AB  - Recent studies demonstrated that Rumex species exhibit various
pharmacological activities, such as antiinflamatory, antioxidant, anticancer,
antimicrobial, but there are almost no data on pharmacological activities of Balcan
endemic species Rumex balcanicus. Thus, the aim was to examine antioxidant
activity, using DPPH and FRAP method, antimicrobial activity via broth
microdilution method and α-amylase inhibitory activity using DNS reagent of
aqueous and methanol extracts of roots and aerial parts of R. balcanicus, collected on
Vlasina Lake in 2014. All tested extracts exhibited high antioxidant activity which
has been correlated with the high content of polyphenols. Aqueous extracts exhibited
lower activity compared to methanol extracts. Methanol extract ofR. balcanicus roots
showed the highest activity, both in DPPH (IC50= 9.57 μg/mL) and FRAP test
(IC50=3.98 μmol Fe/mg of extract), while aqueous extract of aerial parts showed the
lowest activity (IC50= 24.73 μg/mL; IC50=2.11 μmol Fe/mg). Only methanol extract
of aerial parts showed α-amylase inhibitory activity with IC50=252 μg/mL. All
extracts had low antimicrobial activity against almost all tested bacterial strains except
against Bacillus subtilis. Positive results obtained in this study show that R. balcanisus
contains potential as a new raw material and for the development of drugs efficient in
treating diabetes.

AB  - Rumex balcanicus Rech.f. (Polygonaceae) is an endemic species that inhabits
central parts of Serbia and Kosovo. The genus Rumex is characterizied by the
accumulation of anthraquinones, naphthalene-1,8-diols, flavonoids and stilbenoids.
Aerial parts of R. balcanicus, collected on Vlasina Lake (Serbia) in 2014, were used
to determine the chemical composition and cytotoxic activity. Cyclohexane,
dichloromethane, methanol and aqueous extracts were prepared and tested using
MTT-test, against human epithelial cervical cancer cells (HeLa), human colon
carcinoma cell line (LS174), human non-small cell lung cancer cells (A549) and
healthy MRC-5 human embryonic lung fibroblast cell line. Methanol and aqueous
extracts were analyzed using LC-MS. In tested extracts, caffeic acid derivates and
flavonol derivatives were identified. The main components in both extracts were rutin,
miquelianin, quercitrin and quercetin 3-O-acetyl-rhamnoside. Caffeic acid and its
glycosides were also present in analyzed extracts. Aqueous extracts had no
cytotoxicity against tested cell lines, while cyclohexane, dichlormethane and
methanol extracts were active against all tested cell lines with IC50 ranged from
116.48±3.26 μg/mL to 178.21±1.30 μg/mL. Due to the presence of flavonoids and
exhibited cytotoxic effect, R. balcanicus herba could be interesting as a plant material
for further pharmacological analysis and isolation of the anti-tumor compounds.

AB  - The aim of study was to model and optimize the extraction of polyphenols and flavonoids with expressed antioxidant activity from chickpea seeds using a central composite design. The optimal extraction conditions were the extraction time of 145.5 min, ethanol concentration of 83.7% and liquid-to-solid ratio of 24.9 (expressed as cm(3) per g). The content of total polyphenol and flavonoid was 2.75 g gallic acid equivalent per kilogram of the dried extract and 0.147 g rutin equivalent per kilogram of the dried extract, respectively. Using 2,2-diphenyl-1-picrylhydrazyl assay, it was found that the optimal extract had the IC50 of 1.55 mg cm(-3).

AB  - The essential oil from birch (Betula pendula Roth.) buds was obtained by Clevenger-type hydrodistillation. Its qualitative and quantitative composition was determined by gas chromatography in combination with mass spectrometry (GC/MS) and flame ionization detection (GC/FID), respectively. Twenty-seven compounds, mainly sesquiterpene hydrocarbons (78.7%) and oxygenated sesquiterpenes (14.8%), were identified comprising 93.5% of total identified components in the essential oil. The most abundant compounds were germacrene D (21.7%) and -cadinene (17.0%). Antimicrobial activity of isolated essential oil against Gram-positive and Gram-negative bacteria as well as fungus Candida albicans was determined using the disc-diffusion method. The isolated essential oil has shown antimicrobial activity against the following microorganisms: Proteus vulgaris, Bacillus luteus and Klebsiella pneumoniae. Antioxidative activity of the essential oil was determined using the DPPH assay, after adding DPPH radical and after 20min, 30min and 60min incubation with radical. Essential oil showed antioxidant activity.

abstract = "The essential oil from birch (Betula pendula Roth.) buds was obtained by Clevenger-type hydrodistillation. Its qualitative and quantitative composition was determined by gas chromatography in combination with mass spectrometry (GC/MS) and flame ionization detection (GC/FID), respectively. Twenty-seven compounds, mainly sesquiterpene hydrocarbons (78.7%) and oxygenated sesquiterpenes (14.8%), were identified comprising 93.5% of total identified components in the essential oil. The most abundant compounds were germacrene D (21.7%) and -cadinene (17.0%). Antimicrobial activity of isolated essential oil against Gram-positive and Gram-negative bacteria as well as fungus Candida albicans was determined using the disc-diffusion method. The isolated essential oil has shown antimicrobial activity against the following microorganisms: Proteus vulgaris, Bacillus luteus and Klebsiella pneumoniae. Antioxidative activity of the essential oil was determined using the DPPH assay, after adding DPPH radical and after 20min, 30min and 60min incubation with radical. Essential oil showed antioxidant activity.",

This work presents the selective extraction of EA, as a natural antioxidant, from the aerial parts of Alchemilla vulgaris L. EA and A. vulgaris extract are incorporated into p(NIPAM) and p(NIPAM-HPMet) hydrogels and characterized by FTIR method. The EA content in the extract by the UHPLC-DAD-HESI-MS/MS method is determined (0.64mgcm(-3)). The total flavonoids content in the A. vulgaris extract was determined by the spectrophotometric method. Antioxidant activity of the A. vulgaris extract and EA is examined using the 2,2-diphenyl-1-picrylhydrazyl assay. The p(NIPAM-HPMet) shows a better incorporation and release at 37 degrees C of EA standard and A. vulgaris extract (98.87 and 96.45% respectively), compared with p(NIPAM). [GRAPHICS]

Elemental composition and nutritional value of three edible mushrooms from Serbia

AB  - Nutrition value, as well as macro- and microconstituents of three edible mushrooms (Pleurotus ostreatus, Lentinula edodes, Macrolepiota procera) collected in Serbia were tested. It has been determined that all three mushroom samples were low in energy, fat and carbohydrates, but rich in β-glucans content. The most abundant elements in these three mushrooms were essential marcroelements Ca, K and P, but also there is a significant amount of Li, Se and Zn. On the other hand, toxic elements, such as Pb, As and Cd were also detected in level higher than allowed.
AB  - Nutritivna vrednost, kao i sadržaj makro- i mikroelemenata testirana je u uzorcima tri jestive pečurke, sakupljene u Srbiji (Pleurotus ostreatus, Lentinula edodes, Macrolepiota procera). Pokazano je da su sve tri gljive bile siromašne energijom, mastima i ugljenimhidratima, ali bogate β-glukanima. Najzastupljeniji elementi u sva tri uzorka bili su esencijalni makroelementi Ca, K i P, ali je takođe je detektovana značajna količina Li, Se i Zn. Sa druge strane, koncentracija toksičnih metala, Pb, As i Cd, bila je iznad dozvoljenih granica.

Conceptual model of collaborative pharmaceutical practice in healthcare and social care for the elderly

AB  - Background/Aim. In the new millennium, the focus has been increasingly shifting to optimisation by enhancing the collaborative (common, joint) practice of healthcare professionals, for the purpose of achieving effectiveness and efficiency. Pharmacists are the last link in the healthcare services providing chain. The aim of this study was to present a critical analysis of the published models of the collaborative pharmacy practice along with development of a conceptual model of collaborative pharmacy practice in the healthcare and social care for the elderly population. Methods. Using two search algorithms that were created to search articles published in English, a comprehensive search of the bibliographic databases Web of Science and PubMed was undertaken (up to June 2015). Afterwards, articles were independently assessed by two authors, against predetermined inclusion and exclusion criteria. Results. Regulations on pharmacy collaboration are present in many developed countries. However, the implementation of the collaborative practice is still not widespread. Therefore, a conceptual model of the collaborative healthcare and social care of the elderly provides an insight into a multi-layer structure that has to be established in order to achieve a functioning system of the collaborative healthcare practice. The model concluded that aspirations towards teamwork, communication and above all - the system of regulators and payers, who acknowledge a healthcare collaboration, are crucial for establishment of a collaborative healthcare practice. Conclusion. This research provides a tool in the form of a guide and check-list for decision-makers and policy-makers in order to achieve the preferred effects generated from the collaborative practice by selecting the models and activities that need to be undertaken for implementation of the collaborative healthcare and social care of the elderly that is best suited for their country.
AB  - Uvod/Cilj. U cilju postizanja efektivnosti i efikasnosti, u novom milenijumu se sve više govori o optimizaciji zdravstvene zaštite, kroz jačanje udružene prakse zdravstvenih profesionalaca. Farmaceuti predstavljaju poslednju kariku u lancu pružanja zdravstvene zaštite. Cilj ove studije bio je da prikaže kritičku analizu publikovanih modela kolaborativne farmaceutske prakse, kao i razvoj konceptualnog modela farmaceutske kolaborativne prakse u zdravstvenoj i socijalnoj zaštiti starije populacije. Metode. Koristeći dva algoritma koja su kreirana za pretragu, pretraženi su radovi (do juna 2015. godine) iz bibliografija elektronskih baza podataka Web of Science i PubMed. Nakon pretrage, radovi su nezavisno procenjeni u odnosu na predefinisane kriterijume za uključenje i isključenje, od strane dva autora. Rezultati. Regulativa o farmaceutskoj kolaboraciji je zastupljena u mnogim razvijenim zemljama. Međutim, implementacija kolaborativne prakse još uvek nije široko rasprostranjena. Stoga, konceptualni model kolaborativne farmaceutske prakse u zdravstvenoj i socijalnoj zaštiti starijih osoba obezbeđuje uvid u višeslojnu strukturu koja treba biti uspostavljena kako bi se osigurao funkcionalni sistem kolaborativne zdravstvene prakse. Model zaključuje da su aspiracije prema timskom radu, komunikacija i nadasve sistem regulatora i platioca koji prepoznaju zdravstvenu kolaboraciju, ključni za uspostavljanje kolaborativne zdravstvene prakse. Zaključak. Ovo istraživanje donosi alat u obliku vodiča i čekliste za donosioce odluka i regulativa, kako bi se istim omogućilo da postignu preferirane efekte prikupljenih kolaborativnih praksi, kroz označavanje modela i aktivnosti koje treba preduzeti za implementaciju one kolaborativne zdravstvene i socijalne zaštite starijih osoba koji su najprimenjiviji u njihovoj zemlji.

Farmaceuti predstavljaju poslednju kariku u lancu pružanja zdravstvene zaštite. Cilj ove studije bio je da prikaže kritičku analizu publikovanih modela kolaborativne farmaceutske prakse, kao i razvoj konceptualnog modela farmaceutske kolaborativne prakse u zdravstvenoj i socijalnoj zaštiti starije populacije. Metode. Koristeći dva algoritma koja su kreirana za pretragu, pretraženi su radovi (do juna 2015. godine) iz bibliografija elektronskih baza podataka Web of Science i PubMed. Nakon pretrage, radovi su nezavisno procenjeni u odnosu na predefinisane kriterijume za uključenje i isključenje, od strane dva autora. Rezultati. Regulativa o farmaceutskoj kolaboraciji je zastupljena u mnogim razvijenim zemljama. Međutim, implementacija kolaborativne prakse još uvek nije široko rasprostranjena. Stoga, konceptualni model kolaborativne farmaceutske prakse u zdravstvenoj i socijalnoj zaštiti starijih osoba obezbeđuje uvid u višeslojnu strukturu koja treba biti uspostavljena kako bi se osigurao funkcionalni sistem kolaborativne zdravstvene prakse. Model zaključuje da su aspiracije prema timskom radu, komunikacija i nadasve sistem regulatora i platioca koji prepoznaju zdravstvenu kolaboraciju, ključni za uspostavljanje kolaborativne zdravstvene prakse. Zaključak. Ovo istraživanje donosi alat u obliku vodiča i čekliste za donosioce odluka i regulativa, kako bi se istim omogućilo da postignu preferirane efekte prikupljenih kolaborativnih praksi, kroz označavanje modela i aktivnosti koje treba preduzeti za implementaciju one kolaborativne zdravstvene i socijalne zaštite starijih osoba koji su najprimenjiviji u njihovoj zemlji.",

Chemical composition and biological activity of the plum seed extract

AB  - The aim of this paper was to estimate the biological activity of the plum seed extract and to define the chemical composition by using the ESI-MS method. During the investigation of the antioxidant activity, the extract showed a better ability to inhibit DPPH radicals compared with amygdalin standard. The results of the antimicrobial study indicate that the extract has a greater effect on Gram-negative bacteria compared with amygdalin. Gram-positive bacteria and fungi remained resistant in both cases. Unlike amygdalin, the extract showed a greater antimicrobial activity against the same microbiological strains. Amygdalin was isolated from the extract before the investigation of antiproliferative activity against MDA-MB-361, MDA-MB-453, HeLa and LS174 cell lines using MTT assay. Unlike the MDA-MB-453 cell line, the activity of standard and isolated amygdalin was not observed at the concentrations lower than 400 μg cm-3 on MDA-MB-361 cell line. The isolated amygdalin has not shown the activity in other cell lines. The antiproliferative activity of amygdalin standard was also noticed on HeLa and LS-174 cell lines at the concentrations of 333.27 and 333.17 μg cm-3, respectively.
AB  - Cilj ovog rada bila je procena biološke aktivnosti ekstrakta semena šljive i definisanje hemijskog sastava primenom ESI-MS metode. Tokom ispitivanja antioksidativne aktivnosti, ekstrakt je pokazao bolju sposobnost inhibiranja DPPH radikala u poređenju sa standardom amigdalina. Fröhner), A. fissa Günther & Schummel (A. sect. Calycinae (Buser) Buser) and A. viridiflora Rothm. (A. sect. Calycinae), and identified ellagic acid and
quercetin-3-O-β-D-glucoside. Anti-H. pylori activity was tested against ten clinical isolates and one reference strain (ATCC 43504). The methanol extracts
were more active than the dichloromethane and cyclohexane extracts. The ranges of concentrations were between 4 μg/mL for methanol extracts of
A. viridiflora, A. glabra and A. monticola, and 256 μg/mL for cyclohexane extracts of A. viridiflora, A. glabra and A. fissa. The best overall activity was
obtained with A. monticola extracts. No significant difference was found in the ellagic acid contents of the methanol extracts of the tested Alchemilla species
(0.2-0.3 mg/mL), and anti-H. pylori activity was similar (4 -32 μg/mL). Ellagic acid exhibited strong activity at very low concentrations (0.125-0.5
μg/mL), while the second identified compound, quercetin-3-O-β-D-glucoside, was also very active in concentration of 2-16 μg/mL.

AB  - The commercial samples of aerial parts of Origanum vulgare L. and Ocimum basilicum L. (Lamiaceae) were tested for antimicrobial activity. The activity of the extracts with different polarity was tested against a panel of microorganisms, including laboratory strain Helicobacter pylori NCTC 12868. The tested extracts showed a moderate activity. The extracts of O. vulgare were more active against bacteria, especially against Gram positive bacteria with the minimal inhibitory concentration (MIC) between 62.5 and 125 μg/mL, than the extracts of O. basilicum which were active against Candida albicans (MIC 125 μg/mL). Cyclohexane extract of O. vulgare did not show any activity against tested H. pylori, while all other tested extracts were active with MICs between 250 μg/mL and 500 μg/mL. Identified and quantified rosmarinic acid and other polar compounds could be active antibacterial compounds in these spices.
AB  - Komercijalni uzorci nadzemnih delova Origanum vulgare L. i Ocimum basilicum L. (Lamiaceae) su korišćeni za određivanje antimikrobne aktivnosti. Aktivnost ekstrakata različite polarnosti je testirana protiv izabranih mikroorganizama, uključujući laboratorijski soj Helicobacter pilori NCTC 12868. Testirani ekstrakti su imali umerenu aktivnost. Ekstrakti O. vulgare su bili aktivniji protiv bakterija, posebno Gram pozitivnih bakterija sa minimalnim inhibitornim koncentracijama (MIK) između 62,5 i 125 μg/mL, od ekstrakata O. basilicum koji su pokazali aktivnost protiv Candida albicans (MIK 125 μg/mL). Cikloheksanski ekstrakt O. vulgare nije pokazao antimikrobnu aktivnost na testirani H. pylori, dok su ostali testirani ekstrakti bili aktivni sa MIK vrednostima između 250 μg/mL i 500 μg/mL. Identifikovana i kvantifikovana ruzmarinska kiselina i druga polarna jedinjenja mogu predstavljati aktivna jedinjenja sa antibakterijskom aktivnošću u ovim začinima.

The extracts of O. vulgare were more active against bacteria, especially against Gram positive bacteria with the minimal inhibitory concentration (MIC) between 62.5 and 125 μg/mL, than the extracts of O. basilicum which were active against Candida albicans (MIC 125 μg/mL). Cyclohexane extract of O. vulgare did not show any activity against tested H. pylori, while all other tested extracts were active with MICs between 250 μg/mL and 500 μg/mL. Identified and quantified rosmarinic acid and other polar compounds could be active antibacterial compounds in these spices., Komercijalni uzorci nadzemnih delova Origanum vulgare L. i Ocimum basilicum L. (Lamiaceae) su korišćeni za određivanje antimikrobne aktivnosti. Aktivnost ekstrakata različite polarnosti je testirana protiv izabranih mikroorganizama, uključujući laboratorijski soj Helicobacter pilori NCTC 12868. Testirani ekstrakti su imali umerenu aktivnost. Ekstrakti O. vulgare su bili aktivniji protiv bakterija, posebno Gram pozitivnih bakterija sa minimalnim inhibitornim koncentracijama (MIK) između 62,5 i 125 μg/mL, od ekstrakata O. basilicum koji su pokazali aktivnost protiv Candida albicans (MIK 125 μg/mL). Cikloheksanski ekstrakt O. vulgare nije pokazao antimikrobnu aktivnost na testirani H. pylori, dok su ostali testirani ekstrakti bili aktivni sa MIK vrednostima između 250 μg/mL i 500 μg/mL. A full factorial design in the formulation of diazepam parenteral nanoemulsions: Physicochemical characterization and stability evaluation

AB  - Using the experimental design methodology, we have developed and characterized nanoemulsions for a parenteral delivery using diazepam as a model drug. The formulations containing 20 or 30% (w/w) of medium chain triglycerides or the mixture of medium chain triglycerides and soybean oil as the oil phase, soybean lecithin and polysorbate 80 as emulsifiers, and a phosphate buffer solution as the aqueous phase were prepared by cold high pressure homogenization. The obtained nanoemulsions were evaluated in terms of droplet size, size distribution, surface charge, drug-vehicle interactions and physical stability. To evaluate the effects of the oil phase type, oil content and drug presence, as well as their interactions on critical quality attributes of nanoemulsions, a three-factor two-level full factorial design was applied. After the preparation, all nanoemulsions revealed small spherical droplets in the range 170-210 nm, with the narrow droplet size distribution ( lt  0.15) and the surface charge about -60 mV. The experimental design results indicated that not only factors alone (oil type, oil content, presence of drug), but their interactions also had a significant effect on the nanoemulsion droplet size, polydispersity index, and zeta potential. During two months of storage at 25°C, all nanoemulsions formulated with the medium chain triglycerides-soybean oil mixture (4:1, w/w) remained physically stable, without considerable changes in monitored parameters. Physicochemical characteristics and stability of these nanoemulsions demonstrated their suitability for parenteral drug delivery.
AB  - Cilj ovog rada bio je da se primenom metodologije eksperimentalnog dizajna razviju parenteralne nanoemulzije sa diazepamom kao model lekovitom supstancom i da se sprovede njihova sveobuhvatna fizičkohemijska karakterizacija. Metodom homogenizacije pod visokim pritiskom na sobnoj temperaturi izrađene su placebo i nanoemulzije sa lekom, stabilizovane smešom lecitina i polisorbata 80, variranjem udela i vrste uljane faze - 20 i 30% (m/m) triglicerida srednje dužine lanca ili smeše triglicerida srednje dužine lanca i sojinog ulja u odnosu 4:1. Dobijene nanoemulzije okarakterisane su u pogledu veličine i raspodele veličina kapi, površinskog naelektrisanja, interakcija lek-nosač i fizičke stabilnosti. U cilju procene istovremenog uticaja vrste uljane faze, udela ulja i prisustva leka, kao i njihovih interakcija, na kritične atribute kvaliteta nanoemulzija, primenjen je pun faktorski dizajn sa tri faktora na dva nivoa. Nakon izrade, sve formulacije nanoemulzija imale su malu veličinu kapi u opsegu 170-210 nm, sa veoma uskom raspodelom veličina (ispod 0,15) i površinskim naelektrisanjem oko -60 mV. Rezultati eksperimentalnog dizajna pokazali su da ne samo pojedinačni faktori (vrsta ulja, koncentracija ulja, prisustvo leka), nego i njihove interakcije, značajno utiču na veličinu kapi, indeks polidisperznosti i zeta potencijal ispitivanih nanoemulzija. Tokom 2 meseca čuvanja na 25°C, sve nanoemulzije formulisane sa smešom triglicerida srednje dužine lanca i sojinog ulja kao uljanom fazom bile su fizički stabilne, bez značajnih promena u praćenim parametrima. Fizičkohemijske karakteristike i stabilnost navedenih nanoemulzija ukazuju da one mogu biti potencijalni nosači za parenteralnu isporuku lekovitih supstanci.

Antioxidant and antimicrobial activity of a new generation phyto-gel

AB  - Antioxidant and antimicrobial activities of a new phyto-gel were studied in this work. The formulated gel is composed of propilene glycol extracts of St. John's worth (H. perforatum L.), comfrey (S. officinale L.), yarrow (A. millefolium L.), horsetail (E. arvense L.) and Na-usniate. The antioxidant activity was determined for the gel and each investigated extract of the gel separately by the DPPH test. The total phenols and flavonoids content was determined spectrophotometrically, for the gel and each investigated extracts. The antimicrobial activity of the gel and each compound separately was tested in vitro on the following microorganisms: Staphylococcus aureus, Micrococcus luteus, Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger. The antioxidant activity of the investigated samples decreases in the series: H. perforatum (EC50 = 1.04 mg/mL) > Na-usniate (EC50 = 4.16 mg/mL) > A. millefolium (EC50 = 7.60 mg/mL) > gel (EC50 = 23.47 mg/mL) > S. officinale (EC50 = 30.10 mg/mL) > E. arvense (EC50 = 44.70 mg/mL). The formulated phyto-gel showed the antimicrobial activity. The results suggest that the obtained phyto-gel is a good source of natural antioxidants and antimicrobial agents, and it has potential as an alternative to synthetic products in pharmaceutical and cosmetic industries.
AB  - U radu je ispitivana antioksidativna i antimikrobna aktivnost fitogela - formulacija gela sa propilen-glikolnim ekstraktima H. perforatum, S. officinale, A. millefolium, E. arvense i Na-usninatom. Optimization of technological procedure for amygdalin isolation from plum seeds (Pruni domesticae semen)

AB  - The process of amygdalin extraction from plum seeds was optimized using central composite design (CCD) and multilayer perceptron (MLP). The effect of time, ethanol concentration, solid-to-liquid ratio, and temperature on the amygdalin content in the extracts was estimated using both mathematical models. The MLP 4-3-1 with exponential function in hidden layer and linear function in output layer was used for describing the extraction process. MLP model was more superior compared with CCD model due to better prediction ability. According to MLP model, the suggested optimal conditions are: time of 120 min, 100% (v/v) ethanol, solid-to liquid ratio of 1:25 (m/v) and temperature of 34.4 degrees C. The predicted value of amygdalin content in the dried extract (25.42 g per 100 g) at these conditions was experimentally confirmed (25.30 g per 100 g of dried extract). Amygdalin (>90%) was isolated from the complex extraction mixture and structurally characterized by FT-IR, UV, and MS methods.

лeчeњу мaлигних тумoрa, oвa бoлeст и дaљe односи велики број живота. Дaнaс сe нa
тржишту мoгу нaћи брojни хeмoтeрaпeутици у циљу лeчeњa oвe бoлeсти. Због своје
токсичности вeћинa лекова и лековитих производа нe мoжe да се aпликује у дoзaмa
кoje у пoтпунoсти сузбиjају рaст ћелија тумора у oргaнизму. Зa трeтмaн oвe бoлeсти
данас се примeњује велики број фармаколошки активних aгeнaсa изолованих из
биљака, мeђу кojимa су квeрцeтин и aмигдaлин.
Стога је циљ ове дисертације њихова изолација из биљних материјала (листа
зеленог чаја и семена шљиве). У раду je испитан утицај времена екстракције, природе
растварача и односа биљне сировине и растварача на принос кверцетина, односно
укупних флавоноида из листа зеленог чаја применом техника експерименталног
дизајна и вештачких неуронских мрежа. Код екстракције амигдалина из семена шљиве
поред наведених параметара испитан је и утицај температуре екстракције. Садржај
амигдалина и кверцетина у добијеним екстрактима одређен је развијеним HPLC
методама. Садржај укупних флавоноида испитан је спектрофотометријском методом,
која се заснива на грађењу комплекса са алуминијум хлоридом.
Амигдалин је изолован из етанолног екстракта семена шљиве након додатка
диетил-етра. Чистоћа овако добијеног амигдалина је 90% и дефинисана је у односу на
расположиви стандард применом HPLC методе. Структурна карактеризација
изолованог амигдалина извршена је применом UV, IC и ESI-MS/MS метода.
У циљу одређивања фармаколошких активности екстраката зеленог чаја и
семена шљиве, изолованог aмигдaлинa, стандарда квeрцeтинa и амигдалина примeњене
су одговарајуће методе за одређивање антиоксидативне (DPPH тест), aнтимикрoбне
(диск дифузиона метода) и aнтипролиферативне активности (МТТ тест).
На основу ових фармаколошких испитивања показано је да стандард кверцетина
и екстракт зеленог чаја имају бољу антиоксидативну активност у односу на стандард
амигдалина и екстракт семена шљиве. Микробиолошким испитивањима утврђено је да
стандард амигдалина и екстракт семена шљиве показују антимикробну активност само
на сојевима Pseudomonas aeruginosa (>23 mm код оба узорка) и Escherichia coli (17-19
mm и 20-22 mm, респективно). Стандард кверцетина и екстракта зеленог чаја показују
додатно дејство и на сој Staphylococcus aureus са зонама инхибиције 20-22 mm и 17-19
mm, респективно.
МТТ тестом утврђено је да су добијене IC50 вредности за изолат и стандард
амигдалина (>379,5 μg cm-3 и 304,65-333,27 μg cm-3, респективно) на испитиваним
ћелијским линијама више у поређењу са екстрактом зеленог чаја и стандардом
кверцетина (47,41-114,39 μg cm-3 и 19,24-44,07 μg cm-3, респективно). Вредности за IC50
су очекиване и у складу са литературним подацима. Цитотоксичност амигдалина може
да се повећава уз додатак ензима β-D-глукозидазе.
AB  - The plant extracts and compounds isolated from natural sources are commonly used
for prevention of chronic diseases. These bioactive compounds have an increased application
in modern conventional medicine. Despite a great progress in the treatment of malignant
tumors, this disease still takes many lives. Today, a huge number of chemotherapeutics for
tumor treatment can be found in the market. Because of their toxicity most drugs and drug
products can not be applicated in the doses that completely inhibit the cells growth in
organism. Quercetin and amygdalin belong to the most commonly used pharmacological
agents for tumor treatment.
Therefore, the subject of this dissertation is isolation of amygdalin and quercetin from
two plant materials, i.e. green tea leaves and plum seeds. In this study, the effect of extraction
time, solvent nature and solid to liquid ratio was investigated for quercetin, i.e. total
flavonoids from green tea leaves using experimental design and artificial neural network. In
addition to these parameters, the impact of extraction temperature was also studied during
amygdalin extraction from plum seeds. The content of amygdalin, as well as quercetin in the
extracts was determined using the developed HPLC methods. Total flavonoids content was
determined by a spectrophotometric method based on the formation of complex with
aluminium chloride.
Amygdalin was isolated from the ethanolic extract of plum seeds after addition of
diethyl-ether. The obtained amygdalin purity of 90% was defined with regard to the available
amygdalin standard using HPLC method. The structural characterization of isolated
amygdalin was carried using UV, IR and ESI-MS/MS methods.
Adequate methods for determination of antioxidant activity (DPPH assay),
antimicrobial activity (disc diffusion method) and antiproliferative activity (MTT assay) were
used in order to define the pharmacological activities of green tea and plum seeds extracts,
isolated amygdalin, quercetin and amygdalin standards.
Based on all these pharmacological studies, it was shown that quercetin and green tea
extract have better antioxidant activity compared to amygdalin and plum seeds extract.
Amygdalin standard and plum seeds extract have antibacterial activity only on the strain of P.
аeruginosa (>23 mm for both samples) and E. coli (17-19 mm and 20-22 mm, respectively).
Quercetin standard and green tea extract have an additional effect on the strain of S. aureus
with an inhibition zone of 20-22 mm and 17-19 mm, respectively.
IC50 values of isolated amygdalin and its standard (>379.5 μg cm-3 and 304.65-333.27
μg cm-3, respectively) for the investigated cell lines were higher compared to green tea
extract and quercetin standard (47.41-114.39 μg cm-3 and 19.24-44.07 μg cm-3, respectively).
These values were determined using MTT assay and are in accordance with the literature.
The cytotoxicity of amygdalin can be increased by addition of β-D-glucosidase enzyme.

AB  - New formulation for treatment a copper deficiency in human organism was developed and optimized by application of mathematical modeling. This formulation contained copper (II) complex with polysaccharide pullulan, as active substance. The binder concentration [polyvinyl pyrrolidone (PVP %)], the disintegrant concentration (corn starch %) and the resistance to crushing (hardness) were taken as independent variables. In vitro measured drug release characteristics of the tablets at pH 1.20 and 7.56 were studied as response variables. Initially, the created full factorial 2(3) model showed that the resistance to crushing has the most significant effect on copper (II) complex release from the formulation. Optimal tablet formulation F2, with lower Hardness (50 N), lower Starch (20.0%) and higher PVP (2.7%) concentrations, is selected using the partial least squares (PLS) regression modeling. The selected formulation F2 has expressed the best drug release profile at both pH (98.66% pH = 1.20; 93.35% pH = 7.56), and the lowest variation of tablets weight. The presented theoretical approach and created PLS model can be readily applied in future copper complexes studies and formulation design.

Inclusion complexes with cyclodextrin and usnic acid

AB  - Molecular inclusion complexes of usnic acid (UA) with beta-cyclodextrin (beta-CD) and 2-hydroxypropyl beta-cyclodextrin (HP beta-CD) were prepared by the co-precipitation method in the solid state in the molar ratio of 1:1. Structural complexes characterization was based on different methods, FTIR, H-1 NMR, XRD and DSC. Parallel to the complex by the above methods, corresponding physical mixtures of UA with cyclodextrins and complexing agents (beta-CD, HP beta-CD and UA) were analyzed. The results of DSC analysis showed that, at around 200 A degrees C, the endothermal peak in the complexes with cyclodextrins originating from the UA melting has disappeared. Complex diffractogram patterns do not contain peaks characteristic for the pure UA. They are more appropriate to cyclodextrin diffractogram. This fact points to the molecular encapsulation of UA in the cyclodextrin cavity. Chemical shifts in H-1 NMR spectra after the inclusion of UA into the cyclodextrin cavity, especially H-3 protons (0.0012 and 0.0102 ppm in the beta-CD and HP beta-CD, respectively) and H-5 and H-6 (0.0134 ppm) and hydrogen from CH3 (0.0073 ppm) HP beta-CD also points to the formation of molecular inclusion complexes. The improved solubility of UA in water was achieved by molecular incapsulation. In the complex with beta-CD the solubility is 0.3 mg/cm(3), with HP beta-CD 4.2 mg/cm(3) while the uncomplexed UA solubility is 0.06 mg/cm(3). The microbial activity of UA and both complexes was tested against eight bacteria and two fungi and during the test no reduced activity of UA in the complexes was observed.

AB  - The aim of this study was to design and optimize a new tablet formulation for treatment of copper deficiency in human organism by using an experimental design. The new no-veneered tablets, prepared by a wet granulation technique, are containg active substance, a copper(II) complex with polysaccharide pullulan. The binder concentration, the disintegrant concentration and the resistance to crushing were used as independent variables in the formulation; while in vitro measured drug release characteristics of the tablets was response variable in a full factorial design 2(3) modeling. A cubic model for data fitted was used to examine the obtained results. They showed that the resistance to crushing has the most significant effect on copper(II) complex release from the formulation, while the disintegrant concentration has smaller influence on dissolution profile of copper(II) complex and the binder concentration had minor impact in this study. Lower value of resistance to crushing has influence on better dissolution profile. Furthermore, physical characteristics of the tablets were evaluated, viz., drug content, hardness, thickness, friability, disintegration time, mass variation, particle size and size distribution.

AB  - Quality by design (QbD) is a concept first outlined by Juran, who believed that quality could be planned and that most quality crises and problems relate to the way in which quality was planned in the first place. Experimental design is a powerful technique and tool for QbD, used for exploring new processes, gaining increased knowledge of the existing processes and optimizing these processes for achieving internationally competitive performance. It is also used for the investigation of relationship between parameters of ill-defined process. In this paper, the experimental design principles in pharmaceutical development and impact of these principles on pharmaceutical legislation have been reviewed. Also, slow implementation of QbD in pharmaceutical industries has been discussed. Pharmaceutical legislation is necessary for companies to continue benefiting from knowledge gained and to continually improve throughout the process lifecycle by making adaptations to assure that root causes of manufacturing problems are quickly corrected.

AB  - Background: Venous ulcers (ulcus cruris venosum) are a common chronic disease that requires continuing treatment and significantly influences a patient's way of life. The therapeutic effects of the ointment Herbadermal on epithelialization and microbial flora of venous ulcers in 25 patients (10 men and 15 women) were tested over a 7-week period. The major components of the ointment are extracts of garlic, St. John's wort, and calendula. Material and Methods: The patients were over 18 years of age, with ulceration of the lower leg no longer than 2 months or recurrent ulceration during the last 6 months. The involved patients did not use any other phytomedicines or supportive therapies. Parameters were evaluated before the treatment and every 2 weeks during the period of 7 weeks. The total treatment response was evaluated on the basis of epithelialization, granulation, fibrin deposits, exudation, and edema. Results: The percentage of epithelialization was 99.1% after 7 weeks, without significant effects on the microbial flora. Conclusion: This combination of extracts can be recommended as topical treatment for wound healing because of its epithelizing, anti-erythematous, and anti-edematous properties.

Antimicrobial activity of lozenge with garlic bulb powder

AB  - According to the tested antimicrobial activity of constituents and their chemical characteristics, a new formulation of oral antiseptic was made based on garlic bulb powder and its active principles with strong antimicrobial activity against a wide range of bacteria and fungi. The antimicrobial activity of garlic bulb powder, allicin and the lozenge with 15% of garlic powder was tested using broth microdilution method. The tested garlic powder, as well the lozenge, have shown very high antimicrobial activity with MIC 1.25-5.00 mg/ml, and 1.87-7.50 mg/ml, respectively. The major compound, allicin, was highly active at a very low concentration. The minimal inhibitory concentration of allicin was from 6.25-12.50 μg/ml for antibacterial activity and 0.4 μg/ml for antifungal activity. Those concentrations are comparable with concentrations of commercially available antibiotics and fungicides. The formulation of an antiseptic with herbal constituents, high antibacterial and antifungal activity and pleasant taste could be an alternative to classical pharmaceutical oral antiseptics.
AB  - Na osnovu ispitane antimikrobne aktivnosti komponenata i njihovih hemijskih karakteristika, napravljena je nova formulacija oralnog antiseptika koji kao aktivnu komponentu ima prašak belog luka čiji sastojci pokazuju veoma izraženu aktivnost protiv širokog spektra bakterija i gljive Candida albicans. Antimikrobna aktivnost praška belog luka, alicina i lozengi sa 15% praška belog luka je ispitivana bujon mikrodilucionom metodom. Testiran prašak belog luka, kao i lozenge imaju veoma jaku antimikrobnu aktivnost sa minimalnim inhibitornim koncentracijama (MIC) 1,25-5,00 mg/ml (prašak belog luka) i 1,87-7,50 mg/ml (lozenge). Glavna aktivna komponeta belog luka, alicin, pokazuje antimikrobnu aktivnost u veoma niskim koncentracijama. Minimalne inhibitorne koncentracije alicina se kreću od 6,25-12,5 μg/ml protiv bakterija i 0,4 μg/ml protiv gljivice Candida albicans. Ove koncentracije se mogu uporediti sa koncentracijama komercijalnih antibiotika i fungicida. Naši rezultati su u skladu sa prethodnim ispitivanjima alicina i različitih ekstrakta belog luka, i potvrđuju najvažniju ulogu alicina u antimikrobnoj aktivnosti. Veoma je važna visoka aktivnost ovako formulisanog oralnog preparata protiv gljivice C. albicans koja je čest uzročnik infekcija usne duplje. Formulisan proizvod sa antiseptičkim osobinama karakteriše stanje čvrstog rastvora gde su aktivne supstance molekularno-disperzno ugrađene u obliku dobro rastvornog polimernog matriksa (sorbitola) velike specifične površine. Dobra rastvorljivost polimernog matriksa i velika specifična površina omogućuju zadovoljavajuću brzinu otpuštanja aktivnih principa iz belog luka (alicin). Pored visoke antimikrobne aktivnosti, lozenge poseduju prijatan ukus i osvežavaju dah, pa se mogu koristiti kao prirodni antiseptik u prevenciji i terapiji infekcija sluzokože usta i ždrela. Formulacija antiseptika sa biljnim sastojcima i visokom antibakterijskom i antifungalnom aktivnošću može biti alternativa farmaceutskim oralnim antisepticima.

Composition of Satureja kitaibelii Essential Oil and its Antimicrobial Activity

AB  - The composition of the essential oil obtained by hydrodistillation from the aerial parts of Satureja kitaibelii from Rtanj mountain (Serbia), collected during three years, was studied. Thirty-nine components were identified in each sample of S. kitaibelii essential oil, representing about 87% of the oils. p-Cymene was the most dominant compound in all three oils (27.9%, 14.7% and 24.4%, respectively). The simple formulation of a lozenge with 0.2% of S. kitaibelii essential oil was prepared and the antimicrobial activity of the essential oil and the lozenge with essential oil was tested using a broth microdilution assay. Both essential oil and lozenge possessed strong antimicrobial activity with MIC values of 0.10-25 mu g/mL, and 0.97-15.6 mg/mL, respectively.
