AB  - A method has been developed for separation of nitrendipine and its impurities of reaction partners and side reaction products by high-performance liquid chromatographic method on a RP-ls column and detection at 238 nm. The mobile phase composition that provided an acceptable nitrendipine resolution, in large excess and possible impurities, in a short elution time, is methanol:water (70:30) and pH 3. Linearity (r greater than or equal to 0.999), reproducibility (RSD = 0.8-1.4%), determination limit (0.5 2%,) and recovery (99.8-102.3) were validated and found to be satisfactory. This method enables monitoring of the process of synthesis, as well as the choice of the synthetic design.

Apolipoprotein E polymorphism and severity of angiographically verified coronary artery disease

AB  - Apolipoprotein E is an important genetic determinant of serum lipoprotein concentrations and coronary artery disease risk. The apo E polymorphism is a major determinant of interindividual variation in the initiation and progression of atherosclerosis. Differences in the gene sequence cause structural changes strongly affecting binding properties of the corresponding apo E isoforms. The presence of the epsilon 2 allele leads to LDL receptor upregulation and increased LDL clearance. In contrast the presence of the epsilon 4 allele leads to receptor down regulation and decreased LDL clearance. For most populations, individuals with the apo epsilon 2 allele display lower levels of plasma cholesterol compared with individuals carrying the apo epsilon 3 allele, whereas individuals with the apo epsilon 4 allele show higher levels of plasma cholesterol, especially LDL-cholesterol and an increased risk for cardiovascular disease. ApoE polymorphism explains near 7% of the variation in total, LDL cholesterol and apo B levels. To examine whether the polymorphism in the apo E gene is associated with CAD in our population, we analyzed 94 patients with angiographically verified CAD and in 45 patients with excluded CAD as a control group. The apo E phenotype was determined with isoelectric Focusing and Western immunoblotting technique using commercial antibodies. Patients were divided into three groups according to arteriograms as category: 1. mild, evidence of narrowing of one coronary artery >60% (39); 2. moderate, narrowing of two major coronary arteries (30) and 3. Severe, narrowing of all three major coronary arteries (25). The apo E allele frequencies of patients with CAD vs without CAD were 4.2% vs 7.7% For epsilon 2; 86.7% vs 85.5% for epsilon 3 and 9.1% vs 6.6%; for epsilon 4. The frequency of allele epsilon 4 was higher in patients with 3 vessel CAD than in patients without CAD (12% vs 6.6%) but it did not reach the statistical significance (chi(2)= 0.83, p = 0.36, d.f. 1). The Odds ratio for risk of CAD associated with the presence of apo epsilon 4 allele was 1.39 and for the phenotype E3/4 1.60. Our preliminary results suggest that the presence of apo epsilon 4 allele may impose some risk for developing CAD and that apo E determines the severity of CAD.

Low frequency of PIM3 gene in patients with monoclonal gammopathies

AB  - The distribution of PI (protease inhibitor) phenotypes and PI M subtypes was studied in 200 patients with monoclonal gammopathies and 320 healthy blood donors by isoelectric focusing in thin-layer polyacrylamide gels, pH range 4-5. The distribution of PI phenotypes and gene frequencies in the patients differed significantly from the corresponding values found in the blood donors, as shown by the chi(2) test (chi(2) = 16.5, p = 0.02 and chi(2) = 17.6, p = 0.0014, respectively). A lower frequency of the M3 variant was observed in patients, both in homozygous (PI M3) and heterozygous forms (PI M1M3 and PI M2M3). No significant difference between PIz allele frequencies in patients and healthy controls was found.

Well‐defined, diffusion‐controlled, cathodic waves were observed for F‐1 only in strong acidic solution (pH 1.25 HCl) and for F‐2 in the H range 1.25–5.33 (HCl and Britton–Robinson buffer). The overall reduction of the azomethine group to a primary amine, a process that involves the uptake of four electrons, is irreversible. For both oximes a mechanism of reduction has been proposed. The rapid and accurate determination of these quinolinium and isoquinolinium oximes is possible by dc polarography and by DPP. Limits of detection are 5 × 10−7 M for both oximes using DPP.

Potentiometric investigation of the stability of palladium(II) complex of pralidoxime chloride in aqueous solution

AB  - The formation of a complex between palladium(II) chloride and pralidoxime chloride (PAM-2Cl) has been studied by means of potentiometric pH measurements. The real stability constant of the complex in aqueous medium of ionic strength 0.3 M (KCl) at 25.0°C was log Ks = 7.29. This value was close to that (log Ks = 7.02) obtained previously by spectrophotometric methods after appropriate correction with respect to the corresponding value of the acidic constant of PAM-2Cl (pKca = 8.05), which was also determined under the same experimental conditions.

AB  - Pralidoxime chloride (PAM-2Cl) has been determined spectrophotometrically in Britton-Robinson buffer solution at pH = 6.45; the method is based on measurement of the absorbance of the Pd(II)-pralidoxime complex at 327 nm. Studies of the composition of the complex by Job's continuous variation method, the molar ratio method and Bent-French's method yielded a Pd(II):pralidoxime ratio of 1:1. The conditional stability constant (K′) of the complex at the optimum pH of 6.45 and an ionic strength (μ) of 0.3 M was found to be 105.2. The molar absorptivity was 1.05 × 104 1 mol-1 cm-1. Beer's law was obeyed at concentrations up to 60 μM. The detection limit was 0.55 μg ml-1. The relative standard deviation (N = 10) was 0.28-1.03%. The method was accurate and sensitive for the analysis of PAM-2Cl in water and tablets.

AB  - It has been established that obidoxime chloride reacts with palladium(II) chloride in the pH range 3.9-8.0 and forms a yellow water-soluble (1:1) complex with maximum absorbance at 355 nm. By applying the methods of Sommer, Nash and Job involving non-equimolar solutions the conditional stability constant of the complex, at the optimum pH of 6.3 and an ionic strength μ = 0.55 M, is found to be 105.3. The molar absorptivity at 355 nm is 3.16 × 104 l mol-1 cm-1 at pH 6.3. Beer's law is obeyed up to 20 μmol l-1 obidoxime chloride concentration. The detection limit of the method is 0.36 μg ml-1. The relative standard deviation (n = 10) varies over the range 1.10-2.66%. The proposed method was found to be suitable for the accurate and sensitive analysis of obidoxime chloride in water and injection solutions.

AB  - The polarographic behavior of 1‐[2‐phenyl‐2‐(hydroxyimino)ethyl]‐6‐methylquinolinium chloride has been investigated. The pH of the Britton‐Robinson buffer, used as supporting electrolyte, influences the position of the half‐wave potential which shifts towards more negative values with increasing pH. Well defined, diffusively controlled cathodic waves were observed in acid medium for the overall irreversible reduction of the azomethine group to a primary amino group. A single four‐electron reduction is obtained and a mechanism has been proposed for the reduction. The ratio id/c remains constant and allows a rapid and reproducible determination of the compound (CV = 0.54–2.10 %).

AB  - It has been established that 1-(2-phenyl-2-hydroxyiminoethyl)-1-quinolinium chloride, 1-(2-phenyl-2-hydroxyiminoethyl)-1-isoqui-nolinium chloride, 1-(2-phenyl-2-hydroxyiminoethyl)-1-(4′-methyl)-quinolinium chloride and 1-(2-phenyl-2-hydroxyiminoethyl)-1-(6′-methyl)quinolinium chloride react with palladium(II) chloride in the pH range 3.3-7.1 and form yellow water-soluble 1:1 complexes with maximum absorbance at 413 nm. The conditional stability constants of the complexes at the optimum pH of 6.5 are all about 104.7, and the molar absorptivities are in the range 2.2-2.6×103 l·l mole-1·cm-1 at pH 6.5 and 413 nm. Beer's law is obeyed up to 3-4×10-4M oxime concentration, depending on the oxime determined.
