rates at 22 and 8 %, respectively.

AB  - Allyl acetates were synthesised from allenes utilising methodology based on the general reactivity of pi-allyl palladium intermediates which participate efficiently in transformations involving nucleophiles. Reactions of allenes and aryl iodides in the presence of AcONa and Pd(OAc)(2)/PPh(3) as the catalytic system afforded allyl acetates in moderate to good yields. Monosubstituted allenes, depending on their structure, produced either a separable mixture of two regioisomeric products or a single regioisomer. As allylic acetates can be easily hydrolysed, the methodology is applicable for the synthesis of allyl alcohols as well.

Study of forced degradation behavior of Eletriptan hydrobromide by LC and LC-MS and development of stability-indicating method

AB  - The objective of the present study was to report the stability profile of novel antimigrain drug Eletriptan hydrobromide based on the information obtained from forced degradation studies. The drug was subjected to acid (0.1-1 mol L-1 HCl), neutral and base (0.1-1 mol L-1 NaOH) hydrolysis and to oxidative decomposition (3-15% (v/v) H2O2). Photolysis and thermo degradation at 75 degrees C were carried out in methanol solution and in solid state with both Eletriptan hydrobromide bulk drug and the tablet formulation. The products formed under different stress conditions were investigated by LC and LC-MS. The experimental conditions for LC were chosen by employing experimental design and multicriteria decision making methodology. These powerful tools enabled the accomplishment of satisfactory resolution with the shortest possible analysis time. Analytes were separated on a C-18 column (XTerra (TM), 150 mm x 3.9 mm, 5 mu m) with the mobile phase composed of methanol-water solution of TEA (pH 6.52, 1%, v/v) (30:70, v/v) pumped at 1 mL min(-1) flow rate. The column temperature was set at 50 degrees C and the detection at 225 nm using DAD detector. The LC method was suitably modified for LC-MS analysis which was further used to characterize the arisen degradation products. The possible degradation pathway was outlined based on the results. The drug appeared to be instable towards every stress condition but oxidation. The stability was not jeopardized even under more exaggerated conditions such as increased temperature of the solutions to 75 degrees C, increased strength of acid/alkali solutions and prolonged testing period. Validation of the LC-DAD method was carried out in accordance with ICH guideline. The method met all required criteria and was applied when testing the commercially available tablets.

AB  - The aim of the present work was to investigate and compare phenolic compounds and antioxidant activity of methanol extracts of Quercus robur and Quercus cerris acorn kernels obtained before and after thermal treatment. Content of total phenolics, tannins, non-tannin phenolics and flavonoids was determined spectrophotometrically and content of gallic acid with HPLC. Antioxidant activity of the samples was assayed through FRAP (Ferric Reducing Antioxidant Power), DPPH scavenging test and inhibition of Fe2+/ascorbate induced lipid peroxidation. Extracts of native and thermally treated kernels showed high antioxidant activity, with extracts of thermally treated kernels being more active than extracts of native ones. Hydrolysable tannins and gallic acid were identified in all samples. Non-tannin phenolics, including gallic acid, were present in significantly higher quantities in thermally treated samples, whilst tannin content decreased. This indicates that during thermal treatment hydrolysable tannins were degraded. As the result of this degradation and consequent increase of non-tannin phenolics content, and amongst them especially gallic acid, thermally treated samples possess higher antioxidant activity than do the native ones. The obtained results have provided further grounds for establishing Q. robur and Q. cerris acorn kernels as a source for functional food preparation,

spectrophotometrically using FC-reagentr, while gallic acid content was estimatecl usipg
HPLC2. After thermal treatment tannin content was decreased (from 20,4 to 19,70 ),
with significant increase of simple polyphenols and gallic acid content (from 1,9 to
4,004, and from 0,5 to 5,lYo, respectively).
Antioxidant activity was estimated using FRAP test (total antioxidant activity,
TAA), scavenging of DPPH radical and TBA test (inhibition of lipid peroxidation, LP
in liposomes)r.
Sample-s QN and QT showed substantial antioxidant activify. TAA was 2.44 and.
3.76 pmol F.'*/mg, respectively. Comparing to quercetin and L-ascorbic acid these
values were ca 2-3 times lower. Both samples concentration-dependently scavenged
stable DPPH radical (IC5s:8.04 and 5.77 pglml), and inhibited LP (IC50:32.13 and
I 6.39 1tg/m| respectively).
Higher antioxidant activity of extract of thermally treated kernels (QT) could be
explained as a consequence of thermal degradation of hydrolysable tannins and,
therefore, increased gallic acid content.
AB  - Ispitivani su polifenolni sastojci i antioksidantna svojstva suvih MeOI.l
ekstrakata nativnog (QN) i termidki tretiranog (QT) semena luZnjaka, euercus robur.
SadrZaj ukupnih i netaninskih polifenola i tanina odredivan je spektiofotometrijski,
kori5cenjem Folin-Ciocalteu reagensa', a sadraaj galne kiseline tecnonr
hromatografijom (HPLC)2. Konstatovano je da nakon termicktg tretmana sadrZaj tanina
opada (sa 20,4 na 19,7o ), a znalajno raste kolidina netaninskih polifenola 1sa f ,9 na
4,0oA) i galne kiseline (sa 0,5 na 5 ,loh).
Za procenu antioksidante aktivnosti odredivani su: ukupna antioksidanta
aktivnost, UAA (FRAP test), sposobnost neutralizacije DPPH radikala i inhibitorni
efekat na lipidnu peroksidaciju (LP) u lipozomima (TBA tesr).r
Uzorci QN i QT ispoljili su znadajnu antioksidantnu aktivnost, i to UAA je
iznosila 2,44 i 3,76 pmol F.'*/rg, respektivno. Dobijene vrednosti aktivnosti su oko 2-
3 puta niZe u poredenju sa kvercetinom i vitaminom C. lJzorci eN i eT su
koncentraciono zavisno neutralisali DPPH radikal (IC5s:8,04 i 5,77 pglml) i inhibirali
LP (ICs0:32,13 i I 6,39 ltglml, respektivno).
Veia antioksidantna aktivnost ekstrakta termidki tretiranog semena (eT), moLe
se objasniti veiim sadrZajem galne kiseline, koja se oslobada razgradnjom
hidrolizujucih tanina tokom termidke obrade.
