Wild mint (Mentha longifolia) extracts in the production of non-alcoholic beverages

AB  - Isolates of medicinal and aromatic herbs are used as additives in non-alcoholic drinks to improve the sensory characteristics and antioxidant potential. The method of drying the plant material, among other factors, has a profound influence on the chemical composition and pharmacological activities of plant extracts. This paper analyzes the effect of the drying technique (I - natural drying, II - in the laboratory oven, III - in low-temperature condensational drier) on the volatile fraction of the wild mint (Mentha longifolia (L.) Hudson) extract, in order to select an extract of the best quality for preparation of non-alcoholic drinks. The volatile profile of the extracts was determined by the GC-FID and GC-MS techniques, the antimicrobial activity by the microdilution technique, the antioxidant activity by the DPPH and FRAP assays, and the sensory acceptance according to the Regulation on quality for refreshing non-alcoholic beverages. The results showed that piperitone was the major component of the volatile fraction extract of the natural drying herb and low-temperature condensational drier herb (53.9% and 61.1%, respectively), while the extract of laboratory oven herb was rich in menthone (35.5%). At the concentrations in the range of 0.8-3.2 mg/mL the extracts better inhibited the Gram (+) bacteria. The beverage to which the extract of naturally dried wild mint was added, showed the antioxidant activity (9.09 +/- 0.17 mu mol Fe2+/mL by FRAP and 14.00 +/- 3.00 mu L/mL by DPPH method) and good sensorial characteristics (concentration of the extract 0.8 g/L).

AB  - The way of drying the fresh herbal material influences the chemical content and the biological activities of their essential oils. The influence of the different drying methods of the herb Mentha longifolia (L.) Hudson on the antioxidant and antimicrobial activity of the extracted essential oils has been analyzed in this study. Drying has been carried out in three ways: in the natural way, in the laboratory oven (45 degrees C) and in the absorptional low-temperature condensational drier (35 degrees C). The antioxidant activity of the essential oil has been estimated by FRAP and DPPH assays, while the antimicrobial activity has been estimated by the diffusible and micro-delusional method, testing on the nine types of bacteria and two types of fungi. The essential oil obtained from the herb dried in the natural way has shown the highest antioxidant activity and the lowest from the herb dried in the laboratory oven. Bacillus subtilis, Micrococcus luteus and Enterococcus faecalis have shown the highest sensitivity on the three samples. The oil obtained from the herb dried in the absorptional low-temperature drier has shown the strongest antimicrobial effect.

AB  - This study examines the essential oil and the ethanol extract of Artemisia alba Turra herb, in terms of chemical composition, antioxidant and antimicrobial activities in order to select the active pharmacological isolate for making phytopreparation from the group of bitter aromatics and/or bitter liqueurs. Chemical composition of the essential oils and volatile fractions of the extract was determined by GC-FID/GC-MS and non-volatile fractions of the extract by HPLC. The content of total phenolics and flavonoids in the extracts was determined by Folin-Ciocalteu reagent and aluminum chloride, respectively. The antioxidant activity of isolates was tested by FRAP and DPPH tests and the antimicrobial by microdilution method. Major constituents in the essential oil were camphor, artemisia ketone and 1,8-cineole (23.7%, 15.2% and 14.1%, respectively), and in the volatile fraction of the extract, scopoletin (14.0%) and corymbolone (10.3%). The main components in non-volatile fraction of the extract were kaempferol 3-O-(6 ''-O-malonylglucoside)- 7-O-rhamnoside, chlorogenic acid and rutin (16.1%, 11.4% and 9.5% respectively). The content of total phenolics in the extract, expressed as gallic acid was 77.18 mg/g and flavonoids, expressed as rutin 53.80 mg/g of dry extract. Extract exhibited stronger antioxidant activity than essential oil and better antimicrobial activity on a larger number of tested microorganisms.

AB  - Antioxidant and antimicrobial activities as well as total phenols and flavonoids contents of Salvia glutinosa L. (glutinous sage) and Salvia officinalis L. (sage) extracts were studied. Methanol and aqueous ethanol (70% v/v) were used for extraction of bioactive compounds, both in the presence and the absence of ultrasound, from herb and the spent plant material remaining after the essential oil hydrodistillation. The ratio of plant material to extracting solvent was 1:10 g/mL. Antioxidant and antimicrobial activities of the extracts were found to depend on the type of plant material and the extraction conditions. The plant materials from which essential oil had been recovered were proven to be valuable raw materials for making various herbal preparations.
AB  - Ispitivana je antioksidantna i antimikrobna aktivnost, kao i sadržaj ukupnih fenola i flavonoida, ekstrakta lepljive žalfije (Salvia glutinosa L.) i žalfije (kadulje) (Salvia officinalis L.). Ekstrakcija bioaktivnih komponenata je vršena ultrazvukom i klasičnom ekstrakcijom (maceracijom) iz suve herbe, kao i biljnog materijala koji je ostao nakon izolacije etarskih ulja postupkom hidrodestilacije. Za ekstrakciju su korišćeni rastvarači metanol i vodeni rastvor etanola (70% v/v) pri hidromodulu 1:10 (g/mL), pri čemu je upotrebom drugog rastvarača dobijen veći prinos ekstraktivnih supstanci. Antioksidantna aktivnost dobijenih ekstrakata ispitana je DPPH testom, dok je njihova antimikrobna aktivnost određena disk difuzionom metodom. Pokazalo se da navedene aktivnosti ekstrakata zavise od tipa biljnog materijala i uslova ekstrakcije. Nezavisno od tipa rastvarača, biljnog materijala i tehnike ekstrakcije, S. glutinosa je pokazala snažniju antioksidantnu aktivnost, dok je S. officinalis bila bogatija fenolnim komponentama. Takođe, ekstrakti herbe S. glutinosa sadržali su više flavonoida od ekstrakata S. officinalis. Svi etanolni ekstrakti pokazali su veću antioksidantnu aktivnost od metanolnih ekstrakata. Posebno se ističu ekstrakti herbe, koji su bogatiji fenolnim komponentama i flavonoidima od ekstrakata rezidualnog biljnog materijala. Za razliku od ekstrakata S. officinalis, ekstrakti S. glutinosa su pokazali bolji antioksidantni kapacitet. Generalno, ekstrakti S. glutinosa dobijeni iz rezidualnog biljnog materijala, pokazali su jaču antimikrobnu aktivnost protiv svih mikroorganizama u poređenju sa ekstraktima herbe. I u ovom slučaju, etanolni ekstrakti su pokazali jaču antimikrobnu aktivnost od metanolnih ekstrakata. Biljni materijal (SPM) iz kojeg je prethodno izolovano etarsko ulje sadrži i dalje komplekse aktivnih materija koje pokazuju značajne biološke aktivnosti, što ga svrstava u vrednu sirovinu za izradu raznovrsnih fitopreparata.

Monitoring of the photochemical stability of carvedilol and its degradation products by the RP-HPLC method

AB  - A sensitive, selective, precise and stability-indicating, new high-performance liquid chromatographic method for the analysis of carvedilol both as a bulk drug and in formulations was developed and validated. As the method could effectively separate the drug from its degradation products, it can be employed as a stability-indicating one. The method was validated for linearity, selectivity, precision, robustness, LOD, LOQ and accuracy. The chromatographic separation was achieved on a Chromolit RP8e, 100x4.6 mm, analytical column. The mobile phase consisted of a mixture of acetonitrile and water (45:55, V/V) (pH 2.5), pH adjusted with formic acid. The absorbance was monitored with a UV detector at 280 nm and the temperature of the analyses was 40°C. The flow rate was 0.5 mL/min. The linearity (r≥ 0.999), reproducibility (0.68-1.27 %) and recovery (99.71-101.58) were found to be satisfactory. This method enables the simultaneous determination of carvedilol and its degradation products, as well as stability.
AB  - Karvedilol je neselektivni beta blokator sa vazodilatatornim i antioksidativnim svojstvima. Primenjuje se u terapiji hipertenzije, angine pektoris i kongestivne srčane insuficijencije. Razvijena je i validirana nova HPLC metoda za istovremenu kvalitativnu i kvantitativnu analizu karvedilola i njegovih nečistoća kao i za praćenje fotohemijske stabilnosti ovog jedinjenja. Metoda je validirana na linearnost, selektivnost, preciznost, robustnost, LOD, LOQ i tačnost. Optimalni uslovi postignuti su korišćenjem hromatografske kolone Chromolit RP 8e, 100x4,6 mm. Mobilnu fazu predstavlja smeša acetonitrila i vode u odnosu 45:55 pri pH 2,5. Analiza se odvija na temperaturi od 40°C pri protoku 0,5 ml/min a talasna dužina UV detektora iznosi 280 nm. Linearnost (r≥0,999), reproduktivnost (0,68-1,27%), i rikaveri (99,71-101,58) pokazuju zadovoljavajuće vrednosti. Hromatografska analiza je pokazala da se kao glavni proizvod degradacije karvedilola javlja 4-hidroksikarbazol, dok nečistoća C nije prisutna pod ovim uslovima. Predložena RP-HPLC metoda je brza, precizna, tačna, osetljiva, pouzdana i primenljiva za kvalitativnu i kvantitativnu analizu KarvileksR tablete, kao i za praćenje stabilnosti ovog leka.
