AB  - Oxidative stress has been implicated in numerous pregnancy-related disorders. Biologically active plant sec-
ondary metabolites, which are present in everyday diet, could prove effective therapeutic agents in preventing
these disorders. This study evaluated effects of taxifolin (dihydroquercetin) on ROS production, markers of
oxidative damage to lipids and proteins, activity of antioxidant enzymes and production of pro-inflammatory
cytokines in H2O2-induced oxidative stress in trophoblast HTR-8/SVneo cells. Taxifolin in 10 μM and 100 μM
concentrations attenuated oxidative damage to lipids and proteins, as evidenced by a decrease in MDA content,
extracellular LDH activity, carbonyl groups and nitrite contents. A reduction in the activity of antioxidant en-
zymes SOD, CAT and GPx in cells pre-treated with taxifolin, prior to H2O2 exposure, was also observed, along
with a reduction in intracellular ROS production. Both evaluated concentrations of taxifolin showed anti-
inflammatory activity in trophoblast cells, by reducing production of pro-inflammatory cytokines IL-1β and IL-
6. In this model of H2O2-induced oxidative stress, taxifolin showed marked antioxidative and anti-
inflammatory activities in trophoblast cells, adding further evidence of its protective effects and showing po-
tential as a therapeutic agent in preventing adverse pregnancy outcomes.

AB  - INTRODUCTION: Imaging  techniques  based  on  X-ray  phase-contrast  (XPC)  have  shown  tremendous  promise  for applications involving biomaterials and soft tissue formation [1,2].XPC imaging can be applied at higher energy offering the  potential  for  lower  dose  imaging.  Essential  to  the  development  of  this  technique  and  its  routine  use  is  an understanding of the potential damage of X-ray dose on cells and tissues. EXPERIMENTAL: In this study the comet assay, a sensitive assay for DNA damage, was used to evaluate DNA damage on  mesenchymal  stem  cells  (MSCs) exposedto X-ray  irradiation.  We  examined  the  effects  of  early  (immediately following  irradiation)  and  delayed  (24h  post-irradiation) X-ray  effects  caused  by  low  (15mGy)  and  intermediate (150mGy  and  1.5  Gy)  exposure  on  MSCs  during  a  monitoring  period  of  4  weeks  (five  irradiations,  one  weekly).  Cells were submitted to apolychromatic X-ray source (Thermo Fisher PXS10 conditions: voltage 45 kV, source current 160A, source power 7.2 W, source spot size 9 um, photon flux on the sample 7.66106photonss-1mm-2irradiation).Statistical analysis was performedby using Two-way analysis of variance (ANOVA) with Tukey’s multiple comparisons posttest in GraphPad Prism 5.0.A difference at p< 0.05 was considered statistically significant.RESULTS AND DISCUSSION: Resultsof the DNA comet assay indicated that early effects of low-and intermediate-dose of XPC induced an increase in the number of cells with DNA damage after each irradiation, where intermediate-dose (150 mGy and 1.5 Gy) produced significantly higher damage relative to controls. DNA damage induced by low and intermediate doses returned to the control value 24h after the irradiation exposure, suggesting a strong protection of MSCs at the tested doses of XPC irradiation. CONCLUSIONS: The data presented in this studyshows that 24 h after the last of five weekly low and intermediate doses XPC irradiation, the harmful effects on DNA in MSCs were notdetected. The current study reinforces the need of investigating consequences of low and intermediate doses of X-ray PC irradiation in the field of tissue engineering and provide new basis for MSCs using in the clinics.

AB  - Copper(ii) complexes with an α-diimine show a wide variety of biological activities, such as antibacterial, antifungal, antioxidant and anticancer. In this work, we synthesized and structurally characterized two novel Cu(ii) complexes with methyl 3-formyl-4-hydroxybenzoate (HL) and α-diimines: 2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen). Crystal structure analysis shows that the formulas of the compounds are [Cu(bipy)(L)(BF4)] (1) and [Cu(phen)(L)(H2O)](BF4)·H2O (2), with BF4− as a ligand in complex 1, which is rarely coordinated to metals. Both complexes have a square pyramidal geometry, while DFT calculations showed that the most stable structures of complexes 1 and 2 in a water/DMSO mixture are square-planar derivatives [Cu(bipy)(L)]+ and [Cu(phen)(L)]+. The antibacterial activity of compounds was evaluated in vitro on four Gram-negative and four Gram-positive bacterial strains. Complex 2 showed greater antibacterial activity towards all bacterial strains comparable to the control compound Amikacin. Complex 2 exerted a strong cytotoxic effect against the tested cancer cell lines (IC50 values ranging from 0.32 to 0.44 μM). Both complexes caused apoptotic cell death in HeLa cells and a noticeable in vitro antiangiogenic effect. In the concentration range of 5 to 100 μM, the complexes showed the absence of a genotoxic effect and displayed a protective effect against oxidative DNA damage induced by H2O2 in human peripheral blood cells. The interaction between the compounds and calf-thymus DNA was evaluated by diverse techniques suggesting a tight binding, which was also confirmed by molecular docking. In addition, it was found that the complexes bind tightly and reversibly to bovine and human serum albumin.

Prospective study of cytotoxic and genotoxic effects of Combretastatin

AB  - Combretastatins are a class of natural phenols found in the bark of Combretum
caffrum, commonly known as South African Bush Willow. Despite having a similar
name, combretastatins are unrelated to statins, a family of cholesterol-lowering drugs.
Combretastatin A4 have been shown to be one of the most potent tubulin-depolymerizing
agent. Microtubules control chromosomal segregation and cytokinesis during mitosis
in both cancer and stromal cells and contribute to overall tumor growth. Consequently,
microtubule inhibitors interfere with cell cycle progression and induce apoptosis in
cancer cells in vitro.
The aim of this study was to investigate the potential gentoxic effect of Comretastatin
A4 (CA4) in isolated peripheral blood mononuclear cells (PBMC) in Comet assay in
order to establish is there any DNA damage in healty non-dividing cells. The aim also
was to explore potential cytotoxic activity of CA4 against human cervical carcinoma
(HeLa) cell line.
Genotoxicity of CA4 was evaluated on PBMC in a range of 9 concentrations
(from 1 nM to 200μM). Non of the tested concentrations showed genotoxiceffect.
The same range of different concentrations of CA4 (from 1 nM to 200μM) were
applied to evaluate potential cytotoxicity in a monolayer culture of HeLa cells using

abstract = "Combretastatins are a class of natural phenols found in the bark of Combretum
caffrum, commonly known as South African Bush Willow. Despite having a similar
name, combretastatins are unrelated to statins, a family of cholesterol-lowering drugs.
Combretastatin A4 have been shown to be one of the most potent tubulin-depolymerizing
agent. Microtubules control chromosomal segregation and cytokinesis during mitosis
in both cancer and stromal cells and contribute to overall tumor growth. Consequently,
microtubule inhibitors interfere with cell cycle progression and induce apoptosis in
cancer cells in vitro.
The aim of this study was to investigate the potential gentoxic effect of Comretastatin
A4 (CA4) in isolated peripheral blood mononuclear cells (PBMC) in Comet assay in
order to establish is there any DNA damage in healty non-dividing cells. The aim also
was to explore potential cytotoxic activity of CA4 against human cervical carcinoma
(HeLa) cell line.
Genotoxicity of CA4 was evaluated on PBMC in a range of 9 concentrations
(from 1 nM to 200μM). Non of the tested concentrations showed genotoxiceffect.
The same range of different concentrations of CA4 (from 1 nM to 200μM) were
applied to evaluate potential cytotoxicity in a monolayer culture of HeLa cells using

AB  - Background: Chromosome instability (CIN) in Alzheimer’s disease has been found in peripheral blood lymphocytes, and also in neurons of affected individuals. CIN is a result of errors in chromosome segregation during mitosis leading to numerical and structural chromosomal abnormalities. Material and Methods: Fluorescent In Situ Hybridization. (FISH) has been a method of choice for the evaluation of CIN in various diseases. We have studied a specific type of X chromosome instability, or premature centromere division (PCD) in interphase nuclei of the hippocampus brain tissue neurons from sporadic AD females. PCD was studied by using a new approach to FISH. Results: Namely, we have established a methodology that allows quantitative analysis of centromere fluorescence intensity, i.e. the size of individual FISH spots, thus giving a more focused view of the centromere region of interest. By using the micro image system, our results show that quantitative FISH can distinguish PCD positive signals vs. PCD negative signals in cells of brain tissue. Conclusion: This could be a specific biomarker in affected cells in order to verify CIN, thus supporting the established qualitative analysis of FISH spots in the study of centromere and chromosomal alterations in interphase nuclei of patients affected by AD.

AB  - Dry olive leaf extract (DOLE) and its active component oleuropein (OLE) were applied as reducing and stabilizing agents to prepare colloidal 20-25 nm silver nanoparticles (Ag NPs). The Ag NPs were characterized using transmission electron microscopy, X-ray diffraction analysis, and absorption spectroscopy. The cytotoxic actions of coated Ag NPs, and their inorganic and organic components, were examined against trophoblast cells and human peripheral blood lymphocytes (PBLs), Gram-positive, Gram-negative bacteria, and yeast. The genotoxic potential was evaluated in PBLs in vitro with the comet assay. Ag/DOLE and Ag/OLE induced cytotoxic effects in both types of cells after 24 h exposure when silver concentrations were 0.025-0.2 mM. However, the most pronounced cytotoxicity exhibits Ag/OLE. Both colloids also caused reduced ROS production in both cell types at 0.1 mM and 0.2 mM, while bare Ag NPs did not alter ROS levels at any of the conditions. Functionalized Ag/DOLE and Ag/OLE did not show genotoxic effects in PBLs, while bare AgNPs increased DNA damage significantly only at 0.2 mM. Regarding the antimicrobial effects, the Ag/OLE had MIC values for all evaluated microorganisms from 0.0625 to less than 0.0312 mM. Also, the antimicrobial effect of Ag/DOLE was significantly higher on Gram-negative bacteria and yeast than on Gram-positive bacteria. Obtained results indicate that Ag/OLE induced the most pronounced biological effects, beneficial for its application as an antimicrobial agent, but with potential risks from exposure to high concentrations that could induce cytotoxicity in healthy human cells.

AB  - Background: Antioxidants and prooxidants have an impact on the intracellular oxidative equilibrium. Overproduction of prooxidants leads to oxidative stress caused by imbalances in oxidative reduction pathways. The body can be supplied with non-enzymatic, low molecular weight antioxidants through diet.The edible medicinal mushroom Chaga, Inonotus obliquus (Ach. ex Pers.) Pilat, has long been long used to treat or prevent various health conditions and disorders. The bioactive compounds of Chaga exhibit antitumor, anti-inflammatory, hypoglycemic, immunomodulatory, antioxidant, and antigenotoxic effects. Material and Methods: DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging activity, FRAP (ferric reducing antioxidant power) total antioxidant activity, and hydroxyl radical scavenging capacity were measured.
Results: Commercial Biochaga (B), a water extract of Biochaga mushroom, was obtained from Sibpribor Ooo, Irkutsk, Russia. B (IC =5.9 mg/ mL) showed moderate reducing power compared in comparison to vitamin C and strong compared to BTH. B (IC =1.78 mg/mL) showed remarkable free radical scavenging and moderate hydroxyl scavenging activity (IC =8.473 mg/mL). Conclusion: We can place Biochaga in the radical scavenging category because it efficiently eliminates hydroxyl radicals against which the body has insufficient antioxidant defenses.

AB  - Mushrooms have been evaluated for their nutritional and medicinal properties for
centuries. We evaluated the biological activities of commercial products of Cordyceps
Sinensis (CS), Agaricus blazei (AB), and Immune Assist (IA) (all produced by Aloha
Medicinals). Immune Assist (IA) is made from extract of six species of medical
mushrooms: Agaricus blazei - Cordycepssinensis - Grifola frondosa - Ganoderma
lucidum - Coriolus versicolor - Lentinula edodes.
The antioxidant evaluation showed that CS had strong OH scavenging properties and
moderate reducing power, while its DPPH scavenging ability was weak. AB displayed
remarkable ·OH scavenging properties, moderate reducing power, and modest DPPH
scavenging activity, similar to IA. Also, CS, AB and IA displayed significant antigenotoxic
effects in human peripheral blood cells against H2O2-induced DNA damage.
Further, the cytotoxicity of CS, AB, and IA to HS-5 (bone marrow stroma cells), MCF-
7 (human breast epithelial cell line), and MDA-MB-231 (human breast carcinoma cell
line) were assessed using MTT assay. AB and IA could inhibit the proliferative action
of mentioned cancer cell lines after 72h in a dose-dependent manner, while there were
no effects on HS-5 cell lines proliferation, while CS did not show anti-proliferative/
cytotoxic activity on cancer cell lines.
Current findings remain a significant challenge for the usage of medicinal mushrooms
in the field of cancer prevention/treatment, and thus an active area of future research.

7 (human breast epithelial cell line), and MDA-MB-231 (human breast carcinoma cell
line) were assessed using MTT assay. AB and IA could inhibit the proliferative action
of mentioned cancer cell lines after 72h in a dose-dependent manner, while there were
no effects on HS-5 cell lines proliferation, while CS did not show anti-proliferative/
cytotoxic activity on cancer cell lines.
Current findings remain a significant challenge for the usage of medicinal mushrooms
in the field of cancer prevention/treatment, and thus an active area of future research.",

AB  - The antibacterial performance and cytotoxic examination of in situ prepared silver nanoparticles (Ag NPs), on inorganic-organic hybrid nanopowder consisting of zirconium dioxide nanoparticles (ZrO2 NPs) and dihydroquercetin (DHQ), was performed against Gram (−) bacteria Escherichia coli and Gram (+) bacteria Staphylococcus aureus, as well as against human cervical cancer cells HeLa and healthy MRC-5 human cells. The surface modification of ZrO2 NPs, synthesized by the sol-gel method, with DHQ leads to the interfacial charge transfer (ICT) complex formation indicated by the appearance of absorption in the visible spectral range. The prepared samples were thoroughly characterized (TEM, XRD, reflection spectroscopy), and, in addition, the spectroscopic observations are supported by the density functional theory (DFT) calculations using a cluster model. The concentration- and time-dependent antibacterial tests indicated a complete reduction of bacterial species, E. coli and S. aureus, for all investigated concentrations of silver (0.10, 0.25, and 0.50 mg/mL) after 24 h of contact. On the other side, the functionalized ZrO2 NPs with DHQ, before and after deposition of Ag NPs, do not display a significant decrease in the viability of HeLa MRC-5 cells in any of the used concentrations compared to the control

AB  - More than 30 types of amyloids are linked to close to 50 diseases in humans, the most prom- inent being Alzheimer’s disease (AD). AD is brain-related local amyloidosis, while another amyloidosis, such as AA amyloidosis, tends to be more systemic. Therefore, we need to know more about the biological entities’ influencing these amyloidosis processes. However, there is currently no support system developed specifically to handle this extraordinarily complex and demanding task. To acquire a systematic view of amyloidosis and how this may be relevant to the brain and other organs, we needed a means to explore "amyloid net- work systems" that may underly processes that leads to an amyloid-related disease. In this regard, we developed the DES-Amyloidoses knowledgebase (KB) to obtain fast and rele- vant information regarding the biological network related to amyloid proteins/peptides and amyloid-related diseases. This KB contains information obtained through text and data min- ing of available scientific literature and other public repositories. The information compiled into the DES-Amyloidoses system based on 19 topic-specific dictionaries resulted in 796,409 associations between terms from these dictionaries. Users can explore this infor- mation through various options, including enriched concepts, enriched pairs, and semantic similarity. We show the usefulness of the KB using an example focused on inflammasome- amyloid associations. To our knowledge, this is the only KB dedicated to human amyloid- related diseases derived primarily through literature text mining and complemented by data mining that provides a novel way of exploring information relevant to amyloidoses.

AB  - In this article, cytotoxicity, the mechanisms of cytotoxic activity, genotoxicity, and interaction with DNA and proteins, of two Cu(II) complexes with a salicylaldehyde derivative (4-(diethylamino)salicylaldehyde) and α-diimine (2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen)) are reported. Both Cu(II) complexes performed cytotoxic effects against all tested malignant cell lines. Complexes exerted highest cytotoxicity against HeLa and A375 malignant cell lines. The cytotoxic activity of Cu(II) complex with phen as a α-diimine co-ligand was significantly higher in comparison with cytotoxic activity of Cu(II) complex with bipy. Pretreatment with specific inhibitors of caspase-3, caspase-8 or caspase-9, in order to clear up the mode of cell death triggered by two Cu(II) complexes in HeLa cells, indicated the ability of these complexes to induce apoptosis through activation of target caspases. Cu(II)-phen complex exhibited significant antioxidant activity compared with Cu(II)-bipy complex, and showed a better effect on reducing intracellular ROS levels in HeLa cells. Tested complexes did not display genotoxic potential in human peripheral blood leucocytes, but exhibited an antigenotoxic effect in post-treatment, after H2O2 exposure. The study of the in vitro biological properties regarding their affinity towards CT (calf-thymus) DNA and serum albumins showed that the compounds can intercalate to CT DNA, and bind reversibly and tightly to the albumins. Molecular docking studies of the ability of compounds to bind to biomacromolecules are consistent with in vitro studies.

AB  - Abstract:Hyssopus officinalisL. is a well-known aromatic plant used in traditional medicine and the foodand cosmetics industry. The aim of this study is to assess the antioxidant, genotoxic, antigenotoxic andcytotoxic properties of characterized hyssop essential oils and methanol extracts. Chemical compositionwas analyzed by gas chromatography - mass spectrometry (GC-MS) and liquid chromatography with diodearray detection and mass spectrometry (LC-DAD-MS), respectively. Antioxidant activity was examinedby 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing/antioxidant power (FRAP) tests; genotoxicand antigenotoxic activity were examined by the comet assay, while cytotoxicity was evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide dye (MTT) test against tumor cell lines (SW480,MDA-MB 231, HeLa) and non-transformed human lung fibroblast cell lines (MRC-5). The essential oilswere rich in monoterpene hydrocarbons (e.g., limonene; 7.99–23.81%), oxygenated monoterpenes (1,8-cineole; 38.19–67.1%) and phenylpropanoids (methyl eugenol; 0.00–28.33%). In methanol extracts, the mostabundant phenolics were chlorogenic and rosmarinic acid (23.35–33.46 and 3.53–17.98 mg/g, respectively).Methanol extracts expressed moderate to weak antioxidant activity (DPPH IC50= 56.04–199.89μg/mL,FRAP = 0.667–0.959 mmol Fe2+/g). Hyssop preparations significantly reduced DNA damage in humanwhole blood cells, induced by pretreatment with hydrogen peroxide. Methanol extracts exhibited selectiveand potent dose- and time-dependent activity against the HeLa cell line.  Results of the current studydemonstrated notableH.officinalismedicinal potential, which calls for further investigation.

AB  - Olive leaf (OL) is a precious source of bioactive compounds for versatile biomedical applications. Oleuropein, the main constituent from olive leaves, and its metabolite hydroxytyrosol are responsible for the majority of the observed biological effects. However, joined components of OL achieved better biological effects than its isolated individual constituents, indicating their synergistic effect. Predominantly free radical scavenging and metal chelation properties of OL compounds are responsible for the protective effects against oxidative damage and could be endorsed for its antioxidant and antigenotoxic effects. Local environment in cells such as the presence of transition metals, oxygen radicals, change in pH value, and the concentration of the flavonoid compounds can modify their activity from antioxidant to prooxidant and from antigenotoxic to genotoxic. Effects of complexes formed between flavonoids, metals, and DNA molecule, in heavy metal intoxicated individuals that are treated with ethylenediaminetetraacetic acid chelation therapy, require further research attention considering the demonstrated prooxidant and genotoxic effects.

Data on protein and lipid oxidative damage in spontaneously hypertensive rats are numerous, but there is no information on DNA damage in tissues measured by comet assay. The aim of this study was to determine the baseline damage to DNA, protein, and lipid macromolecules in different organs of spontaneously hypertensive rats. Markers of lipid peroxidation, protein oxidation, and DNA damage were measured in blood, heart, kidney, and liver of 24-week-old spontaneously hypertensive rats. Plasma prooxidant and antioxidant status were determined as well. Age-matched normotensive Wistar rats were used as control. A rise in markers of lipid peroxidation and protein oxidation, malondialdehyde, and advanced oxidation protein products, was detected in all tissues of spontaneously hypertensive rats, with particularly high values in the liver. DNA damage, measured by the comet assay, was significantly higher in all the studied tissues of spontaneously hypertensive rats compared to normotensive control, with more severe damage in the cardiac and renal cells. Significant depletion of the plasma antioxidant barrier in spontaneously hypertensive rats was also observed. This study showed increased damage to all macromolecules in all studied samples of spontaneously hypertensive rats in comparison with control Wistar rats.

AB  - The comet assay or single cell gel electrophoresis, is the most common method used to measure strand breaks and a variety of other DNA lesions in human populations. To estimate the risk of overall mortality, mortality by cause, and cancer incidence associated to DNA damage, a cohort of 2,403 healthy individuals (25,978 person-years) screened in 16 laboratories using the comet assay between 1996 and 2016 was followed-up. Kaplan–Meier analysis indicated a worse overall survival in the medium and high tertile of DNA damage (p < 0.001). The effect of DNA damage on survival was modelled according to Cox proportional hazard regression model. The adjusted hazard ratio (HR) was 1.42 (1.06–1.90) for overall mortality, and 1.94 (1.04–3.59) for diseases of the circulatory system in subjects with the highest tertile of DNA damage. The findings of this study provide epidemiological evidence encouraging the implementation of the comet assay in preventive strategies for non-communicable diseases.

abstract = "The comet assay or single cell gel electrophoresis, is the most common method used to measure strand breaks and a variety of other DNA lesions in human populations. To estimate the risk of overall mortality, mortality by cause, and cancer incidence associated to DNA damage, a cohort of 2,403 healthy individuals (25,978 person-years) screened in 16 laboratories using the comet assay between 1996 and 2016 was followed-up. Kaplan–Meier analysis indicated a worse overall survival in the medium and high tertile of DNA damage (p < 0.001). The effect of DNA damage on survival was modelled according to Cox proportional hazard regression model. Romina),  rich  in anthocyanin polyphenols, has been demonstrated to have favorable effects on health due to its antioxidant properties. The present study investigated the antigenotoxic potential of anthocyanin-enriched fraction of Romina strawberry methanolic extract (ACY) against DNA damage on human peripheral blood leukocytes, induced by hydrogen peroxidein vitro. Five concentrations of the ACY extract were used in all experiments (2.5, 5, 10, 15, 20 μg/ml). The results of the alkaline comet assay showed no genotoxic effect of the ACY. After the pre-exposureof the leukocytes to the ACY, and subsequent incubation with H2O2, a decreased number of DNA damaged cells was recorded in all the tested concentrations, compared to controls. In the post-treatment, there was a concentration-dependent DNA damage reduction, while a statistically significant decrease was achieved with 15 and 20 μg/ml concentrations. The results indicate that ACY is efficient in oxidative DNA damage reduction, and it is more potent as a post-applicative than a protective agent.

U ovoj studiji ispitivan je antigenotoksični potencijal frakcije metanolnog ekstrakta jagode Romina koji je obogaćen antocijaninom (ACY) na oštećenja DNK na leukocitima humane periferne krvi, izazvanih vodonik-peroksidom in vitro. U svim eksperimentima je korišćeno i testirano pet koncentracija ekstrakta ACY (2.5, 5, 10, 15, 20 µg /ml). Rezultati ispitivanja alkalnim komet testom nisu pokazali genotoksični efekat ACY. Nakon izloženosti leukocita ACY i potom njihovog tretmana i inkubacije sa H2O2, smanjen je broj ćelija sa oštećenom DNK, u svim ispitivanim koncentracijama u poređenju sa kontrolama. U post-tretmanu, došlo je do smanjenja oštećenja DNK u zavisnosti od koncentracije ekstrakta, dok je statistički značajno smanjenje postignuto sa koncentracijama od 15 i 20 µg/ml. Rezultati ukazuju da je ACY efikasan u smanjenju oksidativnih oštećenja DNK i da pokazuje snažniji efekat kao postaplikativno nego kao zaštitno sredstvo.

AB  - The alkaline comet assay, or single cell gel electrophoresis, is one of the most popular methods for assessing DNA damage in human population. One of the open issues concerning this assay is the identification of those factors that can explain the large inter-individual and inter-laboratory variation. International collaborative initiatives such as the hCOMET project - a COST Action launched in 2016 - represent a valuable tool to meet this challenge. The aims of hCOMET were to establish reference values for the level of DNA damage in humans, to investigate the effect of host factors, lifestyle and exposure to genotoxic agents, and to compare different sources of assay variability. A database of 19,320 subjects was generated, pooling data from 105 studies run by 44 laboratories in 26 countries between 1999 and 2019. A mixed random effect log-linear model, in parallel with a classic meta-analysis, was applied to take into account the extensive heterogeneity of data, due to descriptor, specimen and protocol variability. As a result of this analysis interquartile intervals of DNA strand breaks (which includes alkali-labile sites) were reported for tail intensity, tail length, and tail moment (comet assay descriptors). A small variation by age was reported in some datasets, suggesting higher DNA damage in oldest age-classes, while no effect could be shown for sex or smoking habit, although the lack of data on heavy smokers has still to be considered. Finally, highly significant differences in DNA damage were found for most exposures investigated in specific studies. In conclusion, these data, which confirm that DNA damage measured by the comet assay is an excellent biomarker of exposure in several conditions, may contribute to improving the quality of study design and to the standardization of results of the comet assay in human populations.

abstract = "The alkaline comet assay, or single cell gel electrophoresis, is one of the most popular methods for assessing DNA damage in human population. One of the open issues concerning this assay is the identification of those factors that can explain the large inter-individual and inter-laboratory variation. International collaborative initiatives such as the hCOMET project - a COST Action launched in 2016 - represent a valuable tool to meet this challenge. The aims of hCOMET were to establish reference values for the level of DNA damage in humans, to investigate the effect of host factors, lifestyle and exposure to genotoxic agents, and to compare different sources of assay variability. A database of 19,320 subjects was generated, pooling data from 105 studies run by 44 laboratories in 26 countries between 1999 and 2019. A mixed random effect log-linear model, in parallel with a classic meta-analysis, was applied to take into account the extensive heterogeneity of data, due to descriptor, specimen and protocol variability. As a result of this analysis interquartile intervals of DNA strand breaks (which includes alkali-labile sites) were reported for tail intensity, tail length, and tail moment (comet assay descriptors). A small variation by age was reported in some datasets, suggesting higher DNA damage in oldest age-classes, while no effect could be shown for sex or smoking habit, although the lack of data on heavy smokers has still to be considered. Finally, highly significant differences in DNA damage were found for most exposures investigated in specific studies. In conclusion, these data, which confirm that DNA damage measured by the comet assay is an excellent biomarker of exposure in several conditions, may contribute to improving the quality of study design and to the standardization of results of the comet assay in human populations.",

AB  - Increased levels of oxidative stress and oxidative DNA damage are common features in the pathology of Alzheimer’s disease (AD) found in neurons and peripheral cells like peripheral blood lymphocytes (PBL). Natural products such as strawberry cultivar Alba are an important source of bioactive nutrients that could help in lowering both the oxidative stress and DNA damage levels. The objective was to estimate the effects of Alba extract on DNA damage in peripheral blood lymphocytes of sporadic AD (aged 60–84 years) patients, and healthy elderly (aged 69–83 years) and young (aged 21–30 years) individuals in in vitro conditions. Comet assay was used as a sensitive technique for the evaluation of PBL DNA damage levels. Reduction of basal DNA damage level in PBL was shown in the young group after the incubation with Alba extract ranging from 25 to 200 μg/ml, with 100 μg/ml being the most effective concentration. Selected Alba extract of 100 μg/ml was further used for PBL treatment of AD and healthy elderly age matched group, displaying potential to significantly attenuate DNA damage levels in both groups (p <.05). Alba extract displayed biological activity against oxidative DNA damage, suggesting that its functional ingredients may have beneficial health effects. Practical applications: The data obtained in this preliminary study displayed that strawberry Alba extract is efficient against DNA damage induced by endogenous and exogenous oxidative stress in peripheral blood lymphocytes of Alzheimer`s disease in vitro. An active area of future research of Alba cultivar should be to determine the trials in in vivo systems. Our findings also suggest that Alba cultivar’s functional ingredients potentially may have beneficial health effects in AD.

AB  - Oxidative stress and inflammation are DNA instability factors for rheumatoid arthritis (RA) patients. The aims of this study were to evaluate cytogenetic alterations in Peripheral Blood Lymphocytes (PBL) in two groups of RA patients: the early and the long-term RA group; and to examine potential of concomitant treatment with Methotrexate (MTX) and Dry olive leaf extract (DOLE) against cytogenetic damage in RA patients after a 3-weeks treatment. A total of 32 RA patients and 10 healthy individuals were included. RA patients were equally divided into four groups: two groups with early phase RA (one treated with MTX alone, the other in combination with DOLE); and two long-term phase RA groups (group with active disease and group with low disease activity)-both treated with MTX and DOLE combination. PBL cultures were screened for chromosome aberrations and micronuclei frequencies. Significantly increased frequencies of micronuclei were shown in active phase RA disease (both early and long-term) but not in the group with low disease activity, as compared to controls. Chromosome aberrations were detected for all 4 RA groups. The highest frequencies of micronuclei and chromosome aberrations were found in the long-term active RA group. After 3 weeks-treatment, there were no significant decrease of the micronuclei frequencies compared to baseline, although they were reduced in all RA groups, except for the group with the long-term active disease. High level of cytogenetic damage in RA patients was concordant with duration and activity of the RA disease. At 3 weeks of therapy, neither the combined treatment (MTX+DOLE), nor MTX alone did not affect the frequency of micronuclei formation.

Analysis of tiazofurin-induced dna damage in human whole blood cells using an in vitro comet assay

AB  - Objective. Inosine 5’-monophosphate dehydrogenase (IMPDH) activity in cancer cells is increased. Tiazofurin selectively inhibits the activity of IMPDH, and it has been granted for the treatment of different cancers and new viral diseases. Its widespread use was limited because exposure to tiazofurin under certain circumstances was found to have a higher frequency of severe non-hematologic toxicity. Therefore, the objective of this study was to examine genotoxic action and inducement of DNA damage of tiazofurin using the comet assay. Methods. The ability of tiazofurin to induce DNA damage was evaluated using single-cell gel electrophoresis (SCGE) technique/comet assay. Human whole blood cells were exposed to three final concentrations of tiazofurin (1 µM/mL, 2 µM/mL, and 5 µM/mL) for 30 min in vitro. Results. Our results indicate that tiazofurin produced a significant level of DNA damage on whole blood cells after 30 min of exposure vs. control. All tested concentrations were significantly comet-forming, in a concentration-dependent manner. Conclusion. Our investigation on the tiazofurin-treated cells and their relationship to the formation of DNA damage demonstrated that the genotoxic effect was induced after exposure to tiazofurin under described conditions.
AB  - Cilj. Aktivnost inozin 5'-monofosfat dehidrogenaze (IMPDH) povec'ana je u c'elijama karcinoma. Tiazofurin selektivno inhibira aktivnost IMPDH i odobren je za lečenje različitih karcinoma i novih virusnih bolesti. Njegova široko rasprostranjena upotreba bila je ograničena jer je utvrđeno da je izloženost tiazofurinu pod određenim okolnostima imala vec'u incidencu ozbiljne nehematološke toksičnosti. Stoga je cilj ove studije bio da se pomoc'u komet testa ispita genotoksično delovanje i izazivanje DNK oštec'enja tiazofurinom. Metode. Sposobnost tiazofurina da izazove DNK oštec'enje procenjena je primenom elektroforeze DNK pojedinačnih ćelija (SCGE) / komet testa. Ćelije pune krvi su bile izložene trima konačnim koncentracijama tiazofurina (1 µM/mL, 2 µM/mL, and 5 µM/mL) tokom 30 minuta in vitro. Rezultati. Naši rezultati ukazuju na to da je tiazofurin proizveo značajan nivo DNK oštec'enja na c'elijama pune krvi nakon 30 minuta izlaganja u odnosu na kontrolu. Sve ispitivane koncentracije su dovele do značajnog nastanka kometa, pri čemu je nivo oštećenja rastao s koncentracijom. Zaključak. Naše istraživanje c'elija tretiranih tiazofurinom i njihova reakcija na izazivanje DNK oštec'enja pokazalo je da je tiazofurin ispoljio genotoksični efekat pod opisanim uslovima.

AB  - Alzheimer’s disease (AD) is a neurodegenerative disease that affects millions of individuals worldwide and can occur relatively early or later in life. It is well known that genetic components, such as the amyloid precursor protein gene on chromosome 21, are fundamental in early-onset AD (EOAD). To date, however, only the apolipoprotein E4 (ApoE4) gene has been proved to be a genetic risk factor for late-onset AD (LOAD). In recent years, despite the hypothesis that many additional unidentified genes are likely to play a role in AD development, it is surprising that additional gene polymorphisms associated with LOAD have failed to come to light. In this review, we examine the role of X chromosome epigenetics and, based upon GWAS studies, the PCDHX11 gene. Furthermore, we explore other genetic risk factors of AD that involve X-chromosome epigenetics.

AB  - To remedy carotid artery stenosis and prevent stroke surgical intervention is commonly used, and the gold standard being carotid endarterectomy (CEA). During CEA cerebrovascular hemoglobin oxygen saturation decreases and when this decrease reaches critical levels it leads to cerebral hypoxia that causes neuronal damage. One of the proposed mechanism that affects changes during CEA and contribute to acute brain ischemia (ABI) is oxidative stress. The increased production of reactive oxygen species and reactive nitrogen species during ABI may cause an unregulated inflammatory response and further lead to structural and functional injury of neurons. Antioxidant activity are involved in the protection against neuronal damage after cerebral ischemia. We hypothesized that neuronal injury and poor outcomes in patients undergoing CEA may be results of oxidative stress that disturbed function of antioxidant enzymes and contributed to the DNA damage in lymphocytes.
