Chemical Analysis and Investigation of Antimicrobial and Antibiofilm Activities of Prangos trifida (Apiaceae)

AB  - Plants of the genus Prangos are intensively investigated as potential new sources of bioactive isolated products. In this work, the chemical composition of volatile constituents (essential oils and headspace volatiles) and dichloromethane extracts, as well as antimicrobial and antibiofilm activities of essential oils and MFDEs (methanol fractions of dichloromethane extracts) of Prangos trifida from Serbia, were investigated. Volatiles of roots, leaves, stems and fruits, and fatty acids and phytosterols in dichloromethane extracts of roots and fruits were analyzed by GC-FID-MS, whereas coumarins in MFDEs by LC–MS and some isolated coumarins by 1H-NMR. Minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations/minimum fungicidal concentrations (MBCs/MFCs) of essential oils and MFDEs were determined against 13 microorganisms. Antibiofilm activity was assessed against four microorganisms. Additionally, congo red and ergosterol binding assays were conducted to elucidate selected mechanisms of antibiofilm action in the case of Candida albicans. Total of 52 volatile constituents, 16 fatty acids, eight phytosterols and 10 coumarins were identified. Essential oils demonstrated significant activity, surpassing that of commercial food preservatives, against six tested molds from the Aspergillus, Penicillium and Trichoderma genera, as well as against bacteria Staphylococcus aureus and Bacillus cereus. Most of the oils strongly inhibited the formation of biofilms by S. aureus, Listeria monocytogenes and Escherichia coli. MFDEs exhibited noteworthy effects against B. cereus and the tested Aspergillus species, particularly A. niger, and significantly inhibited C. albicans biofilm formation. This inhibition was linked to a marked reduction in exopolysaccharide production, while antifungal mechanisms associated with ergosterol remained unaffected.

stems and fruits of Prangos trifida (Mill.) Herrnst. & Heyn s.l. (Apiaceae) were investigated.
Plant material was collected in Sićevo Gorge (Kusača) in 2020. Essential oils were isolated by
hydrodistillation in Clevenger-type apparatus and analyzed by GC-FID-MS. Antimicrobial
activity was tested by microdilution method against food contaminants: Gram-positive
(Staphylococcus aureus, Bacillus cereus and Listeria monocytogenes), Gram-negative bacteria
(Escherichia coli, Enterobacter cloacae and Salmonella Typhimurium), and molds (Aspergillus
fumigatus, A. niger, A. versicolor, Penicillium funiculosum, P. verrucosum var. cyclopium and
Trichoderma harzianum); artificial food preservatives E211 and E224 were used as controls.
Dominant constituents of essential oils were monoterpenes (66.2-87.2%). Terpinolene
(36.2%) and p-cymene (11.5%) prevailed in root oil, (E)-β-ocimen (23.2%) and terpinolene
(18.1%) in leaf oil, p-cimen-8-ol (21.8%) and p-cimen (14.1%) in stem oil, and p-cimen
(25.4%) and limonene (14.4%) in fruit oil. Essential oils showed significant antifungal
activity (MIC=0.10-0.78 mg/mL; MFC=0.20-1.56 mg/mL), better compared to both tested
commercial preservatives, and interesting antibacterial activity (MIC=0.20-6.25 mg/mL;
MBC=0.39-12.5 mg/mL), which was in the case of root and stem oils better than activity of
both preservatives against S. aureus and B. cereus. It should be noted that composition of
tested oils was in accordance with composition of the oils of this plant from vicinity of
Madrid (1) and from Crimea (2), while their antimicrobial activity was tested in this work for
the first time. Obtained results justify further research of P. trifida essential oils as new
natural food preservatives.
AB  - Cilj ovog rada bio je da se ispitaju hemijski sastav i antimikrobna aktivnost etarskih
ulja korena, listova, stabljika i plodova Prangos trifida (Mill.) Herrnst. & Heyn s.l. (Apiaceae).
Biljni materijal je prikupljen na području Sićevačke klisure (Kusača) 2020. god. Etarska ulja
su dobijena destilacijom vodenom parom u aparaturi po Klevendžeru i analizirana metodom
GC-FID-MS. Antimikrobna aktivnost testirana je mikrodilucionom metodom na
kontaminantima hrane: Gram-pozitivnim (Staphylococcus aureus, Bacillus cereus i Listeria
monocytogenes), Gram-negativnim bakterijama (Escherichia coli, Enterobacter cloacae i
Salmonella Typhimurium), i plesnima (Aspergillus fumigatus, A. niger, A. versicolor,
Penicillium funiculosum, P. verrucosum var. cyclopium i Trichoderma harzianum); kao
kontrole korišćeni su sintetski konzervansi E211 i E224. Dominantni sastojci etarskih ulja
bili su monoterpeni (66,2-87,2%). Najzastupljeniji u ulju korena bili su terpinolen (36,2%) i
p-cimen (11,5%), u ulju listova (E)-β-ocimen (23,2%) i terpinolen (18,1%), u ulju stabljika p-cimen-
8-ol (21,8%) i p-cimen (14,1%), a u ulju plodova p-cimen (25,4%) i limonen (14,4%).
Etarska ulja su pokazala značajnu antifungalnu aktivnost (MIK=0,10-0,78 mg/mL;
MFK=0,20-1,56 mg/mL), bolju u poređenju sa oba testirana komercijalna konzervansa, i
interesantnu antibakterijsku aktivnost (MIK=0,20-6,25 mg/mL; MBK=0,39-12,5 mg/mL),
koja je u slučaju ulja korena i stabljika prema S. aureus i B. cereus bila bolja u odnosu na oba
konzervansa. Treba istaći da je sastav ispitivanih etarskih ulja bio u skladu sa sastavom ulja
ove biljke iz okoline Madrida (1) i sa Krima (2), dok je njihova antimikrobna aktivnost
ispitivana u ovom radu po prvi put. Dobijeni rezultati predstavljaju dobru osnovu za dalja
istraživanja etarskih ulja P. trifida kao novih prirodnih konzervanasa.

AB  - Antimicrobial properties of extracts of underground parts of three Laserpitium L. (Apiaceae) species, namely Laserpitium latifolium L., Laserpitium zernyi Hayek and Laserpitium ochridanum Micevski, were investigated. The investigated species are widely used as functional foods, as spices and for preparations in traditional medicine for treating complaints connected with infection and inflammation. Furthermore, antimicrobial and antibiofilm effects of laserpitine, the most abundant compound in the chloroform extract of Laserpitium latifolium, and guaianolide sesquiterpene lactones, such as, isomontanolide, montanolide and tarolide, principal components of the extracts of Laserpitium zernyi and Laserpitium ochridanum were assessed. The antimicrobial activity was tested using the microdilution method against five pathogenic bacteria and five fungi, as well as in the microplate biofilm assay on two Candida clinical isolates (C. albicans and C. krusei). Among the extracts, Laserpitium latifolium showed the most prominent activity. Isolated metabolites exerted higher effects against fungal than against bacterial strains, isomontanolide being the most active. Interestingly, all constituents showed higher potential on inhibition of biofilm formation than fluconazole, a reference compound. Tested metabolites may be good novel agents with high antifungal and antibacterial potential that might find practical applications in food industry as food preservatives in order to retard the growth of food spoiling microbes, but only after detailed safety assessments.

AB  - The phenolic profile and antibacterial, antimutagenic and antitumour activities of the methanolic extracts of Veronica urticifolia Jacq. were evaluated. HPLC-DAD/ESI-MS analysis revealed the presence of phenolic acids, flavonoids and phenylethanoids, with acteoside as the main component (14.9 mg/g of extract). Antibacterial effect was determined using the microbroth dilution assay and Staphylococcus aureus was the most sensitive strain (MIC and MBC = 7.5 mg/mL). Antimutagenic activity was evaluated by Ames mutagenicity assay. At 1 mg/plate, the tested extract afforded high protection against the mutagenicity of nitroquinoline-N-oxide (4NQO) to Salmonella typhimurium strain TA100 (inhibition rate 48.3%). Antitumour activity was screened in Ehrlich ascites carcinoma (EAC) model. Pretreatment with 2 mg/kg body weight showed statistically significant decrease in tumour cell viability, while ascites volume and tumour cell count became slightly decreased, but not to a statistically significant extent. The results indicate that V. urticifolia deserves further research into its potential chemoprevention effects.

abstract = "The phenolic profile and antibacterial, antimutagenic and antitumour activities of the methanolic extracts of Veronica urticifolia Jacq. were evaluated. HPLC-DAD/ESI-MS analysis revealed the presence of phenolic acids, flavonoids and phenylethanoids, with acteoside as the main component (14.9 mg/g of extract). Antibacterial effect was determined using the microbroth dilution assay and Staphylococcus aureus was the most sensitive strain (MIC and MBC = 7.5 mg/mL). Antimutagenic activity was evaluated by Ames mutagenicity assay. At 1 mg/plate, the tested extract afforded high protection against the mutagenicity of nitroquinoline-N-oxide (4NQO) to Salmonella typhimurium strain TA100 (inhibition rate 48.3%). Antitumour activity was screened in Ehrlich ascites carcinoma (EAC) model. Pretreatment with 2 mg/kg body weight showed statistically significant decrease in tumour cell viability, while ascites volume and tumour cell count became slightly decreased, but not to a statistically significant extent. The results indicate that V. urticifolia deserves further research into its potential chemoprevention effects.",

AB  - Here we report on the antimicrobial and antioxidant activity of several extracts of Ruscus aculeatus L. and Ruscus hypoglossum L., as well as of some compounds previously isolated from them (p-coumaric and caffeic acids, rutin). For antimicrobial activity, the microdilution technique was used against eight bacterial and five fungal strains, along with standard drugs (streptomycin, ampicillin, bifonazole, ketoconazole). Investigated extracts and isolated compounds showed antibacterial and especially antifungal activity. In some cases this activity was better than standard drugs (streptomycin, ampicillin, bifonazole, ketoconazole) and was more evident for some of isolated compounds. Antioxidant activity was investigated by DPPH and 2-deoxyribose assays, along with spectrophotometrical determination of total phenolics content (TPC) using Folin-Ciocalteu reagent. In DPPH test obtained SC50 values were in a range of 173-541 mu g/mL, while inhibition of 2-deoxyribose for some extracts reached up to maximal 50%. Results of antioxidant assays were strongly correlated with total phenolics content. The most active was the EtOAc fraction of the MeOH extract of R. aculeatus herb regarding all investigated parameters.

AB  - Background Three pure compounds that naturally occur in plants were of particular interest to our study regarding the possibility of using them as food preservatives: p-coumaric acid (found in peanuts, tomatoes, carrots, garlic, wine, vinegar, etc.), caffeic acid (found in argan oil, oats, wheat, rice and olive oil) and rutin (found in asparagus, citrus fruits, berries, apple, apricot, asparagus, beef and beer). In the following study we investigated in situ antioxidant and antimicrobial activities of three pure compounds, namely caffeic acid, p-coumaric acid and rutin, naturally occurring in plants. Results Two food systems were used in order to obtain information on how these compounds react in actual food systems rather than microbiological media. The results indicated good antioxidant activity in in situ food systems. For tested phenolic compounds it was further shown that they successively inhibited the development of the isolated food contaminant Staphylococcus aureus in chicken soup. Panelist found that organoleptic characteristics of chicken soup and pork meat improved after treatment with phenolics. Conclusion Our findings alone, along with the potential use of phenolic compounds that are widespread in nature, may imply their potential use as preservatives in the food industry.

AB  - This study was designed to evaluate the antibacterial activity of the Veronica montana L. water extract and its main phenolic compound, protocatechuic acid. The antibacterial activity was determined by microdilution assay against six strains of Gram-positive and Gram-negative bacteria. Listeria monocytogenes was the most sensitive of the tested bacterial species. Antibacterial preserving properties of protocatechuic acid were also evaluated after its incorporation in cream cheese, using L. monocytogenes as commonly cheese contaminant. The compound successfully inhibited L monocytogenes development in cream cheese, at room temperature and in refrigerator (25 degrees C and 4 degrees C, respectively), after 3 days of inoculation. Sensory evaluation was carried out in order to validate the mentioned food system. A possible mode of action of the tested compound towards bacterial cells was assessed and appears to be direct lysis of pathogenic cytoplasmic membrane. Prediction of pharmacokinetic properties was also performed using computational analyzes. The obtained results can serve as an important platform for the development of effective natural preservatives.

AB  - The whole, fresh involucral bracts of cardoon, Cynara cardunculus L. (Compositae), were extracted with EtOH and an aqueous suspension of the obtained EtOH extract was partitioned successively with CHCl3, EtOAc and n-BuOH, leaving a residual water extract. All obtained extracts were evaluated for their antioxidant and antimicrobial properties. The antioxidant potential was evaluated using following in vitro methods: FRAP (ferric reducing antioxidant power) assay, and scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. Antimicrobial activity was estimated using a microdilution technique against food-borne, mycotoxin producers and human pathogenic bacteria and micromycetes. The following bacteria were tested: Salmonella typhimurium, Escherichia coli, Bacillus subtilis, Staphylococcus epidermidis, Staphylococcus aureus, as well as micromycetes: Aspergillus niger, Aspergillus ochraceus, Aspergillus flavus, Penicillium ochrochloron, Penicillium funiculosum, Trichoderma viride, Fusarium tricinctum and Alternaria alternata. Results showed that all extracts possessed concentration-dependent antioxidant activity. In biological assays, C. cardunculus extracts showed antimicrobial activity comparable with standard antibiotics.

AB  - The chemical composition and antimicrobial and antiradical activities of the essential oils isolated from unripe and ripe fruits of S. globiferum Vis. (Apiaceae) have been determined. The most abundant constituent in the essential oils of unripe and ripe fruits was sabinene (53.1% and 65.3%), followed by gamma-terpinene (7.7% and 6.6%), alpha-pinene (7.2% and 4.4%), and beta-phellandrene (5.0% and 4.9%). Antibacterial and antifungal properties of these oils were evaluated using a modified microdilution technique. Scavenging activity was determined by the DPPH radical assay. The essential oils exhibited significant antimicrobial, but low antiradical activity.
