Antioxidative and anti-inflammatory activities of Erica spiculifolia extracts and fractions

AB  - There is little data on the phytochemical/pharmacological properties of Erica spiculifolia Salisb. (syn. Bruckentalia spiculifolia (Salisb.) Rchb.). This study examines the antioxidative and anti-inflammatory activities of different extracts and fractions of E. spiculifolia in vitro on isolated rat peritoneal macrophages, in the carrageenan-induced rat paw oedema test, BSA test, and two complementary antioxidant assays. Ethanolic extracts of leaves, flowers, and aboveground parts, and petroleum ether, ether, ethyl acetate, and water fractionations of the ethanol extract of E. spiculifolia applied at doses of 50–200 mg/kg p.o. exhibited dose-dependent anti-inflammatory activity comparable with indomethacin. All tested samples, except for the petroleum ether fraction, exerted excellent in vitro antioxidant activity, and all of them exhibited significant and similar inhibition of BSA denaturation comparable with diclofenac. Ethanolic extract of the aboveground parts obtained by percolation, ethyl acetate and water fractions had the highest efficiency, attenuating inflammation by more than 50% in the lowest applied concentration alongside exceptional radical scavenging activity.

AB  - Kao jedan od najvećih javnozdravstvenih izazova diabetes mellitus predstavlja razlog
konstantnog unapređenja farmakoterapije (1). Savremena terapija diabetes mellitus-a može
promeniti tok bolesti, prevenirati komplikacije i doprineti uštedi u zdravstvu (2). Cilj rada bio je da
se analizira potrošnja antidijabetika u Srbiji, u periodu od 2012. do 2021. godine i usklađenost sa
preporukama Nacionalnog vodiča. Podaci o potrošnji antidijabetika prikupljeni su iz publikacija
Agencije za lekove i medicinska sredstva Srbije. Za svaki internacionalni nezaštićen naziv (INN),
praćena je potrošnja izražena u standardizovanoj jedinici - definisanoj dnevnoj dozi na 1000
stanovnika u danu (DDD/1000 stanovnika/dan). Najveća potrošnja antidijabetika tokom ispitivanog
perioda primećena je 2020. godine, dok je najniža bila 2012. godine. Potrošnja antidijabetika u 2021.
godini iznosila je 92,18 DDD/1000 stanovnika/dan što je za 23,73% više u odnosu na 2012. godinu.
Među ovom grupom lekova dominira upotreba oralnih antidijabetika koji se u proseku primenjuju
4,45 puta više u odnosu na insulin i analoge. Najzastupljeniji lekovi u ukupnoj potrošnji antidijabetika
do 2017. godine bili su sulfonamidi i derivati sulfonilureje kada su u proseku činili 43,39%. Nakon
2017. godine prvo mesto po potrošnji zauzima metformin, čiji udeo u ukupnoj potrošnji
antidijabetika dostiže 48,75% u 2021. godini. Potrošnja novih oralnih antidijabetika je veoma mala i
ne prelazi 0,1% ukupne potrošnje tokom ispitivanog perioda. Ipak najveći porast je primećen u
upotrebi inhibitora natrijum-glukoznog kotransportera 2 (SGLT2) (3,19 DDD/1000 stanovnika/dan
u 2021. godini nasuprot 0,001 DDD/1000 stanovnika/dan u 2015. godini). Rastući trend potrošnje
posebno je izražen kod metformina i SGLT2 inhibitora. Rezultati ukazuju na usklađenost primene
antidijabetika sa preporukama Nacionalnog vodiča.
AB  - As one of the biggest public health challenges, diabetes mellitus represents the reason for the
constant pharmacotherapy improvement (1). Modern therapy of diabetes mellitus can alter the
course of the disease, prevent complications, and contribute to healthcare savings (2). This study
aimed to analyze the consumption of antidiabetic drugs in the Republic of Serbia from 2012 to 2021
and compliance with the recommendations of the National Guide. The consumption of all registered
antidiabetic drugs was obtained from publications of the Medicines and Medical Devices Agency of
Serbia. For each International Nonproprietary Name (INN), consumption was monitored and
expressed in a standardized unit-defined daily dose per 1000 inhabitants per day (DDD/1000
inhabitants/day). During the study period, the highest consumption was observed in 2020, while the
lowest was in 2012. Antidiabetic drug consumption in 2021 amounted to 92.18 DDD/1000
inhabitants/day, which is 23.73% higher compared to 2012. The consumption of oral antidiabetics
was dominant, on average 4.45 times more than insulin and analogs. The most commonly used drugs
until 2017 in total antidiabetic consumption were sulfonylureas and sulfonylurea derivatives,
constituting an average of 43.39%. After 2017, metformin took the lead in consumption, reaching
48.75% of total antidiabetic consumption in 2021. New oral antidiabetic drugs do not exceed 0.1%
of the total consumption during the examined period. However, sodium-glucose cotransporter-2
(SGLT2) inhibitors (3.19 DDD/1000 inhabitants/day in 2021 compared to 0.001 DDD/1000
inhabitants/day in 2015) noted the most significant increase. The increasing consumption trend is
particularly pronounced for metformin and SGLT2 inhibitors. The results indicate compliance in the
use of antidiabetic drugs with the National Guide recommendations.

Spasmolytic, Gastroprotective and Antioxidant Activities of Dry Methanol Extract of Ferula heuffelii Underground Parts

AB  - Dry MeOH extract of Ferula heuffelii (Apiaceae) underground parts was tested for spasmolytic, gastroprotective and antioxidant activities. HPLC analysis revealed that chlorogenic acid (CGA; 34.6 mg/g) was its main constituent. Extract in vitro exhibited notable total antioxidant activity (FRAP value=1.0 μmol Fe2+/mg), and scavenging of DPPH (SC50=62.5 μg/ml) and •OH radicals (49.5 % at 20 μg/ml in 2-deoxyribose assay). In vitro on isolated rat ileum, extract exhibited significant spasmolytic activity, i. e., it showed 124.6 % of maximal atropine effect on spontaneous contractions (at 100 μg/ml), and reduced spasmogenic effect of KCl (80 mm) to 44.4 % (at 60 μg/ml) and of highest applied concentration of ACh to 26.3 % (at 120 μg/ml). In parallel experiments, spasmolytic effect of CGA was also demonstrated. In acute EtOH-induced gastric ulceration model in rats, extract (100 mg/kg p.o.) showed significant gastroprotective effect (gastric damage score 0.50), similar to ranitidine (20 mg/kg p.o.). Obtained results showed that tested F. heuffelii polar extract represents new herbal preparation with potential use against some gastrointestinal complaints.

In vitro cytotoxic and anti-inflammatory activity of Bruckenthalia spiculifolia extracts and fractions

AB  - According to our previous studies, Bruckenthalia spiculifolia Rchb. (Ericaceae) possess
strong antioxidant activity and hydrogel loaded with 2% ethanolic extract from its flowers
and leaves is a potential southing and wound healing agent (1). Ethanolic extracts of leaves,
flowers, and herba, alongside fractions obtained by systematic solvent extraction method
with solvents of the increasing polarity of liquid-liquid fractionation of areal parts of B.
spiculifolia, were prepared. To estimate in vitro anti-inflammatory activities of eight different
extracts and fractions, isolated rat peritoneal macrophages were employed. MTT test was
used for the cytotoxicity determination, while the anti-inflammatory potential
lipopolysaccharide-induced NO production model was utilized (2). Experimental procedures
with animals complied with the rules of the European Union Normative. Resident rat
peritoneal macrophages were harvested by peritoneal lavage and used for both procedures.
Phytochemical analysis was done by RP-HPLC coupled with DAD detection. Tested samples
statistically significantly decreased macrophage ability to metabolize MTT, mainly in a
concentration-dependent manner. All of them also significantly inhibited NO production
from macrophages stimulated with lipopolysaccharide, which can be related to either their
cytotoxic or anti-inflammatory activity. Quantified polar compounds (especially quercetin-3-
O-glucoside) could be associated with better antioxidant and anti-inflammatory effects, as
well as with a lower cytotoxic potential. Indeed, fractions obtained with ethyl acetate and
water, but also ethanol extract obtained after lipids removal with petroleum ether, showed
the most promising effects. These results point out that B. spiculifolia is worth further
investigation in order to detect new naturally occurring anti-inflammatory agents.
AB  - Prema našim prethodnim istraživanjima, Bruckenthalia spiculifolia Rchb. (Ericaceae)
poseduje jaku antioksidativnu aktivnost, a hidrogel sa 2% etanolnog ekstrakta cveta i lista
ove biljne vrste pospešuje zarastanje rana (1). Pripremljeni su etanolni ekstrakti listova,
cvetova i nadzemnog dela biljne vrste B. spiculifolia, kao i frakcije dobijene sistematskom
ekstrakcijom rastvaračima rastuć eg polariteta tečno-tečno frakcionisanjem nadzemnih
delova biljke. Za procenu lokalne antiinflamatorne aktivnosti ukupno osam različitih
ekstrakata i frakcija korišćeni su izolovani peritonealni makrofagi pacova. MTT test je služio
za određivanje citotoksičnosti, a model produkcije NO izazvane lipopolisaharidom za in vitro
kvantifikaciju antiinflamatornog potencijala (2). Eksperimentalne procedure sa životinjama
su usklađeni sa zahtevima Evropske unije. Peritonealni makrofagi pacova su sakupljeni
peritonealnim ispiranjem i korišćeni za obe procedure. Fitohemijska analiza je izvršna

sposobnost makrofaga da metabolišu MTT i to uglavnom na dozno-zavisan način. Svi oni su
takođe značajno inhibirali proizvodnju NO iz makrofaga stimulisanih lipopolisaharidom, što
se može dovesti u vezu sa njihovom citotoksičnom ili antiinflamatornom aktivnošć u.
Kvantifikovana polarna jedinjenja (naročito kvercetin-3-O-glukozid) mogu biti povezana sa
boljim antioksidativnim i antiinflamatornim efektima, kao i sa nižim citotoksičnim
potencijalom. Zaista, frakcije dobijene sa etil acetatom i vodom, ali i etanolni ekstrakt dobijen
nakon uklanjanja lipida petroletrom, su pokazale najbolje efekte. Izloženi rezultati ukazuju
na to da vrstu B. spiculifolia treba dalje ispitati u cilju pronalaženja novih prirodnih
antiinflamatornih agensa.

abstract = "According to our previous studies, Bruckenthalia spiculifolia Rchb. (Ericaceae) possess
strong antioxidant activity and hydrogel loaded with 2% ethanolic extract from its flowers
and leaves is a potential southing and wound healing agent (1). Ethanolic extracts of leaves,
flowers, and herba, alongside fractions obtained by systematic solvent extraction method
with solvents of the increasing polarity of liquid-liquid fractionation of areal parts of B.
spiculifolia, were prepared. To estimate in vitro anti-inflammatory activities of eight different
extracts and fractions, isolated rat peritoneal macrophages were employed. MTT test was
used for the cytotoxicity determination, while the anti-inflammatory potential
lipopolysaccharide-induced NO production model was utilized (2). Experimental procedures
with animals complied with the rules of the European Union Normative. Resident rat
peritoneal macrophages were harvested by peritoneal lavage and used for both procedures.
Phytochemical analysis was done by RP-HPLC coupled with DAD detection. Tested samples
statistically significantly decreased macrophage ability to metabolize MTT, mainly in a
concentration-dependent manner. All of them also significantly inhibited NO production
from macrophages stimulated with lipopolysaccharide, which can be related to either their
cytotoxic or anti-inflammatory activity. Quantified polar compounds (especially quercetin-3-
O-glucoside) could be associated with better antioxidant and anti-inflammatory effects, as
well as with a lower cytotoxic potential. Indeed, fractions obtained with ethyl acetate and
water, but also ethanol extract obtained after lipids removal with petroleum ether, showed
the most promising effects. These results point out that B. spiculifolia is worth further
investigation in order to detect new naturally occurring anti-inflammatory agents., Prema našim prethodnim istraživanjima, Bruckenthalia spiculifolia Rchb. (Ericaceae)
poseduje jaku antioksidativnu aktivnost, a hidrogel sa 2% etanolnog ekstrakta cveta i lista
ove biljne vrste pospešuje zarastanje rana (1). Pripremljeni su etanolni ekstrakti listova,
cvetova i nadzemnog dela biljne vrste B. spiculifolia, kao i frakcije dobijene sistematskom
ekstrakcijom rastvaračima rastuć eg polariteta tečno-tečno frakcionisanjem nadzemnih
delova biljke. Za procenu lokalne antiinflamatorne aktivnosti ukupno osam različitih
ekstrakata i frakcija korišćeni su izolovani peritonealni makrofagi pacova. MTT test je služio
za određivanje citotoksičnosti, a model produkcije NO izazvane lipopolisaharidom za in vitro
kvantifikaciju antiinflamatornog potencijala (2). Eksperimentalne procedure sa životinjama
su usklađeni sa zahtevima Evropske unije. Peritonealni makrofagi pacova su sakupljeni
peritonealnim ispiranjem i korišćeni za obe procedure. Fitohemijska analiza je izvršna

Macroelements versus toxic elements in selected wild edible mushrooms of the Russulacea family from Serbia

AB  - Three edible mushrooms of the Russulacea family (Lactarius deliciosus, Lactarius sanguifluus and Lactarius semisanguifluus), most frequently consumed in Serbia, were analyzed using the ICP-OES technique to evaluate the content of K, P, Ca, Mg, Na, Al, As, Cd and Pb, both in cap and stipe. Corresponding soils were analyzed, too. Based on the obtained values for the elemental composition of the mushrooms and the soil, bioaccumulation and translocation factors were calculated. All the examined mushrooms species were recognized as bioexclusors of analyzed toxic elements, but bioaccumulators of K, P and Ca. The studied mushrooms are good sources of macroelements. One portion of 300 g of fresh mushrooms had a significant contribution of K and P, exceeding 15 % of the recommended daily intake for the elements. On the contrary, mushrooms had a low potential to bioaccumulate toxic elements, and presented results indicated the regular consumption of wild edible mushrooms is safe for human health. Correlation analysis was applied to determine phosphorus’s influence on the elements’ content in the mushrooms and corresponding soils, demonstrating the most remarkable mushrooms' tendency to accumulate phosphorus.
AB  - Циљ овог рада било је одређивање садржаја K, P, Ca, Mg, Na, Al, As, Cd и Pb у три
јестиве, самоникле печурке (Lactarius deliciosus, Lactarius sanguifluus и Lactarius semisanguifluus)
које припадају породици Russulacea. Такође, одређен је и садржај поменутих
елемената у земљишту на коме су расле анализиране печурке. На основу добијених
резултата, за сваки елемент је израчунат биоакумулациони и транслокациони фактор.
Будући да је утврђено да печурке акумулирају одговарајуће макроелементе, резултати
су приказани и као унос (%) одговарајућих елемената на основу препоручене дневне
дозе, прерачунато на порцију од 300 g свежих печурака. За токсичне елементе израчунат
је садржај уноса елемената на основу прихватљивог недељног уноса. Корелациона ана-
лиза је коришћена како би се утврдио утицај фосфора на садржај елемената у печур-
кама и одговарајућим земљиштима, обзиром да је фосфор показао најзначајнију тен-
денцију акумулације.
