Cream-gel with ceramides (part II): An evaluation of the effects on skin hydration and transepidermal water loss

AB  - Taking into account the latest discoveries in the structure and functions of ceramides (sphingolipids, sphingoids), manufacturers of cosmetic raw materials propagate to use ceramides (nature-identical ceramides, ceramide analogues, pseudoceramides) for the purpose of restoration of the natural intercellular lipid barrier. Despite putative importance, there is no consistent data on the benefits of ceramides when used in conventional, consumer products on skin that has suffered minor barrier damage. Transepidermal water loss (TEWL) and skin moisture content are two parameters frequently used for assessment of the influence of cosmetic/pharmaceutical products on barrier function. The ceramides-containing cream-gel has been formulated, and its influence on barrier function during 14 days, compared to “placebo” containing no sphingolipids, assessed on six volunteers with no apparent barrier damage. It was shown that the cream-gel improved skin moisture content, as well as the overall barrier function quality. The lack of significant differences in the influence of the cream-gel and “placebo” on the measured parameters indicated that, for such a kind of product, there was no evidence for the beneficial effects of ceramides on the skin that has suffered minor barrier damage.
AB  - Novija otkrića u vezi strukture i funkcije ceramida (sfingolipida, sfingoida) kože, poslužila su proizvođačima kozmetičkih sirovina da često nekontrolisano i neargumentovano propagiraju upotrebu ceramida u kozmetičkim proizvodima u svrhe reparacije prirodne intercelularne lipidne barijere. Rezultati ispitivanja efekata ceramida na koži često ne daju čvrste dokaze da primena konvencionalnih kozmetičkih proizvoda sa ceramidima može da popravi neznatno oštećenu kožu. Pouzdana metoda za procenu uticaja kozmetičkih/farmaceutskih preparata na funkciju kožne barijere, odnosno efikasnosti i/ili prihvatljivosti formulacije predstavlja ispitivanje transepidermalnog gubitka vode (TEGV) iz kože. Savremena formulacija krem-gela sa ceramidima je aplikovana na kožu i ispitan je njen uticaj na funkciju kožne barijere na panelu od 6 ispitanika sa zdravom kožom, u vremenskom periodu od 14 dana, prema unapred zadatom programu u odnosu na “placebo” formulaciju. Pokazano je da ispitivani krem-gel pozitivno utiče na hidratisanost (vlažnost) stratum corneuma (SC) i transepidermalni gubitak vode, odnosno kvalitet kožne barijere. Sa druge strane, odsustvo statistički značajnih razlika TEGV i vlažnosti kože pod uticajem ispitivanih formulacija ukazuje da, u ovom slučaju, nema dovoljno dokaza o prednostima prisustva ceramida u "aktivnom" krem-gelu u odnosu na “placebo” formulaciju, kada su primenjeni na neznatno oštećenu ili neoštećenu kožu ispitanika.

AB  - In this work results of model creams investigation containing polymeric emulsifier in the form of „Liquid Dispersion Polymers”, Salcare® SC 91 (Sodium acrylates/paraffinum liquidum/PPG-1 Trideceth-6; 3 % (m/m)) and 30 % (m/m) of different polarity oil phases (paraffin, light liqiud; medium-chain triglycerides and almond oil) are presented. For physico-chemical characterization and physical stability assessment of prepared creams investigations of appearance, physical stability, pH value and electrical conductivity alongside with rheological measurements (rotational and oscillatory) in the period of three months were performed. All the tested samples had good macroscopic behavior and physical stability and their pH values were acceptable for topical application. Rheological measurements revealed that oil phase type had great influence on applicative characteristics and stability of emulsion systems. Model cream samples prepared with more polar oil phases had better stability than cream containing less polar phase.
AB  - U radu su prikazani rezultati ispitivanja model kremova izrađenih sa polimernim emulgatorom u formi tečne disperzije, Salcare® SC 91 (Natrijum Akrilat Kopolimer/Tečni parafin/PPG-1 Tridecet-6; 3 % (m/m)) i 30 % (m/m) masne faze različite polarnosti (tečni laki parafin, trigliceridi srednje dužine lanca i bademovo ulje). U cilju fizičko-hemijske karakterizacije i procene stabilnosti izrađenih kremova sprovedena su ispitivanja izgleda, fizičke stabilnosti, pH vrednosti i električne provodljivosti kao i reološka analiza (kontinualna i oscilatorna) u periodu od 3 meseca. Svi ispitivani uzorci pokazali su zadovoljavajući izgled i fizičku stabilnost a vrednosti pH bile su u opsegu prihvatljivom za preparate za primenu na kožu. Reološka merenja su pokazala da tip upotrebljene masne faze može imati znatan uticaj na aplikativne karakteristike i stabilnosti datih emulzionih sistema. Uzorci model kremova izrađenih sa masnim fazama veće polarnosti pokazali su se stabilnijim u odnosu na krem sa najmanje polarnom masnom fazom.

Liquid crystals in pharmaceutical and cosmetic preparations

AB  - Liquid crystals are physical state, gathering the features of liquids and solids, at the same time. In common property with liquids is more or less pronaunced capability of flowing, whereas, like the solids, they have arranged, crystal state. In that way, liquid crystals phase represents intermediate state of matter, also signed as mesomorphic phase, mesophase. Thermotropic and liotropic crystals could be distinguished. For the pharmaceutical application the liotropic liquid crystals are more important phase. Very polar tensids (ethoxilated fatty alcohols,..) could make hydrated micelles, and in such a condesated form they could be structured as cubic liquid crystals. When determined tensids are included into the gel formulation, a transparent and elastic tenside-gel with liquid crystals is formed. A high tenside content in form of liquid crystals, existed in these preparations, influences the skin lipids structure, improving stratum corneum permeability for mentioned substances and thus provide better therapeutic effect. In cosmetics, liquid crystals are used with decorative purpose. For the reason of irritation caused by some colours, the cholesteric liquid crystals are particularly convenient, commonly used as colour vehicles, in nail lacquers, eye shadows and lip pencils.
AB  - Tečni kristali su agregatno stanje koje objedinjuje osobine tečnosti i čvrstih tela. Zajedničko svojstvo sa tečnostima je više ili manje izražena sposobnost proticanja, a sa čvrstim telima raspoređeno, kristalno stanje. Tako faze tečnih kristala reprezentuju međustanje, zbog čega se označavaju i kao mezofaze. Razlikuju se termotropni i liotropni tečni kristali. Za farmaciju imaju značaj uglavnom liotropni tečni kristali. Veoma polarni tenzidi (etoksilirani masni alkoholo…) mogu da stvaraju hidratisane micele koje zgusnute obrazuju strukturu kubičastog tečnog kristala. Kada se određeni tenzidi uključe u formulaciju gela, obrazuje se tečno-kristalni tenzid - gel koji je optički transparentan i elastičan. Visok sadržaj tenzida u obliku tečnih kristala u ovim preparatima utiče na strukturu lipida kože, povećava permeabilnost stratum corneum-a za lekovite supstance i obezbeđuje bolji terapijski efekat. U kozmetičkoj industriji se tečni kristali koriste uglavnom u dekorativne svrhe. Zbog iritacije do koje dovode neke boje, posebno su pogodni holesterolski tečni kristali, koji se koriste kao nosači boja u lakovima za nokte, senkama za oči i olovkama za usta.

For the reason of irritation caused by some colours, the cholesteric liquid crystals are particularly convenient, commonly used as colour vehicles, in nail lacquers, eye shadows and lip pencils., Tečni kristali su agregatno stanje koje objedinjuje osobine tečnosti i čvrstih tela. Zajedničko svojstvo sa tečnostima je više ili manje izražena sposobnost proticanja, a sa čvrstim telima raspoređeno, kristalno stanje. Tako faze tečnih kristala reprezentuju međustanje, zbog čega se označavaju i kao mezofaze. Razlikuju se termotropni i liotropni tečni kristali. Za farmaciju imaju značaj uglavnom liotropni tečni kristali. Veoma polarni tenzidi (etoksilirani masni alkoholo…) mogu da stvaraju hidratisane micele koje zgusnute obrazuju strukturu kubičastog tečnog kristala. Kada se određeni tenzidi uključe u formulaciju gela, obrazuje se tečno-kristalni tenzid - gel koji je optički transparentan i elastičan. Visok sadržaj tenzida u obliku tečnih kristala u ovim preparatima utiče na strukturu lipida kože, povećava permeabilnost stratum corneum-a za lekovite supstance i obezbeđuje bolji terapijski efekat. U kozmetičkoj industriji se tečni kristali koriste uglavnom u dekorativne svrhe. Zbog iritacije do koje dovode neke boje, posebno su pogodni holesterolski tečni kristali, koji se koriste kao nosači boja u lakovima za nokte, senkama za oči i olovkama za usta.",
