AB  - Introduction/Objective Biofilm and pyocyanin production are essential components of Pseudomonas aeruginosa virulence and antibiotic resistance.Our objective was to examine inhibitory effect of synthetized propafenone derivatives 3-(2-Fluoro-phenyl)-1-(2- (2-hydroxy-3-propylamino-propoxy)-phenyl)-propan-1-one hydrochloride (5OF) and3-(2-Trifluoromethyl-phenyl)-1-(2-(2-hydroxy-3-propylamino-propoxy)-phenyl)-propan-1-one hydrochloride (5CF3) on biofilm and pyocyanin in Pseudomonas aeruginosa clinical strains.Methods Effects were tested on nine clinical isolates and one control laboratory strain of P. aeruginosa. In vitro analysis of biofilm growing was performed by incubating bacteria (0.5 McFarland) with 5OF and 5CF3 (500–31.2 μg/ml) and measuring optical density (OD) at 570 nm. Bacteria in medium without com-pounds were positive control. Blank medium (an uninoculated medium without test compounds) was used as negative control. Pyocyanin production was estimated by OD at 520 nm, after bacteria incubated with 5CF3 and 5OF (250 and 500 μg/ml), treated with chloroform, and chloroform layer mixed with HCl. Results A total of 500 μg/ml of 5OF and 5CF3 completely inhibited biofilm formation in 10/10 and 4/10 strains, respectively. A total of 250 μg/ml of 5OF and 5CF3 strongly inhibited biofilm formation in 7/10 strains, while inhibition with 125 μg/ml of 5OF and 5CF3 was moderate. Lower concentrations had almost no effect on biofilm production. Pyocyanin production was reduced to less than 40% of the control value in 6/9, and less than 50% of the control in 7/9 strains with 500 μg/ml of 5OF and 5CF3, respectively. At 250 μg/ml 5OF and 5CF3, most strains had pyocyanin production above 50% of the control value.Conclusion Synthetized propafenone derivatives, 5OF and 5CF3, inhibited biofilms and pyocyanin produc-tion of Pseudomonas aeruginosa clinical strains. Presented results suggest that propafenone derivatives are potential lead-compounds for synthesis of novel antipseudomonal drugs.
AB  - Увод/Циљ Производња биофилма и пиоцијанина je важан фактор вируленције и антибиотске резистенције бактерије Pseudomonas aeruginosa. Циљ рада је био да се испита инхибиторни ефекат синте-тисаних пропафенонских деривата, 3-(2-флуоро-фенил)-1-[2-(2-хидрокси-3-пропиламино-пропокси)-фенил]-про-пан-1-он-хидрохлорид) (5OF) и 3-(2-трифлуорометилфенил)-1-[2-(2-хидрокси-3-пропиламино-пропокси)-фенил]-про-пан-1-он-хидрохлорид) (5CF3), на продукцију биофилма и пиоцијанина код клиничких изолата бактерије Pseudomonas aeruginosa.Методе Ефекат пропафенонских деривата испитан је на девет клиничких изолата и једном стандардном соју бакте-рије P. aeruginosa. Утицај на продукцију биофилма испитан је in vitro, инкубацијом бактерија (0,5 по Макфарланду) са 5OF и 5CF3 (500–31,2 μg/ml), и мерењем оптичке густине на 570 nm. Бактерије у медијуму без испитиваних једињења су биле позитивна контрола, а сам медијум негативна контро-ла. Производени пиоцијанин, који је одређиван мерењем оптичке густине на 520 nm, на коинкубације бактерија са 5CF3 или 5OF (250 и 500 μg/ml), третиран је хлороформом и мешањем хлороформског слојa са HCl.Резултати При концентрацији од 500 μg/ml 5OF је довео до потпуне инхибиције продукције биофилма код свих испи-тиваних сојева (10/10). Инхибиција биофилма са 500 μg/ml5CF3 била је потпуна код 4/10 сојева. При концентрацији 5OF и 5CF3 од 250 μg/ml продукција биофилма код већине испитаних изолата била је слаба, док је при концентрацији 125 μg/ml 5OF односно 5CF3 продукција била умерена. Ниже концентрације 5OF и 5CF3 нису имале инхибиторни ефекат на формирање биофилма. У присуству 500 μg/ml 5OF у 6/10 испитиваних сојева продукција пиоцијанина пала је на мање од 40% у односу на контролну вредност. Иста концентрација (500 μg/ml) 5CF3 снизила је продукцију пиоцијанина на мање од 50% од контроле у 7/9 сојева. При концентрацији 250 μg/ml 5OF или 5CF3 већина сојева продуковала је пиоцијанин изнад 50% у односу на позитивну контролу.Закључак Синтетисани пропафенонски деривати, 5OF и 5CF3, инхибирају продукцију биофилма и пиоцијанина код клиничких сојева бактерије Pseudomonas aeruginosa. До-бијени резултати указују на то да пропафенонски деривати представљају могућа полазна једињења за синтезу нових антипсеудомонасних агенаса.

Oleic acid - Health benefits and status in plasma phospholipids in the serbian population

AB  - Oleic acid has many benefi cial eff ects on human health. One of the main dietary sources of oleic acid is olive oil. Non-Mediterranean European countries, including Serbia, have low habitual olive oil consumption, but other vegetable oils also contain diff erent amounts of oleic acid. In infants and young children milk is the most important source of fatty acids, including oleic acid. Furthermore, fatty acid composition of plasma phospholipids refl ects dietary intake and fatty acid metabolism. In patients with serious chronic diseases fatty acid status is altered independently on the intake. Here we reviewed status of oleic acid in healthy persons as well as in patients with diff erent chronic diseases in Serbia.

AB  - Nutritivne deficijencije prepoznate su kao jedan od glavnih preventabilnih faktora rizika za nastanak kongenitalnih anomalija i drugih neželjenih ishoda trudnoće. Naučni napredak u oblasti etiopatogeneze kongenitalnih anomalija doveo je do zaključka da je optimalno izbalansirana ishrana majki uz adekvatan unos makro i mikronutrijenata izuzetno važna za obezbeđivanje normalnog embrionalnog razvoja. Zaštitni efekat perikoncepcijske primene suplemenata folne kiseline u redukciji rizika od nastanka i rekurence defekata nervne cevi naučno je potvrđen mnogim obzervacionim i interventnim studijama. Folati učestvuju kao koenzimi u transferu monougljeničnih grupa u reakcijama metabolizma aminokiselina i sintezi i reparaciji nukleinskih kiselina, pa je uloga ovog hidrosolubilnog B9 vitamina posebno značajna za uspešno odvijanje intenzivne ćelijske deobe i proliferacije tokom embriogeneze. Ipak, uprkos postignutom naučnom konsenzusu o koristima folne kiseline u obezbeđivanju zdravog potomstva i činjenici da su mnoga regulatorna tela u oblasti zdravlja izdala preporuke o primeni folne kiseline u periodu planiranja začeća i ranim fazama trudnoće, studije ukazuju da je među ženama reproduktivne dobi unos ovog esencijalnog nutrijenta i dalje suboptimalan. Na varijabilnost situacije u različitim zemljama utiče niz faktora kao što su: stepen obaveštenosti žena o merama prevencije defekata nervne cevi, postojanje, primena i obuhvat strategija za unapređenje unosa i statusa folata u ciljnoj populacionoj grupi, dostupnost i priuštivost suplemenata folne kiseline i konačno, izloženost stanovništva namirnicama koje su fortifikovane ovim vitaminom.Procena nutritivnog statusa danas je prepoznata kao važan deo zdravstvene nege na individualnom i populacionom nivou. Prvi korak u suočavanju sa izazovom ostvarivanja adekvatnog unosa folata kao esencijalnih nutrijenata je pouzdana i objektivna procena nutritivnog statusa zasnovana na primeni standardizovanih, validiranih instrumenata, koji su prilagođeni specifičnim karakteristikama populacije od interesa. Iz toga je proistekao prvi cilj ove doktorske disertacije – razvoj i evaluacija validnosti Upitnika o učestalosti konzumiranja namirnica za procenu dijetarnog unosa folata kod žena reproduktivne dobi u Republici Srbiji (F-FFQ). U razvoju upitnika primenjena je kombinacija validiranog upitnika za procenu unosa folata iz Republike Hrvatske i NCI/ Block Health Habits and History Questionnaire uz dodatnu adaptaciju odnosno dopunu lokalno dostupnim namirnicama i tradicionalnim jelima iz Srbije. Analizu pregledne (eng. face validity) isadržajne validnosti (eng. content validity) upitnika sprovela je grupa eksperata iz Centra izuzetne vrednosti u oblasti istraživanja ishrane i metabolzma Instituta za medicinska istraživanja, Univerziteta u Beogradu...
AB  - Nutritive deficiencies are well-recognized preventable risk factors for the development of congenital anomalies and other adverse pregnancy outcomes. Scientific progress in the comprehension of congenital anomalies has led to the conclusion that optimally balanced maternal diet with adequate intake of macro- and micronutrients can contribute to reducing the incidence of these disorders. Protective effect of the periconceptional folic acid supplementation in reducing the risk for both occurrence and recurrence of neural tube defects (NTDs) is scientifically confirmed through many observational and interventional studies. Folate derivatives are involved as coenzymes in the single-carbon units’ transfer in metabolism of amino acids and synthesis and reparation of nucleic acids. Accordingly, the role of this water-soluble B9 vitamin is of special importance for intense cell maturation, division and proliferation during embryogenesis. However, regardless the unequivocal epidemiological evidence, achieved scientific consensus on the folic acid benefits in ensuring healthy offspring, and the fact that many health authorities officially recommended the use of folic acid prior to conception and during early phases of pregnancy, numerous studies revealed suboptimal intake of this essential nutrient among women of reproductive age. The variability of the situation in different countries is affected by a number of factors such as: the awareness of women of prevention measures for NTDs; the existence, application and coverage of strategies for improving folate intake and status in the targeted population group; the availability and affordability of folic acid supplements; and finally, the population exposure to folic acid- fortified foods.Nutritional status assessment is nowadays recognized as an important part of medical care at individual and population level. In facing the challenge of achieving folate adequacy, the first step is reliable and objective nutritional assessment performed with standardized and validated instruments adapted to specific characteristics of the population of interest. Therefore, the aim of the first study in the present doctoral dissertation was to develop and validate the Food Frequency Questionnaire for the assessment of folate intake (F-FFQ) among women of reproductive age in Serbia. The FFQ for folate (F-FFQ) was developed by combining a validated FFQ for folate in Croatia region with the NCI/Block Health Habits and History Questionnaire with further adaptation based on inclusion of folate-rich traditional foods consumed in Serbia. Iz toga je proistekao prvi cilj ove doktorske disertacije – razvoj i evaluacija validnosti Upitnika o učestalosti konzumiranja namirnica za procenu dijetarnog unosa folata kod žena reproduktivne dobi u Republici Srbiji (F-FFQ). U razvoju upitnika primenjena je kombinacija validiranog upitnika za procenu unosa folata iz Republike Hrvatske i NCI/ Block Health Habits and History Questionnaire uz dodatnu adaptaciju odnosno dopunu lokalno dostupnim namirnicama i tradicionalnim jelima iz Srbije. Analizu pregledne (eng. face validity) isadržajne validnosti (eng. content validity) upitnika sprovela je grupa eksperata iz Centra izuzetne vrednosti u oblasti istraživanja ishrane i metabolzma Instituta za medicinska istraživanja, Univerziteta u Beogradu..., Nutritive deficiencies are well-recognized preventable risk factors for the development of congenital anomalies and other adverse pregnancy outcomes. Scientific progress in the comprehension of congenital anomalies has led to the conclusion that optimally balanced maternal diet with adequate intake of macro- and micronutrients can contribute to reducing the incidence of these disorders. Protective effect of the periconceptional folic acid supplementation in reducing the risk for both occurrence and recurrence of neural tube defects (NTDs) is scientifically confirmed through many observational and interventional studies. Folate derivatives are involved as coenzymes in the single-carbon units’ transfer in metabolism of amino acids and synthesis and reparation of nucleic acids. Accordingly, the role of this water-soluble B9 vitamin is of special importance for intense cell maturation, division and proliferation during embryogenesis. However, regardless the unequivocal epidemiological evidence, achieved scientific consensus on the folic acid benefits in ensuring healthy offspring, and the fact that many health authorities officially recommended the use of folic acid prior to conception and during early phases of pregnancy, numerous studies revealed suboptimal intake of this essential nutrient among women of reproductive age. The variability of the situation in different countries is affected by a number of factors such as: the awareness of women of prevention measures for NTDs; the existence, application and coverage of strategies for improving folate intake and status in the targeted population group; the availability and affordability of folic acid supplements; and finally, the population exposure to folic acid- fortified foods.Nutritional status assessment is nowadays recognized as an important part of medical care at individual and population level. In facing the challenge of achieving folate adequacy, the first step is reliable and objective nutritional assessment performed with standardized and validated instruments adapted to specific characteristics of the population of interest. Therefore, the aim of the first study in the present doctoral dissertation was to develop and validate the Food Frequency Questionnaire for the assessment of folate intake (F-FFQ) among women of reproductive age in Serbia. Correlation coefficients were calculated between each of the dietary methods and three pair-wise correlations were applied for the calculation of validity coefficients. Correlation coefficients observed between F-FFQ and three 24 h recalls were r = 0.56 (p  lt  0.001) and r = 0.57 ( p  lt  0.001) for total sample and validation group, respectively. Bland-Altman plot and cross-classification analyses indicated good agreement between methods. High validity coefficients were determined between the true intake (I) and dietary assessment methods, F-FFQ (Q) and 24 h dietary recalls (R) (rho QI(rbc) = 0.871 and rho QI(ser) = 0.814; rho RIrbc = 0.652 and rho RIser = 0.698), and moderate ones for biomarkers (B) (rho BIrbc = 0.428 and rho BIser = 0.421). The F-FFQ is valid instrument for the assessment of dietary folate intake in women living in Serbia, a country without mandatory folic acid food fortification.

AB  - Adiponectin is an adipocyte-derived plasma protein with insulin-sensitizing and anti-inflammatory properties and is suggested to be a biomarker of metabolic disturbances. The aim of this study was to investigate the effects of alphalipoic acid (ALA) on plasma adiponectin and some metabolic risk factors in patients with schizophrenia. The plasma adipokine levels (adiponectin and leptin), routine biochemical and anthropometric parameters, markers of oxidative stress, and the serum phospholipid fatty acid profile in eighteen schizophrenic patients at baseline, in the middle, and at the end of a 3-month long supplementation period with ALA (500 mg daily) were determined. A significant increase in the plasma adiponectin concentrations, as well as a decrease in fasting glucose and aspartate aminotransferase activity (AST), was found. Baseline AST activity was independently correlated with the adiponectin concentrations. Our data show that ALA can improve plasma adiponectin levels and may play a potential role in the treatment of metabolic risk factor in patients with schizophrenia. Future randomized controlled trials are needed to confirm these preliminary investigations.

In vitro assessment of antiproliferative action selectivity of dietary isothiocyanates for tumor versus normal human cells

AB  - Background/Aim. Numerous epidemiological studies have shown beneficial effects of cruciferous vegetables consumption in cancer chemoprevention. Biologically active compounds of different Brassicaceae species with antitumor potential are isothiocyanates, present in the form of their precursors - glucosinolates. The aim of this study was to determine the selectivity of antiproliferative action of dietary isothiocyanates for malignant versus normal cells. Methods. Antiproliferative activity of three isothiocyanates abundant in human diet: sulforaphane, benzyl isothiocyanate (BITC) and phenylethyl isothiocyanate, on human cervix carcinoma cell line - HeLa, melanoma cell line - Fem-x, and colon cancer cell line - LS 174, and on peripheral blood mononuclear cells (PBMC), with or without mitogen, were determined by MTT colorimetric assay 72 h after their continuous action. Results. All investigated isothiocyanates inhibited the proliferation of HeLa, Fem-x and LS 174 cells. On all cell lines treated, BITC was the most potent inhibitor of cell proliferation with half-maximum inhibitory concentration (IC50) values of 5.04 mmoL m-3 on HeLa cells, 2.76 mmol m-3 on Fem-x, and 14.30 mmol m-3 on LS 174 cells. Antiproliferative effects on human PBMC were with higher IC50 than on malignant cells. Indexes of selectivity, calculated as a ratio between IC50 values obtained on PBMC and malignant cells, were between 1.12 and 16.57, with the highest values obtained for the action of BITC on melanoma Fem-x cells. Conclusion. Based on its antiproliferative effects on malignant cells, as well as the selectivity of the action to malignant vs normal cells, benzyl isothiocyanate can be considered as a promising candidate in cancer chemoprevention. In general, the safety of investigated compounds, in addition to their antitumor potential, should be considered as an important criterion in cancer chemoprevention. Screening of selectivity is a plausible approach to the evaluation of safety of both natural isothiocyanates and synthesised analogues of these bioactive compounds.
AB  - Uvod/Cilj. Brojne epidemiološke studije pokazale su povoljne efekte konzumiranja povrća iz familije kupusnjača (Brassicaceae) u hemioprevenciji karcinoma. Osnovni biološki aktivni sastojci ovog povrća su izotiocijanati, prisutni u obliku prekursora - glukozinolata. Cilj ovog rada bio je određivanje selektivnosti antiproliferativnog delovanja dijetetskih izotiocijanata na maligne ćelije u odnosu na normalne ćelije. Metode. Antiproliferativna aktivnost tri izotiocijanata zastupljena u ljudskoj ishrani: sulforafana (SFN), benzil-izotiocijanata (BITC) i feniletil-izotiocijanata (FEITC) na humane maligne ćelijske linije, HeLa, ćelijsku liniju karcinoma grlića materice, Fem-x, ćelijsku liniju melanoma, i LS 174, ćelijsku liniju karcinoma kolona, kao i na mononuklearne ćelije periferne krvi (MNĆPK), sa ili bez delovanja mitogena, određivana je MTT kalorimetrijskim testom, 72 h nakon kontinuiranog delovanja agenasa. Svi ispitivani izotiocijanati pokazali su citotoksično delovanje na MNĆPK, ali sa višim IC50 vrednostima u odnosu na maligne ćelije. Indeksi selektivnosti antitumorkog delovanja, izraženi kao odnos IC50 vrednosti dobijenih na MNĆPK i malignim ćelijama, bili su između 1,12 i 16,57, sa najvišom vrednosti pri delovanju BITC na Fem-x ćelije. Zaključak. Na osnovu antiproliferativne aktivnosti na maligne ćelije i selektivnosti antiproliferativnog delovanja na maligne u odnosu na normalne ćelije, benzil izotiocijanat se ističe kao perspektivni agens u hemioprevenciji karcinoma. Generalno, pored antitumorskog delovanja, bezbednost primene ovih jedinjenja treba da predstavlja važan kriterijum u izboru odgovarajućih izotiocijanata za primenu u primarnoj, sekundarnoj i tercijarnoj hemioprevenciji kancera. Ispitivanje selektivnosti predstavlja pogodan pristup oceni bezbednosti i prirodnih izotiocijanata i sintetskih analoga.

Indeksi selektivnosti antitumorkog delovanja, izraženi kao odnos IC50 vrednosti dobijenih na MNĆPK i malignim ćelijama, bili su između 1,12 i 16,57, sa najvišom vrednosti pri delovanju BITC na Fem-x ćelije. Zaključak. Na osnovu antiproliferativne aktivnosti na maligne ćelije i selektivnosti antiproliferativnog delovanja na maligne u odnosu na normalne ćelije, benzil izotiocijanat se ističe kao perspektivni agens u hemioprevenciji karcinoma. Generalno, pored antitumorskog delovanja, bezbednost primene ovih jedinjenja treba da predstavlja važan kriterijum u izboru odgovarajućih izotiocijanata za primenu u primarnoj, sekundarnoj i tercijarnoj hemioprevenciji kancera. Ispitivanje selektivnosti predstavlja pogodan pristup oceni bezbednosti i prirodnih izotiocijanata i sintetskih analoga.",

Chemical composition and nutritional quality of wholegrain bread from Belgrade market

AB  - Introduction: There is a growing number of epidemiological evidences that wholegrain products may reduce the risk of cardiovascular
diseases, type 2 diabetes, some forms of cancers, as well as obesity. The
beneficial health effects are due to the presence of many biologically
active compounds, which are located in the outer layer and germ fraction of the grain. Therefore, the higher intake of wholegrain foodstuffs
is recommended. Wholegrain bread, locally known as brown bread, is
the most common foodstuff of this type in Serbian diet.
Objectives: The objective of this paper was to assess the quality
indicators, basic chemical composition, and fiber profile of wholegrain
breads sampled from Belgrade supermarkets and bakeries.
Method / Design: In total 8 different samples of wholegrain
bread were analyzed. The nutritional quality of samples was evaluated by common physicochemical analyses (contents of protein, ash, water, crude fiber, acid degree, sodium chloride, starch). Enzymatic
methods were used for determination of fiber profile (resistant starch,
beta-glucan, fructans, and arabinoxylan). Also, sensory characteristics
were assessed.
Results: During evaluation of the sensory quality all samples
fulfilled the regulatory requirements. All analyzed samples received
high marks for each individual property of quality. However, the varia-
bility of acid degree, as well as some nutrient content (protein, crude
fiber, starch) among samples was great. On the other hand, other
parameters were similar between different bread samples (content of
water, ash, sodium chloride). The fiber fractions that were analyzed in
wholegrain bread samples revealed that resistant starch, fructans and
arabonoxylan were the major fiber fractions, while cellulose content
was unexpectedly low.
Conclusions: Obtained results indicated that wholegrain bread
from Belgrade market is of the standard quality and could be an important source of specific fiber fractions in everyday diet.

Evaluation of angiogenic and neuroprotective potential of different extracts from three Veronica species

AB  - Methanol and 70% acetone extracts from Veronica jacquinii, Veronica urticifolia and Veronica teucrium were evaluated regarding their neuroprotective/antioxidant effects on the human neuroblastoma SH-SY5Y cell line. All extracts exhibited modest protective activity by increasing cell survival in cells stressed with sodium nitroprusside (9-12%) and hydrogen peroxide (16-21%) compared to non-treated cells. These activities were accompanied by reductions in the amount of superoxide radicals and index of lipid peroxidation. Extracts were further analysed for the content of total phenolics, phenylpropanoid glycosides and iridoids, and the major compounds acteoside and aucubin. The highest amount of total phenolics was observed in V. jacquinii, while V. teucrium was richest in total iridoids, acteoside and aucubin. Angiogenic properties on EA.hy926 human endothelial cells were also examined. At the highest non-toxic concentration (25 mu g/ml), the tested extracts inhibited spontaneous tube formation of endothelial cells on the extracellular matrix, implying their possible antiangiogenic activity. The most potent inhibitory effects were shown by methanol extract of V. jacquinii and aqueous acetone extract of V. teucrium. The extracts did not significantly change the adhesive or migratory capacity of EA.hy926 cells. Considering the traditional use of Veronica species, these results suggest a need for further assessment of their supposed wound-healing properties.
