AB - Significance of pharmacognostic studies on previously untested or scarcely tested
wild plants is constantly rising, considering growing demand of pharmaceutical industry for
new bioactive isolates, including essential oils and extracts. Accordingly, comprehensive
research that included all nine taxa of genus Heracleum L. (Apiaceae) (three of which are
endemic) from Serbia, Montenegro, North Macedonia and Slovenia was performed (60
samples of various plant organs collected from 14 localities in period 2009-2016). Plants of
this genus (cow parsnips) got their Latin name after ancient hero Heracles, because of their
habitus and use in folk medicine in treatment of various diseases. Also, some of them were
the main ingredient of borscht, a traditional dish in Eastern European countries. They are
interesting because of wide range of metabolites: from essential oil components, coumarins,
flavonoids, phenolic acids, triterpenes and phytosterols, to some specific fatty acids. Analysis
of these secondary and primary metabolites was performed using modern chromatographic
and spectroscopic techniques (GC-FID-MS, LC-MS and NMR). In the case of 35 selected
essential oils, antimicrobial, cytotoxic and/or antioxidant activities were also tested. It was
revealed that studied plants represent potential new sources of compounds of importance
not only for pharmaceutical, but also for cosmetic, food and chemical industries. For
components of essential oils and furanocoumarins, by multivariate statistical analysis,
chemosystematic significance was also established. Results of pharmacological
investigations of essential oils indicated their importance as potential new medicinal plant
raw materials, under condition of respecting established maximum daily intakes for those
oils in which furanocoumarins were detected.
flavonoida, fenolkarboksilnih kiselina, triterpena i fitosterola, sve do nekih specifičnih
masnih kiselina. Ispitivanje navedenih sekundarnih i primarnih metabolita ovih taksona
sprovedeno je uz korišćenje savremenih hromatografskih i spektroskopskih tehnika (GC-
FID-MS, LC-MS i NMR).
prehrambenu i hemijsku industriju.
potencijalno novih lekovitih biljnih sirovina, uz uslov poštovanja ustanovljenih maksimalnih

Plant essential oils (EOs) can have a significant antibacterial effect especially through additive or synergistic action as antibiotic adjuvants. We investi-gated the composition and activity of EOs of two species of genus Salvia (S) from Greece with the aim to deter-mine their antimicrobial activity as well as the activity in combination with selected antibiotics. Methods. The aerial parts of wild-growing S. fruticosa and S. ringens were subjected to a steam distillation and the obtained EOs were analyzed by gas chromatography and gas chroma-tography/mass spectrometry. The broth-microdilution method was used in order to determine the minimum in-hibitory concentrations (MICs) of EOs on seven strains of bacteria and one yeast. Antimicrobial activity of the combination of EO and antibiotics was investigated by checkerboard method and estimated by calculating frac-tional inhibitory concentration (FIC) of each component and fractional inhibitory concentration index (FICI). Re-sults. Dominant component of S. fruticosa EO was trans-thujone (54.2%) and for S. ringens EO it was α-pinene (28.1%). The MICs of EOs of both species were in the range from 200 μg/mL to > 500 μg/mL. The strongest antimicrobial effect was achieved against Bacillus subtilis and Candida albicans. According to FICI values, EO of S. fruticosa had additive effect with ciprofloxacin against most of bacterial strains but not with amikacin. Conclu-sion. The essential oils of S. ringens and S. fruticosa showed modest antimicrobial activity. However, the es-sential oil of S. fruticosa showed a promising additive ef-fect in combination with ciprofloxacin.
Resultati.

AB - The hawkweed genus Hieracium L. s. str. (Asteraceae) is one of the species-richest and
systematically the most complex genera of flowering plants. The genus traditional
classification, beside Hieracium, included, among others, subgenus Pilosella, which is
currently considered as separate genus. In folk medicine, application of some Hieracium s.
str. species was reported for various respiratory, gastrointestinal, urogenital, and some skin
disorders. The 28 species from the Balkan Peninsula were investigated, and majority was
collected on the Mt Durmitor (Montenegro), where the greatest diversity of taxa of this genus
was registered. In flowering aerial parts, composition of pharmaceutically significant
secondary metabolites - flavonoids, phenolic acids, sesquiterpene lactones and triterpenes
was established, and by multivariate statistical analysis their chemosystematic significance
was demonstrated. Corresponding metabolites isolation was performed using column
chromatography and semi-preparative liquid chromatography (LC-MS), identification using
mass spectrometry, NMR spectroscopy and refractometry, and extracts comparative
qualitative and quantitative analysis using LC-MS or gas chromatography (GC-FID-MS). In all
28 species, sesquiterpene lactones were identified for the first time. Three of them were
previously undescribed proline conjugates. Sesquiterpene lactone-amino acid conjugates are
quite rare in nature, and formerly were identified only in a few species of other Asteraceae
genera. Results obtained by investigation of pharmacological activities (in vitro antioxidant,
antimicrobial, cytotoxic, anticholinesterase, and in vivo antihyperalgesic and antiedematous),
as well as safety profiles of selected, chemically characterized extracts and/or secondary
metabolites, indicated the medicinal potential of the analysed plants and gave justification
for further research aiming to find new natural medicinal raw materials.
kiselina, seskviterpenskih laktona i triterpena, i multivarijantnom statističkom analizom
pokazan i njihov hemosistematski značaj. Izolovanje odgovarajućih metabolita izvršeno je
primenom hromatografije u koloni i semi-preparativne tečne hromatografije (LC-MS),
identifikacija primenom masene spektrometrije, NMR spektroskopije i refraktometrije, a uporedna
kvalitativna i kvantitativna analiza ekstrakata primenom LC-MS ili gasne hromatografije (GC-FID-
MS).
vrsta drugih rodova familije Asteraceae. Rezultati dobijeni ispitivanjem farmakološke
aktivnosti (in vitro antioksidantne, antimikrobne, citotoksične, antiholinesterazne, i in vivo
lekoviti potencijal ispitivanih biljaka i opravdanost daljih istraživanja u cilju iznalaženja
novih prirodnih lekovitih sirovina.

AB - Phytochemical and pharmacological in-vestigations of essential oils isolated from plant species of the genus Seseli have been intensified recently. These plant species have long-term use in nutrition and traditional medicine in the treatment of various disorders. Volatile secondary metabolites of Seseli gracile Waldst. & Kit (Apiaceae) have not been pharma-cologically examined so far. The aim of the conducted research was to assess the antiradical, antimicrobial and spasmolytic ac-tivities of S. gracile essential oil isolated from the aerial parts of the plant. Methods. The antiradical activity was determined us-ing the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scaveng-ing test, antimicrobial activity using broth microdilution meth-od with standard strains of Gram (+), Gram (-) bacteria and yeast, while spasmolytic activity was evaluated on isolated rat il-eum, pre-contracted with a high concentration of potassium. Results. The results showed moderate antiradical (SC50 = 78.60 μg/mL) and good spasmolytic activity (IC50 = 271.4 nL/mL) of S. gracile essential oil. In the range of tested concen-trations, minimal inhibitory concentration (MIC) was deter-mined only for the strain of Escherichia coli (500 μg/mL). Con-clusion. Results obtained in this study justify the need for fur-ther studies to elucidate exact molecular mechanism underlay this spasmolytic effect of S. gracile herb essential oil.
Rezultati. Rezultati pokazuju umerenu antiradikalsku (SC50 = 78.60 μg/mL) i dobru spazmolitičnu aktivnost (IC50 = 271.4 nL/mL) etarskog ulja S. gracile.

Among 12 testedcompounds, three flavonoid aglycones and 8-epiixerisamine A exhibited the highestanticholinesterase activity. Selected dried MeOH and CH2Cl2 herb extracts of six species showedantimicrobial activity against 18, whereas 8-epiixerisamine A and crepiside E against fourmicroorganisms. The same selected isolates exhibited cytotoxic activity against three and onecancer cell lines, respectively, with negligible inhibition of the normal cells growth. In thecarrageenan-induced localized inflammation model in rats, three selected MeOH and CH2Cl2extracts exhibited antihyperalgesic, but not antiedematous activity; safety of the oral administrationof two extracts that exhibited significant antihyperalgesic activity was demonstrated in vivo inrotarod test and acute oral toxicity study.

AB - Present study investigated triterpene profile, antihyperalgesic and antiedematous activities of Hieracium scheppigianum flowering aerial parts dichloromethane extract (SCH), and antihyperalgesic and antiedematous activities of previously chemically characterised polyphenol-rich H. glabratum and H. calophyllum flowering aerial parts methanol extracts (GLA and CAL, respectively). α- and β-Amyrin and their acetates, and lupeol acetate were identified and quantified in SCH by GC-FID and GC-MS. In carrageenan-induced localised inflammation model in rats, SCH and GLA (50–200 mg/kg, p.o.) produced significant and dose-dependent antihyperalgesic effect of 26.9%–56.2% (ED50=163.0 ± 26.5 mg/kg) and 25.3%–51.6% (ED50=211.6 ± 70.6 mg/kg), respectively, and CAL (200 mg/kg, p.o.) exhibited effect of 38.1%. Extracts did not significantly reduce paw edema. SCH and GLA, which demonstrated higher (over 50%) antihyperalgesic efficacy, were tested in a rotarod test (200 mg/kg, p.o.) and no alteration of motor coordination was observed. Also, acute administration of SCH and GLA in mice (2000 mg/kg, p.o.) caused neither mortality nor toxicity.

AB - The composition of leaf and flower essential oils of eight Heracleum taxa (populations collected in Serbia, Montenegro, North Macedonia and Slovenia) was statistically analyzed to evaluate its chemosystematic significance. Investigated taxa included H. orphanidis and the representatives of H. sphondylium group: H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii and H. verticillatum. Hydrodistilled essential oils were analyzed by GC–FID and GC–MS. Chemosystematic significance was evaluated using multivariate statistics: principal component analysis (PCA), non-metric multidimensional scaling (NMDS) and unweighted pair-group arithmetic averages clustering (UPGMA). Statistical analyses included our previously published data on the composition of essential oils of eight leaf and three flower samples, as well as the data on the composition of the oils of additional eight leaf and five flower samples obtained in the current work. Leaf and flower essential oils of H. sphondylium group members were dominated by various sesquiterpenes, phenylpropanoids and/or monoterpenes. Heracleum orphanidis leaf and flower essential oils were rich in aliphatic esters, mostly octyl acetate. Statistical analysis of the composition of leaf essential oils, as well as of flower oils, demonstrated the grouping of investigated populations of these Heracleum taxa according to their systematics, i.e., separation of H. orphanidis from the representatives of H. sphondylium group, and grouping of H. pyrenaicum subspecies within this group. Morphologically related H. sibiricum and H. ternatum were closely located in PCA and NMDS and in UPGMA even shared the same cluster.

AB - The extraction yield of Juglans nigra L. leaves was assessed at different ethanol concen-trations (0–96% (v/v)) and solvent-to-solid ratios (5–20 kg kg−1). The response surfacemethodology (RSM) and artificial neural network with genetic algorithms (ANN-GA) weredeveloped to optimize the extraction variables. The RSM and ANN-GA models determined50% (v/v) ethanol concentration and 20 kg kg−1solvent-to-solid ratio as optimal conditions, ensuring an extraction yield of 27.69 and 27.19 g 100 g−1of dry leaves. The phenolic com-pounds in optimal extract were quantified: 3-O-caffeoylquinic acid (2.27 mg g−1of dry leaves),quercetin-3-O-galactoside (10.99 mg g−1of dry leaves) and quercetin-3-O-rhamnoside (15.07mg g−1of dry leaves) using high-performance liquid chromatography (HPLC). The mineralsin optimal extract were quantified: macro-elements (the relative order by content was: K> Mg > Ca) using inductively coupled plasma optical emission spectrometry (ICP-OES) andmicro-elements (the relative order by content was: Zn > Rb > Mn > I>Sr > Ni > Cu > Co >V > Ag > Se) using inductively coupled plasma mass spectrometry (ICP-MS). The extrac-tion coefficients for minerals were determined and were highest for K (64.3%) and I (53.5%).Optimization of extraction process resulted in high extraction yield from J. nigra leaves andoptimal extract containing different phytochemical compounds.

AB - Geranium species are widely used in traditional medicine of Balkan. The aim of this work was to investigate and compare chemical composition of volatile fractions obtained by hydrodistillation from aerial parts of G. macrorrhizum, G. phaeum, G. sanguineum, G. robertianum, G. palustre, G. pyrenaicum, G. columbinum and G. lucidum as well as from underground parts of G. macrorrhizum and G. phaeum, originated from Vlasina plateau in South Eastern Serbia. The volatiles were analyzed using GC/MS and GC-FID. G. palustre volatiles have been studied for the first time with β-selinene (18.6 %) as a characteristic compound. The cluster analysis revealed separation of volatiles into two main groups. Volatile fractions of G. macrorrhizum were separated from all other samples due to high sesquiterpene content (92.3 % in aerial and 94.6 % in underground parts). The volatile fractions of other samples were mainly composed of sesquiterpenes (10.8–61.8 %), diterpenes (12.9–43.0 %) and fatty acids and their derivatives (6.6–21.6 %) with the exception of volatile fraction of G. phaeum underground parts which was dominated only by fatty acids and their derivatives (76.6 %). The results presented in this article contribute to the knowledge on the chemistry of this genus and advances the knowledge on flora of southeast Serbia.

Rezultati.
Rumex crispus (curly dock) is a wild perennial herbaceous plant, which products are considered as a rich source of biologically active molecules with antioxidant and cytotoxic activities. The aim of this study was to estimate of antioxidant and cytotoxic activities of aqueous extract of curly dock fruits. Methods. The aqueous extract of curly dock fruits was evaluated for its antioxidant activity by in vitro assays for ferric-reducing antioxidant power (FRAP), NO•, OH• and 2,2-diphenyl-1-picrylhydrazyl (DPPH)-free radical scavenging activities and the influence on lipid peroxidation in liposomes. The cytotoxicity of tested extract was examined in vitro in human cervix carcinoma (HeLa), colon adenocarcinoma (HT-29) and breast adenocarcinoma (MCF7) cells. Results. The tested extract showed a potential antioxidant activity manifested in scavenging of free radicals as well as an ability to decrease lipid peroxidation in liposomes. The results indicated tissue-selective cytotoxicity of R. crispus fruit extract in vitro. The most prominent antitumor activity was observed towards HeLa and MCF7 cell lines. Conclusion. The investigated aqueous fruit extract of R. crispus had potential antioxidant and cytotoxic activities, with necrosis as a main mechanism of induced cell-death. Different methods of extraction of R. crispus fruits, apart from aqueous, are recommended for further investigations.

AB - Three seaweeds (Halimeda tuna, Codium bursa and Cystoseira barbata) and one seagrass (Cymodocea nodosa) were collected from the Coast of Montenegro, Gulf of Boka Kotorska and their chemical analysis was performed. In seagrass C. nodosa, three phenolic compounds were identified (diosmetin 7-sulfate, caftaric and coutaric acid). The content of β-glucan, fatty acids, sterols and micro- and macro-elements were investigated among all samples. The highest content of β-glucan was detected in C. nodosa seagrass (13.04±0.42 g/100 g). The highest polyunsaturated fatty acids (PUFAs) level was reported in C. barbata, the brown alga (7.157 mg/g), which also had the significant sterol content (fucosterol, 21.76±0.1 μg/g). Green algae, C. bursa and H. tuna, showed the highest level of sterols (β-sitosterol, 95.21±0.16 μg/g and 73.90±0.08 μg/g, respectively). H. tuna had the highest content of calcium (Ca) in amount of 55125 μg/g. In C. bursa, C. barbata and C. nodosa, the Na/K ratio was low (0.43, 0.46 and 0.69, respectively).

H. sphondylium (H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii i H. verticillatum), i H. orphanidis, svi iz (tipske) sect. Wendia.
Hemosistematski značaj komponenti etarskih ulja (u slučaju podzemnih organa i plodova i komponenti njihovih headspace frakcija) i furanokumarina utvrđen je multivarijantnom statističkom analizom (PCA, nMDS i UPGMA).

AB - For essential oils of roots, leaves, flowers and fruits of eight Heracleum taxa (H.sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii, H. verticillatum and H. orphanidis), we previously demonstrated antimicrobial, cytotoxic (selective to cancer cells) and/or antioxidant activities. In this work, for these essential oils maximum daily intake related to total furanocoumarins (FCs) content was estimated, according to Committee on Herbal Medicinal Products of European Medicines Agency (EMA/HMPC) recommendations. FCs were quantified using gas chromatography, and their sum equivalent to xanthotoxin (8-methoxypsoralen, 8-MOP) was calculated. It was shown that daily intake, not contributing significantly to overall risk (equivalent to intake of 1.5 mg FCs) for root essential oils was in the range of 1.94-5.23 mL, for fruit oils of 5.23-15.68 mL and for leaf or flower oils of 2.90-15.68 mL. Daily intake, not posing any unacceptable risk (equivalent to intake of 15 μg FCs) for root oils was in the range of 0.02-0.05 mL, for fruit oils of 0.05-0.16 mL and for leaf or flower oils of 0.03-0.16 mL. This work demonstrates the application of current EMA/HMPC recommendations, in order to establish safety profile of herbal preparations containing FCs.
AB - Za etarska ulja korena, listova, cvasti i plodova osam taksona roda Heracleum
(H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum,
demonstrira primenu aktuelnih EMA/HMPC preporuka u cilju utvrđivanja bezbednosnog
profila biljnih preparata u kojima su detektovani furanokumarini.

AB - Fatty acids (FAs), sterols, and triterpenes of dichloromethane extracts of the fruits of 8 Heracleum L. taxa (Apiaceae) from southeastern Europe were investigated by gas chromatography with flame ionization detector and gas chromatography with mass spectrometry. In order to analyze the FAs, their volatile methyl esters were obtained by saponification and subsequent esterification of oily supernatants of the fruit extracts. Dominant was petroselinic acid (42.8%-56.5%), followed by linoleic (20.3%-33.3%) and oleic acids (12.3%-13.7%). Sterols and triterpenes were analyzed as volatile derivatives obtained by the silanization of residual unsaponifiable fractions. Among them, the most abundant was β-sitosterol (44.9%-56.9%), followed by stigmasterol (15.7%-25.0%), Δ7-stigmastenol (6.6%-12.5%), and campesterol (5.2%-8.1%). The quantity of petroselinic acid was also determined by the external standard method (298.8-433.4 mg/g of oily supernatant). The obtained results show that the investigated plants are potential valuable sources of the compounds utilized in different industries.

AB - The antimicrobial activity of the essential oils of root, aerial parts and fruit from two natural populations of Seseli rigidum was tested against 28 isolates of Candida albicans and one laboratory control strain of C. albicans ATCC 10259. Strains of C. albicans were isolated from oropharyngeal swabs, vaginal and/or cervical swabs, eye swabs, wound swabs, feces and urine. The investigated strains showed high level of resistance towards the activity of control antimycotic agent fluconazole and to a lower extent towards amphotericin B. The S. rigidum essential oils, in the concentrations of 50-200 μg/ml, inhibited the growth of investigated strains (28.6-46.4% of tested strains). The essential oils of root from both localities showed antifungal effect at the concentration 50 μg/ml. The dominant compound in the root essential oils was falcarinol (57.5% and 88.8%). The lower activity was obtained with the essential oil of aerial parts or fruit. The 100 μg/ml of essential oil of aerial parts showed significant effect on C. albicans strains. The essential oil of fruit from population from Brdjanska gorge exerted somehow higher effect. It was characterised by higher α-pinene and β-phellandrene content and lower amount of sabinene. The obtained results revealed the significant antifungal effects of S. rigidum root, aerial parts and fruit essential oils against resistant strains of C. albicans.

AB - Sweet flag rhizome (Acorus calamus L., Araceae) exhibits various biological activities such as sedative, anticonvulsant,
immunosuppressant, antidiabetic, and anti-inflammatory. This plant is protected in Serbia, but due to
its high market requirements, the cultivation of this species could be the final solution. The content and composition
of essential oils from 24 cultivated samples and five samples of sweet flag from natural populations, as
well as the influence of nitrogen fertilization on the composition of essential oils were studied. Essential oils were
analysed using GC and GC/MS. Statistical analysis included the ANOVA and PCA analysis. The total content of
essential oil was not significantly different between the natural (0.80–1.10%) and cultivated samples
(1.15–1.30%). In all essential oils main compounds were β-asarone (4.82–17.98%), cadinadienol
(10.14–13.95%), camphor (3.55–13.20%), and acorenone (9.68–12.64%). The cultivation of sweet flag showed
significant (p < 0.05) impact on the composition of essential oil and the content of β-asarone (4.82–17.07%,
mean 11.95±4.76 vs. 17.51–17.98%, mean 17.71 ± 2.23). The PCA analysis also separated the essential oils
from natural and cultivated conditions based on the content of β-asarone. On the other hand, the amount of the
fertilizer did not show any effect (p > 0.05) on the composition of investigated samples. The process of cultivation
increased the level of active, but potentially genotoxic and carcinogenic compound β-asarone. It would be
significant to determine the content of β-asarone routinely in the sweet flag rhizome, especially in the cultivated
plants.

AB - Filipendula ulmaria i F. vulgaris se tradicionalno koriste kod peptičkog ulkusa i bolnih stanja.Cilj: Liofilizovani infuzi cvasti (LIC) ovih vrsta i izolovana jedinjenja F. vulgaris su podvrgnuti hemijskim i farmakološkim ispitivanjima u cilju preliminarne procene terapijskog potencijala i opravdanosti zabeležene etnomedicinske primene.
AB - Filipendula ulmaria and F. vulgaris are traditionally used in the treatment of peptic ulcer and pain.Aim of the study: Lyophilized flower infusions (LFIs) of these species and isolated compounds of F. vulgaris were subjected to chemical and pharmacological investigations with the goal to preliminary assess therapeutic potential and validity of the recorded ethnomedicinal use. Methods: Chemical composition of LFIs was analyzed by HPLC-UV method. Antioxidant and cytotoxic activities were assessed in vitro, anti-inflammatory effect and anti-aggregation potential were studied ex vivo in human platelets, whereas antihyperalgesic, antiedematous, gastroprotective activities and acute toxicity were investigated in vivo. Results and discussion: LFIs were characterized by high polyphenolic content, pronounced antioxidant activity, low capacity to inhibit biosynthesis of eicosanoids and cytotoxicity. Antihyperalgesic effect of LFIs (100–300 mg/kg) was statistically significant and dose-dependent: ED50 ± SEM values, obtained at the time of peak effects, were 164.8 ± 15.4 mg/kg (F. ulmaria) and 172.2 ± 6.2 mg/kg (F. vulgaris). LFIs (100–300 mg/kg), tellimagrandin II (40 mg/kg) and spiraeoside (50 mg/kg) reduced intensity of gastric lesions induced by ethanol. After acute application of high LFIs doses, signs of toxicity in animals were not observed. In performed experiments, it was shown that flavonoids, gallic acid and tellimagrandin II contributed to the pharmacological actions of LFIs.Conclusion: The obtained data support use of F. ulmaria and F. vulgaris flowers in traditional medicine. Polyphenols are active constituents, and hence potential quality markers. Significant similarity of chemical and pharmacological profiles of tested LFIs suggests that F. ulmaria and F. vulgaris could be equivalent biological sources of herbal drug Filipendulae flos.

AB - Ethnopharmacological relevance: Meadowsweet (Filipendula ulmaria (L.) Maxim.) and dropwort (Filipendula vulgaris Moench) are herbaceous perennials employed in folk medicine for their antirheumatic, antipyretic and antiulcer properties. Aim of the study: To assess ethnomedicinal claims through investigation of antioxidant, anti-inflammatory and gastroprotective effects of F. ulmaria and F. vulgaris lyophilized flower infusions (LFIs) as well as the F. vulgaris isolated flavonoids spiraeoside, kaempferol 4'-O-glucoside, astragalin 2'-O-gallate, mixture of hyperoside 2'-O-gallate and isoquercitrin 2'-O-gallate, and a tannin tellimagrandin IL Materials and methods: Free radical scavenging activity of the tested samples was determined by examining their ability to neutralize DPPH and OH radicals in vitro, whereas reducing properties were assessed in Ferric Reducing Antioxidant Power (FRAP) assay. Anti-inflammatory activity was studied ex vivo in human platelets by monitoring the effect on eicosanoid biosynthesis.. Gastroprotective action was estimated in animal model of acute gastric injury induced by ethanol. Results: LFIs and spiraeoside exerted activities comparable to those of positive control in DPPH-radical scavenging and FRAP antioxidant assays, whereas notable hydroxyl radical scavenging ability was demonstrated only for spiraeoside (IC50 = 5.1 mu g/mL). Among tested samples, astragalin 2 ''-O-gallate (IC50 = 141.1 mu g/mL) and spiraeoside (IC50 = 4.69 mu g/mL) the most markedly inhibited production of pro -inflammatory prostaglandin E-2 and 12(S)-hydroxy-(5Z,8Z,10E,14Z)-eicosatetraenoic acid in human platelets, respectively. Examination of LFIs (100-300 mg/kg, p.o.) gastroprotective action in rats revealed their capacity to preserve mucosal integrity. In addition, spiraeoside (50 mg/kg, p.o.) and tellimagrandin II (40 mg/kg, p.o.) showed ulcer preventive ability. Conclusion: Current study supports documented traditional use of investigated herbs and indicates that flavonoid and tannin components are partially responsible for the demonstrated pharmacological activities.

AB - IntroductionHieracium s. str. represents one of the largest and most complex genera of flowering plants. As molecular genetics seems unlikely to disentangle intricate relationships within this reticulate species complex, analysis of flavonoids and phenolic acids, known as good chemosystematic markers, promise to be more reliable. Data about pharmacological activity of Hieracium species are scarce. ObjectiveEvaluation of the chemosystematic significance of flavonoids and phenolic acids of methanol extracts of aerial flowering parts of 28 Hieracium species from the Balkans. Additionally, investigation of antioxidant potentials of the extracts. MethodsComparative qualitative and quantitative analysis of flavonoids and phenolic acids was performed by LC-MS. Multivariate statistical data analysis included non-metric multidimensional scaling (nMDS), unweighted pair-group arithmetic averages (UPGMA) and principal component analysis (PCA). Antioxidant activity was evaluated using three colorimetric tests. ResultsDominant phenolics in almost all species were luteolin type flavonoids, followed by phenolic acids. Although the investigated Hieracium species share many compounds, the current classification of the genus was supported by nMDS and UPGMA analyses with a good resolution to the group level. Hieracium naegelianum was clearly separated from the other investigated species. Spatial and ecological distances of the samples were likely to influence unexpected differentiation of some groups within H. sect. Pannosa. The vast majority of dominant compounds significantly contributed to differences between taxa. The antioxidant potential of the extracts was satisfactory and in accordance with their phenolics composition. ConclusionsComparative LC-MS analysis demonstrated that flavonoids and phenolic acids are good indicators of chemosystematic relationships within Hieracium, particularly between non-hybrid species and groups from the same location. abstract = "IntroductionHieracium s. str. represents one of the largest and most complex genera of flowering plants. Spatial and ecological distances of the samples were likely to influence unexpected differentiation of some groups within H. sect. Pannosa. AB - The composition of the essential oils and the furanocoumarin profiles of CH2Cl2 extracts from underground parts and fruits of nine Heracleum taxa (Apiaceae) from Southeastern Europe were statistically analyzed to evaluate their chemosystematic significance. Eight investigated taxa (H. orphanidis and members of H. sphondylium group: H. sphondylium, H. sibiricum, H. montanum, H. ternatum, H. pyrenaicum subsp. pollinianum, H. pyrenaicum subsp. orsinii and H. verticillatum) belong to the type section of the genus. Additionally analyzed taxon, H. austriacum subsp. siifolium, belongs to H. sect. Wendia. Qualitative and quantitative analysis of the essential oils was performed by GC-FID and GC/MS, and of the furanocoumarins by LC/MS. Furanocoumarins were identified using standards and/or based on UV, MS, 1H- and ROESY NMR spectra. Multivariate statistics (PCA, nMDS, UPGMA) of analyzed metabolites showed that the investigated taxa were grouped according to their taxonomic classification. PCA revealed the significance of some monoterpenes and (Z)-falcarinol of the underground parts oils, octyl esters of the fruit oils, and most of the total of 12 identified furanocoumarins.

AB - Four sesquiterpene lactones (SLs), including three undescribed proline-SL conjugates, the guaianolides calophyllamine A and 8-epiixerisamine A, and the eudesmanolide calophyllamine B, were isolated from the methanol extract of Hieracium calophyllum R. Uechtr. (Compositae) flowering heads. Another known guaianolide, crepiside E, was detected in Hieracium L. species for the first time. Their structures were elucidated using extensive 1D and 2D NMR spectroscopy in combination with HRMS. The isolated SLs were used as external standards for qualitative and quantitative LC-MS analysis of the dry methanol extracts of the flowering aerial parts of 28 Hieracium species from the Balkan Peninsula. Guaianolides were the dominant SLs in 27 species studied. The chemosystematic significance of detected SLs was evaluated using multivariate statistics (PCA, nMDS and UPGMA). Differentiation between the main groups was well supported. All four compounds significantly and equally contributed to the differences between the species. In addition, the eudesmanolide calophyllamine B could be a significant chemosystematic marker for H. sect. Villosa (Griseb.) Gremli s.l. and Glauciformia (Freyn) Zahn-Italica (Fr.) Av. Touv.

AB - The subject of the study was the investigation of the chemical composition and antimicrobial activity of the essential oils (EOs) isolated from Calamintha sylvatica, C. vardarensis, C. nepeta and C. glandulosa, as well as their antibacterial activity in combination with antibiotics. The quantitative and qualitative analysis of EOs was performed using the GC/FID and GC/MS methods. The antimicrobial activity of EOs against six standard bacterial strains and one strain of yeast was tested using the broth microdilution method, while the antimicrobial activity of a combination of essential oils and gentamicin/ciprofloxacin was tested by the checkerboard method. The dominant components (> 10%) of the essential oils were: cis-piperitone epoxide and menthone (C. sylvatica), pulegone and menthone (C. vardarensis), pulegone and piperitenone (C. nepeta), pulegone, piperitenone, menthone and piperitone (C. glandulosa). EOs did not exhibit significant antimicrobial activity except the essential oil of C. vardarensis which was selectively active against Staphylococcus aureus (MIC - 21.25 mu g/mL). The overall effect of essential oil-antibiotic combinations varied from synergistic (FICI lt = 0.5) to antagonistic (FICI >= 2) depending on the bacterial strain tested.
