AB  - Copper(ii) complexes with an α-diimine show a wide variety of biological activities, such as antibacterial, antifungal, antioxidant and anticancer. In this work, we synthesized and structurally characterized two novel Cu(ii) complexes with methyl 3-formyl-4-hydroxybenzoate (HL) and α-diimines: 2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen). Crystal structure analysis shows that the formulas of the compounds are [Cu(bipy)(L)(BF4)] (1) and [Cu(phen)(L)(H2O)](BF4)·H2O (2), with BF4− as a ligand in complex 1, which is rarely coordinated to metals. Both complexes have a square pyramidal geometry, while DFT calculations showed that the most stable structures of complexes 1 and 2 in a water/DMSO mixture are square-planar derivatives [Cu(bipy)(L)]+ and [Cu(phen)(L)]+. The antibacterial activity of compounds was evaluated in vitro on four Gram-negative and four Gram-positive bacterial strains. Complex 2 showed greater antibacterial activity towards all bacterial strains comparable to the control compound Amikacin. Complex 2 exerted a strong cytotoxic effect against the tested cancer cell lines (IC50 values ranging from 0.32 to 0.44 μM). Both complexes caused apoptotic cell death in HeLa cells and a noticeable in vitro antiangiogenic effect. In the concentration range of 5 to 100 μM, the complexes showed the absence of a genotoxic effect and displayed a protective effect against oxidative DNA damage induced by H2O2 in human peripheral blood cells. The interaction between the compounds and calf-thymus DNA was evaluated by diverse techniques suggesting a tight binding, which was also confirmed by molecular docking. In addition, it was found that the complexes bind tightly and reversibly to bovine and human serum albumin.

Pharmacokinetic characterization, benefits and barriers of subcutaneous administration of monoclonal antibodies in oncology

AB  - Objective: Therapeutic monoclonal antibodies in oncology are slowly becoming the dominant treatment option for many different cancer types. The main route of administration, infusion, requires extensive product preparations, patient hospitalization and close monitoring. Patient comfort improvement, staff workload reduction and cost savings dictated the development of subcutaneous formulations. The aim of this review is to present pharmacokinetic characteristics of subcutaneous products, discuss the differences between intravenous and subcutaneous routes and to point out the advantages as well as challenges of administration route shift from the formulation development and pharmacometric angle. Data sources: Food and Drug administration's Purple book database and electronic medicines compendium were used to identify monoclonal antibodies in oncology approved as subcutaneous forms. Using keywords subcutaneous, monoclonal antibodies, pharmacokinetics, model, as well as specific drugs previously identified, both PubMed and ScienceDirect databases were researched. Data summary: There are currently six approved subcutaneous onco-monoclonal antibodies on the market. For each of them, exposure to the drug was similar in relation to infusion, treatment effectiveness was the same, administration was well tolerated by the patients and costs of the medical service were reduced. Conclusion: Development of subcutaneous forms for existing and emerging new monoclonal antibodies for cancer treatment as well as shifting from administration via infusion should be encouraged due to patient preference, lower costs and overall lack of substantial differences in efficacy and safety between the two routes.

St. George’s mushroom, Calocybe gambosa (Fr.) Donk: A promising source of nutrients and biologically active compounds

AB  - The composition of biologically active compounds of Calocybe gambosa (Lyophyllaceae) was analysed, and
the antioxidant and cytotoxic activities were tested in vitro. C. gambosa was low in energy, fat, and car-
bohydrates, but rich in proteins and fibres. The total polyphenol content in the extracts was low (23.08 ±
0.67 in aqueous and 24.99 ± 4.25 mg GA mg 1 of extract in methanolic extract). The methanolic extract
showed anti-DPPH radical activity with an IC 50 of 626.10 ± 25.20 mg mL 1 . The sample of C. gambosa is
rich in nucleotides and amino acids responsible for its pleasant taste. The nucleoside and 50-mono-
phosphates contents were 0.97 mg g 1 and 2.32 mg g 1 of dry mushroom, respectively. The contents of
essential and non-essential amino acids were 18.41 ± 0.06 and 41.75 ± 0.30 mg g 1 d.w., respectively. The
percentages of saturated and unsaturated fatty acids were 42.6% and 57.4%, respectively. The most
abundant water-soluble and fat-soluble vitamins were B1 and E, respectively. Cytotoxic effect of the extracts was examined against different cancer cell lines, and the best cytotoxicity was showed by the
peptide extract against colon cancer cell line LS174.

Black Trumpet, Craterellus cornucopioides (L.) Pers.: Culinary Mushroom with Angiotensin Converting Enzyme Inhibitory and Cytotoxic Activity

Pers.) were tested in vitro. C. cornucopioides was low in energy, fat and carbohydrate contents, but rich in dietary fibre, especially β-glucan as well as niacin and α-tocopherol. The content of essential and non-essential free amino acids was 1.49 and 5.48 mg/g dry weight (dw). The nucleosides and 5'-nucleotides were determined at 1.84 and 3.99 mg/g dw, respectively. The share of unsaturated fatty acids (UFAs) was 75.92% with oleic acid as the major UFA. Cyclohexane and dichloromethane extracts expressed significant cytotoxic activity against selected cell lines, human epithelial cervical cancer cells (HeLa), adenocarcinomic human alveolar basal epithelial cells (A549), colorectal cancer cells (LS174) and normal MRC-5 human embryonic lung fibroblast cells (IC50 of 78.3-155.6 μg/mL). ACE inhibitory activity of the aqueous extract was strong with an IC50 of 0.74 μg/mL. It can be concluded that black trumpet is a good source of nutrients, such as vitamins, dietary fibres, amino acids, nucleotides and fatty acids, which contribute to the overall nutritional value of this fungus with potential for ACE inhibitory activity and use in anti-hypertensive diet.

It can be concluded that black trumpet is a good source of nutrients, such as vitamins, dietary fibres, amino acids, nucleotides and fatty acids, which contribute to the overall nutritional value of this fungus with potential for ACE inhibitory activity and use in anti-hypertensive diet.",

AB  - Antimicrobial and cytotoxic activities were tested for dried MeOH extracts of Hieracium calophyllum (CAL), H. coloriscapum (COL), H. pseudoschenkii (PSE), H. valdepilosum (VAL) and H. glabratum (GLA) herbs (flowering aerial parts), their 2 sesquiterpene lactones (SLs) 8-epiixerisamine A and crepiside E, and dried CH2Cl2 extract of H. scheppigianum (SCH) herb. In microdilution test, extracts showed activity on all tested microorganisms (8 bacteria, 10 fungi). The best effect was exhibited by SCH and CAL on Salmonella Typhimurium (MIC=1.7–2.5 mg/mL MBC=3.4–5.0 mg/mL), and SCH and VAL on Candida albicans (MIC=2.5 mg/mL MFC=5.0 mg/mL). SLs showed notable effect on all tested fungi Aspergillus ochraceus, Penicillium funiculosum, C. albicans and C. krusei (MIC=0.15–0.4 mg/mL MFC=0.3–0.8 mg/mL). In MTT test, extracts inhibited growth of all tested cancer cells (HeLa, LS174 and A549), with the best effect on HeLa (IC50=148.1 μg/mL for SCH, and 152.3–303.2 μg/mL for MeOH extracts); both SLs were active against HeLa cells (IC50=46.2 μg/mL for crepiside E and 103.8 μg/mL for 8-epiixerisamine A). Extracts and SLs showed good safety profile on normal MRC-5 cells.

AB  - Selected tetrahydropyrimidines (THPMs) were investigated by means of cytotoxic activities on selected cancer (HeLa, A549, and LS174) and non-cancerous cell lines (MRC-5). Among evaluated compounds, two of them ( B7 and B8 ) showed good cytotoxic activity on the tested cell lines and were selected for fur- ther evaluation that included mechanism of action, DNA and BSA interactions and molecular docking study. Calculated parameters from fluorescence quenching studies indicated that B7 and B8 bind on mi- nor groove of DNA and have great ability to bind on carrier protein. Three-dimensional quantitative struc- ture anti-HeLa activity study was performed with data set of THPMs. Molecular Interaction Fields were used to derive Grid independent descriptors (GRIND), as independent variables in Pentacle software. The quality and predictive capacity of the model was proved by internal statistical parameters: R 2 = 0.992, Q 2 = 0.51, as well as external parameters such as R 2 pred = 0.804 and r m 2 , r / 2 m and r 2 m , that were higher than 0.5. The structural determinants significant for anti-HeLa activity of compounds were identified by using developed 3D-QSAR model. Interpretation of the most impactful GRIND variables on the anti-HeLa activity generated several hypotheses for design of novel and more potent anti-HeLa tetrahydropyrim- idines. Additional molecular targets for the most active synthesized derivatives ( B7 and B8 ) are predicted by use of online web-based tool-SwissTargetPrediction.

AB  - In this article, cytotoxicity, the mechanisms of cytotoxic activity, genotoxicity, and interaction with DNA and proteins, of two Cu(II) complexes with a salicylaldehyde derivative (4-(diethylamino)salicylaldehyde) and α-diimine (2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen)) are reported. Both Cu(II) complexes performed cytotoxic effects against all tested malignant cell lines. Complexes exerted highest cytotoxicity against HeLa and A375 malignant cell lines. The cytotoxic activity of Cu(II) complex with phen as a α-diimine co-ligand was significantly higher in comparison with cytotoxic activity of Cu(II) complex with bipy. Pretreatment with specific inhibitors of caspase-3, caspase-8 or caspase-9, in order to clear up the mode of cell death triggered by two Cu(II) complexes in HeLa cells, indicated the ability of these complexes to induce apoptosis through activation of target caspases. Cu(II)-phen complex exhibited significant antioxidant activity compared with Cu(II)-bipy complex, and showed a better effect on reducing intracellular ROS levels in HeLa cells. Tested complexes did not display genotoxic potential in human peripheral blood leucocytes, but exhibited an antigenotoxic effect in post-treatment, after H2O2 exposure. The study of the in vitro biological properties regarding their affinity towards CT (calf-thymus) DNA and serum albumins showed that the compounds can intercalate to CT DNA, and bind reversibly and tightly to the albumins. Molecular docking studies of the ability of compounds to bind to biomacromolecules are consistent with in vitro studies.

AB  - Mahonia aquifolium and its secondary metabolites have been shown to have anticancer potential. We performed MTT, scratch, and colony formation assays; analyzed cell cycle phase distribution and doxorubicin uptake and retention with flow cytometry; and detected alterations in the expression of genes involved in the formation of cell-cell interactions and migration using quantitative real-time PCR following treatment of lung adenocarcinoma cells with doxorubicin, M. aquifolium extracts, or their combination. MTT assay results suggested strong synergistic effects of the combined treatments, and their application led to an increase in cell numbers in the subG1 phase of the cell cycle. Both extracts were shown to prolong doxorubicin retention time in cancer cells, while the application of doxorubicin/extract combination led to a decrease in MMP9 expression. Furthermore, cells treated with doxorubicin/extract combinations were shown to have lower migratory and colony formation potentials than untreated cells or cells treated with doxorubicin alone. The obtained results suggest that nontoxic M. aquifolium extracts can enhance the activity of doxorubicin, thus potentially allowing the application of lower doxorubicin doses in vivo, which may decrease its toxic effects in normal tissues.

AB  - Rumex balcanicus Rech.f. (Polygonaceae) is an endemic species that inhabits
central parts of Serbia and Kosovo. The genus Rumex is characterizied by the
accumulation of anthraquinones, naphthalene-1,8-diols, flavonoids and stilbenoids.
Aerial parts of R. balcanicus, collected on Vlasina Lake (Serbia) in 2014, were used
to determine the chemical composition and cytotoxic activity. Cyclohexane,
dichloromethane, methanol and aqueous extracts were prepared and tested using
MTT-test, against human epithelial cervical cancer cells (HeLa), human colon
carcinoma cell line (LS174), human non-small cell lung cancer cells (A549) and
healthy MRC-5 human embryonic lung fibroblast cell line. Methanol and aqueous
extracts were analyzed using LC-MS. In tested extracts, caffeic acid derivates and
flavonol derivatives were identified. The main components in both extracts were rutin,
miquelianin, quercitrin and quercetin 3-O-acetyl-rhamnoside. Caffeic acid and its
glycosides were also present in analyzed extracts. Aqueous extracts had no
cytotoxicity against tested cell lines, while cyclohexane, dichlormethane and
methanol extracts were active against all tested cell lines with IC50 ranged from
116.48±3.26 μg/mL to 178.21±1.30 μg/mL. Due to the presence of flavonoids and
exhibited cytotoxic effect, R. balcanicus herba could be interesting as a plant material
for further pharmacological analysis and isolation of the anti-tumor compounds.

AB  - Three seaweeds (Halimeda tuna, Codium bursa and Cystoseira barbata) and one seagrass (Cymodocea nodosa) were collected from the Coast of Montenegro, Gulf of Boka Kotorska and their chemical analysis was performed. In seagrass C. nodosa, three phenolic compounds were identified (diosmetin 7-sulfate, caftaric and coutaric acid). The content of β-glucan, fatty acids, sterols and micro- and macro-elements were investigated among all samples. The highest content of β-glucan was detected in C. nodosa seagrass (13.04±0.42 g/100 g). The highest polyunsaturated fatty acids (PUFAs) level was reported in C. barbata, the brown alga (7.157 mg/g), which also had the significant sterol content (fucosterol, 21.76±0.1 μg/g). Green algae, C. bursa and H. tuna, showed the highest level of sterols (β-sitosterol, 95.21±0.16 μg/g and 73.90±0.08 μg/g, respectively). H. tuna had the highest content of calcium (Ca) in amount of 55125 μg/g. In C. bursa, C. barbata and C. nodosa, the Na/K ratio was low (0.43, 0.46 and 0.69, respectively).

AB  - Cytotoxic and antimicrobial activities were investigated for MeOH extracts of flowering aerial parts of Hieracium calophyllum R. Uechtr. (CAL), H. coloriscapum Rohlena & Zahn (COL), H. pseudoschenkii (Rohlena & Zahn) Niketić (PSE), H. valdepilosum Vill. s.l. (VAL) and H. glabratum Willd. (GLA), their two isolated sesquiterpene lactones 8-epiixerisamine A and crepiside E, as well as for CH2Cl2 extract of flowering aerial parts of H. scheppigianum Freyn (SCH). Crepiside E (IC50 = 46.00 μg/mL), COL (IC50 = 152.28 μg/mL) and SCH (IC50 = 148.12 μg/mL) showed 
the highest cytotoxic activity against HeLa cells in MTT test. The cytotoxic activity against LS174, A549, as well as against normal MRC-5 cells was significantly weaker. In microdilution test, crepiside E and 8-epiixerisamine A exhibited noteworthy antifungal effect against Aspergillus ochraceus, Penicillium funiculosum, Candida albicans and C. krusei (MIC = 0.15 - 0.4 mg/mL, MFC = 0.3 - 0.8 mg/mL). Among the extracts, the best antibacterial activity was shown by SCH and CAL against Pseudomonas aeruginosa (MIC = 1.68 and 2.52 mg/mL, MBC = 3.36 and 5.04 mg/mL), and the highest antifungal activity by SCH and VAL against C. albicans (MIC = 2.48 and 2.52 mg/mL, MFC = 4.96 and 5.04 mg/mL).

abstract = "Cytotoxic and antimicrobial activities were investigated for MeOH extracts of flowering aerial parts of Hieracium calophyllum R. Uechtr. (CAL), H. coloriscapum Rohlena & Zahn (COL), H. pseudoschenkii (Rohlena & Zahn) Niketić (PSE), H. valdepilosum Vill. s.l. (VAL) and H. glabratum Willd. (GLA), their two isolated sesquiterpene lactones 8-epiixerisamine A and crepiside E, as well as for CH2Cl2 extract of flowering aerial parts of H. scheppigianum Freyn (SCH). Crepiside E (IC50 = 46.00 μg/mL), COL (IC50 = 152.28 μg/mL) and SCH (IC50 = 148.12 μg/mL) showed 
the highest cytotoxic activity against HeLa cells in MTT test. The cytotoxic activity against LS174, A549, as well as against normal MRC-5 cells was significantly weaker. In microdilution test, crepiside E and 8-epiixerisamine A exhibited noteworthy antifungal effect against Aspergillus ochraceus, Penicillium funiculosum, Candida albicans and C. krusei (MIC = 0.15 - 0.4 mg/mL, MFC = 0.3 - 0.8 mg/mL). Among the extracts, the best antibacterial activity was shown by SCH and CAL against Pseudomonas aeruginosa (MIC = 1.68 and 2.52 mg/mL, MBC = 3.36 and 5.04 mg/mL), and the highest antifungal activity by SCH and VAL against C. albicans (MIC = 2.48 and 2.52 mg/mL, MFC = 4.96 and 5.04 mg/mL).",

Rezultati i diskusija: Gasnom hromatografijom sa FID i MS detekcijom analiziran je sastav etarskih ulja (ukupno 59 uzoraka) izolovanih destilacijom vodenom parom iz podzemnih organa, listova, cvasti i plodova. U uljima podzemnih organa dominirali su monoterpeni i/ili fenilpropani (taksoni grupe H. sphondylium), ili (Z)-falkarinol (H. orphanidis i H. austriacum); u uljima plodova alifatični estri oktilacetat (sect. Heracleum) ili oktilheksanoat (H. austriacum), a u uljima listova i cvasti alifatični estri (H. orphanidis) ili seskviterpeni i/ili fenilpropani. Antimikrobna aktivnost 30 odabranih uzoraka etarskih ulja ispitivana je mikrodilucionom metodom na po osam sojeva bakterija i mikromiceta (u slučaju 17 uzoraka ulja MIK vrednosti su prema barem jednom mikroorganizmu bile niže ili bliske 100 µg/mL); selektivna citotoksičnost 26 uzoraka ulja MTT testom na tumorske HeLa, LS174 i A549 ćelije (u slučaju 13 uzoraka ulja IC50 vrednosti su prema barem jednoj ćelijskoj liniji bile niže ili bliske 30 µg/mL; ulja uglavnom nisu inhibirala rast normalnih MRC-5 ćelija). Ukupno 8 od ispitivanih 14 uzoraka ulja pokazalo je slab anti-DPPH potencijal. Sastav aktivnih zona u TLC-DPPH testu ispitan je gasnom hromatografijom (dominirali su oksidovani terpeni i fenilpropani). S obzirom da je u pojedinim uljima utvrđeno prisustvo furanokumarina koji mogu delovati fototoksično, definisan je bezbednosni profil ulja prema preporuci Evropske agencije za lekove (utvrđeno je da dnevni unos 1,94-15,68 mL ulja ne doprinosi značajno ukupnom riziku, koji postoji kao posledica unosa izvesnih količina furanokumarina hranom).
U dihlormetanskim ekstraktima podzemnih organa i plodova svih taksona identifikovano je ukupno 12 furanokumarina (LC-MS metodom pomoću standardnih supstanci ili na osnovu UV, MS, 1H i ROESY NMR spektara), od kojih su dominantni bili pimpinelin, bergapten, biakangelikol, heraklenin i/ili imperatorin. U uljastim supernatantima dihlormetanskih ekstrakata plodova gasnom hromatografijom (GC-FID i GC-MS) analizirane su masne kiseline (dominantna je bila petroselinska), steroli (najzastupljeniji je bio β-sitosterol) i triterpeni. 
Hemosistematski značaj komponenti etarskih ulja (u slučaju podzemnih organa i plodova i komponenti njihovih headspace frakcija) i furanokumarina utvrđen je multivarijantnom statističkom analizom (PCA, nMDS i UPGMA).
Zaključak: Sprovedena hemijska karakterizacija značajno je doprinela boljem poznavanju kompozicije sekundarnih i primarnih metabolita odabranih predstavnika sekcije Heracleum, dok su ispitivanja H. austriacum subsp. siifolium iz sekcije Wendia sprovedena po prvi put. Utvrđeno je da su ispitivani taksoni potencijalno novi biljni izvori jedinjenja značajnih za farmaceutsku, kozmetičku, prehrambenu ili hemijsku industriju. Rezultati ispitivanja farmakoloških aktivnosti etarskih ulja ukazali su na opravdanost daljih istraživanja ovih izolata kao potencijalno novih lekovitih biljnih sirovina, uz uslov poštovanja ustanovljenih maksimalnih dozvoljenih dnevnih unosa za ona etarska ulja u kojima su detektovani furanokumarini.

Phenolic constituents, antioxidant, α-amylase and α-glucosidase inhibitory activities of Pyrus × velenovskyi bark

AB  - Pyrus × velenovskyi Dostálek (Rosaceae) [P. pyraster (L.) Burgsd. × P. spinosa Forssk.] is a deciduous tree up to 5 m height. Leaf-blades 2.2–4.8 × 1.3–2.7 cm, ± glabrate, ellip tic, gradually narrowed to a rather long petiole. Th is natural hybrid plant was described from Bulgaria, and has been also found in Serbia. The objective of this work was to investigate the phenolic profile, as well as in vitro antioxidant, α-amylase and α-glucosidase inhibitory activities of dried methanol ex tract from the bark of this tree. The plant material was collected in eastern Serbia (Jelašnička Klisura gorge). In the dried bark, the contents of different classes of phenolics were spectrophotometrically determined: total polyphenols (10.36%), tannins (8.78%), procyanidins (4.21%) and phenolic glycosides (3.54%). After pre-extraction with dichloromethane, powdered bark was extracted with methanol by bimaceration at room temperature. In obtained dried methanol extract, using aforementioned spectrophotometric tests, the contents of total polyphenols (33.42%) and tannins (21.55%) were determined, and by HPLC, arbutin, chlorogenic acid, catechin and procyanidin B2 were identifi ed. Its antioxidant activity, i.e. ferric reducing capacity (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH radical scavenging ability, as well as the inhibition of the enzymes α-amylase and α-glucosidase were assessed using corresponding colorimetric assays. Tested dried methanol extract exhibited signifi cant DPPH and OH radical scavenging abilities (SC50 = 6.85 and 12.21 μg/mL, respectively), and ferric reducing capacity (10.74 mmol Fe (II)/g of dried extract), as well as the inhibition of α-amylase (IC50 = 11.4 μg/mL) and α-glucosidase (IC50 = 5.48 μg/mL).

AB  - A series of 23 novel anthraquinone-chalcone hybrids containing amide function was synthesized and structurally characterized. Sixteen compounds exerted strong cytotoxic activities against K562, Jurkat and HL-60 leukemia cell lines and significantly lower cytotoxic effects against normal MRC-5 cells, indicating very high selectivity in their anticancer action. The compounds 6g, 6u and 6v activate apoptosis in K562 cells through the extrinsic and intrinsic apoptotic pathway. The compound 6e triggered apoptosis in K562 cells only through the extrinsic apoptotic pathway. Treatment of K562 cells with each of these four compounds caused decrease in the expression levels of MMP2, MMP9, and VEGF, suggesting their anti-invasive, antimetastatic and antiangiogenic properties. The compounds 6g and 6v downregulated expression levels of miR-155 in K562 cells, while compounds 6e and 6u upregulated miR-155 levels in treated cells, in comparison with control cells. The structurebased 3-D QSAR models for 6f, 6e, 6i and 61 describe pro-apoptotic activity against caspase-3.

AB  - The aim of the present study was to reveal the potential biological activities of the dichlorometane: methanol (1:1) dry extract (DME) and the nutritional value of selected seaweeds: Cymodocea nodosa, Halimeda tuna, Cystoseira barbata and Codium bursa, collected from the Adriatic coast, Montenegro. We assessed the chemical composition and several biological activities such as: anti-a-glucosidase, antitumour, antimicrobial and antioxidative activity. H. tuna had the best cytotoxic activity against human colon carcinoma cell line, LS174 (IC50 = 17.92 +/- 1.54 mu g/mL). C. nodosa demonstrated strong cytotoxicity against human adenocarcinoma cell line, HeLa (IC50 = 13.28 +/- 0.39 mu g/mL) and human chronic myelogenous leukaemia cell line, K562 (IC50 = 19.64 +/- 1.55 mu g/mL). C. barbata had the best anti a-glucosidase (IC50 = 9.98 +/- 3.34 mu g/mL) and antimicrobial activity (minimal inhibitory concentration of 100 mu g/mL) for Staphylococcus aureus and Bacillus subtilis. C. bursa showed the highest nutritional value (490.4 kcal).

AB  - Antibacterial and cytotoxic activities of cyclohexane, dichloromethane, methanol, and aqueous extracts of Cantharellus cibarius were tested. Broth microdilution assay was performed against 10 bacterial strains (Staphylococcus aureus, S. epidermidis, Micrococcus luteus, Bacillus subtilis, Enterococcus feacalis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella abony), with emphasis on Helicobacter pylori. Methanol extract was the most active against H. pylori strains with minimal inhibitory concentration values between 4 and 32 mu g/mL. All extracts were active against antibiotic resistant H. pylori. Methanol and aqueous extracts had no cytotoxicity against tested cell lines, whereas cyclohexane and dichloromethane extracts were active against HeLa and N87 cells, but also against healthy MRC-5 cells (IC50 39.26 +/- 1.24-134.79 +/- 0.01 mu g/mL). The tested aqueous extracts have shown 68% of angiotensin-converting enzyme inhibitory activity in doses of 1.25 mg/mL. Chemical analysis has shown the presence of linoleic, cis-vaccenic, and oleic acids, sterols, beta-glucans, and polyphenolic compounds.

AB  - Many Heracleum L. taxa (Apiaceae) are used as food and spices, and in traditional medicine. In this work, the chemical composition of Heracleum pyrenaicum subsp. orsinii (Guss.) F. Pedrotti and Pignatti root, leaf and fruit essential oils, their antimicrobial activity and cytotoxic effect on malignant and normal cells were investigated for the first time. The composition of the oils was analyzed by GC and GC-MS. Monoterpenes prevailed in the root oil, with beta-pinene (38.6%) being dominant, while in the leaf oil, sesquiterpenes, mostly (E)-nerolidol (20.5%) and (E)-caryophyllene (17.0%), were the most abundant constituents. The fruit oil contained the majority of aliphatic esters, mainly octyl acetate (36.8%) and octyl hexanoate (22.1%). The antimicrobial activity was determined by microdilution method against eight bacteria and eight fungi (standard strains, clinical or food isolates). The best antibacterial activity, better than the activity of ampicillin, was shown by the root oil against Salmonella typhimurium, Escherichia coli and Pseudomonas aeruginosa. The strongest antifungal activity, stronger than the activity of ketoconazole, was exhibited by the leaf and root oils against Trichoderma viride, and by the root oil against Aspergillus ochraceus. The cytotoxic effect of the oils, determined by MTT test, was prominent against malignant HeLa, LS174 and A549 cells (IC50 = 6.49-14.56 µg/mL). On the other hand, the oils did not show toxicity against normal MRC-5 cells at tested concentrations (IC50 > 200.00 µg/mL). It can be concluded that investigated H. pyrenaicum subsp. orsinii oils represent potential new raw materials for food and pharmaceutical industry.

AB  - Although some widespread, native cow parsnips (Heracleum L. spp., Apiaceae) had broad medicinal and culinary applications throughout history, the knowledge about their volatile constituents is insufficient. This work investigates the composition and bioactivities of H. sphondylium L. (HSPH), H. sibiricum L. (HSIB) and H. montanum Schleich. ex Gaudin (HMON) essential oils. The composition was tested by GC and GC-MS. (Z)-β-Ocimene was the most abundant in HSPH (28.9%) and HMON (20.4%) root oils, while in HSIB root oil, β-pinene (26.2%), methyl eugenol (22.3%) and elemicin (25.6%) prevailed. Leaf and flower oils were dominated by various sesquiterpenes (germacrene D, β-sesquiphellandrene, (E)-β-farnesene and/or (E)-caryophyllene) and/or phenylpropanoids (apiole, methyl eugenol, elemicin and/or (Z)-isoelemicin). Octyl acetate (57.5-67.1%) was the main constituent of all fruit oils. The antimicrobial activity was screened by a microdilution method against eight bacteria and eight fungi. The strongest antimicrobial effect, in several cases better than the activity of antibiotics, was shown by HSPH (MICs = 0.12-3.30 mg mL-1) and HMON (MICs = 0.10-1.30 mg mL-1) flower oils against bacteria, and HSIB fruit oil against fungi (MICs = 0.15-0.40 mg mL-1). AB  - In this work, the chemical composition, antimicrobial and cytotoxic activity of Heracleum verticillatum Pančic and H. ternatum Velen. root, leaf, and fruit essential oils were investigated. The composition was analyzed by GC and GC/MS. Heracleum verticillatum and H. ternatum root oils were dominated by monoterpenes, mostly β-pinene (23.5% and 47.3%, respectively). Heracleum verticillatum leaf oil was characterized by monoterpenes, mainly limonene (20.3%), and sesquiterpenes, mostly (E)-caryophyllene (19.1%), while H. ternatum leaf oil by the high percentage of phenylpropanoids, with (Z)-isoelemicin (35.1%) being dominant constituent. Both fruit oils contained the majority of aliphatic esters, mostly octyl acetate (42.3% in H. verticillatum oil and 49.0% in H. ternatum oil). The antimicrobial activity of the oils was determined by microdilution method against eight bacterial and eight fungal strains. The strongest effect was exhibited by H. verticillatum root oil, particularly against Staphylococcus aureus, Salmonella typhimurium (MICs = 0.14 mg/ml, MBCs = 0.28 mg/ml), and Trichoderma viride (MIC = 0.05 mg/ml, MFC = 0.11 mg/ml). Cytotoxic effect was determined by MTT test against malignant HeLa, LS174, and A549 cells (IC50 = 5.9 - 146.0 μg/ml), and against normal MRC-5 cells (IC50> 120.1 μg/ml). The best effect was exhibited by H. verticillatum root oil on A549 cells (IC50= 5.9 μg/ml), and H. ternatum root oil against LS174 cells (IC50= 6.7 μg/ml).

In vitro assessment of antiproliferative action selectivity of dietary isothiocyanates for tumor versus normal human cells

AB  - Background/Aim. Numerous epidemiological studies have shown beneficial effects of cruciferous vegetables consumption in cancer chemoprevention. Biologically active compounds of different Brassicaceae species with antitumor potential are isothiocyanates, present in the form of their precursors - glucosinolates. The aim of this study was to determine the selectivity of antiproliferative action of dietary isothiocyanates for malignant versus normal cells. Methods. Antiproliferative activity of three isothiocyanates abundant in human diet: sulforaphane, benzyl isothiocyanate (BITC) and phenylethyl isothiocyanate, on human cervix carcinoma cell line - HeLa, melanoma cell line - Fem-x, and colon cancer cell line - LS 174, and on peripheral blood mononuclear cells (PBMC), with or without mitogen, were determined by MTT colorimetric assay 72 h after their continuous action. Results. All investigated isothiocyanates inhibited the proliferation of HeLa, Fem-x and LS 174 cells. On all cell lines treated, BITC was the most potent inhibitor of cell proliferation with half-maximum inhibitory concentration (IC50) values of 5.04 mmoL m-3 on HeLa cells, 2.76 mmol m-3 on Fem-x, and 14.30 mmol m-3 on LS 174 cells. Antiproliferative effects on human PBMC were with higher IC50 than on malignant cells. Indexes of selectivity, calculated as a ratio between IC50 values obtained on PBMC and malignant cells, were between 1.12 and 16.57, with the highest values obtained for the action of BITC on melanoma Fem-x cells. Conclusion. Based on its antiproliferative effects on malignant cells, as well as the selectivity of the action to malignant vs normal cells, benzyl isothiocyanate can be considered as a promising candidate in cancer chemoprevention. In general, the safety of investigated compounds, in addition to their antitumor potential, should be considered as an important criterion in cancer chemoprevention. Screening of selectivity is a plausible approach to the evaluation of safety of both natural isothiocyanates and synthesised analogues of these bioactive compounds.
AB  - Uvod/Cilj. Brojne epidemiološke studije pokazale su povoljne efekte konzumiranja povrća iz familije kupusnjača (Brassicaceae) u hemioprevenciji karcinoma. Osnovni biološki aktivni sastojci ovog povrća su izotiocijanati, prisutni u obliku prekursora - glukozinolata. Cilj ovog rada bio je određivanje selektivnosti antiproliferativnog delovanja dijetetskih izotiocijanata na maligne ćelije u odnosu na normalne ćelije. Metode. Antiproliferativna aktivnost tri izotiocijanata zastupljena u ljudskoj ishrani: sulforafana (SFN), benzil-izotiocijanata (BITC) i feniletil-izotiocijanata (FEITC) na humane maligne ćelijske linije, HeLa, ćelijsku liniju karcinoma grlića materice, Fem-x, ćelijsku liniju melanoma, i LS 174, ćelijsku liniju karcinoma kolona, kao i na mononuklearne ćelije periferne krvi (MNĆPK), sa ili bez delovanja mitogena, određivana je MTT kalorimetrijskim testom, 72 h nakon kontinuiranog delovanja agenasa. Antimicrobial and Cytotoxic Activities of the Sulphur Shelf Medicinal Mushroom, Laetiporus sulphureus (Agaricomycetes), from Serbia

AB  - Cyclohexane, dichloromethane, and methanol extracts of the medicinal mushroom Laetiporus sulphureus were tested for their antimicrobial, cytotoxic, and radical scavenging activities. Total polyphenol content was also determined. Regarding the amount of the phenolic compounds in the methanol extract, there were 14.1 +/- 0.6 mg gallic acid/g dry extract and a concentration of 0.66 +/- 0.15 mg/mL resulted in the removal of 50% of the DPPH radicals (half-maximal inhibitory concentration [IC50]) . The average minimum inhibitory concentrations (MICs) against a select panel of microorganisms were between 125 and 250 mu g/mL, and the methanolic extract significantly inhibited the growth of Staphylococcus aureus (MIC, 31.25 mu g/mL). Also, cyclohexane and dichloromethane extracts inhibited the growth of Helicobacter pylori (MIC, 62.5 mu g/mL). Cyclohexane and dichloromethane extracts showed higher antiproliferative action compared with the methanol extract. The lowest IC50 was induced on HeLa cells by dichloromethane extract (IC50,67.20 +/- 5.49 mu g/mL), followed by cyclohexane extract (IC50 75.91 +/- 1.57 mu g/mL). The tested cyclohexane and dichloromethane extracts possessed moderate antibacterial and significant cytotoxic activity against both malignant (HeLa and N87) and healthy (MRC5) cell lines. Methanol extracts exhibited promising anti-Helicobacter activity without cytotoxicity against N87 and MRC5 cell lines.

Chemical composition and biological activity of the plum seed extract

AB  - The aim of this paper was to estimate the biological activity of the plum seed extract and to define the chemical composition by using the ESI-MS method. During the investigation of the antioxidant activity, the extract showed a better ability to inhibit DPPH radicals compared with amygdalin standard. The results of the antimicrobial study indicate that the extract has a greater effect on Gram-negative bacteria compared with amygdalin. Gram-positive bacteria and fungi remained resistant in both cases. Unlike amygdalin, the extract showed a greater antimicrobial activity against the same microbiological strains. Amygdalin was isolated from the extract before the investigation of antiproliferative activity against MDA-MB-361, MDA-MB-453, HeLa and LS174 cell lines using MTT assay. Unlike the MDA-MB-453 cell line, the activity of standard and isolated amygdalin was not observed at the concentrations lower than 400 μg cm-3 on MDA-MB-361 cell line. The isolated amygdalin has not shown the activity in other cell lines. The antiproliferative activity of amygdalin standard was also noticed on HeLa and LS-174 cell lines at the concentrations of 333.27 and 333.17 μg cm-3, respectively.
AB  - Cilj ovog rada bila je procena biološke aktivnosti ekstrakta semena šljive i definisanje hemijskog sastava primenom ESI-MS metode. Tokom ispitivanja antioksidativne aktivnosti, ekstrakt je pokazao bolju sposobnost inhibiranja DPPH radikala u poređenju sa standardom amigdalina. Rezultati antimikrobne studije pokazali su da ekstrakt ima veći uticaj na rast Gram-negativnih bakterija u odnosnu na amigdalin. Gram-pozitivne bakterije i gljivice ostale su rezistentne u oba slučaja. Za razliku od amigdalina, ekstrakt je pokazao veću antimikrobnu aktivnost na iste sojeve mikroorganizama. Amigdalin je izolovan iz ekstrakta pre ispitivanja antiproliferativne aktivnosti na MDA-MB-361, MDA-MB-453, HeLa i LS174 ćelijskim linijama upotrebom MTT testa.
