Analysis of vitamin content in products intended for infants and children on the Serbian market

AB  - Infant formula and follow-on formula, baby food and food supplements are important sources of essential nutrients for infants and young children. It is therefore important to monitor the nutrient content of these products. The content of vitamins A, D, E, C and B group was determined in 121 samples (44 samples of infant and follow-on formulas, baby foods, foods for special medical purposes and 77 of food supplements for children. High Performance Liquid Chromatography with Fluorescence and Diode Array Detector (HPLC-FLD-DAD) methods, were used for analysis. Among these samples, almost every third sample was outside of the defined tolerance range. The biggest deviations were found for vitamins A and E. The obtained values in foods for infants and young children for vitamins A and E was 17–147 % and 24–140 % of the declared value, respectively, whereas for food supplements, they were 5–150 % and 52–205 %, respectively. The highest values for vitamin E were in instant drinks (80–203 % of the declared value). The content of vitamins D and B2 in food supplements in liquid form was 10–127 % and 11–110 % of the declared value, respectively. Considering that these foods are often the only source of essential nutrients for infants and children, these results show the need for regular monitoring of the composition of these products.

Generic approach in a gradient elution HPLC method development that enables troubleshooting free method transfer

AB  - Nowadays, method development is strongly focused on reducing time needed for method development and execution. This subject specially concerns gradient elution methods regarding the usual need for trouble shooting assistance with uncertain outcome during the method transfer from one laboratory to another. One of the main reasons for this situation is the dwell volume difference between HPLC systems. Therefore, the aim of this study was to propose a novel method development methodology that would integrate the dwell volumes differences in the optimization process. The proposed approach could be quite useful in industry that has insight in HPLC instruments planned to be used during the method life cycle. It was tested on the model mixture consisting of dabigatran etexilate mesylate and its nine impurities by use of perimental design methodology. Three different (U)HPLC instruments with high dwell volume differences were selected to challenge the methodology. Plan of experiments was defined with Plackett-Burman design for screening phase and D-optimal design for optimization phase. Initial and final amount of organic modifier, time of the gradient elution and pH value of the aqueous phase were selected as variables nificant for the gradient programme profile and included in the optimization stage along with dwell volume values. The separation criteria s between critical peak pairs was selected as output for method optimization while indirect modelling together with Monte Carlo simulations enabled selection of optimal and robust chromatographic conditions. They included 24% (v/v) of initial amount of acetonitrile, 54% (v/v) of the final amount of acetonitrile, 15 min of gradient elution run time and pH value equal to 4.9. The proposed method was successfully validated, met all validation criteria and thus proved its utility.

AB  - Phthalates are commonly used plasticizers for plastic materials. Number of toys are
made from plastic materials or are coated with plastic layer. No existence of chemical bound
between phthalates and polymer layers means their easy migration. Children’s exposure to
phthalates is recognised as a threat to their health, since phthalates interfere with hormones
level (endocrine disrupter chemicals – EDC), but can also induce hepatotoxicity, renal
toxicity, allergic diseases and astma and disrupt thyroid function. Rulebook on Toy safety
(“Official Gazette of RS” No. 78/2019) was published in 2019, to set rules on toys safety and
their free movement on the market. Phthalates are prohibited, individually or in combination
in a concentration equal to or greater than 0.1% (m/m). Butylbenzyl phthalate (BBP), Di-n-
butyl phthalate (DBP), Bis(2-ethylhexyl) phthalate (DEHP) and Diisobutyl phthalate (DIBP)
are listed as prohibited, but DIBP restrictions start from July 2023 (1). In accordance with
this legislative, laboratory of Institute of Public Health of Serbia “Dr Milan Jovanovic Batut”
analyses part of these toys present on the market in Serbia. Total of 232 toy samples were
analysed in the last 5 years (2017-2021), by HPLC-DAD. Phthalates are found in 40 samples
(17.2%), 10 samples (4.3%) were bellow and 30 (12.9%) exceeded concentration of 0.1%
(m/m). The most common elevated phthalates were DEHP and DBP. During 2019 and 2021
no phthalates are found in samples. Constant appearance of new toy manufacturers and
control of existing ones demand monitoring of toys present on RS market, for the purpose of
children's safety.
AB  - Ftalati spadaju u najčešće korišćene plastifikatore za plastične materijale. Veliki broj
igračaka izrađen je od plastike ili je plastificiran. Usled nepostojanja hemijske veze između
ftalata i slojeva polimera, lako može doći do migracije ftalata. Izloženost dece ftalatima
prepoznata je kao pretnja po zdravlje, budući da su ftalati klasifikovani u grupu jedinjenja
endokrinih ometača (EDC), pošto utiču na promene nivoa hormona, ali takođe mogu dovesti i
do oštećenja jetre, bubrega, poremećaja rada štitaste žlezde, alergijskih bolesti i astme.
Pravilnik o bezbednosti igračaka Republike Srbije („Službeni glasnik RS” broj 78/2019)
donešen je u cilju uspostavljanja pravila o bezbednosti igračaka i njihovom slobodnom
kretanju na tržištu. Upotreba ftalata kao plastifikatora zabranjena je, pojedinačno ili u smeši,
ako je koncentracija jednaka ili veća od 0,1% (m/m). Na listi zabranjenih ftalata su:
butilbenzil ftalat (BBP), di-n-butil ftalat (DBP), di-2-etilheksil ftalat (DEHP), kao i di-izo-butil
ftalat (DIBP) čija zabrana stupa na snagu od jula 2023. godine (1). U skladu sa postojećim
propisima, laboratorija Instituta za javno zdravlje Srbije „Dr Milan Jovanović Batut“ analizira
deo igračaka prisutnih na tržištu u Srbiji. Tokom prethodnih 5 godina (2017-2021)
analizirano je ukupno 232 uzoraka igračaka. Ftalati su nađeni u 40 uzoraka (17,2%), od kojih
je 10 uzoraka (4,3%) imalo ispod, a 30 (12,9%) preko granične koncentracije od 0,1%
(m/m). Najčešće povišeni ftalati su DEHP i DBP. Tokom 2019. i 2021. godine u uzorcima nije
utvrđeno prisustvo ftalata. Usled stalne pojave novih proizvođača igračaka, kao i kontrole
već prisutnih, neophodan je redovan monitoring igračaka na tržištu RS radi bezbednosti
dece.

(“Official Gazette of RS” No. 78/2019) was published in 2019, to set rules on toys safety and
their free movement on the market. Phthalates are prohibited, individually or in combination
in a concentration equal to or greater than 0.1% (m/m). Butylbenzyl phthalate (BBP), Di-n-
butyl phthalate (DBP), Bis(2-ethylhexyl) phthalate (DEHP) and Diisobutyl phthalate (DIBP)
are listed as prohibited, but DIBP restrictions start from July 2023 (1). In accordance with
this legislative, laboratory of Institute of Public Health of Serbia “Dr Milan Jovanovic Batut”
analyses part of these toys present on the market in Serbia. Total of 232 toy samples were
analysed in the last 5 years (2017-2021), by HPLC-DAD. Phthalates are found in 40 samples
(17.2%), 10 samples (4.3%) were bellow and 30 (12.9%) exceeded concentration of 0.1%
(m/m). The most common elevated phthalates were DEHP and DBP. During 2019 and 2021
no phthalates are found in samples. Constant appearance of new toy manufacturers and
control of existing ones demand monitoring of toys present on RS market, for the purpose of
children's safety., Ftalati spadaju u najčešće korišćene plastifikatore za plastične materijale. Veliki broj
igračaka izrađen je od plastike ili je plastificiran. Usled nepostojanja hemijske veze između
ftalata i slojeva polimera, lako može doći do migracije ftalata. Izloženost dece ftalatima
prepoznata je kao pretnja po zdravlje, budući da su ftalati klasifikovani u grupu jedinjenja
endokrinih ometača (EDC), pošto utiču na promene nivoa hormona, ali takođe mogu dovesti i
do oštećenja jetre, bubrega, poremećaja rada štitaste žlezde, alergijskih bolesti i astme.
Pravilnik o bezbednosti igračaka Republike Srbije („Službeni glasnik RS” broj 78/2019)
donešen je u cilju uspostavljanja pravila o bezbednosti igračaka i njihovom slobodnom
kretanju na tržištu. Upotreba ftalata kao plastifikatora zabranjena je, pojedinačno ili u smeši,
ako je koncentracija jednaka ili veća od 0,1% (m/m). Na listi zabranjenih ftalata su:
butilbenzil ftalat (BBP), di-n-butil ftalat (DBP), di-2-etilheksil ftalat (DEHP), kao i di-izo-butil
ftalat (DIBP) čija zabrana stupa na snagu od jula 2023. godine (1). U skladu sa postojećim
propisima, laboratorija Instituta za javno zdravlje Srbije „Dr Milan Jovanović Batut“ analizira
deo igračaka prisutnih na tržištu u Srbiji. Tokom prethodnih 5 godina (2017-2021)
analizirano je ukupno 232 uzoraka igračaka. Ftalati su nađeni u 40 uzoraka (17,2%), od kojih
je 10 uzoraka (4,3%) imalo ispod, a 30 (12,9%) preko granične koncentracije od 0,1%
(m/m). Najčešće povišeni ftalati su DEHP i DBP. Tokom 2019. i 2021. godine u uzorcima nije
utvrđeno prisustvo ftalata. Usled stalne pojave novih proizvođača igračaka, kao i kontrole
već prisutnih, neophodan je redovan monitoring igračaka na tržištu RS radi bezbednosti

AB  - Nowadays, one of the ultimate requirements in drug analysis is rapid method
development, prompt chromatographic run time and long lasting method’s lifecycle. Special
attention should be paid to development of gradient elution (U)HPLC methods that are
commonly related to troubleshooting during the inter-laboratory method transfer. The dwell
volume difference between (U)HPLC systems is the main reason for this issue (1). The aim of
this study was to propose new gradient method development methodology with integrated
dwell volumes values in the optimization process. This is especially useful approach for
industry where is frequently known which (U)HPLC instruments will be applied for drug
analysis. Proposed methodology was tested on the model mixture which encompassed
dabigatran etexilate mesylate and its nine impurities. Experimental design methodology and
three different (U)HPLC instruments with significant dwell volume differences were utilized.
Statistically significant variables were selected with Plackett-Burman design, and along with
dwell volume values were included in D-optimal design. The separation criteria s between
adjacent peaks was selected as output for method optimization. Indirect modelling together
with Monte Carlo simulations enabled selection of optimal and robust chromatographic
conditions joint for all instruments. The optimal conditions included 24% (v/v) of initial
amount of acetonitrile, 54% (v/v) of the final amount of acetonitrile, 15 min of gradient
elution run time and pH value equal to 4.9. The proposed method utility was proved since it
was successfully validated and met all validation criteria (2).
AB  - U današnje vreme, jedan od važnih zahteva prilikom razvoja hromatografskih metoda
jeste da se razvoj uradi brzo, da vreme trajanja hromatografske analize bude kratko i da se
obezbedi dug životni ciklus metode. Posebnu pažnju treba obratiti na (U)HPLC metode sa
gradijentnim eluiranjem kod kojih se često javljaju problemi prilikom međulaboratorijskog
transfera. Glavni razlog za zahtevan transfer gradijentnih (U)HPLC metoda je razlika u
vrednostima dwell volume između (U)HPLC uređaja (1). Cilj ovog rada je bio da predloži
novu metodologiju razvoja gradijentnih (U)HPLC metoda, pri čemu će vrednosti dwell
volumes biti integrisane sa drugim ulaznim promenljivama u fazi optimizacije. Ovo bi bio
posebno koristan pristup u industriji gde se obično zna na kojim će sve (U)HPLC
instrumentima metoda biti korišćena. Metodologija je testirana na smeši dabigatran eteksilat

AB  - Introduction: Gradient elution HPLC finds its
purpose in simultaneous analyses of solutes
covering wide range of polarities. However, the
instrument related factors, especially dwell volume, are frequently responsible for fizzy transfer and
short life cycle of the gradient elution method.
Therefore, it is advisable to incorporate dwell
volume into the optimization stage and avoid
transfer related failures. The chemometric
approach would enable selection of optimal
chromatographic conditions for different HPLC
instruments. The aim of this study was to propose
and test this approach in gradient elution method’s
development.
Materials and Methods: The experiments were
carried out on three chromatographic systems
(UPLC, UHPLC and HPLC), while the separation
was achieved on Kinetex C18 Core-shell column
(100 mm × 2.1 mm, 2.6 μm particle size). Design
of experiments was constructed in Design-Expert
11.0. Indirect modeling, grid point search and
graphical presentations were done in Matlab
7.10.0.
Results: Dabigatrane etexilate mesylate and nine
structurally related compounds were selected as
suitable model mixture due to its complexity and
polarity. Method development was supported with
experimental design methodology, Placket –
Burman for screening and D-optimal design for
optimization purposes. Dwell volumes were
included in the optimization phase and in this way
the same optimal chromatographic conditions for
all three instruments were selected. They included 10 mM ammonium acetate buffer
with pH set to 4.9 using acetic acid, and
acetonitrile. The components of the mobile phase
were pumped into chromatographic system with
flow rate of 400 μL min-1 in a linear gradient mode:
at 0 minutes 24% (v/v) acetonitrile and 76% (v/v) of
buffer solution, at 15 minutes 54% (v/v) acetonitrile
and 46% (v/v) buffer solution. At 16 minutes the
acetonitrile content was back to 24% (v/v) and 76%
(v/v) of buffer solution. The re-equilibration time
was set to 5 minutes. The examined
chromatographic region is graphically presented
and optimal conditions are noticed as the cross
sections (yellow dots). The method was validated
and confirmed its utility on all instruments.
Conclusions: The proposed methodology
demonstrated its ability to predict joint optimal
chromatographic conditions for instruments with different values of dwell volume. The potential was
confirmed on complex model mixture and
instruments significantly differing in dwell volume
values. In this way the gap between developing
and routine needs could be overwhelmed, followed
by facilitated transfer of methods.

AB  - The aim of this study was to explore the mechanisms of bis(2- ethylhexyl) phthalate (DEHP), dibutyl phthalate (DBP) and bisphenol A (BPA) mixture-induced asthma development and test probiotic as a potential positive intervention. Comparative Toxicogenomics Database (CTD) and ToppGene Suite were used as the main tools for in silico analysis. In vivo 28-day experiment was conducted on rats - seven groups (n = 6): (1) Control: corn oil, (2) P: probiotic (8.78 * 108 CFU/kg/day); (3) DEHP: 50 mg/kg b.w./day, (4) DBP: 50 mg/kg b.w./day, (5) BPA: 25 mg/kg b.w./day; (6) MIX: DEHP + DBP + BPA; (7) MIX + P. Lungs, thymus and kidneys were extracted and prepared for redox status and essential metals analysis. By conducting additional in vitro experiment, probiotic phthalate and BPA binding ability was explored. There were 24 DEHP, DBP and BPA asthma-related genes, indicating the three most probable mechanisms - apoptosis, inflammation and oxidative stress. In vivo experiment confirmed that significant changes in redox status/essential metal parameters were either prominent, or only present in the MIX group, indicating possible additive effects. In vitro experiment confirmed the ability of the multy-strain probiotic to bind DEHP/DBP/BPA mixture, while probiotic administration ameliorated mixtureinduced changes in rat tissue.

abstract = "The aim of this study was to explore the mechanisms of bis(2- ethylhexyl) phthalate (DEHP), dibutyl phthalate (DBP) and bisphenol A (BPA) mixture-induced asthma development and test probiotic as a potential positive intervention. Comparative Toxicogenomics Database (CTD) and ToppGene Suite were used as the main tools for in silico analysis. In vivo 28-day experiment was conducted on rats - seven groups (n = 6): (1) Control: corn oil, (2) P: probiotic (8.78 * 108 CFU/kg/day); (3) DEHP: 50 mg/kg b.w./day, (4) DBP: 50 mg/kg b.w./day, (5) BPA: 25 mg/kg b.w./day; (6) MIX: DEHP + DBP + BPA; (7) MIX + P. Lungs, thymus and kidneys were extracted and prepared for redox status and essential metals analysis. By conducting additional in vitro experiment, probiotic phthalate and BPA binding ability was explored. There were 24 DEHP, DBP and BPA asthma-related genes, indicating the three most probable mechanisms - apoptosis, inflammation and oxidative stress. In vivo experiment confirmed that significant changes in redox status/essential metal parameters were either prominent, or only present in the MIX group, indicating possible additive effects. In vitro experiment confirmed the ability of the multy-strain probiotic to bind DEHP/DBP/BPA mixture, while probiotic administration ameliorated mixtureinduced changes in rat tissue.",
