AB - Background/Aim. Plant essential oils (EOs) can have a significant antibacterial effect especially through additive or synergistic action as antibiotic adjuvants. We investi-gated the composition and activity of EOs of two species of genus Salvia (S) from Greece with the aim to deter-mine their antimicrobial activity as well as the activity in combination with selected antibiotics. Methods. The aerial parts of wild-growing S. fruticosa and S. ringens were subjected to a steam distillation and the obtained EOs were analyzed by gas chromatography and gas chroma-tography/mass spectrometry. The broth-microdilution method was used in order to determine the minimum in-hibitory concentrations (MICs) of EOs on seven strains of bacteria and one yeast. Antimicrobial activity of the combination of EO and antibiotics was investigated by checkerboard method and estimated by calculating frac-tional inhibitory concentration (FIC) of each component and fractional inhibitory concentration index (FICI). Re-sults. Dominant component of S. fruticosa EO was trans-thujone (54.2%) and for S. ringens EO it was α-pinene (28.1%). The MICs of EOs of both species were in the range from 200 μg/mL to > 500 μg/mL. The strongest antimicrobial effect was achieved against Bacillus subtilis and Candida albicans. According to FICI values, EO of S. fruticosa had additive effect with ciprofloxacin against most of bacterial strains but not with amikacin. Conclu-sion. The essential oils of S. ringens and S. fruticosa showed modest antimicrobial activity. However, the es-sential oil of S. fruticosa showed a promising additive ef-fect in combination with ciprofloxacin.
Resultati.

AB - The hawkweed genus Hieracium L. s. str. (Asteraceae) is one of the species-richest and
systematically the most complex genera of flowering plants. The genus traditional
classification, beside Hieracium, included, among others, subgenus Pilosella, which is
currently considered as separate genus. In folk medicine, application of some Hieracium s.
str. species was reported for various respiratory, gastrointestinal, urogenital, and some skin
disorders. The 28 species from the Balkan Peninsula were investigated, and majority was
collected on the Mt Durmitor (Montenegro), where the greatest diversity of taxa of this genus
was registered. In flowering aerial parts, composition of pharmaceutically significant
secondary metabolites - flavonoids, phenolic acids, sesquiterpene lactones and triterpenes
was established, and by multivariate statistical analysis their chemosystematic significance
was demonstrated. Corresponding metabolites isolation was performed using column
chromatography and semi-preparative liquid chromatography (LC-MS), identification using
mass spectrometry, NMR spectroscopy and refractometry, and extracts comparative
qualitative and quantitative analysis using LC-MS or gas chromatography (GC-FID-MS). In all
28 species, sesquiterpene lactones were identified for the first time. Three of them were
previously undescribed proline conjugates. Sesquiterpene lactone-amino acid conjugates are
quite rare in nature, and formerly were identified only in a few species of other Asteraceae
genera. Results obtained by investigation of pharmacological activities (in vitro antioxidant,
antimicrobial, cytotoxic, anticholinesterase, and in vivo antihyperalgesic and antiedematous),
as well as safety profiles of selected, chemically characterized extracts and/or secondary
metabolites, indicated the medicinal potential of the analysed plants and gave justification
for further research aiming to find new natural medicinal raw materials.
kiselina, seskviterpenskih laktona i triterpena, i multivarijantnom statističkom analizom
pokazan i njihov hemosistematski značaj. Izolovanje odgovarajućih metabolita izvršeno je
primenom hromatografije u koloni i semi-preparativne tečne hromatografije (LC-MS),
identifikacija primenom masene spektrometrije, NMR spektroskopije i refraktometrije, a uporedna
kvalitativna i kvantitativna analiza ekstrakata primenom LC-MS ili gasne hromatografije (GC-FID-
MS).
vrsta drugih rodova familije Asteraceae. Rezultati dobijeni ispitivanjem farmakološke
aktivnosti (in vitro antioksidantne, antimikrobne, citotoksične, antiholinesterazne, i in vivo
lekoviti potencijal ispitivanih biljaka i opravdanost daljih istraživanja u cilju iznalaženja
novih prirodnih lekovitih sirovina.

AB - Significance of pharmacognostic studies on previously untested or scarcely tested
wild plants is constantly rising, considering growing demand of pharmaceutical industry for
new bioactive isolates, including essential oils and extracts. Accordingly, comprehensive
research that included all nine taxa of genus Heracleum L. (Apiaceae) (three of which are
endemic) from Serbia, Montenegro, North Macedonia and Slovenia was performed (60
samples of various plant organs collected from 14 localities in period 2009-2016). Plants of
this genus (cow parsnips) got their Latin name after ancient hero Heracles, because of their
habitus and use in folk medicine in treatment of various diseases. Also, some of them were
the main ingredient of borscht, a traditional dish in Eastern European countries. They are
interesting because of wide range of metabolites: from essential oil components, coumarins,
flavonoids, phenolic acids, triterpenes and phytosterols, to some specific fatty acids. Analysis
of these secondary and primary metabolites was performed using modern chromatographic
and spectroscopic techniques (GC-FID-MS, LC-MS and NMR). In the case of 35 selected
essential oils, antimicrobial, cytotoxic and/or antioxidant activities were also tested. It was
revealed that studied plants represent potential new sources of compounds of importance
not only for pharmaceutical, but also for cosmetic, food and chemical industries. For
components of essential oils and furanocoumarins, by multivariate statistical analysis,
chemosystematic significance was also established. Results of pharmacological
investigations of essential oils indicated their importance as potential new medicinal plant
raw materials, under condition of respecting established maximum daily intakes for those
oils in which furanocoumarins were detected.
flavonoida, fenolkarboksilnih kiselina, triterpena i fitosterola, sve do nekih specifičnih
masnih kiselina. Ispitivanje navedenih sekundarnih i primarnih metabolita ovih taksona
sprovedeno je uz korišćenje savremenih hromatografskih i spektroskopskih tehnika (GC-
FID-MS, LC-MS i NMR).
prehrambenu i hemijsku industriju.
potencijalno novih lekovitih biljnih sirovina, uz uslov poštovanja ustanovljenih maksimalnih

AB - Phytochemical and pharmacological in-vestigations of essential oils isolated from plant species of the genus Seseli have been intensified recently. These plant species have long-term use in nutrition and traditional medicine in the treatment of various disorders. Volatile secondary metabolites of Seseli gracile Waldst. & Kit (Apiaceae) have not been pharma-cologically examined so far. The aim of the conducted research was to assess the antiradical, antimicrobial and spasmolytic ac-tivities of S. gracile essential oil isolated from the aerial parts of the plant. Methods. The antiradical activity was determined us-ing the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scaveng-ing test, antimicrobial activity using broth microdilution meth-od with standard strains of Gram (+), Gram (-) bacteria and yeast, while spasmolytic activity was evaluated on isolated rat il-eum, pre-contracted with a high concentration of potassium. Results. The results showed moderate antiradical (SC50 = 78.60 μg/mL) and good spasmolytic activity (IC50 = 271.4 nL/mL) of S. gracile essential oil. In the range of tested concen-trations, minimal inhibitory concentration (MIC) was deter-mined only for the strain of Escherichia coli (500 μg/mL). Con-clusion. Results obtained in this study justify the need for fur-ther studies to elucidate exact molecular mechanism underlay this spasmolytic effect of S. gracile herb essential oil.
Rezultati. Rezultati pokazuju umerenu antiradikalsku (SC50 = 78.60 μg/mL) i dobru spazmolitičnu aktivnost (IC50 = 271.4 nL/mL) etarskog ulja S. gracile.

Chemosystematic significance ofthe compounds identified in the herbs of 28 species was determined using multivariate statistics(PCA, nMDS and UPGMA).Dried MeOH herb extracts in vitro exhibited significant total antioxidant activity andscavenging of DPPH•, ABTS•+ and OH• radicals (28 species), as well as the ability to inhibitacetylcholinesterase (27 species) and butyrylcholinesterase (7 species). Among 12 testedcompounds, three flavonoid aglycones and 8-epiixerisamine A exhibited the highestanticholinesterase activity. Selected dried MeOH and CH2Cl2 herb extracts of six species showedantimicrobial activity against 18, whereas 8-epiixerisamine A and crepiside E against fourmicroorganisms. The same selected isolates exhibited cytotoxic activity against three and onecancer cell lines, respectively, with negligible inhibition of the normal cells growth. In thecarrageenan-induced localized inflammation model in rats, three selected MeOH and CH2Cl2extracts exhibited antihyperalgesic, but not antiedematous activity; safety of the oral administrationof two extracts that exhibited significant antihyperalgesic activity was demonstrated in vivo inrotarod test and acute oral toxicity study.

AB - Present study investigated triterpene profile, antihyperalgesic and antiedematous activities of Hieracium scheppigianum flowering aerial parts dichloromethane extract (SCH), and antihyperalgesic and antiedematous activities of previously chemically characterised polyphenol-rich H. glabratum and H. calophyllum flowering aerial parts methanol extracts (GLA and CAL, respectively). α- and β-Amyrin and their acetates, and lupeol acetate were identified and quantified in SCH by GC-FID and GC-MS. In carrageenan-induced localised inflammation model in rats, SCH and GLA (50–200 mg/kg, p.o.) produced significant and dose-dependent antihyperalgesic effect of 26.9%–56.2% (ED50=163.0 ± 26.5 mg/kg) and 25.3%–51.6% (ED50=211.6 ± 70.6 mg/kg), respectively, and CAL (200 mg/kg, p.o.) exhibited effect of 38.1%. Extracts did not significantly reduce paw edema. SCH and GLA, which demonstrated higher (over 50%) antihyperalgesic efficacy, were tested in a rotarod test (200 mg/kg, p.o.) and no alteration of motor coordination was observed. Also, acute administration of SCH and GLA in mice (2000 mg/kg, p.o.) caused neither mortality nor toxicity.

AB - Geranium species are widely used in traditional medicine of Balkan. The aim of this work was to investigate and compare chemical composition of volatile fractions obtained by hydrodistillation from aerial parts of G. macrorrhizum, G. phaeum, G. sanguineum, G. robertianum, G. palustre, G. pyrenaicum, G. columbinum and G. lucidum as well as from underground parts of G. macrorrhizum and G. phaeum, originated from Vlasina plateau in South Eastern Serbia. The volatiles were analyzed using GC/MS and GC-FID. G. palustre volatiles have been studied for the first time with β-selinene (18.6 %) as a characteristic compound. The cluster analysis revealed separation of volatiles into two main groups. Volatile fractions of G. macrorrhizum were separated from all other samples due to high sesquiterpene content (92.3 % in aerial and 94.6 % in underground parts). The volatile fractions of other samples were mainly composed of sesquiterpenes (10.8–61.8 %), diterpenes (12.9–43.0 %) and fatty acids and their derivatives (6.6–21.6 %) with the exception of volatile fraction of G. phaeum underground parts which was dominated only by fatty acids and their derivatives (76.6 %). The results presented in this article contribute to the knowledge on the chemistry of this genus and advances the knowledge on flora of southeast Serbia.

AB - The extraction yield of Juglans nigra L. leaves was assessed at different ethanol concen-trations (0–96% (v/v)) and solvent-to-solid ratios (5–20 kg kg−1). The response surfacemethodology (RSM) and artificial neural network with genetic algorithms (ANN-GA) weredeveloped to optimize the extraction variables. The RSM and ANN-GA models determined50% (v/v) ethanol concentration and 20 kg kg−1solvent-to-solid ratio as optimal conditions, ensuring an extraction yield of 27.69 and 27.19 g 100 g−1of dry leaves. The phenolic com-pounds in optimal extract were quantified: 3-O-caffeoylquinic acid (2.27 mg g−1of dry leaves),quercetin-3-O-galactoside (10.99 mg g−1of dry leaves) and quercetin-3-O-rhamnoside (15.07mg g−1of dry leaves) using high-performance liquid chromatography (HPLC). The mineralsin optimal extract were quantified: macro-elements (the relative order by content was: K> Mg > Ca) using inductively coupled plasma optical emission spectrometry (ICP-OES) andmicro-elements (the relative order by content was: Zn > Rb > Mn > I>Sr > Ni > Cu > Co >V > Ag > Se) using inductively coupled plasma mass spectrometry (ICP-MS). The extrac-tion coefficients for minerals were determined and were highest for K (64.3%) and I (53.5%).Optimization of extraction process resulted in high extraction yield from J. nigra leaves andoptimal extract containing different phytochemical compounds.

AB - The composition of leaf and flower essential oils of eight Heracleum taxa (populations collected in Serbia, Montenegro, North Macedonia and Slovenia) was statistically analyzed to evaluate its chemosystematic significance. Investigated taxa included H. orphanidis and the representatives of H. sphondylium group: H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii and H. verticillatum. Hydrodistilled essential oils were analyzed by GC–FID and GC–MS. Chemosystematic significance was evaluated using multivariate statistics: principal component analysis (PCA), non-metric multidimensional scaling (NMDS) and unweighted pair-group arithmetic averages clustering (UPGMA). Statistical analyses included our previously published data on the composition of essential oils of eight leaf and three flower samples, as well as the data on the composition of the oils of additional eight leaf and five flower samples obtained in the current work. Leaf and flower essential oils of H. sphondylium group members were dominated by various sesquiterpenes, phenylpropanoids and/or monoterpenes. Heracleum orphanidis leaf and flower essential oils were rich in aliphatic esters, mostly octyl acetate. Statistical analysis of the composition of leaf essential oils, as well as of flower oils, demonstrated the grouping of investigated populations of these Heracleum taxa according to their systematics, i.e., separation of H. orphanidis from the representatives of H. sphondylium group, and grouping of H. pyrenaicum subspecies within this group. Morphologically related H. sibiricum and H. ternatum were closely located in PCA and NMDS and in UPGMA even shared the same cluster.

Rezultati.
Rumex crispus (curly dock) is a wild perennial herbaceous plant, which products are considered as a rich source of biologically active molecules with antioxidant and cytotoxic activities. The aim of this study was to estimate of antioxidant and cytotoxic activities of aqueous extract of curly dock fruits. Methods. The aqueous extract of curly dock fruits was evaluated for its antioxidant activity by in vitro assays for ferric-reducing antioxidant power (FRAP), NO•, OH• and 2,2-diphenyl-1-picrylhydrazyl (DPPH)-free radical scavenging activities and the influence on lipid peroxidation in liposomes. The cytotoxicity of tested extract was examined in vitro in human cervix carcinoma (HeLa), colon adenocarcinoma (HT-29) and breast adenocarcinoma (MCF7) cells. Results. The tested extract showed a potential antioxidant activity manifested in scavenging of free radicals as well as an ability to decrease lipid peroxidation in liposomes. The results indicated tissue-selective cytotoxicity of R. crispus fruit extract in vitro. The most prominent antitumor activity was observed towards HeLa and MCF7 cell lines. Conclusion. The investigated aqueous fruit extract of R. crispus had potential antioxidant and cytotoxic activities, with necrosis as a main mechanism of induced cell-death. Different methods of extraction of R. crispus fruits, apart from aqueous, are recommended for further investigations.

AB - Teucrium × rohlenae Maly is a natural hybrid between T. montanum and T.
polium. It was found in the surrounding of Kotor (Montenegro) by L. Čelakovski fil.
and reported by J. Rohlena as T montanum × polium in 1922. This hybrid is validly
described by K. Maly in 1951. After the original description, Teucrium × rohlenae
have not been reported in the Balkan Peninsula, but hybrids between T montanum and
T. polium have been cited in Italy (as Teucrium x castrense) and Moldova (as
Teucrium × bogoutdinovae). In sympatric population of T. montanum and T. polium
in the surrounding of Trilj near Split (Croatia), we found intermediate individuals with
potentially hybridogenic origin. The aim of the study was to analyse 44 morphoanatomical
characters of stem, leaf, inflorescens and calyx, and chemical composition
of volatile compounds of parental species and their potential hybrid Teucrium ×
rohlenae. The composition of hexane extracts of aerial parts was determined using
GC-FID and GC-MS methods. A detailed morpho-anatomical analysis has shown that
parental species are well differentiated, while potential hybrids have an intermediate
position. Volatiles of potential hybrid represented both species, main hydrocarbon
compounds were alike volatiles of T. polium while the terpene profile was more
similar to T. montanum terpenes.

AB - Lavender flower and essential oil are traditionally used for relief of mild
mental stress and to aid sleep. Staphylococcus spp. are part of skin microbiota and
frequent contaminants of the wounds. The aim of the study was to investigate the
composition and antimicrobial activity of lavender oils on standard strains of bacteria,
wound isolates of Staphylococcus epidermidis and S. aureus and also the activity of
the oil in the combination with gentamicin. The essential oils were analysed by GCFID/
GC-MS. Sensitivity of seven standard bacterial strains and eight wound isolates
of staphylococci were examined by disk-diffusion or broth-microdilution method. The
dominant components in the essential oils of lavender flower were: linalool, linalyl
acetate and camphor and in oil of leaves and stems borneol, 1,8-cineol and camphor.
The essential oil of lavender inhibited the growth of laboratory bacterial strains (MIC
0.125 – 1 mg/ml). The best activity (0.125 mg/ml) was obtained with commercial
samples that contained higher content of camphor and borneol. The essential oil of
lavender flower (2%) inhibited the growth of multiresistant wound isolates of
staphylococci. Significant synergistic effects against Staphylococcus spp. were
observed in combinations of subinhibitory concentrations of essential oils (15.6 – 31.2
μg/ml) and gentamicine (0.06 – 0.25 μg/ml).

AB - This study explores the shoot anatomical structure, content and composition of
the essential oils of the vegetative organs and fruit of the Pimpinella alpina. The plant
material was collected at Mt. Besna Kobila in South-Eastern Serbia, and Šar planina
in Metohija. The anatomical studies were conducted on permanent slides obtained by
standard method of preparation. The essential oils obtained by hydrodistillation from
flowering aerial parts, fruits and roots (from flowering and fruiting period), were
analysed by GC-FID and GC-MS. Anatomical analysis showed that the rhizome has
a secondary structure in the bark zone, while in the zone of vascular tissues it has a
primary structure. The stem has a primary structure, with vascular bundles. The leaf
is dorsiventral, amphistomatic. Secretory channels are numerous in all organs.
Sesquiterpene hydrocarbons with (E)-β-farnesene (23.1-52.1%) were dominant in the
essential oils from aerial parts, fruit and roots, except in the root sample from Šar
planina, which was characterised by high amount of phenylpropanoids (44.2%) with
epoxy-pseudoisoeugenyl 2-methylbutyrate (25.9%). Phenylpropanoids with
pseudoisoeugenol skeleton, which are unique to Pimpinella species essential oils,
were also present in prominent amounts (7.9-26.8%) in other samples. This is the first
report on content and composition of P. alpina essential oil.

AB - Recent studies demonstrated that Rumex species exhibit various
pharmacological activities, such as antiinflamatory, antioxidant, anticancer,
antimicrobial, but there are almost no data on pharmacological activities of Balcan
endemic species Rumex balcanicus. Thus, the aim was to examine antioxidant
activity, using DPPH and FRAP method, antimicrobial activity via broth
microdilution method and α-amylase inhibitory activity using DNS reagent of
aqueous and methanol extracts of roots and aerial parts of R. balcanicus, collected on
Vlasina Lake in 2014. All tested extracts exhibited high antioxidant activity which
has been correlated with the high content of polyphenols. Aqueous extracts exhibited
lower activity compared to methanol extracts. Methanol extract ofR. balcanicus roots
showed the highest activity, both in DPPH (IC50= 9.57 μg/mL) and FRAP test
(IC50=3.98 μmol Fe/mg of extract), while aqueous extract of aerial parts showed the
lowest activity (IC50= 24.73 μg/mL; IC50=2.11 μmol Fe/mg). Only methanol extract
of aerial parts showed α-amylase inhibitory activity with IC50=252 μg/mL. All
extracts had low antimicrobial activity against almost all tested bacterial strains except
against Bacillus subtilis. Positive results obtained in this study show that R. balcanisus
contains potential as a new raw material and for the development of drugs efficient in
treating diabetes.

AB - Rumex balcanicus Rech.f. (Polygonaceae) is an endemic species that inhabits
central parts of Serbia and Kosovo. The genus Rumex is characterizied by the
accumulation of anthraquinones, naphthalene-1,8-diols, flavonoids and stilbenoids.
Aerial parts of R. balcanicus, collected on Vlasina Lake (Serbia) in 2014, were used
to determine the chemical composition and cytotoxic activity. Cyclohexane,
dichloromethane, methanol and aqueous extracts were prepared and tested using
MTT-test, against human epithelial cervical cancer cells (HeLa), human colon
carcinoma cell line (LS174), human non-small cell lung cancer cells (A549) and
healthy MRC-5 human embryonic lung fibroblast cell line. Methanol and aqueous
extracts were analyzed using LC-MS. In tested extracts, caffeic acid derivates and
flavonol derivatives were identified. The main components in both extracts were rutin,
miquelianin, quercitrin and quercetin 3-O-acetyl-rhamnoside. Caffeic acid and its
glycosides were also present in analyzed extracts. Aqueous extracts had no
cytotoxicity against tested cell lines, while cyclohexane, dichlormethane and
methanol extracts were active against all tested cell lines with IC50 ranged from
116.48±3.26 μg/mL to 178.21±1.30 μg/mL. Due to the presence of flavonoids and
exhibited cytotoxic effect, R. balcanicus herba could be interesting as a plant material
for further pharmacological analysis and isolation of the anti-tumor compounds.

central parts of Serbia and Kosovo.

AB - Three seaweeds (Halimeda tuna, Codium bursa and Cystoseira barbata) and one seagrass (Cymodocea nodosa) were collected from the Coast of Montenegro, Gulf of Boka Kotorska and their chemical analysis was performed. In seagrass C. nodosa, three phenolic compounds were identified (diosmetin 7-sulfate, caftaric and coutaric acid). The content of β-glucan, fatty acids, sterols and micro- and macro-elements were investigated among all samples. The highest content of β-glucan was detected in C. nodosa seagrass (13.04±0.42 g/100 g). The highest polyunsaturated fatty acids (PUFAs) level was reported in C. barbata, the brown alga (7.157 mg/g), which also had the significant sterol content (fucosterol, 21.76±0.1 μg/g). Green algae, C. bursa and H. tuna, showed the highest level of sterols (β-sitosterol, 95.21±0.16 μg/g and 73.90±0.08 μg/g, respectively). H. tuna had the highest content of calcium (Ca) in amount of 55125 μg/g. In C. bursa, C. barbata and C. nodosa, the Na/K ratio was low (0.43, 0.46 and 0.69, respectively).

AB - Sweet flag rhizome (Acorus calamus L., Araceae) exhibits various biological activities such as sedative, anticonvulsant,
immunosuppressant, antidiabetic, and anti-inflammatory. This plant is protected in Serbia, but due to
its high market requirements, the cultivation of this species could be the final solution. The content and composition
of essential oils from 24 cultivated samples and five samples of sweet flag from natural populations, as
well as the influence of nitrogen fertilization on the composition of essential oils were studied. Essential oils were
analysed using GC and GC/MS. Statistical analysis included the ANOVA and PCA analysis. The total content of
essential oil was not significantly different between the natural (0.80–1.10%) and cultivated samples
(1.15–1.30%). In all essential oils main compounds were β-asarone (4.82–17.98%), cadinadienol
(10.14–13.95%), camphor (3.55–13.20%), and acorenone (9.68–12.64%). The cultivation of sweet flag showed
significant (p < 0.05) impact on the composition of essential oil and the content of β-asarone (4.82–17.07%,
mean 11.95±4.76 vs. 17.51–17.98%, mean 17.71 ± 2.23). The PCA analysis also separated the essential oils
from natural and cultivated conditions based on the content of β-asarone. On the other hand, the amount of the
fertilizer did not show any effect (p > 0.05) on the composition of investigated samples. The process of cultivation
increased the level of active, but potentially genotoxic and carcinogenic compound β-asarone. It would be
significant to determine the content of β-asarone routinely in the sweet flag rhizome, especially in the cultivated
plants.

AB - The ability of the dried MeOH extracts of aerial flowering parts of 28 Hieracium s. str. species from Balkan Peninsula and their selected metabolites (seven flavonoids, three phenolic acids and two sesquiterpene lactones) to inhibit acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) was determined using colorimetric Ellman method. The study included: H. gymnocephalum, H. orieni, H. blecicii, H. paratrichum, H. spirocaule, H. mokragorae, H. pannosum s.l., H. plumulosum, H. villosum, H. pilosum, H. pseudoschenkii, H. naegelianum, H. anastrum, H. calophyllum, H. scheppigianum, H. durmitoricum, H. guentheri-beckii, H. mirificissimum, H. coloriscapum, H. pyricephalum, H. albopellitum, H. glabratum, H. scorzonerifolium s.l., H. dentatum s.l., H. neilreichii, H. valdepilosum s.l., H. tommasinianum and H. macrodontoides. The extracts were mainly more active towards AChE, i.e. all inhibited more than 50% AChE, with H. pseudoschenkii extract being the most potent (IC50=0.64 mg/mL). Seven extracts reached 50% inhibition of
BuChE, and H. pilosum extract was the most active (IC50=0.56 mg/mL). The observed activity could be attributed to some tested constituents. Flavonoid aglycones apigenin, luteolin and diosmetin significantly inhibited both enzymes (IC50AChE=47.12-89.89 μg/mL; C50BuChE=18.40-73.44 μg/mL). Sesquiterpene lactone 8-epiixerisamine A selectively inhibited AChE (IC50=80.01 μg/mL). The other tested metabolites did not reach 50% inhibition of both enzymes.

AB - Cytotoxic and antimicrobial activities were investigated for MeOH extracts of flowering aerial parts of Hieracium calophyllum R. Uechtr. (CAL), H. coloriscapum Rohlena & Zahn (COL), H. pseudoschenkii (Rohlena & Zahn) Niketić (PSE), H. valdepilosum Vill. s.l. (VAL) and H. glabratum Willd. (GLA), their two isolated sesquiterpene lactones 8-epiixerisamine A and crepiside E, as well as for CH2Cl2 extract of flowering aerial parts of H. scheppigianum Freyn (SCH). Crepiside E (IC50 = 46.00 μg/mL), COL (IC50 = 152.28 μg/mL) and SCH (IC50 = 148.12 μg/mL) showed
the highest cytotoxic activity against HeLa cells in MTT test. The cytotoxic activity against LS174, A549, as well as against normal MRC-5 cells was significantly weaker. In microdilution test, crepiside E and 8-epiixerisamine A exhibited noteworthy antifungal effect against Aspergillus ochraceus, Penicillium funiculosum, Candida albicans and C. krusei (MIC = 0.15 - 0.4 mg/mL, MFC = 0.3 - 0.8 mg/mL). Among the extracts, the best antibacterial activity was shown by SCH and CAL against Pseudomonas aeruginosa (MIC = 1.68 and 2.52 mg/mL, MBC = 3.36 and 5.04 mg/mL), and the highest antifungal activity by SCH and VAL against C. albicans (MIC = 2.48 and 2.52 mg/mL, MFC = 4.96 and 5.04 mg/mL).

AB - Headspace fractions of fruits and/or roots of Heracleum sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii and H. verticillatum, belonging to group H. sphondylium, and H. orphanidis, all from sect. Heracleum, as well as H. austriacum subsp. siifolium from sect. Wendia (from Serbia, Montenegro, North Macedonia or Slovenia) were isolated using automatic headspace sampler and analyzed by GC-FID and GC-MS. Chemosystematic significance of identified constituents was evaluated using multivariate statistics: PCA, nMDS and UPGMA. GC analysis of headspace fractions from 17 fruit samples of all investigated taxa showed that octyl acetate or α-pinene (sect. Heracleum), or n-octanol (H. austriacum) were the most abundant. The analysis of the fractions from 13 root samples of investigated taxa belonging to sect. Heracleum revealed prevalence of β-pinene or (Z)-β-ocimene (group H. sphondylium), or n-nonane (H. orphanidis). Statistical analysis of fruit fractions revealed separation of H. austriacum and H. orphanidis from investigated members of group H. sphondylium. Morphologically related H. sphondylium and H. montanum were grouped together and H. verticillatum was well separated from other representatives of group H. sphondylium. Complete separation of H. sibiricum, H. ternatum and H. pyrenaicum samples was noticed in root fractions statistical analysis.

AB - Osteoarthritis (OA) is the most common form of arthritis, which represents a substantial economic burden for society and significantly affects patients’ quality of life. Current conventional treatments of OA may be insufficiently effective and unsafe. In an attempt to overcome these limitations, many patients use herbal medicinal products (HMPs) and dietary supplements. A considerable number of herbal drugs and preparations (e.g., willow bark, Salicis cortex; devil’s claw root, Harpagophyti radix; blackcurrant leaf, Ribis nigri folium; nettle leaf/herb, Urticae folium/herba; meadowsweet/meadowsweet flower, Filipendulae ulmariae herba/flos; rosemary leaf/oil, Rosmarini folium/aetheroleum; and juniper oil, Juniperi aetheroleum) are traditionally employed to relieve minor articular pain. Active constituents (e.g., sesquiterpene lactones, triterpenic acids, diarylheptanoids, iridoid glycosides, phenolic glycosides, procyanidins, and alkaloids) are not often fully known. Experimental studies suggest that herbal extracts/compounds are able to suppress inflammation, inhibit catabolic processes, and stimulate anabolic processes relevant to OA. Therapeutic benefit of most HMPs is expected solely from the experience of their long-standing traditional use. Efficacy and safety of several HMPs were assessed in clinical trials. The growing body of preclinical and clinical evidence provides rationale for the use of herbal products in the treatment of OA. However, at present, they cannot be recommended to patients with confidence.
