AB  - Testis cancer (TC) is the most common malignancy of young men. Current evidence from studies, alongside genetics and hormonal status, suggests a significant role of toxic metals, cadmium (Cd) and lead (Pb), in the origin and development of TC. Besides oxidative stress and endocrine disruption, interaction with bioelements is one of the critical mechanisms of Cd and Pb toxicity and malign transformation. This study aimed to investigate metal levels in blood, healthy, and tumor testis tissue and to reveal hormone, oxidative status, and bioelements levels in patients with TC. The study enrolled 52 patients with TC and 61 healthy volunteers. Toxic metals and bioelements levels were analyzed by atomic absorption spectrophotometry (AAS) while electrochemiluminescence immunoassay (ECLIA) and spectrophotometry methods were used for hormone and oxidative parameters evaluation. Significantly higher blood Cd levels were depicted in TC cohort. Furthermore, blood Cd elevation was associated with a 1.98 higher probability of TC developing. However, a metal concentration between healthy and tumor testis tissue did not differ significantly. Lower levels of estradiol and testosterone, established in a cohort of TC patients, followed the significant role of hormones in TC development. At the same time, ischemia-modified albumin (IMA) has been recognized as a parameter with very good accuracy as a potential diagnostic marker for TC. The study revealed different distribution patterns of copper (Cu) and zinc (Zn) in the three compartments of the patients, as well significant correlation between essential metals Cu/Zn and toxic metals Cd/Pb indicating metal-metal interactions as pivotal mechanisms of metals toxicity.

AB  - Recent data indicate that lead (Pb) can induce adverse effects even at low exposure levels. Moreover, the corresponding mechanisms of low Pb toxicity have not been well identified. In the liver and the kidneys, Pb was found to induce various toxic mechanisms leading to organ physiological disruption. Therefore, the purpose of the study was to simulate low-dose Pb exposure in an animal model with the aim of assessing oxidative status and essential element levels as the main mechanism of Pb toxicity in the liver and kidneys. Furthermore, dose–response modelling was performed in order to determine the benchmark dose (BMD). Forty-two male Wistar rats were divided into seven groups: one control group, and six groups treated for 28 days with 0.1, 0.5, 1, 3, 7, and 15 mg Pb/kg b.w./day, respectively. Oxidative status parameters (superoxide dismutase activity (SOD), superoxide anion radical (O2−), malondialdehyde (MDA), total sulfhydryl groups (SHG), and advanced oxidation protein products (AOPP)) and Pb, copper (Cu), zinc (Zn), manganese (Mn), and iron (Fe) levels were measured. Lowering Cu levels (BMD: 2.7 ng/kg b.w./day), raising AOPP levels (BMD: 0.25 µg/kg b.w./day) in the liver, and inhibiting SOD (BMD: 1.3 ng/kg b.w./day) in the kidneys appear to be the main mechanisms of Pb toxicity. The lowest BMD was derived for a decrease in Cu levels in liver, indicating that this effect is the most sensitive.

AB  - With global urbanization and industrialization, air pollution has become an inevitable problem. Among air pollutants, toxic metals bound to particulate matter (PM) have a high hazardous potential, contributing to the development of several diseases, including various types of cancer. Due to PM pollution, Serbia is considered to be among the most polluted countries in Europe. Therefore, the objective of the study was to assess and characterize the non-carcinogenic and carcinogenic risks of children’s and adults’ exposure to metal(oid)s (Pb, Cd, Ni, and As) bound to PM10 in five of the most polluted areas in the Republic of Serbia (Subotica, Smederevo, Bor, Valjevo, and Kraljevo). Non-carcinogenic (HQ and HI) and carcinogenic risk (CR) were calculated using USEPA methodology. Our results show that PM10 concentrations exceeded the annual limit of 40 μg/m3 at four out of five monitoring sites (ranging from 44.33 to 63.25 μg/m3). Results obtained from Bor monitoring station show that safe limits were exceeded for both children and adults, indicating an unacceptable risk (> 1) obtained for inhalation exposure to the As (HQ = 6.14) and Cd (HQ = 1.17), while total HI was 7.43, which characterized the risk as unacceptable. For the same station, the CR value was 1.44E−04 (> 1 × 10−4). In other sites, the risks were acceptable. The characterized risk from exposure to the toxic elements via PM10 in critical locations in Serbia contributes to improving air quality by requiring regulatory organs to take new actions and adopt new measures to reduce air pollution.

AB  - The objectives of the study were to simulate low-level Pb exposure scenario in an animal model and to examine reproductive adverse effects. Based on obtained data, we have performed Benchmark dose (BMD)-response modelling. Male Wistar rats were randomized in seven groups (n = 6): one control and six treated with: 0.1, 0.5, 1, 3, 7, and 15 mg Pb/kg body weight, daily for 28 days by oral gavage. The rats were sacrificed and the blood and testes were used for further analysis of testosterone levels in serum, testicular essential metal levels and histological analysis. The Pb treatment led to a dose-dependent decrease of serum testosterone levels with a negative trend (BMDI 0.17–6.13 mg Pb/kg). Increase of Zn (dose-dependent, BMDI 0.004–19.7 mg Pb/kg) and Cu and a decrease of Mn testicular levels were also detected with unscathed histology of the testes. The presented results might be used in further evaluation of the point of departure in human health risk assessment for Pb.

The rats were sacrificed and the blood and testes were used for further analysis of testosterone levels in serum, testicular essential metal levels and histological analysis. The Pb treatment led to a dose-dependent decrease of serum testosterone levels with a negative trend (BMDI 0.17–6.13 mg Pb/kg). Increase of Zn (dose-dependent, BMDI 0.004–19.7 mg Pb/kg) and Cu and a decrease of Mn testicular levels were also detected with unscathed histology of the testes. The presented results might be used in further evaluation of the point of departure in human health risk assessment for Pb.",

Sulforaphane (SFN), isothiocyanate from cruciferous vegetables, diminishes chemical carcinogenesis susceptibility, but has been shown to act as a friend or a foe depending on various factors. By conducting the mechanistic toxicogenomic data mining approach, this research aimed to determine if SFN can alleviate toxic-metal and/or phthalate/BPA mixture-induced CRC at the gene level. Comparative Toxicogenomics Database, ToppGene Suite portal, Cytoscape software, InteractiVenn and Gene Expression Omnibus (GEO) database (GEO2R tool) was used. Among the mutual genes for all the investigated substances, SFN had a protective impact only through PTGS2. Other proposed protective SFN-targets included ABCA1, ALDH2, BMP2, DPYD, MYC, SLCO2A1, and SOD2, only in the case of phthalates/BPA exposure. The only additional gene relevant for SFN protection against the toxic metal mixture-induced CRC was ABCB1. Additionally, the majority of the top 15 molecular pathways extracted for SFN impact on phthalate and BPA mixture-linked CRC development were directly linked with cancer development, which was not the case with the toxic metal mixture. The current research has indicated that SFN is a more effective chemoprotective agent against CRC induced by phthalates/BPA mixture than by toxic-metal mixture. It has also presented the value of computational methods as a simple tool for directing further research, selecting appropriate biomarkers and exploring the mechanisms of toxicity.

AB  - Sulforaphane (SFN) is a naturally occurring molecule present in plants from Brassica family. It becomes bioactive after hydrolytic reaction mediated by myrosinase or human gastrointestinal microbiota. Sulforaphane gained scientific popularity due to its antioxidant and anti-cancer properties. However, its toxicity profile and potential to cause adverse effects remain largely unidentified. Thus, this study aimed to generate SFN-triggered adverse outcome pathway (AOP) by looking at the relationship between SFN-chemical structure and its toxicity, as well as SFN-gene interactions. Quantitative structure-activity relationship (QSAR) analysis identified 2 toxophores (Derek Nexus software) that have the potential to cause chromosomal damage and skin sensitization in mammals or mutagenicity in bacteria. Data extracted from Comparative Toxicogenomics Database (CTD) linked SFN with previously proposed outcomes via gene interactions. The total of 11 and 146 genes connected SFN with chromosomal damage and skin diseases, respectively. However, network analysis (NetworkAnalyst tool) revealed that these genes function in wider networks containing 490 and 1986 nodes, respectively. The over-representation analysis (ExpressAnalyst tool) pointed out crucial biological pathways regulated by SFN-interfering genes. These pathways are uploaded to AOP-helpFinder tool which found the 2321 connections between 19 enriched pathways and SFN which were further considered as key events. Two major, interconnected AOPs were generated: first starting from disruption of biological pathways involved in cell cycle and cell proliferation leading to increased apoptosis, and the second one connecting activated immune system signaling pathways to inflammation and apoptosis. In both cases, chromosomal damage and/or skin diseases such as dermatitis or psoriasis appear as adverse outcomes.

Comparative Toxicogenomics Database (CTD), ShinyGO, ToppCluster and Cytoscape were used for bioinformatic analysis. In vivo subacute study was performed on rats - five groups (n = 6): (1) Control: corn oil, (2) DEHP: 50 mg/kg b.w./day, (3) DBP: 50 mg/kg b.w./day, (4) BPA: 25 mg/kg b.w./day, (5) MIX: DEHP + DBP + BPA. Zebrafish embryos were exposed to the investigated substances in different doses, singularly and combined (binary and ternary mixtures). Liver injury was linked to 75 DEHP, DBP, and BPA genes, mostly connected to inflammation/oxidative stress. In rats, significant alterations in redox status/bioelements and pathohistology were most notable or exclusively present in MIX (probable additive effects). BPA decreased liver area (LA) index in dose-dependent manner. DEHP (< 2 µg/mL) and DBP (≤ 5 µg/mL) reduced LA values, while their higher doses increased LA index. The effect of DBP in binary mixtures led to a lethal outcome at the two highest concentrations, while the hepatotoxicity of DEHP/DBP/BPA mixture was dictated by BPA (confirmed by the benchmark dose analysis).

AB  - The current study aimed to assess the connection between the mixture of lead (Pb), cadmium (Cd), arsenic (As), methylmercury (MeHg) and decabrominated diphenyl ether (decaBDE) and thyroid function, by using in silico toxicogenomic data-mining approach. To obtain the linkage between investigated toxic mixture and thyroid diseases (TDs), the Comparative Toxicogenomics Database (CTD) was used, while gene ontology (GO) enrichment analysis was performed by ToppGeneSuite portal. The analysis has shown 10 genes connected to all chemicals present in the mixture and TDs (CAT, GSR, IFNG, IL1B, IL4, IL6, MAPK1, SOD2, TGFB1, TNF), most of which were in co-expression (45.68%), or belonged to the same pathway (30.47%). Top 5 biological processes and molecular functions affected by the investigated mixture emphasized the role of two common mechanisms – oxidative stress and inflammation. Cytokines and inflammatory response was listed as the main molecular pathway that may be triggered by simultaneous exposure to toxic metal(oid)s and decaBDE and connected to TDs. The direct relations between Pb/decaBDE and redox status impairment in thyroid tissue was confirmed by our chemical-phenotype interaction analysis, while the strongest linkage between Pb, As and decaBDE and thyroid disorders was found. The obtained results provide better understanding of molecular mechanisms involved in the thyrotoxicity of the investigated mixture, and can be used to direct further research.

To obtain the linkage between investigated toxic mixture and thyroid diseases (TDs), the Comparative Toxicogenomics Database (CTD) was used, while gene ontology (GO) enrichment analysis was performed by ToppGeneSuite portal. The analysis has shown 10 genes connected to all chemicals present in the mixture and TDs (CAT, GSR, IFNG, IL1B, IL4, IL6, MAPK1, SOD2, TGFB1, TNF), most of which were in co-expression (45.68%), or belonged to the same pathway (30.47%). Top 5 biological processes and molecular functions affected by the investigated mixture emphasized the role of two common mechanisms – oxidative stress and inflammation. Cytokines and inflammatory response was listed as the main molecular pathway that may be triggered by simultaneous exposure to toxic metal(oid)s and decaBDE and connected to TDs. The direct relations between Pb/decaBDE and redox status impairment in thyroid tissue was confirmed by our chemical-phenotype interaction analysis, while the strongest linkage between Pb, As and decaBDE and thyroid disorders was found. The obtained results provide better understanding of molecular mechanisms involved in the thyrotoxicity of the investigated mixture, and can be used to direct further research.",

AB  - The present study investigated the effects of PCBs on the rat kidneys with attention given to the determination critical effect dose (CED) using the Benchmark dose (BMD) approach. Male albino Wistar rats (7 animals per group) were given by oral gavage Aroclor 1254 dissolved in corn oil at doses of 0.0, 0.5, 1, 2, 4, 8, or 16 mg/kg b.w./day for 28 days. The PCB nephrotoxicity was manifested by a dose-dependent changes in serum urea levels. The study has also revealed PCB-induced oxidative stress induction in kidneys. The observed nephrotoxic effects can be partly explained by oxidative damage of lipids and proteins in the kidneys due to observed reduced CuZnSOD activity and disturbances in antioxidant protection. Аll the renal oxidative stress parameters showed dependence on PCB oral doses as well as internal, measure kidney PCB levels. Calculated BMDL values were lower than estimated no observed adverse effect levels (NOAEL) based on the study, suggesting the importance of BMD approach use in future risk assessment.

AB  - This study aimed to investigate the effect of 150 mg/L sodium fluoride (NaF) on redox status parameters and essential metals [copper (Cu), iron (Fe), and zinc (Zn)] in the blood, liver, kidney, brain, and spleen of Wistar rats and to determine the protective potential of selenium (Se) against fluoride (F-) toxicity. Male Wistar rats were randomly distributed in groups of five (n=5) receiving tap water (control) or water with NaF 150 mg/L, NaF 150 mg/L + Se 1.5 mg/L, and Se 1.5 mg/L solutions ad libitum for 28 days. Fluorides caused an imbalance in the redox and biometal (Cu, Fe, and Zn) status, leading to high superoxide anion (O2 .-) and malondialdehyde (MDA) levels in the blood and brain and a drop in superoxide dismutase (SOD1) activity in the liver and its increase in the brain and kidneys. Se given with NaF improved MDA, SOD1, and O2 .- in the blood, brain, and kidneys, while alone it decreased SH group levels in the liver and kidney. Biometals both reduced and increased F- toxicity. Joint impact of key air pollutants on COVID-19 severity: prediction based on toxicogenomic data analysis

AB  - Considering that some researchers point to a possible influence of air pollution on COVID-19 transmission, severity, and death rate, the aim of our in silico study was to determine the relationship between the key air pollutants [sulphur dioxide (SO2), carbon monoxide (CO), particulate matter (PMx), nitrogen dioxide (NO2), and ozone (O3)] and COVID-19 complications using the publicly available toxicogenomic analytical and prediction tools: (i) Comparative Toxicogenomic Database (CTD) to identify genes common to air pollutants and COVID-19 complications; (ii) GeneMANIA to construct a network of these common and related genes; (iii) ToppGene Suite to extract the most important biological processes and molecular pathways; and (iv) DisGeNET to search for the top gene-disease pairs. SO2, CO, PMx, NO2 , and O 3 interacted with 6, 6, 18, 9, and 12 COVID-19-related genes, respectively. Four of these are common for all pollutants (IL10, IL6, IL1B, and TNF) and participate in most (77.64 %) physical interactions. Further analysis pointed to cytokine binding and cytokine-mediated signalling pathway as the most important molecular function and biological process, respectively. Other molecular functions and biological processes are mostly related to cytokine activity and inflammation, which might be connected to the cytokine storm and resulting COVID-19 complications. The final step singled out the link between the CEBPA gene and acute myelocytic leukaemia and between TNFRSF1A and TNF receptor-associated periodic fever syndrome. This indicates possible complications in COVID-19 patients suffering from these diseases, especially those living in urban areas with poor air quality.
AB  - COVID-19 (engl. coronavirus disease 2019) respiratorna je bolest prouzročena infekcijom SARS-CoV-2 virusom (engl. severe acute respiratory
syndrome coronavirus 2). Pretpostavlja se da postoji utjecaj atmosferskih čimbenika, uključujući i onečišćenje zraka, na prenošenje koronavirusa,
njegovu težinu i stopu smrtnosti. Stoga je cilj ovoga in silico istraživanja bio utvrditi odnos između ključnih onečišćivača zraka [sumporova
dioksida (SO2), ugljikova monoksida (CO), lebdećih čestica (PMx), dušikova dioksida (NO2), ozona (O3)] i komplikacija COVID-a 19
korištenjem: (i) komparativne toksikogenomičke baze podataka (engl. Comparative Toxicogenomic Database, CTD) za dobivanje gena, međusobno
povezanih s onečišćivačima zraka i komplikacijama COVID-a 19, (ii) GeneMANIA servera za konstruiranje mreže između dobivenih i
srodnih gena, (iii) ToppGene Suite za izdvajanje najvažnijih bioloških procesa/molekularnih puteva i (iv) DisGeNET baze podataka za

abstract = "Considering that some researchers point to a possible influence of air pollution on COVID-19 transmission, severity, and death rate, the aim of our in silico study was to determine the relationship between the key air pollutants [sulphur dioxide (SO2), carbon monoxide (CO), particulate matter (PMx), nitrogen dioxide (NO2), and ozone (O3)] and COVID-19 complications using the publicly available toxicogenomic analytical and prediction tools: (i) Comparative Toxicogenomic Database (CTD) to identify genes common to air pollutants and COVID-19 complications; (ii) GeneMANIA to construct a network of these common and related genes; (iii) ToppGene Suite to extract the most important biological processes and molecular pathways; and (iv) DisGeNET to search for the top gene-disease pairs. SO2, CO, PMx, NO2 , and O 3 interacted with 6, 6, 18, 9, and 12 COVID-19-related genes, respectively. Four of these are common for all pollutants (IL10, IL6, IL1B, and TNF) and participate in most (77.64 %) physical interactions. Further analysis pointed to cytokine binding and cytokine-mediated signalling pathway as the most important molecular function and biological process, respectively. Other molecular functions and biological processes are mostly related to cytokine activity and inflammation, which might be connected to the cytokine storm and resulting COVID-19 complications. The final step singled out the link between the CEBPA gene and acute myelocytic leukaemia and between TNFRSF1A and TNF receptor-associated periodic fever syndrome. This indicates possible complications in COVID-19 patients suffering from these diseases, especially those living in urban areas with poor air quality., COVID-19 (engl. coronavirus disease 2019) respiratorna je bolest prouzročena infekcijom SARS-CoV-2 virusom (engl. severe acute respiratory
syndrome coronavirus 2). Pretpostavlja se da postoji utjecaj atmosferskih čimbenika, uključujući i onečišćenje zraka, na prenošenje koronavirusa,
njegovu težinu i stopu smrtnosti. Stoga je cilj ovoga in silico istraživanja bio utvrditi odnos između ključnih onečišćivača zraka [sumporova
dioksida (SO2), ugljikova monoksida (CO), lebdećih čestica (PMx), dušikova dioksida (NO2), ozona (O3)] i komplikacija COVID-a 19
korištenjem: (i) komparativne toksikogenomičke baze podataka (engl. Comparative Toxicogenomic Database, CTD) za dobivanje gena, međusobno
povezanih s onečišćivačima zraka i komplikacijama COVID-a 19, (ii) GeneMANIA servera za konstruiranje mreže između dobivenih i

AB  - One of the most polluted cities in Serbia and Europe is Bor, due to mining and
smelting processes in Mining-Metallurgical Complex in Bor. Mining activities release mainly
SO2 and paritculate matter in air (1) and, as arsenic is usually found in copper and lead ores,
air quality monitoring in Bor has been showing its presence in exceeded annual limit
concentrations in past few years. Exposure to arsenic is related with anemia, skin changes,
peripheral neuropathy and cancer development. This study was carried out to assess
children’s and adult’s exposure to arsenic bound to PM10 and to charcterize risk from their
exposure through the air in the city of Bor. Mean yearly concentrations of arsenic were used
from Reports of the Serbian Enviromental Protection Agency, for measuring stations: Bor-
City park, Bor 1, Bor 2 and Bor-Jugopetrol. The USEPA method was used to assess non-
carcinogenic and carcinogenic risk, which classifies non-carcinogenic risk as acceptable (<1)
or unacceptable (>1) based on hazard quotient (HQ) values for three main exposure
pathways, while cancer risk classification is based on CR values also as acceptable (<1×10 -4 )
or unacceptable (>1×10 -4 ) (2). HQinh values exceeded safe level both for children and adults
on all measuring stations, describing risk as unacceptable via inhalation exposure.
Unacceptable carcinogenic risk for children was calculated on measuring station Bor-
Jugopetrol and for adults on measuring stations Bor-City park, Bor 1 and Bor-Jugopetrol.
Actions for reduction of arsenic release into the air should be taken due to obtained results
for city of Bor.
AB  - Grad Bor spada među najzagađenije gradove u Republici Srbiji i Evropi. Najveći izvor
zagađenja u ovom gradu predstavlja rudnik bakra, Rudarsko-topioničarski basen Bor, usled
emisije, predominantno, SO2 i suspendovanih čestica (1). Arsen je čest pratilac ruda bakra i
olova, te se iz godine u godinu monitoringom kvaliteta vazduha uočava njegovo prisustvo u
koncentracijama koje prelaze dozvoljene godišnje granice. Izloženost arsenu može dovesti
do razvoja anemije, promena na koži, periferne neuropatije, ali i razvoja karcinoma. Cilj rada
bio je da se proceni izloženost i okarakteriše rizik dece i odraslih od izloženosti arsenu iz
PM10 čestica tj. putem zagađenog vazduha u gradu Boru. Vrednosti prosečnih godišnjih
koncentracija arsena u vazduhu su preuzete iz godišnjih izveštaja Agencije za zaštitu životne
sredine, za merne stanice: Bor-Gradski park, Bor 1, Bor 2 i Bor-Jugopetrol. Zdravstveni rizik
je procenjen metodologijom koju preporučuje Američka agencija za zaštitu životne sredine.
Na osnovu vrednosti količnika hazarda (HQ) za oralni, inhalacioni i dermalni put unosa, ne-
karcinogeni rizik se klasifikuje kao prihvatljiv (<1) ili neprihvatljiv (>1), dok se karcinogeni
rizik klasifikuje na osnovu vrednosti CR, takođe kao prihvatljiv (<1×10-4 ) ili neprihvaljiv
(>1×10 -4 ) (2). Izračunate vrednosti HQinh za decu i odrasle prelaze vrednost 1 ukazujući na
neprihvatljiv rizik od izloženosti arsenu preko PM10 čestica iz zagađenog vazduha na svim
razmatranim mernim stanicama. Neprihvatljiv karcinogeni rizik za decu je izračunat na
stanici Bor-Jugopetrol, a za odrasle na stanicama Bor-Gradski park, Bor 1 i Bor-Jugopetrol.
Dobijeni rezultati ukazuju na preku potrebu preduzimanja mera u cilju smanjenja emisije
arsena u vazduh u Boru.

AB  - Polluted air is significant factor with a negative impact on human health. Solving the
problem of polluted air requires involvement of the health system and health workers,
among whom pharmacists in public pharmacies can stand out as the most accessible health
workers with the highest knowledge in the field of toxicology. Considering the indisputable
role of pharmacists in improving health and disease prevention, raising public awareness of
air pollution health impact and advising patients on activities in case of air pollution,
pharmacists would contribute to maintaining health, reducing the burden on the health
system and its efficiency and sustainability. The pharmaceutical profession can make a
significant contribution to combating the impact of air pollution on human health by
increasing the availability of guidelines in the case of polluted air, increasing the availability
of air quality monitoring tools, strengthening educational capacity models, participating in
regulatory frameworks and improving funding for pharmaceutical services. In order for
these responsibilities to be fully incorporated into pharmaceutical practice, it is necessary to:
implement education in the field of toxicology, both at basic and higher levels of study in
order to develop the necessary competencies; increase public awareness of the health
consequences of air pollution, as well as the new roles of pharmacists; develop appropriate
guidelines, regulatory and financial support as well as screening tools. With this in mind, the
role of pharmacists in reducing the impact of air pollution on human health should be
recognized and developed in order to protect human health.
AB  - Zagađen vazduh predstavlja značajan faktor koji ima negativan uticaj na zdravlje ljudi.
Rešavanje problema zagađenog vazduha zahteva uključivanje zdravstvenog sistema i
zdravstvenih radnika, među kojima se mogu istaći farmaceuti u javnim apotekama kao
najdostupniji zdravstveni radnici, sa najvišim znanjima iz oblasti toksikologije. Imajući u
vidu neospornu ulogu farmaceuta u unapređenju zdravlja i prevenciji bolesti, podizanjem
svesti javnosti o uticaju zagađenja vazduha na zdravlje ljudi i davanjem saveta pacijentima o
aktivnostima u slučaju zagađenja vazduha, farmaceuti bi doprineli očuvanju zdravlja,
smanjenju opterećenja zdravstvenog sistema i njegovoj efikasnosti i održivosti.
Farmaceutska profesija može značajno doprineti suzbijanju uticaja zagađenja vazduha na
zdravlje ljudi i to: povećanjem dostupnosti smernica u slučaju zagađenog vazduha,
povećanjem dostupnosti alata za monitoring kvaliteta vazduha, jačanjem obrazovnih
kapaciteta, učestvovanjem u donošenju regulatornih okvira i poboljšanjem modela
finansiranja farmaceutskih usluga kako bi se obezbedila održivost. Da bi navedene
odgovornosti bile u potpunosti inkorporirane u farmaceutsku praksu, potrebno je:
implementirati edukaciju iz oblasti toksikologije, kako na osnovnim tako i na višim nivoima
studija kako bi se razvile neophodne kompetencije; povećati svest javnosti o zdravstvenim
posledicama zagađenja vazduha, kao i o novim ulogama farmaceuta; razviti odgovarajuće
smernice, regulatornu i finansijsku podršku kao i alate za skrining. Imajući navedeno u vidu,
uloga farmaceuta u smanjenju uticaja zagađenja vazduha na zdravlje ljudi treba biti
prepoznata i razvijena sa ciljem očuvanja zdravlja.

vidu neospornu ulogu farmaceuta u unapređenju zdravlja i prevenciji bolesti, podizanjem
svesti javnosti o uticaju zagađenja vazduha na zdravlje ljudi i davanjem saveta pacijentima o
aktivnostima u slučaju zagađenja vazduha, farmaceuti bi doprineli očuvanju zdravlja,
smanjenju opterećenja zdravstvenog sistema i njegovoj efikasnosti i održivosti.
Farmaceutska profesija može značajno doprineti suzbijanju uticaja zagađenja vazduha na
zdravlje ljudi i to: povećanjem dostupnosti smernica u slučaju zagađenog vazduha,
povećanjem dostupnosti alata za monitoring kvaliteta vazduha, jačanjem obrazovnih
kapaciteta, učestvovanjem u donošenju regulatornih okvira i poboljšanjem modela
finansiranja farmaceutskih usluga kako bi se obezbedila održivost. Da bi navedene
odgovornosti bile u potpunosti inkorporirane u farmaceutsku praksu, potrebno je:
implementirati edukaciju iz oblasti toksikologije, kako na osnovnim tako i na višim nivoima
studija kako bi se razvile neophodne kompetencije; povećati svest javnosti o zdravstvenim
posledicama zagađenja vazduha, kao i o novim ulogama farmaceuta; razviti odgovarajuće
smernice, regulatornu i finansijsku podršku kao i alate za skrining. Imajući navedeno u vidu,
uloga farmaceuta u smanjenju uticaja zagađenja vazduha na zdravlje ljudi treba biti
prepoznata i razvijena sa ciljem očuvanja zdravlja.",

AB  - According to the current regulations in Serbia, the cultivation and sale of hemp-based
products (Cannabis sativa L.) containing up to 0.3% of substances from the group of
tetrahydrocannabinol is allowed. ∆-9-tetrahydrocannabinol (THC) is responsible for the
psychoactive effects and the development of cannabis addiction. A growing number of products
based on parts or the whole cannabis plant are on free sale. Since cannabis is the most
commonly used psychoactive substance, there is a greater need for quick and easy detection
in the body. The Aim of this project was to examine the effect of commercial cannabis-based
products consuption on urine THC detection by using immunochromatographic test strips.
Eight healthy volunteers consumed commercially available hemp-based products (four tea
products, two ground and two whole seeds products). The products were consumed according to
the instructions given by the manufacturer, for three consecutive days, and on the fourth day the
first morning urine was tested for the presence of THC using multicomponent One Step Rapid
Test®-Ameritek® test strips. Urine samples from five subjects gave a negative test result, and three
samples, from subjects who consumed hemp ground seeds and tea in bulk, gave a positive THC
test. Further analysis of positive urine samples by the conformative method (HPLC-PDA) did not
prove the presence of THC. Consumption of certain commercial cannabis-based products can lead
to false-positive results in the detection of THC in urine by immunochromatographic test strips.
Therefore, it is necessary to perform further analysis of urine by conformative methods.
AB  - Prema važećoj regulativi u Srbiji je dozvoljeno gajenje i prodaja proizvoda na bazi
konoplje (Cannabis sativa L.) koji sadrže do 0,3% supstanci iz grupe tetrahidrokanabinola od
kojih se ∆-9-tetrahidrokanabinolu (THC) pripisuju psihoaktivni efekti i razvoj zavisnosti
povezan sa zloupotrebom kanabisa. U slobodnoj prodaji se nalazi sve veći broj proizvoda
baziranih na delovima ili celoj biljci kanabisa. S obzirom na to da je kanabis najčešće
korišćena psihoaktivna supstanca postoji i veća potreba za brzom i lakom detekcijom u
organizmu. Cilj ovog rada je ispitati uticaj konzumiranja komercijalnih proizvoda na bazi
konoplje na dokazivanje THC u urinu imunohromatografskim test trakama (1,2). U
ispitivanje je uključeno osam zdravih dobrovoljaca koji su konzumirali komercijalno
dostupne preparate na bazi konoplje (četiri čaja, dva mlevena i dva cela semena). Preparati
su konzumirani prema uputstvu datom od strane proizvođača, tri uzastopna dana, a četvrtog
dana je vršeno testiranje prvog jutarnjeg urina na prisustvo THC, upotrebom
višekomponentnih One Step Rapid Test®‐Ameritek® test traka. Uzorci pet ispitanika su dali
negativan rezultat testa, a tri uzorka, poreklom od ispitanika koji su konzumirali mleveno
seme konoplje i čaj u rinfuzu, dala su pozitivan test na THC. Daljom analizom urina koji su
dali pozitivan rezultat, konfirmativnom metodom (HPLC-PDA) nije dokazano prisustvo THC.
Konzumacija pojedinih preparata na bazi konoplje, koji se mogu naći u slobodnoj prodaji,
može da dovede do pojave lažno pozitivnih rezultata pri dokazivanju THC u urinu
imunohromatografskim test trakama, zbog čega je neophodno izvršiti dalju analizu urina
konfirmativnim metodama.

AB  - PM10 particles present in excessive concentrations in polluted air can negatively affect
respiratory and cardiovascular system, whereas lead exposure shows adverse effects on
renal, hematological, immunological, reproductive and neurological system, especially in
children (1). This study was carried out to assess children’s exposure to PM 10 and lead
through the air in critical districts of Serbia. Mean yearly concentrations of PM10 and lead
were used from reports of the Serbian Enviromental Protection Agency, for selected
measuring sites in the cities of Subotica, Smederevo, Bor, Valjevo and Kraljevo. Measuring
sites were selected based on air quality (over-polluted air) and being close to places where
children spend their time during the day. The US EPA method was used to assess health risk,
which classifies risk as acceptable (≤1) or unacceptable (>1) based on hazard quotient (HQ)
and hazard index (HI) values (2). HQ for PM10 particles was higher than 1 in all critical
districts, indicating risk as unacceptable. Values of HI for lead were lower than 1 in all critical
districts, indicating risk as acceptable. Results are showing that actions for reduction of air
pollution in Serbia should be taken due to obtained results for particulate matter and lead.
AB  - PM10 čestice prisutne u prekomernim koncentracijama u zagađenom vazduhu
ispoljavaju štetne efekte na respiratorni i kardiovaskularni sistem, dok olovo negativno utiče
i na renalni, hematopoetski, endokrini, imunološki, reproduktivni i nervni sistem, naročito
kod dece (1). Cilj ovog rada bio je da se proceni izloženost dece PM10 česticama i olovu putem
zagađenog vazduha u kritičnim područjima Srbije. Vrednosti prosečnih godišnjih
koncentracija PM 10 čestica i olova su preuzete iz godišnjih izveštaja Agencije za zaštitu
životne sredine, za odabrane merne stanice u gradovima Subotica, Smederevo, Bor, Valjevo i
Kraljevo. Kriterijum za izbor područja, odnosno mernih stanica, bio je kvalitet vazduha
(prekomerno zagađen vazduh) i blizina mesta gde deca provode značajan deo vremena.
Zdravstveni rizik je procenjen metodologijom koju preporučuje Američka agencija za zaštitu
životne sredine. Na osnovu vrednosti količnika hazarda (HQ) i indeksa hazarda (HI) rizik se
klasifikuje kao prihvatljiv (≤1) ili neprihvatljiv (>1) (2). HQ za PM10 čestice prelazi vrednost 1
u svim razmatranim područjima, ukazujući na neprihvatljiv rizik dok su vrednosti HI za
olovo manje od 1 ukazujući na prihvatljiv rizik u svim područjima. Dobijeni rezultati ukazuju
na potrebu preduzimanja mera u cilju smanjenja zagađenja vazduha u Republici Srbiji na
osnovu izračunatog zdravstvenog rizika od izloženosti suspendovanim česticama i olovu.

AB  - Opioids are a wide group of natural, semi-synthetic and synthetic substances, used in
the pharmacotherapy of painful conditions. However, due to their psychoactive properties,
they are often abused. Immunochromatographic test panels are used for substance abuse
monitoring and are quick, simple and based on the principle of competitive binding. Main
disadvantage of immunochromatographic analysis is cross-reactivity, because of sufficient
specificity of test antibodies for the analyte of interest. This study aimed to examine the
effect of codeine-based OTC preparation (Caffetin®) on false positive results of preliminary
opiate urineanalysis, and determine the time period during which these results may occur.

AB  - Opioidi predstavljaju široku grupu supstanci prirodnog, polusintetskog i sintetskog
porekla, koje se osim primene u farmakoterapiji bolnih stanja, , često zloupotrebljavaju. Za
kontrolu zloupotrebe koriste se imunohromatografske test trake, brza i jednostavna analiza
zasnovana na principu kompetitivnog vezivanja. Glavni nedostatak im je pojava ukrštene
reaktivnosti usled niske specifičnosti antitela za analit od interesa. Cilj ovog rada je bio
ispitati uticaj primene OTC preparata koji sadrži kodein (Caffetin®) na pojavu lažno
pozitivnih rezultata preliminarne imunohromatografske analize opijata u urinu i utvrditi
vremenski period njihove pojave od trenutka primene analgetika. U istraživanju su

AB  - Acute poisonings by drugs used for cardiovascular diseases treatment are less
frequent than poisonings by other drugs, but often lead to serious disorders in various organ
systems and are characterized by relatively high mortality. When it comes to polydrug
poisoning by drugs with a depressant effect on the cardiovascular system, hospital care of
the patients is necessary. This research aimed to present data on acute poisonings by drugs
used in the cardiovascular diseases treatment in the Republic of Serbia in the period 2010-
2018 based on the published Annual reports of the National Poison Control Center of the
Military Medical Academy (1). Number of patients examined on suspicion of poisoning by
drugs for cardiovascular diseases treatment, in the period 2010-2018, ranged from 4-6% of
all examined due to acute drug poisoning per year, while number of hospitalizations due to
drug poisoning ranged from 7 to 11% of all hospitalized patients. Of the total number of
deaths resulting from drug poisoning, 27% are patients who died due to the poisoning by
drugs used for cardiovascular diseases treatment. The most common causes of poisoning in
the examined patients were beta blockers (45%), followed by Ca 2+ channel blockers (25%)
and ACE inhibitors (21%), and these three groups of drugs are the most common causes in
hospitalized patients as well. Lethal outcomes are most often the result of acute poisoning by
Ca 2+ channel blockers (50%) and beta blockers (40%), mainly in combination with drugs
used in psychiatric illnesses treatment, such as benzodiazepines, antiepileptics and
antipsychotics.
AB  - Akutna trovanja lekovima za lečenje kardiovaskularnih bolesti su ređa u odnosu na
trovanja drugim lekovima, ali često dovode do ozbiljnih poremećaja u funkcionisanju
različitih sistema organa i karakterišu se relativno visokim mortalitetom. Kada su u pitanju
polimedikamentozna trovanja lekovima koji imaju depresorno dejstvo na kardiovaskularni
sistem, neophodno je bolničko zbrinjavanje pacijenata. Cilj ovog rada je bio prikazati analizu
podataka o akutnim trovanjima lekovima koji se koriste u terapiji kardiovaskularnih
oboljenja u Republici Srbiji u periodu od 2010 do 2018. godine na osnovu publikovanih
Godišnjaka Nacionalanog centra za kontrolu trovanja Vojnomedicinske akademije (1). Broj
pacijenata koji su pregledani pod sumnjom na trovanje lekovima za lečenje

Causes of acute poisoning with a fatal outcome in the Republic of Serbia in the period from 2010 to 2018

AB  - The aim of this investigation was to present data analysis on acute poisonings with
fatal outcome in the Republic of Serbia in from 2010 to 2018, based on the published Annual
reports of the National Poison Control Center of the Military Medical Academy (1). In the
observed 9-year period, the most common cause of acute poisoning was alcohol (50%),
followed by drugs (30.1%), while in the third and fourth place were psychoactive substances
(7.1%) and gases 4%. Corrosive and pesticide poisoning was present in only 2% of the total
number of examined patients per year. The most common reason for the hospitalization was
due to drug poisoning (71%), corrosives (9%), pesticides (5%), psychoactive substances
(4%), gases (4%) and other pathogens (7%). From 2010 to 2018, number of deaths due to
acute poisoning ranged from 26 to 43 annually with an average value of 33. Of 290 deceased
patients, 101 (34.9%) were of drug poisoning, 108 (38.5%) corrosives, 28 (9.8%) pesticides,
4 (1.4%) gases, 14 (4.6%) psychoactive substances and 37 (12.4%) other substances,
predominantly alcohols and mushrooms. Combination of two or more drugs most often led
to death (n=51), while among corrosives, hydrochloric acid was the most common (n=40). Of
the pesticides, the most common were organophosphorus insecticides (n=14), while heroin
was the most common psychoactive substance (n=7). The analyzed data indicated the
importance of preventive measures that need to be carried out in our country to reduce the
number of acute poisonings, especially the most severe forms that lead to death.
AB  - Cilj ovog rada bio je da se prikaže analiza podataka o akutnim trovanjima sa smrtnim
ishodom u Republici Srbiji u periodu od 2010. do 2018. godine na osnovu publikovanih
Godišnjaka Nacionalanog centra za kontrolu trovanja Vojnomedicinske akademije (1). U
posmatranom devetogodišnjem periodu najčešći uzročnik akutnih trovanja je alkohol (50%),
zatim lekovi (30,1%), dok su na trećem i četvrtom mestu psihoaktivne supstance (7,1%) i
gasovi 4%. Samo oko 2% od ukupnog broja pregledanih pacijenata godišnje su slučajevi
trovanja korozivima i pesticidima. Najčešći razlog hospitalizacije je usled trovanja lekovima
(71%), zatim korozivima (9%), pesticidima (5%), psihoaktivnim supstancama (4%),
gasovima (4%) i drugim uzročnicima (7%). Od 2010. do 2018. godine broj letalnih ishoda
usled akutnog trovanja se kretao od 26 do 43 slučaja godišnje sa prosečnom vrednošću od 33
slučaja po godini. Od 290 preminula pacijenta, 101 (34,9%) slučaj je bilo trovanje lekovima,
108 (38,5%) korozivima, 28 (9,8%) pesticidima, 4 (1,3%) gasovima, 14 (4,6%)
psihoaktivnim supstancama i 37 (12,4%) ostalim supstancama, dominantno alkoholima i
gljivama. Kombinacija dva i više lekova je najčešće dovela do smrtnog ishoda (n=51), dok je
među korozivima, najzastupljenija bila hlorovodonična kiselina (n=40). Od pesticida kao
uzročnika akutnih trovanja sa smrtnim ishodom najzastupljeniji su organofosforni
insekticidi (n=14), a prema broju smrtnih ishoda od psihoaktivnih supstanci najviše je
zastupljen heroin (n=7). Analizirani podaci ukazuju na značaj preventivnih aktivnosti koje je
u našoj zemlji potrebno sprovesti kako bi se smanjio broj akutnih trovanja, a time i onih
najtežih oblika koji dovode do smrtnog ishoda.

(7.1%) and gases 4%. Corrosive and pesticide poisoning was present in only 2% of the total
number of examined patients per year. The most common reason for the hospitalization was
due to drug poisoning (71%), corrosives (9%), pesticides (5%), psychoactive substances
(4%), gases (4%) and other pathogens (7%). From 2010 to 2018, number of deaths due to
acute poisoning ranged from 26 to 43 annually with an average value of 33. Of 290 deceased
patients, 101 (34.9%) were of drug poisoning, 108 (38.5%) corrosives, 28 (9.8%) pesticides,
4 (1.4%) gases, 14 (4.6%) psychoactive substances and 37 (12.4%) other substances,
predominantly alcohols and mushrooms. Combination of two or more drugs most often led
to death (n=51), while among corrosives, hydrochloric acid was the most common (n=40). Of
the pesticides, the most common were organophosphorus insecticides (n=14), while heroin
was the most common psychoactive substance (n=7). The analyzed data indicated the
importance of preventive measures that need to be carried out in our country to reduce the
number of acute poisonings, especially the most severe forms that lead to death., Cilj ovog rada bio je da se prikaže analiza podataka o akutnim trovanjima sa smrtnim
ishodom u Republici Srbiji u periodu od 2010. do 2018. godine na osnovu publikovanih
Godišnjaka Nacionalanog centra za kontrolu trovanja Vojnomedicinske akademije (1). U
posmatranom devetogodišnjem periodu najčešći uzročnik akutnih trovanja je alkohol (50%),
zatim lekovi (30,1%), dok su na trećem i četvrtom mestu psihoaktivne supstance (7,1%) i
gasovi 4%. Samo oko 2% od ukupnog broja pregledanih pacijenata godišnje su slučajevi
trovanja korozivima i pesticidima. Najčešći razlog hospitalizacije je usled trovanja lekovima
(71%), zatim korozivima (9%), pesticidima (5%), psihoaktivnim supstancama (4%),
gasovima (4%) i drugim uzročnicima (7%). Od 2010. do 2018. godine broj letalnih ishoda
usled akutnog trovanja se kretao od 26 do 43 slučaja godišnje sa prosečnom vrednošću od 33
slučaja po godini. Od 290 preminula pacijenta, 101 (34,9%) slučaj je bilo trovanje lekovima,
108 (38,5%) korozivima, 28 (9,8%) pesticidima, 4 (1,3%) gasovima, 14 (4,6%)
psihoaktivnim supstancama i 37 (12,4%) ostalim supstancama, dominantno alkoholima i
gljivama. Kombinacija dva i više lekova je najčešće dovela do smrtnog ishoda (n=51), dok je
među korozivima, najzastupljenija bila hlorovodonična kiselina (n=40). Od pesticida kao
uzročnika akutnih trovanja sa smrtnim ishodom najzastupljeniji su organofosforni
insekticidi (n=14), a prema broju smrtnih ishoda od psihoaktivnih supstanci najviše je
zastupljen heroin (n=7). Analizirani podaci ukazuju na značaj preventivnih aktivnosti koje je
u našoj zemlji potrebno sprovesti kako bi se smanjio broj akutnih trovanja, a time i onih
najtežih oblika koji dovode do smrtnog ishoda.",

AB  - Alternative form of cancer treatment includes targeting natural compounds such as
sulfur-rich dietary phytochemical sulforaphane (SFN). However, data on SFN safety,
interactions on the protein level and target of SFN in human organism are limited (1). The
aim of this study was to elucidate the target interactions of SFN in human body in order to
rationalize possible side-effects and predict off-targets by using in silico approach. STITCH
database (http://stitch.embl.de) was used to obtain the information about chemical–protein
interactions, while Metascape (https://metascape.org/) highlighted protein-protein
interaction enrichment (PPIE).
SwissTargetPrediction (http://www.swisstargetprediction.ch/) indicated the target
molecule classes of SFN in human. Human genes that had the strongest interaction with SFN
were NQO1, NFE2L2, CASP3, HSP90AA1, MAPK14, HDAC6, HPGDS, KEAP1, GSTA1 and
GSTM1. PPIE analysis singled out fluid shear stress and atherosclerosis, NRF2 pathway and
chemical carcinogenesis - reactive oxygen species (ROS) as the most significant interactions.
The most represented class of SFN targeted molecules in human organism were enzymes
(26.7%). Epidermal growth factor receptor erbB1, macrophage migration inhibitory factor,
nitric oxide synthase (inducible) showed the highest probability target rate. In our previous
study (2), we pointed out that the genome of cancer patients could affect SFN safety. The
current study provides a set of target genes, emphasizes the importance of oxidative stress in
the suggested genetic interactions and predicts classes of target molecules, which should
further be examined.
AB  - Alternativni oblik lečenja raka uključuje upotrebu prirodnih jedinjenja kao što je
fitohemikalija bogata sumporom, sulforafan (SFN). Međutim, podaci o interakcijama SFN na
nivou proteina i ciljnih mesta dejstva SFN u ljudskom organizmu su ograničeni (1). Cilj ove
studije bio je da se ukaže na ciljne interakcije SFN kod ljudi kako bi se racionalizovali mogući
neželjeni efekti i predvidela nova ciljna mesta toksičnosti korišćenjem in silico pristupa.

Effect of Lead and Polychlorinated-biphenyls Mixture on Sperm Cells in Rats

AB  - Benchmark dose (BMD) approach extends the use of data and allows a quantification
of the uncertainties and confirmation of the effect. The objective of this study was to
investigate whether a mixture of lead (Pb) and polychlorinated-biphenyls (PCBs)
has any effect on sperm cells, and whether these effects are dose dependent. The
experiment was conducted in accordance with the approval of Ethical Committee No. 323-
07-11822/2018-05. Male Wistar rats were exposed to the mixture of Pb and PCBs by oral
gavage for 28 days. Doses od Pb in mixture were 0.1 mg/kg/day, 0.5 mg/kg/day and 1
mg/kg/day, while doses of PCBs in mixture were 0.25 mg/kg/day, 0.5 mg/kg/day, and 1
mg/kg/day respectively. Rats were anesthetized by the combination of ketamine/xylazine.
Through the scrotum section, the right testis and epididymis were collected, while sperm
cells were taken from the cauda of epididymis by diffusion in saline solution at 37°C. Sperm
motililty was determined by the light microscope. The aliquot of the sperm cells was fixated
with the combination of formalin in the phosphate buffer and used for morphology and
concentration sperm analysis. The benchmark dose lower confidence limit (BMDL40) of
0.00013 mg/kg/day was derived for the sperm motility effect of Pb and PCBs as co-variant.
From our results, we can conclude that the mortality was the most sensitive parameter. This
value is also one of the lowest BMDL for Pb, which can lead to conclusion that PCBs increase
the potency of Pb toxicity.
AB  - Pristup računanja Benchmark doze (BMD) proširuje upotrebu podataka, omogućava
kvantifikaciju nesigurnosti i potvrđuje efekat. Cilj ove studije bio je da se ispita da li smeša
olova (Pb) i polihlorovanih-bifenila (PCBs) ispoljava efekat na ćelije sperme, kao i da se ispita
da li su efekti dozno zavisni. Eksperiment je sproveden u skladu sa odobrenjem Etičkog

often abused substances. Immunochromatographic test panels for preliminary urineanalysis
are used to track its abuse. Immunochromatographic analysis is quick, simple and based on
the principle of competitive binding. Its main disadvantage is cross reactivity - tendency of
antibodies to bind structurally similar compounds, including pseudoephedrine. This study
aimed to examine the effect of pseudoephedrine-based OTC preparations on the occurrence
of false-positive results of preliminary amphetamine urineanalysis and determine the time
period during which these results may occur. The study included two subjects. TylolHot®
was used for two days, while, on the first day, single dose was administered. On the second
day, preparation was administered according to the dosing regimen recommended by the
manufacturer (three times a day). Urine samples were collected for three consecutive days.
Analysis was performed by immunochromatographic test strips "DIAQUICK" Multi-Drug Test
Panels. TylolHot® use led to false positive results for amphetamine 3.75 h after single
administration, and lasted for 27 h with repeated administration. After the application of the
tested preparation, false positive results on amphetamine were obtained in different time
periods, which depended on the intra- and interindividual variations, as well as the dosing
regimen. Although test strips are used for quick preliminary analysis, it is necessary to check
the results via confirmation methods (gas or liquid chromatography coupled to mass
spectrometry). The obtained results are significant during routine testing of patients,
employees, students, athletes and members of the army.
AB  - Amfetamin je snažan stimulans centralnog nervnog sistema i jedna od supstanci koja
se najčešće zloupotrebljavaju. Za praćenje njegove zloupotrebe koriste se
imunohromatografske test trake za preliminarnu analizu urina. Imunohromatografska
analiza je brza i jednostavna metoda koja se zasniva na principu kompetitivnog vezivanja.
Njen glavni nedostatak je unakrsna reaktivnost – sklonost antitela da vezuju strukturno
slična jedinjenja, između ostalih i pseudoefedrin. Cilj ovog rada bio je ispitati uticaj primene
OTC preparata na bazi pseudoefedrina na pojavu lažno pozitivnih rezultata preliminarne
analize amfetamina u urinu i utvrditi vremenski period pojave ovih rezultata od trenutka
primene preparata. Ispitivanje je uključivalo dva ispitanika. Tokom dva dana korišćen je
preparat TylolHot® , pri čemu je prvi dan primenjena jedna doza, a drugi dan se preparat
primenjivao prema režimu doziranja preporučenom od strane proizvođača (tri puta
dnevno). Uzorci urina sakupljani su tokom tri uzastopna dana, a analiza je izvršena
imunohromatografskim test trakama "DIAQUICK" Multi-Drug Test Panels. Dobijeni su lažno
pozitivni rezultati na amfetamin već u periodu od 3,75 h nakon jednokratne primene
TylolHot ® preparata, i trajali su tokom 27 h pri višekratnoj primeni. Utvrđeno je da se nakon
primene TylolHot® preparata u različitim vremenskim periodima mogu dobiti lažno pozitivni
rezultati, što zavisi od intra- i interindividualnih varijacija i režima doziranja. Iako se test
trake koriste za brzu preliminarnu analizu, neophodno je potvrditi rezultate konfirmativnim
metodama (gasna ili tečna hromatografija spregnuta sa masenom spektrometrijom).
Dobijeni rezultati su značajni prilikom rutinskih testiranja pacijenata, zaposlenih, učenika,
sportista i pripadnika vojske.

Awareness of the citizens from the municipality of Bor on the influence of pharmaceutical waste on human health and environment

AB  - The rapid global growth of drugs production and consumption undoubtedly leads to
contamination of the environment with pharmaceutical substances, as well as the products
of their decomposition and metabolism. Therefore, the ecosystem is exposed to
pharmaceutical waste, what may result in various unwanted effects, such as the
development of microorganism’s resistance to antibiotics. A brief review of the European
Union regulations, as well as the United States of America and Australia amadments,
revealed the diversity of approaches and regulatory measures adopted with the purpose of
proper hazardous waste handling. However, none of proposed systems show enviable
results. We conducted the research in the period of March-May 2020, to find out how citizens
of Bor municipality handle the pharmaceutical waste. A total of 226 responses were collected
by surveying patients, in person and in electronic form. The obtained results were analyzed
using the statistical software SPSS 21.0. The presented results show that the awareness of
the potential toxicity of pharmaceutical waste is developed in more than a third of
respondents (37.5%), but also low level of knowleadge about the proper way of disposing
generated waste. Over 36% of men and 42% of women throw away their medicines together
with other household waste, while 9% of men and 11% of women take them to pharmacies.
The legal regulations and accompanying guidelines, together with appropriate media
campaign, would contribute to a better understanding of the given problem and the adoption
of new citizens habits when it comes to hazardous waste handling.
AB  - Brz rast proizvodnje i potrošnje lekova na globalnom nivou, nesumnjivo dovodi do
kontaminacije životne sredine farmaceutskim supstancama, njihovim metabolitima, kao i
proizvodima njihove dodatne razgradnje u životnoj sredini. Posledično, živi svet biva izložen
uticaju farmaceutskog otpada usled čega dolazi do ispoljavanja različitih neželjenih efekata,
kao, na primer, razvoja rezistencije mikroorganizama na antibiotike. Kratkim pregledom
regulative zemalja članica Evropske unije, Sjedinjenih Američkih Država i Australije, uočena
je različitost pristupa i regulatornih mera donetih u cilju pravilnog rukovanja otpadom,
između ostalih i farmaceutskim otpadom, pri čemu ni jedan sistem ne pokazuje zavidne
rezultate. Istraživanjem rađenim u periodu mart-maj 2020. godine pokušali smo da dođemo
do saznanja koliko je razvijena svest građana opštine Bor, o postojanju farmaceutskog
otpada kao posebne kategorije koja može da ugrozi zdravlje ljudi ali i životnu sredinu.
Anketiranjem pacijenata, kako ličnim kontaktom, tako i u elektronskoj formi prikupljeno je
ukupno 226 odgovora, a dobijeni rezultati analizirani su pomoću statističkog softvera SPSS
21.0. Dobijeni rezultati pokazuju da je svest o štetnosti farmaceutskog otpada razvijena kod
više od trećine ispitanika (37,5%), ali i postojanje niskog stepena informisanosti o pravilnom
načinu zbrinjavanja generisanog otpada. Zbog toga, preko 36% muškaraca i 42% žena lekove
baca u komunalni otpad., dok ih 9% muškaraca i 11% žena odnosi u apoteke. Dopuna
zakonske regulative kao i pratećih pravilnika, propraćena odgovarajućom medijskom
kampanjom doprinela bi boljem razumevanju datog problema i usvajanju novih navika kod
građana u načinu rukovanja neiskorišćenim lekovima iz domaćinstva.

AB  - Humans are exposed to the chemicals from very different sources. Therefore, the
exposure assessment should be focused on mixtures rather than single chemicals. In this
study, bioelements (Zn and Cu) homeostasis effects of mixture of Cd and BDE-209, widely
spread persistent environmental pollutants, has been evaluated. In order to evaluate the
potential of mixture on Zn and Cu homeosthasis in rats kidneys, Wistar rats were treated by
oral gavage during 28 days with three doses of Cd 2.5, 7.5, and 15 mg/bw/day, three
combinations of Cd with 1000 mg BDE-209/kg/day, and water as a control group. Kidney
tissue samples were collected, mineralised, and Cu and Zn levels were measured by flame
atomic absorbtion spectrophotometer. Possible interactions were evaluated comparing the
dose-response curves slopes and lower confidence limit of Benchmark dose of 10%
(BMDL10) were calculated by PROAST software (RIVM, Netherlands). Cd alone can disturb
Zn in kidneys and calculated BMDL10 was 1.713 mg Cd/kg/day, while in combination with

potent activity of the mixture than single Cd.
AB  - Ljudi mogu biti izloženi hemikalijama iz različitih izvora, te je potrebno procenjivati
efekte na bazi izloženosti smešama hemikalija, pre nego pojedinačnim hemikalijama. U
studiji je analiziran uticaj smeše Cd i BDE-209, široko rasprostranjenih zagađujućih
perzistentnih supstanci, na homeostazu bioelemenata, cinka (Zn) i bakra (Cu). U cilju
evaluacije potencijala smeše da naruši homeostazu Zn i Cu u bubrezima pacova, Wistar
pacovi su oralnom gavažom bili tretirani tokom 28 dana dozama Cd od 2,5, 7,5 i 15
mg/kg/dan, kombinacijama ove tri doze sa 1000 mg BDE-209/kg/dan i vodom kao
kontrolom. Tkivo bubrega je izdvojeno, zatim mineralizovano, a sadržaj Zn i Cu je meren
atomskom apsorpcionom spektrofotometrijom. Moguće interakcije su procenjivane
poređenjem nagiba krivih doza-odgovor, a izračunate su i donje granice pouzdanosti
Benchmark doze od 10% (BMDL10) korišćenjem PROAST softvera (RIVM, Holandija). Cd
primenjen sam utiče na homeostazu Zn u bubregu i izračunata BMDL10 iznosi 1,713
mg/kg/dan, dok u kombinacji sa BDE-209 iznosi 1,719 mg/kg/dan. Cd ometa homeostazu Cu
u bubregu pacova i BMDL10 iznosi 5,679 mg/kg/dan, dok je u kombinaciji sa BDE-209 BMDL
10 puta niža i iznosi 0,517 mg/kg/dan. Nagib krive doza odgovor za smešu Cd i BDE-209 bio
je daleko niži od nagiba krive doza-odgovor za Cd. S obzirom na to da Cu igra značajnu ulogu
u nastanku krvnih ćelija, ali i u očuvanju durgih sistema organa, dobijeni rezultati iziskuju
pažnju zbog daleko potentnijeg uticaja smeše na nivo Cu u odnosu na uticaj samog Cd.

abstract = "Humans are exposed to the chemicals from very different sources. Therefore, the
exposure assessment should be focused on mixtures rather than single chemicals. In this
study, bioelements (Zn and Cu) homeostasis effects of mixture of Cd and BDE-209, widely
spread persistent environmental pollutants, has been evaluated. In order to evaluate the
potential of mixture on Zn and Cu homeosthasis in rats kidneys, Wistar rats were treated by
oral gavage during 28 days with three doses of Cd 2.5, 7.5, and 15 mg/bw/day, three
combinations of Cd with 1000 mg BDE-209/kg/day, and water as a control group. Kidney
tissue samples were collected, mineralised, and Cu and Zn levels were measured by flame
atomic absorbtion spectrophotometer. Possible interactions were evaluated comparing the
dose-response curves slopes and lower confidence limit of Benchmark dose of 10%
(BMDL10) were calculated by PROAST software (RIVM, Netherlands). Cd alone can disturb
Zn in kidneys and calculated BMDL10 was 1.713 mg Cd/kg/day, while in combination with

AB  - The aim of this study was to: (1) examine subacute toxicity of bis (2-ethylhexyl)
phthalate (DEHP), dibutyl phthalate (DBP) and bisphenol A (BPA) mixture in rats and
compare it with individual substance effects; (2) investigate the mechanisms of toxicity on
critical target organ, testes (in vivo/in silico study); (3) examine multibiotic ability to mitigate
mixture toxicity. Male rats were divided into groups (n = 6): (1) Control; (2) P (probiotic
(8.78*10 8 CFU/kg bw/day)); (3) DEHP (50 mg/kg bw/day); (4) DBP (50 mg/kg bw/day); (5)
BPA (25 mg/kg bw/day); and (6) MIX (50 mg/kg bw/day DEHP + 50 mg/kg bw/day DBP +
25 mg/kg bw/day BPA) (7) MIX + P. In silico toxicogenomic analysis was performed by
Comparative Toxicogenomic Database (CTD), Citoscape software and ToppGene Suite portal.
MIX led to significant changes in tissue structure (liver, kidney, spleen and testis),
biochemical parameters and testosterone level of rats compared to the control group and
individual substances. In silico analysis revealed 20 genes associated with DEHP/DBP/BPA
and male reproductive system disorders, while the most probable mechanisms included
metabolism, aryl hydrocarbon receptor pathway, apoptosis, and oxidative stress. Oxidative
stress was confirmed in vivo, and changes in parameters in testicular tissue homogenates
were most pronounced or present only in MIX group. Probiotic annulled/mitigated changes
in biochemical, hematological, and oxidative stress parameters, relative liver mass, food
consumption, and organ pathohistology. Hence, the obtained results indicate the possibility
of future considerations of protective probiotic effects against phthalates and BPA mixture
toxicity.

(DEHP), dibutil ftalata (DBP), bisfenola A (BPA) na pacovima i uporediti je sa efektima
