Antioxidant and antimicrobial activity of a new generation phyto-gel

AB  - Antioxidant and antimicrobial activities of a new phyto-gel were studied in this work. The formulated gel is composed of propilene glycol extracts of St. John's worth (H. perforatum L.), comfrey (S. officinale L.), yarrow (A. millefolium L.), horsetail (E. arvense L.) and Na-usniate. The antioxidant activity was determined for the gel and each investigated extract of the gel separately by the DPPH test. The total phenols and flavonoids content was determined spectrophotometrically, for the gel and each investigated extracts. The antimicrobial activity of the gel and each compound separately was tested in vitro on the following microorganisms: Staphylococcus aureus, Micrococcus luteus, Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger. The antioxidant activity of the investigated samples decreases in the series: H. perforatum (EC50 = 1.04 mg/mL) > Na-usniate (EC50 = 4.16 mg/mL) > A. millefolium (EC50 = 7.60 mg/mL) > gel (EC50 = 23.47 mg/mL) > S. officinale (EC50 = 30.10 mg/mL) > E. arvense (EC50 = 44.70 mg/mL). The formulated phyto-gel showed the antimicrobial activity. The results suggest that the obtained phyto-gel is a good source of natural antioxidants and antimicrobial agents, and it has potential as an alternative to synthetic products in pharmaceutical and cosmetic industries.
AB  - U radu je ispitivana antioksidativna i antimikrobna aktivnost fitogela - formulacija gela sa propilen-glikolnim ekstraktima H. perforatum, S. officinale, A. millefolium, E. arvense i Na-usninatom. Primenom DPPH testa određena je antioksidativna aktivnost svih ispitivanih ekstrakata, Na-usninata i gela. Primenom spektrofotometrijskih metoda određen je sadržaj ukupnih fenola i flavonoida u ekstraktima i formulisanom gelu. Antimikrobna aktivnost gela, kao i njegovih pojedinačnih komponenata, je ispitivana in vitro na sledećim mikroorganizmima: Staphylococcus aureus, Micrococcus luteus, Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Candida albicans i Aspergillus niger. Application of mathematical modeling for the development and optimization formulation with bioactive copper complex

AB  - New formulation for treatment a copper deficiency in human organism was developed and optimized by application of mathematical modeling. This formulation contained copper (II) complex with polysaccharide pullulan, as active substance. The binder concentration [polyvinyl pyrrolidone (PVP %)], the disintegrant concentration (corn starch %) and the resistance to crushing (hardness) were taken as independent variables. In vitro measured drug release characteristics of the tablets at pH 1.20 and 7.56 were studied as response variables. Initially, the created full factorial 2(3) model showed that the resistance to crushing has the most significant effect on copper (II) complex release from the formulation. Optimal tablet formulation F2, with lower Hardness (50 N), lower Starch (20.0%) and higher PVP (2.7%) concentrations, is selected using the partial least squares (PLS) regression modeling. The selected formulation F2 has expressed the best drug release profile at both pH (98.66% pH = 1.20; 93.35% pH = 7.56), and the lowest variation of tablets weight. The presented theoretical approach and created PLS model can be readily applied in future copper complexes studies and formulation design.

Inclusion complexes with cyclodextrin and usnic acid

AB  - Molecular inclusion complexes of usnic acid (UA) with beta-cyclodextrin (beta-CD) and 2-hydroxypropyl beta-cyclodextrin (HP beta-CD) were prepared by the co-precipitation method in the solid state in the molar ratio of 1:1. Structural complexes characterization was based on different methods, FTIR, H-1 NMR, XRD and DSC. Parallel to the complex by the above methods, corresponding physical mixtures of UA with cyclodextrins and complexing agents (beta-CD, HP beta-CD and UA) were analyzed. The results of DSC analysis showed that, at around 200 A degrees C, the endothermal peak in the complexes with cyclodextrins originating from the UA melting has disappeared. Complex diffractogram patterns do not contain peaks characteristic for the pure UA. They are more appropriate to cyclodextrin diffractogram. This fact points to the molecular encapsulation of UA in the cyclodextrin cavity. Chemical shifts in H-1 NMR spectra after the inclusion of UA into the cyclodextrin cavity, especially H-3 protons (0.0012 and 0.0102 ppm in the beta-CD and HP beta-CD, respectively) and H-5 and H-6 (0.0134 ppm) and hydrogen from CH3 (0.0073 ppm) HP beta-CD also points to the formation of molecular inclusion complexes. The improved solubility of UA in water was achieved by molecular incapsulation. In the complex with beta-CD the solubility is 0.3 mg/cm(3), with HP beta-CD 4.2 mg/cm(3) while the uncomplexed UA solubility is 0.06 mg/cm(3). The microbial activity of UA and both complexes was tested against eight bacteria and two fungi and during the test no reduced activity of UA in the complexes was observed.

The new no-veneered tablets, prepared by a wet granulation technique, are containg active substance, a copper(II) complex with polysaccharide pullulan. The binder concentration, the disintegrant concentration and the resistance to crushing were used as independent variables in the formulation; while in vitro measured drug release characteristics of the tablets was response variable in a full factorial design 2(3) modeling. A cubic model for data fitted was used to examine the obtained results. They showed that the resistance to crushing has the most significant effect on copper(II) complex release from the formulation, while the disintegrant concentration has smaller influence on dissolution profile of copper(II) complex and the binder concentration had minor impact in this study. Lower value of resistance to crushing has influence on better dissolution profile. Furthermore, physical characteristics of the tablets were evaluated, viz., drug content, hardness, thickness, friability, disintegration time, mass variation, particle size and size distribution.

Treatment of Venous Ulcers with the Herbal-Based Ointment Herbadermal (R): A Prospective Non-Randomized Pilot Study

AB  - Background: Venous ulcers (ulcus cruris venosum) are a common chronic disease that requires continuing treatment and significantly influences a patient's way of life. The therapeutic effects of the ointment Herbadermal on epithelialization and microbial flora of venous ulcers in 25 patients (10 men and 15 women) were tested over a 7-week period. The major components of the ointment are extracts of garlic, St. John's wort, and calendula. Material and Methods: The patients were over 18 years of age, with ulceration of the lower leg no longer than 2 months or recurrent ulceration during the last 6 months. The involved patients did not use any other phytomedicines or supportive therapies. Parameters were evaluated before the treatment and every 2 weeks during the period of 7 weeks. The total treatment response was evaluated on the basis of epithelialization, granulation, fibrin deposits, exudation, and edema. Results: The percentage of epithelialization was 99.1% after 7 weeks, without significant effects on the microbial flora. Conclusion: This combination of extracts can be recommended as topical treatment for wound healing because of its epithelizing, anti-erythematous, and anti-edematous properties.

The therapeutic effects of the ointment Herbadermal on epithelialization and microbial flora of venous ulcers in 25 patients (10 men and 15 women) were tested over a 7-week period. The major components of the ointment are extracts of garlic, St. John's wort, and calendula. Material and Methods: The patients were over 18 years of age, with ulceration of the lower leg no longer than 2 months or recurrent ulceration during the last 6 months. The involved patients did not use any other phytomedicines or supportive therapies. Parameters were evaluated before the treatment and every 2 weeks during the period of 7 weeks. The total treatment response was evaluated on the basis of epithelialization, granulation, fibrin deposits, exudation, and edema. Results: The percentage of epithelialization was 99.1% after 7 weeks, without significant effects on the microbial flora. Conclusion: This combination of extracts can be recommended as topical treatment for wound healing because of its epithelizing, anti-erythematous, and anti-edematous properties.",

AB  - According to the tested antimicrobial activity of constituents and their chemical characteristics, a new formulation of oral antiseptic was made based on garlic bulb powder and its active principles with strong antimicrobial activity against a wide range of bacteria and fungi. The antimicrobial activity of garlic bulb powder, allicin and the lozenge with 15% of garlic powder was tested using broth microdilution method. The tested garlic powder, as well the lozenge, have shown very high antimicrobial activity with MIC 1.25-5.00 mg/ml, and 1.87-7.50 mg/ml, respectively. The major compound, allicin, was highly active at a very low concentration. The minimal inhibitory concentration of allicin was from 6.25-12.50 μg/ml for antibacterial activity and 0.4 μg/ml for antifungal activity. Those concentrations are comparable with concentrations of commercially available antibiotics and fungicides. The formulation of an antiseptic with herbal constituents, high antibacterial and antifungal activity and pleasant taste could be an alternative to classical pharmaceutical oral antiseptics.
AB  - Na osnovu ispitane antimikrobne aktivnosti komponenata i njihovih hemijskih karakteristika, napravljena je nova formulacija oralnog antiseptika koji kao aktivnu komponentu ima prašak belog luka čiji sastojci pokazuju veoma izraženu aktivnost protiv širokog spektra bakterija i gljive Candida albicans. Antimikrobna aktivnost praška belog luka, alicina i lozengi sa 15% praška belog luka je ispitivana bujon mikrodilucionom metodom. Testiran prašak belog luka, kao i lozenge imaju veoma jaku antimikrobnu aktivnost sa minimalnim inhibitornim koncentracijama (MIC) 1,25-5,00 mg/ml (prašak belog luka) i 1,87-7,50 mg/ml (lozenge). Glavna aktivna komponeta belog luka, alicin, pokazuje antimikrobnu aktivnost u veoma niskim koncentracijama. Minimalne inhibitorne koncentracije alicina se kreću od 6,25-12,5 μg/ml protiv bakterija i 0,4 μg/ml protiv gljivice Candida albicans. Ove koncentracije se mogu uporediti sa koncentracijama komercijalnih antibiotika i fungicida. Naši rezultati su u skladu sa prethodnim ispitivanjima alicina i različitih ekstrakta belog luka, i potvrđuju najvažniju ulogu alicina u antimikrobnoj aktivnosti. Veoma je važna visoka aktivnost ovako formulisanog oralnog preparata protiv gljivice C. albicans koja je čest uzročnik infekcija usne duplje. Formulisan proizvod sa antiseptičkim osobinama karakteriše stanje čvrstog rastvora gde su aktivne supstance molekularno-disperzno ugrađene u obliku dobro rastvornog polimernog matriksa (sorbitola) velike specifične površine. Dobra rastvorljivost polimernog matriksa i velika specifična površina omogućuju zadovoljavajuću brzinu otpuštanja aktivnih principa iz belog luka (alicin). Pored visoke antimikrobne aktivnosti, lozenge poseduju prijatan ukus i osvežavaju dah, pa se mogu koristiti kao prirodni antiseptik u prevenciji i terapiji infekcija sluzokože usta i ždrela. Formulacija antiseptika sa biljnim sastojcima i visokom antibakterijskom i antifungalnom aktivnošću može biti alternativa farmaceutskim oralnim antisepticima.

Composition of Satureja kitaibelii Essential Oil and its Antimicrobial Activity

AB  - The composition of the essential oil obtained by hydrodistillation from the aerial parts of Satureja kitaibelii from Rtanj mountain (Serbia), collected during three years, was studied. Thirty-nine components were identified in each sample of S. kitaibelii essential oil, representing about 87% of the oils. p-Cymene was the most dominant compound in all three oils (27.9%, 14.7% and 24.4%, respectively). The simple formulation of a lozenge with 0.2% of S. kitaibelii essential oil was prepared and the antimicrobial activity of the essential oil and the lozenge with essential oil was tested using a broth microdilution assay. Both essential oil and lozenge possessed strong antimicrobial activity with MIC values of 0.10-25 mu g/mL, and 0.97-15.6 mg/mL, respectively.
