AB  - Laurus nobilis L., Lauraceae, u narodu poznat kao lovor, je cvetni, zimzeleni žbun koji
samoniklo raste u većem delu Crne Gore, a često se uzgaja i koristi kao aromatična, začinska i lekovita
biljka. Cilj ovog istraživanja je bio da se analizira hemijski sastav etarskog ulja (EU) ploda i lista, a
zatim testira antibakterijska i antifungalna aktivnost EU na sledećim sojevima: Staphylococcus aureus
ATCC25923, Enterococcus faecalis ATCC29212, Bacillus subtilis ATCC6633 Escherichia coli
ATCC25922, Klebsiella pneumoniae ATCC13083, Salmonella Abony NCTC6017, Pseudomonas
aeruginosa ATCC27853 i Candida albicans ATCC10231. Ispitivanje kvalitativnog i kvantitativnog
sastava etarskih ulja lista i plodova lovora izvršeno je gasnohromatografskom tehnikom spregnutom
sa masenom spektrometrijom (GC-MS). Utvrđeno je da je etarsko ulje ploda bogato 1,8-cineolom
(33,3%), α-terpenil acetatom (7,0%), α-pinenom (5,8%), β-elemenom (5,7%), sabinenom (5,3%), β-
pinenom (4,0%), borneolom (2,1%) i terpinen-4-olom (2,1%). Najvažnije komponente
identi�ikovane u EU listova su 1,8-cineol (35,1%), α-terpinil acetat (10,4%) i linalol (7,6%), između
ostalih, a u skladu su sa prethodno dobijenim literaturnim podacima (1). Dodatno, ispitivana su
antibakterijska i antifungalna svojstva EU lista i ploda lovora bujon mikrodilucionom metodom. EU
ploda je pokazao antibakterijsko i antifungalno svojstvo protiv skoro svih testiranih sojeva
mikroorganizama, dok je EU lista inhibiralo sledeće sojeve: Staphylococcus aureus ATCC25923,
Enterococcus faecalis ATCC29212, Bacillus subtilis ATCC6633 i Candida albicans ATCC10231.
Escherichia coli ATCC25922 je bakterijski soj koji je pokazao najveću otpornost na etarska ulja lovora,
kako ploda, tako i lista. Dobijeni rezultati za etarska ulja lovora i 1,8 -cineol, koji je njihov glavni
sastojak, su u skladu sa prethodnim studijama (2).
AB  - Laurus nobilis L., Lauraceae, popularly known as laurel, is a flowering, evergreen bush which
grows wildly in most parts of Montenegro, and is often cultivated and used as an aromatic, flavoring
spice and medicinal plant. The aim of this research was to analyze the chemical composition of the
essential oil (EO) of fruit and leaf, and afterwards antibacterial and antifungal activity of EOs on the
following strains: Staphylococcus aureus ATCC25923, Enterococcus faecalis ATCC29212, Bacillus
subtilis ATCC6633 Escherichia coli ATCC25922, Klebsiella pneumoniae ATCC13083, Salmonella Abony
NCTC6017, Pseudomonas aeruginosa ATCC27853 i Candida albicans ATCC10231. The qualitative and
quantitative composition of the EOs of fruit and leaves was carried out by using a gas
chromatographic analysis coupled with mass spectrometry (GC-MS). The EO of fruit was found to be
rich 1,8-cineole (33.3%), α-terpinyl acetate (7.0%), α-pinene (5.8%), β-elemene (5.7%), sabinene
(5.3%), β-pinene (4.0%), borneol (2.1%) and terpinene-4-ol (2.1%). The most important
components identified in the leaves EO were 1,8-cineol (35.1%), α-terpinyl acetate (10.4%) and
linalool (7.6%), among others, are in line with previous literature reports (1). In addition, the
antibacterial and antifungal properties of leaf and fruit laurel EOs were examined according to the
broth microdilution method. The fruit EO showed antibacterial and antifungal activities against
almost all strains of the microorganisms tested, whereas the leaf EO was able to inhibit:
Staphylococcus aureus ATCC25923, Enterococcus faecalis ATCC29212, Bacillus subtilis ATCC6633 and
Candida albicans ATCC10231. Escherichia coli ATCC25922 was bacterial strain that showed the
highest resistance to both tested laurel EOs. The results obtained herein for laurel EO and 1,8-cineole
as its main constituent are in accordance with previous studies (2).

AB  - Usnic acid is one of the most investigated lichen secondary metabolites, with several proven biological properties with potential medical relevance. However, its unfavorable physico-chemical properties, as well as observed hepatotoxicity, have discouraged wide-range utilization of usnic acid as a promising therapeutic agent. In accordance with the growing research interest in the development of nanotechnology, especially in the arena of preparations based on natural sources of medicinal compounds, usnic acid incorporated into nano-and microsized colloidal carriers has been a subject of a large number of publications. Therefore, this review discusses the overall results of the studies dealing with usnic acid encapsulated into lipid-based, polymeric and nonorganic micro-and/or nanocarriers, as potential drug delivery systems for this natural compound, in an attempt to introduce its usage as a potential antitumor, antimicrobial, wound-healing, antioxidative and anti-inflammatory drug.

Cytotoxic activity of supercritical CO2 extract of old man's beard in L929 fibrosarcoma cell line

AB  - Old man’s beard (Usnea barbata) is one of the most investigated lichens which biological properties
(such as antimicrobial, anti-inflammatory and cytotoxic activities) have been proven in several scien-
tific studies. Stated properties of old man’s beard have been attributed mainly to its major secondary
metabolite - a dibenzofuran derivative, usnic acid. The aim of the current study was to examine cyto-
toxic activity of the quantified supercritical CO2 extract and pure usnic acid in parallel against L929
fibrosarcoma cells. Our results indicated the tested extract to be more toxic towards the investigated
cell line compared to usnic acid per se, despite the high amount of this compound (81.41% (w/w))
revealed in the extract using HPLC. Such result may be connected to the unidentified compounds
present in the supercritical CO2 extract that could be responsible for apoptosis and oxidative stress in
the exerted cytotoxicity of the investigated extract, but not usnic acid. Our study supports further in-
vestigations of supercritical CO2 extract of U. barbata as a prospective therapeutic agent with potential
relevance in the treatment of sarcoma.

Stability, antioxidant activity, in vivo safety and efficacy of creams with standardized wild apple fruit extract: a comparison of conventional and biodegradable emulsifiers

AB  - Objective The aim of the study was in vitro and in vivo characterization of cosmetic cream with 6% of standardized wild apple fruit extract, stabilized by conventional non-ionic emulsifier-CEW, in order to determine the influence of emulsifiers (conventional vs. biodegradable) on the characteristics of creams and their effects on the skin. Methods Organoleptic and physico-chemical (pH values and electrical conductivity) analysis was performed, determination of fruit acids-FAs content (using HPLC analysis) and estimation of its antioxidant activity-AA (using DPPH test) during 180 days. In vivo study included following examinations: screening of safety profile (after creams application under occlusion during 24 h at human skin); skin moisturizing potential, transepidermal water loss-TEWL, skin pH after 28 days of cream application and hypopigmentation efficacy 7 days of cream application at artificially induced skin hyperpigmentation. Results Investigated cosmetic cream-CEW showed satisfactory organoleptic, physico-chemical characteristics, stability, FAs content (0.13%) and AA (19.25 +/- 0.67 %RSC) after preparation, which remained unchanged over the study period. In vivo investigation revealed absence of skin irritation after CEW's application under occlusion. An increase of skin moisturization (after 14 days Delta EC was 18.52 +/- 11.51 and after 28 days of applications 16.52 +/- 9.36) during 28 day-study, with unchanged TEWL and skin pH values was shown. Decrease of melanin index was revealed, too (after 7 days Delta MI was -31.40 +/- 16.50). Conclusion Cosmetic cream stabilized by conventional emulsifier showed better antioxidant potential and weaker moisturizing and hypopigmentation effects related to the cream with same composition but stabilized by biodegradable emulsifiers. Based on all mentioned above, investigated cosmetic cream might be considered for potential use as modern, stable, safe and efficient cosmetic product in the prevention and/or treatment of oxidative stress-related skin changes and/or damages, for moisturization of dry, even irritated skin as well as for lightening of hyperpigmented skin.

AB  - Lichens represent a promising source of antimicrobial, cytotoxic and antioxidant agents. Their great pharmacological potential lies in the fact that they represent specific symbiotic organisms and thus possess natural roles allowing them to be highly adaptable to different environmental conditions. On the other hand, stated biological activities of lichens with prospective medicinal significance may be connected to their long-term use in traditional treatment of various ailments. Genus Usnea from the Parmeliaceae family is certainly one of the best studied in terms of chemical composition and biological properties of its extracts and/or isolated compounds. In the first part of the study, a detailed review of literature has been performed yielding a detailed report on the investigations of biological activities of the lichens belonging to this genus. In the second part of the study, chemical composition of the lichens from the genus was described and, additionally, a survey of the biological properties of the most representative secondary metabolites in these lichens has been reported. It could be concluded that the extracts and/or isolated compounds from the lichens belonging to the genus Usnea may be considered a valuable source of prospective drug candidates with potential clinical relevance.

AB  - The objective of this work was to investigate and profoundly characterize low-energy nanoemulsions as multifunctional carriers, with slight reference to dermal administration. An evidence-based approach was offered for deepening the knowledge on their formation via spontaneous emulsification. Curcumin, a compound of natural origin, potentially powerful therapeutic, was chosen as a model API. Due to curcumin's demanding properties (instability, poor solubility, low permeability), its potentials remain unreached. Low-energy nanoemulsions were considered carriers capable of overcoming imposed obstacles. Formulation consisting of Polysorbate 80 and soybean lecithin as stabilizers (9:1, 10%), medium-chain triglycerides as the oil phase (10%) and ultrapure water was selected for curcumin incorporation in 3 different concentrations (1, 2 and 3 mg/mL). Physicochemical stability was demonstrated during 3 months of monitoring (mean droplet size: 111.3-146.8 nm; PDI  lt  0.2; pH: 4.73-5.73). Curcumin's release from developed vehicles followed Higuchi's kinetics. DPPH (IC50 = 0.1187 mg/ mL) and FRAP (1.19 +/- 0.02 mmol/g) assays confirmed that curcumin acts as a potent antioxidant through different mechanisms, with no alterations after incorporation in the formulation. High biocompatibility in line with antigenotoxic activity of curcumin-loaded formulations (protective and reparative) was estimated through Comet assay. A multidisciplinary approach is needed to fully characterize developed systems, directing them to more concrete application possibilities.

AB  - Topical treatment of skin infections is often limited by drawbacks related to both antimicrobial agents and their vehicles. In addition, considering the growing promotion of natural therapeutic products, our objective was to develop and evaluate naturally-based emulsion system, as prospective topical formulation for skin infections-treatment. Therefore, alkyl polyglucoside surfactants were used for stabilization of a vehicle serving as potential carrier for supercritical CO2-extract of Usnea barbata, lichen with well-documented antimicrobial activity, incorporated using two protocols and three concentrations. Comprehensive physicochemical characterization suggested possible involvement of extract's particles in stabilization of the investigated system. Raman spectral imaging served as the key method in disclosing extract's particles potential to participate in the microstructure of the tested emulsion system via three mechanisms: (1) particle-particle aggregation, (2) adsorption at the oil-water interface and (3) hydrophobic particle-surfactant interactions. Stated extract-vehicle interaction proved to be correlated to the preparation procedure and extract concentration on one hand and to affect the physicochemical and biopharmaceutical features of investigated system, on the other hand. Thereafter, formulation with the best preliminary stability and liberation profile was selected for further efficiency and in vivo skin irritation potential evaluation, implying pertinent in vitro antimicrobial activity against G+ bacteria and overall satisfying preliminary safety profile.

AB  - There is a worldwide ongoing investigation for novel natural constituents with cytotoxic and antioxidant properties. The aim of this study was to investigate chemical profile and stated biological activities of the supercritical CO2 extract (SCE) of old man's beard compared to the extracts obtained using the conventional techniques (Soxhlet extracts and macerate). The most abundant compound identified was usnic acid, which content was inversely proportional to the polarity of the solvent used and was the highest in the SCE, which was the sample revealing the highest cytotoxic activity in tested tumor cell lines (B16 mouse melanoma and C6 rat glioma), with lower IC50 values compared to pure usnic acid. Further investigations suggested both SCE and usnic acid to induce apoptosis and/ or autophagy in B16 and C6, indicating higher cytotoxicity of SCE to be related to the higher degree of ROS production. A good correlation of usnic acid content in the extracts and their antioxidant capacity was established, extricating SCE as the most active one. Presented results support further investigations of SCE of old man's beard as a prospective therapeutic agent with potential relevance in the treatment of cancer and/ or in oxidative stress-mediated conditions.

AB  - Antimicrobial activity of Usnea barbata especially against bacteria involved in pathogenesis of various skin conditions has been well documented in literature. Nevertheless, there are no papers dealing with formulation of its isolates into topical preparations for treatment of skin infections. In present study, alkyl polyglucoside (APG)-based vehicle was developed as carrier of U. barbata CO2-supercritical extract (U-SE) that demonstrated the best anti- microbial potential in preliminary screening. For comparison, chosen extract in the same concentration and using the same procedure was incorporated into a pharmacopoeial vehicle. Comparative evaluation of physicochemical stability, efficiency and safety proved APG-based vehicle to possess certain preferential features as carrier of U-SE compared to the reference one, composing a topical formulation with potential clinical relevance in treatment of skin infections.
AB  - Antimikrobna aktivnost vrste Usnea barbata naročito protiv bakterija koje učestvuju u patogenezi različitih bolesti kože, dobro je dokumentovana u naučnoj literaturi. Uprkos tome, ne postoje radovi koji se bave formulacijom topikalnih preparata na bazi ovog lišaja namenjenih lečenju kožnih infekcija. U ovoj studiji, razvijena je podloga stablizovana alkil poliglukozidnim (APG) emulgatorom, kao potencijalni nosač za ekstrakt vrste U. barbata koji je pokazao najbolji antimikrobni potencijal u preliminarnom istraživanju nekoliko ekstrakata dobijenih upotrebom različitih ekstragenasa/postupaka. Radi poređenja, odabrani CO2-natkritični ekstrakt je inkorporiran u istoj koncentraciji (koja odgovara 2 mas.% usninske kiseline) i istim postupkom izrade u često korišćenu podlogu farmakopejskog kvaliteta, a zatim je sprovedeno uporedno istraživanje fizičko-hemijske stabilnosti, efikasnosti i bezbednosti na obe grupe uzoraka. Rezulati našeg istraživanja pokazali su da se razvijena podloga stabilizovana APG emulgatorom može smatrati pogodnim nosačem za CO2-natkritični ekstrakt vrste U. barbata, čineći topikalnu formulaciju sa potencijalnim kliničkim značajem u terapiji kožnih infekcija. Reološka istraživanja pokazala su zadovoljavajuću sposobnost emulzije stabilizovane APG emulgatorom da ostane fizički stabilna nakon dodatka CO2-natkritičnog ekstrakta vrste U. barbata, u poređenju sa farmakopejskom. Međutim, konduktometrijska merenja su pokazala određena neslaganja sa reološkim nala- zima, namećući potrebu za dodatnim istraživanjima radi konačne procene fizičke stabilnosti testiranih emulzionih sistema. Izmerene vrednosti pH tokom perioda od 90 dana čuvanja uzoraka na sobnoj temperaturi ukazale su na zadovoljavajuću preliminarnu hemijsku stabilnost svih uzoraka. Dalja istraživanja efikasnosti i bezbednosti pokazala su bolji antimikrobni potencijal uzoraka sa podlogom stabilizovanom APG emulgatorom u odnosu na tradicionalno upotrebljavanu farmakopejsku bazu poznatog kvaliteta. Zadovoljavajući preliminarni bezbedonosni profili su pokazani kako za uzorke stabilizovane APG emulgatorom tako i za referentne krem uzorke. Na osnovu navedenog, može se zaključiti da je istraživana podloga bazirana na APG emulgatoru pokazala određene povoljnije karakteristike kao nosač za CO2-natkritični ekstrakt vrste U. barbata u poređenju sa konvencionalno korišćenom podlogom, naročito uzimajući u obzir njene poboljšane senzorne karakteristike, kao osobinu koja je često odlučujuća za komplijansu pacijenata.

Uprkos tome, ne postoje radovi koji se bave formulacijom topikalnih preparata na bazi ovog lišaja namenjenih lečenju kožnih infekcija. U ovoj studiji, razvijena je podloga stablizovana alkil poliglukozidnim (APG) emulgatorom, kao potencijalni nosač za ekstrakt vrste U. barbata koji je pokazao najbolji antimikrobni potencijal u preliminarnom istraživanju nekoliko ekstrakata dobijenih upotrebom različitih ekstragenasa/postupaka. Optimization of extraction of antioxidant components from yarrow herb

AB  - Recently, research efforts have been directed toward medicinal plants and their extracts, as important sources of natural antioxidant. Lots of biologically active compounds are responsible for the antioxidant effects of yarrow - Achillea millefilium L. extracts. The aim of our study was to determine which of the process parameters of pressure enhanced solvent extraction of Millefolii herba is significant for its efficiency and weather there are interactions between the examined parameters. Compression time, decompression time and the number of cycles were identified as independent variables, while the content of total flavonoids, tannins and total polyphenols were selected as dependent variables. For obtaining the extract of M. herba, rich in antioxidative ingredients, compression time should be set on its higher level (2.0 min), decompression time on its lower level (1.30 min) and the number of cycles on its higher level (99).
AB  - Oksidativna degradacija sastojaka hrane dovodi do promene njenog mirisa i ukusa, čime se narušava njen nutritivni i senzorni kvalitet. Iz navedenog razloga je neophodno dodavati antioksidante tokom tehnološkog procesa izrade namirnica. Lekovito bilje i njihovi izolati predmet su istraživanja, pored ostalog i kao izvor antioksidantnih materija i alternativa sintetičkim antioksidansima za primenu u industriji hrane. Istraživanja velikog broja biljaka ukazala su da skoro sve poseduju određena antioksidativna svojstava. Hajdučka trava - Achillea millefilium L. (Asteraceae) i njeni ekstrakti, zahvaljujući prisustvu, pre svega, polifenola, flavonoida i tanina, ispoljavaju antioksidativni efekat. Sadržaj ovih materija u izolatima zavisi od njihove koncentracije u polaznom biljnom materijalu ali i od načina/uslova ekstrakcije. Cilj našeg ispitivanja bio je da utvrdimo optimalne vrednosti procesnih parametara za izvođenje pritiskom ubrzane ekstrakcije iz herbe hajdučke trave (Millefolii herba), radi dobijanja ekstrakata bogatih antioksidativnim materijama, kao i da utvrdimo zavisnost koja postoji između procesnih parametara. Ekstrakcija je obavljena u Timatic mikro ekstraktoru. Takozvano vreme kompresije, vreme dekompresije i broj ciklusa kruženja ekstrakcionog sredstva predstavljaju nezavisno promenljive veličine, dok su sadržaj ukupnih flavonoida, tanina, ukupnih polifenola kao i indeks refrakcije, suvi ostatak, relativna gustina, sadržaj etanola i pH vrednost zavisno promenljive veličine. Vreme kompresije bilo je 1 odnosno 2 minuta, vrednost vremena dekompresije 1,3 i 2 min, dok su ekstrakcije obavljene uz minimalno 20 odnosno maksimalno 99 ciklusa. Kao ekstragens korišćen je 70% (V/V) etanol. Droga: ekstrakt odnos iznosio je 1:3. Radni pritisak kretao se u opsegu od 1 do 99 bar. Primenjen je 23 faktorski dizajn, odnosno urađeno je osam ekstrakcija herbe hajdučke trave. Statistička analiza eksperimentalnih rezultata izvršena je upotrebom softvera Design Expert 7.0. U cilju potpune optimizacije primenjena je multivarijantna metodologija upotrebom Derringer funkcije poželjnih odgovora. Proces optimizacije postupka ekstrakcije pokazao je da je za izradu ekstrakata herbe hajdučke trave bogatih sadržajem antioksidativnih materija (flavonoidi, tanini i polifenoli), potrebno podesiti vreme kompresije na najviši nivo (2,0 min), vreme dekompresije na najniži (1,30 min) i vršiti ekstrakciju uz maksimalni broj ciklusa (99).

Bioactivity assays on Carlina acaulis and C. acanthifolia root and herb extracts

AB  - The present study describes investigation on chemical profile and pharmacological properties of hydromethanolic extracts of Carlina acaulis and C. acanthifolia roots and herbs. In order to investigate the antioxidant properties several methods were applied: FRAP (Ferric Reducing Antioxidant Power) assay, TBA assay (Fe2+/ascorbic induced lipid peroxidation) and scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH radicals. In assessing anti-inflammatory activity the carrageenan-induced rat paw oedema test was used, with indomethacin as a reference drug. Anti-ulcer activity of the extracts was investigated using ethanol-induced stress ulcer in rats and ranitidine as a reference drug. Antimicrobial activity was tested on five bacterial strains and one strain of fungi using the agar diffusion method. Investigated extracts exhibited considerable dose-dependent antioxidant, anti-inflammatory and anti-ulcer activities. Extracts of herbs were more active than the corresponding root extracts. All extracts exhibited moderate activity against all tested microorganisms. Herb extracts of C. acaulis and C. acanthifolia, as well as their root extracts exhibited similar bioactivity, which could be explained with their similar chemical composition revealed by HPLC analysis. The pharmacological activities of investigated Carlina extracts demonstrated here will serve as the basis for their further investigations in order to estimate their impact on human health and nutrition.

AB  - Sea buckthorn (Hippophae rhamnoides L., Elaeagnaceae), is a thorny bush or small tree, native to Europe and Asia. Sea buckthorn fruit is rich in oil and carotenes which are found both in pericarp and seeds. Fruit and seed oils have significantly different fatty acid profiles and carotene levels. Previously reported data on the content of fatty acids and the carotenes in the oil are based on the fresh fruits. However, sea buckthorn fruits for traditional herbal use are available commonly in dry form. The goal of work was to analyze the content of fatty acids and carotenes in the fruit pericarp oil and seed oil obtained from dry sea buckthorn fruits. Physical and Chemical constants of the oils were also determined. It was found that the oil yield from pericarp and seeds were 18,8 and 7,4 respectively (expressed in g of oil per 100 g of dry plant material). The composition of fatty acids was determined by GC in the methyl ester form. The main fatty acids in pericarp oil were palmitic, palmitoleic, and oleic acids (35,2 %, 28,5 %, and 29,9 % respectively). Seed oil was, on the other hand, rich in oleic, linoleic and alpha-linolenic acids (23,7 %, 37,6 %, 20,5 % respectively). The content of carotenes in oil obtained from fresh sources varies extremely within different subspecies and it is influenced strongly by methods of oil isolation. The common reported levels of carotenes in pericarp and seed oil obtained from fresh fruits are 100-500mg/100g and 20-100 mg/100 g respectively. Our findings have significantly lower values then the average reported data, which points that dehydration of sea buckthorn berries influences negatively upon the content of carotenes, and then fatty oil content remains roughly the same as in fresh fruits. From medical point of view and dried fruits are a valuable source of polyunsaturated fatty acids and monounsaturated you have a favorable effect primarily on the cardiovascular system.
