AB - Teucrium montanum L. and T. capitatum L. are two plant species with overlapping distribution
in the Balkan Peninsula, especially in Croatia, where several occurrences of their putative hybrid
species T. × rohlenae have been recorded. Next-generation sequencing of both species and de novo
assembly was carried out resulting in 120 contigs for T. montanum and 1685 contigs for T. capitatum
assembled. The Dig-up primers pipeline was used for SSR mining of both assemblies, applying
different criteria that resulted in 112 SSR candidates for testing. A subset of 41 SSRs was selected and
after two rounds of testing, twelve SSRs were developed and characterized. A total of 232 alleles
were detected with 5 to 29 alleles per locus, based on the test sample. The genetic diversity analysis
of three Teucrium taxa from a single location revealed a higher level of diversity in T. montanum
than in T. capitatum with intermediate values for the hybrid species. The NeighborNet diagram and
genetic structure analysis grouped the taxa into two separate clusters, the first of which consisted
exclusively of T. montanum, while the other was composed of intermixed T. capitatum and T. × rohlenae
individuals. The availability of SSR markers for two Teucrium taxa will allow in-depth analysis
of genetic diversity and structure, as well as molecular identification of their putative hybrids in
the future.

veoma izazovnim u taksonomskom smislu. Vrsta Teucrium montanum pripada sekciji Polium i zajedno
godina u srednjem Pleistocenu. Teucrium montanum je poluodrveneli žbun, rasprostranjen u južnoj i
centralnoj Evropi, Maloj Aziji, i Alžiru u Severnoj Africi.
uglavnom smatraju njenim sinonimima.
rasprostranjena, uz veliki diverzitet staništa na kojima se javlja ispoljava i veliku morfološku
varijabilnos.
bliskim vrstama unutar sekcije Polium.
Analizom klimatskih parametara i prikupljenih
Filogenetski odnosi unutar populacija T. montanum sensu lato utvrđeni su AFLP metodom, dok su
genetički odnosi između populacija potencijalnih roditelja i hibrida urađena analizom mikrosatelita
(SSR).
Pet morfoloških grupa (montanum, pannonicum, skorpili, helianthemoides i
dok dve morfološke grupe (skadarensis i luteolum) poseduju specifičnu kombinaciju morfoloških
AFLP analiza
utvrdila je prisustvo dve genetičke grupe (A i B), dok je unutar morfološki vrlo varijabilne genetičke
grupe A utvrđeno prisutvo još šest genetičkih klada (cl.A1, cl.A2A, cl.A2B, cl.A3, cl.A4 i cl.A5), koje
su prostorno jasno razdvojene. Genetička diverzifikacija samo delimično odgovara morfološkoh
diverzifikaciji.
subsp., T. pannonicum subsp. pannonicum, T. pannonicum subsp. skorpili comb. et stat nova, T.
helianthemoides subsp. helianthemoides, T. helianthemoides subps. hirsutum comb. et stat nova, T.
AB - The genus Teucrium is paraphyletic and includes about 250 species, with the center of diversity in the
Mediterranean region. The often unclear boundaries between species and the presence of interspecific
hybridization make this genus very challenging from a taxonomic point of view. Teucrium montanum
belongs to section Polium and together with T. polium and T. capitatum, forms a phylogenetic lineage
that diverged about 3 million years ago in the middle Pleistocene. Teucrium montanum is a semi-woody
shrub, distributed in southern and central Europe, Asia Minor, and Algeria in northern Africa. This
species has a broad ecological valence and occurs in different vegetation types from coastal areas of the
Mediterranean to alpine and subalpine belts. The species is morphologically very variable, and a large
number of taxa have been described throughout its range, most of which are now considered synonyms.
In the Balkan Peninsula, the species T. montanum is widespread, exhibits great morphological variability,
and is found in a wide range of habitats. A large number of taxa, whose status is not completely resolved
and which are associated with the T. montanum sensu lato, have been recorded in the Balkan Peninsula.
Taking all this into account, the main aims of this doctoral dissertation are: to (1) describe the morpho-
anatomical, ecological, and genetic variability of T. montanum sensu lato in the Balkan Peninsula, (2)
determine the phylogeographic relationships within T. montanum sensu lato in the Balkan Peninsula, (3)
determine the status of unaccepted taxa associated with T. montanum sensu lato in the Balkan Peninsula,
and (4) to determine the interspecific hybridization of T. montanum sensu lato with closely related
species within the section Polium in the Balkan Peninsula. A total of 765 individuals from 60 populations
were analyzed, as well as 30 individuals from three populations of T. capitatum and 25 individuals with
morphological transitional characters between T. montanum and T. capitatum. Morphological and
anatomical preparations were measured using a standard morphometric method. The ecology of the
habitats of T. montanum populations in the Balkan Peninsula was determined by analyzing climatic
parameters and data collected at the localities. Phylogenetic relationships within populations of T.
montanum sensu lato were determined using the AFLP, while genetic relationships between potential
parents and hybrids were analyzed using microsatellites (SSR). Within T. montanum, the presence of
seven morphological groups was determined (montanum, skadarensis, pannonicum, skorpili, luteolum,
helianthemoides, and parnassicum), which can be distinguished based on morphological characters. Five
morphological groups (montanum, pannonicum, skorpili, helianthemoides, and parnassicum) correspond
to previously described taxa associated with T. montanum, while two morphological groups (skadarensis
and luteolum) have a specific combination of morphological traits that has not been observed before. The
identified morphological groups are more or less well geographically separated. The diversification of
the morphological groups corresponds only partially with the ecological diversification of the analyzed
habitats. The AFLP analysis revealed the presence of two genetic groups (A and B), while within the
morphologically highly variable genetic group A, the presence of six additional genetic clades was
detected (cl.A1, cl.A2A, cl.A2B, cl.A3, cl.A4, and cl.A5), which are spatially clearly separated. Genetic
diversification only partially corresponds to morphological diversification. In localities where
populations of T. montanum and T. capitatum grow sympatrically, individuals with transitional
morphological and genetic characteristics between them were recorded. Introgressions were detected
between the analyzed parents and hybrids, which were expressed to a different extent and in different
patterns at different localities. Based on the obtained morphological and genetic results, a new taxonomic
concept within T. montanum sensu lato was proposed, which includes three taxa at the species level (T.
montanum, T. pannonicum, and T. helianthemoides) and seven additional taxa at the subspecies level (T.
montanum subsp. montanum, T. montanum subsp. skadarensis nova subsp., T. pannonicum subsp.
pannonicum, T. pannonicum subsp. skorpili comb. et stat nova, T. helianthemoides subsp.
helianthemoides, T. helianthemoides subsp. hirsutum comb. et stat nova, T. helianthemoides subsp.
parnassicum comb. et stat nova). A new taxon, T. montanum subsp. skadarensis subsp. nova Zbiljić &
D. Lakušić, was described, which is distributed on the southern slopes of Prokletije on ultramafic rocks
in the vicinity of the city of Skadar in northern Albania.

AB - In this work, the overall morpho-anatomical variability of Teucrium montanum sensu lato on the Balkan Peninsula was described. A total of 60 populations were collected all over the Balkans. For each analysed individual, the morphological characters of the stem, leaves, and flowers, as well as the anatomical characters of leaves were measured. The habitats of each population were characterized by altitude zone, type of geological substrate, vegetation, and climate type. Basic descriptive statistics were calculated for each character state, and multivariate analyses were used to compare all assessed characters and groups. The results showed that the analysed populations exhibit high morphological variability and broad ecological preferences. Five of the seven morphologically defined groups fit the already described taxa (T. montanum subsp. montanum, T. montanum subsp. helianthemoides, T. pannonicum, T. skorpili, T. montanum var. parnassicum), whereas two groups are morphologically specific but did not fit the existing taxonomic concepts. The patterns of observed morphological variability only partially match the patterns of ecological differentiation of the habitats of the analysed populations. The occurrence of different morphological groups inhabiting similar habitats in the same biogeographic region suggests that the morpho-anatomical differentiation of T. montanum s.l. may be due to genetic or evolutionary factors.

abstract = "In this work, the overall morpho-anatomical variability of Teucrium montanum sensu lato on the Balkan Peninsula was described. AB - Teucrium montanum L. is a semi-woody evergreen small shrub distributed in southern and central Europe, Asia Minor, and Algeria in North Africa. The species has a very wide ecological valence occurring in various Mediterranean, sub-Mediterranean, continental as well as oro-Mediterranean and Alpine habitats. Due to its wide ecological valence and broad distribution, T. montanum displays high morphological variability, which is reflected in a large number of described taxa associated with it. In Europe, about thirty taxa have been described at specific and infraspecific levels, of which only T. montanum subsp. montanum and T. montanum subsp. helianthemoides (Adamović) Baden are accepted, while all the others are considered synonyms of T. montanum. The observed morphological diversity is reflected in the presence of six taxa described from the Balkan Peninsula: T. montanum subsp. helianthemoides, T. montanum var. hirsutum Boiss., T. montanum var. modestum Heldr., T. montanum var. parnassicum Čelak., T. pannonicum A. Kern., and T. skorpili Velen. During field research throughout the Balkans, seven morphologically more or less well-defined groups with relatively solid geographical differentiation were observed. Five of the seven groups fit the already described infraspecific taxa, while two groups are morphologically specific but do not correspond to the existing taxa. The aim of this study was to describe the overall morpho-anatomical variability of T. montanum sensu lato in the Balkans and to determine whether the seven identified groups can be distinguished based on morpho-anatomical characters. The most typical populations of these groups could be separated quite well based on the quantitative morphological traits. Multivariate analysis of all samples showed a solid separation of three groups and a significant number of individuals with intermediate positions. Other groups did not show statistical diversification but possess a specific combination of morphological characters by which they can be identified.

AB - Satureja kitaibelii Wierzb. ex Heuff. (Rtanj tea) has distinct morphology and distribution which separate it from highly polymorphic S. montana L. In this work, we investigated phenolic composition of S. kitaibelii and compared it to that of S. montana. Flowering aerial parts of four S. kitaibelii samples were collected in Serbia and Bulgaria and one commercial sample of Rtanj tea was obtained from Serbia. Three samples of S. montana were collected in North Macedonia and Serbia and the fourth one was cultivated sample. Herbs were extracted with 50% ethanol. Dry extract solutions were analyzed by liquid chromatography with UV and MS detection. Rosmarinic acid (RA) and clinopodic acid O (CAO) were quantified using RA as the external standard. All extracts were characterized by presence of phenolic acids, RA (16.5-47.9 mg/g) and CAO (1.6-29.3 mg/g, calculated as RA), and flavonoids. The extracts of S. kitaibelii distinguished by high contents of both RA and CAO, which were present in similar quantities within each extract. On the other hand, extracts of S. montana contained at least 3 times less CAO than RA. The obtained results further confirm distinct properties of S. kitaibelii and give additional arguments to its status as a separate species.

AB - Teucrium montanum L. is a semi-woody, evergreen small shrub, widely distributed in Europe occurring from the seacoast to the subalpine and alpine belts. The study aimed to investigate and compare the chemical composition of essential oil of aerial parts of T. montanum from 24 populations collected from West, Central, and South West Balkans. The essential oils were obtained by hydrodistillation and qualitative and quantitative analysis was performed by GC-FID and GC-MS. The dominant compounds were oxidized sesquiterpenes (24.83–85.75%), then sesquiterpene hydrocarbons (3.00–62.81%), and oxidized monoterpenes (0–27.78%). Cluster analysis revealed 10 clearly separated groups. First group (Žumberak) is completely separated and characterized by high germacrene-D-4-ol (63.17%), the second (Gjergjevice) by high shyobunol (51.23%), the third (Vratna) by high (dehydro)-sesquicineole-acetate (45.29%), the fourth (Ostrovice) by high epi-α-cadinol (36.80%), the fifth (Nera canyon) by high limonene (36.65%), sixth group (Orovica, Sićevo, Galičica) by high cis-sesquisabinene hydrate (18.73–29.05%), seventh group (Štrpce, Dobrilovina) by high γ-cadinene (13.43–23.06%), eighth group (Tara, Durmitor, Biokovo, Fužine) by moderate germacrene-D-4-ol (19.76–23.44%), and the ninth large group (Potoci, Lovćen, Orjen, Premantura, Vodice, Pletvar, Senj, Oštarije, Trebinje, Korita) by germacren D (6.15–33.87%). Such a high variability needs more research to define relationships among populations.

AB - Several natural hybrids of Teucrium montanum L. and T. polium L. have been described in certain parts of the area where these two species are sympatric. A new population with intermediate individuals that share morphological characteristics of both T. montanum and T. polium has been found in Croatia (Bisko near Trilj), and most likely represents a hybridogenous taxon. The aim of this study is to compare the morpho-anatomical and phytochemical characteristics of the two parental species and their putative hybrid, as well as to determine the most important distinguishing characters between these three groups. In order to describe the variability and significance of morpho-anatomical and phytochemical differentiation of the analysed groups several multivariate statistical analysis were conducted (PCA, CDA, DFA, UPGMA-clustering). All analyses confirm the separation of two species, T. polium and T. montanum, and reveal the intermediate position of the putative hybrid. In this way, data on morphological and anatomical analysis together with data on the composition of the essential oils support the assumption of the hybridogenous origin of the intermediate individuals. Based on descriptions and original indication of parental species of previously described hybrids T. × castrense Verg., T. × bogoutdinovae Melinkov and T. × rohlenae K.Malý, we found that the intermediate specimens collected in Croatia perfectly correspond to T. × rohlenae, a hybridogenous taxon discovered by Rohlena in the vicinity of Kotor in Montenegro, and thus prove the presence of a new natural hybrid in the flora of Croatia.

AB - Teucrium × rohlenae Maly is a natural hybrid between T. montanum and T.
polium. It was found in the surrounding of Kotor (Montenegro) by L. Čelakovski fil.
and reported by J. Rohlena as T montanum × polium in 1922. This hybrid is validly
described by K. Maly in 1951. After the original description, Teucrium × rohlenae
have not been reported in the Balkan Peninsula, but hybrids between T montanum and
T. polium have been cited in Italy (as Teucrium x castrense) and Moldova (as
Teucrium × bogoutdinovae). In sympatric population of T. montanum and T. polium
in the surrounding of Trilj near Split (Croatia), we found intermediate individuals with
potentially hybridogenic origin. The aim of the study was to analyse 44 morphoanatomical
characters of stem, leaf, inflorescens and calyx, and chemical composition
of volatile compounds of parental species and their potential hybrid Teucrium ×
rohlenae. The composition of hexane extracts of aerial parts was determined using
GC-FID and GC-MS methods. A detailed morpho-anatomical analysis has shown that
parental species are well differentiated, while potential hybrids have an intermediate
position. Volatiles of potential hybrid represented both species, main hydrocarbon
compounds were alike volatiles of T. polium while the terpene profile was more
similar to T. montanum terpenes.

AB - Teucrium montanum L. is widely distributed in the Balkan
Peninsula, extending from the sea coast to altitudes of over
2100 m a. s. l. with the highest number of occurrences in the
zone between 500 and 1000 m. The aim of the study was to
investigate the chemical composition of essential oil of aerial
parts of T. montanum from 14 different populations from Balkan Peninsula, ten from Serbia (SR-Trešnjica canyon, SR-Brdjanska gorge, SR-Goč, SR-Gornjak gorge, SR-Grza canyon,
SR-Jelašnica, SR-Kopaonik, SR-Maglič, SR-Rtanj, SR-Sićevačka gorge), two from Greece (GR-Olimp, GR-Ossa) and two
from Albania (AL-Deja, AL-Skadar). The essential oils were
obtained by hydrodistillation and qualitative and quantitative analysis was performed by GC-FID and GC-MS. The
aerial parts of T. montanum contained only small amounts
of oil (traces-0.7%), predominantly composed by sesquiterpene compounds (58.0-99.0%). One population from Albania
(Deja), two from Greece (Olimp, Ossa) and two from Serbia
(Sićevačka gorge, Trešnjica canyon) contained more than 90%
of sesquiterpenes. The composition of essential oils was quite
variable and the main compounds in almost all oils were germacrene D (trace-45.5%), sabinene (trace-23.1%), α-pinene
(trace-20.7%), limonene (trace-20.4%), (E)-caryophyllene (2.9-14.5%), γ-cadinene (trace-13.8%) and δ-cadinene
(trace-12.0%). The cluster analysis revealed the separation
of investigated essential oils to five clusters. The first cluster
(AL-Skadar, SR-Brdjanska gorge, SR-Gornjak gorge, SR-Grza canyon, SR-Goč) is characterized by high germacrene D
content (17.6-45.5%), the second (SR-Maglič, SR-Kopaonik,
SR-Sićevačka gorge) by high sesquiterpene alcohol shyobunol content (14.6-55.2%), the third (AL-Deja, SR-Jelašnica)
by high α-bisabolol content (31.2%-43.7%) and shyobunol
(3.2%) in population AL-Deja, the fourth (SR-Rtanj) by high
cis-sesquisabinene hydrate content (42.4%) and the fifth (GR-Olimp, GR-Ossa, SR-Trešnjica canyon) by high germecrene
D-4-ol (5.96%-61.9%), and β-eudesmol (14.9%) and epi-α-cadinol (10.2%) content in GR-Olimp population. Such a high
variability needs more research to define relationships among
populations.

(trace-20.7%), limonene (trace-20.4%), (E)-caryophyllene (2.9-14.5%), γ-cadinene (trace-13.8%) and δ-cadinene
(trace-12.0%). The cluster analysis revealed the separation
of investigated essential oils to five clusters. The first cluster
(AL-Skadar, SR-Brdjanska gorge, SR-Gornjak gorge, SR-Grza canyon, SR-Goč) is characterized by high germacrene D
content (17.6-45.5%), the second (SR-Maglič, SR-Kopaonik,
SR-Sićevačka gorge) by high sesquiterpene alcohol shyobunol content (14.6-55.2%), the third (AL-Deja, SR-Jelašnica)
by high α-bisabolol content (31.2%-43.7%) and shyobunol
(3.2%) in population AL-Deja, the fourth (SR-Rtanj) by high
cis-sesquisabinene hydrate content (42.4%) and the fifth (GR-Olimp, GR-Ossa, SR-Trešnjica canyon) by high germecrene
D-4-ol (5.96%-61.9%), and β-eudesmol (14.9%) and epi-α-cadinol (10.2%) content in GR-Olimp population. Such a high
variability needs more research to define relationships among
populations.",
