Rise of Pregabalin Poisoning and Abuse Cases in Serbia: A Ten-Year Retrospective Study

AB  - Purpose: The aim of this study was to analyze data on gabapentinoid-related attendances to the National Poison Control Center of Serbia (NPCC), particularly abuse cases; to estimate its changes and to compare it with trends in national consumption rates of these drugs. We also aimed to analyze the main characteristics of the study population and to investigate the major clinical effects in poisoned patients. Patients and Methods: This is a retrospective study of patients admitted to the NPCC for acute poisoning involving gabapentinoids from 1 May 2012 to 1 October 2022. Results: There were 357 (95.5%) pregabalin-related and 17 (4.5%) gabapentin-related poisoning cases in 302 patients. Abuse of pregabalin was detected in 27.8% (84/302), while gabapentin abuse occurred in 0.7% (2/302) of all patients. A steady increase in rates of pregabalin poisoning and abuse cases strongly correlated with the increase in overall consumption of this drug, while there were no significant changes in rates of gabapentin consumption, poisoning and abuse rate during the study period. Most patients who abused pregabalin pregabalin were males (84.5%) and the median age was 26 years (range: 15–45 years). Almost 60% of patients who abused pregabalin (48/84) belonged to the migrant population. Co-ingestions occurred in 89.4% of pregabalin-related cases (319/357), resulting in more severe poisoning. The most often co-ingested drugs were benzodiazepines and among them clonazepam was detected in the largest number of cases. Conclusion: The poisoning and abuse cases involving pregabalin are on the rise in Serbia, which coincided with an increase in its overall consumption during the study period. Isolated pregabalin ingestions resulted in mild poisoning, although severe symptoms such as coma and bradycardia were recorded. When prescribing pregabalin to patients at risk of abuse caution is needed. Strengthening the measures for dispensing of pregabalin may reduce the risks associated with its abuse.

AB  - Various dual inhibitors of COX-2 and 5-LOX enzymes have been developed so far in order to obtain more effective and safer anti-inflammatory drugs. The aim of this study was to design and synthesize new dual COX-2 and 5-LOX inhibitors, and to evaluate their enzyme inhibition potential and redox properties. Thirteen compounds (1–13) were designed taking into account structural requirements for dual COX-2 and 5-LOX inhibition and antioxidant activity, synthesized, and structurally characterized. These compounds can be classified as N-hydroxyurea derivatives (1, 2 and 3), 3,5-di-tert-butylphenol derivatives (4, 5, 6, 7 and 13), urea derivatives (8, 9 and 10) and “type B hydroxamic acids” (11 and 12). COX-1, COX-2 and 5-LOX inhibitory activities were evaluated using fluorometric inhibitor screening kits. The evaluation of the redox activity of newly synthesized compounds was performed in vitro in the human serum pool using redox status tests.
The prooxidative score, the antioxidative score and the oxy-score were calculated. Seven out of thirteen synthesized compounds (1, 2, 3, 5, 6, 11 and 12) proved to be dual COX-2 and 5-LOX inhibitors. These compounds expressed good COX-2/COX-1 selectivity. Moreover, dual inhibitors 1, 3, 5, 11 and 12 showed good antioxidant properties.

AB  - Introduction: Telomeres are protective chromosomal ends. Short telomeres are a proven biomarker of biological aging. We aimed to find an association of telomere length and telomerase activity in circulating leukocytes and thromboaspirates of patients with acute myocardial infarction. Furthermore, association of the telomere-telomerase system with oxidative stress markers (as common risk factors for coronary artery disease (CAD)) was tested. Material and methods: Patients were selected from the patients admitted to the intensive care unit with acute myocardial infarction with ST-segment elevation (STEMI), with the following inclusion criteria – STEMI patients between 18 and 80 years old of both genders and candidates for primary percutaneous coronary intervention, with infarction pain present for a maximum of 12 h. In all the patients leukocyte telomere length, telomerase activity and scores related to oxidative-stress status (Protective, Damage and OXY) were evaluated. Results: Patients were divided into different groups: with stable angina pectoris (AP) (n = 22), acute myocardial infarction with: STEMI (n = 93), non-obstructive coronary arteries (MINOCA) (n = 7), blood vessel rupture (n = 6) at three time points, and compared to the group of 84 healthy subjects. Telomerase activity was significantly higher in all CAD sub-groups compared to the control group (AP = 0.373 (0.355–0.386), STEMI = 0.375 (0.349–0.395), MINOCA = 0.391 (0.366–0.401), blood vessel rupture = 0.360 (0.352–0.385) vs. CG = 0.069 (0.061–0.081), p < 0.001), while telomeres were significantly shorter in STEMI, MINOCA and blood vessel rupture groups compared to the control group (STEMI = 1.179 (0.931–1.376), MINOCA = 1.026 (0.951–1.070), blood vessel rupture = 1.089 (0.842–1.173) vs. CG = 1.329 (1.096–1.624), p = 0.030]. Values of OXY score were significantly higher in STEMI and MINOCA patients compared to the control group and AP patients (5.83 (4.55–7.54) and 10.28 (9.19–10.72) vs. 4.94 (3.29–6.18) and 4.18 (2.58–4.86), p < 0.001). Longer telomeres and higher telomerase activity were found in thromboaspirates, compared to the peripheral blood leukocytes in the same patients (1.25 (1.01–1.84) vs. 1.18 (0.909–1.516), p = 0.036; and 0.366 (0.367–0.379) vs. 0.366 (0.367–0.379), p < 0.001, respectively). In addition, telomere length and telomerase activity had good diagnostic ability to separate STEMI patients from healthy persons. Conclusions: Leukocyte telomere length and telomerase activity can differentiate CAD patients from healthy persons, and relate CAD to oxidative stress.

AB  - A comparative in vitro study of the antioxidant potential of natural phenols (zingerone, curcumin, raspberry ketone, magnolol) and their synthesized derivatives was performed. The antioxidant efficiency was evaluated in blood serum obtained from healthy individuals, by means of spectrophotometry, before and after the addition of pro-oxidant tert-butyl hydroperoxide (TBH). Moreover, the antioxidant effect of an equimolar mixture of curcumin and zingerone was investigated. Interpretation of our results reveals that in the blood serum of healthy individuals curcumin (C1), raspberry ketone (RK1), magnolol (M1) and synthesized derivative of zingerone (Z2) demonstrate remarkable antioxidant effects (p < 0.05). However, in the state of TBH-induced excessive oxidative stress natural magnolol and synthesized derivatives C1, Z1 and RK1 show powerful antioxidant activity and thus can be further investigated to obtain information about their metabolic transformations and their potential influence at the cellular level. Results obtained from measurements in an equimolar mixture of zingerone and curcumin indicate synergism (p < 0.05) between the two compounds. This combination is especially successful due to the fast and efficient neutralization of added pro-oxidant TBH. The commercial availability of turmeric and ginger and their frequent combined use in diet suggest ideas for further broader utilization of the beneficial synergistic effect of their phenolic components.

-, targeted or immunotherapy, or cancer by itself, 
and it represents a great challenge for clinicians in everyday practice. A wide spectrum of factors 
related t
o chemotherapy (type of drug, dose during each cycle, cumulative dose, schedule, method 
of  application,  combination  with  other  cardiotoxic  drugs  or  association  with  radiotherapy)  and  
patient  characteristics  (age,  presence  of  cardiovascular  risk  factors,  pr
evious  cardiovascular  
disease)  are  the  determining  factors  that  influence  the  frequency  of  CT.  Imaging  methods  for  
morphological  and  functional  monitoring  of  the  heart  muscle  are  used  for  monitoring  CT.  The  
quest  for  diagnostic  tools  for  early  CT  detection 
is  of  great  significance.  In  line  with  this,  the  
measurement  of  some  cardiac  biomarkers  has  found  its  place  in  clinical  settings  as  an  early  
determinant of myocardial injury. Therefore, in this review article, special attention will be paid 
to  certain  wel
l-established,  as  well  as  certain  novel  cardiac  biomarkers,  and  their  role  in  
recognizing asymptomatic CT, in order to gain deeper insight into their diagnostic utility.
AB  - Klinička  efikasnost  hemioterapije,  kao  priznatog  terapeutskog  pristupa  malignim 
bolestima, obično ima određena ograničenja zbog svoje kardiotoksičnosti (KT), te posledične 
kardiomiopatije, pa čak i srčane insuficijencije. KT se definiše kao svaka povreda miokarda 
povezana  s  lečenjem  raka,  uzimajući  u  obzir  hemioterapiju,  radioterapiju,  biološku  ili 
imunoterapiju,  kao  i  sam  karcinom,  te  predstavlja veliki  izazov  za  kliničare 
u  svakodnevnoj  
praksi. Širok spektar faktora povezanih s hemioterapijom (vrsta leka, doza tokom svakog ciklusa, 
kumulativna doza, raspored, način primene, kombinacija s drugim kardiotoksičnim lekovima ili 
povezanost s radioterapijom) i karakteristike bolesnika (dob, prisutnost faktora kardiovaskularnog 
rizika, prethodne kardiovaskularne bolesti), odlučujući su faktori koji utiču na učestalost KT. Za 
praćenje KT koriste se radiološke metode za morfološko i funkcionalno praćenje srčanog mišića. 
Potraga za dij
agnostičkim alatima za rano otkrivanje KT od velikog je značaja. U skladu s tim, 
merenje nekih srčanih biomarkera kao rane determinante povrede miokarda našlo je svoje mesto 
u kliničkim okruženjima. Stoga 
će u ovom preglednom članku posebna pažnja biti po
sve
ć
ena 
nekim  dobro  utvrđenim,  kao  i  nekim  novim  srčanim  biomarkerima,  i  njihovoj  ulozi  u 
prepoznavanju asimptomatske KT, kako bi se stekao dublji uvid u njihovu dijagnostičku korisnost 
i neželjene efekte lekova koji se primenjuju u terapiji karcinoma.

AB  - Background: Inflammation, oxidative stress and an imbalance between proteases and protease inhibitors are recognized pathophysiological features of chronic obstructive pulmonary disease (COPD). The aim of this study was to evaluate serum levels of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in patients with COPD and to assess their relationship with lung function, symptom severity scores and recent acute exacerbations. Methods: In this observational cohort study, serum levels of MMP-9 and TIMP-1 and the MMP-9/TIMP-1 ratio in the peripheral blood of COPD patients with stable disease and healthy controls were determined, and their association with lung function (postbronchodilator spirometry, body plethysmography, single breath diffusion capacity for carbon monoxide), symptom severity scores (mMRC and CAT) and exacerbation history were assessed. Results: COPD patients (n = 98) had significantly higher levels of serum MMP-9 and TIMP-1 and a higher MMP-9/TIMP-1 ratio than healthy controls (n = 47) (p ≤ 0.001). The areas under the receiver operating characteristic curve for MMP-9, TIMP-1 and the MMP-9/TIMP-1 ratio for COPD diagnosis were 0.974, 0.961 and 0.910, respectively (all p < 0.05). MMP-9 and the MMP-9/TIMP-1 ratio were both negatively correlated with FVC, FEV1, FEV1/FVC, VC, and IC (all p < 0.05). For MMP-9, a positive correlation was found with RV/TLC% (p = 0.005), and a positive correlation was found for the MMP-9/TIMP-1 ratio with RV% and RV/TLC% (p = 0.013 and 0.002, respectively). Patients with COPD GOLD 3 and 4 presented greater MMP-9 levels and a greater MMP-9/TIMP-1 ratio compared to GOLD 1 and 2 patients (p ≤ 0.001). No correlation between diffusion capacity for carbon monoxide and number of acute exacerbations in the previous year was found. Conclusions: COPD patients have elevated serum levels of MMP-9 and TIMP-1 and MMP-9/TIMP-1 ratio. COPD patients have an imbalance between MMP-9 and TIMP-1 in favor of a pro-proteolytic environment, which overall indicates the importance of the MMP-9/TIMP-1 ratio as a potential biomarker for COPD diagnosis and severity.

Electrochemical Characterisation and Confirmation of Antioxidative Properties of Ivermectin in Biological Medium

AB  - Ivermectin (IVM) is a drug from the group of anthelmintics used in veterinary and human medicine. Recently, interest in IVM has increased as it has been used for the treatment of some malignant diseases, as well as viral infections caused by the Zika virus, HIV-1 and SARS-CoV-2. The electrochemical behaviour of IVM was investigated using cyclic (CV), differential pulse (DPV) and square wave voltammetry (SWV) at glassy carbon electrode (GCE). IVM showed independent oxidation and reduction processes. The effect of pH and scan rate indicated the irreversibility of all processes and confirmed the diffusion character of oxidation and reduction as an adsorption-controlled process. Mechanisms for IVM oxidation at the tetrahydrofuran ring and reduction of the 1,4-diene structure in the IVM molecule are proposed. The redox behaviour of IVM in a biological matrix (human serum pool) showed a pronounced antioxidant potential similar to that of Trolox during short incubation, whereas a prolonged stay among biomolecules and in the presence of an exogenous pro-oxidant (tert-butyl hydroperoxide, TBH) resulted in a loss of its antioxidant effect. The antioxidant potential of IVM was confirmed by voltametric methodology which is proposed for the first time.

The effect of pH and scan rate indicated the irreversibility of all processes and confirmed the diffusion character of oxidation and reduction as an adsorption-controlled process. Mechanisms for IVM oxidation at the tetrahydrofuran ring and reduction of the 1,4-diene structure in the IVM molecule are proposed. The redox behaviour of IVM in a biological matrix (human serum pool) showed a pronounced antioxidant potential similar to that of Trolox during short incubation, whereas a prolonged stay among biomolecules and in the presence of an exogenous pro-oxidant (tert-butyl hydroperoxide, TBH) resulted in a loss of its antioxidant effect. The antioxidant potential of IVM was confirmed by voltametric methodology which is proposed for the first time.",

AB  - Background: The uneven lipid-lowering statin effects and
statin intolerance raise interest regarding the involvement
of coadministration of statins and dietary supplements.
This study aimed to evaluate the effects of octacosanol
supplementation on markers of redox status in cardiovascular patients on chronic atorvastatin therapy.
Methods: A double-blind, randomized, placebo-controlled,
single-centre study was conducted. Redox status home-
ostasis parameters  i.e., advanced oxidation protein products (AOPP), pro-oxidant-antioxidant balance (PAB), total
oxidant status (TOS), total antioxidant status (TAS), superoxide dismutase activity (SOD), total protein sulfhydryl (SH-
groups), and paraoxonase 1 (PON1) activity  were
assessed in 81 patients. According to favorable changes in
lipid profile, patients were classified into two groups:
responders (n = 35) and non-responders (n = 46), and
followed for 13 weeks. A principal component analysis
(PCA) was applied to explore the effect of octacosanol supplementation and the relationship between investigated
parameters as predictors of responders’ and non-responders’ status.
Results: Significant decrease in Oxy-score value was found
at the endpoint compared to baseline in responders’ group (21.0 (13.4–25.5) versus 15.1 (12.4–18.0); P < 0.01).
PCA analysis extracted 4 significant factors in the both
groups, whereas extracted factors containing »octacosanol
status« variable explained 14.7% and 11.5% of the variance
in responders’ and non-responders’ subgroups, respectively.
Conclusions: Octacosanol supplementation leads to an
improvement of lipid profile and markers of redox status in
responders’ group. New studies are needed to validate our
results in order to find the best approach for personalized
supplementation as a useful adjunct to standard statin
therapy.
AB  - Uvod: Neujednačeni hipolipemijski efekti primene statina kao i intolerancija na statine dovode do porasta inte resovanja u vezi sa zajedničkom primenom statina i dijetetskih suplemenata. Ova studija je imala za cilj da proceni efekte suplementacije oktakozanolom na markere redoks statusa kod kardiovaskularnih pacijenata na hroničnoj terapiji atorvastatinom. Metode: Sprovedena je dvostruko slepa, randomizovana, placebo kontrolisana studija. U serumu 81 pacijenta određivani su parametri redoks statusa [produkti uznapredovale oksidacije proteina (AOPP), prooksidativno-antioksidativni balans (PAB), totalni oksidativni status (TOS), totalni antioksidativni status (TAS), aktivnost superoksid dismutaze (SOD), sulfihidrilne grupe (SH-grupe) i aktivnost paraoksonaze 1 (PON1)]. PCA analiza je izdvojila po 4 značajna faktora u obe grupe, pri čemu faktori koji sadrže varijablu "status oktakozanola" objašnjavaju 14,7% i 11,5% ukupne varijacije kod respondera i nerespondera, redom. Zaključak: Suplementacija oktakozanolom dovodi do poboljšanja lipidnog profila i markera redoks statusa u grupi respondera. Buduće studije su potrebne za potvrdu dobijenih rezultata a u cilju pronalaženja optimalnog pristupa za personalizovanu suplementaciju uz standardnu terapiju statinima.

Chalcone derivatives as potential antiseptics and disinfectants

AB  - The global market for antiseptics and disinfectants is growing rapidly, fueled by a
coronavirus pandemic and a possible outbreak of infectious diseases in the future. The
antimicrobial, anti-inflammatory and redox activity of chalcone (1,3-diaryl-2-propen-1-one)
is well documented in the literature. The aim of this study is screening of antimicrobial
activity of chalcones and their saturated derivatives as active pharmaceutical ingredients,
synthesized by Claisen-Schmidt condensation (1). The redox potential of the investigated
compounds was tested in the biological environment using spectrophotometric methods to
determine prooxidant/antioxidant parameters. Within preformulation studies solubility and
compatibility with excipients commonly used in liquid pharmaceutical dosage forms was
performed. All the tested chalcones and their saturated derivatives showed satisfactory
antimicrobial activity, but two saturated chalcones showed the best MIC (0.156 - 1.25 mM)
and were categorized as compounds with strong bacteriostatic activity (2). Solubility of the
chalcons with moderate antimicrobial activity and redox potential was higher than its
minimum bacteriostatic and bactericidal concentration in all the tested solvents (ethanol,
isopropyl alcohol and propylene glycol). Based on the chemical structure and predicted logP
values for saturated chalcones that show stronger antimicrobial activity, better water
solubility is expected. These data could be a starting point for formulations of antiseptics and
disinfectants with lower concentrations of alcohol-based solvents, as the synergism between
saturated chalcone and ethanol and isopropyl alcohol was previously shown. Considering the
lighter color of the saturated chalcone, better aesthetic acceptability of the developed
formulations is expected, since it will not stain the skin and objects during application.
AB  - Globalno tržište antiseptika i dezinfekcionih sredstava ubrzano raste podstaknuto
pandemijom koronavirusa i moguć im širenjem drugih zaraznih bolesti u buduć nosti.
Antimikrobna, antiinflamatorna i redoks aktivnost halkona (1,3-diaril-2-propen-1-ona) je

AB  - Nonalcoholic fatty liver disease (NAFLD) is the most common chronic liver disease,
present in up to 30% of the adult population worldwide (1). Triglycerides accumulation in
hepatocytes (steatosis) represents the root cause of NAFLD and is associated with oxidative
stress, which could further lead to fibrosis and cell death of hepatocytes (2). The aim of this
research was to identify redox status markers for predicting the risk of developing steatosis.
158 participants were included. Steatosis was confirmed by ultrasound in 101 subjects,
while the remaining 57 were in the control group. The following markers of redox status
were determined in serum and plasma samples of all subjects: superoxide dismutase 1
(SOD1), paraoxonase (PON1), malondialdehyde (MDA) and superoxide anion (O2.-). For this
purpose, spectrophotometric methods and enzyme immunosorbent assays were used. SOD1
was statistically significantly higher (P<0.001), while O2 .- was significantly lower in the
patient group (P<0.001). SOD1 was significantly negatively correlated with O2.- (ρ= -0.494,
P<0.001) and MDA (ρ= -0.242, P=0.002). Univariate binary logistic regression analysis
showed a positive association between SOD1 and the presence of steatosis (OR=1.018, 95%
CI 1.005-1.031; P=0.005), as well as a negative association between O2 .- and the presence of
steatosis (OR=0.959, 95% CI 0.941-0.978; P<0.001). Multivariate analysis singled out SOD1
(OR=1.024, 95% CI 1.006-1.041; P=0.007) and O2.- (OR=0.965, 95% CI 0.942-0.989;
P=0.004) as independent predictors for the presence of steatosis in our subjects. The redox
status parameters, SOD1 and O2, respectively, showed a positive and negative prediction of
the presence of statosis in our subjects.
AB  - Nealkoholna masna bolest jetre (eng. nonalcoholic fatty liver disease, NAFLD) je
najčešće hronično oboljenje jetre, prisutno i u do 30% adultne populacije širom sveta (1).
Značajnu ulogu u nastanku NAFLD ima akumulacija triglicerida u hepatocitima – steatoza,
koja je povezana sa oksidativnim stresom, a koji dalje vodi fibrozi i ćelijskoj smrti hepatocita
(2). Cilj ovog istraživanja bio je identifikacija markera redoks statusa za predviđanje rizika za
nastanak steatoze. Studija je obuhvatila 158 ispitanika iz Kliničko bolničkog centra Zemun.
Steatoza je potvrđena ultrazvukom kod 101 ispitanika, dok je preostalih 57 činilo kontrolnu
grupu. U uzorcima seruma i plazme svih ispitanika određeni su sledeći markeri redoks

AB  - The pathogenesis of the COVID-19 infection caused by the SARS-CoV-2 coronavirus is
the subject of active research worldwide. As a complex disease, it is suspected that oxidative
stress plays a role in its pathogenesis and contributes to the progressive shortening of
telomeres. Patients aged 25–84 years (n=31) were monitored at the time of diagnosis, as
well as at control on the 14th and 21st days. Redox status parameters were determined: total
oxidant status (TOS), superoxide anion radical (O2.-), prooxidant-antioxidant balance (PAB),
advanced protein oxidation products (AOPP), malondialdehyde (MDA), and ischemia-
modified albumin (IMA) as prooxidants and total antioxidant status (TAS), superoxide-
dismutase (SOD) and paraoxonase 1 (PON1) enzyme activity, total antioxidant and total
oxidant status ratio (TAS/TOS), and total sulfhydryl groups (SHG) as antioxidants. Patients
have the shortest telomeres at diagnosis and their length increases during recovery. A
statistically significant difference in rTL was shown in patients at the time of diagnosis and
after 21 days (p=0.008). rTL has a statistically significant negative correlation with SHG in
patients at the time of diagnosis (ρ=-0.386, p=0.038), as well as in patients after 14 days (ρ =-
0.389, p=0.037). After 21 days, a statistically significant positive correlation was shown
between rTL and SOD (ρ =0.381, p=0.046). The obtained results indicate progressive
shortening of telomeres caused by COVID-19 infection, impaired redox status, as well as the
change of these parameters during recovery, which confirms the link between oxidative
stress and rTL, as well as the participation of these processes in the pathogenesis of COVID-
19.

aktivnog istraživanja širom sveta. Kako je u pitanju složena bolest, smatra se da i u njenoj
patogenezi leži oksidativni stres, koji doprinosi progesivnom skraćivanju telomera. Pacijenti
starosti 25-84 godina (n=31) praćeni su u trenutku postavljanja dijagnoze, kao i na kontroli
14. i 21. dana. Parametri redoks statusa određivani su iz seruma ili plazme odgovarajućim
spektrofotometrijskim metodama, dok je iz leukocita periferne krvi merena relativna dužina
telomera (rTL) pomoću qPCR metode. Od parametara oksidativnog statusa određivani su:
totalni oksidantni status (TOS), superoksidni anjon radikal (O2.- ), prooksidantno-
antioksidantni balans (PAB), uznapredovali produkti oksidacije proteina (AOPP),
malondialdehid (MDA), ishemijom modifikovan albumin (IMA). Kao parametri
antioksidativne zaštite određivani su: totalni antioksidantni status (TAS), aktivnost enzima
superoksid-dismutaze (SOD) i paraoksonaze 1 (PON1), odnos totalnog antioksidantnog i

AB  - Non-enzymatic glycation, oxidative stress (OS) and dyslipidemia are the main
metabolic alterations behind the development of macrovascular
complications (cardiovascular diseases) of diabetes mellitus (DM).
However, clinical relevance of biomarkers of these processes in patients with microvascular
complications (nephropathy, neuropathy, retinopathy) is less understood. Therefore, the aim
of this study was to examine advanced glycation products (AGEs), biomarkers of OS, and
dyslipidemia in 100 DM patients (33 without microvascular complications and 77 with
complications) and 30 subjects without DM. AGEs levels were higher in patients with
complications than in those without complications (median: 5.72; interquartile range: 4.60-
6.54 U/mL vs. median: 4.84; interquartile range: 4.10-5.40 U/L; P<0.05). In addition to
AGEs, the group with diabetic retinopathy had higher plasma total antioxidant capacity
(P<0.05), while the group with diabetic nephropathy had smaller LDL size than the patients
without these complications (25.48±1.26 nm vs. 26.21±1.19 nm; P<0.05). The patients
with co-existing cardiovascular disease were further characterized by dysfunctional HDL
particles, as evidenced by increased small HDL particles (P<0.05) and reduced paraoxonase-
1 activities. Significant increase in both pro-oxidant-antioxidant balance and ischemia-
modified albumin (P<0.05), with simultaneously decreased activity of superoxide-dismutase
(P<0.05) was found in patients with progressive diabetic neuropathy, indicating the highest
degree of oxidative damage. It can be concluded that patients with microvascular
complications of DM have aggravated redox imbalance and lipid profile alterations. In
addition to metabolic control, strategies aimed at lowering OS and correcting dyslipidemia
can contribute to the prevention of microvascular complications of diabetes.

nije dovoljno razjašnjen. Cilj ovog istraživanja je bio ispitivanje produkata uznapredovale

AB  - The discovery of HIV and the study of molecular mechanisms crucial for the virus
replication cycle have led to the identification of important protein structures - potential
targets of drug action in AIDS therapy. One of the most significant discoveries is HIV-1
protease (PR), an enzyme that plays a key role in the HIV replication cycle (1). This study
aimed to test and demonstrate the interactions of newly synthesized chalcones (1,3-diaryl-2-
propen-1-one) as well as three commercial drugs (lopinavir, ritonavir and darunavir) in the
active site of HIV-1 protease using in sillico methods. and that the results obtained correlate
with the results of in vitro tests on the enzyme itself. The twenty structurally similar
chalcone derivatives were synthesized and their physico-chemical characterization was
performed. Docking calculations were performed using the Autodock Wine program in the
3D structure of the enzime catalytic site (pdb code: 6B36). The inhibition of enzyme activity
was monitored by fluorimetric method (2). The obtained results revealed that all compounds
showed anti-HIV-1 protease activity. Compound C1 showed the highest inhibitory activity
with an IC 50 values of 0.001 μM which is comparable with commercial product Darunavir.
The results obtained indicate that the synthesized compounds can be classified as potential
anti-HIV-1 protease inhibitors. Further research is focused on testing the ADMET properties
of the synthesized compounds as well as the synthesis of their analogues for which in silico
tests have shown satisfactory results.
AB  - Otkrić e HIV-a i istraživanje molekularnih mehanizama ključnih za ciklus replikacije
virusa dovelo je do identifikacije važnih proteinskih struktura - potencijalnih ciljnih mesta
dejstva lekova u terapiji AIDS-a. Jedno od najznačajnijih otkrića je HIV-1 proteaza (PR),
enzim koji ima ključnu ulogu u ciklusu replikacije HIV-a (1). Ova studija imala je za cilj da
primenom in sillico metoda ispita i prikaže interakcije novosintetisanih halkona (1,3-diaril-2-
propen-1-ona) kao i tri komercijalna leka (lopinavira, ritonavira i darunavira) u aktivnom

Antioxidant properties of HDL: beyond cardiovascular protection

AB  - High-density lipoprotein (HDL) is the most complex lipoprotein particle, containing
lipids and dozens of various functional proteins. Such sophisticated composition enables
numerous activities of HDL; from the reverse cholesterol transport, to antioxidative, anti-
inflammatory, anti-aggregation, antiadhesive, and vasodilatory effects. Accordingly, the
significance of HDL goes far beyond its cardioprotective properties and novel research
points towards its role in etiopathogenesis of various other diseases. Antioxidative
properties of HDL are primarily attributed to the enzyme paraoxonase 1 (PON1), whose
principal role is to protect low-density lipoprotein (LDL) and cell membranes against
harmful effects of reactive oxygen species (1). PON1 is located on HDL particles and its
activity largely depends on HDL structure. Our investigations have shown that PON1 is not
equally distributed across the entire population of serum HDL subfractions. Namely, our
results suggest that the allocation of PON1 on specific HDL subclasses changes in
pathophysiological conditions, such as chronic kidney disease, polycystic ovary syndrome, or
hypertension in pregnancy. Similarly, a shift in HDL subclasses distribution toward smaller,
dysfunctional particles is reported in diabetes, metabolic syndrome, obstructive sleep apnea,
sarcoidosis, but also in malignant diseases, such as colorectal cancer. In parallel,
antioxidative defense mechanisms were diminished in all these categories of patients, which
was evident as decreased PON1 activity and rise of oxidative stress. Structural modifications
of HDL particles affect their functions, thus antioxidative capability of PON1 depends on
qualitative properties of its lipoprotein carrier. Such complex interaction is highly significant
for the initiation and progression of numerous diseases.
AB  - Lipoprotein visoke gustine (high‐density lipoprotein, HDL) je najkompleksnija
lipoproteinska čestica koja, pored lipidnih komponenti, sadrži i desetine različitih
funkcionalnih proteina. Ovakav složen sastav omogućava brojne funkcije HDL; od reverznog
transporta holesterola, do antioksidativnih, antiinflamatornih, antiagregacijskih,
antiadhezivnih i vazodilatatornih svojstava. U skladu s tim, značaj HDL u mnogome
prevazilazi kardioprotektivne efekte, a savremena otkrića ukazuju na ulogu ovih čestica u
etiopatogenezi različitih oboljenja. Antioksidativna svojstva HDL prevashodno su vezana za
enzim paraoksonazu 1 (PON1), čija je osnovna funkcija zaštita lipoproteina niske gustine
(low‐density lipoprotein, LDL) i ćelijskih membrana od oksidativnih oštećenja (1). PON1 je
locirana na HDL česticama i uočeno je da aktivnost ovog enzima u velikoj meri zavisi od
strukture samog HDL. U našim istraživanjima pokazali smo da PON1 nije ravnomerno
zastupljena na svim subfrakcijama HDL, te da se ova zastupljenost menja u različitim
patofiziološkim stanjima, kao što su hronične bubrežne bolesti, sindrom policističnih jajnika,
ili hipertenzija u trudnoći. Slično tome, pomeranje raspodele HDL subfrakcija ka manjim,
disfunkcionalnim česticama uočeno je kod pedijatrijskih i odraslih pacijenata sa dijabetesom,
metaboličkim sindromom, opstruktivnom apnejom u toku spavanja, sarkoidozom, ali i sa
malignim bolestima, kao što je kolorektalni karcinom. Istovremeno, nivo antioksidativne
zaštite kod ovih pacijenata je bio snižen, što je bilo vidljivo i kao smanjenje aktivnosti PON1,
te sledstveno povećanje nivoa oksidativnog stresa. Strukturne modifikacije HDL čestica utiču
na njihovu funkciju, pa tako i antioksidativni kapacitet enzima PON1 zavisi od kvalitativnih
svojstava njegovog lipoproteinskog nosača. Ovakva složena interakcija između HDL i
pridruženih funkcionalnih proteina značajna je za nastanak i progresiju brojnih oboljenja.

principal role is to protect low-density lipoprotein (LDL) and cell membranes against
harmful effects of reactive oxygen species (1). PON1 is located on HDL particles and its
activity largely depends on HDL structure. Our investigations have shown that PON1 is not
equally distributed across the entire population of serum HDL subfractions. Namely, our
results suggest that the allocation of PON1 on specific HDL subclasses changes in
pathophysiological conditions, such as chronic kidney disease, polycystic ovary syndrome, or
hypertension in pregnancy. Similarly, a shift in HDL subclasses distribution toward smaller,
dysfunctional particles is reported in diabetes, metabolic syndrome, obstructive sleep apnea,
sarcoidosis, but also in malignant diseases, such as colorectal cancer. In parallel,
antioxidative defense mechanisms were diminished in all these categories of patients, which
was evident as decreased PON1 activity and rise of oxidative stress. Structural modifications
of HDL particles affect their functions, thus antioxidative capability of PON1 depends on
qualitative properties of its lipoprotein carrier. Such complex interaction is highly significant
for the initiation and progression of numerous diseases., Lipoprotein visoke gustine (high‐density lipoprotein, HDL) je najkompleksnija
lipoproteinska čestica koja, pored lipidnih komponenti, sadrži i desetine različitih
funkcionalnih proteina. Ovakav složen sastav omogućava brojne funkcije HDL; od reverznog
transporta holesterola, do antioksidativnih, antiinflamatornih, antiagregacijskih,
antiadhezivnih i vazodilatatornih svojstava. U skladu s tim, značaj HDL u mnogome
prevazilazi kardioprotektivne efekte, a savremena otkrića ukazuju na ulogu ovih čestica u
etiopatogenezi različitih oboljenja. Antioksidativna svojstva HDL prevashodno su vezana za
enzim paraoksonazu 1 (PON1), čija je osnovna funkcija zaštita lipoproteina niske gustine
(low‐density lipoprotein, LDL) i ćelijskih membrana od oksidativnih oštećenja (1). PON1 je
locirana na HDL česticama i uočeno je da aktivnost ovog enzima u velikoj meri zavisi od
strukture samog HDL. U našim istraživanjima pokazali smo da PON1 nije ravnomerno
zastupljena na svim subfrakcijama HDL, te da se ova zastupljenost menja u različitim
patofiziološkim stanjima, kao što su hronične bubrežne bolesti, sindrom policističnih jajnika,
ili hipertenzija u trudnoći. Slično tome, pomeranje raspodele HDL subfrakcija ka manjim,
disfunkcionalnim česticama uočeno je kod pedijatrijskih i odraslih pacijenata sa dijabetesom,
metaboličkim sindromom, opstruktivnom apnejom u toku spavanja, sarkoidozom, ali i sa
malignim bolestima, kao što je kolorektalni karcinom. Istovremeno, nivo antioksidativne
zaštite kod ovih pacijenata je bio snižen, što je bilo vidljivo i kao smanjenje aktivnosti PON1,
te sledstveno povećanje nivoa oksidativnog stresa. Strukturne modifikacije HDL čestica utiču
na njihovu funkciju, pa tako i antioksidativni kapacitet enzima PON1 zavisi od kvalitativnih
svojstava njegovog lipoproteinskog nosača. Ovakva složena interakcija između HDL i
pridruženih funkcionalnih proteina značajna je za nastanak i progresiju brojnih oboljenja.",

AB  - Oxidative stress is a phenomenon that occurs due to the disturbance in the balance
between the production of reactive oxygen species and the ability of biological systems to
remove the resulting compounds. Oxidative stress is involved in the pathogenesis of many
disorders in the human organism. This indicates the importance of quantification of
oxidative stress parameters in biological samples. Traditionally, these parameters are
determined by biochemical tests. Although these tests are routinely performed, they have
many drawbacks. To determine the exact concentration of selected compounds, more
sensitive analytical methods are becoming more important. In the modern scientific
literature, liquid chromatography-tandem mass spectrometry (LC-MS/MS) is increasingly
mentioned (1). This method, with adequate selection of stationary and mobile phases,
enables quantification of very low concentrations of selected parameters. In addition, these
methods can simultaneously determine the concentration of many selected components.
However, it is necessary to take into account that LC-MS/MS methods require a very purified
biological sample from which the proteins have been maximally removed. In this work, we
will present the results of determination of cysteine, cystine, reduced and oxidized
glutathione in patients with microcellular lung cancer. The use of LC-MS/MS methods is
becoming increasingly common for the analysis of oxidative stress markers in biological
fluids. In the future, we are expected to move to even more modern, fully automated
methods, which simultaneously purify and analyze samples.
AB  - Oksidativni stres je pojava koja nastaje usled narušavanja ravnoteže između
proizvodnje i nagomilavanja reaktivnih jedinjenja kiseonika u organizmu i sposobnosti
bioloških sistema da ukloni nastala jedinjenja. Oksidativni stres učestvuje u patogenezi
mnogih poremećaja kao što su kardiovaskularne bolesti, dijabetes i bolesti bubrega. Ovo
ukazuje na značaj određivanja odabranih parametara oksidativnog stresa u uzorcima
biološkog materijala. Tradicionalno, najveći broj parametara se određuje biohemijskim
testovima. Iako se ovi testovi rutinski izvode, oni imaju mnoge nedostatke. Da bi se odredila
tačna koncentracija izabranih parametara, koji se u uzorcima nalaze u veoma niskim
koncentracijama, sve veću prednost imaju osetljivije analitičke metode. U savremenoj
naučnoj literaturi se u te svrhe sve više spominje tečna hromatografija spregnuta sa
masenim detektorom (LC-MS/MS) (1). Ova metoda, uz adekvatan izbor stacionarne i
mobilne faze, omogućava kvantifikaciju veoma niskih koncentracija odabranih jedinjenja.
Sem toga, ovim metodama se istovremeno, u jednom uzorku, može odrediti koncentracija
većeg broja odabranih komponenti. Ipak, neophodno je voditi računa o tome da tečna
hromatografija spregnuta sa masenim detektorom zahteva dobro prečišćen uzorak biološkog
materijala iz kog su maksimalno uklonjeni proteini. U ovom radu biće prikazani rezultati
određivanja cisteina, cistina, redukovanog i oksidovanog glutationa kod pacijenata sa
mikrocelularnim karcinomom pluća. Upotreba LC-MS/MS metoda je sve uobičajenija za
analizu markera oksidativnog stresa u biološkim tečnostima zbog svoje osetljivosti. U
budućnosti se očekuje razvoj ka još savremenijim, potpuno automatizovanim metodama,
kojima se istovremeno uzorci prečišćavaju i analiziraju.

AB  - Oxidative stress is a pathphysiological phenomenon that occurs during various
physiological processes, as a consequence of aging, but also in many diseases. Oxidative
stress "eruption" occurs when its factors - free radicals are increased in pathological
conditions, when their creation overcomes the mechanisms of antioxidant protection.
Measurement of redox status parameters gives new biomarkers of various diseases. During
more than 25 years of dealing with this topic, we analyzed the redox status of a large number
of patients with various diseases: cardiovascular, stroke, kidney disease, diabetes, cancer,
obesity. Redox status was determined in a large number of healthy subjects, and in special
physiological conditions: pregnancy, athletes, children, elderly, in subjects after antioxidants'
supplementation. Oxidative stress markers measurement is applied in pharmacological
studies on several different animal models: atherosclerosis, multiple sclerosis, blast injuries,
poisoning, rheumatoid arthritis. These types of studies provide insight into cellular oxidative
stress development, mechanisms involved in the disease development, insight into the
possibility of its modulation under the therapy influence. Many years of experience in the
oxidative stress analysis, enabled us to develop a system for testing the antioxidant potential
of biologically active compounds in the serum pool of healthy individuals. The redox status of
the respondents is "chemical ID card", which changes in various diseases. "Redox imbalance"
diagnosis would allow determining the type of antioxidants' application, dosing regimen,
monitoring the therapy effects, which is the application of the principles of precision
(personalized) medicine, as the ideal of modern therapy.
AB  - Oksidativni stres je patofiziološki fenomen koji se javlja tokom različitih fizioloških
procesa, kao posledica starenja, ali i u mnogim bolestima. “Erupcija” oksidativnog stresa
nastaje kada se njegovi činioci tzv. slobodni radikali povećano stvaraju u određenim
patološkim stanjima, kada njihovo stvaranje nadvlada mehanizme i činioce antioksidativne
zaštite. Merenje parametara redoks statusa daje nove biomarkere različitih bolesti . Tokom
više od 25 godina bavljenja ovom temom analizirali smo redoks status velikog broja
pacijenata sa različitim bolestima: kardiovaskularnim, moždanim udarom, akutnim i
hroničnim bubrežnim bolestima, dijabetesom, kancerom, gojaznošću. Redoks status je
određivan i kod velikog broja zdravih ispitanika, kao i u posebnim fiziološkim stanjima:
trudnice, sportisti, deca, stare osobe, kao i kod ispitanika nakon suplementacije
antioksidansima. Merenje markera oksidativnog stresa je primenjeno i u farmakološkim
studijama na više različitih animalnih modela: ateroskleroza, multipla skleroza, blast
povreda, trovanja, reumatoidni artritis. Ovakve vrste studija omogućavaju uvid u razvoj
ćelijskog oksidativnog stresa, mehanizme koji su uključeni u razvoj bolesti, kao i uvid u
mogućnosti njegove modulacije pod uticajem terapije. Dugogodišnje iskustvo u analitici
oksidativnog stresa nam je omogućilo razvoj sistema za testiranje antioksidativnog
potencijala biološki aktivnih jedinjenja u serumskom pulu zdravih osoba. Rеdoks status
ispitanika je njegova “hеmijska lična karta”, koja se menja u različitim stanjima i
bolestima. Dijagnoza “rеdoks disbalansa“ kod odrеđеnе osobе, omogućila bi odrеđivanjе
vrstе i načina primеnе antioksidanasa, rеžim doziranja, praćеnjе еfеkata primеnе, što bi
predstavljalo primenu principa prеciznе (pеrsonalizovanе) medicine, kao ideala moderne
terapije.

AB  - Background and Objectives: Diabetic foot (DF) development is driven by complex interactions of hyperglycemia, inflammation, and oxidative stress (OS). We aimed to investigate OS and inflammatory biomarkers in patients with DF and their potential to improve early diagnosis and management of DF. Materials and Methods: The prooxidant–antioxidant balance (PAB), superoxide dismutase (SOD), total oxidative status (TOS), total sulfhydryl groups (SHG), routine biochemical parameters, and complete blood count were determined in 42 patients with type-2 DM, of which 23 patients had DF, while 19 patients were without DF complications. The neutrophils-to-lymphocyte ratio (NLR) was evaluated as a biomarker of inflammation. Results: Patients with DF had significantly higher (p < 0.05) PAB levels (170 ± 33.9 U/L) compared to those without DF complications (142 ± 31.3 U/L). In addition, patients with DF had significantly reduced SOD activities (p < 0.01). A positive correlation was found between the PAB and NLR index (r = 0.449; p < 0.05). Conclusions: In conclusion, the development of DF is associated with enhanced OS and inflammation processes. PAB and NLR could be useful non-invasive biomarkers of DF development.

AB  - Background. Metabolic alterations, particularly disorders of lipoprotein metabolism in COVID-19, may affect the course and outcome of the disease. This study aims at evaluating the lipoprotein profile and redox status in SARS-CoV-2 infected patients with different pneumonia severity and their association with lethal outcomes. Methods. The prospective cohort study was performed on 98 COVID-19 patients with mild, moderate, and severe pneumonia. Lipid and inflammatory parameters, lipoprotein subclasses, and redox status biomarkers were determined at the study entry and after one week. Results. Compared to patients with mild and moderate pneumonia, severely ill patients had higher oxidised low-density lipoprotein (oxLDL) and malondialdehyde levels and lower high-density lipoprotein cholesterol (HDL-C) concentrations and paraoxonase 1 activity. Reduction in the proportion of large HDL 2a subclasses with a concomitant increase in the proportion of smallest HDL 3c and small dense LDL (sdLDL) particles was observed in patients with severe disease during the time. However, these changes were reversed in the mild and moderate groups. The results showed a positive association between changes in oxLDL and total antioxidative status. However, prooxidants and antioxidants in plasma were lower in patients with lethal outcomes. Conclusions. Increased levels of oxLDL and sdLDL particles may contribute to the severity of COVID-19. The role of oxidative stress should be clarified in further studies, mainly its association with lethal outcomes.

Redox and biometal status in Wistar rats after subacute exposure to fluoride and selenium counter-effects

AB  - This study aimed to investigate the effect of 150 mg/L sodium fluoride (NaF) on redox status parameters and essential metals [copper (Cu), iron (Fe), and zinc (Zn)] in the blood, liver, kidney, brain, and spleen of Wistar rats and to determine the protective potential of selenium (Se) against fluoride (F-) toxicity. Male Wistar rats were randomly distributed in groups of five (n=5) receiving tap water (control) or water with NaF 150 mg/L, NaF 150 mg/L + Se 1.5 mg/L, and Se 1.5 mg/L solutions ad libitum for 28 days. Fluorides caused an imbalance in the redox and biometal (Cu, Fe, and Zn) status, leading to high superoxide anion (O2 .-) and malondialdehyde (MDA) levels in the blood and brain and a drop in superoxide dismutase (SOD1) activity in the liver and its increase in the brain and kidneys. Se given with NaF improved MDA, SOD1, and O2 .- in the blood, brain, and kidneys, while alone it decreased SH group levels in the liver and kidney. Biometals both reduced and increased F- toxicity. Nadomjesni Se u kombinaciji s NaF pozitivno je utjecao na razine MDA, SOD1 i O2.- u krvi, mozgu i bubrezima, a sâm Se smanjio je razine SH skupina u jetri i bubrezima. Izloženost fluoridu uzrokovala je sniženje, ali i porast koncentracija biometala. Nužna su dodatna istraživanja kako bi se ispitali antioksidacijski učinci Se na toksičnost izazvanu F-., Cilj ovog istraživanja bio je utvrditi djelovanje 150 mg/L natrijevog fluorida (NaF) na redoks-status i koncentracije esencijalnih elemenata [bakar (Cu), željezo (Fe) i cink (Zn)] u krvi, jetri, bubrezima, mozgu i slezeni Wistar štakora te mogući zaštitni učinak selena (Se) od toksičnosti prouzročene fluoridom (F-). Mužjaci Wistar štakora nasumično su razvrstani u četiri skupine (n=5), nakon čega su 28 dana konzumirali običnu vodu ili vodu s otopinom NaF 150 mg/L, NaF 150 mg/L + Se 1,5 mg/L ili Se 1,5 mg/L. Izloženost fluoridu dovela je do poremećaja redoks-parametara i koncentracija istraživanih biometala. Utvrđene su povišene razine superoksid aniona (O2.-) i malondialdehida (MDA) u krvi i mozgu, smanjena aktivnost superoksid dismutaze (SOD1) u jetri te njezin porast u mozgu i bubrezima. AB  - In parallel with the rapid growth of obesity, there is also an increase in the prevalence of type 2 diabetes mellitus (T2D) worldwide. Due to its compli- cations, cardiovascular diseases are the leading cause of death in those patients. In the last two decades, special attention has been given to ox- idative stress and inflammation, as the underlying mechanisms related to T2D occurrence and progression. Moreover, micro-ribonucleic acids (miRNAs) as new genetic biomarkers take an important place in the investigation of different metabolic pathways of insulin signaling. In this review article, we discuss microRNA modulation with oxidative stress and inflammation in pa- tients with T2D. Better insight into the novel potential therapeutic targets for treatment of diabetes and its complications is of utmost importance for public health.

AB  - Fast inverted, oil-in-water (o/w) emulsions, also known as SWitch-Oil-Phase (SWOP) emulsions, express the performances of both o/w and water-in-oil (w/o) emulsions during application to the skin, favoring their use as cosmetic carriers in sunscreen products. The objective of this study was to investigate the antioxidant potential (by 2 different methods) and the ultraviolet (UV) absorption ability of SWOP emulsion (S) with incorporated plant-based antioxidants dihydroquercetin (DHQ) and β-carotene (βC), using quercetin (Q) in a reference emulsion, in addition to the evaluation of their physicochemical properties and stability. A new biochemical extra- cellular model for in vitro assessment of antioxidative properties for the SWOP emulsions (S, SQ, SDHQ, and SDHQβC) was developed and compared with the results of 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. The analyses were performed at 20 °C and 37 °C, and oxidative stress parameters were monitored and statistically analyzed. The sun protection factor (SPF) of the samples was determined in vitro. Q and DHQ incorporated into the SWOP emulsion exhibited a strong DPPH radical scavenging ability. Neither incorporated nor pure βC showed DPPH radical scavenging ability at the tested concentrations. Contrary to that, in the bioenvironment conditions, SDHQβC showed minor antioxidative effects increase and also a significant decrease in exogenous pro-oxidative effects, caused by pro-oxidant, when compared to SDHQ. The obtained SPFs of SDHQβC, SDHQ, and SQ were 5.19, 4.65, and 3.35, respectively. The phys- icochemical stability of the emulsions was satisfactory during 1 month storage. The presented results demonstrated that the SWOP emul- sion is a suitable carrier for antioxidants with a photoprotective ability. The novel biochemical approach could be used in addition to DPPH assay with several advantages, relevant for the testing of antioxidant activity of potential active ingredients in cosmetic products.

AB  - Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a highly
transmittable and heterogenic infection of the respiratory tract, characterized by a broad spectrum
of clinical manifestations with a different degree of severity. Medical laboratories play an
important role in early diagnosis and management of Coronavirus Disease 2019 (COVID-19)
patients. Indeed, the results of several laboratory tests are essential for assessing the severity of
the disease, selecting appropriate therapeutic procedures and monitoring treatment response.
Routine laboratory testing in COVID-19 patients includes biomarkers of acute phase reaction,
hematological and biochemical parameters that indicate tissue injury. The aim of this review
paper is to describe the role of these biomarkers in the diagnostics and management of adult and
pediatric COVID-19 patients.
AB  - Novi koronavirus SARS-CoV-2 je uzročnik lako prenosive i veoma heterogene infekcije respiratornog trakta, koju karakteriše širok spektar kliničkih manifestacija, različitog stepena težine. Medicinske laboratorije igraju važnu ulogu u ranoj dijagnostici i praćenju pacijenata sa COVID-19. Rezultati nekoliko laboratorijskih testova su od suštinskog značaja za procenu težine bolesti, odabir odgovarajućih terapijskih procedura i praćenje odgovora na terapiju. Rutinska laboratorijska ispitivanja kod pacijenata sa COVID-19 uključuju biomarkere akutne faze, hematološke i biohemijske parametre koji ukazuju na oštećenje tkiva. Cilj ovog rada je da se prikaže uloga tih biomarkera u dijagnostici i praćenju toka bolesti kod odraslih i pedijatrijskih pacijenata sa COVID-19.

AB  - Due to the increased prevalence of obesity-related complications, especially type 2 diabetes
mellitus (DM2), great attention has been paid to the variety of adipokines and cytokines, i.e.
biomarkers, which are secreted by the enlarged visceral adipose tissue. Those pro-inflammatory
mediators diminish insulin sensitivity via impairing insulin signalling pathways. If these
processes persist, insulin resistance and DM2 occur, with a typical finding of low-grade chronic
inflammation. The current review focuses on some hematological indices that reflect the
inflammation level, such as white blood cell count, its subsets, and derived indexes. Those
parameters are easy to measure, cost-effective, and widely available in a primary care setting. A
deeper knowledge of the changes in the mentioned hematological parameters in DM2, in addition
to glucoregulation biomarkers, may enable physicians to act promptly when patients with this
metabolic disorder are concerned.
AB  - Zbog povećane prevalence komplikacija povezanih sa gojaznošću, naročito diabetes mellitus-a tip (DM2), posebna pažnja se posvećuje različitim adipokinima i citokinima, tj. biomarkerima koje luči uvećano visceralno masno tkivo. Pomenuti medijatori inflamacije smanjuju insulinsku senzitivnost vršeći uticaj na signalne puteve insulina. Ukoliko ovi procesi potraju duže dolazi do pojave insulinske rezistencije i DM2, sa tipičnim nalazom hronične inflamacije niskog stepena. Ovaj revijalni članak akcenat stavlja na određene hematološke parametre koji odražavaju stepen inflamacije, kao što je broj leukocita, njihove podvrste, kao i indeksi izvedeni iz istih. Ovi parametri su lako merljivi, ekonomski isplativi i široko dostupni u ambulantama primarne zdravstvene zaštite. Bolje poznavanje promena navedenih hematoloških parametara u DM2, pored biomarkera glikoregulacije, može omogućiti lekarima pravovremenu reakciju kada se radi o pacijentima sa ovim metaboličkim poremećajem.

deeper knowledge of the changes in the mentioned hematological parameters in DM2, in addition
to glucoregulation biomarkers, may enable physicians to act promptly when patients with this
metabolic disorder are concerned., Zbog povećane prevalence komplikacija povezanih sa gojaznošću, naročito diabetes mellitus-a tip (DM2), posebna pažnja se posvećuje različitim adipokinima i citokinima, tj. biomarkerima koje luči uvećano visceralno masno tkivo. Pomenuti medijatori inflamacije smanjuju insulinsku senzitivnost vršeći uticaj na signalne puteve insulina. Ukoliko ovi procesi potraju duže dolazi do pojave insulinske rezistencije i DM2, sa tipičnim nalazom hronične inflamacije niskog stepena. Ovaj revijalni članak akcenat stavlja na određene hematološke parametre koji odražavaju stepen inflamacije, kao što je broj leukocita, njihove podvrste, kao i indeksi izvedeni iz istih. Ovi parametri su lako merljivi, ekonomski isplativi i široko dostupni u ambulantama primarne zdravstvene zaštite. Bolje poznavanje promena navedenih hematoloških parametara u DM2, pored biomarkera glikoregulacije, može omogućiti lekarima pravovremenu reakciju kada se radi o pacijentima sa ovim metaboličkim poremećajem.",
