Anthropogenic influence on seasonal and spatial variation in bioelements and non-essential elements in honeybees and their hemolymph

AB  - Honeybee colony losses have been a focus of research in the last years, due to the importance of managed honeybee colonies for economy and ecology. Different unfavorable conditions from the outside environment have a strong impact on the hive health. The majority of losses occur mainly during winter and the exact reason is not completely understood. Only a small number of studies are dealing with content of bioelements, their function and influence on honeybee physiology. The aim of the present study was to determine seasonal and spatial variations in content of bioelements and non-essential elements, in hemolymph and whole body of honeybees originating from three regions with different degrees of urbanization and industrialization. Concentrations of 16 elements were compared: macroelements (Ca, K, Mg, Na), microelements (Cu, Fe, Mn, Zn) and non-essential elements (Al, Ba, Cd, Co, Cr, Ni, Pb, Sr) in samples collected from 3 different environments: Golija (rural region), Belgrade (urban region) and Zajača (industrial region). Content of bioelements and non-essential elements in honeybees was under noticeable influence of the surrounding environment, season and degree of honeybee activity. Hemolymph was proven to be helpful in differentiating air pollution from other sources of honeybee exposure. The results of our study demonstrated that bees can be successfully used as biomonitors since we have observed statistically significant differences among observed locations, but unless compared locations are exposed to excessively different pollution pressures, it is essential that all bees should be collected at the same season.

Influence of Dental Restorations on Oxidative Stress in Gingival Crevicular Fluid

AB  - Biocompatibility of dental materials (DM) can be evaluated by gingival crevicular fluid (GCF) oxidative stress (OS) status. The goal of the study was to ascertain influence of dental caries degree, teeth position, and type and amount of applied DM on GCF OS profile. For this purpose, we tested six DMs that were sealed in one session: amalgam (Amg), composites: Tetric EvoCeram and Beautifil (BF), phosphate cement-zinc phosphate and polycarboxylate cements zinc polycarboxylate cements, and glass ionomer cement (GIC). The study included 88 dental outpatients. Follow-up was scheduled at 7th and 30th day. Oxidative stress parameters (malondialdehyde (MDA) and glutathione (GSH) levels and total superoxide dismutase (tSOD) activity) were measured before (0th day) and after the treatment (7th and 30th day) in GCF. Control teeth were mirror-positioned healthy teeth. The DM accomplished the following effects (listed in descending order): increase of GSH in GCF was realized by ZPoC > BF > GIC > Amg; tSOD activity increase by ZPoC > BF > Amg; and MDA decrease by ZPoC > ZPhC > Amg > TEC. Dental caries provokes insignificant rise of OS in GCF. ZPoC and ZPhC showed the highest antioxidant effect, contrary to GIC. Restorations with antioxidant properties may reduce gum diseases initiated by caries lesion, what is of great clinical relevance in dentistry.

Correlating household second-hand smoke exposure and respiratory symptoms in school children

AB  - Objective. Aim of this paper was to correlate exposure to household second-hand smoke and respiratory symptoms in participating school children, who are attending ten primary schools in Belgrade, Serbia. Method. In order to determine correlation between second-hand smoke exposure at home and respiratory symptoms in primary school pupils, a comparative cross-sectional study has been conducted. The study was conducted in ten primary schools on 735 children, in 44 classrooms in Belgrade, Serbia. A standardized questionnaire was distributed to parents/caregivers, on child's respiratory health, socioeconomic status and parents' lifestyle habits, primarily cigarette smoking. Results. 78% of all exposed children have parents smoking up to 20 cigarettes/day, while more than 22% are exposed to parents' household second-hand smoke of more than 21 cigarettes per day (p lt 0.0001). Respiratory disorders appear in 45.9% of children exposed to domestic second-hand smoke (CI 1.04-1.89), asthmatic symptoms in 33.5% (CI 0.94-1.78), while the least incidence of disorders is reported for continuous cough, which have lasted for 3 months (3.3%, CI 0.44-2.36), wheezing after exercise (4.5%, CI 0.38-1.41) and being awaken by wheezing in last 12 months (7.4%, CI 0.89-3.28). Conclusion. Univariate logistic regression analysis proved that parental smoking exposure is a statistically significant predictor for childhood respiratory symptoms' manifestation.
AB  - Cilj. Cilj ovog rada je uspostavljanje korelacije između izloženosti pasivnom pušenju u kućnom okruženju i pojave respiratornih simptoma kod dece koja pohađaju deset osnovnih škola u Beogradu, Srbija. Metod. Istraživanje je sprovedeno po tipu komparativne studije preseka, u deset osnovnih škola, na uzorku od 735 dece, u 44 učionice, u Beogradu, Srbija. Roditeljima/starateljima distribuiran je standardizovani upitnik, sa pitanjima o respiratornom zdravlju dece, socio-ekonomskom statusu i navikama roditelja, pre svega o pušenju cigareta. Rezultati. 78% dece koja su izložena duvanskom dimu ima roditelje koji puše do 20 cigareta/dan, dok je više od 22% izloženo duvanskom dimu od popušenih preko 21 cigareta/dan, od strane roditelja (p lt 0,0001). Respiratorni poremećaji prisutni su kod 45,9% dece izložene pasivnom pušenju u kućnom okruženju (CI 1.04-1.89), astmatični simptomi kod 33,5% (CI 0,94-1.,78), dok je najniža incidenca ovih poremećaja prijavljena u slučaju hroničnog kašlja, u trajanju od 3 meseca (3,3%, CI 0,44-2,36), vizinga nakon fizičke aktivnosti (4,5%, CI 0,38-1,41), kao i za slučaj buđenja usled viziga u poslednjih 12 meseci (7,4%, CI 0,89-3,28). Zaključak. Univarijantnom logističkom regresionom analizom dokazano je da je izloženost roditeljskom pušenju duvana statistički značajan prediktor za pojavu respiratornih simptoma kod dece.

AB  - The objective of our study was to examine testicular toxicity of cadmium (Cd), focusing on oxidative stress (OS), essential metals and androgenic status and morphological changes. Male Wistar rats [controls and four Cd-subgroups (n = 6) organized according to the exposure (1, 3, 10 and 21 days)] were intraperitoneally (i.p.) treated with 1 mg CdCl2/kg/day. Testicular Cd deposition was noticed from the 1st day. After 10 and 21 days, copper (Cu) and iron (Fe) increased by 60-109% and 43-67%, respectively, while zinc (Zn) decreased by 24-33%. During 121 days of the exposure, decrease in testicular total superoxide dismutase (SOD) and total glutathione-s-transferase (GST) activities occurred gradually by 30-78% and 15-84%, respectively, while superoxide anion radical (O-2(center dot-)) increased gradually by 114-271%. After 10-21 days, decrease in testicular catalase (CAT) activity appeared by 13-31%. After 21 days, malondialdehyde (MDA) decreased by 44% and the ratio of oxidized glutathione/reduced glutathione (GSSG/GSH) increased by 130% in testes of the rats exposed to Cd. Additionally, decreased testicular testosterone level and the relative testes mass, along with induced microscopic and macroscopic changes were occured, what can be explained as the consequence of instantly developed OS, impaired essential metals status and Cd testicular deposition.

Testicular Cd deposition was noticed from the 1st day. After 10 and 21 days, copper (Cu) and iron (Fe) increased by 60-109% and 43-67%, respectively, while zinc (Zn) decreased by 24-33%. During 121 days of the exposure, decrease in testicular total superoxide dismutase (SOD) and total glutathione-s-transferase (GST) activities occurred gradually by 30-78% and 15-84%, respectively, while superoxide anion radical (O-2(center dot-)) increased gradually by 114-271%. After 10-21 days, decrease in testicular catalase (CAT) activity appeared by 13-31%. After 21 days, malondialdehyde (MDA) decreased by 44% and the ratio of oxidized glutathione/reduced glutathione (GSSG/GSH) increased by 130% in testes of the rats exposed to Cd. Additionally, decreased testicular testosterone level and the relative testes mass, along with induced microscopic and macroscopic changes were occured, what can be explained as the consequence of instantly developed OS, impaired essential metals status and Cd testicular deposition.",

AB  - Understanding the cellular stress response in honey bees will significantly contribute to their conservation. The aim of this study was to analyze the response of the antioxidative enzymes superoxide dismutase and catalase in honey bees related to the presence of toxic metals in different habitats. Three locations were selected: (i) Tunovo on the mountain Golija, as control area, without industry and large human impact, (ii) Belgrade as urban area, and (iii) Zajaca, as mining and industrial zone. Our results showed that the concentrations of lead (Pb) in whole body of bees vary according to habitat, but there was very significant increase of Pb in bees from investigated industrial area. Bees from urban and industrial area had increased expression of both Sod1 and Cat genes, suggesting adaptation to increased oxidative stress. However, in spite increased gene expression, the enzyme activity of catalase was lower in bees from industrial area suggesting inhibitory effect of Pb on catalase.

AB  - Introduction: Contact herbicide diquat (DQ), redox cycling compound, mediates its systemic toxicity throughout the enlarged production of free radicals. Target organs are liver and kidney in humans. To-date, the mechanism of DQ-induced neurotoxicity has not been rationalized. Objective: The objectives of the study were to examine the ability of DQ to induce oxidative stress (OS) and/or nitrosative stress (NS) upon intrastriatal (i.s.) administration and to investigate the role of nitric oxide (NOx) using NG-nitro-L-arginine methyl ester (L-NAME), a non-selective inhibitor of nitric oxide synthase (NOS) in the pretreatment of DQ i.s. administration. Material and Methods: The experiment was conducted on Wistar rats, randomly divided in experimental groups, receiving different treatments i.s. applied. Parameters of OS/NS such as: superoxide anion radical (O-2(center dot-)), superoxide dismutase (SOD), malondialdehyde (MDA) and nitrates (NO3-) were measured in the cortex (bilaterally), at 30th min, 24 hours and 7 days after the treatments. Results: Lethargy and high mortality rate were observed only in the DQ group (within 24 hours and 2-3 hours, respectively) after awakening from anesthesia. Markedly increased production of NOx and O-2(center dot-) along with elevated lipid peroxidation altogether contributed to DQ neurotoxicity. The most importantly, the L-NAME i.s. pretreatment protected treated animals from dying and diminished OS/NS response against DQ-induced neurotoxicity. Conclusion: The i.s. pretreatment with L-NAME resulted in neuroprotection against DQ neurotoxity, based on animal survival and reduced LPO in the cortex.

AB  - Most commonly observed central nervous system (CNS) effects induced by systemic toxicity of herbicide diquat (DQ) are general depression and lethargy. Generally, it is accepted that DQ exerts its toxicity through the production of superoxide anion radical (O2 ●-) during its redox metabolism in the presence of molecular oxygen, which further initiates radical chain reaction, contributing developing of oxidative stress (OS) as well. Mechanisms of DQ neurotoxic effect is not rationalized till now. The objective of the study was to examine whether OS contributes to DQ neurotxicity. For this purpose, male Wistar rats were intrastriataly (i.s.) treated with DQ and oxidative status parameters such as: superoxide anion radical (O2 ●-); nitrates (NO3 -), as a final metabolite of reactive nitogen species; malondialdehyde (MDA), an indicator of lipid peroxidation; activity of superoxide dismutase (SOD); glutathione peroxidase (GPx), and glutathione (GSH), were measured in the hippocampus at 30 minutes, 24 hours and 7 days post treatment. Noteworthy, mortality rate (30 - 40 %) was observed in the group of rats treated with DQ, within 2-3 hours after awakening from anesthesia. Additionally, lethargy was the only neurological symptom observed in that group. Analyzed parameters indicate that OS mediates DQ neurotxicity, which is documented with significant increase of lipid peroxidation.
AB  - Najčešće zapaženi efekti sistemskog trovanja herbicidom dikvatom (DK) na centralni nervni sistem (CNS) su opšta depresija i letargija. Opšte je prihvaćeno da se toksičnost DK ostvaruje posredstvom povećanog stvaranja superoksid anjon radikala (O2 ●-) tokom njegovog redoks metabolizma, u prisustvu molekularnog kiseonika, koji dalje inicira lančanu reakciju radikalskog tipa i razvoj oksidativnog stresa (OS). Do danas mehanizmi neurotoksičnog efekta DK nisu u potpunosti poznati. Cilj ove studije je bio da ispitamo da li OS posreduje u neurotoksičnosti indukovanoj DK. Eksperiment je sproveden na mužjacima Wistar pacova, intrastrijatalno tretiranih (i.s.) DK. Parametri oksidativnog statusa, kao što su: superoksid anjon radikal (O2 ●-), nitrati (NO3 -), kao finalni metaboliti reaktivnih vrsta azota; malondialdehid (MDA), indikator lipidne peroksidacije; aktivnost enzima; superoksid dizmutaze (SOD) i glutation peroksidaze (GPx); i glutation (GSH) mereni su u hipokampusu, 30 minuta, 24 sati i 7 dana posle tretmana. Effect of Magnesium Supplementation on the Distribution Patterns of Zinc, Copper, and Magnesium in Rabbits Exposed to Prolonged Cadmium Intoxication

AB  - The present study is designed to investigate whether magnesium (Mg) supplementation may prevent Cd-induced alterations in zinc (Zn), copper (Cu), and magnesium (Mg) status in rabbits. For this purpose, the concentrations of Zn, Cu, and Mg were estimated in blood, urine, and organs (brain, heart, lungs, liver, kidney, spleen, pancreas, skeletal muscle, and bone) of rabbits given Cd (10 mg/kg b.w.) and rabbits cotreated with Mg (40 mg/kg b.w.) orally, as aqueous solutions of Cd chloride and Mg acetate every day for 4 weeks. Samples were mineralized with conc. HNO3 and HClO4 (4:1) and metals concentrations were determined by atomic absorption spectrophotometry (AAS). Magnesium supplementation succeeded to overcome Cd-induced disbalance of investigated bioelements. Beneficial effects of Mg were observed on Zn levels in blood and urine, on Cu levels in urine, and on Mg levels in blood. Magnesium pretreatment also managed to counteract or reduce all Cd-induced changes in levels of Cu and Mg in organs, while it did not exert this effect on Zn levels. These findings suggest that enhanced dietary Mg intake during Cd exposure can have at least partly beneficial effect on Cd-induced alterations in homeostasis of zinc, copper, and magnesium.

abstract = "The present study is designed to investigate whether magnesium (Mg) supplementation may prevent Cd-induced alterations in zinc (Zn), copper (Cu), and magnesium (Mg) status in rabbits. For this purpose, the concentrations of Zn, Cu, and Mg were estimated in blood, urine, and organs (brain, heart, lungs, liver, kidney, spleen, pancreas, skeletal muscle, and bone) of rabbits given Cd (10 mg/kg b.w.) and rabbits cotreated with Mg (40 mg/kg b.w.) orally, as aqueous solutions of Cd chloride and Mg acetate every day for 4 weeks. Samples were mineralized with conc. HNO3 and HClO4 (4:1) and metals concentrations were determined by atomic absorption spectrophotometry (AAS). Magnesium supplementation succeeded to overcome Cd-induced disbalance of investigated bioelements. Beneficial effects of Mg were observed on Zn levels in blood and urine, on Cu levels in urine, and on Mg levels in blood. Magnesium pretreatment also managed to counteract or reduce all Cd-induced changes in levels of Cu and Mg in organs, while it did not exert this effect on Zn levels. These findings suggest that enhanced dietary Mg intake during Cd exposure can have at least partly beneficial effect on Cd-induced alterations in homeostasis of zinc, copper, and magnesium.",

AB  - Lead manifests toxic effects in almost all organs and tissues, especially in: the nervous system, hematopoietic system, kidney and liver. This metal has a special affinity for deposition in hard tissue, i.e., bones and teeth. It is generally believed that the main mechanism of its toxicity relies on its interaction with bioelements, especially with Ca and Mg. This article analyses the influence of Pb poisoning on Ca and Mg content in hard tissues, (mandible, femur, teeth and skull) of female and young rats. Experiments were carried out on 60 female rats, AO breed, and on 80 of their young rats (offspring). Female rats were divided into three groups: the first one was a control group, the second one received 100 mg/kg Pb2+ kg b.wt. per day in drinking water, the third one received 30 mg/kg Pb2+ kg b.wt. per day in drinking water. Young rats (offspring) were divided into the same respective three groups. Lead, calcium and magnesium content in hard tissues (mandible, femur, teeth-incisors and skull) was determined by flame atomic absorption spectrophotometry in mineralized samples. There was a statistically significant Pb deposition in all analyzed female and young rat hard tissues. Ca and Mg contents were significantly reduced in all female and young rat hard tissues. These results show that Pb poisoning causes a significant reduction in Ca and Mg content in animal hard tissues, which is probably the consequence of competitive antagonism between Pb and Ca and Mg.

The influence of chronic lead poisoning on the activity of some serum enzymes in rats

AB  - In this paper, the influence of chronic lead intoxication on the activity of serum enzymes aspartate and alanine aminotransferases (AST and ALT) and alkaline phosphatase (ALP) was examined. The experiment was performed on 130 adult female DA rats and 80 young rats. Rats were treated by lead-acetate 100 and 30 mg Pb per kg body weight for 10, 20, 30, 40, 50 and 60 days. Young rats (offspring of studied female rats) were treated with lead only through the placenta and mother's milk. The activities of serum AST, ALT and ALP were determined spectrophotomerically by IFCC method. The activity of examined serum enzymes was significantly increased in conditions of chronic lead intoxication in female rats and their offspring in relation to the control group. The activity of serum AST, ALT and ALP was in a positive correlation with the time of intoxication. There were no significant differences between the activities of enzymes AST and ALT in the serum and the amount of lead. The activity of ALP was significantly higher in serum of rats treated with higher amounts of lead. Increased AST, ALT and ALP activity in serum is most likely the consequence of lead hepatotoxicity.
AB  - U ovom radu su prikazani rezultati ispitivanja uticaja hroničnog trovanja olovom na aktivnost enzima aspartat i alanin aminotransferaze (AST i ALT) kao i alkalne fosfataze (ALP) u krvnom serumu pacova. Eksperiment je izveden na 130 ženki pacova i njihovih 80 mladunaca. Ženke su tretirane olovo-acetatom oralno putem pijaće vode u dve doze-100 i 30 mg/kg TM dnevno u trajanju od 10,20,30, 40, 50 i 60 dana, dok su njihovi mladunci olovo primali samo preko placente i putem mleka. Nakon žrtvovanja, od životinja je uzimana krv i u krvnom serumu je spektrofotometrijski, na automatskom analizatoru Hitachi 911, određivana aktivnost enzima AST, ALT i ALR Dobijeni rezultati su ukazali da hronično trovanje olovom ima za posledicu značajno povećanje aktivnosti enzima AST, ALT i ALP u krvnom serumu ženki pacova, pri čemu se aktivnost navedenih enzima linearno povećavala sa povećanjem vremena intoksikacije. Nisu zapažene značajne razlike u aktivnosti enzima AST i ALT u odnosu na primenjenu dozu olova, dok je u slučaju ALP ustanovljeno da veća doza olova ima za posledicu značajnije povećanje aktivnosti ovog enzima u krvnom serumu. U mladunaca ženki koje su tokom trudnoće i laktacije primale olovo, takođe je ustanovljeno značajno povećanje aktivnosti navedenih enzima u krvnom serumu u odnosu na kontrolnu grupu. Nisu ustanovljene značajne razlike u aktivnosti AST, ALT i ALP u serumu mladunaca u pogledu doze olova koje su njihove majke primale. Povećanje aktivnosti enzima AST, ALT i ALP u krvnom serumu ženki i mladunaca je najverovatnije posledica hepatotoksičnog dejstva olova udruženog sa intrahepatičkom holestazom.

Our previous results suggested that oral Mg treatment have better effect on investigation biochemical parameters (protoporphyrins, aminolevulinic acid - ALA and d-aminolevulinic dehydrataseALA-D) used in evaluating Pb intoxication, then CaNa(2)EDTA, chelation agens, currently used in therapy of Pb intoxication. The toxic effect of Pb induced considerably modifies the activity of many other enzymes. In this work we have examined the influence of Mg (as alternative therapy of Ph poisoning) on enzymes activity biochemical markers for general health conditions - aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) in condition of lead intoxication. Many studies showed disturbances of activity ALT, AST and ALP The aim of this study was to confirm positive effects of Mg intake in condition of such intoxication at the level on activity of investigated enzymes. The experiment was performed on 45 male Wister rats, divided in three groups. I - control group; II - group treated daily for 30 days with 100 mg Pb, per kg body weight and next 60 without Pb treatment (spontaneous detoxication); III group - the same treatment as II group for the first 30 days, but next 60 days rats were treated orally with 40 mg Mg/kg body weight. Activity of AST and ALT was significant increased in condition of Pb poisoning, but ALP activity was significant reduced. Influence of excessive oral Mg treatment was positive: decrease of AST activity and ALT activity, which was probably in correlation with significant elimination of Ph from liver and increase of ALT enzyme activity at the normal level.

Contribution to interaction between magnesium and toxic metals: The effect of prolonged cadmium intoxication on magnesium metabolism in rabbits

AB  - The results obtained up to now indicate that increased intake of some heavy metals causes disorder of bioelements metabolism leading to their blood and organs decrease and higher elimination via the urine. As to lead and magnesium, our investigations indicated that the reaction is reversible and that increased intake of Mg eliminates Pb via urine. Some other findings suggest similar relations between Mg and certain heavy metals, thus pointing to significant role of Mg in toxicology of heavy metals and announcing a new chapter in the toxicology of metals entitled 'Mg in professional and ecotoxicology'. In this report we present the results of our investigations on the effect of Cd on Mg metabolism. The experiment was performed on rabbits given orally, every day, for 4 weeks 10 mg Cd/kg b.w. as aqueous solution of CdCl 2 . Magnesium content was determined in blood, urine, soft tissues and bones by the AAS method. Under the experimental conditions, Cd lead to statistically significant decrease of blood Mg (p < 0.001 after day 16) which was associated with increased Mg elimination via urine (p < 0.01). Statistically significant changes were not detected in the tissues, except in the liver where we found enhanced Mg content (p < 0.05), while its level in the muscles decreased (p < 0.01).

abstract = "The results obtained up to now indicate that increased intake of some heavy metals causes disorder of bioelements metabolism leading to their blood and organs decrease and higher elimination via the urine. As to lead and magnesium, our investigations indicated that the reaction is reversible and that increased intake of Mg eliminates Pb via urine. Some other findings suggest similar relations between Mg and certain heavy metals, thus pointing to significant role of Mg in toxicology of heavy metals and announcing a new chapter in the toxicology of metals entitled 'Mg in professional and ecotoxicology'. In this report we present the results of our investigations on the effect of Cd on Mg metabolism. The experiment was performed on rabbits given orally, every day, for 4 weeks 10 mg Cd/kg b.w. as aqueous solution of CdCl 2 . Magnesium content was determined in blood, urine, soft tissues and bones by the AAS method. Under the experimental conditions, Cd lead to statistically significant decrease of blood Mg (p < 0.001 after day 16) which was associated with increased Mg elimination via urine (p < 0.01). Statistically significant changes were not detected in the tissues, except in the liver where we found enhanced Mg content (p < 0.05), while its level in the muscles decreased (p < 0.01).",

Compared effects of high oral Mg supplements and of EDTA chelating agent on chronic lead intoxication in rabbits

AB  - Our previous experiments showed that excessive oral Mg intake has beneficial effects in chronic Pb poisoning, inducing decrease of Pb body burden and its increased elimination via urine. The aim of this work was to investigate and to compare the effect of Mg on urinary Pb elimination with the effect of calcium-disodium edetate (CaNa(2)EDTA) - chelating agent currently used in therapy of chronic Pb intoxication. Besides, under the same experimental conditions, biochemical parameters protoporphyrin IX (ppIX), zinc protoporphyrin (Znpp) and delta-aminolevulinic acid (ALA) were determined. Experiments were carried out on rabbits preciously intoxicated for 4 weeks with 20 mg Pb/kg b.w. per day. After the period of intoxication, one group of animals was given per os, for 28 days, 40 mg Mg/kg, the other one was i.v. treated for 7 days with 15 mg CaNa(2)EDTA/kg (therapeutic doses), while the third one [rabbits without therapy] served as a control. During the period of detoxication, lead was determined in blood and urine samples, and ppIX and Znpp were determined in blood and ALA in urine. Results suggest that oral treatment with magnesium, although inducing later Pb elimination than EDTA, has even better effect on investigated biochemical parameters than chelating therapy.
