Cherry-the source of potential functional food

AB  - Serbia is among the top world producers of high-quality cherry fruit. Beside it refreshing taste cherries are rich source of many natural active principles. The most interesting among them are those one which exhibit antioxidant properties. It is possible to produce many derived products from cherries and one certainly is fruit wine. Such wine, is rich source of those natural active principles present in cherry fruit and the aim of this study is to investigate antioxidant properties of cherry wine. Fruit wines were produced from Serbian autochthonous sort of cherry. Microvinification was conducted with pure selected yeast culture and enzymatic preparation glycosidase. In the half of microvinifications pits were left, without cracking. Determinations of total phenolic content (TPC) were conducted using Folin-Ciocalteu method. Antiradical activity estimated by DPPH method, while FRAP method was also applied. Selected phenolic compounds were quantified by UPLC TQ-MS/MS. The TPC for the cherry wines were in interval 1552-1787 mg GAE/L while FRAP was 45.3 – 73.5 mmol/L Fe2+. The IC50 anti DPPH radical activity was from 3.8 to 7.2%. Also, were quantified compounds which exhibit antioxidant properties which are hydroxybenzoic acid derivatives, such as vanillic, gallic, protocatehuic and parahydroxybenzoic acid. The obtain results indicate that cherry wine is good source of antioxidant compounds. Antioxidant properties and quantity of phenolic compounds depends from the technological process applied in the production of fruit wines. Cherries and their derived products have beneficial health effect for overall health.

AB  - This study aimed to estimate the potential of the selected berry (strawberry) and drupe (apricot, plum and sweet cherry) fruits as the substrates for the production of new fruit wines enriched with phenolic compounds. Sweet cherry wine (cultivar Burlat) stood out both for the profound content of phenolics (followed by their chemical profile) and potent anti-DPPH radical activity. The same fruit wine samples exhibited high redox potentials, slightly lower than strawberry and plum wine samples. Therefore, sweet cherry cultivar Burlat may be well recommended for development of novel fruit-based products endowed with naturally occurring phenolics. In any case, the remaining four cultivars are also worth further research efforts, particularly apricot cultivar Keckemetska ruza, which is one of the representatives of greatly underestimated fruit type thus far as a substrate for the production of the relevant fruit wines.

AB  - Introduction - Fruit and their products, including fruit wines, represent a rich source of natural bioactive compounds. This study focusing on fruit wines (prepared from commercially grown fruits by Serbian producers) has included the investigation of their chemical composition and biological activity. Materials and methods - Black chokeberry, blueberry, raspberry, blackberry and cherry were used for wine production by innovative vinification procedure, with or without using sugar and enzymatic preparation glycosidase, respectively. Selected phenolics were identified and quantified by UPLC/MS-MS analysis, while Total Phenolic Content (TPC) was determined by the Folin-Ciocalteu method. In addition to this, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and FRAP (Ferric Reducing Ability of Plasma) methods were applied for the preliminary evaluation of anti-DPPH radical activity and redox potential respectively at in vitro conditions. Results and discussion - Among the fruit wines examined within this study, the blackberry one stood out for profound FRAP (115.23 mmol L-1 Fe2+), DPPH (1.11%) and TPC values (2,395 mg GAE L-1). On the other hand, the raspberry wine showed the lowest potential towards the aforementioned parameters. Using principal component analysis, these fruit wines were chemically differentiated, according to the predominant phenolic compounds. Conclusions - All fruit wine samples displayed a good antioxidant potential with the blackberry one being most potent. Such a finding is of particular importance for Serbia as one of the leading producers of this edible fruit both in Europe and rest of the world.

Phenolic profile of some fruit wines and their antioxidant properties

AB  - The composition and content of individual phenolic compounds in various fruit wines and overall antioxidant properties of these fruit wines were studied. Phenolic compounds were separated by reverse phase (RP) HPLC techniques, and their content was determined by means of mass spectrometer (MS) with triple quadrupole (TQ) analyser, which recorded specific precursor-product transitions. Antioxidant activity was monitored by the following spectrophotometric methods: DPPH, FRAP and Folin-Ciocalteu assay, respectively. Four types of berries (raspberry, blackberry, chokeberry and blueberry), one type of pome (apple) and one type of stone fruit (cherries) were used for the production of fruit wines. Corresponding fruit wines were produced by microvinification with or without adding sugar into the must before fermentation. Increase in alcohol level was responsible for the increased content of phenolic compounds in fruit wine due to improved extraction conditions. Produced fruit wines were preserved by adding SO2 which interferes with the determination of antioxidant activity of phenolic compounds. In this case, the development of a correlation method based on FRAP assay was introduced. Fruit wines are a rich source of substances which show beneficial effects on human health. Depending on the fruit type, different antioxidant compounds were predominant in wine samples.

AB  - Procedure for isolation of pyrrolizidine alkaloids (PAs) from Rindera umbellata Bunge plant species was optimised. Different extraction media (methanol, ethanol and sulphuric acid), concentration and volume of sulphuric acid, pH of PA solution for alkaline extraction, extraction time and techniques (maceration, ultrasonic and overhead rotary mixer assisted extraction) were investigated. The yields of six PAs (7-angeloyl heliotridane, 7-angeloyl heliotridine, lindelofine, 7-angeloyl rinderine, punctanecine and heliosupine) were monitored by GC-MS/FID. The best results for the isolation all of six PAs were obtained when the extraction was performed with 1M sulphuric acid (30mL per 1.00g of dried sample) by overhead rotary mixer during three days. Optimal pH value for alkaline extraction of PAs with CH2Cl2 was 9, and the extraction should be performed with four portions of 30mL of CH2Cl2. This procedure could be also useful for a plant sample preparation for GC and LC analyses of PAs.

Different extraction media (methanol, ethanol and sulphuric acid), concentration and volume of sulphuric acid, pH of PA solution for alkaline extraction, extraction time and techniques (maceration, ultrasonic and overhead rotary mixer assisted extraction) were investigated. The yields of six PAs (7-angeloyl heliotridane, 7-angeloyl heliotridine, lindelofine, 7-angeloyl rinderine, punctanecine and heliosupine) were monitored by GC-MS/FID. The best results for the isolation all of six PAs were obtained when the extraction was performed with 1M sulphuric acid (30mL per 1.00g of dried sample) by overhead rotary mixer during three days. Optimal pH value for alkaline extraction of PAs with CH2Cl2 was 9, and the extraction should be performed with four portions of 30mL of CH2Cl2. This procedure could be also useful for a plant sample preparation for GC and LC analyses of PAs.",

Stone Fruit Wines as a Sources of Antioxidants

AB  - Introduction: The consumption of fruit and their product is
very important for balanced nutrition. They are rich source of natural
compounds which show beneficial health effects on human organism.
Fruit wines are rich sources of phenolic compounds which possess an-
tioxidant properties and play role in health protection from oxidative
stress.
Objectives: The aim of this study was to investigate the profile
and in vitro antioxidant properties of fruit wines.
Method / Design: Total polyphenol content was conducted by
using Folin-Ciocalteu method. Anioxidant capacity of samples was
determined by DPPH method, by calculating the percent of inhibition
of DPPH radical. Discoloration was measured on spectrofotometer at
518 nm. Also was used modern FRAP procedure for determination of
antioxidant properties of wine samples. For identification and quantification of some antioxidant compounds HPLC TQ-MS/MS method
was used.
Results: Wines were made from apricot, peach and plum in
winery according to the wine making procedure. From each kind of
fruit were made four different types of wine. All determinations were
conducted in these 12 samples of wine. The total polyphenol content
determined by Folin-Ciocalteu method was in range 358-1389 mg/L
expressed through concentration of gallic acid. The results of DPPH
analyze of samples were in range 35.69-68.50%. The results of antioxidant capacity analyzed by FRAP procedure were in range 21.75-45.15
mmol/Fe2+. Results of HPLC TQ-MS/MS analysis showed that our
samples are sources of compounds such as catechin, epicatechin and
phenolic acids. All those compounds have antioxidant properties.
Conclusions: The obtained results indicate that fruit wines are
sources of antioxidant compounds. Antioxidant properties of fruit
wines depend from which kind of fruit they were made. Natural compounds from fruit wines are particularly important in the prevention
of non-communicable disease caused by oxidative stress.

AB  - The examination of the aerial parts, roots, and seeds of the endemic plant Rindera umbellata is reported in this paper for the first time. Phytochemical investigation of R. umbellata led to the isolation and characterization of ten pyrrolizidine alkaloids and eleven fatty acids in the form of triglycerides. Pyrrolizidine alkaloids 1-9 were found in the aerial parts, 7 and 8 in the roots, and 6-10, together with eleven fatty acids, in the seeds of this plant species. The structures of compounds 1-10 were established based on spectroscopic studies (H-1- and C-13-NMR, 2D NMR, IR and CI-MS). After trans-esterification, methyl esters of the fatty acids were analyzed using GC-MS. The effect of lindelofine-N-oxide (7) on tubulin polymerization was determined.

Methylprednisolone and its related substances in freeze-dried powders for injections

AB  - In this work, the behavior of the active pharmaceutical substances methylprednisolone (in a form of methylprednisolone sodium succinate) in finished pharmaceutical dosage form, i.e., freeze-dried powder for injections, was examined. The goal was to evaluate the chemical stabilities of methylprednisolone sodium succinate packaged in a dual chamber vial, as a specific container closure system. The effect of different parameters: temperature, moisture and light were monitored. The method proposed by United States Pharmacopeia was used to determine concentrations of methylprednisolone, as the sum of the concentration of methylprednisolone esters (17-hydrogen succinate and 21-hydrogen succinate) and free methylprednisolone. The HPLC method was used for stability evaluation of the active substance and determination of related substances. Four main degradation products were registered. Temperature has a major impact on the degradation process with the appearance of 3 degradation products (impurities B, C and D), while the presence of light caused an increasing content of impurity A. Identification of impurity B, C and D has been realized using mass and NMR spectroscopy. All three substances are substances related to methylprednisolone.
AB  - U ovom radu ispitivane su osobine farmakološki aktivne supstance metilprednizolona (u obliku metilprednizolon-natrijum-sukcinata) u gotovom proizvodu - liofilizatu za rastvor za injekcije. Cilj rada je ispitivanje hemijske stabilnosti metilpredni-zolon-natrijum-sukcinata u dvokomornoj bočici, kao specifičnom sistemu kontaktnog pakovanja. Ispitan je efekat različitih parametara: temperature, vlage i svetlosti. Za određivanje koncentracije metilprednizolona, kao zbirne koncentracije metilprednizolon estara (17-hidrogen-sukcinata i 21-hidrogen-sukcinata) i slobodnog metilprednizolona, korišćena je metoda opisana u Američkoj farmakopeji. Za ispitivanje srodnih supstanci primenjena je HPLC metoda. Uočena su 4 degradaciona proizvoda. Dokazano je da povećanje temperature ima najveći značaj na proces degradacije i utiče na povećanje sadržaja nečistoća B, C i D, dok prisustvo svetlosti dovodi do povećanja sadržaja nečistoće A. Nečistoće B, C i D su identifikovane primenom masene i NMR spektroskopije. Sve tri nečistoće su identifikovane kao srodne supstance metilprednizolona.

Pyrrolizidine alkaloids from seven wild-growing Senecio species in Serbia and Montenegro

AB  - The genus Senecio (family Asteraceae) is one of the largest in the world. It comprises about 1100 species which are the rich source of pyrrolizidine alkaloids. Plants containing pyrrolizidine alkaloids are among the most important sources of human and animal exposure to plant toxins and carcinogens. The pyrrolizidine alkaloids of seven Senecio species (S. erucifolius, S. othonnae, S. wagneri, S. subalpinus, S. carpathicus, S. paludosus and S. rupestris) were studied. Fourteen alkaloids were isolated and their structures determined from spectroscopic data (1H- and 13C-NMR, IR and MS). Five of them were identified in S. erucifolius, four in S. othonnae, two in S. wagneri, four in S. subalpinus, two in S. carpathicus, three in S. paludosus and three in S. rupestris. Seven pyrrolizidine alkaloids were found for the first time in particular species. The results have chemotaxonomic importance. The cytotoxic activity and antimicrobial activity of some alkaloids were also studied.
AB  - Jedan od najbrojnijih rodova na svetu je rod Senecio (familija Asteraceae). Obuhvata oko 1100 vrsta koje predstavljaju bogat izvor pirolizidinskih alkaloida. Najznačajnija izloženost ljudi i životinja biljnim toksinima i karcinogenima potiče od biljaka koje sadrže pirolizidinske alkaloide. U ovom radu izolovani su pirolizidinski alkaloidi iz sedam vrsta roda Senecio (S. erucifolius, S. othonnae, S. wagneri, S. subalpinus, S. carpathicus, S. paludosus i S. rupestris), a njihove strukture određene na bazi spektroskopskih podataka (1H- i 13C-NMR, IR i MS). Izolovano je ukupno 14 alkaloida, pet iz S. erucifolius, četiri iz S. othonnae, dva iz S. wagneri, četiri iz S. subalpinus, dva iz S. carpathicus, tri iz S. paludosus i tri iz S. rupestris. Sedam pirolizidinskih alkaloida je po prvi put izolovano iz nekih pojedinačnih vrsta. Dobijeni rezultati imaju hemotaksonomski značaj. Ispitana je citotoksičnost i antimikrobna aktivnost izolovanih alkaloida.

Five of them were identified in S. erucifolius, four in S. othonnae, two in S. wagneri, four in S. subalpinus, two in S. carpathicus, three in S. paludosus and three in S. rupestris. Seven pyrrolizidine alkaloids were found for the first time in particular species. The results have chemotaxonomic importance. The cytotoxic activity and antimicrobial activity of some alkaloids were also studied., Jedan od najbrojnijih rodova na svetu je rod Senecio (familija Asteraceae). Obuhvata oko 1100 vrsta koje predstavljaju bogat izvor pirolizidinskih alkaloida. Najznačajnija izloženost ljudi i životinja biljnim toksinima i karcinogenima potiče od biljaka koje sadrže pirolizidinske alkaloide. U ovom radu izolovani su pirolizidinski alkaloidi iz sedam vrsta roda Senecio (S. erucifolius, S. othonnae, S. wagneri, S. subalpinus, S. carpathicus, S. paludosus i S. rupestris), a njihove strukture određene na bazi spektroskopskih podataka (1H- i 13C-NMR, IR i MS). Izolovano je ukupno 14 alkaloida, pet iz S. erucifolius, četiri iz S. othonnae, dva iz S. wagneri, četiri iz S. subalpinus, dva iz S. carpathicus, tri iz S. paludosus i tri iz S. rupestris. Sedam pirolizidinskih alkaloida je po prvi put izolovano iz nekih pojedinačnih vrsta. Dobijeni rezultati imaju hemotaksonomski značaj. Ispitana je citotoksičnost i antimikrobna aktivnost izolovanih alkaloida.",

AB  - The objective of the present study was to report the stability profile of novel antimigrain drug Eletriptan hydrobromide based on the information obtained from forced degradation studies. The drug was subjected to acid (0.1-1 mol L-1 HCl), neutral and base (0.1-1 mol L-1 NaOH) hydrolysis and to oxidative decomposition (3-15% (v/v) H2O2). Photolysis and thermo degradation at 75 degrees C were carried out in methanol solution and in solid state with both Eletriptan hydrobromide bulk drug and the tablet formulation. The products formed under different stress conditions were investigated by LC and LC-MS. The experimental conditions for LC were chosen by employing experimental design and multicriteria decision making methodology. These powerful tools enabled the accomplishment of satisfactory resolution with the shortest possible analysis time. Analytes were separated on a C-18 column (XTerra (TM), 150 mm x 3.9 mm, 5 mu m) with the mobile phase composed of methanol-water solution of TEA (pH 6.52, 1%, v/v) (30:70, v/v) pumped at 1 mL min(-1) flow rate. The column temperature was set at 50 degrees C and the detection at 225 nm using DAD detector. The LC method was suitably modified for LC-MS analysis which was further used to characterize the arisen degradation products. The possible degradation pathway was outlined based on the results. The drug appeared to be instable towards every stress condition but oxidation. The stability was not jeopardized even under more exaggerated conditions such as increased temperature of the solutions to 75 degrees C, increased strength of acid/alkali solutions and prolonged testing period. Validation of the LC-DAD method was carried out in accordance with ICH guideline. The method met all required criteria and was applied when testing the commercially available tablets.

AB  - The aporphine alkaloid magnoflorine, known as a constituent of Asian Epimedium species, was isolated for the first time from the wild growing European species, Epimedium alpinum L. Identification was done by comparison with literature data. The HPLC method was applied for the determination of the content of the alkaloid in methanolic extracts of plant material. The underground part of E. alpinum contains 1-2 % of magnoflorine, while its content in methanolic extracts was 9.2-11.8 %. The aerial part of the plant contains less alkaloid (0.06-0.12 %), while the magnoflorine content in methanolic extracts of the aerial part was 0.4-0.8 %. Probably, the high content of magnoflorine in the methanolic extracts of underground parts of E. alpinum influences its dose-dependent cytotoxic activity (300 μg/ml; 39 % survival of cells in an experimental K562 cell culture).

Ovo jedinjenje je korišćeno kao standard u HPLC analizi metanolnih ekstrakata E. alpinum. Sadržaj magnoflorina je znatno viši u ekstraktima podzemnih organa (9,2-11,8%, što odgovara sadržaju od 1-2 % magnoflorina u biljnom materijalu), nego u ekstraktima nadzemnih organa prevolca (0,4-0,8%,što odgovara sadržaju od 0,06-0,12%magnoflorina u biljnom materijalu). AB  - Tanacetum larvatum (Gris.) Kanitz. is an endemic perennial herb distributed on a rocky terrain in Serbia(Kosovo), Montenegro and Albania. The chemical composition of T larvatum essential oil, obtained from two locations in Montenegro (Mt. Komovi [Sample II] and Mt. Prokletije [Sample II]) was investigated. The oils obtained by hydrodistillation were analyzed by GC and GC/MS, on the two different capillary columns. The aerial parts of T. larvatum yielded 0.3% and 0.2% of a yellowish oil (Sample I and Sample II, respectively). About 40 compounds were identified, representing similar to 89% and 96% of the total oil content in the Samples I and II, respectively. tran's-Sabinyl acetate was found to be the dominant component (51.2% and 69.7%). Among the rest of compounds beta-pinene (7.7% and 4.3%) and camphor (6.3% and 4.3%) were the most abundant in both samples.

AB  - An investigation of the coumarin content of plant species Angelica silvestris (roots and fruits) and Peucedanum austriacum (Jacq.) Koch (roots), both belonging to the family Apiaceae is reported. A. silvestris contained only linear furocoumarins (four in the roots and five in the fruits), two of them (heraclenol and heraclenin) found for the first time in this species. The photoactive compound bergapten (also known as 5-methoxypsoralen) was also among the isolated coumarins. In P. austriacum only reduced (dihydro) coumarins were detected (four compunds): linear dihydrofurocoumarins, deltoin and sekorin, an angular dihydrofurocoumarin, zosimin and a linear dihydropyranoeoumarin, agasyllin. None of them has been found before in this plant species.
