AB  - Copper(ii) complexes with an α-diimine show a wide variety of biological activities, such as antibacterial, antifungal, antioxidant and anticancer. In this work, we synthesized and structurally characterized two novel Cu(ii) complexes with methyl 3-formyl-4-hydroxybenzoate (HL) and α-diimines: 2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen). Crystal structure analysis shows that the formulas of the compounds are [Cu(bipy)(L)(BF4)] (1) and [Cu(phen)(L)(H2O)](BF4)·H2O (2), with BF4− as a ligand in complex 1, which is rarely coordinated to metals. Both complexes have a square pyramidal geometry, while DFT calculations showed that the most stable structures of complexes 1 and 2 in a water/DMSO mixture are square-planar derivatives [Cu(bipy)(L)]+ and [Cu(phen)(L)]+. The antibacterial activity of compounds was evaluated in vitro on four Gram-negative and four Gram-positive bacterial strains. Complex 2 showed greater antibacterial activity towards all bacterial strains comparable to the control compound Amikacin. Complex 2 exerted a strong cytotoxic effect against the tested cancer cell lines (IC50 values ranging from 0.32 to 0.44 μM). Both complexes caused apoptotic cell death in HeLa cells and a noticeable in vitro antiangiogenic effect. In the concentration range of 5 to 100 μM, the complexes showed the absence of a genotoxic effect and displayed a protective effect against oxidative DNA damage induced by H2O2 in human peripheral blood cells. The interaction between the compounds and calf-thymus DNA was evaluated by diverse techniques suggesting a tight binding, which was also confirmed by molecular docking. In addition, it was found that the complexes bind tightly and reversibly to bovine and human serum albumin.

AB  - In this article, cytotoxicity, the mechanisms of cytotoxic activity, genotoxicity, and interaction with DNA and proteins, of two Cu(II) complexes with a salicylaldehyde derivative (4-(diethylamino)salicylaldehyde) and α-diimine (2,2′-bipyridine (bipy) and 1,10-phenanthroline (phen)) are reported. Both Cu(II) complexes performed cytotoxic effects against all tested malignant cell lines. Complexes exerted highest cytotoxicity against HeLa and A375 malignant cell lines. The cytotoxic activity of Cu(II) complex with phen as a α-diimine co-ligand was significantly higher in comparison with cytotoxic activity of Cu(II) complex with bipy. Pretreatment with specific inhibitors of caspase-3, caspase-8 or caspase-9, in order to clear up the mode of cell death triggered by two Cu(II) complexes in HeLa cells, indicated the ability of these complexes to induce apoptosis through activation of target caspases. Cu(II)-phen complex exhibited significant antioxidant activity compared with Cu(II)-bipy complex, and showed a better effect on reducing intracellular ROS levels in HeLa cells. Tested complexes did not display genotoxic potential in human peripheral blood leucocytes, but exhibited an antigenotoxic effect in post-treatment, after H2O2 exposure. The study of the in vitro biological properties regarding their affinity towards CT (calf-thymus) DNA and serum albumins showed that the compounds can intercalate to CT DNA, and bind reversibly and tightly to the albumins. Molecular docking studies of the ability of compounds to bind to biomacromolecules are consistent with in vitro studies.

AB  - The isothiocyanato Fe(III) complex with (E)-N, N, N-trimethyl-2-oxo- 2-(2-(1-(pyridin-2-yl)ethylidene)hydrazinyl)ethan-1-aminium was synthesized and characterized by elemental analysis, IR spectroscopy and X-ray analysis. The octahedral geometry of the Fe(III) complex consists of the deprotonated form of the hydrazone ligand coordinated through the pyridine nitrogen, azomethine nitrogen and carbonyl oxygen atoms with three isothiocyanato ligands in the remaining coordination places. The measured effective magnetic moment for Fe(III) complex corresponds to high spin Fe(III) ion. The hydrazone ligand and its Fe(III) complex showed lower activity against the tested microbial strains than standard antimicrobial drugs.

Quantum chemical calculations of the molecular structures and the relative stabilities of linkage isomers of the Cd(II) complex showed that the isomer with N-Cd-N coordination of OCN- is the most stable. The investigated Cd(II) complexes showed lower activity than standard antimicrobial drugs. [GRAPHICS] .

AB  - Two Zn(II) complexes with the condensation product of 2-quinolinecarboxaldehyde and trimethylammonium acetohydrazide chloride (Girard's T reagent) (HLCl) and monodentate pseudohalides (azide and cyanate) have been synthesized and characterized by elemental analysis, IR and NMR spectroscopy, and single-crystal X-ray diffraction. In both complexes, the coordination surroundings of the Zn(II) ions consist of a deprotonated hydrazone ligand coordinated through an NNO set of donor atoms and two monodentate pseudohalides (N-3(-) or NCO-) at the remaining coordination sites. The Zn(II) complexes showed low to moderate activity against laboratory control strains of pathogenic bacteria and fungi. [GRAPHICS] .

AB  - A dinuclear double end-on azido bridged KM complex [Ni2L2(mu-(1),(1)-N-3)(2)(N-3)(2)] (1) with the condensation product of 2-quinolinecarboxaldehyde and trimethylammonium acetohydrazide chloride (Girard's T reagent) (HLCl) has been synthesized and characterized by elemental analysis, IR spectroscopy, single-crystal X-ray diffraction, magnetic measurements and DFT studies. In complex 1 intra-dimer ferromagnetic coupling between Ni2+ ions (J= +12.0(2) cm(-1)) and inter-dimer antiferromagnetic interaction (delta = -0.8(3) cm(-1)) were observed. DFT-BS calculations provided explanation of ferromagnetic exchange coupling in complex 1.

The crystal structure of chloro Cd(II) complex (1) was determined. In 1 and 2, coordination surrounding of Cd(II) consists of deprotonated hydrazone ligand coordinated through NNO-donor atoms and two monodentates at the rest of the coordination places. Quantum chemical calculations of the molecular structures and the relative stabilities of linkage isomers of the Cd(II) complex showed that the isomer with N-Cd-N coordination of OCN- is the most stable. The investigated Cd(II) complexes showed lower activity than standard antimicrobial drugs. [GRAPHICS] .

AB  - Isothiocyanate complexes of Zn(II) and Cd(II) with the condensation product of 2,6-diacetylpyridine and trimethylammoniumacetohydrazide (Girard's T reagent) were synthesized, characterized, and their antimicrobial activities were evaluated. The structures of the complexes were determined by elemental analysis, IR, and NMR spectroscopy. The crystal structure of the Zn(II) complex was also determined. Quantum-chemical calculations of the geometry and total energy of isomers of 2,6-diacetylpyridine-bis(trimethylammoniumacetohydrazone) were performed in vacuum and methanol solution, with the aim to explain conformational behavior and E/Z isomerism of this compound. DFT calculations of the molecular structures and the relative stabilities of linkage isomers of the Cd(II) complex showed that the isomer with N-Cd-N coordination of SCN- is the most stable. Complexes of Zn(II) and Cd(II) exhibited low to moderate activity against the tested microbial strains. [GRAPHICS]

AB  - Complexes of Pd(II) and Co(III) with the condensation derivative of 2-(diphenylphosphino)benzaldehyde and Girard T reagent were synthesized, characterized, and their antimicrobial activities were evaluated. The ligand and the complexes were characterized by elemental analysis, IR and NMR spectroscopies, and X-ray crystallography. In both complexes, the deprotonated ligand was coordinated to the metal through the phosphorus, the imine nitrogen, and the carbonyl oxygen atoms. In the octahedral Co(III) complex, two molecules of ligands were coordinated to metal ion, while square-planar environment of Pd(II) complex was constituted of one tridentate ligand and chloride in the fourth coordination place. The ligand and complexes showed moderate antibacterial activity. AB  - Three square-planar complexes of Ni(II) with condensation derivative of 2-(diphenylphosphino)benzaldehyde and 4-phenylsemicarbazide and monodentate pseudohalides have been synthesized and characterized on the basis of the results of X-ray. NMR and IR spectroscopy and elemental analysis. Investigated complexes exhibited moderate antibacterial and cytotoxic activity. The most pronounced cytotoxic activity (in the range of cisplatin) to HeLa cell line was observed for ligand and all the complexes. Azido complex and ligand induced concentration dependent cell cycle arrest in the S phase, as well as decrease of percentage of cells in G1 phase, without significant increase of apoptotic fraction of cells. The interaction of the azido complex and ligand with CT-DNA results in changes in UV-Vis spectra typical for non-covalent bonding. The observed intrinsic binding constant of azido complex CT-DNA and ligand-CT-DNA were 3.22 x 10(5) M-1 and 2.79 x 10(5) M-1. The results of DNA cleavage experiments showed that azido complex nicked supercoiled plasmid DNA.

The structures of the Pd(II) and Fe(III) complexes in solid state were determined by IR, elemental analysis and X-ray analysis. The structure of Pd(II) complex in solution was determined by NMR spectroscopy. Results of magnetic measurements for Fe(III) complex were reported. In both complexes the ligand is coordinated as tridentate via the phosphorus, the imine nitrogen and the carbonyl oxygen atoms. Results of antimicrobial activity investigation indicate that the activity of the ligand is enhanced upon complexation, and that the MIC values of the iron complex to some bacterial strains are not much higher than those of cefotaxime.

AB  - A Schiff base of 3-acetylpyridine with semicarbazide as well as the corresponding tetrahedral Zn(II) complex were synthesized and characterized by X-ray crystal structure analysis and spectroscopic methods. It is interesting to note that the ligand coordinated as a monodentate although there are several donor atoms in it. Computational studies showed that such structure is more stable than the hypothetical structure with one ligand bound as a bidentate. The complex exibited moderate antibacterial, antifungal and cytotoxic activities while the ligand was mostly inactive. The complex strongly induced formation of reactive oxygen species in tumor cell lines. It also influenced cell cycle progression in tumor cell lines, and induced autophagy. The latter effect is, at least in part, a protective one.

AB  - Acid-base equilibria of Zn(II) and Fe(III) complexes with N',N'2-bis- [(1E)-1-(2-pyridyl)ethylidene]ethanedihydrazide (ligand L1) and N',N'2-bis[(1E)- -1-(2-pyridyl)ethylidene]propanedihydrazide (ligand L2), i.e., [Fe(L1)Cl2(H2O)], [Fe(L2)Cl(H2O)]2+, [Zn(L1)(H2O)3]+ and [Zn(L2)(H2O)2]2+, which expressed cytotoxic activity, were investigated in aqueous media. The equilibrium constants were determined potentiometrically at 25°C at a constant ionic strength of 0.10 mol/dm3 (Na2SO4). The results showed that at pH  lt  8 both the Fe(III) complexes studied here have three, while [Zn(L1)(H2O)3]+ and [Zn(L2)(H2O)2]2+ have one and two titratable protons, respectively. Based on the obtained values for the equilibrium constants, protonation schemes of the examined complexes are proposed.
AB  - U ovom radu su proučavane kiselinsko-bazne ravnoteže u vodenim rastvorima kompleksa Zn(II) i Fe(III) sa N',N'2-bis[(1E)-1-(2-piridil)etiliden]etandihidrazidom (ligand L1) i N',N'2-bis[(1E)-1-(2-piridil)etiliden]propandihidrazidom (ligand L2), tj. [Fe(L1)Cl2(H2O)], [Fe(L2)Cl(H2O)]2+, [Zn(L1)(H2O)3]+, [Zn(L2)(H2O)2]2+, koji pokazuju citotoksičnu aktivnost. Ravnotežne konstante su određivane potenciometrijski, na temperaturi 25°C i pri konstantnoj jonskoj jačini 0,10 mol/dm3 Na2SO4. Utvrđeno je da u intervalu pH  lt  8 ispitivani Fe(III) kompleksi imaju tri, [Zn(L1)(H2O)3]+ jedan, a [Zn(L2)(H2O)2]2+ dva protona koji mogu da se titrišu. Na osnovu dobijenih vrednosti za konstante, pretpostavljene su i odgovarajuće protonacione sheme.

AB  - In this paper the first complexes of M2+ ions (M2+ = Zn2+, Cd2+ and Co2+) with N-benzyloxycarbonyl-S-phenylalaninato ligand (1-3) are described. The new complexes were characterized by means of elemental analysis, IR and UV-vis spectroscopy, molar conductivity measurements and H-1, C-13 and N-15 NMR spectroscopy (1D and 2D). The Co(II) complex adopts an octahedral geometry, and the Zn(II) and Cd(II) complexes adopt a tetrahedral one. For the first time, the antimicrobial activity of N-benzyloxycarbonyl-S-phenylalaninato ligand (N-Boc-S-phe) and the complexes 1-3 was investigated against Gram-positive: Micrococcus luteus, Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis and Bacillus subtilis and Gram-negative bacteria: Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and two strains of the yeast Candida albicans. It was shown that the complexes were effective against most strains. The best activity was detected against the yeast C. albicans.
