AB  - Endokrini ometači, u koje se između ostalih ubrajaju i toksični metali poput kadmijuma (Cd), se dovode u vezu sa sve češćom pojavom neplodnosti i poremećajima reproduktivnog zdravlja. Kako je jedan od najznačajnijih mehanizama toksičnosti Cd njegova interakcija sa bioelementima, dalje ispitivanje ovog mehanizma moglo bi da doprinese rasvetljavanju mehanizama toksičnosti ovog metala u testisima, kao i uloge izloženosti ovom metalu u reproduktivnom zdravlju.
AB  - Endocrine disruptors, which include, but are not limited to, toxic metals such as Cd, have been linked to the increasing incidence of infertility and reproductive health disorders. As one of the most important mechanisms of Cd toxicity is its interaction with bioelements, further investigation of this mechanism could contribute to a better understanding of mechanisms of endocrine-disrupting Cd activities in testes and its role in human reproductive health.

AB  - The present study investigated the effects of PCBs on the rat kidneys with attention given to the determination critical effect dose (CED) using the Benchmark dose (BMD) approach. Male albino Wistar rats (7 animals per group) were given by oral gavage Aroclor 1254 dissolved in corn oil at doses of 0.0, 0.5, 1, 2, 4, 8, or 16 mg/kg b.w./day for 28 days. The PCB nephrotoxicity was manifested by a dose-dependent changes in serum urea levels. The study has also revealed PCB-induced oxidative stress induction in kidneys. The observed nephrotoxic effects can be partly explained by oxidative damage of lipids and proteins in the kidneys due to observed reduced CuZnSOD activity and disturbances in antioxidant protection. Аll the renal oxidative stress parameters showed dependence on PCB oral doses as well as internal, measure kidney PCB levels. Calculated BMDL values were lower than estimated no observed adverse effect levels (NOAEL) based on the study, suggesting the importance of BMD approach use in future risk assessment.

Influence of the mixture of Cd and BDE-209 on Zn and Cu homeostasis in rats

AB  - Humans are exposed to the chemicals from very different sources. Therefore, the
exposure assessment should be focused on mixtures rather than single chemicals. In this
study, bioelements (Zn and Cu) homeostasis effects of mixture of Cd and BDE-209, widely
spread persistent environmental pollutants, has been evaluated. In order to evaluate the
potential of mixture on Zn and Cu homeosthasis in rats kidneys, Wistar rats were treated by
oral gavage during 28 days with three doses of Cd 2.5, 7.5, and 15 mg/bw/day, three
combinations of Cd with 1000 mg BDE-209/kg/day, and water as a control group. Kidney
tissue samples were collected, mineralised, and Cu and Zn levels were measured by flame
atomic absorbtion spectrophotometer. Possible interactions were evaluated comparing the
dose-response curves slopes and lower confidence limit of Benchmark dose of 10%
(BMDL10) were calculated by PROAST software (RIVM, Netherlands). Cd alone can disturb
Zn in kidneys and calculated BMDL10 was 1.713 mg Cd/kg/day, while in combination with
BDE-209 it was 1.719 mg/kg/day. Cd disturbs Cu in kidney in significantly higher dose of
5.679 mg Cd/kg/day. However, in combination with the BDE-209, derived BMDL10 was 10
times lower, 0.517 mg Cd/kg/day. Dose-response curve slope for the effect on Cu was
significantly lower in the case of mixture effect than in the case of Cd effect. Since Cu plays a
major role in the blood cells creation and health of other systems, our results indicate more
potent activity of the mixture than single Cd.
AB  - Ljudi mogu biti izloženi hemikalijama iz različitih izvora, te je potrebno procenjivati
efekte na bazi izloženosti smešama hemikalija, pre nego pojedinačnim hemikalijama. U
studiji je analiziran uticaj smeše Cd i BDE-209, široko rasprostranjenih zagađujućih
perzistentnih supstanci, na homeostazu bioelemenata, cinka (Zn) i bakra (Cu). U cilju
evaluacije potencijala smeše da naruši homeostazu Zn i Cu u bubrezima pacova, Wistar
pacovi su oralnom gavažom bili tretirani tokom 28 dana dozama Cd od 2,5, 7,5 i 15
mg/kg/dan, kombinacijama ove tri doze sa 1000 mg BDE-209/kg/dan i vodom kao
kontrolom. Tkivo bubrega je izdvojeno, zatim mineralizovano, a sadržaj Zn i Cu je meren
atomskom apsorpcionom spektrofotometrijom. Moguće interakcije su procenjivane
poređenjem nagiba krivih doza-odgovor, a izračunate su i donje granice pouzdanosti
Benchmark doze od 10% (BMDL10) korišćenjem PROAST softvera (RIVM, Holandija). Cd
primenjen sam utiče na homeostazu Zn u bubregu i izračunata BMDL10 iznosi 1,713

AB  - The extent of human exposure to chemicals, their endocrine-disrupting mechanisms of action, and the relationship between exposure and various human diseases raise significant scientific and public health concerns. Polychlorinated biphenyls (PCBs) belong to the group of organic compounds known as persistent organic pollutants, characterized by long-range transport, persistence, bioaccumulation, and high toxicity. They have been identified as endocrine-disrupting chemicals. In vivo and in vitro studies have shown that endocrine-disrupting effects of PCBs mainly involve thyroid and reproductive function. In the review presented, these effects were placed in the context of the most recent findings on PCB-induced endocrine-disrupting mechanisms and modes of action.

AB  - Fully brominated diphenyl ether (BDE-209) is a flame retardant widely used in plastics and textiles. Because of its high persistence, humans are exposed to it continuously, mainly via dust ingestion. We investigated effects of BDE-209 on renal function and oxidative stress development in the kidney after subacute exposure in rats. Five groups of animals were given by oral gavage 31.25-500 mg BDE-209/kg b.w./day for 28 days, and relative kidney weight, serum urea and creatinine, and oxidative stress parameters in the kidney were determined. Benchmark-dose approach was used for dose response modeling. Serum creatinine was increased, while results obtained for serum urea were inconclusive. Relative kidney weight was not affected by BDE-209. Kidney reduced glutathione was elevated, while superoxide dismutase activity was not changed after BDE-209 treatment. Also, levels of thiobarbituric acid reactive substances (TBARS) were increased and total -SH groups were decreased, which indicated oxidative imbalance. The critical effect dose (CED)/CEDL ratios for the effects on TBARS and total -SH groups indicated estimated CEDs for these markers can be used in risk assessment of BDE-209. Our study results have shown that a relatively low dose of BDE-209 affects kidney function and that oxidative stress is one of the mechanisms of its nephrotoxicity.

AB  - The objective of this study was to assess toxicity of Cd and BDE-209 mixture on haematological parameters in subacutely exposed rats and to determine the presence and type of interactions between these two chemicals using multiple factorial regression analysis. Furthermore, for the assessment of interaction type, an isobologram based methodology was applied and compared with multiple factorial regression analysis. Chemicals were given by oral gavage to the male Wistar rats weighing 200-240g for 28 days. Animals were divided in 16 groups (8/group): control vehiculum group, three groups of rats were treated with 2.5, 7.5 or 15 mg Cd/kg/day. These doses were chosen on the bases of literature data and reflect relatively high Cd environmental exposure, three groups of rats were treated with 1000, 2000 or 4000 mg BDE-209/kg/bw/day, doses proved to induce toxic effects in rats. Furthermore, nine groups of animals were treated with different mixtures of Cd and BDE-209 containing doses of Cd and BDE-209 stated above. Blood samples were taken at the end of experiment and red blood cells, white blood cells and platelets counts were determined. For interaction assessment multiple factorial regression analysis and fitted isobologram approach were used. In this study, we focused on multiple factorial regression analysis as a method for interaction assessment. We also investigated the interactions between Cd and BDE-209 by the derived model for the description of the obtained fitted isobologram curves. Current study indicated that co-exposure to Cd and BDE-209 can result in significant decrease in RBC count, increase in WBC count and decrease in PLT count, when compared with controls. Multiple factorial regression analysis used for the assessment of interactions type between Cd and BDE-209 indicated synergism for the effect on RBC count and no interactions i.e. additivity for the effects on WBC and PLT counts. On the other hand, isobologram based approach showed slight antagonism for the effects on RBC and WBC while no interactions were proved for the joint effect on PLT count. These results confirm that the assessment of interactions between chemicals in the mixture greatly depends on the concept or method used for this evaluation.

AB  - It is known that kidneys are target tissue for Cd toxicity but there is not enough literature data on kidney effects of BDE-209. Moreover, Cd as toxic metal and BDE-209 as organic halogenated pollutant can share similar mechanism of action like, oxidative stress which is already assumed. However, how will Cd act as prooxidant in presence of BDE-209 is still not investigated, therefore, the objective of this study was to assess this BDE-209 influence. Results of this study indicated slight decrease in MDA in rat's kidney homogenates after subacute exposure to Cd and/or BDE-209, while SOD activity was increased and content of -SH groups decreased. Based on dose-response assessment, BDE-209 did not influence Cd toxicity mediated by chosen oxidative stress parameters, namely the derived CEDL5 values were the same for single Cd and for Cd given in combination with the BDE-209. In conclusion, dynamic of oxidative process has influence on the parameters levels. Co-exposure with polybrominated organic pollutant did not influence Cd effects on oxidative stress parameters. Having in mind that this is pilot study, performed with only one dose of BDE-209, further investigations are necessary, including changes in dose ranges, duration of exposure etc. Moreover, results contributes to the issue of mixture toxicology even BDE-209 did not influenced oxidative stress as mechanism of Cd toxicity.
AB  - Poznato je da su bubrezi ciljni organ toksičnosti kadmijuma, ali je mnogo manje literaturnih podataka o toksičnim efektima BDE-209 na bubrege. Kadmijum, kao toksičan metal i BDE-209, kao organska halogenovana zagađujuća supstanca mogu imati sličan mehanizam dejstva posredovan oksidativnim stresom. Međutim, kako će Cd da deluje kao prooksidant u prisustvu BDE-209 još uvek nije proučavano, te je cilj ovog rada bio da se proceni uticaj BDE-209 na oksidativni stres posredovan kadmijumom. Rezultati ovog istraživanja ukazuju na blagi pad koncentracije MDA u homogenatima bubrega pacova nakon subakutne ekspozicije Cd i/ili BDE-209, dok se aktivnost SOD povećava i sadržaj - SH grupa smanjuje. Na osnovu procene odnosa doza-odgovor, BDE-209 nije imao uticaja na toksičnost Cd izazvanu posredstvom oksidativnog stresa (posmatrajući odabrane parametre). Potvrda je dobijena i iz izračunatih donjih granica pouzdanosti doze od 5% - CEDL5 jer su vrednosti su bile iste za sam Cd i Cd primenjen u kombinaciji sa BDE-209. Može se zaključiti da dinamika oksidativnih procesa ima uticaj na vrednosti parametara oksidativnog stresa, da stoga istovremena ekspozicija Cd i polibromovanom organskom polutantu nije promenila uticaj samog Cd na parametre oksidativnog stresa. Imajući u vidu da je ovo pilot studija, izvršena sa samo jednom dozom BDE-209, potrebna su dalja istraživanja, uključujući i promene opsega doza, trajanje ekspozicije itd. Pored toga, rezultati ove studije doprinose tumačenju toksikologija smeša, iako BDE-209 nije imao uticaja na oksidativni stres kao mehanizam Cd toksičnosti za date uslove ekspozicije.

Može se zaključiti da dinamika oksidativnih procesa ima uticaj na vrednosti parametara oksidativnog stresa, da stoga istovremena ekspozicija Cd i polibromovanom organskom polutantu nije promenila uticaj samog Cd na parametre oksidativnog stresa. Imajući u vidu da je ovo pilot studija, izvršena sa samo jednom dozom BDE-209, potrebna su dalja istraživanja, uključujući i promene opsega doza, trajanje ekspozicije itd. Pored toga, rezultati ove studije doprinose tumačenju toksikologija smeša, iako BDE-209 nije imao uticaja na oksidativni stres kao mehanizam Cd toksičnosti za date uslove ekspozicije.",

AB  - Hepatotoxicity is one of the well-documented adverse health effects of polychlorinated biphenyls (PCBs)persistent organic pollutants widely present in the environment. Although previous studies suggest possible role of oxidative stress, the precise mechanisms of PCB-induced ROS production in liver still remain to be fully assessed. The aim of this study was to evaluate the effects of different doses of PCBs on the parameters of oxidative stress and to investigate whether these effects are dose dependent. Furthermore, a comparison between calculated benchmark doses (BMD) and estimated NOAEL values for investigated parameters, was made. Six groups of male albino Wistar rats (7 animals per group) were receiving Aroclor 1254 dissolved in corn oil in the doses of 0.5, 1, 2, 4, 8, 16 mg PCBs/kg b.w./day by oral gavage during 28 days while control animals were receiving corn oil only. The following parameters of oxidative stress were analyzed in liver homogenates: superoxide dismutase activity, glutathione, malondialdehyde (MDA) and total protein thiol levels. Hepatic enzymes AST, ALT, ALP and protein albumin were also determined in serum as clinical parameters of liver function. Collected data on the investigated parameters were analyzed by the BMD method. The results of this study demonstrate that subacute exposure to PCBs causes induction of oxidative stress in liver with dose-dependent changes of the investigated parameters, although more pronounced adverse effects were observed on enzymatic than on non-enzymatic components of antioxidant protection. The obtained values for BMD and NOAEL support the use of BMD concept in the prediction of health risks associated with PCBs exposure. Furthermore, our results implicate possible use of MDA in PCBs risk assessment, since MDA was the most sensitive investigated parameter with calculated low critical effect dose of 0.07 mg/kg b.w.

AB  - Endocrine-disrupting chemicals currently represent one of the major concerns and this study was aimed to investigate the effects of different doses of cadmium, widespread toxic metal, on the levels of thyroid hormones and to calculate Benchmark doses for these effects. Furthermore, the effects of co-exposure to cadmium and polychlorinated biphenyls on thyroid function were investigated. Six orally-treated groups of rats were receiving 0.3, 0.6, 1.25, 2.5, 5 and 10 mg Cd/kg b.w./day, five groups were orally treated with 0.5, 1, 2, 4 and 8 mg PCBs/kgb.w./day, while nine groups of rats were orally-treated with different dose combinations of Cd and PCBs (0.6, 1.25 and 2.5 mg Cd/kgb.w. and 2,4 and 8 mg PCBs/kg b.w./day), during 28 days. Thyroid hormones were adversely affected by cadmium, with most prominent effect observed on triiodothyroxine levels indicating Cd interference with thyroid function at extrathyroidal level. Calculated Benchmark doses for Cd effects on thyroid hormones indicate triiodothyroxine as the most sensitive one that can be used as a basis for risk assessment. This study also implicates possible synergistic effects of Cd and PCBs on thyroid function as a consequence of their interference at different levels of thyroid homeostasis.

Toxicity mechanisms and interactions of polychlorinated biphenyls and polybrominated diphenylethers

AB  - Polychlorinated biphenyls (PCBs) and brominated flame retardants, polybrominated diphenylethers (PBDEs), are widespread environmental contaminants as a result of anthropogenic activities and due to their persistency and resistance to degradation. Both groups of chemicals are placed on the list of persistent organic pollutants (POPs), covered by the Stockholm convention which is aimed to limit or ban the production, use, emission, import and export of POPs in order to protect human health and the environment. Taking into account the structural similarity between PCBs and PBDEs, and the known effects of PCBs, these two groups of chemicals could have similar mechanisms of action. Also, because of real simultaneous exposure to these compounds, an examination of possible interactions is of great importance. Interactions of the compounds from these groups are likely at the system level of hormones, particularly thyroid and reproductive ones. As there are mechanisms of adverse effects of both groups of chemicals on the nervous system, including functional, neurological and behavioral changes, there are influences on neurotransmiters, changes in signal transduction and apoptosis. There are interactions affecting the occurrence of metabolic disorders as well. Data on the toxicity of mixtures of PCBs and PBDEs, as well as interactions of these chemicals would contribute to the processes of risk evaluation and risk characterisation.
AB  - Polihlorovani bifenili (PCBs) i bromovani usporivači gorenja, polibromovani difenil etri (PBDEs) su široko rasprostranjeni u životnoj sredini kao posledica antropogenih aktivnosti i zahvaljujući njihovoj postojanosti i otpornosti na degradaciju. Obe grupe jedinjenja se nalaze na listi perzistentnih organskih zagađivača (POPs) usvojenoj Stokholmskom konvencijom sa ciljem da zabrani ili ograniči proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs u svrhu zaštite zdravlja ljudi i životne sredine. Uzimajući u obzir strukturnu sličnost između PBDEs i PCBs, kao i činjenicu da je istovremena ekspozicija ovim jedinjenima realna, ispitivanja njihovih interakcija su od značaja za predviđanje rezultujućeg efekta. Saznanja u oblasti toksikologije smeša, takođe doprinose procesu evaluacije i karakterizacije rizika. Interakcije jedinjenja iz ovih grupa su verovatne na nivou endokrinog sistema, naročito tiroidnog i reproduktivnog. S obzirom na mehanizme toksičnosti obe grupe supstanci za nervni sistem, uključujući funkcionalne promene, neurološke poremećaje i promene u ponašanju, moguće je očekivati njihove interakcije i na sistemima neurotransmitera, promene u prenosu signala i izazivanje apoptoze, kao i na pojavu metaboličkih poremećaja.

Determination of morphine, codeine and 6-monoacetylmorphine in saliva of substance-abuse patients using HPLC/MS methods

AB  - Background/Aim. Saliva represents an alternative specimen for substances abuse determination in toxicology. Hence, the aim of this study was to optimize a method for saliva specimen preparation for heroin metabolites, morphine and 6-monoacetylmorphine (6-mam), and codeine determination by liquid chromatography-mass spectrometry (LC/MS), and to apply this method on saliva samples taken from the patients. Methods. Saliva specimen was prepared using liqiud/liquid extraction of morphine, codeine and 6- mam by mixture of chloroform and isopropanol (9 : 1; v/v). Extracts were analysed by HPLC/MS technique: separation column Waters Spherisorb® 5 μm, ODS2, 4.6 × 100 mm; mobile phase: ammonium acetate : acetonitile (80 : 20; v/v), mobile phase flow rate 0.3 mL/min; mass detection range: 100-400 m/z. Regression and correlation analyses were performed with the probalility level of 0.05. Concentrations of morphine, codeine and 6-mam were determined in saliva samples of the patients with 'opiates' in urine identified by the test strips. Results. Calibration for each analysed substance was done in the concentration range from 0.1 to 1 mg/L and the coefficient of correlation was R2 > 0.99. We obtained following calibration curves: y = 385531x + 14584; y = 398036x + 31542; and y = 524162x - 27105, for morphine, codeine and 6-mam, respectively. Recovery for morphine and codeine determination was 99%, while for 6- mam it was 94%. Limits of detection and quantification of a proposed method were 0.01 mg/L and 0.05 mg/L, respectively. Concentration of morphine in the saliva of the heroin users ranged between 0.54 and 5.82 mg/L, concentration of codeine between 0.05 and 5.33, and 6-mam between 0.01 and 0.68 mg/L. A statistically significant correlation between codeine and 6-mam concentrations was obtained. Conclusion. A proposed HPLC/MS method for morphine, codeine and 6-mam determination in saliva is accurate, simple, cheap and suitable for routine analysis and monitoring of heroin abuse.
AB  - Uvod/Cilj. Saliva predstavlja alternativni matriks za identifikaciju sredstava zloupotrebe. Cilj ovog rada bio je optimizacija metode pripreme uzorka salive i određivanja metabolita heroina, morfina i 6-monoacetilmorfina (6-mam), i kodeina liquid chromatography-mass spectrometry (LC/MS) metodom i provera metode u realnim uslovima kod heroinomana. Metode. Priprema uzoraka vršena je tečno-tečnom ekstrakcijom uz smešu hloroforma i izopropil alkohola u odnosu 9 : 1. Ekstrakti su analizirani tehnikom HPLC/MS: razdvajanje na koloni Waters Spherisorb® 5 μm, ODS2, 4,6 × 100 mm, vršeno je primenom mobilne faze amonijum-acetat : acetonitril u odnosu 80 : 20 pri protoku od 0,3 mL/min. Masena detekcija je vršena u opsegu masa od 100 do 400 m/z. Primenjene su regresiona i korelaciona analiza za nivo verovatnoće 0,05. Određivanje prisustva morfina, kodeina i 6-mam vršeno je u uzorcima salive kod osoba kod kojih je test trakama utvrđeno prisustvo 'opijata' u urinu. Rezultati. Kalibracija je vršena u opsegu koncentracija 0,1-1 mg/L sa koeficijentom determinacije R2 > 0,99. Dobijene su kalibracione krive: za morfin, y = 385531x + 14584; kodein, y = 398036x + 31542 i 6- monoacetilmorfin, y = 524162x - 27105.
