AB  - U okviru doktorske disertacije izvršena je hemijska karakterizacija i skrining farmakološke aktivnosti podzemnih i nadzemnih delova osam vrsta roda Geranium (G. macrorrhizum, G. robertianum, G. phaeum, G. palustre, G. sanguineum, G. columbinum, G. pyrenaicum i G. lucidum) poreklom sa Vlasinske visoravni.Isparljive frakcije izolovane destilacijom vodenom parom analizirane su gasnom hromatografijom sa plameno-jonizacionim i masenim detektorom (GC-FID/MS). Seskviterpeni su bili dominantni u isparljivoj frakciji podzemnog dela G. macrorrhizum, kao i u frakcijama nadzemnih delova svih ispitivanih vrsta osim G. phaeum i G. columbinum gde su preovladali diterpeni. U isparljivim frakcijama podzemnih delova svih ispitivanih vrsta osim G. macrorrhizum preovladavale su masne kiseline i njihovi derivati.Spektrofotometrijskom analizom utvrđeno je da su metanolni ekstrakti bogati polifenolnim jedinjenjima, kao i da su tanini dominantni polifenoli u gotovo svim uzorcima.Iz metanolnog ekstrakta nadzemnog dela G. palustre, primenom hromatografije u koloni, izolovana su dva jedinjenja koja su na osnovu UV-Vis, MS i NMR spektara identifikovana kao elagni tanin korilagin i flavonoid astragalin-2′′-O-galat.Primenom tečne hromatografije sa masenom spektrometrijom (LC-MS) u metanolnim ekstraktima utvrđeno je prisustvo i određen sadržaj ukupno sedam fenolkarboksilnih kiselina (elagne, galne, galoilhina, galoilšikiminske, protokatehinske, hlorogenske i brevifolin karboksilne kiseline), 12 flavonoidnih jedinjenja (kvercetina, kemferola, njihovih heterozida i galoilovanih derivata heterozida, kao i metilovanih derivata kemferola) i 14 taninskih jedinjenja, uključujući elagne tanine (geraniin i njegov izomer, korilagin, telimagrandin I i jedinjenja izomerne strukture, telimagrandin II i hebulaginska kiselina) i galne tanine (galoilglukoza, digaloilglukoza, trigaloilglukoza i pentagaloilglukoza). Ispitivanjem redukcione sposobnosti u FRAP testu i antiradikalskog delovanja DPPH testom, utvrđeno je da ekstrakti poseduju značajan in vitro antioksidantni potencijal, uporediv sa L-askorbinskom kiselinom kao poznatim antioksidansom. Ispitivanjem uticaja na parametre oksidativnog stresa u humanom serumu, utvrđeno je da ekstrakti dovode do značajnog povećanja antioksidantnog kapaciteta humanog seruma, pri čemu je efekat većine ekstrakata bio bolji ili uporediv sa efektom troloksa, kao pozitivne kontrole. Antimikrobna aktivnost ekstrakata testirana je mikrodilucionom metodom na sedam standardnih sojeva Gram pozitivnih i Gram negativnih bakterija, standardnom soju gljivice Candida albicans, deset kliničkih izolata Escherichia coli i sedam kliničkih izolata Klebsiella pneumoniae. Većina ekstrakata ispoljila je aktivnost prema svim standardnim sojevima. Značajna antimikrobna aktivnost prema svim kliničkim izolatima E. coli utvrđena je za ekstrakte nadzemnih delova G. robertianum i G. columbinum, dok je aktivnost prema kliničkim izolatima K. pnaeumoniae bila slabija.In vitro citotoksična aktivnost ekstrakata, geraniina i elagne kiseline ispitana je MTT testom na ćelijskim linijama humanog karcinoma prostate (PC-3), kolona (HT-29), grlića materice (HeLa) i melanoma (Hs 294T). Selektivnost testa utvrđena je ispitivanjem na zdravim ćelijama – humanim fibroblastima pluća (MRC-5). Svi ekstrakti ispoljili su citotoksično delovanje prema ćelijama karcinoma grlića materice (HeLa), a ekstrakt nadzemnog dela G. lucidum i na HT-29 ćelijsku liniju karcinoma kolona. Geraniin je najjaču aktivnost ispoljioprema HeLa ćelijskoj liniji, uz izraženu selektivnost, dok je citotoksična aktivnost elagne kiseline bila slabija, uz nisku selektivnost.
AB  - The subject of this doctoral dissertation was the chemical characterization and screening of the pharmacological activity of the underground and aerial parts of eight species of the genus Geranium (G. macrorrhizum, G. robertianum, G. phaeum, G. palustre, G. sanguineum, G. columbinum, G. pyrenaicum and G. lucidum), originating from the Vlasina Plateau.The volatile fractions isolated by hydrodistillation were analyzed by gas chromatography with flame ionization detector and mass detector (GC-FID/MS). Sesquiterpenes dominated in the volatile fraction of the underground parts of G. macrorrhizum as well as in the fractions of the aerial parts of all species studied except G. phaeum and G. columbinum, where diterpenes predominated. Fatty acids and their derivatives predominated in the volatile fractions of the underground parts of all examined species except G. macrorrhizum.Spectrophotometric analysis showed that the methanolic extracts were rich in polyphenolic compounds and that tannins were the dominant polyphenols in almost all samples.Two compounds were isolated from the methanolic extract of the aerial parts of G. palustre by column chromatography and identified as the ellagitannin corilagin and the flavonoid astragalin-2''-O-gallate by UV-Vis, MS - and NMR spectra.In the methanolic extracts, liquid chromatography with mass spectrometry (LC-MS) was used for identification and quantification of seven phenolic acids (ellagic, gallic, galloylquinic, galloylshikimic, protocatechuic, chlorogenic and brevifolincarboxylic acid), 12 flavonoid compounds (quercetin, kaempferol, their heterosides, galloylated derivatives of heterosides and methylated derivatives of kaempferol) and 14 tannin compounds, including ellagitannins (geraniin and its isomer, corilagin, telimagrandin I and isomeric compounds, telimagrandin II, and chebulagic acid) and gallotannins (galloylglucose, digalloylglucose, trigalloylglucose, and pentagalloylglucose).By studying the reducing power in the FRAP assay and the antiradical activity in the DPPH assay, the extracts were found to have significant antioxidant potential in vitro, comparable to L-ascorbic acid as a known antioxidant. When the effect on human serum oxidative stress parameters was examined, it was found that the extracts led to a significant increase in the antioxidant capacity of human serum, and the effect of most of the extracts was better than or comparable to the effect of Trolox as a positive control.The antimicrobial activity of the extracts was tested by the microdilution method on seven standard strains of Gram-positive and Gram-negative bacteria, one standard strain of the yeast Candida albicans, ten clinical isolates of Escherichia coli, and seven clinical isolates of Klebsiella pneumoniae. Most of the extracts showed activity against all standard strains. Significant activity against all clinical isolates of E. coli was observed for the extracts of the aerial parts of G. robertianum and G. columbinum, while activity against clinical isolates of K. pnaeumoniae was weaker.The in vitro cytotoxic activity of the extracts, geraniin and ellagic acid was tested by MTT assay on human prostate (PC-3), colon (HT-29), cervical (HeLa) and melanoma (Hs 294T) cell lines. The selectivity of the assay was determined by testing on healthy cells - human lung fibroblasts (MRC-5). All extracts showed cytotoxic activity against cervical cancer cells (HeLa) and the extract from the aerial parts of G. lucidum also against the HT-29 colon cancer cell line.Geraniin showed the strongest activity against the HeLa cell line, with pronounced selectivity, whereas the cytotoxic activity of ellagic acid was weaker and with lower selectivity.

The Geranium L . species are widely used in ethnomedicine. In the current study, the total phenolic and tannin content, antioxidant and antimicrobial activity of methanol extracts of eight Geranium species were investigated.

In silico prediction of protein targets for volatile compounds of Geranium L. species

AB  - The Geronrum L. species have significant use in traditionaI medicine. Previous studies showed that these plants are rich in phytochemicals, especially in polyphenols and votatile compounds. The aim of this study was to determine potential target proteins for volatile compounds present in Geranium species and to provide a rationale for its current uses. The structures of identified volatile molecules were collated and their potential protein targets were predicted using PIDGIN software. The 2D and 3D structures of these compounds were generated using KingDraw software, while the 3D structures of selected target proteins
were obtained from Protein Data Bank. All volatile compounds were docked against the whole surface of endothelial PAS domain containing protein 1 (EPAS-1; PDBID: F310) and musarinic receptors M1 (PDBID:6WJC), M2 (PDBID:3UON), M4 (PDBID:5DG) and M5 (PDBID: 6OL9) using LeDock software. The study revealed several compounds with the potential to interact with target
proteins. Hexahydrofarnesyl acetone and γ-curcumene formed the most favorable binding poses inside the binding sites of all muscarinic receptors. Linalool and δ-guaiene forned favorable interactions to EPAS-l target protein. Predicted interaction energies are generally lower compared to ligands for these protein targts, mainly due to their size, hydrophobic properties, and the lack of H-bond forming groups. However, these compounds can fit the target site and form favorable interactions. It can be hypothesized that these plants and their extracts exert pharmacological activity via synergistic action of several components that
can interact with multiple proteins in vivo. Additionally, new therapeutic applications of these plants may be proposed based on the other targets predicted by PIDGIN but not reported in this abstract. Further in vitro studies are required to confirm in silico prediction and gain further insights on potential interactions of tested compounds and target proteins.

However, these compounds can fit the target site and form favorable interactions. It can be hypothesized that these plants and their extracts exert pharmacological activity via synergistic action of several components that
can interact with multiple proteins in vivo. Additionally, new therapeutic applications of these plants may be proposed based on the other targets predicted by PIDGIN but not reported in this abstract. Further in vitro studies are required to confirm in silico prediction and gain further insights on potential interactions of tested compounds and target proteins.",

AB  - Geranium species are widely used in traditional medicine of Balkan. The aim of this work was to investigate and compare chemical composition of volatile fractions obtained by hydrodistillation from aerial parts of G. macrorrhizum, G. phaeum, G. sanguineum, G. robertianum, G. palustre, G. pyrenaicum, G. columbinum and G. lucidum as well as from underground parts of G. macrorrhizum and G. phaeum, originated from Vlasina plateau in South Eastern Serbia. The volatiles were analyzed using GC/MS and GC-FID. G. palustre volatiles have been studied for the first time with β-selinene (18.6 %) as a characteristic compound. The cluster analysis revealed separation of volatiles into two main groups. Volatile fractions of G. macrorrhizum were separated from all other samples due to high sesquiterpene content (92.3 % in aerial and 94.6 % in underground parts). The volatile fractions of other samples were mainly composed of sesquiterpenes (10.8–61.8 %), diterpenes (12.9–43.0 %) and fatty acids and their derivatives (6.6–21.6 %) with the exception of volatile fraction of G. phaeum underground parts which was dominated only by fatty acids and their derivatives (76.6 %). The results presented in this article contribute to the knowledge on the chemistry of this genus and advances the knowledge on flora of southeast Serbia.

AB  - The antimicrobial activity of methanol extracts of aerial parts of 8 Geranium

tested against 8 standard strains (Staphylococcus aureus, Enterococcus faecalis,
Bacillus subtilis, Escherichia coli, Klebsiella pneumaniae, Pseudomonas aeruginosa,
Salmonella abony and Candida albicans) as well as 10 clinical isolates of E. coli.
Also, total phenolics and tannins were quantified using spectrophotometric Folin-
Ciocalteu method. The extracts exhibited antibacterial and anticandidal activity with
minimal inhibitory concentrations (MICs) between 12.5 and 200 μg/ml. E. faecalis
was most sensitive strain (MIC 12.5-50 μg/ml), especially to G. columbinum and G.
phaeum extracts (MICs 12.5 μg/ml). The determined amounts of total phenolics and
tannins were in the range of 170-534 and 38-386 mg GAE/g of dry extract,
respectively, with the highest content in G. sanguineum extract. However, the
antimicrobial activity was not related to total polyphenolics or tannin content and
further investigation in regard with phenolic profile is needed.
