AB  - Humans are exposed to the chemicals from very different sources. Therefore, the
exposure assessment should be focused on mixtures rather than single chemicals. In this
study, bioelements (Zn and Cu) homeostasis effects of mixture of Cd and BDE-209, widely
spread persistent environmental pollutants, has been evaluated. In order to evaluate the
potential of mixture on Zn and Cu homeosthasis in rats kidneys, Wistar rats were treated by
oral gavage during 28 days with three doses of Cd 2.5, 7.5, and 15 mg/bw/day, three
combinations of Cd with 1000 mg BDE-209/kg/day, and water as a control group. Kidney
tissue samples were collected, mineralised, and Cu and Zn levels were measured by flame
atomic absorbtion spectrophotometer. Possible interactions were evaluated comparing the
dose-response curves slopes and lower confidence limit of Benchmark dose of 10%
(BMDL10) were calculated by PROAST software (RIVM, Netherlands). Cd alone can disturb
Zn in kidneys and calculated BMDL10 was 1.713 mg Cd/kg/day, while in combination with
BDE-209 it was 1.719 mg/kg/day. Cd disturbs Cu in kidney in significantly higher dose of
5.679 mg Cd/kg/day. However, in combination with the BDE-209, derived BMDL10 was 10
times lower, 0.517 mg Cd/kg/day. Dose-response curve slope for the effect on Cu was
significantly lower in the case of mixture effect than in the case of Cd effect. Since Cu plays a
major role in the blood cells creation and health of other systems, our results indicate more
potent activity of the mixture than single Cd.
AB  - Ljudi mogu biti izloženi hemikalijama iz različitih izvora, te je potrebno procenjivati
efekte na bazi izloženosti smešama hemikalija, pre nego pojedinačnim hemikalijama. U
studiji je analiziran uticaj smeše Cd i BDE-209, široko rasprostranjenih zagađujućih
perzistentnih supstanci, na homeostazu bioelemenata, cinka (Zn) i bakra (Cu). U cilju
evaluacije potencijala smeše da naruši homeostazu Zn i Cu u bubrezima pacova, Wistar
pacovi su oralnom gavažom bili tretirani tokom 28 dana dozama Cd od 2,5, 7,5 i 15
mg/kg/dan, kombinacijama ove tri doze sa 1000 mg BDE-209/kg/dan i vodom kao
kontrolom. Tkivo bubrega je izdvojeno, zatim mineralizovano, a sadržaj Zn i Cu je meren
atomskom apsorpcionom spektrofotometrijom. Moguće interakcije su procenjivane
poređenjem nagiba krivih doza-odgovor, a izračunate su i donje granice pouzdanosti
Benchmark doze od 10% (BMDL10) korišćenjem PROAST softvera (RIVM, Holandija). Cd
primenjen sam utiče na homeostazu Zn u bubregu i izračunata BMDL10 iznosi 1,713
mg/kg/dan, dok u kombinacji sa BDE-209 iznosi 1,719 mg/kg/dan. Cd ometa homeostazu Cu
u bubregu pacova i BMDL10 iznosi 5,679 mg/kg/dan, dok je u kombinaciji sa BDE-209 BMDL
10 puta niža i iznosi 0,517 mg/kg/dan. Nagib krive doza odgovor za smešu Cd i BDE-209 bio
je daleko niži od nagiba krive doza-odgovor za Cd. S obzirom na to da Cu igra značajnu ulogu
u nastanku krvnih ćelija, ali i u očuvanju durgih sistema organa, dobijeni rezultati iziskuju
pažnju zbog daleko potentnijeg uticaja smeše na nivo Cu u odnosu na uticaj samog Cd.

AB  - In order to look into the combined effects of Cd and BDE-209 in vitro, this study was aimed at examining cytotoxic and genotoxic effects using the human colon carcinoma cell line (SW 480) as a biological test system as well as to determine if ROS production was one of the possible mechanisms of their mixture action. This cell line was chosen since ingestion of contaminated food/water represents an important route of exposure to both Cd and BDE-209, which is why intestinal cells are a common target for the contaminants present in food and water. Cells were treated with single Cd in concentrations of 2.5, 7.5 or 15 mu g Cd/mL (corresponding to 22, 67 or 134 mu M), single BDE-209 in concentrations of 2.5, 5 or 10 mu g BDE209/mL (corresponding to 2.5, 5 or 10 mu M), and their mixtures (design 3 x 3). Mixture of Cd and BDE-209 has shown clear potential to reduce the viability of SW 480 cells, as evidenced by cytotoxicity associated with ROS generation. Factorial regression models used to identify type of interaction revealed synergism related to mixture citotoxicity and additive interaction for the effect on ROS production. The results from this introductory study could contribute to the issue of possible adverse effects associated with co-exposure and body burden with two persistent environmental pollutants, Cd and BDE-209.

AB  - In relation to commonly used no-observed-adverse-effect-level (NOAEL) point of departure, because of its statistical power and reliability, Benchmark dose (BMD) approach has been proposed as an alternative in dose-response assessment. The aim of this study was to derive BMD (10%) by means of PROAST software to quantify influence of cadmium (Cd) and/or decabrominated diphenyl ether (BDE209) on thyroid hormones homestasis. Study was conducted on male Wistar rats treated orally by gavage for 28 days by either single substances or their combination. Three groups of animals were dosed with Cd at levels of 2.5, 7.5 and 15 mg/kg b.w./day, three groups of animals were dosed with BDE209 at levels of 1000, 2000 or 4000 mg/kg b.w./day, while 9 groups received different dose mixtures of previously given dose levels of Cd and BDE209 (design 3 x 3). Results of the study have indicated that Cd+BDE209 mixtures are likely more potent to disrupt thyroid function than would be expected from the chemicals individually. Derived BMD - lower confidence limits (BMDL), if ratio BMD/BMDL is  lt  10, were 9.4 mg Cd/kg b.w./day and 2155 mg BDE209/kg b.w./day for the effect on T3; and 6.22 mg Cd/kg b.w./day in the mixture with BDE209 2000 mg/kg b.w./day for the effect on FT3.
AB  - Primena statistički dobijene Benchmark doze (BMD) u proceni rizika predstavlja alternativu najčešće korišćenoj 'dozi bez štetnog efekta' (NOAEL) zbog veće pouzdanosti u analizi odnosa doze i toksičnog efekta. Cilj ovog rada bio je izračunavanje BMD (10%) primenom PROAST softvera radi kvantitativne procene uticaja Cd i/ili BDE209 na homeostazu hormona štitaste žlezde. Ispitivanje je sprovedeno na mužjacima Wistar pacova koji su putem oralne sonde, tokom 28 dana, primali pojedinačne supstance ili njihove kombinacije. Kadmijum je primenjivan u dozama od 2,5, 7,5 i 15 mg Cd/kg t.m./dan, a BDE209 u dozama od 1000, 2000 i 4000 mg BDE209/kg t.m./dan, dok je ostalih devet grupa životinja primalo kombinacije hemikalija (dizajn 3 x 3). AB  - The aim of this work was to assess the risk due to mercury (Hg), dichlorodiphenyltrichloroethane (DDT) and non-dioxin-like polychlorinated biphenyls (ndl PCBs) intake via fish consumption in Serbia. We have developed 24 scenarios using four concentration levels (mean, maximum, 50th and 95th percentile) of contaminants, determined in 521 samples of fish products available on Serbian market; two consumption levels (Food and Agriculture Organization/World Health Organization data and recommendation of American Heart Association); and three body weights (5th, 50th and 95th percentile). All the values concerning the intake of DDT are below the corresponding health based guidance value. Calculated weekly intake of Hg using maximal concentration, intake of 340 g/week and 5th percentile of body weight exceeded the provisional tolerable weekly intake (PTWI). When maximal and 95th percentile concentration of ndl PCBs was used, weekly intakes exceeded a "guidance value" with one exception Le., when 95th percentile of concentration along with 95th percentile of body weight were used. Concerning Hg and ndl PCBs, when extreme concentrations were used, HIs exceeded the value of 1, indicating that fish and fishery products may pose a threat to consumer's health.

AB  - Polybrominated diphenyl ethers (PBDEs) represent group of persistant organic chemicals (POPs), usually used as flame retardant additives in a variety of matherials and commercial products. Since 2009, PBDEs have been placed on the list of POPs chemicals, covered by Stockholm convention which is aimed to limit or ban production, use, emission, import and export of persistent organic pollutants, due to human health and environmental protection. Predominant routes of human exposure by PBDEs are ingestion by food, inhalation or dermal. Based on experimental studies, targets for PBDEs toxicity are nervous system, liver, thyroid gland and development. According to International Agency for Research on Cancer (IARC) PBDEs are not classified as a carcinogens, whereas according to Environmental Protection Agency (EPA) only deca BDE is classified as possible human carcinogen. So far, PBDEs have not been incoporated in national legislative framework. There are no data on environmental contamination, occupational exposure and exposure of general population. Therefore, placing the data on PBDEs in the focus of publics could contribute to the process of risk characterisation, and give directions for risk management in the Republic of Serbia.
AB  - Polibromovani difeniletri (PBDEs) su hemikalije koje se dodaju različitim proizvodima kojima smo okruženi u svakodnevnom životu da bi usporili i/ili sprečili izbijanja požara. Polibromovani difeniletri su 2009. godine pridruženi listi perzistentnih organskih polutanata (POPs) usvojenoj Stokholmskom konvencijom čiji je cilj da ograniči ili zabrani proizvodnju, upotrebu, emisiju ili uvoz i izvoz toksičnih supstanci označenih kao POPs radi zaštite zdravlja ljudi i životne sredine. Do ekspozicije ljudi PBDEs može doći ingestijom, inhalacijom, dermalno. Eksperimentalne studije ukazuju da PBDEs najznačajnije toksične efekte ispoljavaju na jetru, štitnu žlezdu, nervni sistem, rast i razvoj. Međunarodna agencija za istraživanje karcinoma (IARC) ne klasifikuje PBDEs kao karcinogene za ljude, a Američka agencija za zaštitu životne sredine (EPA) klasifikuje samo dekaBDE kao moguć karcinogen za ljude. U našoj zemlji PBDEs nisu inkorporirani u nacionalnu regulativu, nema podataka o nivou zagađenja životne sredine, o sadržaju ovih jedinjenja u radnom okruženju i ekpoziciji opšte populacije, stoga bi saznanja u ovoj oblasti doprinela procesu evaluacije i karakterizacije rizika, ali i dala smernice za upravljanje rizikom PBDEs hemikalijama u Republici Srbiji.
