AB  - Introduction: Chokeberry is a plant belonging to the family Rosaceae which has a long history of edible and medicinal use. The aims of this study are: i) to investigate of HPTLC profile of the berry samples using HPTLC technique based metabolomics approach, ii) to determine the botanical origin of investigated chokeberry adulterations iii) to identify the most important botanical markers responsible for classification. Methods: A simple, rapid, and high-throughput planar chromatography (HPTLC) method was applied in combination with multivariate techniques for differentiating chokeberry samples and identification of the main metabolites of chokeberry and four common adulterants. Images of HPTLC chromatograms obtained using two different detections i.e. visible light and derivatization with natural products reagent were used as a dataset for multivariate analysis. Results: Principal component analysis and Orthogonal Partial Least Squares Discriminant Analysis confirmed the discrimination of five botanically different samples and recognized their main markers such as responsible for differences between chokeberry and four common adulterants. Conclusion: The obtained results showed that the HPTLC-based metabolomics approach can be a very reliable technique for the detection of chokeberry adulteration.

In vitro assessment of the prebiotic potential of selected plant extracts

AB  - In recent years, many preclinical studies have provided evidence that polyphenol compounds
and their metabolites exert prebiotic-like effects by selectively stimulating intestinal bacteria
growth and/or activity [1]. In this study, we investigated the chemical composition,
antioxidant capacity, and prebiotic potential of methanol extracts obtained from Prunus
spinosa L. (blackthorn) and Lycium ruthenicum Murray (black goji) fruits. Polyphenolic
profiles were characterized by using HPLC-DAD-ESI-MS and antioxidant capacity was
estimated with DPPH•, ABTS•+, and FRAP assays. The influence of plant extracts (0.312-5
mg/L) on the growth of three probiotic lactobacilli, probiotic yeast (Saccharomyces
boulardii), and two probiotic mixtures was examined. Studied plant extracts were
characterized by different phenolic compound patterns. Hydroxycinnamic acid derivatives,
quercetin glycosides and anthocyanins were detected in blackthorn extract, while black goji
extract was characterized by the presence of hydroxycinnamic acid derivatives and acylated
anthocyanins. The blackthorn fruit extract showed higher antiradical and reduction activity
than those obtained from black goji fruits. Both extracts have influenced the growth of all
tested probiotics in a concentration-dependent manner, especially of yeast, S. boulardii. The
black goji extract had more stimulatory effects, possibly due to higher anthocyanins content.
When the black goji extract was added to the growth medium at the concentration of 5
mg/mL, about 2-fold stimulation of the growth of S. boulardii was observed. In conclusion,
extracts of the black goji and blackthorn fruits promote the growth of probiotics in vitro.
Further studies are still needed to assess the prebiotic potential of these plant extracts.

AB  - Lycium ruthenicum Murray (Solanaceae), also known as black goji or black wolfberry, has
been used for centuries in traditional medicine and nutrition in Asian countries [1]. In recent
years, black goji berries have been gaining attention as one of the most valuable sources of
anthocyanins with many health-promoting effects [2]. Therefore, this study aimed to
investigate the phytochemical composition and in vitro biological properties of black goji
berry cultivated in southeastern Serbia. The total phenolics, flavonoids, anthocyanins, and
tannins content were determined spectrophotometrically. HPLC-DAD-ESI-MS analysis was
used to identify the phenolic compounds. Methanol extract from black goji berry was
screened for in vitro antioxidant (DPPH∙, ABTS∙+, FRAP, CUPRAC, and β-carotene bleaching
methods), antimicrobial, and hypoglycemic (α-amylase, α-glucosidase) potential.
Phytochemical screening of the extract confirmed the dominant presence of anthocyanins and
hydroxycinnamic acid derivatives, with petunidin 3-p-cumaroylrutinoside-5 glucoside as the
main phenolic compound [3]. The total phenolic content was 18.13 ± 0.15 mg gallic acid
equivalents/g freeze-dried sample. A high correlation is found between the total phenolic
content and the antioxidant activities using different in vitro antioxidant assays. The
antimicrobial activity of the black goji berry extract against eight laboratory control strains
was not pronounced (MIC > 2 mg/ml). The IC50 values were 6.55 ± 0.38 mg/mL and 7.37 ±
0.02 mg/ml for α-amylase, and α-glucosidase inhibition activity, respectively. Overall,
obtained results suggest that the black goji berry cultivated in Serbia should be considered a
valuable source of bioactive compounds for further use in the food, nutraceutical, and
cosmetic industries.

Mahonia aquifolium Extracts Promote Doxorubicin Effects against Lung Adenocarcinoma Cells In Vitro

AB  - Mahonia aquifolium and its secondary metabolites have been shown to have anticancer potential. We performed MTT, scratch, and colony formation assays; analyzed cell cycle phase distribution and doxorubicin uptake and retention with flow cytometry; and detected alterations in the expression of genes involved in the formation of cell-cell interactions and migration using quantitative real-time PCR following treatment of lung adenocarcinoma cells with doxorubicin, M. aquifolium extracts, or their combination. MTT assay results suggested strong synergistic effects of the combined treatments, and their application led to an increase in cell numbers in the subG1 phase of the cell cycle. Both extracts were shown to prolong doxorubicin retention time in cancer cells, while the application of doxorubicin/extract combination led to a decrease in MMP9 expression. Furthermore, cells treated with doxorubicin/extract combinations were shown to have lower migratory and colony formation potentials than untreated cells or cells treated with doxorubicin alone. The obtained results suggest that nontoxic M. aquifolium extracts can enhance the activity of doxorubicin, thus potentially allowing the application of lower doxorubicin doses in vivo, which may decrease its toxic effects in normal tissues.

Methods: Fractions were made from dichloromethane: methanol (1:1) extract of Fucus spiralis: petroleum-ether, ethyl-acetate and n-butanol. Extract and fractions were screened for in vitro cytotoxicity by MTT assay against human cervical adenocarcinoma (HeLa), colorectal adenocarcinoma (LS-174T), lung carcinoma (A549), and normal human lung fibroblasts (MRC-5). Cell cycle distribution of the HeLa cells was evaluated using flow cytometry. Acridine orange (AO)-ethidium bromide (EB) staining was used to assess morphological changes of HeLa cells under fluorescence microscope. Anti-migration and anti-angiogenic properties were investigated using scratch and tube formation assays against human endothelium-derived permanent EA.hy926 cell line. Antidiabetic activity was tested using anti-α-glucosidase assay. Antimicrobial effect was tested using micro- dilution method. Results: Petroleum-ether fraction оf Fucus spiralis rich in fatty acids exerted the highest cytotoxicity against HeLa cells. Ethyl-acetate and petroleum-ether fractions induced the highest accumulation of the HeLa cells in sub-G1 and G2/M phases. Extract and fractions showed proapoptotic effect on HeLa cells under fluorescent microscope. They exhibited antimigratory and antiangiogenic effects in vitro. IC50 value for α-glucosidase inhibitory activity was much stronger than standard acarbose. n-Butanol fraction exerted the highest antibacterial and antifungal activity. Conclusions: The investigation of various biological activities of the extract and fractions obtained from Fucus spiralis may suggest a promising anticancer and pharmacological potential of this edible macroalga.

AB  - A comparative analysis of the phenolic compounds in the 70% EtOH extracts of Hypericum acutum, H. androsaemum, H. barbatum, H hirsutum, H maculatum, and H richeri has been carried out using high-performance liquid chromatography coupled with photodiode array UV detection and high resolution TOF mass spectrometry. Quercetin, astilbin, I3,II8-biapigenin, orientin, 2"-O-acetylorientin, three phenolcarboxylic acids, and eight flavonols 3-O-glycosides were identified in the extracts on the basis of their on-line UV spectra, accurate mass spectral data, and in comparison of retention times with those from the standards. Fingerprint analysis of the extracts revealed significant differences in the qualitative and quantitative chemical composition of the studied species. Antioxidant assays with various reaction mechanisms were used including ferric reducing antioxidant power (FRAP) assay, DPPH, ABTS, superoxide anion radical scavenging capacity and inhibition of liposome peroxidation induced by Fe2+. The most potent were extracts of H acutum and H maculatum indicating this Hypericum species interesting for further research aimed as a potentially new source of biologically active compounds.

Optimization of polyphenols extraction from dried chokeberry using maceration as traditional technique

AB  - Traditional maceration method was used for the extraction of polyphenols from chokeberry (Aronia melanocarpa) dried fruit, and the effects of several extraction parameters on the total phenolics and anthocyanins contents were studied. Various solvents, particle size, solid solvent ratio and extraction time have been investigated as independent variables in two level factorial design. Among examined variables, time was not statistically important factor for the extraction of polyphenols. The optimal extraction conditions were maceration of 0.75 mm size berries by 50% ethanol, with solid solvent ratio of 1:20, and predicted values were 27.7 mg GAE/g for total phenolics and 0.27% for total anthocyanins. Under selected conditions, the experimental total phenolics were 27.8 mg GAE/g, and total anthocyanins were 0.27%, which is in agreement with the predicted values. In addition, a complementary quantitative analysis of individual phenolic compounds was performed using HPLC method. The study indicated that maceration was effective and simple technique for the extraction of bioactive compounds from chokeberry fruit.

The constituents of the extracts were quantified using HPLC-DAD (high-performance liquid chromatography-diode array detector). Isolation of the compounds from n-BuOH and EtOAc extracts was conducted by column chromatography. Antioxidant capacity of the extracts was evaluated by superoxide radical and ABTS root+ scavenging assays and inhibition of lipid peroxidation. The EtOAc extract of H. richeri was the most potent antioxidant in all experiments. Free radical scavenging compounds were identified by HPLC-DPPH root (high-performance liquid chromatography-2,2-diphenyl-1-picrylhydrazyl) post-column derivatisation method, where all phenolic compounds except I3,II8-biapigenin exhibited radical scavenging properties. This is the first report of the presence of myricetin-3-O-rutinoside, myricetin-3-O-galactoside, myricetin-3-O-glucoside, 5-O-caffeoylquinic acid and I3,II8-biapigenin in H. richeri.

AB  - Oil extracts of flowering tops of Hypericum richeri Vill. prepared in three different ways were evaluated for chemical composition, and anti-inflammatory and gastroprotective activities. An HPLC method was developed for determination of two dominant flavonoids, quercetin and I3,II8-biapigenin. The carrageenan-induced rat paw edema test was used for screening the anti-inflammatory activity, while indomethacin-induced rat gastric mucosa damage test was used for evaluation of gastroprotective activity. The oil extract prepared by maceration with 96% ethanol, followed by extraction with sunflower oil by heating on a water bath, exhibited the highest anti-inflammatory (38.4 %) and gastroprotective activities (gastric damage score of 0.9). The same oil extract had the highest content of quercetin (49 mu g/mL) and I3,II8-biapigenin (60 mu g/mL). These results approve the usage of oil extracts of H. richeri as an anti-inflammatory and gastroprotective agent.

AB  - Oil extracts of Hypericum perforatum L. (Oleum Hyperici) were prepared in three different ways according to the prescriptions from traditional medicine. Variability of constituents and biological activity were evaluated in the obtained oil extracts. The carrageenan-induced rat paw edema test has been used for screening the antiinflammatory activity, while the indomethacin-induced rat gastric mucosa damage test was used for evaluation of gastroprotective activity. All examined oil extracts possessed antiinflammatory and gastroprotective activity. Among them, the oil extract prepared by maceration with 96% ethanol, followed by extraction with sunflower oil by heating on a water bath (extract 2), in a dose of 1.25 mL/kg p.o., exhibited the highest antiinflammatory effect (95.24 +/- 11.66%) and gastroprotective activity (gastric damage score of 0.21 +/- 0.12). The same oil extract had the highest content of quercetin and I3,II8-biapigenin (1.29 +/- 9 mu g/mL and 52 +/- 4 mu g/mL, respectively). Quercetin and I3,II8-biapigenin exhibited antiinflammatory activity similar to those of indomethacin as well as significant gastroprotective activity. The results provide evidence for the usage of Oleum Hyperici as an antiinflammatory and gastroprotective agent, Which has been based previously only on ethnopharmacological claims. Copyright
