Morphological diversification of Teucrium montanum sensu lato on the Balkan Peninsula

AB  - In this work, the overall morpho-anatomical variability of Teucrium montanum sensu lato on the Balkan Peninsula was described. A total of 60 populations were collected all over the Balkans. For each analysed individual, the morphological characters of the stem, leaves, and flowers, as well as the anatomical characters of leaves were measured. The habitats of each population were characterized by altitude zone, type of geological substrate, vegetation, and climate type. Basic descriptive statistics were calculated for each character state, and multivariate analyses were used to compare all assessed characters and groups. The results showed that the analysed populations exhibit high morphological variability and broad ecological preferences. Five of the seven morphologically defined groups fit the already described taxa (T. montanum subsp. montanum, T. montanum subsp. helianthemoides, T. pannonicum, T. skorpili, T. montanum var. parnassicum), whereas two groups are morphologically specific but did not fit the existing taxonomic concepts. The patterns of observed morphological variability only partially match the patterns of ecological differentiation of the habitats of the analysed populations. The occurrence of different morphological groups inhabiting similar habitats in the same biogeographic region suggests that the morpho-anatomical differentiation of T. montanum s.l. may be due to genetic or evolutionary factors.

HPTLC-based metabolomics approach for the detection of chokeberry (Aronia melanocarpa (Michx.) Elliott) adulteration

AB  - Introduction: Chokeberry is a plant belonging to the family Rosaceae which has a long history of edible and medicinal use. The aims of this study are: i) to investigate of HPTLC profile of the berry samples using HPTLC technique based metabolomics approach, ii) to determine the botanical origin of investigated chokeberry adulterations iii) to identify the most important botanical markers responsible for classification. Methods: A simple, rapid, and high-throughput planar chromatography (HPTLC) method was applied in combination with multivariate techniques for differentiating chokeberry samples and identification of the main metabolites of chokeberry and four common adulterants. Images of HPTLC chromatograms obtained using two different detections i.e. visible light and derivatization with natural products reagent were used as a dataset for multivariate analysis. Results: Principal component analysis and Orthogonal Partial Least Squares Discriminant Analysis confirmed the discrimination of five botanically different samples and recognized their main markers such as responsible for differences between chokeberry and four common adulterants. Conclusion: The obtained results showed that the HPTLC-based metabolomics approach can be a very reliable technique for the detection of chokeberry adulteration.

Morphological diversification of Teucrium montanum L. sensu lato on the Balkan Peninsula

AB  - Teucrium montanum L. is a semi-woody evergreen small shrub distributed in southern and central Europe, Asia Minor, and Algeria in North Africa. The species has a very wide ecological valence occurring in various Mediterranean, sub-Mediterranean, continental as well as oro-Mediterranean and Alpine habitats. Due to its wide ecological valence and broad distribution, T. montanum displays high morphological variability, which is reflected in a large number of described taxa associated with it. In Europe, about thirty taxa have been described at specific and infraspecific levels, of which only T. montanum subsp. montanum and T. montanum subsp. helianthemoides (Adamović) Baden are accepted, while all the others are considered synonyms of T. montanum. The observed morphological diversity is reflected in the presence of six taxa described from the Balkan Peninsula: T. montanum subsp. helianthemoides, T. montanum var. hirsutum Boiss., T. montanum var. modestum Heldr., T. montanum var. parnassicum Čelak., T. pannonicum A. Kern., and T. skorpili Velen. During field research throughout the Balkans, seven morphologically more or less well-defined groups with relatively solid geographical differentiation were observed. Five of the seven groups fit the already described infraspecific taxa, while two groups are morphologically specific but do not correspond to the existing taxa. The aim of this study was to describe the overall morpho-anatomical variability of T. montanum sensu lato in the Balkans and to determine whether the seven identified groups can be distinguished based on morpho-anatomical characters. The most typical populations of these groups could be separated quite well based on the quantitative morphological traits. Multivariate analysis of all samples showed a solid separation of three groups and a significant number of individuals with intermediate positions. Other groups did not show statistical diversification but possess a specific combination of morphological characters by which they can be identified.

AB  - Satureja kitaibelii Wierzb. ex Heuff. (Rtanj tea) has distinct morphology and distribution which separate it from highly polymorphic S. montana L. In this work, we investigated phenolic composition of S. kitaibelii and compared it to that of S. montana. Flowering aerial parts of four S. kitaibelii samples were collected in Serbia and Bulgaria and one commercial sample of Rtanj tea was obtained from Serbia. Three samples of S. montana were collected in North Macedonia and Serbia and the fourth one was cultivated sample. Herbs were extracted with 50% ethanol. Dry extract solutions were analyzed by liquid chromatography with UV and MS detection. Rosmarinic acid (RA) and clinopodic acid O (CAO) were quantified using RA as the external standard. All extracts were characterized by presence of phenolic acids, RA (16.5-47.9 mg/g) and CAO (1.6-29.3 mg/g, calculated as RA), and flavonoids. The extracts of S. kitaibelii distinguished by high contents of both RA and CAO, which were present in similar quantities within each extract. On the other hand, extracts of S. montana contained at least 3 times less CAO than RA. The obtained results further confirm distinct properties of S. kitaibelii and give additional arguments to its status as a separate species.

AB  - Teucrium montanum L. is a semi-woody, evergreen small shrub, widely distributed in Europe occurring from the seacoast to the subalpine and alpine belts. The study aimed to investigate and compare the chemical composition of essential oil of aerial parts of T. montanum from 24 populations collected from West, Central, and South West Balkans. The essential oils were obtained by hydrodistillation and qualitative and quantitative analysis was performed by GC-FID and GC-MS. The dominant compounds were oxidized sesquiterpenes (24.83–85.75%), then sesquiterpene hydrocarbons (3.00–62.81%), and oxidized monoterpenes (0–27.78%). Cluster analysis revealed 10 clearly separated groups. First group (Žumberak) is completely separated and characterized by high germacrene-D-4-ol (63.17%), the second (Gjergjevice) by high shyobunol (51.23%), the third (Vratna) by high (dehydro)-sesquicineole-acetate (45.29%), the fourth (Ostrovice) by high epi-α-cadinol (36.80%), the fifth (Nera canyon) by high limonene (36.65%), sixth group (Orovica, Sićevo, Galičica) by high cis-sesquisabinene hydrate (18.73–29.05%), seventh group (Štrpce, Dobrilovina) by high γ-cadinene (13.43–23.06%), eighth group (Tara, Durmitor, Biokovo, Fužine) by moderate germacrene-D-4-ol (19.76–23.44%), and the ninth large group (Potoci, Lovćen, Orjen, Premantura, Vodice, Pletvar, Senj, Oštarije, Trebinje, Korita) by germacren D (6.15–33.87%). Such a high variability needs more research to define relationships among populations.

AB  - Ethnopharmacological relevance: Medicinal properties of hyssop have been used in traditional medicine since ancient times, inter alia, in diseases/conditions with an inherent inflammatory process. Aim of the study: Accordingly, the aim of this study was to investigate the anti-inflammatory properties of hyssop herb preparations (essential oil and methanol extracts) in vivo, in vitro and in silico. Materials and methods: For in vitro testing of essential oils and extracts of hyssop herb, the cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzyme assays were used. In vivo anti-inflammatory potential of the extracts (at doses of 50, 100 and 200 mg/kg) was assessed using the carrageenan-induced rat paw edema test. Molecular docking and dynamics were used for in silico testing of the inhibitory activity of chlorogenic (CA) and rosmarinic (RA) acids, as the dominant compounds in the tested methanol extracts against COX-1 and COX-2 enzymes. Results: Significant inhibitory activity was shown in the COX-2 test regarding extracts (essential oils did not exhibit any significant activity). Namely, all analyzed extracts, at a concentration of 20 μg/mL, showed a percentage of inhibition of COX-2 enzyme (54.04–63.04%), which did not indicate a statistically significant difference from the positive control of celecoxib (61.60%) at a concentration of 8.8 μM. In vivo testing showed that all methanol extracts of hyssop herb, at the highest test dose of 200 mg/kg in the third and fourth hours, after carrageenan administration, exhibited a statistically significant (p < 0.05) inhibitory effect on the increase in rat paw edema in relation to control. This activity is comparable or higher in relation to the reference substance, indomethacin, at a concentration of 8 mg/kg. The preliminary in silico results suggest that investigated compounds (RA and CA) showed better inhibitory activity against COX-1 and COX-2 than standard non-steroidal anti-inflammatory drug (NSAID), ibuprofen, as evident from the free binding energy (ΔGbind in kJ mol−1). The binding energies of the docked compounds to COX-1 and -2 were found to be in the range between −47.4 and −49.2 kJ mol−1. Ibuprofen, as the one NSAID, for the same receptors targets, showed remarkably higher binding energy (ΔGbind = −31.3 kJ mol−1 to COX-1, and ΔGbind = −30.9 kJ mol−1 to COX-2). Conclusion: The results obtained not only support the traditional use of hyssop herb in inflammatory conditions in folk medicine, but also open the door to and the need for further in vivo testing of extracts in order to examine the molecular mechanism of anti-inflammatory activity in living systems and possibly develop a new anti-inflammatory drug or supplement.

Composition and antimicrobial activity of essential oils of Salvia fruticosa and Salvia ringens (Lamiaceae)

AB  - Background/Aim. Plant essential oils (EOs) can have a significant antibacterial effect especially through additive or synergistic action as antibiotic adjuvants. We investi-gated the composition and activity of EOs of two species of genus Salvia (S) from Greece with the aim to deter-mine their antimicrobial activity as well as the activity in combination with selected antibiotics. Methods. The aerial parts of wild-growing S. fruticosa and S. ringens were subjected to a steam distillation and the obtained EOs were analyzed by gas chromatography and gas chroma-tography/mass spectrometry. The broth-microdilution method was used in order to determine the minimum in-hibitory concentrations (MICs) of EOs on seven strains of bacteria and one yeast. Antimicrobial activity of the combination of EO and antibiotics was investigated by checkerboard method and estimated by calculating frac-tional inhibitory concentration (FIC) of each component and fractional inhibitory concentration index (FICI). Re-sults. Dominant component of S. fruticosa EO was trans-thujone (54.2%) and for S. ringens EO it was α-pinene (28.1%). The MICs of EOs of both species were in the range from 200 μg/mL to > 500 μg/mL. The strongest antimicrobial effect was achieved against Bacillus subtilis and Candida albicans. According to FICI values, EO of S. fruticosa had additive effect with ciprofloxacin against most of bacterial strains but not with amikacin. Conclu-sion. The essential oils of S. ringens and S. fruticosa showed modest antimicrobial activity. However, the es-sential oil of S. fruticosa showed a promising additive ef-fect in combination with ciprofloxacin.
AB  - Uvod/Cilj. Etarska ulja različitih biljaka mogu imati značajna antibakterijska svojstva, posebno kao adjuvanti antibiotika sa kojima ostvaruju aditivno ili sinergistično dejstvo. Ispitivali smo sastav i aktivnost etarskih ulja dve vrste roda Salvia (S) iz Grčke sa ciljem da odredimo njihovu antimkrobnu aktivnost, kao i dejstvo u kombi-naciji sa odabranim antibioticima. Metode. Nadzemni delovi samoniklih S. fruticosa i S. ringens su destilovani vodenom parom i dobijena etarska ulja su analizirana gasnom hromatografijom i gasnom hromatografijom sa masenom spektrometrijom. Radi određivanja minimalnih inhibitornih koncentracija (MICs) etarskog ulja na sedam sojeva bakterija i na jednoj patogenoj gljivici korišćena je mikrodiluciona metoda. Antimikrobna aktivnost kombi-nacije etarskog ulja i antibiotika ispitana je checkerboard metodom i procenjena je na osnovu frakcione inhibi-torne koncentracije (FIC) svake komponente i indeksa frakcione inhibitorne koncentracije (FICI). Resultati. Dominantna komponenta etarskog ulja S. fruticosa je bio trans-tujon (54,2%), a etarskog ulja S. ringens α-pinen (28,1%). MICs etarskog ulja obe vrste su bile u opsegu od 200 μg/mL do > 500 μg/mL. Najsnažnija anti-mikrobna aktivnost ostvarena je protiv Bacillus subtilis i Candida albicans. Na osnovu FICI vrednosti, etarsko ulje S. fruticosa je sa ciprofloksacinom, ali ne i sa amikacinom imalo aditivni efekat protiv većine bakterijskih sojeva. Zaključak. Etarska ulja S. ringens i S. fruticosa su pokazala skromnu antimikrobnu aktivnost, ali je etarsko ulje S. fru-ticosa u kombinaciji sa ciprofloksacinom ispoljilo značajan aditivni efekat.

The aerial parts of wild-growing S. fruticosa and S. ringens were subjected to a steam distillation and the obtained EOs were analyzed by gas chromatography and gas chroma-tography/mass spectrometry. The broth-microdilution method was used in order to determine the minimum in-hibitory concentrations (MICs) of EOs on seven strains of bacteria and one yeast. Antimicrobial activity of the combination of EO and antibiotics was investigated by checkerboard method and estimated by calculating frac-tional inhibitory concentration (FIC) of each component and fractional inhibitory concentration index (FICI). Re-sults. Dominant component of S. fruticosa EO was trans-thujone (54.2%) and for S. ringens EO it was α-pinene (28.1%). The MICs of EOs of both species were in the range from 200 μg/mL to > 500 μg/mL. The strongest antimicrobial effect was achieved against Bacillus subtilis and Candida albicans. According to FICI values, EO of S. fruticosa had additive effect with ciprofloxacin against most of bacterial strains but not with amikacin. Conclu-sion. The essential oils of S. ringens and S. fruticosa showed modest antimicrobial activity. However, the es-sential oil of S. fruticosa showed a promising additive ef-fect in combination with ciprofloxacin., Uvod/Cilj. Etarska ulja različitih biljaka mogu imati značajna antibakterijska svojstva, posebno kao adjuvanti antibiotika sa kojima ostvaruju aditivno ili sinergistično dejstvo. Ispitivali smo sastav i aktivnost etarskih ulja dve vrste roda Salvia (S) iz Grčke sa ciljem da odredimo njihovu antimkrobnu aktivnost, kao i dejstvo u kombi-naciji sa odabranim antibioticima. Metode. Nadzemni delovi samoniklih S. fruticosa i S. ringens su destilovani vodenom parom i dobijena etarska ulja su analizirana gasnom hromatografijom i gasnom hromatografijom sa masenom spektrometrijom. Radi određivanja minimalnih inhibitornih koncentracija (MICs) etarskog ulja na sedam sojeva bakterija i na jednoj patogenoj gljivici korišćena je mikrodiluciona metoda. Antimikrobna aktivnost kombi-nacije etarskog ulja i antibiotika ispitana je checkerboard metodom i procenjena je na osnovu frakcione inhibi-torne koncentracije (FIC) svake komponente i indeksa frakcione inhibitorne koncentracije (FICI). Resultati. Dominantna komponenta etarskog ulja S. fruticosa je bio trans-tujon (54,2%), a etarskog ulja S. ringens α-pinen (28,1%). MICs etarskog ulja obe vrste su bile u opsegu od 200 μg/mL do > 500 μg/mL. Najsnažnija anti-mikrobna aktivnost ostvarena je protiv Bacillus subtilis i Candida albicans. Na osnovu FICI vrednosti, etarsko ulje S. fruticosa je sa ciprofloksacinom, ali ne i sa amikacinom imalo aditivni efekat protiv većine bakterijskih sojeva. Zaključak. Etarska ulja S. ringens i S. fruticosa su pokazala skromnu antimikrobnu aktivnost, ali je etarsko ulje S. fru-ticosa u kombinaciji sa ciprofloksacinom ispoljilo značajan aditivni efekat.",

Chemical compositionwas analyzed by gas chromatography - mass spectrometry (GC-MS) and liquid chromatography with diodearray detection and mass spectrometry (LC-DAD-MS), respectively. Antioxidant activity was examinedby 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing/antioxidant power (FRAP) tests; genotoxicand antigenotoxic activity were examined by the comet assay, while cytotoxicity was evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide dye (MTT) test against tumor cell lines (SW480,MDA-MB 231, HeLa) and non-transformed human lung fibroblast cell lines (MRC-5). The essential oilswere rich in monoterpene hydrocarbons (e.g., limonene; 7.99–23.81%), oxygenated monoterpenes (1,8-cineole; 38.19–67.1%) and phenylpropanoids (methyl eugenol; 0.00–28.33%). In methanol extracts, the mostabundant phenolics were chlorogenic and rosmarinic acid (23.35–33.46 and 3.53–17.98 mg/g, respectively).Methanol extracts expressed moderate to weak antioxidant activity (DPPH IC50= 56.04–199.89μg/mL,FRAP = 0.667–0.959 mmol Fe2+/g). Hyssop preparations significantly reduced DNA damage in humanwhole blood cells, induced by pretreatment with hydrogen peroxide. Methanol extracts exhibited selectiveand potent dose- and time-dependent activity against the HeLa cell line.  Results of the current studydemonstrated notableH.officinalismedicinal potential, which calls for further investigation.

Isolation and seasonal variation of fruticin in fruits of false indigo-bush (Amorpha fruticosa L. Fabaceae) from Serbia

AB  - Fruticin (amorphin) is a constituent of the fruit of the false indigo-bush (Amorpha fruticosa L.), which
belongs to the class of rotenoid glycosides, and shows several interesting pharmacological activities.
The aim of this study was to isolate and chemically characterize this natural product, as well as to
determine the optimal period of the year for A. fruticosa fruits collection. Fruticin was obtained by
re-crystallization of the precipitate that formed after partial evaporation of the extract, prepared by 3-
fold extraction of powdered plant material by chloroform - ethanol (1:1, v/v). The structure of the final
product was determined by various techniques of instrumental analysis (NMR, UV and MS), and con-
firmed by comparing the obtained spectra with corresponding data in available literature. The content
of fruticin in A. fruticosa fruit was determined by LC-DAD-MS, using the external standard method
based on the constructed calibration curve. Limits of detection (LOD) and quantification (LOQ) were
also determined. A substantial increase in fruticin content was observed during the ripening period
(>50 %). It has also been established that the optimal time for fruit collection is mid-December. Ob-
tained results indicate that the content of fruticin in the A. fruticosa fruit is highly dependable on the
time of collection. Since the biosynthesis of secondary plant metabolites is influenced not only by the
time of collection, but by numerous other factors as well, additional studies are needed to define, with
greater certainty, the conditions that are necessary for design of prospective efficient and sustainable
production process.

AB  - Phenolic groups of steroidal or nonsteroidal estrogens can redox cycle, leading to oxidative stress, where creation of reactive oxygen species are recognized as the main mechanism of their DNA damage properties. Dry olive (Olea europaea L.) leaf extract is known to contain bioactive and antioxidative components and to have an ability to modulate the effects of various oxidants in cells. The main goal of this study was to investigate antigenotoxic potential of a standardized dry olive leaf extract on DNA damage induced by 17β-estradiol and diethylstilbestrol in human whole blood cells in vitro, using comet assay. Our results indicated that both hormones showed a genotoxic effect at a concentration of 100 μM (P  lt  0.05, n = 6). Dry olive leaf extract was efficient in reducing number of cells with estrogen-induced DNA damage at tested concentrations (0.125, 0.5 and 1 mg/mL) (P  lt  0.05, n = 6) and under two experimental protocols, pre-treatment and post-treatment, exhibiting antigenotoxic properties. Analysis of antioxidant properties of the extract revealed moderate ABTS radical scavenging properties and reducing power. Overall, our results suggested that the protective potential of dry olive leaf extract could arise from the synergistic effect of its scavenging activity and enhancement of the cells' antioxidant capacity.

Ispitivanje makroskopskih, mikroskopskih osobina i etarskog ulja lista matičnjaka, Melissa officinalis L.

AB  - Tradicionalno list matičnjaka, Melissae folium, koristi se za ublažavanje blagih
simptoma mentalnog stresa i olakšavanje uspavljivanja, kao i u simptomatskoj terapiji
blagih gastrointestinalnih poremećaja. Etarsko ulje matičnjaka je veoma cenjeno u
prehrambenoj i kozmetičkoj industriji. Cilj rada je bio ispitivanje makroskopskih i
mikroskopskih osobina uzoraka lista samoniklog i gajenog matičnjaka sa različitih
lokaliteta i komercijalno dostupnih čajeva, kao i ispitivanja sastava njihovih etarskih
ulja.
Za analizu etarskih ulja (16 uzoraka), dobijenih destilacijom vodenom parom,
primenjena je gasna hromatografija (GC‐FID) i gasna hromatografija sa masenom
spektrometrijom (GC‐MS). Klaster analizom ispitivane su razlike između etarskih ulja.
Makroskopske i mikroskopske odlike svih uzoraka su odgovarale monografiji
Melissae folium u Ph. Eur. 7.0, ali je u 2 od 5 komercijalnih uzoraka bio veći sadržaj
stranih materija. U svim ispitivanim etarskim uljima glavne komponenate su
predstavljali: geranial (15,58‐53,33%), neral (10,73‐36,64 %), kariofilen oksid (0,76‐
35,57 %), (E)‐kariofilen (tragovi‐16,72 %) i citronelal (0,00‐12,64 %). Sadržaj geraniala
i nerala u ispitivanim uzorcima je bio u opsegu vrednosti dobijenih u prethodnim
ispitivanjima.
Klaster analizom uzorci etarskog ulja su podeljeni na dva klastera. U prvom

for relief of mild symptoms of mental stress, to aid sleep and for symptomatic
treatment of mild gastrointestinal complaints. The essential oil of lemon balm is very
appreciated in food and cosmetic industry. The aim was to examine the macroscopic
and microscopic characteristics of lemon balm leaves of cultivated and wild plants from

Makroskopske i mikroskopske odlike svih uzoraka su odgovarale monografiji
Melissae folium u Ph. Eur. 7.0, ali je u 2 od 5 komercijalnih uzoraka bio veći sadržaj
stranih materija. U svim ispitivanim etarskim uljima glavne komponenate su
predstavljali: geranial (15,58‐53,33%), neral (10,73‐36,64 %), kariofilen oksid (0,76‐
35,57 %), (E)‐kariofilen (tragovi‐16,72 %) i citronelal (0,00‐12,64 %). Sadržaj geraniala
i nerala u ispitivanim uzorcima je bio u opsegu vrednosti dobijenih u prethodnim
ispitivanjima.
Klaster analizom uzorci etarskog ulja su podeljeni na dva klastera. U prvom
klasteru su bila etarska ulja sa visokim sadržajem geraniala i nerala (31,87‐53,33% i
19,81‐36,64%): etarska ulja lista i herbe samoniklog i gajenog matičnjaka u različitim

Quality of selected aromatic herbal drugs available on Belgrade's marketplaces

AB  - Herbal drugs are often administered in the form of monocomponent teas or herbal tea mixtures (tea blends). In order to assess the quality of herbal drugs that are components of the monocomponent teas commercially available on marketplaces in Belgrade, we analyzed 4 samples of Melissa leaf (Melissae folium), 6 samples of Wild thyme, (Serpylli herba), 6 samples of Yarrow (Millefolii herba), 4 samples of Wormwood (Absinthii herba) and 6 samples of Juniper (Juniperi pseudo-fructus), according to the procedures given in European Pharmacopoea. Macroscopic and microscopic features of investigated herbal drugs, as well as their TLC profiles, were in agreement with corresponding data described in Ph. Eur. One sample of Melissa leaf, one sample of Wild thyme and two samples of Juniper, did not fullfil the requirements of general tests. Due to inappropriate size of cut, test on foreign matter could not be performed for some samples. Concerning specific quality, all samples of Melissa leaf, Wild thyme, Wormwood and Juniper were in accordance with Ph. Eur. Regarding the content of essential oil, Yarrow samples met the requirements of Ph. Eur., but the proazulenes content in five samples was lower than required. In conlcusion, the results of these analyses point to high medicinal potential of the most herbal drugs studied. On the other hand, unsatisfying general quality and inappropriate size of cut of several samples could be related to inadequate processing and handling of plant material.
AB  - Biljne droge se vrlo često koriste u obliku monokomponentnih i višekomponentnih čajeva. U cilju ispitivanja kvaliteta biljnih droga koje se mogu naći na pijacama Beograda u obliku monokomponentnih čajeva, analizirana su po četiri uzorka lista matičnjaka (Melissae folium) i herbe belog pelena (Absinthii herba), i po šest uzoraka herbe majčine dušice (Serpylli herba), herbe hajdučke trave (Millefolii herba) i šišarica kleke (Juniperi pseudo-fructus), prema propisu Ph. Eur. 7.0. Makroskopske i mikroskopske karakteristike ispitivanih droga, kao i rezultati TLC analize bili su u skladu sa odgovarajućim podacima datim u monografijama Farmakopeje. U pogledu opšteg kvaliteta, zahteve nisu ispunili jedan uzorak lista matičnjaka, jedan uzorak herbe majčine dušice i dva uzorka šišarica kleke. Zbog neodgovarajućeg stepena usitnjenosti pojedine uzorke nije bilo moguće ispitati u pogledu stranih primesa. Zahteve za specifični kvalitet ispunili su svi uzorci lista matičnjaka, herbe majčine dušice, herbe belog pelena i šišarica kleke. Uzorci herbe hajdučke trave sadržali su odgovarajuću količinu etarskog ulja, ali je sadržaj proazulena u pet uzoraka bio niži od propisanog. Rezultati ispitivanja specifičnog kvaliteta ukazuju na visok lekoviti potencijal većine ispitivanih biljnih droga. Sa druge strane, neispravnost u pogledu opšteg kvaliteta i neodgovarajući stepen usitnjenosti pojedinih uzoraka može se dovesti u vezu sa neadekvatnim rukovanjem biljnim materijalom.

One of the most dangerous invasive plant species in Europe, Heracleum sosnowskyi Manden. H. sosnowskyi, is recorded as a new species in the flora of Serbia. It was found in June 2016 in the area of Belgrade on the left bank of the Danube, near Kovilovo. is Data concerning its morphology, habitat characteristics, flowering period and population size in Serbia are presented in this paper.

A Mediterranean medicinal plant in the continental Balkans: A plastid DNA-based phylogeographic survey of Salvia officinalis (Lamiaceae) and its conservation implications

AB  - Salvia officinalis (Lamiaceae), common or Dalmatian sage, is a Mediterranean aromatic and medicinal plant used in medicine since ancient times. Knowledge on current genetic patterns and genealogical history of its natural populations is required for both breeding efforts and species conservation. We used sequences of two chloroplast intergenic spacers, 3'rps16-5'trnK and rp132-trnL, from 83 individuals from eight natural populations to distinguish between anthropogenic vs natural origin of four disjunct inland populations found outside of the main Adriatic range of the species. We found seven haplotypes, high total gene diversity (H-r = 0.695) and genetic differentiation (G(ST) = 0.682), as well as a phylogeographic structure with two lineages, a sub-structured inland-Adriatic lineage (IAL, comprising inland and Adriatic sub-lineages) and a purely Adriatic lineage (PAL). All four inland and disjunct populations, which comprised the inland sub-lineage of IAL, were almost fixed for a distinct haplotype genealogically closely related to the ancestral haplotype and displayed other features of relict populations. Along with previous biogeographic data and other lines of evidence, assumptions on their anthropogenic origin were rejected. At present, a less diverse IAL (Hd = 0.426, pi = 0.00106) and a more diverse PAL (Hd = 0.403, pi = 0.00257), whose divergence was dated to the Pliocene (3.267 Mya), do not exhibit signs of recent demographic expansions and overlap on the SE Adriatic coast, a region delineated as the main glacial refugium of S. officinalis. Conservation measures accounting for the historical distinctiveness of populations and focusing on currently the most threatened populations are recommended.

Along with previous biogeographic data and other lines of evidence, assumptions on their anthropogenic origin were rejected. At present, a less diverse IAL (Hd = 0.426, pi = 0.00106) and a more diverse PAL (Hd = 0.403, pi = 0.00257), whose divergence was dated to the Pliocene (3.267 Mya), do not exhibit signs of recent demographic expansions and overlap on the SE Adriatic coast, a region delineated as the main glacial refugium of S. officinalis. Conservation measures accounting for the historical distinctiveness of populations and focusing on currently the most threatened populations are recommended.",

However, the indigeneity ofits distant continental disjunctions in Central Balkans is questionable, since thisthermophilous species has been used in medicine for thousands of years and it may havebeen established post-glacially via anthropogenic mediation.To reveal the origin and the age of its disjunctions, 8 and 11 populations weresampled for molecular and morphometrical investigation, respectively. Sequences oftwo chloroplast intergenic spacers, 3’rps16-5’trnK and rpl32-trnL, were used forphylogeographic study. Seven haplotypes, non-randomly distributed in space werefound as well as high total gene diversity (HT = 0.695) and genetic differentiation (GST =0.682) showing a phylogeographic structure.According to the strict clock Bayesian analysis, the divergence of an ancient S.officinalis gene pool to two lineages was dated to the Pliocene (3.267 Ma), whichcoincides with the period when the vast water bodies covered the central part of thePeninsula and presumably divided the paleo-distribution of S. officinalis. AB  - Variations in the yield and composition of essential oil of Dalmatian sage (Salvia officinalis L., Lamiaceae) in various stages of development were analyzed in two individuals (= genotypes) of different geographic origin. Both plants have been successfully grown ten years under identical environmental conditions in a private garden in Belgrade. The amount and composition of the oils were analyzed during one growing season starting from the young to old overwintered leaves. The results of the cluster analysis showed that leaf age and origin of the plants has a significant impact on the composition of the essential oils. All oil samples formed two main clades. The first clade are oils collected from young leaves from April to June and belong to α-humulene type. The second clade includes oil obtained mainly of old leaves, which were collected from August to June, and belonging to the camphor and thujone type. In the second clade samples originating from Učka are completely separated from samples originating from Belgrade. Also, based on these analyzes showed that the same individual (the same genotype) during one growing season significantly change the chemical composition of the essential oils. Thus, it is possible to distinguish three different 'phenological types of essential oils': young leaves ('yl-oils'), early old leaves ('early-ol-oils') and a late old leaves type ('late-ol-oils'). Analysis of the seasonal changes in the composition of the essential oils of Dalmatian sage is shown that different components have different dynamics and different directions of change in the concentration during the season.
AB  - Variranje prinosa i sastava etarskog ulja dalmatinske žalfije (Salvia officinalis L., Lamiaceae) u različitim fazama razvoja analizirano je na dve individue (= genotipa) različitog geografskog porekla. Obe biljke su uspešno gajene deset godina pod identičnom ekološkim uslovima u privatnoj bašti u Beogradu. Količina i sastav ulja su analizirani tokom jedne vegetacione sezone počevši od mladih pa sve do starih prezimelih listova. Rezultati klaster analize su pokazali da starost listova kao i poreklo biljke ima veoma značajan uticaj na sastav etarskog ulja. Svi uzorci ulja su formirali dve osnovne klade. Prvu kladu čine ulja mladih listova sakupljanih od aprila do juna i pripadaju α-humulen tipu. Druga klada obuhvata ulja dobijena uglavnom od starih listova, koji su sakupljani od avgusta do juna, i koji pripadaju kamfor ili tujon tipu. U drugoj kladi uzorci poreklom sa Učke su potpuno odvojeni od uzoraka poreklom iz Beograda. Takođe, na osnovu ovih analiza je utvrđeno da ista individua (isti genotip) u toku jedne vegetacione sezone značajno menja hemijski sastav etarskog ulja. Tako je moguće razlikovati tri različita 'fenološka tipa etarskih ulja': ulja mladih listova, ulja rane faze starih listova i ulja kasne faze starih listova. Analiza sezonskih promena u sastavu etarskih ulja Dalmatinske žalfije je pokazala da različite komponente imaju različitu dinamiku i različite pravce promena u koncentraciji tokom sezone.

Variations of essential oil characteristics of Clinopodium pulegium (Lamiaceae) depending on phenological stage

AB  - The variability of essential oil characteristics in different phenological stages of Clinopodium pulegium from its natural habitat (Svrljiški Timok gorge, Serbia) and from cultivated plants (Niš, Serbia) was determined. The essential oils were obtained from aerial parts of the plants by hydrodistillation and analyzed by GC-MS. These are the first data on the essential oil characteristics of plants of the population from the Svrljiški Timok gorge. Samples originating from both natural and cultivated populations were characterized by high amounts of essential oils in all stages of development (0.8% - 1.4%). Twenty-one compounds were identified, representing 95.3-99.6% of the total oils. Differences in the essential oil composition were more quantitative than qualitative. The quality of the essential oil was stable and did not vary with differences in environmental conditions. Dominance of the main components was modified by phenological stage. Pulegone was dominant in the vegetative (76.1% wild population, 62.7% cultivated population) and the flowering (49.5%, 64.6%) stages, while menthone (48.5%, 65.3%) displaced pulegone (34.7%, 18.4%) at the fruiting stage. Cultivated plants in the vegetative and flowering stages can be considered a significant source of pulegone, and in the fruiting stage a significant source of menthone. Careful selection of the developmental stage of the plant is a potential tool which could be employed to obtain the preferred chemical composition of C. pulegium for commercial use.
AB  - U radu je praćena promenljivost količine i sastava etarkog ulja Clinopodium pulegium kroz različite stadijume razvoja biljke (vegetativni, stadijum cvetanja i plodonošenja). Materijal je sakupljan iz samonikle (klisura Svrljiškog Timoka) i gajene (Niš) populacije. Etarsko ulje je izolovano iz nadzemnih delova biljaka destilacijom vodenom parom i analizirano GC-MS metodom. Ovo su prvi podaci o osobinama etarskog ulja biljaka populacije iz klisure Svrljiškog Timoka. Svi uzorci kako iz samonikle tako i iz gajene populacije su se odlikovali velikim sadržajem ulja (0.8% - 1.4%) u svim stadijumima razvoja. Dvadeset jedna komponenta je identifikovana što predstavlja 95.3-99.6% ukupnog etarskog ulja. Razlike u sastavu ulja su bile više kvantitativne nego kvalitativne. Kvalitet etarskog ulja je bio stabilan a variranja uslovljena promenom sredine relativno mala. Dominacija glavnih komponenata je bila uslovljena promenama fenoloških faza. Pulegon je bio dominantan u vegetativnoj (76.1% u ulju biljaka samonikle populacije, 62.7% u ulju biljaka gajene populacije) i fazi cvetanja (49.5%, 64.6%), dok je menton bio dominantan (48.5%, 65.3%) u fazi plodonošenja. Rezultati su pokazali da se biljke u vegetativnoj i fazi cvetanja mogu smatrati značajnim izvorom pulegona, a u fazi plodonošenja značajnim izvorom mentona. Pažljivim odabirom razvojnog stadijuma biljke moguće je dobiti etarsko ulje željenog sastava koje bi se moglo upotrebiti u komercijalne svrhe.

abstract = "The variability of essential oil characteristics in different phenological stages of Clinopodium pulegium from its natural habitat (Svrljiški Timok gorge, Serbia) and from cultivated plants (Niš, Serbia) was determined. The essential oils were obtained from aerial parts of the plants by hydrodistillation and analyzed by GC-MS. These are the first data on the essential oil characteristics of plants of the population from the Svrljiški Timok gorge. Samples originating from both natural and cultivated populations were characterized by high amounts of essential oils in all stages of development (0.8% - 1.4%). Twenty-one compounds were identified, representing 95.3-99.6% of the total oils. Differences in the essential oil composition were more quantitative than qualitative. The quality of the essential oil was stable and did not vary with differences in environmental conditions. Dominance of the main components was modified by phenological stage. Pulegone was dominant in the vegetative (76.1% wild population, 62.7% cultivated population) and the flowering (49.5%, 64.6%) stages, while menthone (48.5%, 65.3%) displaced pulegone (34.7%, 18.4%) at the fruiting stage. Cultivated plants in the vegetative and flowering stages can be considered a significant source of pulegone, and in the fruiting stage a significant source of menthone. Careful selection of the developmental stage of the plant is a potential tool which could be employed to obtain the preferred chemical composition of C. pulegium for commercial use., U radu je praćena promenljivost količine i sastava etarkog ulja Clinopodium pulegium kroz različite stadijume razvoja biljke (vegetativni, stadijum cvetanja i plodonošenja). Materijal je sakupljan iz samonikle (klisura Svrljiškog Timoka) i gajene (Niš) populacije. Etarsko ulje je izolovano iz nadzemnih delova biljaka destilacijom vodenom parom i analizirano GC-MS metodom. Ovo su prvi podaci o osobinama etarskog ulja biljaka populacije iz klisure Svrljiškog Timoka. Svi uzorci kako iz samonikle tako i iz gajene populacije su se odlikovali velikim sadržajem ulja (0.8% - 1.4%) u svim stadijumima razvoja. Dvadeset jedna komponenta je identifikovana što predstavlja 95.3-99.6% ukupnog etarskog ulja. Razlike u sastavu ulja su bile više kvantitativne nego kvalitativne. Kvalitet etarskog ulja je bio stabilan a variranja uslovljena promenom sredine relativno mala. Dominacija glavnih komponenata je bila uslovljena promenama fenoloških faza. Pulegon je bio dominantan u vegetativnoj (76.1% u ulju biljaka samonikle populacije, 62.7% u ulju biljaka gajene populacije) i fazi cvetanja (49.5%, 64.6%), dok je menton bio dominantan (48.5%, 65.3%) u fazi plodonošenja. Rezultati su pokazali da se biljke u vegetativnoj i fazi cvetanja mogu smatrati značajnim izvorom pulegona, a u fazi plodonošenja značajnim izvorom mentona. Pažljivim odabirom razvojnog stadijuma biljke moguće je dobiti etarsko ulje željenog sastava koje bi se moglo upotrebiti u komercijalne svrhe.",

A Simple and Efficient DNA Isolation Method for Salvia officinalis

AB  - We report an efficient, simple, and cost-effective protocol for the isolation of genomic DNA from an aromatic medicinal plant, common sage (Salvia officinalis L.). Our modification of the standard CTAB protocol includes two polyphenol adsorbents (PVP 10 and activated charcoal), high NaCl concentrations (4 M) for removing polysaccharides, and repeated Sevag treatment to remove proteins and other carbohydrate contaminants. The mean DNA yield obtained with our Protocol 2 was 330.6 mu g DNA g(-1) of dry leaf tissue, and the absorbance ratios 260/280 and 260/230 nm averaged 1.909 and 1.894, respectively, revealing lack of contamination. PCR amplifications of one nuclear (26S rDNA) and one chloroplast (rps16-trnK) locus indicated that our DNA isolation protocol may be used in common sage and other aromatic and medicinal plants containing essential oil for molecular biologic and biotechnological studies and for population genetics, phylogeographic, and conservation surveys in which nuclear or chloroplast genomes would be studied in large numbers of individuals.

Population variability in Thymus glabrescens Willd. from Serbia: Morphology, anatomy and essential oil composition

AB  - In five indigenous populations of Thymus glabrescens Willd. collected in the region of Banat (Serbia), the variability on leaf morphological traits, leaf and stem anatomy, and composition of the essential oil was studied. The major component in the studied populations was either thymol or γ-terpinene. Distinct differentiation of populations with respect to chemical composition of essential oils might be related to spatial distribution of the studied populations. No correlations between morphology, anatomy, and essential oil yield and composition were determined. Both capitate and peltate glandular trichomes were found on calyces, whereas the latter were noticed on the abaxial and adaxial leaf surface.
AB  - U pet autohtonih populacija Thymus glabrescens Willd. sakupljenih u regionu Banata, Srbija, ispitivana je varijabilnost morfoloških karakteristika lista, anatomske građe lista i stabla i sastava etarskih ulja. Glavne komponente etarskih ulja proučavanih populacija bile su timol, γ-terpinen i p-cimen. Uočena je prostorna diferencijacija populacija u odnosu na sastav etarskih ulja. Nije ustanovljena korelacija između morfoloških i anatomskih karakteristika populacija i prinosa i sastava etarskih ulja. Prisustvo i izgled žlezdanih trihoma su utvrđeni posmatranjem nadzemnih delova na svetlosnom i skening elektronskom mikroskopu. Therefore the main objectives of this study were to determine the yield and chemical composition of essential oils from wild-growing T. glabrescens populations, to assess their radical scavenging activity and to correlate the results with published data in order to deduce which components are responsible for the activity. RESULTS: The plant material yielded between 4.0 and 8.0 mL kg(-1) of essential oil. All samples contained considerable but variable concentrations of thymol (22.3-55.1%), depending on the source. Radical-scavenging activities of the oils were estimated by 1,1-diphenyl-2-picrylhydrazyl free radical (DPPH) assay against butylated hydroxytoluene (BHT) and thymol as positive controls. The observed activities (IC50 values ranged from 94 to 230 mu gmL(-1)) were strongly influenced by thymol concentration, as verified by rapid screening for DPPH radical-scavenging activity on thin layer chromatography (TLC) plates and regression analysis. CONCLUSION: These results represent the first report on the free radical-scavenging activity of T. glabrescens essential oil and one of the first comprehensive reports on its composition. Thymus glabrescens could be used in the food industry for seasoning purposes or for preserving processed foods from oxidative degradation.

AB  - Gentiana dinarica Beck. (Gentianaceae) is a rare plant species growing at carbonate soils in subalpine and alpine regions. Acording to the Flora of Serbia G. dinarica is only distributed in Kosovo. In course of our recent pharmacognostic investigations G. dinarica was found at two localities on the mountain Tara, locality Zvezda (1230 m) and Veliki Kraj (1330 m). Plant material (aerial parts and roots) was collected from June to August 2000-2001. Anatomic and phytochemical analyses were carried out. Preliminary screening of the extracts of the roots and aerial parts of G. dinarica showed the presence of secoiridoids, flavonoids and xanthone compounds. From both extracts three secoiridoids were isolated (swertiamarin, gentiopicrin and sweroside).
AB  - Vrsta Gentiana dinarica Beck. (Gentianaceae) je retka biljna vrsta koja je u Srbiji, prema Flori Srbije, do sada konstatovana samo na Kosovu. U toku terenskih istraživanja, ovu vrstu smo zabeležili na dva lokaliteta na planini Tari i to lokalitet Zvezda (1230 m) i lokalitet Veliki Kraj (1330 m).Specifičnost anatomske građe korena G. dinarica je slabo odrveneo kislem, i tzv. unutarksilemski floem, odnosno grupice floemskih elemenata u centralnom delu sekundarnog drveta.Preliminarno ispitivanje ekstrakata korena i nadzemnog dela G. dinarica pokazao je prisustvo sekoiridoida, flavonoida i ksantona. Od sekoiridoida, izolovani su svercijamarin, genciopikrin i sverozid.

Anatomy of the multifloral sage Salvia officinalis L. subsp. multiflora Gajić (Lamiaceae)

AB  - The multifloral sage (Salvia officinalis L. subsp. multiflora Gajić), the subspecies of the common sage, grows only at termophylus limestone in Southeast of Serbia (Gajić, 1973). According to its endemic character, it has being included in the group of Yugoslav taxa of international significance (Stevanović et al. 1995). The research of the structural attributes of leaves and steams of multifloral sage was performed on material collected from Sićevo gorge in S.E. Serbia. The stem and leaves are covered with the dense indumentum, comprised of different, specifically located glandular and non-glandular trichomes. The anatomic structure of the stem and leaves, along with the specific indumentum features indicate xero-mesomorphic nature of multifloral sage.
AB  - Salvia officinalis L. subsp. multiflora Gajić, je endemit jugoistočne Srbije i usled toga uvrštena u listu jugoslovenskih taksona od međunarodnog značaja. Istraživanje strukturnih odlika listova i stabla višecvetne žalfije urađeno je na materijalu prikupljenom sa lokaliteta Sićevačka klisura, odakle je takson i opisan. Stablo i listovi su pokriveni gustim indumentumom sačinjenim od različitih, specifično raspoređenih mehaničkih i žlezdanih trihoma. Anatomska građa stabla i listova, zajedno sa karakterističnim osobinama indumentuma ukazuju na kseromezomorfnu prirodu višecvetne žalfije.

AB  - Iris reichenbachii Heuffel (Iridaceae) is an endemic plant species of Balkan penninsula and Romania. Pharmacognostic study of this species are not done till now. The microscopic preparations of the leaves were analyzed and specific orientation of vessels were noticed. Secondary metabolites were analyzed in the samples of leaves collected in different vegetative phase, flowers, fruits and rhizomes. In the samples, g-pyrones and secoiridoids were detected. Among γ-pyrones, a xanthone C-glycoside mangiferin were one of the most abundant compound. It's content varied from 0.5%-8.34% depending of plant organ or vegetative phase of development.
AB  - Vrsta Iris reichenbachii je endemit Balkanskog poluostrva i Rumunije. Do sada nije farmakognozijski izučavana. U ovom radu je ispitivana anatomska građa lista pri čemu je uočena specifična orijentacija provodnih snopića. Prisustvo sekundarnih metabolita ispitivano je u uzorcima listova, cvetova, plodova i rizoma. Detektovana su jedinjenja g-pironske strukture (ksantoni, flavonoidi) među kojima se po zastupljenosti u uzorcima, kao i po količini ističe ksanton mangiferin. Sadržaj mangiferina varira zavisno od faze razvića i biljnog organa (0.5%-8.34%). Detektovani su i sekoiridoidi od kojih se u značajnijoj količini pojavljuju svercijamarin i genciopikrin.
