The application of tryciclic antidepressants amitriptyline in the therapy of depression is followed by some side effects both those muscarinic and antihistaminic and alpha-adrenergic effects. The manifested degree of side effects directly correlates with the dosage and concentration in plasma, so that maximal effects are manifested when drug concentration in patient's plasma is maximal. When amitriptyline is used in the therapy an effective drug dosage enabling the maximal therapeutic effect and minimal side effects should be determined. By this the maximal outcome of the therapy is made possible.
