AB  - Purpose: Sea macroalgae are an important source of biologically highly valuable compounds. The main aim of this study was to investigate the in vitro anticancer properties and chemical composition of the dichloromethane-methanol extract and three fractions of the Fucus spiralis from coastline of Morocco. Methods: Fractions were made from dichloromethane: methanol (1:1) extract of Fucus spiralis: petroleum-ether, ethyl-acetate and n-butanol. Extract and fractions were screened for in vitro cytotoxicity by MTT assay against human cervical adenocarcinoma (HeLa), colorectal adenocarcinoma (LS-174T), lung carcinoma (A549), and normal human lung fibroblasts (MRC-5). Cell cycle distribution of the HeLa cells was evaluated using flow cytometry. Acridine orange (AO)-ethidium bromide (EB) staining was used to assess morphological changes of HeLa cells under fluorescence microscope. Anti-migration and anti-angiogenic properties were investigated using scratch and tube formation assays against human endothelium-derived permanent EA.hy926 cell line. Antidiabetic activity was tested using anti-α-glucosidase assay. Antimicrobial effect was tested using micro- dilution method. Results: Petroleum-ether fraction оf Fucus spiralis rich in fatty acids exerted the highest cytotoxicity against HeLa cells. Ethyl-acetate and petroleum-ether fractions induced the highest accumulation of the HeLa cells in sub-G1 and G2/M phases. Extract and fractions showed proapoptotic effect on HeLa cells under fluorescent microscope. They exhibited antimigratory and antiangiogenic effects in vitro. IC50 value for α-glucosidase inhibitory activity was much stronger than standard acarbose. n-Butanol fraction exerted the highest antibacterial and antifungal activity. Conclusions: The investigation of various biological activities of the extract and fractions obtained from Fucus spiralis may suggest a promising anticancer and pharmacological potential of this edible macroalga.

AB  - Three seaweeds (Halimeda tuna, Codium bursa and Cystoseira barbata) and one seagrass (Cymodocea nodosa) were collected from the Coast of Montenegro, Gulf of Boka Kotorska and their chemical analysis was performed. In seagrass C. nodosa, three phenolic compounds were identified (diosmetin 7-sulfate, caftaric and coutaric acid). The content of β-glucan, fatty acids, sterols and micro- and macro-elements were investigated among all samples. The highest content of β-glucan was detected in C. nodosa seagrass (13.04±0.42 g/100 g). The highest polyunsaturated fatty acids (PUFAs) level was reported in C. barbata, the brown alga (7.157 mg/g), which also had the significant sterol content (fucosterol, 21.76±0.1 μg/g). Green algae, C. bursa and H. tuna, showed the highest level of sterols (β-sitosterol, 95.21±0.16 μg/g and 73.90±0.08 μg/g, respectively). H. tuna had the highest content of calcium (Ca) in amount of 55125 μg/g. In C. bursa, C. barbata and C. nodosa, the Na/K ratio was low (0.43, 0.46 and 0.69, respectively).

Cytotoxic and antimicrobial activity of selected Hieracium L. extracts and isolated sesquiterpene lactones

AB  - Cytotoxic and antimicrobial activities were investigated for MeOH extracts of flowering aerial parts of Hieracium calophyllum R. Uechtr. (CAL), H. coloriscapum Rohlena & Zahn (COL), H. pseudoschenkii (Rohlena & Zahn) Niketić (PSE), H. valdepilosum Vill. s.l. (VAL) and H. glabratum Willd. (GLA), their two isolated sesquiterpene lactones 8-epiixerisamine A and crepiside E, as well as for CH2Cl2 extract of flowering aerial parts of H. scheppigianum Freyn (SCH). Crepiside E (IC50 = 46.00 μg/mL), COL (IC50 = 152.28 μg/mL) and SCH (IC50 = 148.12 μg/mL) showed 
the highest cytotoxic activity against HeLa cells in MTT test. The cytotoxic activity against LS174, A549, as well as against normal MRC-5 cells was significantly weaker. In microdilution test, crepiside E and 8-epiixerisamine A exhibited noteworthy antifungal effect against Aspergillus ochraceus, Penicillium funiculosum, Candida albicans and C. krusei (MIC = 0.15 - 0.4 mg/mL, MFC = 0.3 - 0.8 mg/mL). Among the extracts, the best antibacterial activity was shown by SCH and CAL against Pseudomonas aeruginosa (MIC = 1.68 and 2.52 mg/mL, MBC = 3.36 and 5.04 mg/mL), and the highest antifungal activity by SCH and VAL against C. albicans (MIC = 2.48 and 2.52 mg/mL, MFC = 4.96 and 5.04 mg/mL).

is also coordinated to metal ions. The complexes were characterized by ele-
mental analysis (C, H, N), FTIR and UV–Vis spectroscopy, molar conductivity
determination and magnetic susceptibility measurement at room temperature.
The analytical data of the complexes show the formation of binuclear
[Ni2(C9H8O4)tpmc](ClO4)2·4H2O and tetranuclear [Zn4(C9H8O4)(tpmc)2](ClO4)6·
·CH3CN·KClO 4·4H2O complexes. In tetranuclear Zn(II) complex bicyclic
dicarboxylate ligand is most likely to be bridge coordinated, and in binuclear
Ni(II) complex it is coordinated in a combined bridged manner with chelate
rings formation. In both complexes macrocyclic ligand was exo coordinated,
out of cyclam ring and adopts a boat conformation. The Zn(II) complex is one
of the rare tetranuclear Zn(II)-tpmc complexes with carboxylate ion bridging
two Zn2tpmc units. The complexes were tested for antibacterial activity against
Gram-positive bacteria Staphylococcus aureus (ATCC 25923) and Bacillus
subtilis (ATCC 6633), Gram-negative bacterium Escherichia coli (ATCC
25922), and yeast Candida albicans (ATCC 10231), and were screened for
antiproliferative activity against human cervix adenocarcinoma (HeLa) and
human myelogenous leukemia (K562) cell lines.

координације. Други начини мостног везивања дикарбоксилата не могу бити у потпуности искључени. У оба комплекса макроциклични лиганд је егзо координован, изван
цикламовог прстена и заузима конформацију лађе. Добијени Zn(II) кoмплекс је један од
ретких тетрануклеарних Zn(II)–tpmc комплекса са карбоксилато лигандом у мосту
између две Zn2tpmc јединице. Добијени комплекси су тестирани на антибактеријску
активност према Грам-позитивним бактеријама Staphylococcus aureus (ATCC 25923) и
Bacillus subtilis (ATCC 6633), Грам-негативној бактерији Escherichia coli (ATCC 25922) и
квасцу Candida albicans (ATCC 10231). Испитивана је њихова антипролиферативна
активност на хуманим малигним ћелијским линијама: цервикалног аденокарцинома

Chemical and pharmacological characterisation of 9 taxa of the genus Heracleum L. (Apiaceae), autochthonous for Southeastern Europe

AB  - Uvod: Predmet istraživanja je devet taksona roda Heracleum (Apiaceae): sedam iz grupe 
H. sphondylium (H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii i H. verticillatum), i H. orphanidis, svi iz (tipske) sect. Heracleum, kao i H. austriacum subsp. siifolium iz sect. Wendia.
Rezultati i diskusija: Gasnom hromatografijom sa FID i MS detekcijom analiziran je sastav etarskih ulja (ukupno 59 uzoraka) izolovanih destilacijom vodenom parom iz podzemnih organa, listova, cvasti i plodova. U uljima podzemnih organa dominirali su monoterpeni i/ili fenilpropani (taksoni grupe H. sphondylium), ili (Z)-falkarinol (H. orphanidis i H. austriacum); u uljima plodova alifatični estri oktilacetat (sect. Heracleum) ili oktilheksanoat (H. austriacum), a u uljima listova i cvasti alifatični estri (H. orphanidis) ili seskviterpeni i/ili fenilpropani. Antimikrobna aktivnost 30 odabranih uzoraka etarskih ulja ispitivana je mikrodilucionom metodom na po osam sojeva bakterija i mikromiceta (u slučaju 17 uzoraka ulja MIK vrednosti su prema barem jednom mikroorganizmu bile niže ili bliske 100 µg/mL); selektivna citotoksičnost 26 uzoraka ulja MTT testom na tumorske HeLa, LS174 i A549 ćelije (u slučaju 13 uzoraka ulja IC50 vrednosti su prema barem jednoj ćelijskoj liniji bile niže ili bliske 30 µg/mL; ulja uglavnom nisu inhibirala rast normalnih MRC-5 ćelija). Ukupno 8 od ispitivanih 14 uzoraka ulja pokazalo je slab anti-DPPH potencijal. Sastav aktivnih zona u TLC-DPPH testu ispitan je gasnom hromatografijom (dominirali su oksidovani terpeni i fenilpropani). S obzirom da je u pojedinim uljima utvrđeno prisustvo furanokumarina koji mogu delovati fototoksično, definisan je bezbednosni profil ulja prema preporuci Evropske agencije za lekove (utvrđeno je da dnevni unos 1,94-15,68 mL ulja ne doprinosi značajno ukupnom riziku, koji postoji kao posledica unosa izvesnih količina furanokumarina hranom).
U dihlormetanskim ekstraktima podzemnih organa i plodova svih taksona identifikovano je ukupno 12 furanokumarina (LC-MS metodom pomoću standardnih supstanci ili na osnovu UV, MS, 1H i ROESY NMR spektara), od kojih su dominantni bili pimpinelin, bergapten, biakangelikol, heraklenin i/ili imperatorin. U uljastim supernatantima dihlormetanskih ekstrakata plodova gasnom hromatografijom (GC-FID i GC-MS) analizirane su masne kiseline (dominantna je bila petroselinska), steroli (najzastupljeniji je bio β-sitosterol) i triterpeni. 
Hemosistematski značaj komponenti etarskih ulja (u slučaju podzemnih organa i plodova i komponenti njihovih headspace frakcija) i furanokumarina utvrđen je multivarijantnom statističkom analizom (PCA, nMDS i UPGMA).
Zaključak: Sprovedena hemijska karakterizacija značajno je doprinela boljem poznavanju kompozicije sekundarnih i primarnih metabolita odabranih predstavnika sekcije Heracleum, dok su ispitivanja H. austriacum subsp. siifolium iz sekcije Wendia sprovedena po prvi put. Utvrđeno je da su ispitivani taksoni potencijalno novi biljni izvori jedinjenja značajnih za farmaceutsku, kozmetičku, prehrambenu ili hemijsku industriju. Rezultati ispitivanja farmakoloških aktivnosti etarskih ulja ukazali su na opravdanost daljih istraživanja ovih izolata kao potencijalno novih lekovitih biljnih sirovina, uz uslov poštovanja ustanovljenih maksimalnih dozvoljenih dnevnih unosa za ona etarska ulja u kojima su detektovani furanokumarini.

Phenolic constituents, antioxidant, α-amylase and α-glucosidase inhibitory activities of Pyrus × velenovskyi bark

AB  - Pyrus × velenovskyi Dostálek (Rosaceae) [P. pyraster (L.) Burgsd. × P. spinosa Forssk.] is a deciduous tree up to 5 m height. Leaf-blades 2.2–4.8 × 1.3–2.7 cm, ± glabrate, ellip tic, gradually narrowed to a rather long petiole. Th is natural hybrid plant was described from Bulgaria, and has been also found in Serbia. The objective of this work was to investigate the phenolic profile, as well as in vitro antioxidant, α-amylase and α-glucosidase inhibitory activities of dried methanol ex tract from the bark of this tree. The plant material was collected in eastern Serbia (Jelašnička Klisura gorge). In the dried bark, the contents of different classes of phenolics were spectrophotometrically determined: total polyphenols (10.36%), tannins (8.78%), procyanidins (4.21%) and phenolic glycosides (3.54%). After pre-extraction with dichloromethane, powdered bark was extracted with methanol by bimaceration at room temperature. In obtained dried methanol extract, using aforementioned spectrophotometric tests, the contents of total polyphenols (33.42%) and tannins (21.55%) were determined, and by HPLC, arbutin, chlorogenic acid, catechin and procyanidin B2 were identifi ed. Its antioxidant activity, i.e. ferric reducing capacity (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH radical scavenging ability, as well as the inhibition of the enzymes α-amylase and α-glucosidase were assessed using corresponding colorimetric assays. Tested dried methanol extract exhibited signifi cant DPPH and OH radical scavenging abilities (SC50 = 6.85 and 12.21 μg/mL, respectively), and ferric reducing capacity (10.74 mmol Fe (II)/g of dried extract), as well as the inhibition of α-amylase (IC50 = 11.4 μg/mL) and α-glucosidase (IC50 = 5.48 μg/mL).

abstract = "Pyrus × velenovskyi Dostálek (Rosaceae) [P. pyraster (L.) Burgsd. × P. spinosa Forssk.] is a deciduous tree up to 5 m height. Leaf-blades 2.2–4.8 × 1.3–2.7 cm, ± glabrate, ellip tic, gradually narrowed to a rather long petiole. Th is natural hybrid plant was described from Bulgaria, and has been also found in Serbia. The objective of this work was to investigate the phenolic profile, as well as in vitro antioxidant, α-amylase and α-glucosidase inhibitory activities of dried methanol ex tract from the bark of this tree. The plant material was collected in eastern Serbia (Jelašnička Klisura gorge). In the dried bark, the contents of different classes of phenolics were spectrophotometrically determined: total polyphenols (10.36%), tannins (8.78%), procyanidins (4.21%) and phenolic glycosides (3.54%). After pre-extraction with dichloromethane, powdered bark was extracted with methanol by bimaceration at room temperature. In obtained dried methanol extract, using aforementioned spectrophotometric tests, the contents of total polyphenols (33.42%) and tannins (21.55%) were determined, and by HPLC, arbutin, chlorogenic acid, catechin and procyanidin B2 were identifi ed. Its antioxidant activity, i.e. ferric reducing capacity (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH radical scavenging ability, as well as the inhibition of the enzymes α-amylase and α-glucosidase were assessed using corresponding colorimetric assays. Tested dried methanol extract exhibited signifi cant DPPH and OH radical scavenging abilities (SC50 = 6.85 and 12.21 μg/mL, respectively), and ferric reducing capacity (10.74 mmol Fe (II)/g of dried extract), as well as the inhibition of α-amylase (IC50 = 11.4 μg/mL) and α-glucosidase (IC50 = 5.48 μg/mL).",

Ispitivanja na endotelijalnim EA.hy926 ćelijama pokazala su antiangiogenetski i antimigratorni efekat ekstrakata.Primena tretmana doksorubicin/ekstrakt ukazala je na izraženo sinergističko dejstvo ekstrakta sa doksorubicinom na A549 ćelijama karcinoma pluća. Ekstrakti povećavaju zadržavanje leka u ćeliji i pojačavaju efekat doksorubicina na smanjenje migracije A549 ćelija i njihovo grupisanje u kolonije. Antimetastatski potencijal kombinacije doksorubicin/ekstrakt potvrđen je analizom ekspresije gena odgovornih za invazivnost.Oba ekstrakta ispoljavaju antimikrobni efekat, koji je najizraženiji na bakterijskoj vrsti Staphylococcus aureus. Ekstrakti dovode do smanjenja produkcije biofilma i glikokaliksa. Sa antibioticima i antimikoticima ekstrakti su delovali sinergistički.Hemijskom analizom potvrđeno je da su za aktivnost ekstrakata M. aquifolium odgovorni alkaloidi tipa berberina. Na intenzitet citotoksičnosti najviše utiču berberin, palmatin i berbamin.Rezultati su pokazali da M. aquifolium može naći svoje mesto u terapiji malignih ili infektivnih oboljenja, pojedinačno i u kombinaciji sa postojećim lekovima.
AB  - The goal of our research was investigation of antitumor and antimicrobial activities of Mahonia aquifolium extracts and it was based on its use in traditional medicine. Water and ethanol extract exerted selective cytotoxic activity on examined malignant cell lines, and the best activity was observed against cervical adenocarcinoma, HeLa, cells. Fluorescence microscopy and cell cycle analysis showed that both extracts induce apoptosis in HeLa cells. Experiments with caspase inhibitors showed that extracts induce a caspase-dependent apoptosis through extrinsic pathway. Both extracts inhibit the migration of EA.hy926 cells, and exerted anti-angiogenic effect.The investigation of the doxorubicine/extract treatment showed synergistic effect of extracts with doxorubicine against lung carcinoma A549, cells. Extracts were shown to prolong doxorubicin retention time in A549 cells. A549 cells treated with doxorubicin/extract were shown to have lower migratory and colony formation potentials than the cells treated with doxorubicin alone. Antimetastatic effect of the treatment was confirmed with analysis of expression of genes responsible for malignant cells invasiveness.Extracts exerted antimicrobial effect. The best effect was observed against Staphylococcus aureus. Extracts showed potential to inhibit production of biofilm and glycocalycs. The use of extracts with antibiotics and antimycotics showed synergistic effect.Chemical analysis confirmed that berberine type alkaloids are the most responsible for activities of M. aquifolium. The compounds with the highest cytotoxic activity are: berberine, palmatine, and berbamine.The results of our research suggested that M. aquifolium may be used in therapy of malignant or infective diseases, alone or in combination with other drugs.

AB  - The aim of the present study was to reveal the potential biological activities of the dichlorometane: methanol (1:1) dry extract (DME) and the nutritional value of selected seaweeds: Cymodocea nodosa, Halimeda tuna, Cystoseira barbata and Codium bursa, collected from the Adriatic coast, Montenegro. We assessed the chemical composition and several biological activities such as: anti-a-glucosidase, antitumour, antimicrobial and antioxidative activity. H. tuna had the best cytotoxic activity against human colon carcinoma cell line, LS174 (IC50 = 17.92 +/- 1.54 mu g/mL). C. nodosa demonstrated strong cytotoxicity against human adenocarcinoma cell line, HeLa (IC50 = 13.28 +/- 0.39 mu g/mL) and human chronic myelogenous leukaemia cell line, K562 (IC50 = 19.64 +/- 1.55 mu g/mL). C. barbata had the best anti a-glucosidase (IC50 = 9.98 +/- 3.34 mu g/mL) and antimicrobial activity (minimal inhibitory concentration of 100 mu g/mL) for Staphylococcus aureus and Bacillus subtilis. C. bursa showed the highest nutritional value (490.4 kcal).

Edible wild plant Heracleum pyrenaicum subsp orsinii as a potential new source of bioactive essential oils

AB  - Many Heracleum L. taxa (Apiaceae) are used as food and spices, and in traditional medicine. In this work, the chemical composition of Heracleum pyrenaicum subsp. orsinii (Guss.) F. Pedrotti and Pignatti root, leaf and fruit essential oils, their antimicrobial activity and cytotoxic effect on malignant and normal cells were investigated for the first time. The composition of the oils was analyzed by GC and GC-MS. Monoterpenes prevailed in the root oil, with beta-pinene (38.6%) being dominant, while in the leaf oil, sesquiterpenes, mostly (E)-nerolidol (20.5%) and (E)-caryophyllene (17.0%), were the most abundant constituents. The fruit oil contained the majority of aliphatic esters, mainly octyl acetate (36.8%) and octyl hexanoate (22.1%). The antimicrobial activity was determined by microdilution method against eight bacteria and eight fungi (standard strains, clinical or food isolates). The best antibacterial activity, better than the activity of ampicillin, was shown by the root oil against Salmonella typhimurium, Escherichia coli and Pseudomonas aeruginosa. The strongest antifungal activity, stronger than the activity of ketoconazole, was exhibited by the leaf and root oils against Trichoderma viride, and by the root oil against Aspergillus ochraceus. The cytotoxic effect of the oils, determined by MTT test, was prominent against malignant HeLa, LS174 and A549 cells (IC50 = 6.49-14.56 µg/mL). On the other hand, the oils did not show toxicity against normal MRC-5 cells at tested concentrations (IC50 > 200.00 µg/mL). It can be concluded that investigated H. pyrenaicum subsp. orsinii oils represent potential new raw materials for food and pharmaceutical industry.

AB  - Although some widespread, native cow parsnips (Heracleum L. spp., Apiaceae) had broad medicinal and culinary applications throughout history, the knowledge about their volatile constituents is insufficient. This work investigates the composition and bioactivities of H. sphondylium L. (HSPH), H. sibiricum L. (HSIB) and H. montanum Schleich. ex Gaudin (HMON) essential oils. The composition was tested by GC and GC-MS. (Z)-β-Ocimene was the most abundant in HSPH (28.9%) and HMON (20.4%) root oils, while in HSIB root oil, β-pinene (26.2%), methyl eugenol (22.3%) and elemicin (25.6%) prevailed. Leaf and flower oils were dominated by various sesquiterpenes (germacrene D, β-sesquiphellandrene, (E)-β-farnesene and/or (E)-caryophyllene) and/or phenylpropanoids (apiole, methyl eugenol, elemicin and/or (Z)-isoelemicin). Octyl acetate (57.5-67.1%) was the main constituent of all fruit oils. The antimicrobial activity was screened by a microdilution method against eight bacteria and eight fungi. The strongest antimicrobial effect, in several cases better than the activity of antibiotics, was shown by HSPH (MICs = 0.12-3.30 mg mL-1) and HMON (MICs = 0.10-1.30 mg mL-1) flower oils against bacteria, and HSIB fruit oil against fungi (MICs = 0.15-0.40 mg mL-1). The MTT test revealed that the oils were not or weakly cytotoxic against human malignant HeLa, LS174 and/or A549 cells (except HSPH root oil; IC50 = 5.72-24.31 μg mL-1) and that tested oils were not toxic against human normal MRC-5 cells (at 200.00 μg mL-1). Significant activity observed against microorganisms that are the common cause of foodborne diseases, food contamination and/or hospital-acquired infections justifies certain traditional uses of the investigated plants and represents a good basis for further research of these Heracleum oils.

AB  - In this work, the chemical composition, antimicrobial and cytotoxic activity of Heracleum verticillatum Pančic and H. ternatum Velen. root, leaf, and fruit essential oils were investigated. The composition was analyzed by GC and GC/MS. Heracleum verticillatum and H. ternatum root oils were dominated by monoterpenes, mostly β-pinene (23.5% and 47.3%, respectively). Heracleum verticillatum leaf oil was characterized by monoterpenes, mainly limonene (20.3%), and sesquiterpenes, mostly (E)-caryophyllene (19.1%), while H. ternatum leaf oil by the high percentage of phenylpropanoids, with (Z)-isoelemicin (35.1%) being dominant constituent. Both fruit oils contained the majority of aliphatic esters, mostly octyl acetate (42.3% in H. verticillatum oil and 49.0% in H. ternatum oil). The antimicrobial activity of the oils was determined by microdilution method against eight bacterial and eight fungal strains. The strongest effect was exhibited by H. verticillatum root oil, particularly against Staphylococcus aureus, Salmonella typhimurium (MICs = 0.14 mg/ml, MBCs = 0.28 mg/ml), and Trichoderma viride (MIC = 0.05 mg/ml, MFC = 0.11 mg/ml). Cytotoxic effect was determined by MTT test against malignant HeLa, LS174, and A549 cells (IC50 = 5.9 - 146.0 μg/ml), and against normal MRC-5 cells (IC50> 120.1 μg/ml). The best effect was exhibited by H. verticillatum root oil on A549 cells (IC50= 5.9 μg/ml), and H. ternatum root oil against LS174 cells (IC50= 6.7 μg/ml).

In vitro assessment of antiproliferative action selectivity of dietary isothiocyanates for tumor versus normal human cells

AB  - Background/Aim. Numerous epidemiological studies have shown beneficial effects of cruciferous vegetables consumption in cancer chemoprevention. Biologically active compounds of different Brassicaceae species with antitumor potential are isothiocyanates, present in the form of their precursors - glucosinolates. The aim of this study was to determine the selectivity of antiproliferative action of dietary isothiocyanates for malignant versus normal cells. Methods. Antiproliferative activity of three isothiocyanates abundant in human diet: sulforaphane, benzyl isothiocyanate (BITC) and phenylethyl isothiocyanate, on human cervix carcinoma cell line - HeLa, melanoma cell line - Fem-x, and colon cancer cell line - LS 174, and on peripheral blood mononuclear cells (PBMC), with or without mitogen, were determined by MTT colorimetric assay 72 h after their continuous action. Results. All investigated isothiocyanates inhibited the proliferation of HeLa, Fem-x and LS 174 cells. On all cell lines treated, BITC was the most potent inhibitor of cell proliferation with half-maximum inhibitory concentration (IC50) values of 5.04 mmoL m-3 on HeLa cells, 2.76 mmol m-3 on Fem-x, and 14.30 mmol m-3 on LS 174 cells. Antiproliferative effects on human PBMC were with higher IC50 than on malignant cells. Indexes of selectivity, calculated as a ratio between IC50 values obtained on PBMC and malignant cells, were between 1.12 and 16.57, with the highest values obtained for the action of BITC on melanoma Fem-x cells. Conclusion. Based on its antiproliferative effects on malignant cells, as well as the selectivity of the action to malignant vs normal cells, benzyl isothiocyanate can be considered as a promising candidate in cancer chemoprevention. In general, the safety of investigated compounds, in addition to their antitumor potential, should be considered as an important criterion in cancer chemoprevention. Screening of selectivity is a plausible approach to the evaluation of safety of both natural isothiocyanates and synthesised analogues of these bioactive compounds.
AB  - Uvod/Cilj. Brojne epidemiološke studije pokazale su povoljne efekte konzumiranja povrća iz familije kupusnjača (Brassicaceae) u hemioprevenciji karcinoma. Osnovni biološki aktivni sastojci ovog povrća su izotiocijanati, prisutni u obliku prekursora - glukozinolata. Cilj ovog rada bio je određivanje selektivnosti antiproliferativnog delovanja dijetetskih izotiocijanata na maligne ćelije u odnosu na normalne ćelije. Metode. Antiproliferativna aktivnost tri izotiocijanata zastupljena u ljudskoj ishrani: sulforafana (SFN), benzil-izotiocijanata (BITC) i feniletil-izotiocijanata (FEITC) na humane maligne ćelijske linije, HeLa, ćelijsku liniju karcinoma grlića materice, Fem-x, ćelijsku liniju melanoma, i LS 174, ćelijsku liniju karcinoma kolona, kao i na mononuklearne ćelije periferne krvi (MNĆPK), sa ili bez delovanja mitogena, određivana je MTT kalorimetrijskim testom, 72 h nakon kontinuiranog delovanja agenasa. Rezultati. Svi ispitivani izotiocijanati inhibirali su proliferaciju HeLa, Fem-x i LS 174 ćelija. Na svim ćelijskim linijama BITC je pokazao najizraženije delovanje sa vrednostima polumaksimalne inhibitorne koncentracije (IC50) od 5.04 mmol m-3 na HeLa ćelijama, 2,76 mmoL m-3 na Fem-x i 14,30 mmol m-3 na LS 174 ćelijama. Svi ispitivani izotiocijanati pokazali su citotoksično delovanje na MNĆPK, ali sa višim IC50 vrednostima u odnosu na maligne ćelije. Indeksi selektivnosti antitumorkog delovanja, izraženi kao odnos IC50 vrednosti dobijenih na MNĆPK i malignim ćelijama, bili su između 1,12 i 16,57, sa najvišom vrednosti pri delovanju BITC na Fem-x ćelije. Zaključak. Na osnovu antiproliferativne aktivnosti na maligne ćelije i selektivnosti antiproliferativnog delovanja na maligne u odnosu na normalne ćelije, benzil izotiocijanat se ističe kao perspektivni agens u hemioprevenciji karcinoma. Generalno, pored antitumorskog delovanja, bezbednost primene ovih jedinjenja treba da predstavlja važan kriterijum u izboru odgovarajućih izotiocijanata za primenu u primarnoj, sekundarnoj i tercijarnoj hemioprevenciji kancera. Ispitivanje selektivnosti predstavlja pogodan pristup oceni bezbednosti i prirodnih izotiocijanata i sintetskih analoga.

Biologically active compounds of different Brassicaceae species with antitumor potential are isothiocyanates, present in the form of their precursors - glucosinolates. The aim of this study was to determine the selectivity of antiproliferative action of dietary isothiocyanates for malignant versus normal cells. Methods. Antiproliferative activity of three isothiocyanates abundant in human diet: sulforaphane, benzyl isothiocyanate (BITC) and phenylethyl isothiocyanate, on human cervix carcinoma cell line - HeLa, melanoma cell line - Fem-x, and colon cancer cell line - LS 174, and on peripheral blood mononuclear cells (PBMC), with or without mitogen, were determined by MTT colorimetric assay 72 h after their continuous action. Results. All investigated isothiocyanates inhibited the proliferation of HeLa, Fem-x and LS 174 cells. On all cell lines treated, BITC was the most potent inhibitor of cell proliferation with half-maximum inhibitory concentration (IC50) values of 5.04 mmoL m-3 on HeLa cells, 2.76 mmol m-3 on Fem-x, and 14.30 mmol m-3 on LS 174 cells. Antiproliferative effects on human PBMC were with higher IC50 than on malignant cells. Indexes of selectivity, calculated as a ratio between IC50 values obtained on PBMC and malignant cells, were between 1.12 and 16.57, with the highest values obtained for the action of BITC on melanoma Fem-x cells. Conclusion. Based on its antiproliferative effects on malignant cells, as well as the selectivity of the action to malignant vs normal cells, benzyl isothiocyanate can be considered as a promising candidate in cancer chemoprevention. In general, the safety of investigated compounds, in addition to their antitumor potential, should be considered as an important criterion in cancer chemoprevention. Screening of selectivity is a plausible approach to the evaluation of safety of both natural isothiocyanates and synthesised analogues of these bioactive compounds., Uvod/Cilj. Brojne epidemiološke studije pokazale su povoljne efekte konzumiranja povrća iz familije kupusnjača (Brassicaceae) u hemioprevenciji karcinoma. Osnovni biološki aktivni sastojci ovog povrća su izotiocijanati, prisutni u obliku prekursora - glukozinolata. Cilj ovog rada bio je određivanje selektivnosti antiproliferativnog delovanja dijetetskih izotiocijanata na maligne ćelije u odnosu na normalne ćelije. Metode. Antiproliferativna aktivnost tri izotiocijanata zastupljena u ljudskoj ishrani: sulforafana (SFN), benzil-izotiocijanata (BITC) i feniletil-izotiocijanata (FEITC) na humane maligne ćelijske linije, HeLa, ćelijsku liniju karcinoma grlića materice, Fem-x, ćelijsku liniju melanoma, i LS 174, ćelijsku liniju karcinoma kolona, kao i na mononuklearne ćelije periferne krvi (MNĆPK), sa ili bez delovanja mitogena, određivana je MTT kalorimetrijskim testom, 72 h nakon kontinuiranog delovanja agenasa. Rezultati. Svi ispitivani izotiocijanati inhibirali su proliferaciju HeLa, Fem-x i LS 174 ćelija. Na svim ćelijskim linijama BITC je pokazao najizraženije delovanje sa vrednostima polumaksimalne inhibitorne koncentracije (IC50) od 5.04 mmol m-3 na HeLa ćelijama, 2,76 mmoL m-3 na Fem-x i 14,30 mmol m-3 na LS 174 ćelijama. Svi ispitivani izotiocijanati pokazali su citotoksično delovanje na MNĆPK, ali sa višim IC50 vrednostima u odnosu na maligne ćelije. Indeksi selektivnosti antitumorkog delovanja, izraženi kao odnos IC50 vrednosti dobijenih na MNĆPK i malignim ćelijama, bili su između 1,12 i 16,57, sa najvišom vrednosti pri delovanju BITC na Fem-x ćelije. Zaključak. Na osnovu antiproliferativne aktivnosti na maligne ćelije i selektivnosti antiproliferativnog delovanja na maligne u odnosu na normalne ćelije, benzil izotiocijanat se ističe kao perspektivni agens u hemioprevenciji karcinoma. Generalno, pored antitumorskog delovanja, bezbednost primene ovih jedinjenja treba da predstavlja važan kriterijum u izboru odgovarajućih izotiocijanata za primenu u primarnoj, sekundarnoj i tercijarnoj hemioprevenciji kancera. Ispitivanje selektivnosti predstavlja pogodan pristup oceni bezbednosti i prirodnih izotiocijanata i sintetskih analoga.",

AB  - The aim of research was to determine the effects of maximally therapeutically achievable concentrations of metformin on malignant cells and healthy peripheral blood mononuclear cells (PBMC). Eight patients with T2D or hyperglycemia and nine healthy volunteers were included in the study. For determination of the influence of metformin on the phenotype of breast carcinoma, 1,410 patients with surgically removed tumors were included. From this group 37 breast cancer patients had DM type 2 or hyperglycemia and were pretreated with metformin alone or sometimes in combination with other antidiabetic drugs. Our results proved that metformin at low concentrations induced mild decrease in survival of malignant cells and PBMC stimulated for proliferation, but it didn't affect survival of resting PBMC. The effects of plasma of hyperglycemic patients who were under metformin therapy on autologous PBMC-induced decrease in survival of MDA-MB-361 cells, was noticeable in some patients. Metformin pretreatment for 24 h of HER2+ MDA-MB-361 cells, which were subsequently treated for 48 h with Herceptin, induced additional decline in cell survival. The analysis of influence of metformin on phenotype of breast cancer cells revealed significantly lower number of diabetic cancer patients treated with metformin with overexpressed HER2+ tumors (p  lt  0.013), while the number of patients with ER+PR+ tumors was not significantly changed (p  lt  0.832). In conclusion, therapeutically used concentrations of metformin exhibit mild cytotoxic action on malignant and dividing normal cells pointing to its preferred role in malignant and autoimmune diseases. The use of metformin was associated with pronounced decrease in HER2 overexpressing tumors.

The results were compared with the corresponding data for the cis-diamminedichloridoplafinum(H) (CDDP) as referent cytostatic, as well as with the free ligands and the solvent dimethyl sulfoxide (DMSO) as controls. The complexes showed considerable antiproliferative effect, although significantly less than CDDP. The thermal decomposition pattern of the complexes was determined by simultaneous TG/DSC measurements. The thermal stability of the compounds 2-7 followed the trend of their antiproliferative activity against the HeLa cell line, as well as their corresponding stability constants. The highest thermal stability and cytotoxicity belonged to complex [Cu(2)tpmc](ClO4)(4), with no anionic co-ligand. Complex [Cu-2(NO2)tpmc](ClO4)(3) exhibited a selective cytotoxicity against LS174 cells, at the level of the most active [Cu(2)tpmc](ClO4)(4).

AB  - The antimicrobial and cytotoxic activities of the essential oil of Satureja montana ssp. pisidica from two localities (mountains Korab and Galicica) were studied. Forty-nine components were identified in the each sample. Oxygenated monoterpene hydrocarbons were the major compounds: carvacrol, thymol, carvacrol methyl ether and beta-linalool. Both tested essential oils showed very high and similar antimicrobial activity. Minimal inhibitory concentrations ranged from 12.5 mu g/mL against S. epidermidis to 50 mu g/mL against P. aerugmosa and C. albicans. The cytotoxic effect of the essential oils was tested against MDA-MB-361, MDA-MB-453, HeLa, LS174 and MRCS cells. The essential oil from Korab demonstrated significantly better results than the oil from Galicica, particularly against HeLa and MDA-MB-453 cell lines, with IC50 values of 63.5 and 72.3 mu g/mL, while the oil from Galicica was the most active on the human epithelial cervical cancer HeLa cells (IC50 99.7 mu g/mL).

Isolation, cultivation, and in vitro susceptibility testing of Borrelia burgdorferi sensu lato: A review

AB  - Lyme borreliosis is the most common vector-borne disease in the northern hemisphere. The agents of Lyme borreliosis are borrelia, bacteria of the family Spirochaetaceae, which are grouped in Borrelia burgdorferi sensu lato species complex. Borreliae are fastidious, slow-growing and biochemically inactive bacteria that need special attention and optimal conditions for cultivation. The isolation of Borrelia from clinical material and their cultivation is a time-consuming and demanding procedure. Cultivation lasts from 9 up to 12 weeks, which is much longer than is necessary to grow most other human bacterial pathogens. Although B. burgdorferi sensu lato is susceptible to a wide range of antimicrobial agents in vitro, up to now the susceptibility of individual Borrelia species to antibiotics is defined only partially.

Antiproliferative activity of mixed-ligand Cu(II) complexes with pendant octaazamacrocycle in vitro

AB  - Four cationic mixed-ligand complexes of Cu (II) with N, N', N'', N''' -tetrakis (2-pyridylmethyl) -1,4,8,11 -tetraazacyclotetradecane (tpmc) and aliphatic dicarboxylic acids: succinic, glutaric , adipic and sebacic were investigated in vitro on the human cervix adenocarcinoma (HeLa), human malignant melanoma (Fem-x), oestrogen-receptor-positive human breast cancer (MCF-7) cells and the human acute monocyte leukemia cell line (THP1), while free ligands, Cu(II) salt and solvent (used as a controls) were inactive on the same conditions. The metabolic activity of the cells was determined by tetrazolium MTS test. All complexes showed cytotoxicity toward the used cell lines, while free ligands, Cu(II) salt and solvent (used as a controls) were inactive on the same conditions. Increasing the number of - CH2- groups in homologous serie of aliphatic dicarboxylate ligands leads to the increasing cytotoxic activity of the complex. The highest cytotoxic effect of azamacrocycle Cu(II) complexes with glutarate, adipate, sebacinate ligand showed on the cell line MCF-7 and the worst on HeLa cells. A significant effect of sebacinate Cu(II)complex to all tested tumor cell lines applied in concentrations of 100 and 200 μM and up to HeLa cells.
AB  - Četiri katjonska mešovito-ligandna kompleksa Cu(II) sa N,N',N'N''-tetrakis(2- piridilmetil)-1,4,8,11-tetraaza-ciklotetradekanom i alifatičnim dikarboksilatima: ćilibarnom, glutarnom, adipinskom i sebacinskom kiselinom su ispitivani na humanim malignim ćelijskim linijama: cervikalnog adenokarcinoma (HeLa), melanoma (Fem-X), estrogen pozitivnog karcinoma dojke (MCF-7) i akutne monocitne leukemjie (THP1). Uporedo su testirani i sami ligandi, proste soli i rastvarači. Metabolička aktivnost ćelija je određena MTS testom. Svi ispitivani kompleksi su pokazali citotoksični efekat prema testiranim ćelijskim linijama, dok su slobodni ligandi, Cu(II) so i rastvarač (koji su korišćeni kao kontrolni) pod istim uslovima, bili neaktivni. Sa povećanjem broja -CH2- grupa homologog niza alifatičnih dikarboksilato liganada dolazi i do povećane citotoksičnosti kompleksa. Najveći citotoksični efekat su pokazali makrociklični Cu(II) kompleksi sa glutarato, adipato i sebacinato ligandima prema MCF-7, a najslabiji prema HeLa ćelijama. Najveći antiproliferativni efekat pokazuje kompleks sa sebacinato ligandom prema svim testiranim linijama tumorskih ćelija u primenjenim koncentracijama od 100 i 200 μM, a najviše prema HeLa ćelijama.
