Antifungal Activity of Lavandula angustifolia Essential Oil against Candida albicans: Time-Kill Study on Pediatric Sputum Isolates

AB  - The aim of our study was to determine the susceptibility of 15 Candida albicans sputum isolates on fluconazole and caspofungin, as well as the antifungal potential of Lavandula angustifolia essential oil (LAEO). The commercial LAEO was analyzed using gas chromatography-mass spectrometry. The antifungal activity was evaluated using EUCAST protocol. A killing assay was performed to evaluate kinetics of 2% LAEO within 30 min treatment. LAEO with major constituents’ linalool (33.4%) and linalyl acetate (30.5%) effective inhibited grows of C. albicans in concentration range 0.5–2%. Fluconazole activity was noted in 67% of the isolates with MICs in range 0.06–1 µg/mL. Surprisingly, 40% of isolates were non-wild-type (non-WT), while MICs for WT ranged between 0.125–0.25 µg/mL. There were no significant differences in the LAEO MICs among fluconazole-resistant and fluconazole-susceptible sputum strains (p = 0.31) and neither among caspofungin non-WT and WT isolates (p = 0.79). The 2% LAEO rapidly achieved 50% growth reduction in all tested strains between 0.2 and 3.5 min. Within 30 min, the same LAEO concentration exhibited a 99.9% reduction in 27% isolates. This study demonstrated that 2% solution of LAEO showed a significant antifungal activity which is equally effective against fluconazole and caspofungin susceptible and less-susceptible strains.

Microencapsulation of Oregano and Thyme essential oils with hydroxypropyl-β cyclodextrin

AB  - Among plant natural products, Oregano and Thyme essential oils (EOs) are well-known for 
their antimicrobial and antioxidant activity. The biological activities of EOs may be reduced 
due to oxidation and volatilization. Stability and preservation of EOs can be assured with the 
microencapsulation method based on complex coacervation technology. Cyclodextrins are 
suitable as carriers for volatile substances insoluble in water, and hydroxypropyl-β cyclodextrin (HPCD) leads the group considering the aqueous solubility and safe toxicity 
profile. The aim of this study was to find conditions for preparing Oregano and Thyme EOs 
microencapsulates with the best technological properties.Oregano and Thyme EOs were 
encapsulated by the freeze drying (lyophilization) method. Nine different combinations were 
prepared, where HPCD content (10, 15 and 20%) and EO:HPCD mass ratio (1:1, 1:5 and 
1:10) were varied. After stirring (200 rpm) for 24 h at a room temperature, suspensions were 
filtered through 0.45 mm PTFE filters. Samples were evaporated under vacuum and frozen (-
80 °C for 1 h), then main drying was carried out (-60 °C, pressure of 0.011 mbar for 40 h), 
and final drying (-65 °C, pressure of 0.054 mbar for 1 h). To ensure the particle purity, 
lyophilizates were washed with acetonitrile and dried at 25 °C. The content of each EO was 
determined spectrophotometrically, encapsulation efficiency (EE%) and yield were 
calculated. Duncan’s post hoc test was used to evaluate the differences between samples. 
The best conditions for both EOs were obtained with 15% HPCD and EO:HPCD mass ratio 
of 1:10. Yields of EOs for chosen Oregano and Thyme EOs complexes were 86.81 ± 2.20% 
and 89.83 ± 2.80%, respectively, with the following EE% of 49.08 ± 1.80% and 49.29 ± 
0.18%. Microencapsulation is a promising method for improving the EOs stability profile, 
and these results could be very useful in the pharmaceutical and food industry for the 
implementation of new products.

Oregano essential oil sensitivity of multiresistant Acinetobacter baumannii strains isolated from COVID-19 patients

COVID-19 pandemic brought a burden to the health care system including a higher rate of patients admitted to the intensive care unit (ICU). Mechanical ventilation, needed in COVID-19 patients admitted to ICU, increases the risk of developing multidrug-resistant (MDR) Acinetobacter baumannii superinfection, which may cause major problems in antibiotic treatment.
OBJECTIVES
The aim of this research was to examine the susceptibility of A. baumannii isolates recovered from COVID-19 patients 
to antibiotics usually used as treatment options, and to Oregano essential oil (EO) as naturally derived compound.
METHODS 
The 54 A. baumannii isolates were recovered from COVID-19 patients admitted to ICU, during period from December 
2020 to February 2021. Biofilm formation ability, as the factor that contributes to bacterial resistance, was examined. Minimum inhibitory concentrations (MICs) were determined for meropenem, imipenem, colistin, gentamicin, tobramycin and levofloxacin. In addition, all 54 isolates of A. baumanii were tested for susceptibility to Oregano EO.
RESULTS 
Results obtained in this study revealed a significant MDR profile of tested A. baumannii isolates. Even 74% (40/54) were strong biofilm producers. A. baumannii isolates were resistant to all tested antibiotics, except to colistin, leaving colistin as the only treatment option. On the other hand, MIC results obtained for Oregano EO were extremely promising: 0.279 mg/mL for all tested isolates. The obtained results emphasize a potentially new treatment approach to infections caused by MDR A. baumannii, using natural products such as EOs.

Optimization and modelling of gentiopicroside, isogentisin and total phenolics extraction from Gentiana lutea L. roots

AB  - Perennial plant Gentiana lutea L. is used worldwide for the preparation of pharmaceutical and food products. Health benefits of G. lutea roots are associated with the presence of major bitter-tasting secoiridoid gentiopicroside and xanthone isogentisin. The aim was to optimize the heat-assisted extraction of gentipicroside (GP), isogentisin (ISG) and total phenolics (TP) from G. lutea roots and develop models with high accuracy and prediction capacity by response surface methodology (RSM) and artificial neural networks (ANN). Extracts were prepared according to central composite design. Significant independent variables which were previously identified by Plackett-Burman screening design were varied at five levels - temperature (20−80 °C), time (8−180 min), solid-to-solvent ratio (1:10−1:50) and ethanol concentration (10–70 %). Contents of GP, ISG (by HPLC-DAD) and TP (by Folin–Ciocalteu method) were analyzed. The optimal conditions for the extraction were temperature of 65 °C, time of 129.08 min, solid-to-solvent ratio of 1:40, and ethanol concentration 49.33 %. Under these conditions, experimentally obtained results for GP (18.03 mg/g dw), ISG (8.15 mg/g dw) and TP (17.46 mg of gallic acid equivalents/g dw) content were in agreement with the values predicted by RSM and ANN. Comparison of models through the coefficient of determination (R2) and the root mean square error (RMSE) showed that ANN approach was superior to RSM in predicting and modelling GP, ISG and TP content, simultaneously. Effective heat-assisted extraction method for the extraction of GP, ISG and TP from the roots of G. lutea was designed, and models with high accuracy and good prediction capacity were developed.

Stability testing of herbal medicines

AB  - Nowdays, the use of products based on medicinal herbs (phytopreparates) are in scientific focus, both in prevention or in treatment of various diseases. Herbal products have been recognized as an excellent source of bioactive compounds which have positive effects on human health. Beside teas, as standard herbal products, phytopreparations are much more represented as final pharmaceutical form on the market today. Phytopreparations must satisfy standard quality, which means that they must be physically, chemically and microbiological stable and have a high degree of purity. In order to determine the stability, storage conditions, shelf-life of the products, stability tests are conducted, which involving tests of environmental factors influence: temperature, relative humidity, light. Stability tests are performed at different stages of development and production. In accordance with the EMEA (European Agency for Drugs and Medical Devices) for a variety of herbal preparations different specific stability tests are conducted.
AB  - Danas, sve je veći akcenat na upotrebi preparata na bazi lekovitog bilja (fitopreparata), kako u prevenciji tako i u lečenju različitih oboljenja. Biljni preparati su prepoznati kao odličan izvor bioaktivnih komponenti, koji imaju pozitivne efekte na zdravlje ljudi. Pored čajeva, kao standardnih biljnih proizvoda, danas su na tržištu mnogo više zastupljeni fitopreparati kao finalne farmaceutske forme. Fitopreparati moraju biti standardnog kvaliteta, što podrazumeva da moraju biti fizički, hemijski i mikrobilološki stabilni, da su standardizovanog sastava i da su visokog stepena čistoće. U cilju utvrđivanja koliko je jedan proizvod stabilan, definisanja uslova čuvanja, određivanja roka trajanja, sprovode se ispitivanja stabilnosti, koja podrazumevaju ispitivanja uticaja faktora okoline: temperature, relativne vlažnosti vazduha, svetlosti na promenu kvaliteta gotovog proizvoda. Ispitivanja stabilnosti se izvode u različitim fazama razvoja i proizvodnje. U skladu sa zahtevima EMEA (Evropska agencija za lekove i medicinska sredstva) za različite biljne preparate postavljaju se različiti specifični testovi stabilnosti.

Pharamaceutical hyaluronic acid based carriers for parenteral use, particularly conjugates with taxol: A review

AB  - In recent years, drug delivery systems are especially interesting because they allow therapeutic efficiency and reduce side effects. Conjugates of hyaluronic acid and chemotherapeutic agents, especially taxol, have been recognized as excellent solution for many therapeutic indications. The aim of conjugation of taxol with hyaluronic acid was the toxicity reduction on healthy cells and increase of selectivity on tumor cells. Hyaluronic acid could be used in various pharmaceutical preparations, also in parenteral, because of its biocompatibility and biodegradability. Hyaluronic acid as a carrier belongs to in situ gelling systems, which is passing a phase transition in response to external conditions such as temperature, different pH value or the presence of ions, enabling prolonged effect of the drug substances. Many tumor cells have a large number of receptors for hyaluronic acid, which has a great affinity for the tumor cells. The incorporation of taxol in gelling system of hyaluronic acid provides sustained drug release, increased anticancer activity, reduced damage of healthy cells and increased selectivity on cancer cells.
AB  - Preparati sa produženim oslobađanjem su poslednjih godina posebno aktuelni jer omogućavaju veću terapijsku efikasnost, smanjuju pojavu neželjenih efekata i omogućavaju ciljano delovanje. Posebno su poslednjih godina interesantni konjugati hijaluronske kiseline i citostatika, a jedan od njih je i taksol, koji pored mnogobrojnih terapijskih indikacija ispoljava i niz neželjenih efekata. Cilj konjugovanja taksola sa hijaluronskom kiselinom je smanjenje toksičnosti na zdravim i povećanje selektivnosti na tumorskim ćelijama. Hijaluronska kiselina se može primenjivati u različitim farmaceutskim preparatima, pa i u parenteralnim, jer ima niz prednosti kao što su biokompatibilnost i biodegradibilnost. Spada u in situ gelirajuće sisteme koji se podvrgavaju faznom prelazu u odgovoru na spoljne stimulanse kao što su temperatura, pH ili prisustvo jona, čime se omogućava produženo dejstvo lekovite supstance. Mnoge tumorske ćelije pokazuju ekspresiju velikog broja receptora za hijaluronsku kiselinu, pa ona ima veliki afinitet za tumorske ćelije. Inkorporiranjem taksola u gelirajući sistem sa hijaluronskom kiselinom omogućava se postepeno oslobađanje leka, povećanje antikancerskog delovanja, smanjuje se oštećenje zdravih ćelija i povećava selektivnost leka.
