Phenolics and Flavonoid Content in Selected Seeds from the Serbian Market

AB  - Objectives: Edible seeds are usually consumed as common food ingredients. They are
considered to have a rich nutrient profile, containing different macro and micronutrients, as well
as some biologically active compounds with positive health effects, such as different phenolics.
The aim of this work was to determine total phenolics (TPC) and total flavonoid content (TFC)
in selected commercial seeds samples from the Serbian market. Methods: Samples of nine seeds
were investigated (sesame and black sesame, raw and roasted sunflower, raw and roasted pumpkin,
hemp, chia and linseed). The samples of native seeds and those defatted using dichloromethane
(maceration and Soxhlet extraction) were extracted with 80% methanol. Obtained hydro-methanol
extracts were dried and further analysed using spectrophotometric methods: TPC was determined
using Folin–Ciocalteu (FC) reagent and expressed as gallic acid equivalents (GAE), while TFC was
measured based on the reaction between flavonoids and aluminium chloride and expressed as
catechin equivalents (KE). Results: In general, hydro-methanol extracts of seed samples defatted
using Soxhlet extraction had the highest TPC and TFC contents. TPC values ranged from 9.47 g
GAE/mg (raw pumpkin seed) to over 170 g GAE/mg (raw sunflower seeds). As for TFC, the
highest amount was measured in extracts of defatted raw sunflower seeds (over 150 g KE/mg),
while roasted pumpkin and hemp seeds’ extracts were practically devoid of flavonoids. Conclusion:
Our results confirmed the fact that certain defatted seeds, which are usually considered as waste
products in oil production, could be considered as valuable sources of certain secondary plant
metabolites, implicating further investigations on their composition and potential in the development
of functional foods.

Dietary Intakes of Sugar in the Student Population from Fruits and Beverages

AB  - Objectives: A healthy diet includes the intake of sugar (up to 5%) from natural sources,
because added sugars increase energy but not nutritional value, and are associated with an increased
risk of developing obesity, cardiovascular diseases and hypertension. Sugars present in fresh fruit,
fruit juices and nectars, refreshing soft drinks and alcoholic drinks usually represent half of the
total sugar intake. The aim of this study was to analyze, through surveys, the habits of the student
population regarding the consumption of food from the sources mentioned and to determine the
amount of sugar consumed. Methods: 123 students (male: 36, female: 87), aged 19 to 32, participated
in this research. Data collection was voluntary and anonymous, and the survey was conducted
using an electronic questionnaire during February 2023. The data on the sugar content of the foods
mentioned were taken from the declarations of the products that are available for free sale. Results:
Based on the processed results from the completed questionnaires, it was concluded that from the
sources mentioned, sugar is represented by 3.7% in relation to the total energy intake, and added
sugar by 2.4% if 2000 kcal/day is consumed. In relation to the representation of carbohydrates of
60% in relation to the total energy intake, the sugar content from the tested sources accounted for
6.3%, and added sugar accounted for 4.0%. Students reported that they most often do not consume
hard liquor (88% of respondents), and that they eat a teaspoon of honey/sugar once a day (14% of
respondents). Conclusion: Although the obtained results are very acceptable and encouraging, it
is necessary to act additionally in terms of increasing awareness of the reduced intake of products
with added sugars and their replacement with those that contain biologically active compounds in
addition to natural sugar composition.

Methods for the determination of antioxidant activity of plant extracts in vitro

AB  - Plants are rich sources of secondary metabolites that exhibit diverse biological and
pharmacological effects. Some plant ingredients, primarily phenolics, have significant in vitro
antioxidant activity, which implies their contribution to the maintenance of redox balance in the
body. These potential antioxidant agents are structurally very diverse, having different
mechanisms of antioxidant activity. Since there is a growing necessity to detect, develop and
understand effective antioxidant compounds, interest in the identification and the measurement
of antioxidants in various plant isolates is persistently growing and many methods are being
established. Most of the available in vitro tests are affordable and easy to perform, but due to the
complex composition of plant extracts, different kinetics, mechanisms and specificity of the
chemical reactions underlying these tests, there is no universal parameter for the assessment of
antioxidant activity. In this paper, some of the currently most used in vitro methods for
investigating and evaluating antioxidant activity of plant extracts are presented, emphasizing their
advantages and weaknesses.
AB  - Biljke su bogati izvori sekundarnih metabolita koji ispoljavaju raznovrsne biološke i
farmakološke efekte. Neki biljni sastojci, u prvom redu polifenolna jedinjenja, ispoljavaju
značajnu in vitro antioksidativnu aktivnost, zbog čega se smatra da mogu doprineti održanju
redoks ravnoteže u organizmu. Ovi potencijalni antioksidativni agensi strukturno su veoma
raznovrsni i deluju različitim antioksidantnim mehanizmima. S obzirom na rastuću potrebu za
iznalaženjem, razvojem i razumevanjem efikasnih antioksidanasa, interesovanje za ispitivanje
antioksidanasa u različitim biljnim izolatima kontinuirano raste, pa su shodno tome i razvijeni
mnogi testovi. Većina dostupnih in vitro testova je pristupačna i jednostavna za izvođenje, ali
zbog složenog sastava biljnih ekstrakata, različite kinetike, mehanizama i specifičnosti hemijskih
reakcija na kojima se ovi testovi zasnivaju, još uvek ne postoji univerzalni parametar za procenu
antioksidativne aktivnosti. U ovom radu su prikazane neke od trenutno najkorišćenijih in vitro
metoda za ispitivanje i procenu antioksidativne aktivnosti biljnih ekstrakata, s naglaskom na
njihove prednosti i nedostatke.

AB  - The highest richness of taxa within genus Stachys was recorded in the wider area of the Mediterranean, where the group S. recta stands out in terms of taxonomic complexity. The center of diversity of this group is on the Balkan Peninsula. Its representatives are still insufficiently defined in taxonomic and horological terms, and their traditional approach includes about fifteen taxa, mostly at the infraspecific level. Despite some radical approaches that were presented at the end of the last century [1], the traditional taxonomic treatment is still prevailed in the relevant global lists.
In this work antiradical potential and phenolic constituents of S. recta L. subsp. recta were investigated. Plant material, collected in Central Serbia (vicinity of Prokuplje), was air-dried, powdered, and then extracted by bimaceration with chloroform and methanol. Obtained dry methanol extract exhibited strong, concentration-dependant anti-DPPH activity (SC50=8.23 μg/mL), with total phenolics content at 0.27 mg GAE/mg of dry extract. In TLC-DPPH test observed yellow zones indicated presence of few compounds which strongly neutralized purple DPPH radical. LC-MS analysis revealed presence of 24 phenolic compounds, belonging to three classes of constituents: phenolic acids, phenylethanol glycosides and flavonoids. Chlorogenic acid (2.63%) was the only phenolic acid, while acteoside was the most dominant phenylethanoid (24.03%). Flavonoid fraction was composed almost entirely of 8-hydroxyflavones, derivatives of hypolaetine and isoscutellareine (53.47%). The most prominent flavonoids in investigated extract were 7-O-[6'''-acetylallosyl(1®2)]-glucopyranosides of 4'-O-methylhypolaetine and hypolaetine (15.92 and 13.01%, respectively).

AB  - Background: Although the use of medicinal plants have been known since ancient times, an increased use of herbal products is evident in the last few decades, mainly as a part of self-care treatments. In addition to the positive health effects, the use of herbal products can cause certain side effects as well as interactions with conventional therapy, especially among elderly patients.
Purpose: To examine the awareness, habits and attitudes of elderly patients towards the use of herbal products in the Republic of Serbia.
Method: The quantitative, non-experimental research was conducted using a questionnaire in public pharmacies during February 2020. Patients older than 60 years were included in the study. The study was anonymous, and patients gave informed consent.
Results: The study involved 436 patients, of whom 62.4% were female. Most of the respondents live in the city (72.9%) and have a high school or lower education degree (61.9%). Most patients (32.3%) use herbal products daily, while only 12.1% rarely or never use them. Every fourth patient (24.5%) uses them for treatment, while 69.5% to improve their health. Although the majority of patients purchase herbal products in pharmacies and in health food stores (53.6% and 20.8%, respectively), a significant number (19.3%) collect medicinal plants and prepare herbal preparations themselves. The main sources of information about medicinal plants / herbal products are pharmacists (39.7%) and people from the immediate area (28.4%), followed by the media (18.2%) and physicians (11.1%). Every fourth patient (24.5%) uses herbal products for the treatment of some health issues, while 69.5% of patients use herbal products to improve their health. 48.9% of them believe that herbal products are safe, but with some side effects, while 35.8% believe herbal products are completely free of any side effects. Half of respondents (51.4%) believe that herbal products are as effective as or more effective than synthetic medicines. Less than one-third of patients (29.8%) indicated that they knew the difference between herbal medicinal products and herbal dietary supplements. Still, a third of patients (36.7%) needed additional explanations for the use of herbal products. 76.6% of patients stated that they always receive advice from a pharmacist when purchasing herbal products.
Conclusion: The survey conducted in this work showed that elderly patients use herbal products to a considerable extent, which means that it is necessary to educate elderly patients on this matter. Pharmacists should play a vital role in this process and actively discuss the characteristics and proper use of herbal products with patients, especially having in mind the high possibility of interactions with conventional medicines and occurrence of side effects. Since the quality of some herbal products sometimes could be questionable, especially non-regulated ones, patients should be encouraged to purchase them from pharmacies and consult with pharmacists.

Structural elucidation of flavonoids isolated from aerial parts of Stachys annua (L.) L. (Lamiaceae)

AB  - Stachys annua (L.) L. (annual yellow-woundwort) is an arrable weed and melliferous archaeophyte, with the native range in Europe to Siberia and Iran [1]. The aim of this work is to implement simple chromatographic technique for isolation of flavonoids from crude methanol extract of S. annua aerial flowering parts (herb).  
Plant material collected in NW Serbia (Mt Sokolske planine) was air-dried, powdered, and then extracted by bimaceration with chloroform and methanol. Dry methanol extract of S. annua herb was applied on preparative TLC plates of silica gel. After developing of chromatogram in methylene chloride:methanol:water (40:10:1, V/V/V), and spraying with NP/PEG reagent, in ultraviolet light at 365 nm three clearly separated fluorescence-quenching zones were observed. Zones were eluted with mixture of methylene chloride and methanol (1:1, V/V), and using LC-MS analysis one dominant compound was detected in each of these fractions. According to their UV, MS and 1H-NMR spectral data, as well as using flavonoids previously isolated from S. alpina subsp. dinarica [2], these compounds (1-3) were identified as 8-hydroxyflavone glycosides, namely: isoscutellarein-7-O-[6'''-acetyl-β-D-allopyranosyl-(1→2)]-β-D-glucopyranoside (compound 1), 3'-hydroxy-4'-O-methylisoscutellareine-7-O-[6'''-acetyl-β-D-allopyranosyl-(1→2)]-β-D-glucopyranoside (compound 2) and 4'-O-methylisoscutellareine-7-O-[6'''-acetyl-β-D-allopyranosyl-(1→2)]-β-D-glucopyranoside (compound 3).
It could be concluded that the developed preparative TLC method is suitable for rapid isolation of the mentioned flavonoid biosides from the crude methanol extract of S. annua without prior fractionation by other chromatographic techniques.

AB  - The focus of research group from Department of Pharmacognosy at the Faculty of
Pharmacy in Belgrade is on natural products, their chemistry and application. Main subjects
of research are analysis of composition and bioactivities of various plant isolates (extracts,
essential oils), isolation and structural analysis of active specialized plant metabolites,
optimization of extraction procedures involving new techniques, and green chemistry
principles. Since the Balkan Peninsula is one of the centers of plant biodiversity, our main
objective is to explore the autochthonous taxa, especially endemic and rare ones, in terms
of their distribution, conservation, morphology, chemistry, pharmacology and potential
utilization. The main outcomes of these studies are the identification of new herbal raw
materials and isolates, their pharmacognostic and pharmacological characterization, and
the development of methods of quality assessment and evaluation of safe use of natural
products, along with exploring other ways of their use. Important segment of research
focuses on identifying natural principles and influences on secondary plant metabolism
and studying the chemotaxonomic potential of certain plant metabolites. Basic research
interest of this group is constantly upgraded trough scientific collaboration with colleagues
from other research institution from Serbia, neighboring regions and abroad, resulting in
many scientific and professional publications, and PhD theses.

AB  - Stachys milanii Petrović (Lamiaceae) is an endemic of central and eastern part of Balkan Peninsula, growing mainly in river valleys on semi-saline soils, often along the edge of cereal fields and abandoned arable lands. It is a critically endangered and strictly protected species in Serbia, with just one proven habitat (1). So far it was investigated on herb essential oil composition and antimicrobial activity, and on fatty acid composition of seeds (2). 
Aim of this work was to investigate antiradical potential and phenolic constituents of dry methanol herb extract. Aerial flowering parts (herb) of S. milanii were collected in North Macedonia (Ovče Pole). Powdered, dried plant material was extracted by bimaceration with chloroform, and then with methanol. Obtained dry methanol extract was used for further analysis. Using appropriate spectrophotometric procedure strong, concentration-dependant anti-DPPH activity of extract was observed (SC50=34.15 μg/mL). Also spectrophotometrically, using Folin-Ciocalteu reagent, significant total phenolics content (0,47 mg gallic acid/mg dry extract) was determined. In TLC-DPPH test observed yellow zones indicated presence of few compounds which strongly neutralized purple DPPH radical. LC-MS analysis revealed presence of one phenolic acid, four phenylethanol glycosides, eight flavonoid glycosides derivatives of 8-hydroxyflavones, and one apigenin glycoside. Using commercial standards in extract were identified and quantified: chlorogenic acid (1.63%) and acteoside (2.65%), as the main phenylethanoid. In 8-hydroxyflavones fraction six hypolaetine and two isoscutellarein derivatives were present; using compounds previously isolated from S. alpina subsp. dinarica Murb. herb, 4'-O-methylhypolaetine-7-O-[6'''-acetylallosyl(1®2)]-glucopyranoside and isoscutellarein-7-O-[6'''-acetylallosyl(1®2)]-glucopyranoside were identified, representing the most prominent flavonoids in investigated extract, respectively.

AB  - The use of medicinal plants has significant impact on human health (1,2). Approximately 13.000 medicinal plants are used worldwide (1), and in developing countries 65-80% of the population uses them for medicinal purposes (2). 
The aim of the study was to examine the awareness of patients in the city of Belgrade about herbal products. Quantitative, non-experimental research was conducted using a purpose-built questionnaire in public pharmacies during February 2020.
The survey involved 954 respondents, mostly women (66.5%); 39.1% of respondents aged 20-40 years. Most of the respondents were employed (65.2%), with secondary school education (45.2%). The vast majority (97.3%) used medicinal plants/herbal products, mostly occasionally, as needed (38.4%). The main motive for their use was health improvement (75.2%), while 16.2% used them to treat diseases. The primary sources of information on medicinal plants/herbal products are pharmacists (45.1%) and the media (24.2%). About a third of the respondents (37.3%) stated that they knew the difference between herbal medicinal products and dietary supplements. 52.9% of respondents claimed that herbal medicinal products are effective, but less than synthetic drugs, while 61.5% claimed that they are safe, but with certain side effects. A third of respondents (32.9%) had difficulty understanding instructions for use and they needed additional explanations. Majority (70.6%) received advice from a pharmacist when buying a herbal product.
Pharmacists are health professionals who have adequate knowledge of medicinal plants/herbal products and have a key role in advising patients in order to implement the proper use of herbal medicinal and other herbal products.

teritoriji grada Beograda o biljnim proizvodima. Sprovedeno je kvantitativno,
neeksperimentalno istraživanje pomoću namenski kreiranog upitnika u javnim apotekama
tokom februara 2020. godine. U istraživanju je učestvovalo 954 ispitanika, od koji 66,5%
ženskog pola; 39,1% starosti 20-40 godina. Većina je bila zaposlena (65,2%), sa
srednjoškolskim obrazovanjem (45,2%). Najveći broj ispitanika (97,3%) je koristio lekovite
biljke/biljne proizvode i to uglavnom povremeno, po potrebi (38,4%). Glavni motiv za

ispitanika (37,3%) je navelo da zna razliku između biljnih lekovitih proizvoda i dijetetskih
suplemenata. Da su biljni lekoviti proizvodi efikasni, ali manje od sintetskih lekova, smatralo
je 52,9% ispitanika, dok je 61,5% smatralo da su oni bezbedni za upotrebu, ali sa mogućim
neželjenim efektima. Trećina anketiranih (32,9%) imala je poteškoće da razume uputstva za
upotrebu biljnih proizvoda, te su im bila potrebna dodatna objašnjenja. Najveći deo
ispitanika (70,6%) dobijao je savet farmaceuta pri kupovini biljnog proizvoda. Farmaceuti su
zdravstveni profesionalci koji poseduju adekvatna znanja o lekovitim biljkama/biljnim
proizvodima i imaju ključnu ulogu u savetovanju pacijenata u cilju sprovođenja pravilne
primene biljnih lekovitih i drugih biljnih proizvoda.

AB  - Nuts and seeds are an indispensable part of the plant-based diet, which is becoming increasingly popular due to the evidence of their health benefits and contribution to sustainability and planetary health. Since the health effects of consuming nuts and seeds directly depend on their nutritional composition and consumed amount, it is essential to know the exact chemical composition of each nut and seed so that appropriate dietary interventions can be adequately planned. The present study aimed to examine the chemical composition of nuts and seeds obtained from the Serbian market and to highlight the similarities and differences in their nutritional composition. In twenty-four samples of nuts and seeds, the content of total lipids, including fatty acid profile, total proteins, including amino acid profile, total carbohydrates (sugars and fibers), phytosterols, and minerals were determined. Content of selected nutrients in grams (g) or milligrams (mg) of nuts and seeds was expressed in one portion (28 g) and as a percentage of contribution of Reference Intakes (% RI) and Dietary Reference Value (% DRV) for macronutrients and minerals, respectively. Some of the seed representatives appeared to be rich sources of essential omega-3 fatty acid, α-linolenic acid (ALA) (flax seed vs. walnut, 6.50 vs. 0.56 g per portion, respectively), dietary fibers (chia seed vs. raw almond, 10.6 vs. 3.4 g per portion, respectively), calcium (black sesame seed vs. almond roasted, 32.4% NRV vs. 8.1% NRV per portion, respectively), magnesium (hemp seed vs. Brazil nut, 38.3% NRV vs. 27.8% NRV per portion, respectively), and zinc (hemp seed vs. pine nut, 21.4% NRV vs. 17.6% NRV per portion, respectively). Our results highlighted the crucial role of seeds in the diet, especially as a better source of nutrients compared to nuts. Furthermore, it was seen that nuts and seeds are different and complementary in their composition. Thus, in order to meet the needs for certain nutrients for which nuts and seeds are used as dietary sources (essential fatty acids, minerals, dietary fibers), it would be beneficial to combine both nuts and seeds as a part of a healthy dietary pattern.

The present study aimed to examine the chemical composition of nuts and seeds obtained from the Serbian market and to highlight the similarities and differences in their nutritional composition. In twenty-four samples of nuts and seeds, the content of total lipids, including fatty acid profile, total proteins, including amino acid profile, total carbohydrates (sugars and fibers), phytosterols, and minerals were determined. Content of selected nutrients in grams (g) or milligrams (mg) of nuts and seeds was expressed in one portion (28 g) and as a percentage of contribution of Reference Intakes (% RI) and Dietary Reference Value (% DRV) for macronutrients and minerals, respectively. Chemical profiling of six Stachys taxa from Balkan Peninsula

AB  - Aim of this work was the chemical profiling of six Stachys taxa from Balkan Peninsula: S. recta subsp. nitens, S. atherocalyx, S. beckeana, S. zepcensis (all four belonging to S. recta group), S. alpina subsp. dinarica and S. plumosa. Dry ethanol extracts of aerial flowering plant parts were qualitatively and quantitatively characterized for their phenolic composition using the HPLC-DAD-ESI-MS technique. In total, 44 phenolic compounds were detected, belonging to three main classes of constituents: phenolic acids, phenylethanol glycosides (PhGs) and flavonoids. Chlorogenic acid was the main phenolic acid in all samples. Regarding PhGs, in all investigated extracts acteoside and lavandulifolioside were present. In S. recta group, extracts of S. atherocalyx, S. recta subsp. nitens and S. beckeana showed similar pattern considering phenylethanol glycosides, while S. zepcensis contained only the two aforementioned compounds. PhGs profile of S. plumosa extract comprised the same compounds from S. recta group, but it was also characterized by the presence of few high-molecular compounds. PhGs composition of S. alpina subsp. dinarica extract was somewhat similar with those of S. recta group and also with S. plumosa, but still specific by the presence of echinacoside and leucosceptoside A. The main flavonoids in extracts of Stachys recta group as well as in S. alpina subsp. dinarica extract were 8-hydroxyflavone glycosides (7-O-allosylglucosides of isoscutellarein and hypolaetin and their 4′-methyl derivatives). In addition, in the extract of S. alpina subsp. dinarica two luteolin-7-O-hexosides were also present. On the other hand, S. plumosa extract differed considerably containing mainly derivatives of chrysoeriol and apigenin, while 8-hydroxyflavones (isoscutellarein derivatives) were detected in traces. The results confirmed the importance of flavone 7-O-allosylglucosides at the sectional level of Stachys genus.

AB  - Many types of vegetable oils produce around the world. They are useful in nutrition,
prevention and treatment of diseases. Some oils traditionally have been well known for a long
time and have been shown many potential health benefits, such as, cardioprotective effect,
effect on digestion, risk of some cancer and it is also used in the treatment of skin infections.
This work was conducted to study some quality criteria of oil extracted from seven different
raw seeds purchased in the Serbian market, namely: sunflowers, sesame seeds, black sesame
seeds, tickles seeds, chia seeds, flax seeds and hemp seeds. Oils are essentially regarded in
terms of quality based on their free fatty acid, iodine value and peroxides value. Oils were
extracted from the seeds by cold extraction with petroleum ether. Quality characteristics,
such as refractive index, acid value, peroxide value, iodine value and saponification value and
unsaponifiable matter were determined. For the determination of these parameters, official
methods (ISO 6320; ISO660; ISO 3960; ISO 3961; ISO 3657 and ISO 18609, respectively) were
used. The contents of investigated parameters in seed samples were determined in the range
of 1.467 to 1.485 nD for refractive index; 1.62 to 4.95 mg KOH/g for acid value; 5.21 to 7.45
for peroxide value; 118 to 197 for iodine value; 180 to 193 for saponification value and 3.14
to 7.26 g/kg for the unsaponifiable matter.
Data obtained in this investigation indicate that all quality parameters of the seed samples,
except peroxide value , comply with the national and Codex standards.

By LC/MS, the contents of arbutin (99.9–131.0 mg/g), chlorogenic acid (2.2–6.3 mg/g), catechin (1.0–5.3 mg/g) and total dimeric and trimeric procyanidins (42.2–61.3 mg/g), including procyanidin B2 (8.9–17.2 mg/g), were determined. Colorimetri- cally, high contents of total phenolics (436.2–533.4 mg GAE/g) and tannins (339.4–425.7 mg GAE/g), as well as strong total antioxidant activities (FRAP values 4.5–5.9 mmol Fe2+ /g), and DPPH (SC50 = 6.6–7.1 μg/ml) and hydroxyl radical (SC50 = 447.1–727.7 μg/ml) scavenging abilities were revealed. In vitro, all extracts exhibited notable inhibition of α-amylase (IC50 = 310.8–617.7 μg/ml) and particularly strong inhibition of α-glucosidase (IC50 = 2.1–3.7 μg/ml). Molecular docking predicted that among identified compounds procyanidin B2 is the best inhibitor of these carbohydrate-digesting enzymes. Obtained results showed that the barks of investigated Pyrus hybrid and its parent taxa have similar composition and bioactivity.

Pumpkin seed cake antioxidant and nutritional value of selected samples

AB  - Pumpkin seed cake, which remains after cold pressing oil extraction, is a nutritionally valuable but inexpensive raw material that is also considered as a potentially rich source of biologically active substances. Thus, the aim of this study was to measure total phenolics content (TPC) of four samples of pumpkin seed cake flour, as well as their antiradical potency by means of DPPH radical scavenging. In order to determine the nutritional value of each sample, moisture, ash, cellulose, lipids, proteins, carbohydrates, mineral content and fatty acid composition were also investigated using conventional methods. Concerning food safety issues the amounts of heavy metals and pesticides were also determined. The results obtained, pointed out differences between samples in their TPC, anti-DPPH activity and nutritional characteristics. TPC, determined as gallic acid equivalents (GA) using the spectrophotometric method with FC reagent, ranged from 24.9-194.1 mg GA/100 g. Correlated with TPC, observed anti-DPPH activity was modest with SC50 values ranged from 0.9-18.5 mg/ml, respectively. As for parameters of nutritional value, obtained results were in the line with previous findings, with protein content ca. 50%. Variations, mainly in the mineral (14.61-30.70 mg/100 g) and the carbohydrate content (9.38-21.86%), could be explained by the different geographical origin of the pumpkins. All tested samples complied with the approved health standards related to the content of heavy metals and pesticides. Nevertheless observed differences, it could be concluded that pumpkin seed cake could be considered as a naturally rich source of proteins, cellulose and minerals (Fe, Cu, Zn, Mg) with a reduced amount of oil, safe for human consumption. It is also a good source of polyphenols, thus its potential as a functional food ingredient should not be neglected.

Prema ovoj monografiji usitnjena herba se koristi oralno u obliku čaja (infuza) za
ublažavanje kašlja povezanog sa prehladom i za simptomatsko ublažavanje blagog
gastrointestinalnog diskomfora, kao i dermalno u obliku infuza ili dekokta za tretman manjih
inflamacija kože i modrica. U Evropskoj farmakopeji (Ph. Eur.) za sada ne postoje
monografije za ovu biljnu drogu i njeno etarsko ulje.
Cilj ovog rada bio je ispitivanje hemijskog sastava etarskog ulja izolovanog iz
komercijalnog uzorka herbe kritskog origana poreklom iz Grčke, kao i procena njegove
antiradikalske aktivnosti.
Etarsko ulje je izolovano iz herbe destilacijom vodenom parom u aparaturi po
Clevenger-u, prema postupku Ph. Eur. Prinos etarskog ulja iznosio je 1,51% V/m, tj. 1,46%
m/m. Hemijski sastav etarskog ulja analiziran je gasnom hromatografijom sa FID i MS
detekcijom (GC-FID i GC-MS). Identifikovane su 42 komponente, koje su činile 94,6% sastava
etarskog ulja. Procentualno najveći udeo u etarskom ulju činili su oksidovani monoterpeni
(58,8%; 9 jedinjenja), a sledili su monoterpenski ugljovodonici (27,4%; 12 jedinjenja),
seskviterpenski ugljovodonici (5,6%; 12 jedinjenja) i oksidovani seskviterpeni (2,3%; 6
jedinjenja). Glavna komponenta etarskog ulja bio je monoterpenski fenol karvakrol (51,4%),
dok su po količini sledili monoterpenski ugljovodonici p-cimen (14,3%) i γ-terpinen (9,3%).
Dobijeni rezultati bili su u skladu sa literaturnim podacima, pri čemu je u slučaju uzorka
analiziranog u ovom radu, prinos etarskog ulja bio veći od prethodno publikovanih vrednosti
(1, 2).
Antiradikalska aktivnost izolovanog etarskog ulja ispitana je spektrofotometrijskim
DPPH testom. Stepen antiradikalske aktivnosti je rastao linearno sa povećanjem
koncentracije etarskog ulja, pri čemu je koncentracija u kojoj je etarsko ulje inhibiralo 50%
DPPH radikala (SC50) iznosila 141,77 μg/mL. Značajan anti-DPPH potencijal etarskog ulja
herbe kritskog origana može se barem delom objasniti velikim udelom karvakrola, čiji je
antiradikalski potencijal ranije pokazan (3).
Rezultati sprovedenih hemijskih ispitivanja mogu biti od značaja za buduće
definisanje parametara farmaceutskog kvaliteta droge Origani dictamni herba i njenog
etarskog ulja.

Medicines Agency (EMA/HMPC) adopted EU monograph on traditional use of Cretan dittany
herb, Origani dictamni herba. According to this monograph, comminuted herb is used orally
as herbal tea (infusion) for the relief of cough associated with cold and for mild
gastrointestinal disorders, as well as cutaneously as infusion or decoction for the treatment
of minor skin inflammations and bruises. European Pharmacopoeia (Ph. Eur.) monographs
for this herbal drug and its essential oil do not exist for now.
The aim of this work was to investigate chemical composition of the essential oil
obtained from a commercial sample of Cretan dittany herb and to evaluate its antiradical
activity.
The essential oil was isolated from the herb by hydrodistillation in a Clevenger‐type
apparatus, according to procedure given in Ph. Eur. The yield was 1.51% V/w, i.e. 1.46%
w/w. Chemical composition of the essential oil was analyzed by gas chromatography with
FID and MS detection (GC-FID and GC-MS). A total of 42 components were identified,
comprising 94.6% of the composition of the essential oil. The dominant were oxygenated
monoterpenes (58.8%; 9 compounds), followed by monoterpene hydrocarbons (27.4%; 12
compounds), sesquiterpene hydrocarbons (5.6%; 12 compounds) and oxygenated
sesquiterpenes (2.3%; 6 compounds). The main component of the essential oil was
monoterpene phenol carvacrol (51.4%), followed by monoterpene hydrocarbons p-cymene
(14.3%) and γ-terpinene (9.3%). Obtained results were in accordance with the literature
data; it should be noted that in the case of the sample analyzed in this work, the essential oil
yield was higher than previously published values (1, 2).
The antiradical activity of the essential oil was investigated by spectrophotometric
DPPH test. The percentage of antiradical activity increased linearly with the increase of
essential oil concentration. The concentration at which the essential oil inhibited 50% DPPH
radicals (SC50) was 141.77 μg/mL. Significant anti-DPPH potential of the essential oil of
Cretan dittany herb can be at least partly explained by the presence of high quantity of
carvacrol, which antiradical potential was demonstrated previously (3).
The results of performed chemical investigations may be important for defining
pharmaceutical quality parameters of the herbal drug Origani dictamni herba and its
essential oil in the future.

The chemical composition, antimicrobial and antiradical properties of the essential oil of Achillea grandifolia aerial parts from Serbia

AB  - Aromatic plants and essential oils have many applications in medicine, pharmaceuticals, cosmetics, and the food industry. The essential oil of the flowering aerial parts of Achillea grandifolia, obtained by hydrodistillation, was analyzed for its constituents and investigated for antimicrobial and radical scavenging activity. The essential oil was characterized by a high amount of oxygenated monoterpenes (72.7%) with 1,8-cineole (29.2%) and camphor (23.4%) being the most abundant. Sesquiterpenes were present in smaller quantities (4.8%). Antimicrobial activity was tested against eight ATCC bacterial strains and two ATCC strains of Candida albicans. The essential oil exhibited highly pronounced antimicrobial activity against Micrococcus luteus with a MIC value of 3.50 μg/mL, as well as significant antimicrobial activity (<100 μg/mL) against Staphylococcus aureus, S. epidermidis and Bacillus subtilis. Gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa were resistant. Achillea grandifolia essential oil exhibited concentration-dependent antiradical activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical with an SC50 value of 5.4 mg/mL. The TLC-DPPH assay revealed two main light yellow spots indicating components with anti-DPPH activity, which after isolation were identified as 1,8-cineole and camphor.
AB  - Aromatične biljke i etarska ulja nalaze primenu u medicini, farmaciji, kozmetičkoj, parfimerijskoj i prehrambenoj industriji. Etarsko ulje iz nadzemnih delova u cvetu Achillea grandifolia izolovano je destilacijom vodenom parom i ispitivano u pogledu hemijskog sastava, antimikrobne i antiradikalske aktivnosti. Etarsko ulje se odlikovalo velikom količinom oksidovanih monoterpena (72,7%) sa 1,8-cineolom (29,2%) i kamforom (23,4%) kao dominantnim jedinjenjima. Seskviterpeni su bili prisutni u manjoj količini (4,8%). Antimikrobna aktivnost je ispitivana prema osam ATCC sojeva bakterija i dva ATCC soja Candida albicans. Etarsko ulje je pokazalo veoma izraženu antimikrobnu aktivnost prema Micrococcus luteus sa MIC vrednošću od 3,50 μg/mL, kao i značajnu aktivnost prema Staphylococcus aureus, S. epidermidis i Bacillus subtilis. Gram-negativne bakterije Escherichia coli i Pseudomonas aeruginosa bile su rezistentne. Etarsko ulje je ispoljilo dozno-zavisnu antiradikalsku aktivnost prema DPPH radikalu sa SC50 vrednošću od 5,4 mg/mL. U TLC-DPPH testu uočene su dve glavne žute zone koje odgovaraju anti-DPPH aktivnim jedinjenjima, a koja su nakon izolovanja identifikovana kao 1,8-cineol i kamfor.

The essential oil exhibited highly pronounced antimicrobial activity against Micrococcus luteus with a MIC value of 3.50 μg/mL, as well as significant antimicrobial activity (<100 μg/mL) against Staphylococcus aureus, S. epidermidis and Bacillus subtilis. Gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa were resistant. Achillea grandifolia essential oil exhibited concentration-dependent antiradical activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical with an SC50 value of 5.4 mg/mL. The TLC-DPPH assay revealed two main light yellow spots indicating components with anti-DPPH activity, which after isolation were identified as 1,8-cineole and camphor., Aromatične biljke i etarska ulja nalaze primenu u medicini, farmaciji, kozmetičkoj, parfimerijskoj i prehrambenoj industriji. Etarsko ulje iz nadzemnih delova u cvetu Achillea grandifolia izolovano je destilacijom vodenom parom i ispitivano u pogledu hemijskog sastava, antimikrobne i antiradikalske aktivnosti. Etarsko ulje se odlikovalo velikom količinom oksidovanih monoterpena (72,7%) sa 1,8-cineolom (29,2%) i kamforom (23,4%) kao dominantnim jedinjenjima. Seskviterpeni su bili prisutni u manjoj količini (4,8%). Antimikrobna aktivnost je ispitivana prema osam ATCC sojeva bakterija i dva ATCC soja Candida albicans. Etarsko ulje je pokazalo veoma izraženu antimikrobnu aktivnost prema Micrococcus luteus sa MIC vrednošću od 3,50 μg/mL, kao i značajnu aktivnost prema Staphylococcus aureus, S. epidermidis i Bacillus subtilis. Gram-negativne bakterije Escherichia coli i Pseudomonas aeruginosa bile su rezistentne. Etarsko ulje je ispoljilo dozno-zavisnu antiradikalsku aktivnost prema DPPH radikalu sa SC50 vrednošću od 5,4 mg/mL. U TLC-DPPH testu uočene su dve glavne žute zone koje odgovaraju anti-DPPH aktivnim jedinjenjima, a koja su nakon izolovanja identifikovana kao 1,8-cineol i kamfor.",

METHOD / DESIGN:
Gas chromatography with flame ionization detector (GC-FID) and gas chromatography with mass spectrometry (GC-MS) analysis of the unsaponifiable fractions was performed on an Agilent 7890A GC equipped with 5975C (inert XL EI/CI) MSD and a FID detector connected by a capillary flow technology two way splitter with make-up (250 °C). A HP-5MS capillary column (Agilent, 30 m × 0.25 mm, 0.25 μm film thickness) was used. The identification of the compounds was based on the comparison of their retention indices (RI), Rt, and mass spectra from NIST/NBS 05, Wiley libraries 8th edition and NIST Chemistry WebBook.29.
RESULTS:
Phytosterols were analyzed as volatile derivatives obtained by the silanization of residual unsaponifiable fractions. Cholesterol standard was used for quantification. Among them, the most abundant was β-sitosterol, followed by stigmasterol, isofucosterol and campesterol (Figure 1).
CONCLUSIONS:
Our results indicate that the seeds (sesame, black sesame, chia seed) contain higher amount
of phytosterols in comparison to the nuts. These results are in correlation with literature
data. The product quality on Serbian market is appropriate according to phytosterols content.

AB  - Numerous epidemiological studies have confirmed that lifestyle and diet can be very effective in preventing and treating many non-communicable diseases. Replacement of unhealthy food intake between the main meals, i.e. snacks, with foods rich in nutrients such as nuts make a significant contribution to the prevention and treatment of some types of cancer, diabetes mellitus type 2 and cardiovascular diseases. Among the essential bioactive components contained in nuts are healthy minerals, such as calcium, magnesium, potassium, phosphorus, zinc, ferrum. The objective of this study was to identify the mineral composition of several varieties of nuts, which are part of everyday diet. The concentrations of 7 elements (Na, K, Ca, Mg, Fe, Zn, and P) were determined in nut samples (peanut, almond, hazelnut, walnut, Brazil nut, cashew, pecan, pistachio and pine nuts). The mineral contents were analyzed by inductively coupled plasma optical emission spectrometry using a Agilend 5110 dual view, ICP-OES, after microwave-assisted acid digestion. The contents of investigated elements in nut samples were determined as mg/kg in the range from 21.4 to 63.0 for Zn; 25.9 to 107.0 for Fe; 84.2 to 1995 for Ca; 981 to 3724 for Mg; 2295 to 4414 for K and 2814 to 7244 for P. The minimum contents of Na were below the detection limit of the method used. The obtained experimental data are another confirmation that nuts, and above all almond and Brazil nut, are important sources of bioactive components i.e. healthy minerals. Knowing the effect on human health, it is necessary the nuts to be recommended and included in the usual daily diet.

AB  - Nuts are nutrient-dense foods with complex matrices rich in unsaturated fatty acids and other bioactive compounds. Numerous reported health benefits of nuts may be attributed to their unique nutrient profiles and synergistic interaction of their constituents, including macronutrients, micronutrients and phytochemicals. The objective of this study was to explore pharmacy students’ nuts consumption practice, as well as their attitudes and knowledge concerning nuts nutritional and health properties. A cross-sectional survey based on the online self-administered questionnaire, was conducted in September 2020. Among respondents (n=136, 90.4% female and 9.6% male, mean age 23.7±2.0 years), more than half agreed or strongly agreed that regular consumption of nuts might have beneficial effects on health, including reducing the risk of cardiovascular disease. Furthermore, the majority of participants perceived nuts as healthy sources of proteins and monounsaturated and polyunsaturated fats. The most common reason cited by respondents for eating nuts were taste (80.2%), followed by health effects (53.1%) and nutrient profiles (45.0%). The most commonly consumed nuts among pharmacy students were almonds (35.2%) and peanuts (28.4%), followed by hazelnuts (19.1%) and walnuts (15.6%). Based on self-reported data, students predominantly consume raw nuts (54.0%) as a snack between meals (78.5%). Nutrition-related courses within the faculty curriculum were the main sources of acquiring information regarding the beneficial effects nuts exert on health outcomes. Although certain gaps in knowledge were determined, in most senior students, the attitudes and dietary practices regarding the consumption of nuts were at an adequate level. Nevertheless, expanded efforts and additional educational programs may be useful in strengthening the capacities of these future health professionals for providing adequate nutrition advice and proactive advocacy of nuts health benefits.

The objective of this study was to explore pharmacy students’ nuts consumption practice, as well as their attitudes and knowledge concerning nuts nutritional and health properties. A cross-sectional survey based on the online self-administered questionnaire, was conducted in September 2020. Among respondents (n=136, 90.4% female and 9.6% male, mean age 23.7±2.0 years), more than half agreed or strongly agreed that regular consumption of nuts might have beneficial effects on health, including reducing the risk of cardiovascular disease. Furthermore, the majority of participants perceived nuts as healthy sources of proteins and monounsaturated and polyunsaturated fats. The most common reason cited by respondents for eating nuts were taste (80.2%), followed by health effects (53.1%) and nutrient profiles (45.0%). The most commonly consumed nuts among pharmacy students were almonds (35.2%) and peanuts (28.4%), followed by hazelnuts (19.1%) and walnuts (15.6%). Based on self-reported data, students predominantly consume raw nuts (54.0%) as a snack between meals (78.5%). Nutrition-related courses within the faculty curriculum were the main sources of acquiring information regarding the beneficial effects nuts exert on health outcomes. Although certain gaps in knowledge were determined, in most senior students, the attitudes and dietary practices regarding the consumption of nuts were at an adequate level. Nevertheless, expanded efforts and additional educational programs may be useful in strengthening the capacities of these future health professionals for providing adequate nutrition advice and proactive advocacy of nuts health benefits.",

Amino acids composition in selected nuts

AB  - In recent years, we are witnessed that a nuts very popular food and is recommended as part of a diet because it contains numerous bioactive compounds. Nuts contain a high quantity of protein, i.e. amino acids that are known to be necessary for important processes such as tissue growth, energy production, immune function and nutrient absorption. In order to check the amount of individual amino acids found in the favorite samples of nuts of our citizens, we determined total protein and amino acid profile (17 amino acids) in nine samples of nuts (peanut, almond, hazelnut, walnut, Brazil nut, cashew, pecan, pistachio, and pine nuts). Total amino acids composition was determined by ion chromatograph with electrochemical detector, manufactured by Thermo, model ICS-5000, with silver reference electrode (Ag / AgCl) and gold (Au) working electrode and chromatographic column AminoPac PA10 and pre-column AminoPac PA10 guard. Analyzed nuts samples contained 11.7-29.2 g/100 g total proteins. Pecan contained the least protein, while peanut was the richest in total protein. Among the amino acids, glutamic acid, aspartic acid and arginine contents were generally high, whereas methionine, cysteine, histidine and tyrosine contents were low. Peanut was the most important source for essential amino acids (lysine 1.08 g/100 g; histidine 0.71 g/100 g; threonine 0.87 g/100 g; leucine 1.97 g/100 g; isoleucine 1.25 g/100 g; valine 1.07 g/100 g; phenylalanine 1,38 g/100 g; and methionine 0.4 g/100 g). Cashew was the biggest source of essential amino acids, 37% (essential/total amino acids), while the smallest source was almond, 27%. Almond was the biggest source of non-essential amino acids, 73%. The obtained results indicate that the nuts are incomplete in the content of amino acids. Knowledge of the amino acid composition of nuts is essential for combination with other food groups when formulating a balanced diet.
