AB - Dry MeOH extract of Ferula heuffelii (Apiaceae) underground parts was tested for spasmolytic, gastroprotective and antioxidant activities. HPLC analysis revealed that chlorogenic acid (CGA; 34.6 mg/g) was its main constituent. Extract in vitro exhibited notable total antioxidant activity (FRAP value=1.0 μmol Fe2+/mg), and scavenging of DPPH (SC50=62.5 μg/ml) and •OH radicals (49.5 % at 20 μg/ml in 2-deoxyribose assay). In vitro on isolated rat ileum, extract exhibited significant spasmolytic activity, i. e., it showed 124.6 % of maximal atropine effect on spontaneous contractions (at 100 μg/ml), and reduced spasmogenic effect of KCl (80 mm) to 44.4 % (at 60 μg/ml) and of highest applied concentration of ACh to 26.3 % (at 120 μg/ml). In parallel experiments, spasmolytic effect of CGA was also demonstrated. In acute EtOH-induced gastric ulceration model in rats, extract (100 mg/kg p.o.) showed significant gastroprotective effect (gastric damage score 0.50), similar to ranitidine (20 mg/kg p.o.). Obtained results showed that tested F. heuffelii polar extract represents new herbal preparation with potential use against some gastrointestinal complaints.

AB - The plants of genus Cymbopogon are well known for their use in traditional medicine and for their high content of essential oils that are widely used as flavoring agents, fragrances, cosmetics, and pharmaceuticals. Essential oils isolated from the dried stems and inflorescence of cultivated Cymbopogon schoenanthus (L.) Spreng. (Poaceae) collected from Khartoum state (Sudan) in two periods were studied. The results of chemical analysis of C. schoenanthus revealed that this plant is rich in essential oil which is a valuable source of the commercially important monoterpene, piperitone (47.7-71.5%). Other abundant constituents of the oils were intermedeol (6.1-17.3%), delta-2-carene (4.5-10.0%) and elemol (2.7-9.0%). The essential oil from the inflorescence was tested for spasmolytic activity using three different experimental models: against spontaneous contractions, contractions induced with acetylcholine and contractions induced with potassium chloride. The oil exhibited strong, significant and dose-dependent spasmolytic activity, indicating the possibility for further investigations of this essential oil for its medicinal purposes or application in food industry.

AB - ObjectivesAs there are no previous studies of the European wild-garlic (Allium ursinum) effects on the gastrointestinal system, despite its traditional applications in gastrointestinal disorders' treatment and regular use in the human diet, we have quantified and compared spasmolytic, antimicrobial and antioxidant activities of its different leaf extracts. MethodsWild-garlic extracts were tested for spasmolytic activity on isolated rat ileum, antimicrobial activity on selected Gram-positive and Gram-negative bacteria and fungi by microdilution method and antioxidant capacity by DPPH radical-scavenging assay. Key findingsWild-garlic extracts were found to decrease ileal basal tone. As the relaxation of K+-induced contractions was similar to one caused by papaverin, the observed spasmolytic effect was most likely mediated through Ca2+-channel inhibition. Ethanolic extract (with the highest phenolic and high alk(en)yl cysteine sulphoxides' levels) produced the strongest spasmolytic activity. In case of acetylcholine-induced contractions, only hydromethanolic extract showed no statistical difference in comparison with positive control. All samples exhibited certain antioxidant potential and strong antimicrobial activity against tested enteropathogenic strains (Salmonella enteritidis was the most sensitive, followed by Escherichia coli, Proteus mirabilis and Enterococcus faecalis). ConclusionBesides other already established health-promoting effects, wild garlic could be useful in treatment of mild gastrointestinal disturbances.

AB - The composition of essential oils obtained by hydrodistillation from dried stems and inflorescences of Cymbopogon nervatus (Hochst.) Chiov. (Poaceae) growing wild in Western and Eastern Sudan and collected at two different periods, was analyzed by GC and GC MS. The most dominant constituents in the oils were p-menthadienols, trans-p-mentha-1(7),8-dien-2-ol (19.9-32.6%), cis-p-mentha-1(7),8-dien-2-ol (18.9-23.3%), trans-p-mentha-2,8-dien-l-ol (10.6-21.0%) and cis-p-mentha-2,8-dien-1-ol (8.1-10.4%). The essential oil of inflorescence from the Western Sudan was tested for spasmolytic activity using three different experimental models: against spontaneous contractions, and contractions induced with ACh and KCI. The oil exhibited strong, significant and concentration-dependent spasmolytic activity. Applied in concentration of 200 mu g/ml, the oil showed 88.44 +/- 16.78% of maximal spasmolytic effect of atropine (6.4 p,,M) against spontaneous contraction. Essential oil (90 mu g/ml) inhibited ACh-induced contractions as well, and reduced the effect of the highest applied concentration of ACh to 37.29 +/- 16.16%. Additionally, the oil exhibited strong activity against contractions induced with KCI (80 mM) and in concentration of 200 mu g/ml, completely abolished the contractile effect of KCI. The essential oil of inflorescence from the Western Sudan was also tested for antioxidant activity by DPPH assay and showed moderate anti-DPPH activity (SC50 23.8 mu l/ml).

AB - Plants from the genus Ferula L. (Apiaceae) were used for various purposes in traditional medicine of different nations throughout the history. Ferula heuffelii GRISEB. ex HEUFFEL is a perennial species endemic for Balkan peninsula. Ten compounds which belong to classes of prenyl-furocoumarin-, prenyl-dihydrofurochromone-, prenyl-benzoyl-and prenyl-benzoylfuranone-type sesquiterpenoids, as well as sesquiterpene coumarins and phenylpropanoids, were, for the first time, isolated from the CHCl3 extract of the underground parts of this plant and identified. Furthermore, extract and three isolated compounds, i.e., latifolone (1), dshamirone (4), and (2S*, 3R*)-2-[(3E)-4,8-dimethylnona-3,7-dien-1-yl]-2,3-dihydro7- hydroxy-2,3-dimethylfuro[3,2-c] coumarin (6) were, for the first time, evaluated for their in vitro antispasmodic activities in three experimental models: spontaneous contraction, and ACh- and KCl-induced contraction of an isolated rat ileum. The extract (0.1-1.3 mg/ml) and compound 6 (1 - 10 mu g/ml) exhibited dose-dependent effect in all three models. Compound 1 (1-6 mu g/ml) affected spontaneous contractions and those induced by KCl, while compound 4 (8 mu g/ml) displayed only moderate activity with ACh-induced contractions. It can be concluded that tested compounds contribute to exhibited antispasmodic activity of crude extract. Additionally, extract (0.1-1.3 mg/ml) was tested for in vitro relaxant activity on an isolated rat trachea, and relaxed the KCl-induced contractions in a dose-dependent manner.

AB - The essential oil from underground parts of Ferula heuffelii from N.E. Serbia, was analysed using GC and GC-MS. The main compounds of the essential oil were elemicin (35.4%) and myristicin (20.6%). The essential oil exhibited the best antimicrobial activity against two strains of Candida albicans (MIC = 7.0 and 13.7 mu g/ml), as well as against Micrococcus luteus (MIC = 13.7 mu g/ml), Staphylococcus epidermidis (MIC = 17.6 mu g/ml), Bacillus subtilis (MIC = 21.1 mu g/ml) and Micrococcus flavus (MIC = 28.2 mu g/ml). In the DPPH radical scavenging assay, essential oil showed substantial activity with SC(50) = 22.43 mu l/ml. The essential oil was also tested for antispasmodic activity. It inhibited spontaneous contraction of isolated rat ileum dose-dependently, and at the concentration of 86.64 mu g/ml exhibited 50% of the maximum effect of atropine. After incubation with 75.00 mu g/ml of essential oil, acetylcholine did not induce contractions of ileum, and at 250.00 mu g/ml, the essential oil almost completely abolished the spasmodic effect of potassium chloride (80 mM).
